Although significant technological advancements have been made in the development of analytical biosensor chips for detecting bacterial strains (E. coli, S. Mutans and B. Subtilis), critical requirements i.e. limit of detection (LOD), fast time of response, ultra-sensitivity with high reproducibility and good shelf-life with robust sensing capability have yet to be met within a single sensor chip. In order to achieve these criteria, we present an electrically-receptive thermally-responsive (ER-TR) sensor chip comprised of simple filter paper used as substrate coated with composite of poly(N-isopropylacrylamide) polymer (PNIPAm) – graphene nanoplatelet (GR) followed by evaporation of Au electrodes for capturing both Gram-positive (S. mutans and B. subtilis) and Gram-negative (E. coli) bacterial cells in real-time. Autoclave water, tap water, lake water and milk samples were tested with ER-TR chip with and without bacterial strains at varying concentration range 10 1 –10 5 cells/mL. The sensor was integrated with in-house built printed circuit board (PCB) to transmit/receive electrical signals. The interaction of E. coli, S. mutans and B. subtilis cells with fibers of PNIPAm-GR resulted in a change of electrical resistance and the readout was monitored wirelessly in real-time using MATLAB algorithm. Forty-one bacterial strains isolated from the headbox water of two machines in a Canadian paper mill were associated with the genera Asticcacaulis, Acidovorax, Bacillus, Exiguobacterium, Hydrogenophaga, Pseudomonas, Pseudoxanthomonas, Staphylococcus, Stenotrophomonas based on the sequence of their 16S rRNA genes. The metabolic profile of these strains were determined using Biolog EcoPlate, and the bacteria were divided into four metabolic groups. Metabolic profiles of the bacterial communities colonizing the headbox water of two paper machines was also determined weekly over a 1 year period. The only compound that was not reduced by the bacterial community was 2-hydroxybenzoic acid. Utilization frequency of the other carbon sources in the Biolog EcoPlate ranged from 3 to 100%. We have developed a new litmus paper test (DipTest) for detecting Escherichia coli (E. coli) in water samples by performing enzymatic reactions directly on the porous paper substrate. The paper strip consists of a long narrow piece of cellulose blotting paper coated with chemoattractant (at bottom edge), wax hydrophobic barrier (at the top edge), and custom formulated chemical reagents (at reaction zone immediately below the wax hydrophobic barrier). When the paper strip is dipped in water, E. coli in the water sample is attracted toward the paper strip due to a chemotaxic mechanism followed by the ascent along the paper strip toward the reaction zone due to a capillary wicking mechanism, and finally the capillary motion is arrested at the top edge of the paper strip by the hydrophobic barrier. The E. coli concentrated at the reaction zone of the paper strip will react with custom formulated chemical reagents to produce a pinkish-red color. Such a color change on the paper strip when dipped into water samples indicates the presence of E. coli contamination in potable water. The performance of the DipTest device has been checked with different known concentrations of E. coli contaminated water samples using different dip and wait times. The DipTest device has also been tested with different interfering bacteria and chemical contaminants. It has been observed that the different interfering contaminants do not have any impact on the DipTest, and it can become a potential solution for screening water samples for E. coli contamination at the point of source. Simple Summary: In biofloc technology systems, organic matter and inorganic substances accumulate in the culture unit mainly due to low water exchange, feed input, high stocking densities, and the level of organic carbon that subsequently increase the bacterial biomass. The organic and inorganic matter in suspension are maintained in a limited concentration to avoid negative effects on animal performance and water quality. This requires the occasional removal of excess solids, which produces an effluent enriched in nutrients, such as nitrogen and phosphorus. The use of integrated aquaculture in which the residues of one species are used as a source of nutrients for another species is an approach which aims to minimize this production of waste. Still, there are aspects of integrated cultures that have to be studied, such as the provision of feed for the different species. Therefore, we evaluated the addition or not of fish feed and microalgae in an integrated shrimp and fish culture regarding animal growth performance and water quality. We found that biofloc is nutritive for fish, but feed is necessary to improve its growth. Moreover, microalgae improved fish survival. However, both of factors did not affect the solids production. This study aims to evaluate a Pacific white shrimp and Nile tilapia integrated system using biofloc technology with or without the addition of the microalgae Scenedesmus obliquus and with or without fish feed supplementation in a two-factor 62-day experiment. The shrimp (2.16 ± 0.01 g) were reared under a density of 400 shrimp m−3 and the fish (1.53 ± 0.12 g) were reared under a density of 522 fish m−3. The microalgae was added to the culture water two times a week. Growth performance, sludge production, and water microbiology were evaluated. Fish feed and the microalgae addition improved fish final biomass in 58% and 14%, respectively (p < 0.05). Fish survival was significantly higher when microalgae was added (93.9 ± 1.8%) compared with the treatments without microalgae addition (86.2 ± 7.6%) (p < 0.05). The yield of the overall system was higher in the treatments with fish feed supplementation (4.2 ± 0.2 kg m−3) compared with no addition (3.9 ± 0.2 kg m−3) (p < 0.05). These results suggest that fish feed supplementation at the rate of 1% of the biomass and microalgae inoculation can improve fish growth performance and system yield, without affecting sludge production and water microbiology. Objectives: This paper describes and comments on contemporary legislation concerning prevention of epidemics caused by contaminated drinking water from public water supplies in the Czech Republic.Methods: Suggestions are made for removing existing legislative shortcomings, clarifying diction of existing laws and expanding sanctions and penalties for health injury caused by providers and operators of public drinking water.Results: The author reflects on improving legislation concerning the compensation of victims of contaminated water with reference to the aftermath of a local epidemic in the Dejvice District of Prague.Conclusion: The issues raised should be addressed since better legislation can significantly contribute to the limitation of water-borne epidemics and their consequences. There is a widespread need for the use of quantitative microbial risk assessment (QMRA) to determine reclaimed water quality for specific uses, however neither faecal indicator levels nor pathogen concentrations alone are adequate for assessing exposure health risk. The aim of this study was to build a conceptual model representing factors contributing to the microbiological health risks of reusing water treated in maturation ponds. This paper describes the development of an unparameterised model that provides a visual representation of theoretical constructs and variables of interest. Information was collected from the peer-reviewed literature and through consultation with experts from regulatory authorities and academic disciplines. In this paper we explore how, considering microbial risk as a modular system, following the QMRA framework enables incorporation of the many factors influencing human exposure and dose response, to better characterise likely human health impacts. By using and expanding upon the QMRA framework we deliver new insights into this important field of environmental exposures. We present a conceptual model of health risk of microbial exposure which can be used for maturation ponds and, more importantly, as a generic tool to assess health risk in diverse wastewater reuse scenarios. Low economic profit usually reduces the incentive of producers to operate their wastewater treatment technologies effectively. It is necessary to investigate the performance of environmentally friendly production technologies that reduce wastewater discharges and generate economic outputs simultaneously (EPTWs) in China over the past decade. In this paper, we apply the Malmquist-Luenberger productivity index widely used in the field of economics to evaluate the productivity change of EPTWs for 30 administrative provinces in China during 2003–2015. The pathways of the productivity change are further identified by decomposing the productivity index into two components: technological change and technical efficiency change. The results show that China’s environmental productivity index associated with wastewater reduction had undergone a downward trend, and evident spatial disparities are observed among the 30 provincial regions. Moreover, the changes of China’s environmental productivity over the whole studied period can mainly be attributed to technological progress, while the technical efficiency component has contributed little, although its annual contributing rate is in an increasing trend. This paper attempts to evaluate the Malaysian water quality indexing method that has been criticized for its ineffectiveness. The indexing method is referred to as the Water Quality Index of the Department of Environment, Malaysia (DOE-WQI). This index was assessed against two other indices (River Ganga Index and Minimal Water Quality Index (WQImin)) and a modified DOE-WQI was also proposed. DOE-WQI and WQImin are physicochemical indices, whereas the River Ganga Index and modified DOE-WQI are indices with the inclusion of a microbiological parameter. The assessment was conducted based on the water quality of tropical rivers in Malaysia, with specific reference to Sarawak River and its tributaries. Water quality in terms of pH, dissolved oxygen, conductivity, turbidity, total suspended solids, biochemical oxygen demand, chemical oxygen demand, ammoniacal nitrogen, and fecal coliform count (FCC) was measured from 18 stations in December 2015, January 2016, and March 2016. Generally, the river was characterized with high FCC with the four indices significantly correlated. The results demonstrated the shortcomings of the existing DOE-WQI; the physicochemical index assigned water quality to a better class than its actual conditions without taking into consideration the FCC. This paper examines the recovery of the enteric adenovirus human adenovirus 40 (HAdV 40) by cross-flow ultrafiltration and interprets recovery values in terms of physicochemical interactions of virions during sample concentration. Prior to ultrafiltration, membranes were either blocked by exposure to calf serum (CS) or coated with a polyelectrolyte multilayer (PEM). HAdV 40 is a hydrophobic virus with a point of zero charge between pH 4.0 and pH 4.3. In accordance with predictions from the extended Derjaguin-Landau-Verwey-Overbeek theory, the preelution recovery of HAdV (rpre) from deionized water was higher with PEMcoated membranes (rprePEM=74.8%±9.7%) than with CS-blocked membranes (rpreCS =54.1%±6.2%). With either membrane type, the total virion recovery after elution (rpost) was high for both deionized water (rpostPEM=99.5%±6.6% and rpost CS =98.8%± 7.7%) and tap water (rpostPEM=89%±15% and rpost CS =93.7%±6.9%). The nearly 100% recoveries suggest that the polyanion (sodium polyphosphate) and surfactant (Tween 80) in the eluent disrupt electrostatic and hydrophobic interactions between the virion and the membrane. Addition of EDTA to the eluent greatly improved the elution efficacy (rpostCS =88.6%±4.3% and rpostPEM=87.0%±6.9%) with surface water, even when the organic carbon concentration in the water was high (9.4±0.1 mg/liter). EDTA likely disrupts cation bridging between virions and particles in the feed water matrix or the fouling layer on the membrane surface. Background: Antibiotic resistance has been referred to as 'the greatest malice of the 21st century' and a global action plan was adopted by the World Health Assembly in 2015. There is a wealth of independent studies regarding antibiotics and resistant bacteria in humans, animals and their environment, however, integrated studies are lacking, particularly ones that simultaneously also take into consideration the health related behaviour of participants and healthcare providers. Such, 'One health' studies are difficult to implement, because of the complex teamwork that they entail. This paper describes the protocol of a study that investigates 'One health' issues regarding antibiotic use and antibiotic resistance in children and their environment in Indian villages.Methods/design: Both quantitative and qualitative studies are planned for a cohort of children, from 6 villages, and their surrounding environment. Repeated or continues data collection is planned over 2 years for quantitative studies. Qualitative studies will be conducted once. Studies include parents' health seeking behavior for their children (1-3 years of age at the onset), prescribing pattern of formal and informal healthcare providers, analysis of phenotypic antibiotic resistance of Escherichia coli from samples of stool from children and village animals, household drinking water, village source water and waste water, and investigation on molecular mechanisms governing resistance. Analysis of interrelationship of these with each other will also be done as basis for future interventions. Ethics approval has been obtained from the Institutional Ethics Committee R.D. A new variation of a method for quantifying cellular adenosine triphosphate (cATP) is described. The variation permits differentiation between cATP from bacteria and cATP from fungi. This paper describes the process by which the new differential cATP method was developed and validate in both laboratory and field tests. Laboratory tests using bacteria, yeasts and filamentous fungi demonstrated that cATP observed in ≥5 μm and <5 μm size-fractions, respectively were consistently within ±10% of predicted concentrations. The field evaluation during which four participating metalworking fluid (MWF) compounders collectively tested >250 used MWF samples, confirmed that the differential protocol reliably detected fungal contamination when it was present. Bloom-forming freshwater cyanobacterial genera pose a major ecological problem due to their ability to produce toxins and other bioactive compounds, which can have important implications in illnesses of humans and livestock. Cyanobacteria such as Microcystis, Anabaena, Oscillatoria, Phormidium, and Aphanizomenon species producing microcystins and anatoxin-a have been predominantly documented from most South Korean lakes and reservoirs. With the increase in frequency of such blooms, various monitoring approaches, treatment processes, and prediction models have been developed in due course. In this paper we review the field studies and current knowledge on toxin producing cyanobacterial species and ecological variables that regulate toxin production and bloom formation in major rivers (Han, Geum, Nakdong, and Yeongsan) and reservoirs in South Korea. In addition, development of new, fast, and high-throughput techniques for effective monitoring is also discussed with cyanobacterial bloom advisory practices, current management strategies, and their implications in South Korean freshwater bodies. Abstract: Atrazine is one of the most persistent herbicides that contaminate watercourses. However, its biodegradation has been scarcely studied in aquatic ecosystems. The present work studies the potential of bacteria collected from freshwater and sludges of three water potabilization plants to degrade atrazine. Bacteria from all samples, rarely exposed to the herbicide, were able to degrade atrazine as the sole N source in microcosms assays. Five bacterial consortia that degraded consistently 1 or 25mgL−1 of atrazine within 48 h were established. T-RFLP analysis showed that three ribotypes accounting for more than 50% of the bacterial communities were common to the five consortia and the complete set of atrazine-degrading genes trzN, atzB, atzC and trzD was detected on them. Two consortia from the same origin but incubated with different atrazine concentrations (SAC 1 and SAC 25) contain isolates affiliated to Sinorhizobium sp. and Pseudomonas citronellolis, but Arthrobacter uraefaciens and Achromobacter xylosoxidans were specific of SAC 1 and SAC 25, respectively. These two consortia degraded atrazine in the presence of nitrate and grew in cyanuric acid. Background The aim of this paper was to develop a reverse transcription loop-mediated isothermal amplification (RT-LAMP) method for rapid, sensitive and inexpensive detection of astrovirus. Results The detection limit of LAMP using in vitro RNA transcripts was 3.6×10 copies•μL-1, which is as sensitive as the presently used PCR assays. However, the LAMP products could be identified as different colors with the naked eye following staining with hydroxynaphthol blue dye (HNB). No cross-reactivity with other gastroenteric viruses (rotavirus and norovirus) was observed, indicating the relatively high specificity of LAMP. The RT-LAMP method with HNB was used to effectively detect astrovirus in reclaimed water samples. Conclusions The LAMP technique described in this study is a cheap, sensitive, specific and rapid method for the detection of astrovirus. The Water-associated Disease Index (WADI) was developed to identify and visualize vulnerability to different water-associated diseases by integrating a range of social and biophysical determinants in map format. In this study vulnerability is used to encompass conditions of exposure, susceptibility, and differential coping capacity to a water-associated health hazard. By assessing these conditions, the tool is designed to provide stakeholders with an integrated and long-term understanding of subnational vulnerabilities to water-associated disease and contribute to intervention strategies to reduce the burden of illness. The objective of this paper is to describe and validate the WADI tool by applying it to dengue. A systemic ecohealth framework that considers links between people, the environment and health was applied to identify secondary datasets, populating the index with components including climate conditions, land cover, education status and water use practices. Data were aggregated to create composite indicators of exposure and of susceptibility in a Geographic Information System (GIS). These indicators were weighted by their contribution to dengue vulnerability, and the output consisted of an overall index visualized in map format. The WADI was validated in this Malaysia case study, demonstrating a significant association with dengue rates at a sub-national level, and illustrating a range of factors that drive vulnerability to the disease within the country. The index output indicated high vulnerability to dengue in urban areas, especially in the capital Kuala Lumpur and surrounding region. However, in other regions, vulnerability to dengue varied throughout the year due to the influence of seasonal climate conditions, such as monsoon patterns. The WADI tool complements early warning models for water-associated disease by providing upstream information for planning prevention and control approaches, which increasingly require a comprehensive and geographically broad understanding of vulnerability for implementation. Abstract: Soil aquifer treatment (SAT) system plays an important role in degrading organic pollutants, removing bacteria, N, P and so on during artificial groundwater recharge with recycling water. In order to discover the possibility for the pathogen going through aerated zone and contaminating groundwater, this paper presents an experiment on the transport and fate of the bacteria (Escherichia coli L.) in SAT system, a packed silt soil column, under saturated conditions. The simulated solution considered the basic control index and maximum limit value of groundwater recharge with recycling water. The concentration of bacteria was 105 Colony-Forming Units L−1 in the secondary discharge standard of wastewater treatment plant effluent. The experiment included two recharging stages (firstly for 288 h and secondly for 528 h) with 264 h drying between them. The results showed that there was no detection of E. coli L., and the electric conductivity, total nitrogen, permanganate index became the same finally between the influent and effluent. The concentration of E. coli L. in the solid phase decreased from up to down of the column. This simulation of recycling water recharge could make people understand the transport and fate of contaminants better, and provided reference to practical projects. [Copyright &y& Elsevier]

Bulk fluorescence measurements could be a faster and cheaper way of enumerating viruses than epifluorescence microscopy, flow cytometry, or transmission electron microscopy (TEM). However, since viruses are not imaged, the background fluorescence compromises the signal, and we know little about its nature. In this paper the size ranges of nucleotides that fluoresce in the presence of SYBR gold were determined for wastewater and a range of freshwater samples using a differential filtration method. Fluorescence excitation-emission matrices (FEEMs) showed that >70% of the SYBR fluorescence was in the < 10-nm size fraction (background) and was not associated with intact viruses. This was confirmed using TEM. The use of FEEMs to develop a fluorescence-based method for counting viruses is an approach that is fundamentally different from the epifluorescence microscopy technique used for enumerating viruses. This high fluorescence background is currently overlooked, yet it has had a most pervasive influence on the development of a simple fluorescence-based method for quantifying viral abundance in water. In recent years, the adsorption of heavy metal cations onto bacterial surfaces has been studied extensively. This paper reports the findings of a study conducted on the heavy metal ions found in mine effluents from a mining plant where Co2+ and Ni2+ bearing minerals are processed. Heavy metal ions are reported to be occasionally present in these mine effluents, and the proposed microbial sorption technique offers an acceptable solution for the removal of these heavy metals. The sorption affinity of microorganisms for metal ions can be used to select a suitable microbial sorbent for any particular bioremediation process. Interactions of heavy metal ions (Co2+ and Ni2+) and light metal ions (Mg2+ and Ca2+) with indigenous microbial cells (Brevundimonas spp., Bacillaceae bacteria and Pseudomonas aeruginosa) were investigated using the Langmuir adsorption isotherm, pseudo second-order reaction kinetics model and a binary-metal system. Equilibrium constants and adsorption capacities derived from these models allowed delineation of the effect of binding affinity and metal concentration ratios on the overall adsorption behaviour of microbial sorbents, as well as prediction of performance in bioremediation systems. Although microbial sorbents used in this study preferentially bind to heavy metal ions, it was observed that higher concentrations (>90 mg/ℓ) of light metal ions in multi-metal solutions inhibit the adsorption of heavy metal ions to the bacterial cell wall. However, the microbial sorbents reduced Ni2+ levels in the mine-water used (93–100% Ni2+ removal) to below the maximum acceptable limit of 350 μg/ℓ, established by the South African Bureau of Standards. Competition among metal ions for binding sites on the biomaterial surface can occur during the bioremediation process, but microbial sorption affinity for heavy metal ions can enhance their remediation in dilute (<5 mg/ℓ heavy metal) wastewaters. The aim of this paper was the syntheses of natural zeolite modified with undoped TiO2 / nitrogen doped TiO2 by calcination method in order to remove Enterococcus faecalis from water. The obtained materials were characterized by X-ray diffraction (XRD), UV-VIS diffuse reflectance spectroscopy (DRUV-VIS), scanning electron microscopy (SEM) and energy dispersive X-ray analysis (EDX). Abstract: In health impact assessments, the sticking efficiency of a bacteria or virus population largely determines the transported distance of that biocolloid population, and hence, the potential health impact. However, at the same time, one of the most difficult parameters to estimate is the lower value of the sticking efficiency that should be used in calculating the health impact. In this paper, we introduce the concept of the minimum sticking efficiency (α i) value of a bacteria population, including a method to determine the minimum sticking efficiency. Thereto, sticking efficiency distributions of 6 environmentally isolated E scherichia coli strains were determined by carrying out laboratory column experiments over a transport distance of about 5m. Experiments were conducted in de-mineralized (DI) water and in artificial groundwater (AGW). Sticking efficiencies were calculated for column segments (at varying distances from top of column) and fractions of total bacteria mass input in each segment were estimated by mass balance. The sticking efficiencies were highest close to the top of the column, near the point of bacteria mass input (0.103–0.352 in DI, and 1.034–9.470 for AGW) and reduced with distance with the lowest α i values (10−5-0.06 in DI and 0.006–0.283 in AGW) determined at the two most distant column segments (between 2.33 and 4.83m from the top of the column). Power–law distributions best described the relationship between fraction of cells retained, F i, and α i . The minimum sticking efficiency was defined as the sticking efficiency belonging to a retained bacteria fraction of 0.001% of the original bacteria mass (total number of cells) flowing into the column (F =10−5), and coinciding with a 99.999% reduction of the original bacteria mass, and minimum sticking efficiencies were extrapolated from the fitted power–law distributions. In the DI experiments, minimum sticking efficiency values ranged from as low as 10−9 (for E. coli strain UCFL-94) to 10−2 (for E. coli strain UCFL-348); in the AGW experiments, minimum sticking efficiency values ranged from 10−6 (for strain UCFL-94) to ≥1(for strain UCFL-348). We concluded that in quantifying health impacts of biocolloids traveling in aquifers, the concept of the minimum sticking efficiencies, and the percentage of individual biocolloids of a total population having such low sticking efficiency, together with an inactivation rate coefficient, can serve as a useful tool to determine the maximum transported distance as a worst case scenario, and, hence, the potential health impact. The surface plasmon resonance (SPR) technique is a well-established method for the measurement of molecules binding to surfaces and the quantification of binding constants between surface-immobilized proteins and proteins in solution. In this paper we describe an extension of the methodology to study bacteriophage-bacterium interactions. A two-channel microfluidic SPR sensor device was used to detect the presence of somatic coliphages, a group of bacteriophages that have been proposed as fecal pollution indicators in water, using their host, Escherichia coli WG5, as a target for their selective detection. The bacterium, E. coli WG5, was immobilized on gold sensor chips using avidin-biotin and bacteriophages extracted from wastewater added. The initial binding of the bacteriophage was observed at high concentrations, and a separate, time-delayed cell lysis event also was observed, which was sensitive to bacteriophage at low concentrations. As few as 1 PFU/ml of bacteriophage injected into the chamber could be detected after a phage incubation period of 120 min, which equates to an approximate limit of detection of around 10² PFU/ml. The bacteriophage-bacterium interaction appeared to cause a structural change in the surface-bound bacteria, possibly due to collapse of the cell, which was observed as an increase in mass density on the sensor chip. Genetic markers are expected to provide better specificity in epidemiological studies and potentially serve as better indicators of water-bome pathogens. Methods used to assess genetic markers of emerging microbial indicators include pulsed field gel electrophoresis, polymerase chain reaction (PCR), and microarrays. This paper outlines a high-throughput approach to screen for such genetic markers using a set of theoretical and experimental screening tools. The theoretical screening involves evaluating genes related to the ribosomal RNA and specific functions from emerging indicator groups, followed by experimental validation with appropriate sampling schemes and high-throughput and economical screening methods, such as microarrays, real time PCR, and on-chip PCR. Analysis of a wide range of samples covering temporal variability in location, host, and waterborne disease outbreaks is essential. The paper presents a new index for assessing water trophy and organic pollution. It is based on only true aquatic macrophytes – being calculated on species score, coefficient of ecological amplitude and degree of cover. The method was tested in an acidic lowland river and an alkaline mountain river, and is shown to be validated by bio-indication scales based on macrophyte communities. The present paper reports the results on the occurrence and composition of steady state phytoplankton assemblages in the shallow water bodies of 18 Bulgarian natural wetlands. They all differ in their morphometry, hydrology and trophic state. Steady states were identified according to Sommer & Padisák (1993) when (i) 1, 2 or 3 species of algae contribute more than 80% of total biomass, (ii) their existence or coexistence persists for long enough (more than 1–2 weeks) and (iii) during that period the total biomass does not increase significantly. These situations were rare and were dominated by 7 stress-tolerant, colony-forming and ruderal disturbance-tolerant cyanoprokaryotes ( Microcystis aeruginosa, Microcystis wesenbergii, Anabaena spiroides, Anabaenopsis arnoldii, Aphanizomenon flos-aquae, Aphanizomenon gracile and Cylindrospermopsis raciborskii). The equilibrium states had been registered only during hypertrophic conditions in three stressed and frequently disturbed shallow water bodies and were related to the summer – late summer season. An estimated 2300 people became seriously ill and 7 died from exposure to microbially contaminated drinking water in the town of Walkerton, Ontario, in May of 2000. The severity of this drinking water disaster resulted in the Government of Ontario calling a public inquiry by Mr. Justice Dennis O'Connor to address the cause of the outbreak, the role, if any, of government policies in contributing to this outbreak, and ultimately, the implications of this experience on the safety of drinking water across the province of Ontario. This paper summarizes relevant evidence presented at the inquiry by the expert panel together with Justice O'Connor's findings addressing the circumstances of the outbreak. These findings are reviewed in relation to the published causes of previous waterborne disease outbreaks that have been reported elsewhere in developed countries. The circumstances surrounding the Walkerton tragedy are an important source of knowledge for those concerned with providing safe drinking water to the public. Although some circumstances are obviously specific to this epidemic, others echo common themes in waterborne outbreaks that have occurred before. These common themes suggest the need for attention to broad issues of drinking water safety in addition to the individual specific details that often command attention. Cryptosporidium, an intestinal protozoan pathogen, is one of the leading causes of diarrhea in healthy adults and death in children. Detection of Cryptosporidium oocysts has become a high priority to prevent potential outbreaks. In this paper, a label-free interdigitated-based capacitive biosensor has been introduced for the detection of Cryptosporidium oocysts in water samples. Specific anti-Cryptosporidium monoclonal antibodies (IgG3) were covalently immobilized onto interdigitated gold electrodes as the capture probes, and bovine serum albumin was used to avoid non-specific adsorption. The immobilization of the antibodies was confirmed by measuring the change in the contact angle. The detection was achieved by measuring the relative change in the capacitive/dielectric properties due to the formation of Cryptosporidium-antibody complex. The biosensor has been tested for different concentrations of Cryptosporidium. The results show that the biosensor developed can accurately distinguish different numbers of captured cells and densities on the surface of the biosensor. The number of Cryptosporidium oocysts captured on the electrode surface was confirmed using a fluorescein isothiocyanate (FITC) immunofluorescence assay. The response from the developed biosensor has been mainly dependent on the concentration of Cryptosporidium under optimized conditions. The biosensor showed a linear detection range between 15 and 153 cells/mm2 and a detection limit of 40 cells/mm2. The label-free capacitive biosensor developed has a great potential for detecting Cryptosporidium in environmental water samples. Furthermore, under optimized conditions, this label-free biosensor can be extended for detection of other biomarkers for biomedical and environmental analyses. [ABSTRACT FROM AUTHOR]
