This paper proposes a mixture regression model-based method for drug sensitivity prediction. The proposed method explicitly addresses two fundamental issues in drug sensitivity prediction, namely, population heterogeneity and feature selection pertaining to each of the subpopulations. The mixture regression model is estimated using the imputation-conditional consistency algorithm, and the resulting estimator is consistent. This paper also proposes an average-BIC criterion for determining the number of components for the mixture regression model. The proposed method is applied to the CCLE dataset, and the numerical results indicate that the proposed method can make a drastic improvement over the existing ones, such as random forest, support vector regression, and regularized linear regression, in both drug sensitivity prediction and feature selection. The p-values for the comparisons in drug sensitivity prediction can reach the order O(10−8) or lower for the drugs with heterogeneous populations. [ABSTRACT FROM AUTHOR]

High dimensional biomedical data contain tens of thousands of features, accurate and effective identification of the core features in these data can be used to assist diagnose related diseases. However, there are often a large number of irrelevant or redundant features in biomedical data, which seriously affect subsequent classification accuracy and machine learning efficiency. To solve this problem, a novel filter feature selection algorithm based on redundant removal (FSBRR) is proposed to classify high dimensional biomedical data in this paper. First of all, two redundant criteria are determined by vertical relevance (the relationship between feature and class attribute) and horizontal relevance (the relationship between feature and feature). Secondly, to quantify redundant criteria, an approximate redundancy feature framework based on mutual information (MI) is defined to remove redundant and irrelevant features. To evaluate the effectiveness of our proposed algorithm, controlled trials based on typical feature selection algorithm are conducted using three different classifiers, and the experimental results indicate that the FSBRR algorithm can effectively reduce the feature dimension and improve the classification accuracy. In addition, an experiment of small sample dataset is designed and conducted in the section of discussion and analysis to clarify the specific implementation process of FSBRR algorithm more clearly. Research: The body composition model is closely related to the physiological characteristics of the human body. At the same time there can be a large number of physiological characteristics, many of which may be redundant or irrelevant. In existing human physiological feature selection algorithms, it is difficult to overcome the impact that redundancy and irrelevancy may have on human body composition modeling. This suggests a role for selection algorithms, where human physiological characteristics are identified using a combination of filtering and improved clustering. To do this, a feature filtering method based on Hilbert-Schmidt dependency criteria is first of all used to eliminate irrelevant features. After this, it is possible to use improved Chameleon clustering to increase the combination of sub-clusters amongst the characteristics, thereby removing any redundant features to obtain a candidate feature set for human body composition modeling. Method We report here on the use of an algorithm to filter the characteristic parameters in INBODY770 (this paper used INBODY 770 as body composition analyzer.) measurement data, which has three commonly-used impedance bands (1 kHZ, 250 kHZ, 500 kHZ). This algorithm is able to filter out parameters that have a low correlation with body composition BFM. The algorithm is also able to draw upon improved clustering techniques to reduce the initial feature set from 29 parameters to 10 parameters for any parameters of the 250 kHZ band that remain after filtering. In addition, we also examined the impact of different sample sizes on feature selection. Result: The proposed algorithm is able to remove irrelevant and redundant features and the resulting correlation between the model and the body composition (BFM which is a whole body fat evaluation can better assess the body's overall fat and muscle composition.) is 0.978, thereby providing an improved model for prediction with a relative error of less than 0.12. Feature selection and classification are the main topics in microarray data analysis. Although many feature selection methods have been proposed and developed in this field, SVM-RFE (Support Vector Machine based on Recursive Feature Elimination) is proved as one of the best feature selection methods, which ranks the features (genes) by training support vector machine classification model and selects key genes combining with recursive feature elimination strategy. The principal drawback of SVM-RFE is the huge time consumption. To overcome this limitation, we introduce a more efficient implementation of linear support vector machines and improve the recursive feature elimination strategy and then combine them together to select informative genes. Besides, we propose a simple resampling method to preprocess the datasets, which makes the information distribution of different kinds of samples balanced and the classification results more credible. Moreover, the applicability of four common classifiers is also studied in this paper. Extensive experiments are conducted on six most frequently used microarray datasets in this field, and the results show that the proposed methods have not only reduced the time consumption greatly but also obtained comparable classification performance. This paper proposes a new support vector machine (SVM) optimization scheme based on an improved chaotic fly optimization algorithm (FOA) with a mutation strategy to simultaneously perform parameter setting turning for the SVM and feature selection. In the improved FOA, the chaotic particle initializes the fruit fly swarm location and replaces the expression of distance for the fruit fly to find the food source. However, the proposed mutation strategy uses two distinct generative mechanisms for new food sources at the osphresis phase, allowing the algorithm procedure to search for the optimal solution in both the whole solution space and within the local solution space containing the fruit fly swarm location. In an evaluation based on a group of ten benchmark problems, the proposed algorithm’s performance is compared with that of other well-known algorithms, and the results support the superiority of the proposed algorithm. Moreover, this algorithm is successfully applied in a SVM to perform both parameter setting turning for the SVM and feature selection to solve real-world classification problems. This method is called chaotic fruit fly optimization algorithm (CIFOA)-SVM and has been shown to be a more robust and effective optimization method than other well-known methods, particularly in terms of solving the medical diagnosis problem and the credit card problem. Cancer classification and feature (gene) selection plays an important role in knowledge discovery in genomic data. Although logistic regression is one of the most popular classification methods, it does not induce feature selection. In this paper, we presented a new hybrid L1/2 +2 regularization (HLR) function, a linear combination of L1/2 and L2 penalties, to select the relevant gene in the logistic regression. The HLR approach inherits some fascinating characteristics from L1/2 (sparsity) and L2 (grouping effect where highly correlated variables are in or out a model together) penalties. We also proposed a novel univariate HLR thresholding approach to update the estimated coefficients and developed the coordinate descent algorithm for the HLR penalized logistic regression model.
