Background: Epidemiological data regarding the incidence and mortality of sepsis are scarcely available. The present study aimed to delineate the epidemiology of sepsis and related mortality among patients hospitalized in Sicily, Italy. Methods: Data on the discharge forms of all patients with sepsis in all Sicilian hospitals from January 2016 to December 2020 were retrospectively collected. Results: In Sicily, 15,373 hospitalizations associated with sepsis occurred from 2016 to 2020, with an overall in-hospital mortality rate of 36.3%. The percentage of hospitalizations associated with sepsis represented 0.65% of all admissions, with an increase over the years. Most patients were admitted to non-surgical and non-infectious disease units, accounting for 50.3% of total admissions. Stratification of patients according to age, intensive care unit (ICU) admission, and presence of septic shock revealed variations in in-hospital mortality rates. Among adult ICU-admitted patients with septic shock, mortality was highest at 81.0%, followed by non-ICU adult patients with septic shock (63.5%), ICU pediatric patients with septic shock (56.7%), ICU adult patients without septic shock (43.9%), non-ICU pediatric patients with septic shock (37.9%), non-ICU adult patients without septic shock (17.9%), ICU pediatric patients without septic shock (10.4%), and non-ICU pediatric patients without septic shock (1%). The mineralized cover preserving soft parts of the Dasycladales, as an external mould, is the only tool allowing to deduce the anatomical parts of these algae. Problems arise when voids within the calcareous skeleton do not reflect accurately the original soft parts due to a loss of mineralization. Based on a rich material collected from the type locality in Languedoc (France), a detailed study was carried out on Chinianella ellenbergeri, the type species of Chinianella. Previous interpretation of voids in the calcified fossil considered the alga to be made of an alternation of fertile and further ramified sterile whorls. Our study shows that large cavities previously interpreted as corresponding to fertile whorls, actually denote empty spaces. Therefore, the alga essentially consists of spaced-out whorls of sterile primary laterals, distally bearing a tuft of three-four phloiophorous secondary laterals. A small number of specimens show the presence of calcified structures interpreted as corresponding to external, subterminal reproductive organs. Consequently, an emended diagnosis is proposed for C. ellenbergeri and for the Chinianella. Present interpretation of Chinianella adds a new step to the understanding of the emergence of external reproductive organs during the geological history of this group of green algae. Chinianella ellenbergeri shows that choristosporate reproduction sensu lato was already well represented and diversified in the Early Jurassic. Other Late Triassic-earliest Jurassic species formerly attributed to Chinianella (namely C. carpatica, C. crosii, C. zanklii, C. micropora, and C. macropora) reveal that the large, fertile primary lateral is a compound pore with a structure like in Cymopolia, i.e., made of a short primary lateral with a terminal gametophore embraced by secondary laterals. In the present paper we provide a new estimate of the body mass (BM) of the dwarf elephant Palaeoloxodon ex gr. P. falconeri from Spinagallo cave (Sicily) at three different ontogenetic stages. The new estimates are based on 3D in-vivo restorations, digitally sculpted on anatomically updated photogrammetric models of the skeletons mounted at the Museo Universitario di Scienze della Terra (MUST), University of Rome (Italy). The new method provided an average BM of 249.98 kg for the adult male, 150.47 kg for the adult female, 38.81 kg for the juvenile male and 7.83 kg for the new-born male. Assuming as reliable these estimates, the values previously obtained by applying the regression formulae based on pad circumferences, shoulder height and long bone circumferences substantially would underestimate or overestimate the Spinagallo elephant BM up to 103%. The results obtained indicate that the volumetric methods can be regarded as the most solid and reliable to estimate the BM in those extinct vertebrates for which accurate mounted skeletons are available. This new method is therefore of critical importance for inferring the BM of taxa characterised by peculiar osteological proportions, such as those shown by the insular dwarf elephant analysed in the present work. This study examines the impact of bottom trawl fishing on the macrobenthic communities inhabiting the coastal terrigenous mud off the northern coast of Sicily (Western Mediterranean). Two intensely trawled gulfs were compared with two gulfs from which trawling has been excluded for 15 years. The results show a significant effect of trawling on the faunal assemblage and when comparing the mean biomass and the whole isotopic composition of the benthic communities. A similar pattern, although not significant, was found for total abundance, biomass, production/biomass ratio and diversity. Higher abundance and lower biomass were found in the untrawled areas, attributable to the presence of more numerous yet smaller individuals, possibly a consequence of more abundant larger predators that are not removed by trawling, and consequent higher predatory pressure on the benthic macrofauna. The SIMPER analysis evidenced a dominance of burrowing deposit feeding worms (Paraonidae and Cossuridae) in trawled areas, as a result of increased mechanical alteration and hence more organic matter available as food. In contrast, the response to trawling as drawn by the use of trophic markers (i.e., stable isotopes) was less clear. While δ 15 N of benthic taxa did not vary significantly between untrawled and trawled areas, δ 13 C was higher in trawled areas possibly due to high sediment resuspension and consequent intense microbial activity. Mixing models confirmed higher reliance to a detritus-based food web for benthic organisms in the trawled areas. Standard Ellipse Areas (SEAc) as a measure of community niche width were slightly larger in trawled areas, likely due to higher generalism triggered by alteration/removal of the original benthic community. DNA samples from 465 blood donors living in 7 towns of Sicily, the largest island of Italy, have been collected according to well defined criteria, and their genetic heterogeneity tested on the basis of 9 autosomal microsatellite and mitochondrial DNA polymorphisms for a total of 85 microsatellite allele and 10 mtDNA haplogroup frequencies. A preliminary account of the results shows that: a) the samples are genetically heterogeneous; b) the first principal coordinates of the samples are correlated more with their longitude than with their latitude, and this result is even more remarkable when one outlier sample (Butera) is not considered; c) distances among samples calculated from allele and haplogroup frequencies and from the isonymy matrix are weakly correlated (r = 0.43, P = 0.06) but such correlation disappears (r = 0.16) if the mtDNA haplogroups alone are taken into account; d) mtDNA haplogroups and microsatellite distances suggest settlements of people occurred at different times: divergence times inferred from microsatellite data seem to describe a genetic composition of the town of Sciacca mainly derived from settlements after the Roman conquest of Sicily (First Punic war, 246 BC), while all other divergence times take root from the second to the first millennium BC, and therefore seem to backdate to the pre-Hellenistic period. A more reliable association of these diachronic genetic strata to different historical populations (e.g. Sicani, Elymi, Siculi), if possible, must be postponed to the analysis of more samples and hopefully more informative uniparental DNA markers such as the recently available DHPLC-SNP polymorphisms of the Y chromosome. In a recent non-destructive analytical campaign at Museo Civico, Castello Ursino, in Catania, Italy, several paintings in the permanent collection were investigated by MA-XRF scanning, with a special focus on Matthias Stomer's production. On one depiction of the Mocking of Christ (ca. 1640) donated to the municipality of Catania by G.B. Finocchiaro in 1826, the analysis documented the use of Naples yellow. Sb with Pb was detected in yellow areas of the Mocking of Christ, but not in his work Tobias healing his father. This finding possibly suggested an early use of lead antimonate yellow in South Italy, although it is generally accepted that this pigment was introduced in painting in the eighteenth century. Further details on his technique and later conservation treatments are provided, as well as literary comparisons with the artistic production during the same period, in Sicily and elsewhere. A systematic study of Stomer's works, for example examining paintings produced while he was in Naples or Rome, might determine whether this material choice depended on local availability. Overall, it would shed light on his technique, as well as on the history of Naples yellow in southern Italy and beyond, before this pigment became so popular in the eighteenth century. The Hunga Tonga-Hunga Ha'apai volcano (Pacific Ocean) generated a cataclysmic explosion on 15 January 2022, triggering several atmospheric disturbances at a global scale, as a huge increase in the total electron content (TEC) in the ionosphere, and a pressure wave travelling in the troposphere. We collected and analysed data over the Mediterranean to study these disturbances, and in particular, (i) data from the barometric and infrasonic stations installed on Italian active volcanoes by the Istituto Nazionale di Geofisica e Vulcanologia (INGV) for investigating the tropospheric pressure waves; (ii) barometric data from the INGV-TROPOMAG and SIAS (Sicilian Agro-meteorological Information System) networks, for investigating the interaction between the orography and pressure waves; (iii) ionograms from the Advanced Ionospheric Sounder-INGV ionosonde at Gibilmanna (Sicily, Italy); (iv) data from the RING (Rete Italiana Integrata GNSS) network, to retrieve the ionospheric TEC; (v) soil CO2 flux data from the INGV surveillance network of Vulcano Island. The analysis of the ground-level barometric data highlights that pressure waves were reflected and diffracted by the topographic surface, creating a complex space–time dynamic of the atmospheric disturbances travelling over Sicily, driven by the interference among the different wavefronts. The ionograms show that a medium-scale travelling ionospheric disturbance (MSTID), with a horizontal wavelength of about 220 km and a period of about 35 min, propagated through the ionospheric plasma in the correspondence of the first barometric variations. Moreover, comparing detrended TEC and barometric data, we further confirmed the presence of the aforementioned MSTID together with its close relation to the tropospheric disturbance. Notification of foodborne outbreaks has been mandatory in Europe since 2005, and surveillance is carried out along the entire food chain. Here we report the results obtained from laboratory investigations about four cases of foodborne outbreaks that occurred in Sicily between 2009 and 2016, deemed to be related to staphylococcal enterotoxins (SEs) and coagulase-positive Staphylococci (CPS) by the Local Public Health Authority. Primosale cheese samples were processed by culture methods for enumeration of CPS and immunoenzymatic assays for detection and differentiation of the SEs possibly contained in food samples. In all cases, the mistrusted foods were found to be contaminated by CPS at bacterial loads between 5 and 8 log CFU/g and contained SE type C (SEC). The reported data confirm the risk of staphylococcal food poisoning associated with the consumption of raw milk cheese. SEC is the most commonly occurring SE in goat milk and dairy products and the most represented enterotoxin in Sicilian dairy products. Our results highlighted the need for improving the current monitoring efficiency and implementing the available laboratory methods to collect more faithful epidemiological data on the current prevalence of staphylococcal toxins in the food chain, including SEs currently not detectable by validated analytical methods. The aim of this research was the study of indigenous yeasts isolated from spontaneous fermentation of Inzolia grapes, one of the most widespread native white grapes in Sicily (Italy). The use of selective medium for the isolation and the screening for sulphur dioxide tolerance were useful for the first selection among 640 isolates. The yeasts characterized by high SO tolerance were identified at species level by restriction analysis of ITS region; although the majority of isolates were identified as S. cerevisiae, some non- Saccharomyces yeasts were found. Forty-seven selected yeasts, both S. cerevisiae and non- Saccharomyces yeasts, were characterized for genetic and technological diversity. The genetic polymorphism was evaluated by RAPD-PCR analysis, whereas the technological diversity was analyzed by determining the main secondary compounds in the experimental wines obtained by inoculating these yeasts. Both the molecular and metabolic profiles of selected yeasts were able to clearly discriminate S. cerevisiae from non- Saccharomyces yeasts. This research was useful for the constitution of a collection of selected indigenous yeast strains, including S. cerevisiae and non- Saccharomyces species possessing interesting enological traits. This collection represents a source of wild yeasts, among of which it is possible to select indigenous starters able to maintain the specific organoleptic characteristics of Inzolia wine. In this study, a first record of Ribes uva-crispa L. (Grossulariaceae) - a new species of the Sicilian flora - from the Madonie Mts. is reported. The autochthonous and relict new population of Ribes uva-crispa L. (Grossulariaceae) is found on Mount Carbonara (Madonie Mts., North-Central Sicily), which is several hundred kilometers away from the Central Apenninic and the South Mediterranean locations of its distribution range. R. uva-crispa shows a distribution similar to other taxa extending from the Euro-Asiatic area to the mountains of North Africa with disjunct populations on the main Sicilian reliefs (Madonie and Nebrodi Mts.), testifying ancient phytogeographical connections. This noteworthy record adds a new species - as well as a new genus and new family - to the vascular flora of Sicily. This paper deals with some cave collembola from Sicily. The analysis has revealed the presence of the species Acherontiella carusoi Dallai, 1978, Heteromurus nitidus (Templeton, 1835), Disparrhopalites patrizii (Cassagnau and Delamare, 1953), and Serrodeurs trinacriae n. sp., new to science. This is the first record of the genus Serroderus for Sicily and Italy. The new species is characterized by the multidentate shape of the mucron, the presence of three to four spines, and by the number of scales on the inner (five) and outer (eight) rows of the dens. The biogeographical distribution of the genus and the taxonomic relationships of S. trinacriae n. sp. are discussed. [ABSTRACT FROM AUTHOR]

Archaeological excavations carried out in the town of Palermo revealed important traces of metallurgical activity related to the Punic Panormos. Five samples of iron slags, recovered during the digging, were characterized by micro-structural, mineralogical and chemical investigations as well as by environmental scanning electron microscopy, energy-dispersive X-ray spectroscopy, X-ray powder diffraction and X-ray fluorescence. The studied remains are spongy, rust colored, with a plano-convex shape and their textural and mineralogical features suggest they are metallurgical slags produced during smithing process. Wüstite, magnetite, fayalite, kirschsteinite, hedembergite, cristobalite and quartz are the main identified mineral phases, while goethite, lepidocrocite and calcite occur as minor secondary phases. The iron slags show heterogeneous structures such as hammering clues, layering in wustite crystals and presence of calcium-rich minerals which are consistent with a broad variation of the forging temperature due to the use of different smithing techniques. This assumption is confirmed by the different crystallization temperatures of the detected mineral phases. Skeletal and dendritic shapes are indicative of rapid cooling and, possibly, quenching. The chemical composition is dominated by iron, silicon and calcium and it is almost constant for all the samples suggesting that they were produced with similar raw materials and technology. The high Ca content suggests the use of carbonate-bearing rocks, widely outcropping over the Palermo territory, probably exploited as fluxing ores for the metallurgical process. The multidisciplinary approach adopted in this study provided new data for a better understanding of the metallurgical techniques evolution in the Sicilian territory in ancient times. Since the studied iron slags represent the unique traces so far known of metallurgical activity in the ancient Palermo town, our results open interesting perspectives for future study of metallurgy in ancient Sicily. Abstract: Late Triassic basaltic rocks crop out in the Lercara area in Western Sicily. Major and trace element composition, as well as Sr–Nd isotopic ratios (87Sr/86Sr i =0.7074−0.7076; εNd i =from −0.69 to −1.09) of the Lercara rocks shows many similarities with Large Ion Lithophile Elements (LILE)- and Light Rare Earth Elements (LREE)-rich tholeiitic basalts of the Central Atlantic Magmatic Province (CAMP), that erupted during the Mesozoic fragmentation of the Pangea supercontinent and subsequent opening of the Central Atlantic Ocean. The geochemical features of the Lercara igneous rocks, together with the spatial distribution of the ~200Ma old CAMP rocks are unlikely to be associated with the arrival of a thermal anomaly in the form of a mantle plume and are more compatible with adiabatic melting of passively upwelling sub-lithospheric mantle. The original melts variably interacted with lower crustal rocks before reaching the surface. AFC modeling suggests two distinct differentiation paths including either simple mixing or assimilation-fractional crystallization processes involving lower crustal rocks. We report the identification of four residual stations forTrifolium michelianumSavi (Fabaceae) in Sicily, whereas the species was documented to be extinct at the Gorgo Cerro station (western Sicily), the only locality previously reported in the literature. In addition to an update on the distribution of the species, a survey on the new Sicilian populations led to their biological, ecological and phytosociological characterization, as well as to an assessment of the risk factors. The species, here relegated to the limit of its distribution area, occurs in particularly sensitive and vulnerable environments, such as the “Mediterranean temporary ponds”, considered “priority” by the Council Directive 92/43/EEC. To assess whether arthropod bites promote Kaposi sarcoma (KS), we determined the seroprevalence of Sicilian (SFSV) and Toscana (TOSV) phlebovirus antibodies in 30 patients with classic KS and 100 controls in Sicily. Nine (6.9%) subjects, all controls, were positive for SFSV, whereas 41 (31.5%) were positive for TOSV. Seroprevalence with immunoglobulin (Ig) M or IgG against either virus was significantly higher in controls (43% vs 13.3% in case patients; P < .01). Adjusted for age, IgG seroprevalence was significantly lower in KS patients compared to controls (adjusted odds ratio, 0.22; 95% confidence interval, .07-.72). Low phlebovirus seroprevalence in patients with KS may reflect incapacity to produce robust, persistent antibody responses, and suggests that arthropod bites do not promote KS. Abstract: The aim of the study was to investigate factors involved in vaccination acceptance among healthcare workers (HCWs) and adverse reactions rates associated with pandemic influenza vaccination. The study was carried out in the major teaching hospital of Sicily from November 2009 to February 2010 on 2267 HCWs. A total of 407 (18%) HCWs were vaccinated against the 2009 pandemic influenza A (H1N1). A logistic regression analysis indicates an increased risk of non-vaccination against pandemic influenza in females (OR=1.6; 95% CI=1.3–2.1) compared to males, in nurses/technicians/administrative workers (OR=1.7; 95% CI=1.3–2.2) compared to doctors/biologists, and in HCWs who were non-vaccinated against seasonal influenza in 2008–2009 (OR=4.9; 95% CI=3.7–6.5) compared to vaccinated HCWs. Overall, 302 (74.2%) out of 407 questionnaires distributed to vaccinated HCWs were returned within the observation period. One hundred fifty-two workers (50.3%) experienced at least one adverse reaction (30.1%, local reactions; 6.6% systemic reactions and 13.6% both of them). The most frequent side effect of vaccination was pain at the injection site (43.4%). Twelve (3.9%) out of 302 HCWs stated they experienced influenza-like illness episodes during the follow-up period. The use of an adjuvanted vaccine against pandemic influenza A (H1N1) appears to be an effective and safe preventive strategy, showing a prevalence of both local and systemic adverse reactions not very different from that seen after vaccination with non-adjuvanted seasonal influenza vaccine. Despite this finding, vaccination coverage among HCWs remains very low, suggesting the need to implement educational campaigns directed to groups with lower coverage rates. [Copyright &y& Elsevier]

Abstract: The present research studied Saccharomyces cerevisiae yeasts isolated from Nero d''Avola grapes, collected in different areas of the Sicily region. RAPD-PCR analysis with M13 primer was used for preliminary discrimination among 341 S. cerevisiae isolates. Inoculated fermentations with S. cerevisiae strains, exhibiting different RAPD-PCR fingerprinting, revealed the impact of selected strains on volatile compound concentration. Two selected strains were used in fermentation at cellar level and the restriction analysis of mtDNA on yeast colonies isolated during fermentation was used to control strain implantation. This study represents an important step to establish a collection of indigenous S. cerevisiae strains isolated from a unique environment, such as Nero d''Avola vineyards. Different starter implantation throughout inoculated fermentation represents an additional character, which might be considered during the selection program for wine starter cultures. Abstract: The progressive and rapid aging of population is the demographic characteristic in the Western countries. This rapid process of aging is causing an increasing burden on the social and health-care services. In this context, the precise knowledge of the environmental, socio-economical and clinical characteristics of the elderly population is mandatory to find the correct strategies to achieve the successful aging. Our study aimed to investigate the functional and clinical characteristics of the elderly (aged 60 to 85 years) of San Teodoro (1500 inhabitants), a rural village of Central Sicily, in particularly considering the dementia prevalence. In 2005, all the elderly between 60 and 85 years old were invited to participate to the study. The list of the potential participants was obtained from the Registry office of the municipality. The final number of the eligible subjects was 374. Rate of participation was 74.9% (280 subjects, 120 M and 160 F). The study was conducted door-to-door. Dementia prevalence was 7.1% (20 subjects, 8 M and 12 F), with 60% Alzheimer''s disease and 15% vascular dementia, slightly higher than that of the European countries (6%). The high prevalence of hypertension (80.3%) and the low education level, two important risk factors for dementia, could explain in part the difference observed. [Copyright &y& Elsevier]

The presence or absence of genetic heterogeneity in Sicily has long been debated. Through the analysis of the variation of Y-chromosome lineages, using the combination of haplogroups and short tandem repeats from several areas of Sicily, we show that traces of genetic flows occurred in the island, due to ancient Greek colonization and to northern African contributions, are still visible on the basis of the distribution of some lineages. The genetic contribution of Greek chromosomes to the Sicilian gene pool is estimated to be about 37% whereas the contribution of North African populations is estimated to be around 6%.In particular, the presence of a modal haplotype coming from the southern Balkan Peninsula and of its one-step derivates associated to E3b1a2-V13, supports a common genetic heritage between Sicilians and Greeks. The estimate of Time to Most Recent Common Ancestor is about 2380 years before present, which broadly agrees with the archaeological traces of the Greek classic era. The Eastern and Western part of Sicily appear to be significantly different by the χ2-analysis, although the extent of such differentiation is not very high according to an analysis of molecular variance. The presence of a high number of different haplogroups in the island makes its gene diversity to reach about 0.9. The site of Rocchicella, near Catania, in eastern Sicily, has yielded important archaeological evidence from prehistoric times to the Middle Ages. Extensive archaeological investigations of cultural layers dating from the Palaeo-Mesolithic to the Copper Age have recently been undertaken, and volcanic glass, mainly obsidian, has been collected in the course of excavation. To determine the provenance of this volcanic glass, a non-destructive elemental analysis was carried out to measure the concentration of characteristic trace elements. The analysis was carried out using a new XRF spectrometer equipped with a beam stability controller and a quantitative method developed at the LANDIS laboratory of the INFN–CNR Institutes of Catania. In addition to the obsidian, it was demonstrated for the first time that a local vitreous material similar to obsidian, but displaying a completely different composition, was used during all the investigated periods. This material was identified as a basaltic glass, characterized by a superficial product of devitrification called palagonite. Analysis of the obsidians has led to the identification of the island of Lipari as the provenance source. Background: Association of thyroid dysfunction with plasma homocysteine levels and vitamin B12 has previously been reported. We evaluated these associations in the elderly in San Teodoro, a mountainous village of Sicily. Methods: Subjects (n=279) aged 60–85 years (119 males and 160 females) were examined using self-reported signs, clinical examination and laboratory tests. Results: Hypothyroidism and/or goiter were two characteristics that were not associated with a significant change in homocysteine when compared with euthyroidism and the absence of goiter. Vitamin B12 was significantly higher in subjects in the first quartile of the thyroid-stimulating hormone distribution, compared with those in the fourth quartile (371±207 vs. 297±196 pmol/L, p=0.0121). Homocysteine was significantly higher in the first quartile of the free tri-iodothyronine distribution compared to the third quartile (18.0±5.7 vs. 16.0±6.2 μmol/L, p=0.0130) and was correlated with log tri-iodothyronine in euthyroid subjects (p=0.0254). In multivariate analysis, homocysteine was associated with vitamin B12 (p=0.0014), folate (p<0.0001), creatinine (p<0.0001) and age (p<0.0001), but not with either free tri-iodothyronine (p=0.7680), tetra-iodothyronine (p=0.5706) or thyroid-stimulating hormone (p=0.2294). Conclusions: Our results suggest that the influence of thyroid hormones on homocysteine is much weaker in elderly subjects than in selected patients with hypothyroidism. Background: Homocysteine is associated with age, folate and vitamin B12. Our study investigated the functional and clinical characteristics of the elderly (aged 60–85 years) of San Teodoro, a village in Central Sicily, and evaluated associations with vitamin B12, folate and homocysteine. Methods: Subjects (n=280) were examined after door-to-door recruitment using interview, physician examination and laboratory tests. Results: A total of 19.3% of the population had a low blood level of folate (<7 nmol/L) and 3.2% had low vitamin B12 concentration (<100 pmol/L). The level of dependency, determined by the Barthel index, influenced homocysteine blood levels (p<0.0001), independent of age (p<0.0001), folate (p=0.0028) and vitamin B12 (p=0.0165). Homocysteine was significantly associated with stroke (p=0.0027) and peripheral arterial vascular disease (p=0.0001), but not with myocardial infarction, angina pectoris, venous thrombosis or cancer. Vitamin B12 was lower in myocardial infarction and higher in diabetes and venous thrombosis compared to the other diseases. Conclusions: The prevalence of deficits in folate and vitamin B12 was paradoxically high in the mountainous northeastern area of Sicily. Abstract: The sarcophagus containing the remains of Federico II, located in the Cathedral of Palermo (Sicily, Italy), was opened on 1998 to perform a multidisciplinary survey [1]. Next to the remains of Federico II and in close contact with them were laying two other skeletons belonging, according to historical records, to Pietro II di Aragona and to an anonymous person (“The Third Individual”), probably a woman. The bones appeared severely deteriorated. Chemical analysis performed on bone samples excluded that the bodies underwent some kind of embalming process. The analysis of mtDNA from bone samples taken from the three skeletons was successful in only one of the two labs involved. The HVR1-mtDNA sequence (region: from nt 16,035 to nt 16,395), obtained from the bone samples of Federico II and “The Third Individual” appear identical but bear double peaks at the same nucleotide positions, suggesting mixing (i.e. contamination) of different mtDNA types. The HVR1 sequence obtained from the bone sample of Pietro II di Aragona does not present double peaks and differ from the Cambridge Reference Sequence (CRS) at six nucleotide positions. Cloning experiment of the Federico II amplicon demonstrated that the mixed mtDNA types are only two: one identical to CRS, the other identical to the sequence of Pietro II di Aragona. A reconstruction of these data are proposed in the Discussion. Due to the problematic context in which this study was carried out (mixed and deteriorated biological material, failure to replicate results in two different labs), our results and reconstruction can only be offered on a tentative basis. It is hoped that the data presented in this study will reveal useful, for future comparison, if further molecular genetics research will be carried out on the royal dynasties that ruled Sicily in the early centuries of the past millennium. Summary In the light of the key role played by interferon (IFN)-γ in the control of tuberculosis, in the present paper we have evaluated the distribution of the functional +874T → A IFN-γ single nucleotide polymorphism (SNP) in Sicilian patients affected by tuberculosis. Our aim was to determine whether there is an association between the TT genotype, which has been suggested to be linked to an increased production of IFN-γ, and resistance to chronic tuberculosis. DNA samples were obtained from 45 patients and 97 healthy controls. Polymorphism at +874 was identified using amplification refractory mutational system methodology. The +874T SNP was less frequent in patients than in controls (0.42 vs. 0.50) but the difference was not significant. The +874TT genotype, which has been suggested to be associated with high IFN-γ production, was significantly decreased in the patients. Thus, resistance to chronic lung tuberculosis might be associated with a genetically determined high IFN-γ production capacity. In conclusion, the present data add another piece of evidence to the complex puzzle of genetic and environmental factors involved in control of infectious diseases. Studies on cytokine gene polymorphisms may elucidate the complex network of trans -interactive genes influencing the type and strength of responses to environmental stressors and may help to identify the genetic factors that affect survival in humans. Abstract: Many Y chromosome short tandem repeats (STRs) have been studied and characterized over the past years. A few Y-STRs multiplex kits have been placed on the market for forensic and population study purposes. The aim of our work was to analyse a sample of Sicilian male individuals to evaluate the allelic frequency and, thus, the possibility to implement a genetic database. An 11 loci Y-STR Typing kit was used to yield haplotype profiles from male DNA; amplification products were detected on ABI PRISM 310 Genetic Analyzer and examined by Genemapper v 3.2 (Applied Biosystems). The population sample subjected to the screening resulted to be very different with regards to certain loci whereas for other loci the allelic profile was less variable. Despite a minor discrimination power within the entire population, Y-STRs represent a valid tool to simplify male/female DNA mixture interpretation which is a major challenge when biological traces are found in case of sexual assault. In the most general terms, assessing tsunami hazard and damage for a given geographic area requires a structured approach involving the identification and the characterization of the potential tsunamigenic sources of interest for that area, the availability of adequate and reliable topographic and bathymetric databases, the quantification of the physical observables related to tsunami impact relevant from the scientific, engineering and civil protection perspectives, a reasonable approach to assess vulnerability and to combine it with the hazard outputs to obtain the damage, and a robust strategy to evaluate the uncertainties to be associated to the final results. When it comes to practical applications, the choice of the methodology that is most appropriate for each specific steps of the procedure above can be strongly dependent on the extension of the area of interest and on the type of tsunamigenic sources involved, not to mention the availability of sufficiently detailed topographies and bathymetries and of appropriate funding/computational resources. A typical example is represented by the computation of high-resolution inundation maps, which can turn out to be a prohibitive task when the geographical scales are regional or even national/transnational.The study presented here is the result of the synergy among three research groups belonging to three Italian institutions that had the possibility to work together in the frame of the recently completed Italian National Project SPOT ("Sismicità Potenzialmente innescabile Offshore e Tsunami"), funded by the Italian Ministry of Economic Development MISE). The main goal of the collaboration was to investigate the possibility of defining and applying a shared approach to the modelling of the propagation and impact of tsunamis generated by earthquakes and landslides over the Italian coasts facing the Adriatic sea, the Ionian sea and the Sicily Channel. The different expertise and know-how of the three research groups allowed to develop a methodology covering all the main steps and approximations required for modelling, with a computationally cheap approach and in the absence of very detailed topo-bathymetric models, tsunami scenarios and their impact on coastal structures in the context of tsunami hazard-to-damage assessment. We briefly comment on the key points of the methodology and then we present some applications related to scenarios of tsunamis generated by earthquakes and landslides and affecting the coasts of the Gulf of Gela (southern Sicily, Italy). The results are provided in terms of maximum water height inland and of damage to buildings and are discussed also in relation to the methodology limitations and to the still open problems. Summary: Objectives: High Toscana virus (TOSV) antibody seropositivity rates have been documented in the last decade, especially in the Mediterranean area. It is unclear if these rates are associated with a recent or past exposure to the virus. This is of importance, as primary infection can cause neurologic complications, especially in adults. The aim of the present study was to assess the current active TOSV circulation in western Sicily. Methods: A cross-sectional seroprevalence study was conducted on 271 individuals aged 4–92 years, sampled from the general population of a small city. Each participant completed a self-administered questionnaire and provided serum, which was analyzed for the presence of specific anti-TOSV IgM and IgG. Results: Anti-TOSV IgM was detected in eight (3.0%) participants, of whom only three had anti-TOSV IgG. The prevalence of anti-TOSV IgM was highest in subjects aged 25–34 and 35–44 years (7.1% and 4.8%, respectively). All subjects positive for anti-TOSV IgM were resident in the suburban area. Conclusions: The detection of IgM documented the circulation of TOSV, a Phlebovirus, in a random population sample of Sicilian adults. The highest risk of TOSV seroconversion in subjects living in the suburbs appears to suggest a high density of TOSV vectors in peri-urban areas. [Copyright &y& Elsevier]
