Background: Pneumonia remains a leading cause of child death in developing countries. The viruses in severe pneumonia remain poorly defined. Methods: The study was conducted at the Eastern Visayas Regional Medical Center in Tacloban City, Philippines from May 2008 to May 2009. Patients aged 8 days to 13 years old who were admitted to the Department of Pediatrics with severe pneumonia were enrolled for the study. Upon admission, polymerase chain reaction was performed using nasopharyngeal swabs and blood cultures to detect respiratory viruses and bacteria, respectively. Result: Among the 819 patients enrolled, at least one virus was detected in 501 cases (61.2%). In addition, 423 cases were positive for a single virus while bacteria were detected in the blood culture sample of 31 cases. The most commonly detected viruses were human rhinoviruses (n = 189), including types A (n = 103), B (n = 17), and C (n = 69), and respiratory syncytial virus (RSV) (n = 165). Novel viruses such as human metapneumovirus, human coronavirus NL63, human bocavirus, and human polyomaviruses WU and KI were also detected. There were 70 deaths, and one or more viruses were detected in 35 (50%) of these cases. Positivity only for influenza A virus (OR = 4.3, 95% CI = 1.3-14.6) was significantly associated with fatal outcome. From the blood culture, Burkholderia cepacia group (n = 9), Streptococcus pneumoniae (n = 4), Staphylococcus aureus (n = 4), Haemophilus influenzae (n = 1), and Salmonella C1 (n = 1) were also isolated. Conclusion: Viruses were commonly detected in children with severe pneumonia in the Philippines. Human rhinovirus (HRV) C was recently identified as the third species of HRV using a molecular technique. Infections caused by previously identified HRVs (A and B) are thought to be limited to the respiratory tract; however, pathogenesis of HRVC is still largely unknown. A total of 816 nasopharyngeal swabs from hospitalized children with severe respiratory infections in the Philippines (May 2008-May 2009) were tested for HRV by reverse transcription polymerase chain reaction (RT-PCR), and 243 samples (29.8%) were positive for HRV. Among these patients, serum samples were also tested to determine whether specific HRV species were associated with viremia. Only 30 serum samples (12.3%) were positive for HRV. However, the HRV positive rates were different among HRV species, 3% (4/135) for HRVA, 0% (0/25) for HRVB, and 31% (26/83) for HRVC, and were the highest on 2 days after the onset of symptoms. These results suggest that HRVC may have a different pathogenicity and can more commonly cause viremia than HRVA and HRVB. Serum positive rates for HRV are affected by age, i.e., higher positive rates for those aged 1 year or more. HRVC that were detected from serum exhibited the same level of sequence diversity as those positive only for nasopharyngeal samples in phylogenetic analysis. However, all HRVA which were detected from serum were clustered in a monophyletic clade based on their 5' non-coding region (NCR) sequences, which is closely related with a certain HRVC genotype (A2) in 5'-NCR. The article focuses on the prevalence of acute respiratory illness related to human enterovirus 68 (HEV68) in Asia, Europe and the U.S. from 2008-2010, which included fatal cases from the Philippines and Japan. It is in the Philippine's Eastern Visayas that an outbreak of HEV68 was seen in late October 2008, which affected 21 patients who were mostly 0-4 years old. In Arizona, the state Department of Health Services detected HEV68 in 5 patients from August - September 2010.

Effects of pH, NH-N, and temperature on basidiospore germination in Coprinopsis austrophlyctidospora from New Zealand, C. phlyctidospora from Japan, C. aff. rugosobispora from Canada, and C. echinospora from Canada were investigated. The Coprinopsis spp. required the presence of ammonium-nitrogen under weak alkaline to neutral conditions for germination, regardless of their different areas of occurrence. The former two species had a wider concentration of NHCl solution and pH range for germination in comparison to the latter two species. The optimum concentration of NHCl solution for the germination was 0.01 M in C. austrophlyctidospora and 0.1 M in the other three species. The pH optimum for germination in the former two species was 8.0 whereas that for germination in the latter two species was 8.0-8.5. The temperature range (5.0-40.0°C) for the former two species was wider than that (5-30°C) for the latter two species. Temperature optima for the germination in the former two species, C. aff. rugosobispora and C. echinospora, were 30, 20-25 and 15°C, respectively. The germination abilities of these Coprinopsis species in a wide range of temperatures are relevant to their natural temperature regime, showing their potential ability to propagate in tropical to subarctic regions. Objective: Pandemic viral infections as emerging infectious diseases are of a great global concern. However, for some viruses, particular strains are endemic to specific areas and can be genetically distinguished from strains in other regions. In contrast, for some other viruses, genetically similar strains can spread and circulate all over the world. This study addresses global dissemination of various viral infections. Methods: We classified 34 viruses as per their ability to cross borders by review. We also described factors responsible for and the dynamics of global dissemination. We examined biological characteristics of viruses, manners or routes of transmission, host responses and epidemiological factors. Results: Factors required for viruses to cross borders include 'non-blood infection', 'short incubation period', 'short infectious period', 'frequent re-infection', 'small basic reproductive number (R0)' and 'high annual incidence'. Conclusion: Knowing the factors responsible for global dissemination of pathogens is useful for controlling and/or containing both classic and emerging infectious diseases. Copyright © 2011 S. Polycystic kidney disease (PKD) is a genetic disorder that is characterized by cyst formation in kidney tubules. PKD arises from abnormalities of the primary cilium, a sensory organelle located on the cell surface. Here, we show that the primary cilium of renal epithelial cells contains a protein complex comprising adenylyl cyclase 5/6 (AC5/6), A-kinase anchoring protein 150 (AKAP150), and protein kinase A. Loss of primary cilia caused by deletion of Kif3a results in activation of AG and increased CAMP levels. Polycystin-2 (PC2), a ciliary calcium channel that is mutated in human PKD, interacts with AC5/6 through its C terminus. Deletion of PC2 increases CAMP levels, which can be corrected by reexpression of wild-type PC2 but not by a mutant lacking calcium channel activity. Phosphodiesterase 4C (PDE4C), which catabolizes CAMP, is also located in renal primary cilia and interacts with the AKAP15O com- plex. Expression of PDE4C is regulated by the transcription factor hepatocyte nuclear factor-1β (HNF-1β), mutations of which produce kidney cysts. PDE4C is down-regulated and CAMP levels are increased in HNF-1β mutant kidney cells and mice. Collectively, these findings identify PC2 and PDE4C as unique components of an AKAP complex in primary cilia and reveal a common mechanism for dysregulation of CAMP signaling in cystic kidney diseases arising from different gene mutations. Abstract: The sudden emergence of the pandemic influenza A (H1N1) 2009 virus in early 2009 has resulted in a rapid transmission of this virus worldwide. Within a short time span, sporadic cases infected with this virus that shows oseltamivir resistance have also been reported. These resistant viruses have an amino acid change from histidine to tyrosine at position 275 (H275Y) of the neuraminidase gene. In this study, a reverse transcriptase PCR/restriction fragment length polymorphism (RT-PCR/RFLP) assay was developed to detect the H275Y mutation. Resistant and sensitive viruses could be differentiated using the RFLP patterns. This RT-PCR/RFLP assay is a simple method and also very specific and sensitive for detecting the H275Y mutation of pandemic influenza A (H1N1) 2009 viruses, and can be used in resource-limited settings. ' Candidatus Helicobacter heilmannii' induce chronic gastritis, which eventually leads to gastric B-cell type mucosa-associated lymphoid tissue (MALT) lymphoma. This study was performed using an animal model of infection with ' Candidatus Helicobacter heilmannii' to elucidate how this chronic inflammation is induced or maintained. BALB/c mice were infected with the ' Candidatus Helicobacter heilmannii' isolate SH4. The animals were examined at 8, 26, 54, and 83 weeks after the infection. The stomach of the animals was resected and immunostained for peripheral lymph node addressin (PNAd) and mucosal addressin cell adhesion molecule 1 (MAdCAM-1), ' Candidatus Helicobacter heilmannii,' and CD45R/B220. An in vitro binding assay with L- and E-selectin·IgM chimeric proteins was performed. Real-time polymerase chain reaction was used to evaluate transcripts of N-acetylglucosamine-6- O-sulfotransferases (GlcNAc6STs), which direct the expression of the PNAd and MAdCAM-1. Chronic gastritis developed in the infected animals, and its severity increased with the duration of the infection. B-cell type MALT lymphoma developed in some animals at 54 and 83 weeks after infection. PNAd- and MAdCAM-1-expressing high endothelial venule (HEV)-like vessels were induced in infected animals which developed chronic gastritis and MALT lymphoma. The number of HEV-like vessels increased as chronic inflammation progressed. The induced HEV-like vessels were bound by L- and E-selectin·IgM chimeric protein. mRNA expressions of GlcNAc6ST-1 and MAdCAM-1 increased in the infected animals. HEV-like vessels expressing GlcNAc6ST-1-mediated L-selectin ligand carbohydrate and MAdCAM-1 may play a crucial role in the pathogenesis of ' Candidatus Helicobacter heilmannii'-induced chronic gastritis and MALT lymphoma. Abstract: In April 2009, pandemic H1N1/09 influenza, which originated from swine influenza, appeared in North America, and it has since spread globally among humans. It is important to know how swine influenza A virus broke the host barrier to cause a pandemic. We analyzed 673 strains of human, avian, and swine influenza viruses and assessed the internal genes PB2, PB1, PA, NP, M, and NS. Here we found accumulation of mutations in segments that were retained as well as introduced due to genetic reassortment of viruses. The retained segments may have to mutate to accommodate new segments. The mutations caused by interaction among segments retained and introduced due to reassortment between swine influenza viruses may have increased the adaptation of the virus to humans, leading to pandemic H1N1/09. Measles, caused by measles virus (MeV), is a common infection in children. MeV is a member of the genus Morbillivirus and is most closely related to rinderpest virus (RPV), which is a pathogen of cattle. MeV is thought to have evolved in an environment where cattle and humans lived in close proximity. Understanding the evolutionary history of MeV could answer questions related to divergence times of MeV and RPV. We investigated divergence times using relaxed clock Bayesian phylogenetics. Our estimates reveal that MeV had an evolutionary rate of 6.0 - 6.5 × 10-4 substitutions/site/year. It was concluded that the divergence time of the most recent common ancestor of current MeV was the early 20th century. And, divergence between MeV and RPV occurred around the 11th to 12th centuries. The result was unexpected because emergence of MeV was previously considered to have occurred in the prehistoric age. MeV may have originated from virus of non-human species and caused emerging infectious diseases around the 11th to 12th centuries. The frequency of the amantadine-resistant H1N1 influenza A virus has been increasing since the 2005-2006 season. It is unclear whether reassortment was involved in this trend. Here, we show that cocirculation of amantadine-resistant and -sensitive strains led to the genesis of amantadine-sensitive reassortant virus during the 2007-2008 season. Thereafter, the reassortant virus predominated. This contrasts with the trend for the H3N2 virus, in which the amantadine-resistant reassortant virus became predominant. The results suggest that it is necessary to monitor genome dynamics to understand the evolution and mechanism of the emergence and spread of antiviral resistance among influenza A viruses. Abstract: To improve crystal quality of InN, an in-situ cyclic rapid pulse annealing during growth was carried out using infrared-lamp-heated molecular beam epitaxy. A cycle of 4min growth of InN at 400°C and 3s pulse annealing at a higher temperature was repeated 15 times on AlN on Si substrate. Annealing temperatures were 550, 590, 620, and 660°C. The back of Si was directly heated by lamp irradiation through a quartz rod. A total InN film thickness was about 200nm. With increasing annealing temperature up to 620°C, crystal grain size by scanning electron microscope showed a tendency to increase, while widths of X-ray diffraction rocking curve of (0002) reflection and E2 (high) mode peak of Raman scattering spectra decreased. A peak of In (101) appeared in X-ray diffraction by annealing higher than 590°C, and In droplets were found on the surface by annealing at 660°C. [Copyright &y& Elsevier]

Summary: Objectives: To determine the location of the parotid duct orifice in relation to the maxillary molars and its influence on oral clearance on the buccal surfaces of the maxillary molars. Methods: A 2-mm hole was made at the centre of an adhesive therapeutic agent for aphthous stomatitis and the agent was placed on the mucosa so that the hole matched the parotid duct orifice. To locate the orifice, an impression of the buccal tooth surfaces and mucosa around the agent was taken with the teeth in centric occlusion. To evaluate the oral clearance rate, 12 subjects who displayed the parotid duct orifice within 1S.D. of the mean values obtained from the original 35 subjects were selected. 1% agar containing 1mol/l potassium chloride was placed into three cylinders positioned horizontally, 6mm apart, in an acrylic holder centred over the mean duct location. The diffusion chambers were taken from the mouth at selected time intervals and the gel transferred quantitatively to flasks containing 300ml of 100ppm NaCl, which was assayed for potassium by atomic absorption spectrophotometry. Half-times for clearance were calculated. Results: The mean location of the parotid duct orifice was −0.4mm (range −7.5 to +6.1mm) mesial to the contact surface between the maxillary first and second molars (where negative values indicate mesial and positive values distal) and 7.2mm (range +3.8 to +10.4mm) above a line touching the buccal cusps of the upper molars. The clearance half-time values were shortest for the central cylinder whether salivary flow was unstimulated or stimulated and when flow was unstimulated the clearance half-time was shorter for the mesial than the distal cylinder. Conclusion: The degree of individual variation in the location of the parotid duct orifice is great and its exact location will markedly affect oral clearance at different positions on the buccal surfaces of the upper molars. Ammonia fungi invade forest floors immediately after a enrichment disturbance by a large input of ammonium-nitrogen. Latent form(s) of the ammonia fungi are spores and/or mycelium fragments. Ammonia fungi are characterized by their rapid germination stimulated by the presence of ammonium-nitrogen under neutral to weakly alkaline conditions. Each ammonia fungus establishes its territory during suppressed combative abilities of other microbes following ammonium-nitrogen disturbance. Earlyphase ammonia fungi (EP fungi) quickly sporulate before nonammonia fungi colonize with the declining of ammonium-nitrogen concentration associated with descending pH. Ectomycorrhizal species of late-phase ammonia fungi (LP fungi) escape from the L-F horizon as a result of interactions between EP fungi and form mycorrhizae in the H-A horizon when other fungal activities are suppressed by the ammonium-nitrogen disturbance. Ectomycorrhizal ammonia fungi initially use ammonium-nitrogen when the pH rises because of the ammonium-nitrogen disturbance and then gradually utilize both ammonium- and nitratenitrogen when the effects of the ammonium-nitrogen disturbance weaken. Early-stage EP fungi are ruderal stress-tolerant strategists whereas late-stage EP fungi are combative ruderal strategists. LP fungi are combative strategists from the standpoint of the interactions between other ammonia fungi. This classification is based on differences in their respective propagation strategies. Abstract: We have carried out a numerical simulation of the hydrothermal process for growing bulk single crystals of ZnO using SC/Tetra as the computational fluid dynamics software. The autoclave is assumed to be axisymmetric. The heat transfer by natural convection is calculated in order to study the effects of the variations in fluid properties. It has been found that the effect of the variations in heat capacity and viscosity is large. The temperature difference and flow rate between the raw-material zone and the crystal-growth zone are also discussed from the viewpoint of the transport phenomena in a crystal-growth process. Background: Influenza A virus infects not only humans, but also other species including avian and swine. If a novel influenza A subtype acquires the ability to spread between humans efficiently, it could cause the next pandemic. Therefore it is necessary to understand the evolutionary processes of influenza A viruses in various hosts in order to gain better knowledge about the emergence of pandemic virus. The virus has segmented RNA genome and 7th segment, M gene, encodes 2 proteins. M1 is a matrix protein and M2 is a membrane protein. The M gene may be involved in determining host tropism. Besides, novel vaccines targeting M1 or M2 protein to confer cross subtype protection have been under development. We conducted the present study to investigate the evolution of the M gene by analyzing its sequence in different species. Results: Phylogenetic tree revealed host-specific lineages and evolution rates were different among species. Selective pressure on M2 was stronger than that on M1. Selective pressure on M1 for human influenza was stronger than that for avian influenza, as well as M2. Site-by-site analyses identified one site (amino acid position 219) in M1 as positively selected in human. Positions 115 and 121 in M1, at which consensus amino acids were different between human and avian, were under negative selection in both hosts. As to M2, 10 sites were under positive selection in human. Seven sites locate in extracellular domain. That might be due to host's immune pressure. One site (position 27) positively selected in transmembrane domain is known to be associated with drug resistance. And, two sites (positions 57 and 89) locate in cytoplasmic domain. The sites are involved in several functions. Conclusion: The M gene of influenza A virus has evolved independently, under different selective pressure on M1 and M2 among different hosts. We found potentially important sites that may be related to host tropism and immune responses. These sites may be important for evolutional process in different hosts and host adaptation. An 80-year-old Japanese man, who had fever and generalized fatigue not improved by antibiotics, was admitted to our hospital. Laboratory data indicative of renal dysfunction and antineutrophil cytoplasmic antibody (ANCA) in the serum led to the consideration of ANCA-associated vasculitis as a differential diagnosis. However, before the diagnostic confirmation, he was found dead on the bed. Autopsy revealed necrotizing crescentic glomerulonephritis in the kidneys. In addition, necrotizing granulomatous vasculitis with infiltration of multinucleated giant cells and neutrophils but not eosinophils was present in multiple organs. The direct cause of death was presumed as cardiac arrest by lethal arrhythmia because vasculitic lesions were distributed widely in the cardiac walls, acute congestion was observed in the systemic organs, and other causes of death were ruled out. Abstract: The basidiomycete Strobilurus ohshimae has fruiting bodies covered with prominently projecting hair-like cells called cystidia; it is not consumed by the collembolan Ceratophysella denisana but is consumed by Mitchellania pilosa. To explain this difference, we examined the effects of S. ohshimae cystidia on collembolan survival. In the field, several collembolan species, including C. denisana, were found dead on S. ohshimae, whereas no dead M. pilosa were found on the fungus. Survival of M. pilosa on S. ohshimae was 100%. In the laboratory, cystidium destruction experiments showed that the cystidia of S. ohshimae were capable of killing C. denisana on contact. A cystidium contact time experiment revealed that the cystidia were also capable of killing M. pilosa, although M. pilosa could survive when the contact time was short. In the field, M. pilosa frequently fed on the interior of S. ohshimae. Interior feeding may enable M. pilosa to feed on S. ohshimae by reducing contact time with the cystidia. The deadly surface of S. ohshimae deters the surface feeder C. denisana but not the interior feeder M. pilosa, resulting in food differentiation of these collembolans. [Copyright &y& Elsevier]

Abstract: Diesel exhaust particles (DEPs) cause many adverse health problems, and reports indicate increased risk of breast cancer in men and women through exposure to gasoline and vehicle exhaust. However, DEPs include vast numbers of compounds, and the specific compound(s) responsible for these actions are not clear. We recently isolated two nitrophenols from DEPs-3-methyl-4-nitrophenol (4-nitro-m-cresol; PNMC) and 4-nitro-3-phenylphenol (PNMPP)—and showed that they had estrogenic and anti-androgenic activities. Here, we tried to clarify the involvement of these two nitrophenols in promoting the growth of the MCF-7 breast cancer cell line. First, comet assay was used to detect the genotoxicity of PNMC and PNMPP in a CHO cell line. At all doses tested, PNMC and PNMPP showed negative genotoxicity, indicating that they had no tumor initiating activity. Next, the estrogen-responsive breast cancer cell line MCF-7 was used to assess cell proliferation. Proliferation of MCF-7 cells was stimulated by PNMC, PNMPP, and estradiol-17β and the anti-estrogens 4-hydroxytamoxifen and ICI 182,780 inhibited the proliferation. To further investigate transcriptional activity through the estrogen receptor, MCF-7 cells were transfected with a receptor gene that allowed expression of luciferase enzyme under the control of the estrogen regulatory element. PNMC and PNMPP induced luciferase activity in a dose-dependent manner at submicromolar concentrations. ICI 182,780 inhibited the luciferase activity induced by PNMC and PNMPP. These results clearly indicate that PNMC and PNMPP do not show genotoxicity but act as tumor promoters in an estrogen receptor α-predominant breast cancer cell line. [Copyright &y& Elsevier]

Basidiospore germination in an ectomycorrhizal ammonia fungus Hebeloma vinosophyllum was stimulated by 10–500 mM NH4Cl aqueous solution at pH 4.5–9.0, but not by pure water. The basidiospores germinated at 10°–35°C with an optimum at 25°–30°C. The highest germination percentage (83.0%) was observed in 100 mM NH4Cl aqueous solution adjusted to pH 8.0 by KOH, when the basidiospores were incubated at a density of 106 spores/ml at 30°C for 14 days. The percent germination value decreased with the increased duration of storage under both dry and wet conditions. Humidity and temperature affected the longevity of H. vinosophyllum basidiospores. The basidiospores maintained their germination ability longer under a dry condition than under a wet condition. The greatest longevity was accomplished by storage at 15°C under a dry condition. Abstract: Cystidia in fruit bodies are taxonomically important characters. However, little is known about their ecological functions. The defensive role of cystidia against the collembolans Ceratophysella denisana and Mitchellania horrida was examined in fruit bodies of Russula bella and Strobilurus ohshimae. Cystidium-destruction experiments demonstrated that R. bella and S. ohshimae cystidia decrease the number of collembola found on gills, although the effects were not significant for R. bella against C. denisana. Furthermore, R. bella cystidia increased collembolan mortality in the laboratory, and in the field, collembola were found dead on parts of the fruit body of this species where cystidia were abundant. In the cystidium-destruction experiment, approximately one-third of collembola appeared to avoid R. bella. Therefore, deadly cystidia may be selected for in R. bella to avoid collembolan attack. Laboratory feeding experiments revealed that collembola can extensively damage R. bella and S. ohshimae basidiospores by feeding. These results suggest that the cystidia of R. bella and S. ohshimae may protect basidiospores from collembolan predation. [Copyright &y& Elsevier]

Five early-phase ammonia fungi (EP fungi) – Amblyosporium botrytis, Ascobolus denudatus, Peziza moravecii, Pseudombrophila petrakii, Coprinopsis phlyctidospora, and Tephrocybe tesquorum, and one late-phase ammonia fungus (LP fungus), Hebeloma vinosophyllum – were co-cultured on malt extract-yeast extract agar media at pH 5.5, 7.0, 8.0, and 9.0. The co-cultures among the early-stage EP fungi Amblyosporium botrytis, Ascobolus denudatus, Peziza moravecii, and Pseudombrophila petrakii, generally did not inhibit or accelerate the reproductive structure formation of the opposed fungi. Among the EP fungi, Am. botrytis, As. denudatus, and Pe. moravecii intermingled with each other. The late-stage EP fungus T. tesquorum inhibited the growth of other EP fungi. Another late-stage EP fungus, C. phlyctidospora, showed ability to invade other EP fungi, but it did not deeply invade into the territories of early-stage EP fungi. The LP fungus H. vinosophyllum tended to accelerate basidioma formation of C. phlyctidospora at pH 5.5 and 9.0. H. vinosophyllum formed the highest numbers of basidomata at pH 5.5. These results show that successive occurrence of ammonia fungi is caused by the interspecific interactions among ammonia fungi as well as by the physiological characteristic of each fungus associated with conditions of its inhabiting soils, such as pH and nitrogen concentration. Abstract: Photo-allylation of various aromatic carbonyl compounds with a penta-coordinated allylsiliconate reagent was remarkably accelerated by the addition of a donor molecule. Abstract: Tomato seedlings grown on nitric medium and treated with various cadmium concentrations (0 to 50 μM) were used. Results obtained show that cadmium remains predominantly located in the roots, which then seem to play the role of trap-organs. Increasing cadmium concentration in the medium leads particularly to a decrease in accumulation, together with a decrease in the activity of glutamine synthetase and in the quantity of plastidic isoform ARNm (GS2), and, on the contrary, to an increase of the cytosolic isoform ARNm (GS1). On the other hand, stimulations were observed for NADH-dependent glutamate synthase, NADH-dependent glutamate dehydrogenase, ARNm quantity of this enzyme, ammonium accumulation, and protease activity. In parallel, stimulations were observed for NAD+ and NADP+-dependent malate dehydrogenase and NADP+-dependent isocitrate dehydrogenase. These results were discussed in relation to the hypothesis attributing to the dehydrogenase enzymes (GDH, MDH, ICDH) an important role in the plant defence processes against cadmium-induced stresses. Abstract: Estimation of overhead production enrichment in continuous foam separation was conducted with a surfactant: sodium n-dodecylbenzenesulfonate (SDBS) and soluble proteins: ovalbumin (OA) and hemoglobin (HB). Axial profiles of the volumetric flow rate and the concentration of the collapsed foam liquid within the column were measured, and the enrichment ratio and the liquid holdup in axial direction were determined experimentally. The proposed model was fitted to the experimental results obtained with various experimental conditions (superficial gas velocity, feed concentration and pH) and was in reasonable agreement with the experimental data by using the least square regression. Abstract: A rapid and simple technique (microthermometry) was developed for the measurement of microbe''s metabolic activities and its inactivation process by an inhibitor. To analyze the results and to determine the parameters for the estimation of the activity and inactivation degree, a simple model was proposed. Yeast cells (Saccharomyces cerevisiae) were used as a microbe. A differential method using two bead-type thermistors as reference and measuring probes was employed for the detection of a temperature change caused by the heat of metabolism of added carbon source (glucose) by the yeast. Experiments were conducted in a 30°C water bath under non-growth conditions (without nitrogen source). The simple thermal response model was applied to obtain two characteristic parameters for the estimation of the yeast activities, i.e., a metabolic heat production rate, ΔQ, and a metabolic heat inhibition rate, ΔK. The proposed model was well in agreement with the experimental results, and the curve fitting gave ΔQ or ΔK. In the case of the addition of glucose to yeast as a carbon source, ΔQ was proportional to the number of live cell (CFU). The slope of CFU versus ΔQ for yeast in exponential phase was larger by about two-fold than that in the stationary phase. In the case of the addition of gultaraldehyde as an inhibitor to the above system, the logarithmic value of ΔK increased with increasing the concentration of gultaraldehyde within the range of 0.02–6.2wt.%. It was suggested that the two model parameters, ΔQ and ΔK, can be regarded as characteristics to estimate the activities of bacteria and the degree of their inactivation by the inhibitor. Abstract: A microreactor for supercritical water has been developed for chemical engineering process. Heat and fluid flow in a T-junction in this system are calculated using STREAM. Since the flow in the tube are in the wide range from laminar to turbulent flow, a nonlinear k–ε model taking into account the low-Reynolds-number effects (proposed by Abe, Kondoh and Nagano) are used. We assume that a supercritical water velocity is 2.0 m/s. The optimum condition for time fluctuation of temperature is when the substrate velocity is 0.4. In this case, the substrate temperature rises above the critical temperature immediately and the main reaction progresses efficiently. When v is 0.1, the temperature remains subcritical region for a while. On the other hand, when the v is larger than 0.4, the temperature does not reach the critical point. [Copyright &y& Elsevier]

Experiments using natural substrates and artificial media were categorized into three groups. Using experiments in these three categories, we investigated dispersion, invasion sequence, and decomposing ability of ammonia fungi in the field on natural substrates. Spore germination and vegetative and reproductive growth of ammonia fungi derived from monocultures and five-species cultures were assessed on natural substrates and on artificial media. These features assist understanding of the colonization and successive occurrence of ammonia fungi in the field based on estimation of interactions among the fungi and the physiological characteristics of each ammonia fungus. The sequential colonization and fruiting of ammonia fungi in the field can be explained primarily by the preference or tolerance of a species to high concentrations of ammonium-N under alkaline to neutral conditions. Succession of ammonia fungi results from their sequential colonization associated with the time needed for each fungus to produce reproductive structures. Colonization likely initiates from mycelia and/or spores preinhabiting the soil and from newly invading airborne propagules. Duration of occurrence of ammonia fungi in the field is shortened by the interactions among them. Summary: To examine the relationship between the spore-breaking capability of collembolans and their localization within fungal sporocarps we investigated the proportion of broken spores in the feces of four collembolan species of the genus Hypogastrura using the spores of Hypsizygus marmoreus and analyzed the spatial distribution of the collembolan species within sporocarps as an index of their feeding habits. The collembolan species with a higher capability to break spores were observed more frequently from the spore-bearing surfaces of sporocarps. The collembolan species with a lower capability to break spores were found more frequently from the non-spore-bearing interiors of the sporocarps. These results suggest that the relative value of spores in the diet varied with the capability of collembolans to break spores. [Copyright &y& Elsevier]

Summary: We examined whether co-existing Hypogastrura species feeding on fruit-bodies show differences in utilization of fruit-bodies, and whether their utilization reflect their preferences for basidiomycete species, by field surveys and bait-trap experiments. In field surveys, the relative densities of the collembolans, Hypogastrura denisana, H. denticulata, and Hypogastrura sp. 1, on the selected basidiomycete species, Cortinarius salor, Lactarius quietus, and Russula emetica, differed significantly between the collembolan species. In bait-trap experiments using fruit-bodies of the three basidiomycete species as bait, the relative densities of the three collembolan species on the basidiomycete species differed significantly between the collembolan species and these differences were similar to the differences observed in the field surveys. These results suggest that the three collembolan species have differential preferences for fruit-bodies of the three selected basidiomycete species, and that the preferences relate to their utilization of fruit-bodies. Abstract: Yeast biomass was methylated in a 0.1 M HCl methyl alcohol solution at room temperature and the methylated yeast (MeYE) was applied to the adsorptive separation of Cr(VI) and As(V) anions from aqueous solutions. At near-neutral pH, while Cr(VI) and As(V) anions were scarcely adsorbed onto unmethylated yeast biomass, the amounts adsorbed increased with increasing methylation degree. The amount of Cr(VI) adsorbed onto MeYE was almost constant at pH 4–6 and decreased with increasing pH above pH 6. The amount of As(V) adsorbed onto MeYE was rather lower than that of Cr(VI) and it had a peak at about pH 7. A metal-binding model was used to describe the adsorption characteristics of Cr(VI) and As(V) on MeYE. The results showed that MeYE has two different types of adsorption sites. The saturated amount of Cr(VI) and As(V) adsorbed onto MeYE having methylation degree 0.94 was 0.55 mmol g-1. [Copyright &y& Elsevier]

PTEN is a tumor suppressor gene mutated in many human cancers, and its expression is reduced or absent in almost half of hepatoma patients. We used the Cre-loxP system to generate a hepatocyte-specific null mutation of Pten in mice (AlbCrePtenflox/floxmice). AlbCrePtenflox/flox mice showed massive hepatomegaly and steatohepatitis with triglyceride accumulation, a phenotype similar to human nonalcoholic steatohepatitis. Adipocyte-specific genes were induced in mutant hepatocytes, implying adipogenic-like transformation of these cells. Genes involved in lipogenesis and β-oxidation were also induced, possibly as a result of elevated levels of the transactivating factors PPARγ and SREBP1c. Importantly, the loss of Pten function in the liver led to tumorigenesis, with 47% of AlbCrePtenflox/flox livers developing liver cell adenomas by 44 weeks of age. By 74-78 weeks of age, 100% of AlbCre tenflox/flox livers showed adenomas and 66% had hepatocellular carcinomas. AlbCrePtenflox/flox mice also showed insulin hypersensitivity. In vitro,AlbCrePtenflox/flox hepatocytes were hyperproliferative and showed increased hyperoxidation with abnormal activation of protein kinase B and MAPK. Control of pandemics requires strong leadership not only by central government but also in local fields, where people actually fight against it. Miyagi Pandemic Influenza Study Group was organized in Sendai area, Japan, in February 2001, and an attempt to study preparation against pandemic influenza was started. Its members are from local governments and hospitals including administrative officers, physicians and laboratory workers, and other functions including manufacturer of an antiviral, army headquarter, academia and local press, although all are volunteer participants. Its main goal is to prepare against coming large-scale epidemic of influenza as concretely as possible in our own local communities. A group meeting has been held every month or two for lectures, presentation and discussion. Documents and other materials were collected and stocked to supply anyone who wants them as references to learn about past pandemics and the present planning. In addition, materials written in English have been translated into Japanese by the members to make them easily accessible for ordinary Japanese people. Discussion in the meetings includes the priority for vaccination or antiviral drugs among staffs of hospitals and communities, several desktop simulations and ways of local governments to manage health and social crises. A series of studies were performed to investigate the ability of rapid antigen-detection kits in diagnosis of influenza. Clinical specimens were collected from a total of more than 3000 cases of influenza-like illness who visited out-patient clinics in Sendai and Fukuoka, Japan, during the 2001–2002 and 2002–2003 influenza seasons. Each clinical specimen was examined with a kit and virus isolation, and results were analyzed retrospectively by several points of interest: sensitivity of the kits based on the virus isolation and the sensitivity by type and subtype of influenza, kinds of specimens, the time course of influenza, and variation of sources of the specimens. Thus, the sensitivities of a kit were 83%, 52% and 58% in general for influenza A(H3N2), A(H1N1), and B, respectively, and those of another kit were 76%, 71%, and 62%, respectively, in 2001–2002 influenza season. The sensitivity using nasal swabs was better than that using throat swab, though only 5% up. Specimens tended to be less sensitive when collected shortly after the onset of illness, and the sensitivity seemed quite varied (17–92%) by source of specimens: staff who collect the specimens, and/or by population of the patients. We have used activation tagging with T-DNA carrying cauliflower mosaic virus 35S enhancers to investigate the complex signaling networks underlying disease resistance in Arabidopsis. From a screen of ~5000 lines, we identified constitutive disease resistance (CDR1) encoding an apoplastic aspartic protease, the overexpression of which causes dwarfing and resistance to virulent Pseudomonas syringae. These phenotypes reflect salicylic-acid-dependent activation of micro-oxidative bursts and various defense-related genes. Antisense CDR1 plants were compromised for resistance to avirulent P. syringae and more susceptible to virulent strains than wild type. CDR1 accumulates in intercellular fluid in response to pathogen attacks. Induction of CDR1 generates a small mobile signal, and CDR1 action is blocked by the protease inhibitor pepstatin and by mutations in the protease active sites. A new biodegradable flocculant was prepared from a common and inexpensive protein. Milk casein was methylated in a 0.05 M HCl methyl alcohol solution at room temperature. The methylated milk casein (MeCS), having a methylation degree of 81%, was applied to the separation or flocculation of diatomite in seawater (pH 8.1±0.1) at room temperature (18–23 °C). The flocculating ability of MeCS was evaluated by a sedimentation balance method (cumulative measurement method). The diatomite suspension was effectively flocculated by the addition of a small amount of MeCS (0.25 wt% of the diatomite weight). The results of sedimentation analysis showed that the size–frequency curve had a very sharp and high peak; thus the diatomite floc formed by MeCS had a rather uniform size. The settling velocity of diatomite floc at the appropriate MeCS dosages (0.25–2 wt% of diatomite) was about 3×10−3 m s−1. [Copyright &y& Elsevier]

A new biosorbent, methylated yeast (MeYE), was prepared for the adsorptive separation of proteins from aqueous solutions. Yeast was methylated in a 0.1 M HCl methyl alcohol solution at room temperature. About 80% of the carboxylic groups of yeast could be methylated within 9 h. The adsorption of egg albumin onto MeYE was studied to evaluate the protein adsorption ability of MeYE. At near neutral pH, egg albumin was scarcely adsorbed onto unmethylated yeast and the adsorbed amount of egg albumin increased with increasing methylation degree. The amount of egg albumin adsorbed onto MeYE increased with increasing pH from 4 to 7 and steeply decreased above pH 7. The Langmuir isotherm was applied to determine the apparent adsorption constant and the saturated adsorbed amount of egg albumin on MeYE. Both the apparent adsorption constant and the saturated adsorbed amount increased with the degree of methylation. The saturated adsorbed amount of egg albumin onto MeYE having methylation degree 77% was 8.41×10−6 mol g−1 or 0.378 g g−1 at near neutral pH. [Copyright &y& Elsevier]

Platelet-derived growth factor plays a critical role to convert bone marrow cells into glomerular mesangial-like cells. Background. Despite increasing interest in bone marrow–derived stem cells, little is known about critical factors that determine their fates both in vitro and in vivo. Recently, we have reported that bone marrow is a reservoir for glomerular mesangial cells in rats. To find a key factor responsible for the differentiation of bone marrow–derived cells into mesangial cells, we established a new culture system of rat bone marrow, which is based on serial replating and differential attachment to collagen types I and IV. Methods. Bone marrow cells that did not adhere to collagen type I within 24 hours were transferred to collagen type IV–coated dishes. Then, the cells attached to collagen type IV in the following 24 hours were maintained in the presence of 2% horse serum, 200 ng/mL of platelet-derived growth factor (PDGF)-BB, and 1 μmol/L of all- trans retinoic acid. In vivo effect of PDGF-B was also examined by introducing human PDGF-B gene into glomeruli. Results. After cultivation under the above condition for 7 days, approximately 14% of cells expressed Thy-1 and desmin, both of which are markers for rat mesangial cells. Thy-1++/desmin+ cells were stellate-shaped, and contracted in response to angiotensin II. When human PDGF-B gene was overexpressed in the glomeruli of chimeric rats whose bone marrow was transplanted from enhanced green florescent protein (EGFP) transgenic rats, the number of EGFP+ mesangial cells increased. This effect was canceled by prior introduction of a neutralizing molecule that is composed of PDGF receptor-β ligand binding site and IgG-Fc. Conclusion. We give an alternative definition of comprehensive Gro¨bner bases in terms of Gro¨bner bases in polynomial rings over commutative Von Neumann regular rings. Our comprehensive Gro¨bner bases are defined as Gro¨bner bases in polynomial rings over certain commutative Von Neumann regular rings, hence they have two important properties which do not hold in standard comprehensive Gro¨bner bases. One is that they have canonical forms in a natural way. Another one is that we can define monomial reductions which are compatible with any instantiation. Our comprehensive Gro¨bner bases are wider than Weispfenning’s original comprehensive Gro¨bner bases. That is there exists a polynomial ideal generated by our comprehensive Gro¨bner basis which cannot be generated by any of Weispfenning’s original comprehensive Gro¨bner bases. A common and inexpensive protein, egg albumin, was applied to the solid–liquid separation or flocculation of diatomite. Egg albumin was methylated in a 0.05 M HCl methyl alcohol solution at room temperature. About 90% of the carboxylic groups of egg albumin could be methylated within 24 h. The adsorption of egg albumin onto diatomite at pH 6.8 was remarkably enhanced by methylation. The adsorption constant of methylated egg albumin to diatomite at 30 °C was about 100-fold larger than that of native egg albumin; however, the adsorption constant of methylated egg albumin decreased to about 1/100 with temperature decreasing from 30 to 6 °C. The saturated adsorption amount of egg albumin was also increased by the methylation. The flocculating ability of methylated egg albumin was examined with a diatomite suspension at 6 and 30 °C in the pH range from pH 2 to 11. The diatomite suspension was effectively flocculated by the addition of small amounts of methylated egg albumin (only 0.5–1 wt% against diatomite) over a wide pH range from pH 3 to 10. A fundamental study about the selective foam separation of protein mixture was carried out. A solution containing two proteins, ovalbumin (OA) and lysozyme (LZ), and an anionic surfactant, sodium dodecyl sulfate (SDS), was adjusted to pH 6.0, which referred to an intermediate state between the isoelectric points of the proteins. The solution was processed by continuous foam separation. The results showed that a proper addition of SDS greatly improved the selective recovery of LZ to OA. The experimental data were well explained by a simple model that most of cationic protein molecules (LZ) are associated with SDS and the adsorption of all the species including LZ–SDS complexes are subjected to Langmuir adsorption isotherm. The results also showed that one of the Langmuir parameters, which means a kind of lyophillic property of adsorbed material, of LZ–SDS complexes was extremely large as compared with that of primary protein. Tornado extraction: A method to enrich and purify RNA from the nephrogenic zone of the neonatal rat kidney. Background. Development of the kidney is a complicated and tightly regulated process. Although several genes responsible for the renal development have been identified to date, the precise mechanisms of spatial and temporal regulation remain to be elucidated. Therefore, expanding our knowledge of molecules that are associated with nephrogenesis will be helpful to understand the whole process. Methods. To extract RNA selectively from the nephrogenic zone of the developing kidney, we developed a simple and reliable method. Results. This method, named “tornado extraction,” enriched RNA of the nephrogenic zone by about 30-fold. In combination with the suppression subtractive hybridization, a considerable number of genes that were differentially expressed in the nephrogenic zone were obtained. These genes included a series of endodermal markers such as albumin and α-fetoprotein as well as GDNF (glia-derived neurotrophic factor), osteoblast-specific factor-2 (OSF-2)/periostin and fetuin (one of the major serum proteins in the fetus). Conclusion. Tornado extraction has great value in studying genes in the nephrogenic zone of the developing kidney. Since the quality of RNA obtained by this method is excellent, tornado extraction is suitable in combination with other techniques including the subtractive hybridization method and DNA microarray analysis. Adsorption of cadmium and lead ions to floc-type biosorbents was reported in this work. Two types of biosorbents containing a marine microalga, Heterosigma akashiwo (Hada) Hada, or a purple non-sulfur bacterium, Rhodobacter sphaeroides, were prepared. The micro-organisms inactivated by steam sterilization were immobilized in casein floc and cross-linked with glutaraldehyde. In the present immobilizing method, we obtained the biosorbents comprising as much as 67% of micro-organism on a dry-weight basis. Simple metal-binding models were applied to explain the adsorption mechanism of bivalent metal ions to the biosorbents. The results showed that casein acted as both the immobilizing material and the adsorbent material. The adsorption of bivalent metal ions to the biosorbents was due to bidentate binding to the acidic sites on casein and monodentate binding to the acidic sites on micro-organisms. The metal-binding constants and the binding capacities of micro-organisms were scarcely influenced by immobilization. [Copyright &y& Elsevier]

A kinetic study of cadmium and lead biosorption to a brown alga, Kjellmaniella crassiforia, was carried out. The shrinking core model derived by M. Gopala Rao and A. K. Gupta (Chem. Eng. J.24, 181, (1982)) was modified and adapted for description of the rate process of cadmium and lead biosorption to the alga. The biosorption rate process was well described and average apparent diffusion coefficient of about 9×10−6 cm2 s−1 was found for both cadmium and lead ions. The value was 20 to 50 times higher than the apparent diffusion coefficients of cadmium and lead ions in strong-acid resins like Dowex 50W-X8. [Copyright &y& Elsevier]

The electronic properties of an Al Σ = 9 tilt grain boundary with segregated Na and Ca impurity atoms have been calculated by a first-principles pseudopotential method. The results show that the boundary expands and the charge density decreases significantly over the whole boundary by the segregation of Na and Ca, and there are no stronger bonds than those associated with metallic bonding in the boundary even with the impurities. It is concluded that the embrittlement promotion mechanism by Na or Ca segregation in the Al boundaries is one kind of 'decohesion model'. [ABSTRACT FROM AUTHOR]

The development of next-generation sequencing (NGS) has enabled the discovery of cancer-specific driver gene alternations, making precision medicine possible. However, accurate genetic testing requires a sufficient amount of tumor cells in the specimen. The evaluation of tumor content ratio (TCR) from hematoxylin and eosin (H&E)-stained images has been found to vary between pathologists, making it an important challenge to obtain an accurate TCR. In this study, three pathologists exhaustively labeled all cells in 41 regions from 41 lung cancer cases as either tumor, non-tumor or indistinguishable, thus establishing a "gold standard" TCR. We then compared the accuracy of the TCR estimated by 13 pathologists based on visual assessment and the TCR calculated by an AI model that we have developed. It is a compact and fast model that follows a fully convolutional neural network architecture and produces cell detection maps which can be efficiently post-processed to obtain tumor and non-tumor cell counts from which TCR is calculated. Its raw cell detection accuracy is 92% while its classification accuracy is 84%. The results show that the error between the gold standard TCR and the AI calculation was significantly smaller than that between the gold standard TCR and the pathologist's visual assessment ( p < 0.05 ). Additionally, the robustness of AI models across institutions is a key issue and we demonstrate that the variation in AI was smaller than that in the average of pathologists when evaluated by institution. These findings suggest that the accuracy of tumor cellularity assessments in clinical workflows is significantly improved by the introduction of robust AI models, leading to more efficient genetic testing and ultimately to better patient outcomes.
