Abstract The bioaccessibility of environmental contaminants has been assessed widely using in vitro simulation; however, the physiological parameters used vary greatly. In this study, we assessed the influence of various physiological parameters and food material on the oral or inhalation bioaccessibility of PM 2.5 -bound hydrophobic organic contaminants (HOCs), including halogenated flame retardants (HFRs), organophosphorus flame retardants (OPFRs), and polycyclic aromatic hydrocarbons (PAHs). The results showed that physiologically based pepsin and pancreatin have a small influence on the HOC liberation from particles. The bioaccessibility increased dramatically when the bile salt concentrations exceeding the critical micelle concentration, and application of porcine bile salts probably lead to underestimated bioaccessibility. Protein and carbohydrates significantly increased the bioaccessibility of most HOCs, while a significant bioaccessibility reduction was caused by green tea. The bioaccessibility of most HOCs was not promoted by liquor under normal physiological condition, but was significantly promoted under fast condition. Long residence time of PM 2.5 in the lung (15 days) would result in higher mobilization of PAHs into the lung fluid than short time (one day). However, the inverse time-dependence for OPFRs suggests degradation in the lung fluid. A mechanism of hydrolysis of organophosphorus ester is hypothesized, and the half lives ranged from 17 to 90 days. Combustion of domestic solid fuels is a significant source of polycyclic aromatic hydrocarbons (PAHs). Some oxygenated PAHs (o-PAHs) and PAHs with molecular weight of 302 (MW302 PAHs) are more toxic than the traditional 16 priority PAHs, whereas their emissions were much less elucidated. This study characterized the size-dependent emissions of parent PAHs (p-PAHs), o-PAHs, and MW302 PAHs from various combustion sources. The estimated emission factors (eEFs) from biomass burning sources were highest for most of the PAHs (391–8928 μg/kg), much higher than that of anthracite coal combustion (43.0–145 μg/kg), both which were operated in an indoor stove. Cigarette smoking had a high eEF of o-PAHs (240 ng/g). MW302 PAHs were not found in the emissions of smoking, cooking, and vehicular exhausts. Particle-size distributions of PAHs were compound- and source-dependent, and the tendency to associate with smaller particles was observed especially in biomass burning and cigarette smoking sources. Furthermore, the inter-source differences in PAH eEFs were associated with their dominance in fine particles. PAH composition profiles also varied with the particle size, showing increasing contributions of large-molecule PAHs with decreasing sizes in most cases. The size distributions of p-PAHs are much more significantly dependent on their n-octanol/air partition coefficients and vapor pressures than those of o-PAHs, suggesting differences in mechanisms governing their distributions. Several molecular diagnostic ratios (MDRs), including two based on MW302 PAHs, specific to these combustion scenarios were identified. However, the MDRs within some sources are also strongly size-dependent, providing a new explanation for the uncertainty in their application for source identification of PAHs. Climate change and geological events have long been known to shape biodiversity, implying that these can likewise be viewed from a biological perspective. To study whether plants can shed light on this, and how they responded to climate change there, we examined Oreocnide , a genus widely distributed in SE Asia. Based on broad geographic sampling with genomic data, we employed an integrative approach of phylogenomics, molecular dating, historical biogeography, and ecological analyses. We found that Oreocnide originated in mainland East Asia and began to diversify ∼6.06 Ma, probably in response to a distinct geographic and climatic transition in East Asia at around that time, implying that the last important geological change in mainland SE Asia might be 1 Ma older than previously suggested. Around six immigration events to the islands of Malesia followed, indicating that immigration from the mainland could be an underestimated factor in the assembly of biotic communities in the region. Two detected increases of diversification rate occurred 3.13 and 1.19 Ma, which strongly implicated climatic rather than geological changes as likely drivers of diversification, with candidates being the Pliocene intensification of the East Asian monsoons, and Pleistocene climate and sea level fluctuations. Distribution modelling indicated that Pleistocene sea level and climate fluctuations were inferred to enable inter-island dispersal followed by allopatric separation, underpinning radiation in the genus. Overall, our study, based on multiple lines of evidence, linked plant diversification to the most recent climatic and geological events in SE Asia. We highlight the importance of immigration in the assembly and diversification of the SE Asian flora, and underscore the utility of plant clades, as independent lines of evidence, for reconstructing recent climatic and geological events in the SE Asian region. Formin-like 3 ( FMNL3 ), a member of diaphanous-related formins subfamily, plays an important role in cytoskeleton reorganization, cell adhesion and cancer cell invasion in vitro. This study aimed to explore the expression of FMNL3 in colorectal carcinoma (CRC) cell-lines and tissues, and further evaluate its prognostic value and correlation with the clinicopathological parameters, and also investigate the effects of FMNL3 gene silencing on the growth and metastasis of CRC in vivo. Immunohistochemical analysis showed that FMNL3 protein was distributed in a punctuate aggregation pattern and located mainly in the cytoplasm of glandular cavity side, close to the nucleus of CRC cells. The positive rate of FMNL3 expression was 87.5% (84/96) in CRC, which was significantly higher than that in adjacent normal mucosa (30%, 9/30). Moreover, FMNL3 protein expressed far more in primary CRC with metastasis and corresponding lymph nodes metastatic CRC than in primary CRC without metastasis. Increased expression of FMNL3 was closely correlated with tumor size, differentiation, serosal invasion, and both lymph node metastasis and distant metastasis. However, it was not correlated with patients' age and gender. According to Kaplan–Meier survival analyses, patients with FMNL3 high expression level had lower overall survival rate than that with FMNL3 low expression level. Univariate and multivariate analyses revealed that high FMNL3 expression was a significant and independent prognostic predictor of patients with CRC. In addition, FMNL3 mRNA and protein levels were substantially up-regulated in CRC-metastasis-derived cell lines, as compared to those in primary-CRC-derived ones. FMNL3 gene silencing suppressed the growth and metastasis of CRC in vivo. In conclusion, FMNL3 plays an important role in the progression and metastasis of CRC and may be a novel potential prognostic predictor and therapeutic target for patients with CRC. Abstract: We have constructed a recombinant adenovirus expression vector carrying the human neurotrophin-3 (NT-3) receptor TrkC (tyrosine protein kinase C) gene (rAd-TrkC; 2478 bp) and confirmed the expression of the encoded TrkC in green fluorescent protein (GFP)-murine neural stem cells (NSCs) by reverse transcription polymerase chain reaction (RT-PCR), Western blot analysis, and immunocytochemistry. The activity of the expressed rAd-TrkC was verified in vitro by evaluating dose-related responses of NSCs to NT-3, a TrkC specific ligand. TrkC-GFP-NSCs had a significantly higher percentage of neuronal differentiation when treated with NT-3 relative to the rAd-LacZ control cells (55.2% vs. 29.8%; P <0.05, χ 2 test). Thus, our rAd-TrkC vector can transfect NSCs and produce functional TrkC receptors to promote neuronal differentiation of NSCs. [Copyright &y& Elsevier]

Recent studies revealed the un-negligible impact of airborne organophosphate esters (OPEs) on phosphorus (P)-limited ecosystems. Subtropical forests, the global prevalence P-limited ecosystems, contain canopy structures that can effectively sequester OPEs from the atmosphere. However, little is known about the behavior and fate of OPEs in subtropical forest ecosystem, and the impact on the P cycling in this ecosystem. OPE concentrations in the understory air (at two heights), foliage, and litterfall were investigated in a subtropical forest in southern China. The median ∑OPE concentrations were 3149 and 2489 pg/m3 in the upper and bottom air, respectively. Foliage exhibited higher ∑OPE concentrations (median = 386 ng/g dry weight (dw)) compared to litter (median = 267 ng/g dw). The air OPE concentrations were ordered by broadleaved forest > mixed forest > coniferous forest, which corresponds to the results of canopy coverage or leaf area index. The spatial variation of OPEs in foliage and litter was likely caused by the leaf surface functional traits. Higher OPE concentrations were found in the wet season for understory air while in the dry season for foliage and litter, which were attributed to the changes in emission sources and meteorological conditions, respectively. The inverse temporal variation suggests the un-equilibrium partitioning of OPEs between leaf and air. The OPE concentrations during the litter-incubation presented similar temporal trends with those in foliage and litter, indicating the strong interaction of OPEs between the litter layer and the near-soil air, and the efficient buffer of litter layer played in the OPEs partitioning between soil and air. The median OPEs-associated P deposition fluxes through litterfall were 270, 186, and 249 μg P/m2·yr in the broadleaved, mixed, and coniferous forests, respectively. Although the fluxes accounted for approximately 0.2% of the total atmospheric P deposition, their significance to this P-limited ecosystem may not be negligible. Accurate prediction and measurement of yield stress are crucial for optimizing sludge treatment and disposal. However, the differences and applicability of various methods for measuring yield stress are subjects of ongoing debate. Meanwhile, literature on measuring sludge yield stress is limited to low solid concentrations (TS <10%), understanding and studying the yield stress of medium to high solid concentration sludge is crucial due to increasingly stringent standards for sludge treatment and disposal. So, this study employed a rotational rheometer to measure sludge yield stress across a wide range of TS (4–50%) using steady shear, dynamic oscillatory shear, and transient shear. The study derived significant conclusions by comparing and summarizing the applicability and limitations of each testing method: Dynamic oscillatory shear methods, including G′- σ curve method, γ - σ curve method, and G** γ c method can measure sludge yield stress ranging from 4% to 40% TS, while other methods are restricted to low or limited solid concentrations; The G' = G″ method, utilizing the intersection of G′ and G″ curves, consistently yields the highest value for yield stress when 4%≤ TS ≤ 12%; The rotational rheometer cannot measure sludge yield stress when the solid concentration exceeds 40% TS; The relationship between sludge yield stress and solid concentration is stronger as a power-law for TS ≤ 25%, transitioning to linear for higher concentrations (28%≤ TS <40%). This study systematically explores the applicability and limitations of various measurement methods for characterizing sludge yield stress across a wide range of solid concentrations, providing valuable guidance for scientific measurement and highlighting challenging research issues. Exploring moisture distribution, especially bound water content, is vital for studying and applying sludge dewatering. The differential scanning calorimetry (DSC) method has been extensively utilized for the quantitative characterization of moisture distribution in sludge. However, this method has certain limitations, such as low reproducibility of results, leading to controversial parameter values in different papers and hindering result comparison. In this study, we investigated the influence of key sample attributes on measuring sludge bound water using the DSC method.The findings demonstrated that the moisture content and mass of sludge samples substantially influenced the reproducibility and stability of DSC test results. To ensure data reliability, the moisture content of the sludge sample should be minimized and kept below 84%, with the mass not exceeding 10 mg. Compared to the influence of sludge moisture content and sample mass, the heating rate (1⁓5 °C/min) minimally affected DSC test results. This study offers a comprehensive insight into how sample attributes and test parameters affect the quantitative characterization of bound water in sludge using the DSC method. Furthermore, practical strategies are presented to enhance the method's applicability in sludge bound water characterization. Hypoxia was proven to cause brain cell apoptosis and autophagy. Herein, we tested the influences of propofol, a commonly used intravenous sedative hypnotic drug, on apoptosis and autophagy aroused by hypoxia stimulation in PC-12 and HT-22 cells. Followed by hypoxia and/or propofol treatment, cell viability of PC-12 and HT-22 cells, apoptosis and autophagy, along with microRNA-137 (miR-137) expression were measured, respectively. Then, miR-137 inhibitor was transfected to silence miR-137. Whether miR-137 took part in the impacts of propofol on hypoxia-exposed cells was explored. Finally, the activities of PI3K/AKT/mTOR and ERK pathways were measured. Hypoxia stimulation aroused cell apoptosis and elevated cell autophagy in PC-12 and HT-22 cells. Propofol weakened the apoptosis and autophagy of PC-12 and HT-22 cells aroused by hypoxia. Moreover, propofol elevated the miR-137 level in PC-12 and HT-22 cells. Silencing miR-137 declined the influences of propofol on hypoxia-induced injuries. Besides, propofol promoted PI3K/AKT/mTOR and ERK pathways activation in hypoxia-exposed cells through raising miR-137. Abstract CSPGs are components of the extracellular matrix in the nervous system, where they serve as cues for axon guidance during development. After a peripheral nerve injury, CSPGs switch roles and become axon inhibitors and become diffusely distributed at the injury site. To investigate whether the spatial distribution of CSPGs affects their role, we combined in vitro DRG cultures with CSPG stripe or coverage assays to simulate the effect of a patterned substrate or dispersive distribution of CSPGs on growing neurites. We observed neurite steering at linear CSPG interfaces and neurite inhibition when diffused CSPGs covered the distal but not the proximal segment of the neurite. The repellent and inhibitory effects of CSPGs on neurite outgrowth were associated with the disappearance of focal actin filaments on growth cones. The application of an actin polymerization inducer, jasplakinolide, allowed neurites to break through the CSPG boundary and grow on CSPG-coated surfaces. The results of our study collectively reveal a novel mechanism that explains how the spatial distribution of CSPGs determines whether they act as a cue for axon guidance or as an axon-inhibiting factor. Increasing our understanding of this issue may promote the development of novel therapeutic strategies that regulate the spatial distributions of CSPGs to use them as an axon guidance cue. Abstract: MicroRNAs (miRNAs) constitute a family of small RNA species which have been demonstrated to be one of key effectors in mediating host–pathogen interaction. In this study, two haemocytes miRNA libraries were constructed with deep sequenced by illumina Hiseq2000 from healthy (L1) and skin ulceration syndrome Apostichopus japonicus (L2). The high throughput solexa sequencing resulted in 9,579,038 and 7,742,558 clean data from L1 and L2, respectively. Sequences analysis revealed that 40 conserved miRNAs were found in both libraries, in which let-7 and mir-125 were speculated to be clustered together and expressed accordingly. Eighty-six miRNA candidates were also identified by reference genome search and stem-loop structure prediction. Importantly, mir-31 and mir-2008 displayed significant differential expression between the two libraries according to FPKM model, which might be considered as promising targets for elucidating the intrinsic mechanism of skin ulceration syndrome outbreak in the species. China's surface mining greatly supported the rapid socio-economic development; however, there was a scarcity in the systematic understanding of national changes in surface mining changes and associated ecosystem services (ESs) losses, which inevitably affected human well-being and limited sustainable ecosystem management and policy optimization. In this study, we quantified the areal changes in surface mining based on the ChinaCover database and performed further analysis of ES loss from expanded surface mining using multidimensional geospatial data from 1990 to 2015, including MODIS products, meteorological records, and statistical datasets. Our observations reveal that China's surface mining was estimated to be 4746 km2 in 2015 and that Inner Mongolia had the largest surface mining area (28%). Surface mining expanded remarkably from 1990 to 2015, with an increase by 2.7 times after 2000. In particular, Inner Mongolia, Shanxi, and Qinghai had the greatest increases in surface mining area. Rapid expansion of surface mining led to obvious declines in natural habitat area, water retention, net primary productivity, and grain production, and these ES losses showed apparent spatiotemporal variations. China has taken many measures to reclaim the abandoned surface mining sites. Given the rapid expansion of surface mining and related ES loss, China should continue to perform ecological restoration for its sustainability. • China's surface mining changes were firstly reported using remote sensing observation. • China's surface mining expanded by 2302 km2 from 1990 to 2015. • China's surface mining caused spatially varied losses in multiple ecosystem services. • Satellite observations reveal China's efforts to make the surface mining area green by 446 km2. Leucine-rich repeat and immunoglobulin domain-containing protein 1 (LINGO-1) is a transmembrane protein that negatively regulates neural regeneration in the central nervous system. LINGO-1 expression is up-regulated after central nerve injury, and is accompanied by cell death. Both LINGO-1 and cell death in the injury microenvironment are thought to limit neural regeneration, but the relationship between LINGO-1 and cell death has not been characterized. To investigate whether LINGO-1 deletion improves the spinal cord microenvironment after spinal cord injury (SCI) and contributes to cell survival, we generated LINGO-1 knockout (KO) mice. These mice and wild-type control mice were subjected to spinal cord transection. Fourteen days after spinal cord transection, cell apoptosis, inflammation, glial scar, and growth of nerve fibers were evaluated by immunostaining. The results showed that LINGO-1 KO mice demonstrated a profound reduction in expression of caspase-3, transferase-mediated deoxyuridine triphosphate biotin nick end labeling (TUNEL), ionized calcium binding adapter molecule 1 (IBA1), glial fibrillary acidic protein (GFAP), and chondroitin sulfate proteoglycans (CSPGs) compared to controls. In contrast, expression of neurofilament (NF) at the SCI site in LINGO-1 KO mice was markedly increased compared to that in wild-type mice. These results suggested that LINGO-1 plays a critical role in the injury microenvironment in processes such as cell death, inflammatory response, and glial scar formation. Importantly, LINGO-1 deletion and a positive microenvironment may exert synergistic effects to promote nerve fiber regeneration.
