Tissue engineering is a field related to regenerative medicine which aims at replacing or regenerating a patient’s tissue, usually using a combination of cells and bioactive material designed to influence cell behaviour. In approaches for bone regeneration, human mesenchymal stem cells (hMSCs) are a common choice because of their ability to proliferate and differentiate into osteoblasts. Harnessing this potential requires biomaterials which promote osteoblastic differentiation, for example by mimicking the conditions in natural bone. Collagen I is a common protein in human bone; it forms fibrils with a characteristic periodic structure, which raises the question whether this morphology has in impact on stem cell fate. Collagen-mimicking nanomaterials can help investigate this question: Gemini surfactants with chiral counterions form twisted bilayers the morphology of which can be tuned by variation of enantiomeric excess, time and temperature. The self-assembled helical nanoribbons which are obtained by this process can be transformed by a sol-gel condensation to form silica nanohelices the size and twist pitch of which resembles that of collagen fibres. The objective of this study is to prepare 2D culture environments featuring these nanomaterals (with and without bioactive peptide functionalisation) to explore the effect of these materials on hMSC differentiation.Silica helices are fabricated by synthesis of surfactants with tartrate as counterion, and organic-inorganic transcription using a silica precursor compound. They can be modified by reaction with APTES and an N-hydroxysuccinimide ester and covalent immobilisation of a peptide. Two peptides were used in this study, one adhesion-promoting peptide and the active domain of the osteogenesis-inducing peptide BMP2. Helices with or without this bioactive functionalisation were covalently grafted to glass substrates using APTES and EDC/NHS-coupling. The presence of peptides on helices was shown by the absorption of helix-grafted peptides bearing the FITC-fluorophore. Successful peptide grafting onto glass surfaces was verified by XPS and fluorescence microscopy. The morphology of helices was monitored with TEM and SEM. SEM images were used to determine the amount of helices on surfaces. HMSCs were cultivated for four weeks on surfaces modified with APTES, peptide(s) or left- or righthanded nanohelices, functionalised or not with bioactive peptide(s). After fixation, the quantities of the osteogenic markers Runx2 and Osteocalcin (OCN) in the cells were evaluated. The results show that BMP2-functionalised surfaces exhibited an elevated level of Runx2 and OCN expression. Some helix-grafted materials exhibited a significantly higher Runx2 and/or OCN expression than the corresponding homogeneous materials, but these differences were not consistent across samples of the same chiral orientation or bioactive functionalisation. Therefore, conclusive general statements about differences in osteogenic effect between helix functionalisations and handednesses aredifficult to make. A potential reason for this is the variability of surface coverage of helix-grafted materials: As the quantity of helices that are immobilised onto the surfaces is lower than expected and varies greatly between the samples, the number of cells that are not in contact with the helices might change as well, which can lead to false negatives.The results of a proteomic experiment have shown which proteins are differentially expressed in cells cultured on helices with or without BMP-functionalisation, compared to bare glass. Comparison with other proteomic studies shows that proteins which are known to be upregulated during osteogenic differentiation are overexpressed most frequently in cells cultured on BMPmodified helices. The proteins that were identified with this method might serve as starting point for future investigations., Un patient peut souffrir d’une perte de substance osseuse de taille critique suite à des accidents ou des pathologies. Comme les cellules souches mésenchymateuses sont capables de différencier en plusieurs phénotypes différents, il est indispensable de les diriger dans la direction désirée. Plusieurs facteurs qui influencent le devenir cellulaire ont été identifiés, comme certains peptides bioactifs, des facteurs mécaniques comme la rigidité, ou la topographie de surface de matériaux.Dans la matrice extracellulaire naturelle du tissu osseux, les cellules souches mésenchymateuses sont entourées d’une variété de principes actifs, dont le plus abondant est le collagène I. Cette protéine s’assemble pour former des nanofibres qui présentent une nanomorphologie périodique avec une périodicité bien définie. La question posée au début de ce travail était: Cette structure a-t-elle un impact sur la différenciation des cellules souches?Pour étudier l’impact de la périodicité nanofibrillaire, nous proposons dans ce travail de recherche l’utilisation d’hélices modèles qui miment en partie la morphologie du collagène. Les hélices nanométriques auto-assemblées des surfactants gemini peuvent avoir un pas d’hélice et un diamètre similaires à ceux du collagène. La modulabilité de ces paramètres et la possibilité de modifier ces structures par des molécules bioactives permettent de moduler les caractéristiques des nanoobjets et d’étudier l’impact de ces nanomatériaux sur les cellules souches mésenchymateuses.

Consider the diagonal action of the projective group $\PGL_3$ on $n$ copies of ${\mathbb P}^2$. In addition, consider the action of the symmetric group $\Sigma_n$ by permuting the copies. In this paper we find a set of generators for the invariant field of the combined group $\Sigma_n \times \PGL_3$. As the main application, we obtain a reconstruction principle for point configurations in ${\mathbb P}^2$ from their sub-configurations of five points. Finally, we address the question of how such reconstruction principles pass down to subgroups.

This dissertation studies invariant rings of finite groups acting linearly on a multivariate polynomial ring over a Dedekind domain or a more general ring. The thesis contains generalizations of several well-known theorems from invariant theory over fields. The question when a ring of invariants over a Dedekind domain is isomorphic to a polynomial ring is studied as well as factoriality and the quasi-Gorenstein property of such rings. Finally the thesis deals with invariant rings of point stabilizers in this setting. Diese Dissertation behandelt Invariantenringe endlicher Gruppen, die linear auf einem multivariaten Polynomring über einem Dedekindring oder einem allgemeineren Ring operieren. Die Arbeit enthält Verallgemeinerungen einiger klassischer Sätze aus der Invariantentheorie über Körpern. Die Frage, wann ein Invariantenring über einem Dedekindring isomorph zu einem Polynomring ist, wird behandelt, ebenso die Faktorialität und die quasi-Gorenstein-Eigenschaft solcher Invariantenringe. Schließlich werden Invariantenringe von Punktstabilisatoren in dieser Situation betrachtet.

This thesis studies separating invariants of finite algebraic groups acting on affine varieties through automorphisms. We investigate the question: what restrictions does the existence of a separating set of small size, or a separating algebra with interesting structural properties impose on the group action. Theorems of Serre, Dufresne, Kac-Watanabe and Gordeev, and Dufresne and Jeffries on invariant rings and separating algebras of linear representations are extended to this more general situation. Die vorliegende Arbeit behandelt separierende Invarianten endlicher algebraischer Gruppen, die auf affinen Varietäten durch Automorphismen operieren. Wir untersuchen die Frage, welche Einschränkungen die Existenz einer separierenden Menge kleiner Mächtigkeit oder einer separierenden Algebra mit interessanten strukturellen Eigenschaften an die Gruppenoperation stellt. Sätze von Serre, Dufresne, Kac-Watanabe und Gordeev, und Dufresne und Jeffries über Invariantenringe bzw. separierende Algebren von linearen Darstellungen werden auf diese allgemeinere Situation übertragen.

The method of Buchberger allows to effectively solve the membership problem in polynomial ideals and many other interesting problems. Mayr and Meyer showed that this is very expensive in the worst case. So the problem has to be specialized for more efficient computations. As previous results show, the complexity of the membership problem is mainly related to the degrees of the representation problem and Gröbner bases which are studied in the first part of the thesis. The main contributions are upper and lower bounds for Gröbner bases depending on the ideal dimension and some results for toric ideals. In the second part, these findings are applied to questions of complexity. The presentation comprises an incremental space-efficient algorithm for the computation of Gröbner bases, an algorithm in polylogarithmic space for the membership problem in toric ideals and the space-efficient computation of the radicals of low-dimensional ideals. Mit Buchberger's Algorithmus lässt sich u.a. das Membership-Problem in Polynomidealen effektiv lösen. Mayr und Meyer haben jedoch gezeigt, dass dies im schlimmsten Fall sehr aufwendig ist. Deshalb müssen einfache Spezialfälle des Problems identifiziert werden. Bekannterweise hängt die Komplexität des Membership-Problems primär von den Graden des Darstellungsproblems und der Gröbner-Basen ab, welche im ersten Teil dieser Arbeit betrachtet werden. Die wichtigsten Beiträge sind Gradschranken für Gröbner-Basen in Abhängigkeit von der Dimension des Ideals und Schranken für torische Ideale. Im zweiten Teil der Arbeit wird darauf aufbauend die Komplexität diverser Algorithmen beschrieben. Diese umfassen die inkrementelle platzeffiziente Berechnung von Gröbner-Basen, einen Algorithums in polylogarithmischem Platz für das Membership-Problem in torischen Idealen und die platzeffiziente Berechnung von Radikalen von niedrigdimensionalen Idealen.

This thesis is composed of several different parts. We start with an investigation of an important problem in computer vision. An appropriate mathematical modeling of this problem motivates a problem in invariant theory, the examination of the natural action of the group PGL_{m+1} x S_n on the set of n-point configurations (P^m_K)^n (for some infinite field K and some m, n in N). This in turn leads to an investigation of algorithms for fields. We develop an algorithm for the intersection of fields (in special cases) and a method for testing whether a field is algebraically closed in another field. Moreover, we give an algorithm for testing simpleness of a field extension as well as finding - if applicable - a generating element. The latter leads to a new proof of a generalized version of the Theorem of Lüroth. Another approach to solve problems in field theory is given by the theory of cross-sections of rational maps. We provide a survey on cross-sections and give a criterion for their existence. This yields an algorithm for testing field membership. Finally, we come back to the examination of the natural action of the group PGL_{m+1} x S_n on the set of n-point configurations. We determine generators of the corresponding invariant field and investigate their separating properties. Die vorliegende Arbeit setzt sich aus verschiedenen Teilen zusammen. Wir beginnen mit der Untersuchung eines wichtigen Problems aus der "computer vision". Eine mathematische Modellierung dieses Problems motiviert ein Problem aus der Invariantentheorie, nämlich die Untersuchung der natürlichen Operation der Gruppe PGL_{m+1} x S_n auf der Menge der n-Punktkonfigurationen (P^m_K)^n (für einen unendlichen Körper K und m, n in N. Das wiederum motiviert eine Untersuchung von Algorithmen für Körper. Wir entwickeln einen Algorithmus zur Berechnung des Schnittes von Körpern (für spezielle Fälle) und ein Verfahren um zu testen, ob ein Körper in einem anderen Körper algebraisch abgeschlossen ist. Ferner geben wir einen Algorithmus an, der eine Körpererweiterung auf Einfachheit testet und gegebenenfalls ein erzeugendes Element findet. Letzteres führt zu einem neuen Beweis einer verallgemeinerten Version des Satzes von Lüroth. Eine weitere Herangehensweise, um Probleme aus der Körpertheorie zu lösen, liefert die Theorie der Sektionen von rationalen Abbildungen. Wir geben einen Überblick über Sektionen und entwickeln ein Kriterium für deren Existenz. Dies führt zu einem Algorithmus, der das Enthaltensein in einem Körper testet. Schließlich kommen wir auf die Untersuchung der Operation der Gruppe PGL_{m+1} x S_n auf der Menge der n-Punktkonfigurationen zurück. Wir bestimmen Erzeuger des zugehörigen Invariantenkörpers und untersuchen deren Trennungseigenschaften.

In this thesis, we present algorithms for the computation of invariant rings. In the first part, we give an algorithm for the computation of invariants of finite groups acting on affine algebras via K-algebra automorphisms. In the second part, we consider linear algebraic groups acting regularly on an irreducible affine variety. In particular, we give an algorithm to compute invariants of unipotent groups. In the third part, we study linear algebraic groups acting regularly on quasi-affine varieties. We present algorithms for the computation of invariant rings for the cases where the group is finite or unipotent. Finally, an outline is given of how the problem of computing invariants of arbitrary linear algebraic groups acting regularly on factorial varieties can be reduced to the problem of computing invariants of one-dimensional tori. Die Arbeit untersucht Algorithmen zur Berechnung von Invariantenringen. Im ersten Teil wird ein Algorithmus entwickelt um Invarianten endlicher Gruppen zu berechnen, die auf affinen Algebren mittels K-Algebra-Automorphismen operieren. Der zweite Teil behandelt reguläre Operationen von linearen algebraischen Gruppen auf irreduziblen affinen Varietäten. Es wird insbesondere ein Algorithmus zur Berechnung von Invariantenringen unipotenter Gruppen angegeben. Im dritten Teil werden reguläre Operationen von linearen algebraischen Gruppen auf quasi-affinen Varietäten untersucht. Es werden Algorithmen zur Berechnung von Invarianten endlicher und unipotenter Gruppen entwickelt. Schließlich wird noch ein Verfahren skizziert, welches das Problem der Berechnung von Invarianten beliebiger linearer algebraischer Gruppenoperationen auf faktoriellen Varietäten zurückführt auf das Problem der Berechnung von Invarianten eindimensionaler Tori.
