Effluent found in the pulp and paper industry can cause considerable damage if it is discharged untreated, because of the high biochemical and chemical oxygen demands. Electrocoagulation is a physicochemical process widely used in industrial wastewater treatment. The removal of different pollutants depends on the sample type and operating conditions. The aim of this research was to evaluate the efficiency of an electrocoagulation system for COD removal from recycled paper production effluent via aluminum and iron electrodes. Different operational parameters, such as the electrolysis time (5 min to 15 min), current density (7 A/m² to 11 A/m²), and distance between each electrode (5 mm to 20 mm), were evaluated. The turbidity, total suspended solids, chlorides, sulfates, and COD had removal efficiencies of 92.7%, 91.3%, 70.4%, 66.6%, and 64%, respectively. A polynomial model was generated to estimate the optimum conditions for COD removal. The optimum times for the current densities 7 A/m², 8 A/m², 9 A/m², 10 A/m², and 11 A/m² were 39.5 min, 39.5 min, 35.7 min, 34.1 min, and 32.8 min, respectively, with a 15-mm electrode gap. We describe herein a method of recharging used commercial spin columns or assembling homemade spin columns using filter paper as binding material for cost-effective, low throughput nucleic acid purification. The efficiency of filter paper-based spin columns was evaluated for purification of nucleic acids from various sources. Following protocols of commercial kits, we found filter paper to be a useful binding material for purification of nucleic acids, including plant genomic DNA, plant total RNA, PCR products, and DNA from agarose gels. However, filter paper has a weak binding affinity to plasmid DNA in tested miniprep protocols. Protocols for the use of filter paper recharged spin columns or homemade spin columns for low throughput purification of plant genomic DNA and total RNA with unused commercial kit buffers or less expensive homemade buffers are presented. Zacharias Dische's discovery of feedback inhibition in metabolism was one of the most important in the history of biochemistry. However, his paper was written and published under very difficult circumstances in wartime and passed almost completely unnoticed. It is almost never cited, and the discovery itself is usually attributed to later work of others. Here I provide a discussion of Dische's work, a translation of his paper into English, and a transcription of the original French version, which is almost unobtainable anywhere. Recent studies have reported incipient morphological caste dimorphism in the Van der Vecht organ size of some temperate Polistes paper wasps. Whether species other than the temperate ones show a similar pattern remains elusive. Here, we have studied some Neotropical Polistes species. By comparing females collected through the year, we showed caste related differences in the size of the Van der Vecht organ in P. ferreri (body size corrected Van der Vech organ size of queens = 0.45 ± 0.06, workers = 0.38 ± 0.07 mm2, p = 0.0021), P. versicolor (body size corrected Van der Vech organ size of queens = 0.54 ± 0.11, workers = 0.46 ± 0.09 mm2, p = 0.010), but not P. simillimus (body size corrected Van der Vech organ size of queens = 0.52 ± 0.05, workers = 0.49 ± 0.06 mm2, p = 0.238). Therefore, it seems that queens and workers of some Neotropical Polistes have diverged in their ontogenic trajectory of the Van der Vecht organ size, providing clear evidence for incipient morphological caste dimorphism. As Polistes are distributed mostly in the tropics, we propose that physical caste differences may be widespread in the genus. Also, we highlight that morphological divergence in the queen–worker phenotypes may have started through differential selection of body structures, like the Van der Vecht organ. Biosensors have emerged as a valuable tool with high specificity and sensitivity for fast and reliable detection of hazardous substances in drinking water. Numerous substances have been addressed using synthetic biology approaches. However, many proposed biosensors are based on living, genetically modified organisms and are therefore limited in shelf life, usability and biosafety. We addressed these issues by the construction of an extensible, cell-free biosensor. Storage is possible through freeze drying on paper. Following the addition of an aqueous sample, a highly efficient cell-free protein synthesis (CFPS) reaction is initiated. Specific allosteric transcription factors modulate the expression of ‘superfolder’ green fluorescent protein (sfGFP) depending on the presence of the substance of interest. The resulting fluorescence intensities are analyzed with a conventional smartphone accompanied by simple and cheap light filters. An ordinary differential equitation (ODE) model of the biosensors was developed, which enabled prediction and optimization of performance. With an optimized cell-free biosensor based on the Shigella flexneri MerR transcriptional activator, detection of 6 μg/L Hg(II) ions in water was achieved. Furthermore, a completely new biosensor for the detection of gamma-hydroxybutyrate (GHB), a substance used as date-rape drug, was established by employing the naturally occurring transcriptional repressor BlcR from Agrobacterium tumefaciens. In this paper, we present a facile and cost-effective method to obtain superhydrophobic filter paper and demonstrate its application for efficient water/oil separation. By coupling structurally distinct organosilane precursors (e.g., octadecyltrichlorosilane and methyltrichlorosilane) to paper fibers under controlled reaction conditions, we have formulated a simple, inexpensive, and efficient protocol to achieve a desirable superhydrophobic and superoleophilic surface on conventional filter paper. The silanized superhydrophobic filter paper showed nanostructured morphology and demonstrated great separation efficiency (up to 99.4%) for water/oil mixtures. The modified filter paper is stable in both aqueous solutions and organic solvents, and can be reused multiple times. The present study shows that our newly developed binary silanization is a promising method of modifying cellulose-based materials for practical applications, in particular the treatment of industrial waste water and ecosystem recovery. The activated sludge process within the pulp and paper industry is generally run to minimize the production of waste activated sludge (WAS), leading to high electricity costs from aeration and relatively large basin volumes. In this study, a pilot-scale activated sludge process was run to evaluate the concept of treating the wastewater at high rate with a low sludge age. Two 150 L containers were used, one for aeration and one for sedimentation and sludge return. The hydraulic retention time was decreased from 24 hours to 7 hours, and the sludge age was lowered from 12 days to 2-4 days. The methane potential of the WAS was evaluated using batch tests, as well as continuous anaerobic digestion (AD) in 4 L reactors in mesophilic and thermophilic conditions. Wastewater treatment capacity was increased almost four-fold at maintained degradation efficiency. The lower sludge age greatly improved the methane potential of the WAS in batch tests, reaching 170 NmL CH4/g VS at a sludge age of 2 days. In addition, the continuous AD showed a higher methane production at thermophilic conditions. Mixed microbial culture from paper mill treatment system was enriched on polyvinyl alcohol (PVA) medium and its PVA biodegradation capacity was investigated as a function of change in the initial pH values and PVA concentrations. The optimum pH value at 0.75 gl-1 initial PVA concentration was determined to be 8. PVA biodegradation by mixed microbial culture was investigated at the range of 0.68 - 15.5 gl-1 initial PVA concentrations. The mixed microbial culture was found to degrade within 5 days 98.6-88.5% of the initial PVA levels which ranged from 0.68 to 1.14 gl-1. The mixed microbial culture was found to remove more than 90% of the initial PVA concentrations, within the range of 3.13 to 6.64 g1-1 in 16-25 days of incubation at 35 °C. [ABSTRACT FROM AUTHOR]

Amphimeriasis, a fish-borne zoonotic disease caused by the liver fluke Amphimerus spp., has recently been reported as an emerging disease affecting an indigenous Ameridian group, the Chachi, living in Ecuador. The only method for diagnosing amphimeriasis was the microscopic detection of eggs from the parasite in patients' stool samples with very low sensitivity. Our group developed an ELISA technique for detection of anti-Amphimerus IgG in human sera and a molecular method based on LAMP technology (named LAMPhimerus) for specific and sensitive parasite DNA detection. The LAMPhimerus method showed to be much more sensitive than classical parasitological methods for amphimeriasis diagnosis using human stool samples for analysis. The objective of this work is to demonstrate the feasibility of using dried stool samples on filter paper as source of DNA in combination with the effectiveness of our previously designed LAMPhimerus assay for successfully Amphimerus sp. detection in clinical stool samples. A total of 102 untreated and undiluted stool samples collected from Chachi population were spread as thin layer onto common filter paper for easily transportation to our laboratory and stored at room temperature for one year until DNA extraction. When LAMPhimerus method was applied for Amphimerus sp. DNA detection, a higher number of positive results was detected (61/102; 59.80%) in comparison to parasitological methods (38/102; 37.25%), including 28/61 (45.90%) microscopy-confirmed Amphimerus sp. infections. The diagnostic parameters for the sensitivity and specificity werecalculated for our LAMPhimerus assay, which were 79.17% and 65.98%, respectively. We demonstrate, for the first time, that common filter paper is useful for easy collection and long-term storage of human stool samples for later DNA extraction and molecular analysis of human-parasitic trematode eggs. This simple, economic and easily handling method combined with the specific and sensible LAMPhimerus assay has the potential to beused as an effective molecular large-scale screening test for amphimeriasis-endemic areas. India has the highest burden of Tuberculosis (TB) and multidrug-resistant TB (MDR-TB) worldwide. Innovative technology is the need of the hour to identify these cases that remain either undiagnosed or inadequately diagnosed due to the unavailability of appropriate tools at primary healthcare settings. We developed and evaluated 3 kits, namely ‘TB Detect’ (containing BioFM-Filter device), ‘TB Concentration and Transport’ (containing Trans-Filter device) and ‘TB DNA Extraction’ kits. These kits enable bio-safe equipment-free concentration of sputum on filters and improved fluorescence microscopy at primary healthcare centres, ambient temperature transport of dried inactivated sputum filters to central laboratories and molecular detection of drug resistance by PCR and DNA sequencing (Mol-DST). In a 2-site evaluation (n = 1190 sputum specimens) on presumptive TB patients, BioFM-Filter smear exhibited a significant increase in positivity of 7% and 4% over ZN smear and LED-FM smear (p<0.05), respectively and an increment in smear grade status (1+ or 2+ to 3+) of 16% over ZN smear and 20% over LED-FM smear. The sensitivity of Mol-DST in presumptive MDR-TB and XDR-TB cases (n = 148) was 90% for Rifampicin (95% confidence interval [CI], 78–96%), 84% for Isoniazid (95% CI, 72–92%), 83% for Fluoroquinolones (95% CI, 66–93%) and 75% for Aminoglycosides (95% CI, 35–97%), using phenotypic DST as the reference standard. Test specificity was 88–93% and concordance was ~89–92% (κ value 0.8–0.9). The patient-friendly kits described here address several of the existing challenges and are designed to provide ‘Universal Access’ to rapid TB diagnosis, including drug-resistant disease. Their utility was demonstrated by application to sputum at 2 sites in India. Recent studies suggest that reactions in aqueous microcompartments can occur at significantly different rates than those in the bulk. Most studies have used electrospray to generate a polydisperse source o f highly charged microdroplets, leading to multiple confounding factors potentially influencing reaction rates (e.g., evaporation, charge, and size). Thus, the underlying mechanism for the observed enhancement remains unclear. We present a new type of electrodynamic balance— the branched quadrupole trap (BQT)—which can be used to study reactions in microdroplets in a controlled environment. The BQT allows for condensed phase chemical reactions to be initiated by colliding droplets with different reactants and levitating the merged droplet indefinitely. The performance of the BQT is characterized in several ways. Sub-millisecond mixing times as fast as ~400 μs are measured for low velocity (~0.1 m /s) collisions of droplets with <40 μm diameters. The reaction of o-phthalaldehyde (OPA) with alanine in the presence of dithiolthreitol is measured using both fluorescence spectroscopy and single droplet paper spray mass spectrometry. The bimolecular rate constant for reaction of alanine with OPA is found to be 84 ± 10 and 67 ± 6 M-1 s-1 in a 30 μm radius droplet and bulk solution, respectively, which demonstrates that bimolecular reaction rate coefficients can be quantified using merged microdroplets and that merged droplets can be used to study rate enhancements due to compartmentalization. Products of the reaction of OPA with alanine are detected in single droplets using paper spray mass spectrometry. Type 1 diabetes (T1D) is increasing in incidence and predictable with measurement of serum islet autoantibodies (iAb) years prior to clinical disease onset. Identifying iAb positive individuals reduces diabetic ketoacidosis and identifies individuals for T1D prevention trials. However, large scale screening for iAb remains challenging as assays have varying sensitivities and specificities, insulin autoantibodies remain difficult to measure and venipuncture is generally required to obtain serum. We developed an approach to reliably measure all four major iAb, including insulin autoantibodies, from dried blood spots (DBS) on filter-paper. By spiking iAb positive serum into iAb negative whole blood in a dose titration, we optimized the conditions for autoantibody elution from filter paper as measured by fluid phase radioimmunoassays. After assessing stability of measuring iAb from DBS over time, we then screened iAb from DBS and the corresponding serum in new-onset T1D (n = 52), and controls (n = 72) which included first-degree relatives of T1D patients. iAb measured from eluted DBS in new-onset T1D strongly correlated with serum measurements (R2 = 0.96 for mIAA, GADA = 0.94, IA-2A = 0.85, ZnT8A = 0.82, p<0.01 for each autoantibody). There were no false positives in control subjects, and 5/6 with previously unknown iAb positivity in sera were detected using DBS. Mass production of vermicompost using suitable species of earthworms and selecting target organic waste materials has appeared to be a great development in the realm of biotechnological research for the sustainable eco-management. Although, for the bioconversion of organic wastes to vermicompost, suitable earthworm species play major roles, a hoard of bacterial assemblages by virtue of production of different enzymes facilitate the process of vermicomposting. The present study has documented the roles of vermicompost associated bacteria in combating, preventing, and controlling of cancer so as to open a new vista not only in the field of vermitechnology but also on biomedical research. Earthworms’ associated bacterial metabolic products having their unique physicochemical excellence have gained importance due to their roles as a facilitator of apoptosis (programed cell death in a MCF-7 cell line). The antioxidant and anticancer activities of ethyl acetate extracts’ of vermicompost associated bacterium Bacillus anthracis were undertaken by antioxidant assay which revealed maximum DPPH radical scavenging effect (75.79 ± 5.41%) of the extracts’ at 9 00 μg ml-1. Furthermore, the crude extracts obtained from the same bacteria were found to decrease the activity of SOD (superoxide dismutase) with the increase in doses. MTT assay showed potent cytotoxic activity against human breast adenocarcinoma cells (MCF-7) with the IC50 value of 46.64 ± 0.79 μg ml-1. It was further confirmed through Hoechst 33258 staining of nuclear fragmentation assay and DNA fragmentation analysis. Western blotting test has confirmed a downregulation of Akt upon application of crude extracts. Increase of SOD activity along with decrease of Akt level reflects that the mode of action is entirely PI-3K dependent. This study tends to indicate that B. anthracis isolated from vermicompost could be potentially explored for the development of new therapeutic agents, especially against cancer. Waste paper can serve as a feedstock for ethanol production due to being rich in cellulose and not requiring energy-intensive thermophysical pretreatment. In this study, an efficient process was developed to convert waste paper to ethanol. To accelerate enzymatic saccharification, pH of waste paper slurry was adjusted to 4.5–5.0 with H 2 SO 4 . Presaccharification and simultaneous saccharification and fermentation (PSSF) with enzyme loading of 40 FPU/g waste paper achieved an ethanol yield of 91.8% and productivity of 0.53 g/(L h) with an ethanol concentration of 32 g/L. Fed-batch PSSF was used to decrease enzyme loading to 13 FPU/g waste paper by feeding two separate batches of waste paper slurry. Feeding with 20% w/w waste paper slurry increased ethanol concentration to 41.8 g/L while ethanol yield decreased to 83.8%. To improve the ethanol yield, presaccharification was done prior to feeding and resulted in a higher ethanol concentration of 45.3 g/L, a yield of 90.8%, and productivity of 0.54 g/(L h). Ethanol fermentation recovered 33.2% of the energy in waste paper as ethanol. The biochemical methane potential of the stillage eluted from ethanol fermentation was 270.5 mL/g VTS and 73.0% of the energy in the stillage was recovered as methane. In this study, the chemical composition, fiber morphology, pulp, and paper properties of the pomegranate tree (Punica granatum L.) were determined. Stem and branch wood were analyzed separately. Kraft and kraft- anthraquinone (AQ) methods were used for the pulping processes. Stem wood showed superiority to branch wood with longer, wider, thicker-walled fibers. The holocellulose, α-cellulose, and lignin contents of branch and stem wood were 72.98%-73.50%, 38.37%-39.92%, and 21.04%-25.29%, respectively. In addition, the yield of kraft pulp made from stem wood was higher than that of branch wood. Many results have been obtained when studying scientific papers citations databases in a network perspective. Articles can be ranked according to their current in-degree and their future popularity or citation counts can even be predicted. The dynamical properties of such networks and the observation of the time evolution of their nodes started more recently. This work adopts an evolutionary perspective and proposes an original algorithm for the construction of genealogical trees of scientific papers on the basis of their citation count evolution in time. The fitness of a paper now amounts to its in-degree growing trend and a “dying” paper will suddenly see this trend declining in time. It will give birth and be taken over by some of its most prevalent citing “offspring”. Practically, this might be used to trace the successive published milestones of a research field. In this paper, three different Zn(II) complexes with (E)-2-(2-(1-(6-bromopyridin-2-yl)ethylidene)hydrazinyl)-N,N,N-trimethyl-2-oxoethan-1-aminium chloride (HLCl) have been synthesized and characterized by single crystal X-ray diffraction, elemental analysis, IR and NMR spectroscopy. All complexes are mononuclear, with the ligand (L) coordinated in a deprotonated formally neutral zwitterionic form via NNO donor set atoms. Complex 1 forms an octahedral geometry with the composition [ZnL2](BF4)2, while complexes 2 [ZnL(NCO)2] and 3 [ZnL(N3)2] form penta-coordinated geometry. Density functional theory (DFT) calculations were performed to enhance our understanding of the structures of the synthesized complexes and the cytotoxic activity of the complexes was tested against five human cancer cell lines (HeLa, A549, MDA-MB-231, K562, LS 174T) and normal human fibroblasts MRC-5. Additionally, antibacterial and antifungal activity of these complexes was tested against a panel of Gram-negative and Gram-positive bacteria, two fungal strains, and a yeast strain. It is noteworthy that all three complexes show selective antifungal activity comparable to that of amphotericin B. Molecular docking analysis predicted that geranylgeranyl pyrophosphate synthase, an enzyme essential for sterol biosynthesis, is the most likely target for inhibition by the tested complexes. Enzymes are protein molecules that play a crucial role in various biological processes in living organisms. They function as catalysts in biological reactions such as digestion, metabolism, DNA replication and other physiological processes. Furthermore, enzymes are widely used in food production, pharmaceuticals and biofuel production. In these industries, they accelerate desired chemical reactions as biocatalysts. Therefore, applying computational methods and data-driven algorithms to predict enzyme properties is essential. Over the past decade, deep learning has made remarkable advancements in science and technology. Deep learning is a subset of machine learning algorithms that rely on artificial neural networks. These algorithms can be employed for supervised, semi-supervised and unsupervised learning. Here, to provide an update on the current literature, we provide an overview of various deep learning algorithms and recent advancements in their application to enzyme science. These applications can generally be categorized into diverse subjects: function prediction, enzyme kinetic parameters prediction, enzyme-substrate identification, condition optimization, thermophilic property prediction, enzyme catalytic site prediction and enzyme design. In conclusion, we discuss the convergence of enzyme science and deep learning, highlighting the potential opportunities and challenges. Artificial intelligence algorithms have diverse applications in enzyme science. Culture conditions for the production of cellulase-free endo-β-1,4-xylanase by an alkalophilic strain of Bacillus subtilis SD8 (BsSD8-xylanase) isolated from paper mill effluent were optimized using 2 factorial design and response surface methodology. Statistical analysis of results showed that the linear and quadratic terms and interaction of 4 variables (temperature, pH, inoculum size and xylan concentration) had significant effects. The optimal conditions for production of the enzyme were: 37.5°C, initial pH of 8.5, inoculum size of 8.5% and xylan concentration of 2%. The predicted and verified BsSD8-xylanase activities under optimal conditions were 8.18 IU/mL. Analysis revealed that the enzyme gene consists of an ORF of 642 bp encoding a protein of 213 amino acids with a molecular mass of 23.5 kDa and pI of 9.67. Secondary and tertiary structure prediction using SOPMA and SWISS model showed a maximum homology of 95.14% with β1,4-xylanase (PDB Id.: 1BVV) of Bacillus circulans obtained from protein data bank. An inexpensive, disposable and highly selective microfluidic paper-based analytical device (μPAD) is described for the determination of ammonia (molecular ammonia and ammonium cation) in wastewaters which implements for the first time a gas-diffusion separation step on a paper-based platform. Its hydrophilic reagent zones were defined by printing filter paper with a hydrophobic paper sizing agent using a conventional inkjet printer. The sample was introduced into the sodium hydroxide impregnated sample zone of the μPAD. This allowed the quantitative conversion of the ammonium ion to molecular ammonia which diffused across the hydrophobic microporous Teflon membrane of the device into an adjacent hydrophilic reagent zone containing the acid-base indicator 3-nitrophenol or bromothymol blue. The change in indicator color was measured using a desktop scanner for ammonia quantification. Under optimal conditions, the μPAD is characterized by a limit of detection of 0.8 and 1.8 mg N L-1 and repeatability of 3.1 and 3.7% (n ⩾ 10, 20 mg N L-1), expressed as relative standard deviation, in the case of 3-nitrophenol or bromothymol blue, respectively. This μPAD was used successfully for the determination of ammonia in sewage and soil water samples. The small dimensions, minimal reagent consumption, low cost, simplicity of operation, and possibility of using a portable scanner make the proposed μPAD suitable for on-site ammonia monitoring in contaminated environmental waters and domestic, agricultural and industrial wastewaters. To explore the citation evolution of papers published in the same year but different month, we selected papers from a discipline (physical geography), a subject (diabetes: endocrine and metabolism) and a journal (Journal of Biological Chemistry) published in 2005 as research objects. These papers were divided into six groups according to the difference in publication month, and we analyzed citations to these papers for the 9 years after publication. The results showed that within 5 years after papers from physical geography were published, the overall differences in citations of papers in different groups were statistically significant (P < 0.05); after that, the differences were not statistically significant. Within 5 years after papers from diabetes (endocrine and metabolism) were published, the overall differences in citations of papers in different groups were statistically significant (P < 0.05); thereafter, the differences were not statistically significant. Within 7 years after papers from the Journal of Biological Chemistry were published, the overall differences in citations of papers in different groups were statistically significant (P < 0.05); thereafter, the differences were not statistically significant. Citations of papers followed the same pattern irrespective of discipline, subject or journal: citations of papers published in the same year but different month were obviously different in the first few publishing years, but as time went on, only the difference in publication month in a calendar year did not affect the papers' longer-term citation. Paper to Plastics (P2P) is an interdisciplinary program that combines chemistry and biology in a research setting. The goal of this project is 2-fold: to engage students in scientific research and to educate them about sustainability and biodegradable materials. The scientific aim of the project is to recycle unwanted office paper to the useful biodegradable polymer poly(lactic acid) (PLA). Through this program, students learn firsthand how chemistry and biology interact to form useful materials from waste. Students combine biological techniques, such as enzymatic digestion and fermentation, with chemical techniques, such as distillation and catalysis, to accomplish the conversion of waste paper into PLA. Through this summer program, students ultimately become familiar with diverse laboratory techniques, while learning how their scientific interests can be used to address important social problems. Abstract: A cost-effective HPLC method for determination of pyrimethamine (PYR) in human whole blood samples dried on filter paper (Whatman®) is reported. Trimethoprim (TMP) was used as an internal standard. Whole blood spiked with PYR was transferred (100μl) onto filter paper and dried at room temperature. Capillary blood samples (100μl) after ingestion of three tablets of sulfadoxine-pyrimethamine (SP) by one subject were also tested. PYR and an internal standard (IS) TMP were extracted into di-isopropyl ether as bases and then re-extracted with 150μl mobile phase. A C-18 column was used and the mobile phase consisted of phosphate buffer (0.05M, pH 5):acetonitrile:concentrated perchloric acid (750:300:2.5, v/v/v). The absorbances of PYR and IS were monitored at 270nm. The limit of quantification was 40ng/ml. The within- and between-assay coefficient of variations were <10% at the limit of quantification. Objectives: Schizophrenia is a group of severe psychiatric disorders with high heritability but only low odds ratios of risk genes. Despite progress in the identification of pathophysiological processes, valid biomarkers of the disease are still lacking. Methods: This comprehensive review summarises recent efforts to identify genetic underpinnings, clinical and cognitive endophenotypes and symptom dimensions of schizophrenia and presents findings from neuroimaging studies with structural, functional and spectroscopy magnetic resonance imaging and positron emission tomography. The potential of findings to be biomarkers of schizophrenia is discussed. Results: Recent findings have not resulted in clear biomarkers for schizophrenia. However, we identified several biomarkers that are potential candidates for future research. Among them, copy number variations and links between genetic polymorphisms derived from genome-wide analysis studies, clinical or cognitive phenotypes, multimodal neuroimaging findings including positron emission tomography and magnetic resonance imaging, and the application of multivariate pattern analyses are promising. Conclusions: Future studies should address the effects of treatment and stage of the disease more precisely and apply combinations of biomarker candidates. Although biomarkers for schizophrenia await validation, knowledge on candidate genomic and neuroimaging biomarkers is growing rapidly and research on this topic has the potential to identify psychiatric endophenotypes and in the future increase insight on individual treatment response in schizophrenia. The effect of pulp and paper-mill effluents on terrestrial animals was investigated using male albino rats as a test organism, under controlled laboratory conditions. Effluent consumption for 15 days caused a reduction in the relative weight of the testis (mg/100 g body weight), compared to the control rats. The effects were not so pronounced in other regions of the male reproductive tract. Biochemical analysis revealed a drop in the activity of epididymal α-glucosidase in the effluent-treated animals. Although sperm collected from the cauda epididymis appeared morphologically normal, a decline in total sperm count and number of motile sperm was seen. The circulating level of testosterone in effluent-treated rats was slightly lower in some groups of rats given effluent treatment compared to controls. The level of progesterone and oestradiol, however, remained unaffected. Hematological and serological analyses suggest that effluents could produce anemia, liver and kidney dysfunction in rats. This inference stems from the altered blood cell counts, enhanced bilirubin and urea concentration in blood and high activity of serum GOT and GPT observed in effluent fed animals compared to controls. Background & objectives: Dried blood spotted on to filter paper has been found suitable for a large number of studies. In tropical countries with varying temperature conditions the use of dried blood needs to be validated. We carried out this study to assess the use of blood spotted filter paper as a transport system to study genotyping ofApo E gene. Methods: Fifty five patients visiting Cardiothoracic Neuroscience Centre (CNC) OPD at the All India Institute of Medical Sciences (AIIMS), New Delhi, and referred for lipid investigations to Cardiac Biochemistry Laboratory were selected at random. Blood was spotted on to Whatman 3 MM filter paper, dried and stored at room temperature. Genomic DNA was extracted and genotyping was carried out at the end of 0, 3 and 12 months. The study was further validated using samples collected on to filter paper from four centres and stored for eight years at room temperature. The temperature and humidity conditions of the centre varied widely. Results: Fifty five samples collected on to filter paper showed exact match of the genotyping when compared to fresh blood. In dried blood samples collected and stored for 1 yr at room temperature DNA extraction and apo E genotyping was done successfully. Interpretation & conclusions: The present results showed the feasibility of using dried blood samples on filter paper for apo E genotyping in tropical temperature. The ability to study chemical dynamics on ultrafast timescales has greatly advanced with the introduction of X-ray free electron lasers (XFELs) providing short pulses of intense X-rays tailored to probe atomic structure and electronic configuration. Fully exploiting the full potential of XFELs requires specialized experimental endstations along with the development of techniques and methods to successfully carry out experiments. The liquid jet endstation (LJE) at the Linac Coherent Light Source (LCLS) has been developed to study photochemistry and biochemistry in solution systems using a combination of X-ray solution scattering (XSS), X-ray absorption spectroscopy (XAS), and X-ray emission spectroscopy (XES). The pump–probe setup utilizes an optical laser to excite the sample, which is subsequently probed by a hard X-ray pulse to resolve structural and electronic dynamics at their intrinsic femtosecond timescales. The LJE ensures reliable sample delivery to the X-ray interaction point via various liquid jets, enabling rapid replenishment of thin samples with millimolar concentrations and low sample volumes at the 120 Hz repetition rate of the LCLS beam. This paper provides a detailed description of the LJE design and of the techniques it enables, with an emphasis on the diagnostics required for real-time monitoring of the liquid jet and on the spatiotemporal overlap methods used to optimize the signal. Background Positive allosteric modulators (PAMs) of the GABAB receptor constitute a new class of GABAB-receptor ligands. GABAB PAMs reproduce several pharmacological effects of the orthosteric GABAB receptor agonist, baclofen, although displaying a better safety profile. Aims This paper reviews the reducing or, frequently, even suppressing effects of all GABAB PAMs tested to date on multiple alcohol-related behaviours in laboratory rodents exposed to validated experimental models of human alcohol use disorder. Results Acute or repeated treatment with CGP7930, GS39783, BHF177, rac -BHFF, ADX71441, CMPPE, COR659, ASP8062, KK-92A, and ORM-27669 reduced excessive alcohol drinking, relapse- and binge-like drinking, operant alcohol self-administration, reinstatement of alcohol seeking, and alcohol-induced conditioned place preference in rats and mice. Conclusions These effects closely mirrored those of baclofen; notably, they were associated to remarkably lower levels of tolerance and toxicity. Providing hands-on practical education without access to laboratories during the Covid-19 pandemic has required creativity and innovation. In this paper, co-authored by academic staff and students, we describe an at-home mobile phone-based 'spectrophotometer' experiment used in an introductory undergraduate biology course. Using colour picker apps, a smartphone can be used to quantify concentration, which was used to compare the strengths of different brands of tea. The protocol is designed to be low-cost and safe to perform outside of a laboratory. Students used the methods to learn important biochemical methods such as preparing dilutions, constructing calibration curves, normalising data and testing a hypothesis. We reflect on the experience of developing and using the protocol from a staff and student perspective, which highlights the advantages of this approach in terms of student independence and inclusivity. We also suggest alternative experiments that could be performed using the protocol. We encourage biology educators to think creatively about the possibilities for using mobile phones or at-home experiments in their teaching. The authors, a chemistry professor and a librarian, used a qualitative survey to assess student perceptions of a Wikipedia editing assignment that they included in a large upper-level biochemistry course. The assignment was initially intended as a public-facing alternative to a short research paper, emphasizing information literacy and scientific literacy. The goal of the survey was to use the results to enhance the assignment. The results of the survey and research for the literature review inspired a novel approach to the assignment using the perspective of metaliteracy. End of life care in dementia is important to avoid patients receiving unnecessary investigations and treatments. In this paper, the authors discuss the final stage of dementia, illustrating this with a case example reflecting what is frequently encountered in hospital practice. The distinction is drawn between 'terminal inanition' of dementia and the more protracted cachexia, sarcopenia and progressive frailty that accompanies advancing dementia, with best practice clinical care considerations for palliative care outlined. Rainbow trout were exposed to a secondary treated, thermomechanical/bleached kraft pulp and paper effluent in 12,000-L, flow-through exposure tanks at an environmental research facility located at a pulp and paper mill in Kawerau, New Zealand. Trout (age, 21 years) were obtained from a local hatchery and exposed either to upstream river water or a nominal concentration of 12% (v/v) effluent diluted in upstream river water. Three treatment groups were used: Effluent exposure that started approximately three months before gonadal growth (eight-month total exposure), effluent exposure that started approximately halfway through gonadal development (two-month total exposure), and trout exposed to reference water alone for the total duration of the experiment. Trout were sacrificed just before spawning; exposure, growth, and reproductive endpoints were assessed during and at the termination of the experiment. Reduction in growth was observed in both sexes in the eight-month treatment group relative to the river water reference treatment group. No differences were observed in condition factor or liver size in either treatment. Females in the eight-month exposure group also had significantly lower ovary weight. The two-month exposure group showed no differences from the reference group in growth or somatic indices. Estradiol and testosterone were reduced in blood samples taken from the eight-month exposure group by four months into the experiment as compared to the reference treatment. Steroid and vitellogenin levels in individual female trout from this treatment were significantly correlated with gonadosomatic indices (GSI) measured at the termination of the experiment. The GSI was not correlated strongly or consistently with pregnenolone, nor were any treatment-related preg-nenolone differences observed, indicating that the steroid hormone reductions likely were not related to cholesterol side-chain cleavage. The xanthones represent an important and widely spread class of specialized metabolites in the plant kingdom, fungi, and lichens. Their aromatic and highly oxygenated structures owe them the ability to undergo a series of chemical reactions to form an important number of derivatives as well as to exert a large scale of excellent activities that particularly increased their interest for several researchers. Amongst the reactions observed on their basic nucleus, C-glycosylation leads to the formation of C-glycosylxanthones, a rare subclass of xanthones encountered in nature. Until July 2023, the literature survey on chemical and pharmacological investigations of medicinal plants indicated that a total of 41 distinct natural C-glycosylxanthones have been isolated from 31 plant species belonging to 10 families. Mangiferin (1) was the most reported one and the most biologically screened. Antioxidant activity was the most performed test on isolated compounds, while some strong and good anti-inflammatory, antiplasmodial, and α-glucosidase inhibitory activities have been also reported in the literature for some C-glycosylxanthones. This paper is a mini-review summarizing the occurrence, chemistry, and biological activities of C-glycosylxanthones. Abstract: A bioanalytical method for the determination of lumefantrine in 100μl blood applied onto sampling paper, by solid-phase extraction and liquid chromatography, has been developed and validated. Whatman 31 ET Chr sampling paper was pre-treated with 0.75M tartaric acid before sampling capillary blood to enable a high recovery of lumefantrine. Lumefantrine was extracted from the sampling paper, then further purified using solid-phase extraction and finally quantified with HPLC. The between-day variation was below 10% over the range 0.4–25μM. The lower limit of quantification was 0.25μM in 100μl capillary blood. No decrease in lumefantrine concentration in dried blood spot is seen after 4 months storage at 22°C. The method was also evaluated in field samples from patients in Tanzania after treatment with lumefantrine/artemether. Lumefantrine could be estimated accurately enough to assess bioavailability and treatment compliance on day 7 (i.e. 4 days after the last dose) after a standard regimen with the lumefantrine/artemether combination. 'Paper-based microfluidics' or 'lab on paper,' as a burgeoning research field with its beginning in 2007, provides a novel system for fluid handling and fluid analysis for a variety of applications including health diagnostics, environmental monitoring as well as food quality testing. The reasons why paper becomes an attractive substrate for making microfluidic systems include: (1) it is a ubiquitous and extremely cheap cellulosic material; (2) it is compatible with many chemical/biochemical/medical applications; and (3) it transports liquids using capillary forces without the assistance of external forces. By building microfluidic channels on paper, liquid flow is confined within the channels, and therefore, liquid flow can be guided in a controlled manner. A variety of 2D and even 3D microfluidic channels have been created on paper, which are able to transport liquids in the predesigned pathways on paper. At the current stage of its development, paper-based microfluidic system is claimed to be low-cost, easy-to-use, disposable, and equipment-free, and therefore, is a rising technology particularly relevant to improving the healthcare and disease screening in the developing world, especially for those areas with no- or low-infrastructure and limited trained medical and health professionals. The research in paper-based microfluidics is experiencing a period of explosion; most published works have focused on: (1) inventing low-cost and simple fabrication techniques for paper-based microfluidic devices; and (2) exploring new applications of paper-based microfluidics by incorporating efficient detection methods. This paper aims to review both the fabrication techniques and applications of paper-based microfluidics reported to date. This paper also attempts to convey to the readers, from the authors' point of view the current limitations of paper-based microfluidics which require further research, and a few perspective directions this new analytical system may take in its development. Abstract: Lipophilic low molar-mass constituents in wood chips for the paper industry result in low quality pulp, pitch deposition, and effluent toxicity. New biotechnological solutions such as fungal pre-treatment of wood chips can reduce pitch problems. This laboratory-scale study focuses on the potential and limitations of a fungal bio-treatment of Norway spruce chips with the white-rot fungus Trametes versicolor. Different fungal treatment conditions were compared. A 4-week fungal treatment reduced the concentration of resin acids and triglycerides by 40% and 100%, respectively, but neither lowered the energy requirements of the TMP process nor significantly affected the morphological fiber characteristics and the physical pulp properties. The pre-treatment led to slightly poorer optical properties. The Trametes versicolor fungal treatment contributed to a less toxic effluent and improved the biodegradability. At the Washington DC Pharmacogenomics in Drug Development and Regulatory Decision-Making: Workshop III – Three Years of Promise, Proposals and Progress on Optimizing the Benefit/Risk of Medicines (11–13 April 2005), one break-out session (Track 2) focused on co-development of therapeutic drug and diagnostic products. The Food and Drug Administration (FDA) released a draft concept paper shortly before the workshop was to convene. Track 2 was a forum for initial discussion of the content of the concept paper, and industry's initial reactions. After the workshop, formal commentaries on the co-development concept paper were submitted by several trade associations (e.g., Pharmaceutical Research and Manufacturers of America (PhRMA), Advanced Medical Technology Association (AdvaMed), American Association for Clinical Chemistry) and individual companies to FDA's Docket No. 2004N-0279. Plants commonly suffer from phosphorus (P) deficiency in calcareous soils. Plant responses to P application on such soils mostly show poor correlation with their soil test P values. Experiments were conducted on 24 different soil samples under laboratory and greenhouse conditions to illustrate the relationship of various inorganic P fractions in different calcareous soils with P uptake by plants, P extraction by iron‐impregnated filter paper, and P soil test values estimated by 0.5M NaHCO 3 and ammonium bicarbonate diethylene triamine penta‐acetic acid. Total P in the 24 soils ranged from 652 to 1245 mgkg -1 with a mean of 922 mgkg -1 . A major proportion (98%) of inorganic P was in HCl‐P (Ca‐bound) form. The HCl‐P (Ca‐bound) ranged from 296 to 729 with a mean of 480 mgkg -1 . The iron (Fe) and aluminum (Al)‐P (NaOH‐P) ranged from 0.92 to 12 mgkg -1 with a mean of 1.57 mgkg -1 . The Fe‐P (citrate‐dithionite bicarbonate) ranged from 0.22 to 4.40 mgkg -1 with a mean of 5.99 mgkg -1 . Data regarding P release from the soil matrix obtained by desorption with iron‐impregnated filter paper was best described by the Elovich equation. Range of slope and intercept values were found to be 5.48 to 17.3 and 17.23 to 56.27 mgkg -1 , respectively. Intercept values calculated for the Elovich equation may be related to labile P initially available for plant uptake in soils. Nearly 100 years ago Michaelis and Menten published their now classic paper [Michaelis, L., and Menten, M. L. (1913) Die Kinetik der Invertinwirkung. Biochem. Z. 49, 333-369] in which they showed that the rate of an enzyme-catalyzed reaction is proportional to the concentration of the enzyme-substrate complex predicted by the Michaelis-Menten equation. Because the original text was written in German yet is often quoted by English-speaking authors, we undertook a complete translation of the 1913 publication, which we provide as Supporting Information. Here we introduce the translation, describe the historical context of the work, and show a new analysis of the original data. In doing so, we uncovered several surprises that reveal an interesting glimpse into the early history of enzymology. In particular, our reanalysis of Michaelis and Menten's data using modern computational methods revealed an unanticipated rigor and precision in the original publication and uncovered a sophisticated, comprehensive analysis that has been overlooked in the century since their work was published. Michaelis and Menten not only analyzed initial velocity measurements but also fit their full time course data to the integrated form of the rate equations, including product inhibition, and derived a single global constant to represent all of their data. Abstract: We revisit an old Russian paper by V.V. Dynnik, R. Heinrich and E.E. Sel’kov (1980a,b) describing: “A mathematical model of carbohydrate energy metabolism. Interaction between glycolysis, the Krebs cycle and the H-transporting shuttles at varying ATPases load”. We analyse the model mathematically and calculate the control coefficients as a function of ATPase loads. We also evaluate the structure of the metabolic network in terms of elementary flux modes. We show how this model can respond to an ATPase load as well as to the glucose supply. We also show how this simple model can help in understanding the articulation between the major blocks of energetic metabolism, i.e. glycolysis, the Krebs cycle and the H-transporting shuttles. Interdisciplinary research in biotechnology and related scientific areas has increased tremendously over the past decade. This rapid pace, in conjunction with advances in microfabricated systems, computer hardware, bioengineering and the availability of low-powered miniature components, has now made it feasible to design bio-inspired materials, sensors and systems with tremendous potential for defence and security applications. To realize the full potential of biotechnology and bio-inspiration, there is a need to define specific requirements to meet the challenges of the changing world and its threats. One approach to assisting the defence and security communities in defining their requirements is through the use of a conceptual model. The distributed or intelligent autonomous sensing (DIAS) system is one such model. The DIAS model is not necessarily aimed at a single component, for instance a sensor, but can include a system, or even a system of systems in the same way that a single organism, a multi-cellular organism or group of organisms is configured. This paper provides an overview of the challenges to and opportunities for bio-inspired sensors and systems together with examples of how they are being implemented. Examples focus on both learning new things from biological organisms that have application to the defence and security forces and adapting known discoveries in biology and biochemistry for practical use by these communities.
