A Simple, precise, accurate and economical spectrophotometric method was developed and validated for simultaneous estimation of Amiloride (AML) and Hydrochlorothiazide (HCZ) in combined dosage form. In simultaneous equation method, AML and HCZ were quantified using their absorptivity values at selected wavelengths, viz., 244nm and 280nm respectively in 0.1 N NaOH. The linearity range was found to be 5-25 μg/ml for AML and HCZ. The accuracy and reproducibility of the proposed method was statistically validated by recovery studies. The simultaneous equation method permits simple, rapid and direct determination of AML and HCZ in commercially available combined dosage form without previous separations and can therefore be used for routine analysis. The paper reports a recent efforts to develop and validate an efficient and rapid analytical assay method by LC/LCMS for a series of 2H-benzoxazinone based 3-aryl-3,4-dihydro-2H-benz[e]-1,3-oxazines. Such compounds posse's potent pharmacological importance, and are of the class of efavirenz, a non-nucleoside reverse transcriptase inhibitor (NNRTI). Series of seven compounds were separated using ODS , 250×4.6mm, 5 μm column with a mobile phase -A consist of 90:10 (v/v) of 0.05M ammonium acetate buffer with pH 5.60 ± 05, (pH adjusted with acetic acid) and acetonitrile. mobile phase -B consisting 90:10 .05M ammonium acetate buffer with pH 5.60 μ 05, pH adjusted with ortho phosphoric acid with a timed gradient program. Detection was carried out at 245 nm and the flow rate 1.0 ml/min. The method has been validated in terms of suitability, specificity, linearity, intra and interday Precision, limits of detection and quantification The method found to be accurate in the concentration range of 1.0-10.0 μg mL-1 for all compounds. LOD and LOQ were from 0.024 to 0.048 μg mL-1 and 0.075 to 0.147 μg mL-1. Method is linear over concentration range 0.1 to 100 μg mL-1. The proposed method was found to be accurate, precise, specific, linear, rugged, robust, and stability indicating for the determination of a series of 3-aryl-3,4-dihydro-2H-benz[e]-1,3-oxazines and has shown to be convenient for routine analysis of 3-aryl-3,4-dihydro-2H-benz[e]-1,3-oxazine derivatives. A new, simple, and practical aromatic bromination process is disclosed wherein NaBr/NaIO4 combination in acidic medium was efficiently utilized for the first time in the bromination of deactivated aromatic compounds, thus, affording the corresponding bromo-derivatives of deactivated aromatics in good yields and purity. Various industrially-important brominated intermediates have been instantly synthesized using aq. CaBr2–Br2system as an efficient and recyclable brominating reagent under aqueous conditions at room temperature without the need for metal catalysts or acidic additives. Structurally-diverse phenol and aniline derivatives with strong electron-withdrawing groups such as carboxylic, nitro and formyl show remarkable reactivity to the brominating reagent and brominated in 92–98% yield with high purity (>99%) in a very short reaction time. Organic solvent-free conditions, a feature of the green chemistry, were successively used not only for the reactions but also for the isolation of products at the end of the reaction. The recycling of HBr by its neutralization, thereby generating additional amounts of industrially-important CaBr2has been designed and developed. The brominating reagent has been recycled and regenerated, and the process was repeated up to 4 cycles after the fresh batch using the regenerated brominating reagent having almost identical selectivity and isolated yields, which seems to be the most promising methodology from the viewpoint of the green approach to organic synthesis.
