Leafminers are a taxonomically diverse group of endophagous insects. A number of them are pests in forestry, horticulture and agriculture, and some of them have become important invasive species. Here, we discuss the characteristics of invasive leafminers of woody plants. We first present 12 cases of invasive leaf-mining species belonging to four different insect orders. For each of them, we briefly describe their invasion, including pathways of introduction, their impact and management methods and their ecology. We then discuss various aspects of these invasions. Leafminers are introduced to new continents and spread through various pathways such as horticultural trade and accidental transport of adults and pre-imaginal stages in containers and vehicles. They may also spread long distances with air currents. A few species have serious economic impacts as orchard pests, such as the citrus leafminer, Phyllocnistis citrella, or as pests of ornamental plants, such as the horse-chestnut leafminer, Cameraria ohridella. The ecological impact of these species should be better studied, especially those killing native trees, such as the birch leaf-mining weevil, Orchestes fagi, in Canada. Compared to other insect groups, invasive leafminers are usually recruited by a range of native parasitoids, which may or may not succeed in controlling the invasive species. Biological control by introduction of parasitoids from the native range has often been successful to control invasive leafminers. This paper provides reviews of the most commonly used methods to detect plant pests belonging to groups of invasive organisms with high economic relevance, including Coleoptera (bark beetles, flathead borers, leaf beetles, longhorn beetles, weevils), Diptera (cone and seed flies, fruit flies), Homoptera (aphids, leafhoppers and psyllids, whiteflies), Lepidoptera (moths and butterflies), Thysanoptera (thrips), bacteria (potato brown rot Ralstonia solanacearum) and fungi (pitch canker disease Gibberella circinata, brown rot disease Monilinia fructicola). Future perspectives in detection methods are discussed, with particular reference to the considerable increase in the volume, commodity type and origins of trade in plant material from third countries, the introduction of new crops, the continuous expansion of the EU with new border countries being added, and the impact of climate change affecting the geographical boundaries of pests and their vectors. 1 The horse-chestnut leaf miner Cameraria ohridella is an invasive moth in Europe and a serious pest of horse-chestnut Aesculus hippocastanum. The moth also occasionally attacks sycamore maple Acer pseudoplatanus, when situated beside infested horse-chestnuts. 2 The main objective of the present study was to provide an overview of the relationship between C. ohridella and A. pseudoplatanus and to determine whether C. ohridella has the potential to shift to this native tree. 3 In the field, females oviposit on different deciduous tree species. Although less frequently attacked than A. hippocastanum, A. pseudoplatanus was clearly preferred for oviposition over 12 other woody species investigated. 4 Surveys in Europe demonstrated that the majority of A. pseudoplatanus trees found beside infested A. hippocastanum had mines of C. ohridella, even though more than 70% of the larvae died within the first two instars. Attack rates and development success greatly varied from site to site. Attack levels on A. pseudoplatanus were not always correlated with those on A. hippocastanum, and mines on A. pseudoplatanus were sometimes observed beside weakly-infested A. hippocastanum. 5 Field observations, experimental exposure of A. pseudoplatanus saplings and rearing trials in a common garden study showed that individual trees may vary in their susceptibility to C. ohridella, whereas there was no evidence that C. ohridella populations vary in their performance on A. pseudoplatanus. 6 To date, there is little evidence that C. ohridella represents a major risk for A. pseudoplatanus. Non-native pests, climate change, and their interactions are likely to alter relationships between trees and tree-associated organisms with consequences for forest health. To understand and predict such changes, factors structuring tree-associated communities need to be determined. Here, we analysed the data consisting of records of insects and fungi collected from dormant twigs from 155 tree species at 51 botanical gardens or arboreta in 32 countries. Generalized dissimilarity models revealed similar relative importance of studied climatic, host-related and geographic factors on differences in tree-associated communities. Mean annual temperature, phylogenetic distance between hosts and geographic distance between locations were the major drivers of dissimilarities. The increasing importance of high temperatures on differences in studied communities indicate that climate change could affect tree-associated organisms directly and indirectly through host range shifts. Insect and fungal communities were more similar between closely related vs. distant hosts suggesting that host range shifts may facilitate the emergence of new pests. Moreover, dissimilarities among tree-associated communities increased with geographic distance indicating that human-mediated transport may serve as a pathway of the introductions of new pests. Chrysomela tremula is a polyvoltine oligophagous leaf beetle responsible for massive attacks on poplar trees. This beetle is an important model for understanding mechanisms of resistance to Bacillus thuringiensis (Bt) insecticidal toxins, because a resistant C. tremula strain has been found that can survive and reproduce on transgenic poplar trees expressing high levels of the Cry3Aa Bt toxin. Resistance to Cry3Aa in this strain is recessive and is controlled by a single autosomal locus. We used a larval midgut transcriptome for C. tremula to search for candidate resistance genes. We discovered a mutation in an ABC protein, member of the B subfamily homologous to P-glycoprotein, which is genetically linked to Cry3Aa resistance in C. tremula. Cultured insect cells heterologously expressing this ABC protein swell and lyse when incubated with Cry3Aa toxin. In light of previous findings in Lepidoptera implicating A subfamily ABC proteins as receptors for Cry2A toxins and C subfamily proteins as receptors for Cry1A and Cry1C toxins, this result suggests that ABC proteins may be targets of insecticidal three-domain Bt toxins in Coleoptera as well. We provide a comprehensive overview of those Lepidopteran invasions to Europe that result from increasing globalisation and also review expansion of species within Europe. A total of 97 non-native Lepidoptera species (about 1% of the known fauna), in 20 families and 11 superfamilies have established so far in Europe, of which 30 alone are Pyraloidea. In addition, 88 European species in 25 families have expanded their range within Europe and around 23% of these are of Mediterranean or Balkan origin, invading the north and west. Although a number of these alien species have been in Europe for hundreds of years, 74% have established during the 20th century and arrivals are accelerating, with an average of 1.9 alien Lepidoptera newly established per year between 2000-2007. For 78 aliens with a known area of origin, Asia has contributed 28.9%, Africa (including Macaronesian islands, Canaries, Madeira and Azores) 21.6%, North America 16.5%, Australasia 7.2% and the neotropics just 5.2%. The route for almost all aliens to Europe is via importation of plants or plant products. Most alien Lepidoptera established in Europe are also confined to man-made habitats, with 52.5% occuring in parks and gardens. We highlight four species in particular, Diaphania perspectalis, Cacyreus marshalli, Cameraria ohridella and Paysandisia archon, as the most important current economic threats. Longhorned beetles (Cerambycidae) are the most diverse group of predominantly wood‐feeding (xylophagous) insects on Earth. Larvae of most species feed within tissues of plants made up of large amounts of plant cell wall (PCW), which is notoriously difficult to digest. To efficiently access nutrients from their food source, cerambycid larvae have to deconstruct PCW polysaccharides – such as cellulose, hemicelluloses and pectin – requiring them to possess a diversity of plant cell wall degrading enzymes (PCWDEs) in their digestive tract. Genomic data for Cerambycidae are mostly limited to notorious forest pests and are lacking for most of the taxonomic groups. Consequently, our understanding of the distribution and evolution of cerambycid PCWDEs is quite limited. We addressed the numbers, kinds and evolution of cerambycid PCWDEs by surveying larval midgut transcriptomes from 23 species representing six of the eight recognized subfamilies of Cerambycidae and each with very diverse host types (i.e., gymnosperms, angiosperms, xylem, phloem, fresh or dead plant tissues). Using these data, we identified 340 new putative PCWDEs belonging to ten carbohydrate active enzyme families, including two gene families (GH7 and GH53) not previously reported from insects. The remarkably wide range of PCWDEs expressed by Cerambycidae should allow them to break down most PCW polysaccharides. Moreover, the observed distribution of PCWDEs encoded in cerambycid genomes agreed more with phylogenetic relationship of the species studied than with the taxonomic origin or quality of the host plant tissues. The number of invasive alien pest and pathogen species affecting ecosystem functioning, human health and economies has increased dramatically over the last decades. Discoveries of invasive pests and pathogens previously unknown to science or with unknown host associations yet damaging on novel hosts highlights the necessity of developing novel tools to predict their appearance in hitherto naïve environments. The use of sentinel plant systems is a promising tool to improve the detection of pests and pathogens before introduction and to provide valuable information for the development of preventative measures to minimize economic or environmental impacts. Though sentinel plantings have been established and studied during the last decade, there still remains a great need for guidance on which tools and protocols to put into practice in order to make assessments accurate and reliable. The sampling and diagnostic protocols chosen should enable as much information as possible about potential damaging agents and species identification. Consistency and comparison of results are based on the adoption of common procedures for sampling design and sample processing. In this paper, we suggest harmonized procedures that should be used in sentinel planting surveys for effective sampling and identification of potential pests and pathogens. We also review the benefits and limitations of various diagnostic methods for early detection in sentinel systems, and the feasibility of the results obtained supporting National Plant Protection Organizations in pest and commodity risk analysis. The Loire River is one of the last European large rivers with important sediment dynamics and numerous sandbanks. The extraction of sediment from the riverbed during decades and the construction of levees for flood prevention have strongly affected and shaped the biodiversity of the Loire River., Many species from pioneer riverbanks have been impacted with particular consequences for psammophilous insects. The ground beetle Harpalus (Acardystus) flavescens (Piller & Mitterpacher, 1783), is considered to have disappeared from the Middle Loire River for 40 years and is endangered everywhere in Europe., In 2012 and 2013, we recorded two specimens of H. flavescens in Région Centre-Val de Loire (France), in the course of a survey dedicated to evaluating the impact of fluvial maintenance operations upon sediment and biodiversity dynamics., The presence of H. flavescens may be linked to the interruption of riverbed extractions and the vegetation removal of sandbanks of the Loire River (ecosystem restoration). Knowing the phylogeographic structure of invasive species is important for understanding the underlying processes of invasion. The micromoth Phyllonorycter issikii, whose larvae damage leaves of lime trees Tilia spp., was only known from East Asia. In the last three decades, it has been recorded in most of Europe, Western Russia and Siberia. We used the mitochondrial cytochrome c oxidase subunit I (COI) gene region to compare the genetic variability of P. issikii populations between these different regions. Additionally, we sequenced two nuclear genes (28S rRNA and Histone 3) and run morphometric analysis of male genitalia to probe for the existence of cryptic species. The analysis of COI data of 377 insect specimens collected in 16 countries across the Palearctic revealed the presence of two different lineages: P. issikii and a putative new cryptic Phyllonorycter species distributed in the Russian Far East and Japan. In P. issikii, we identified 31 haplotypes among which 23 were detected in the invaded area (Europe) and 10 were found in its putative native range in East Asia (Russian Far East, Japan, South Korea and China), with only two common haplotypes. The high number of haplotypes found in the invaded area suggest a possible scenario of multiple introductions. One haplotype H1 was dominant (119 individuals, 67.2%), not only throughout its expanding range in Europe and Siberia but, intriguingly, also in 96% of individuals originating from Japan. We detected eight unique haplotypes of P. issikii in East Asia. Five of them were exclusively found in the Russian Far East representing 95% of individuals from that area. The putative new cryptic Phyllonorycter species showed differences from P. issikii for the three studied genes. However, both species are morphologically undistinguishable. They occur in sympatry on the same host plants in Japan (Sendai) and the Russian Far East (Primorsky krai) without evidence of admixture. [ABSTRACT FROM AUTHOR]

Abstract: The insect midgut is the primary target site for Bt-derived insecticides and Bt alternatives. However, despite extensive recent study, the precise role and nature of different Bt receptors remains a subject of considerable debate. This problem is fuelled by a lack of understanding of the genes expressed in the insect midgut and their physiological roles. The poplar leaf beetle, Chrysomela tremulae, is an important model for understanding the mode of action of, and resistance to, coleopteran-specific Bt toxins and currently shows the only known naturally occurring case of resistance to Cry3A toxins. Moreover it belongs to the same family as the corn rootworm, Diabrotica virgifera, an economically important beetle pest and target of hybrid corn expressing Cry3 toxins. Pyrosequencing is a fast and efficient way of defining the transcriptome of specific insect tissues such as the larval midgut. Here we use 454 based pyrosequencing to sample the larval midgut transcriptome of C. tremulae. We identify candidate genes of putative Bt receptors including transcripts encoding cadherin-like proteins, aminopeptidase N and alkaline phosphatase. We also describe a wealth of new transcripts predicting rapidly evolving gene families involved in plant tissue digestion, which have no homologs in the genome of the stored product pest the Red Flour beetle, Tribolium castaneum. A set of six polymorphic microsatellite markers was developed for the highly invasive horse chestnut leaf-mining moth, Cameraria ohridella. For 96 positive clones, six polymorphic microsatellites with conserved flanking regions were identified. The average number of alleles per locus was eight (range from three to 12). Two of these loci showed significant heterozygosity deficits due to null alleles. The remaining four loci did not depart from Hardy-Weinberg equilibrium. In addition, all six loci were amplified for 20 other gracillariid species belonging to eight different genera, including another invasive species Phyllonorycter platani. Hybrid poplars (Populus tremula × Populus tremuloides) have been genetically engineered via Agrobacterium tumefaciens, to express a synthetic cry3Aa gene derived from the native Bacillus thuringiensis subsp. tenebrionis cry3Aa gene. The presence and the expression of the transgene have been verified in four transgenic poplar lines, using Southern, northern and western analyses. The transgenic poplar's toxicity towards the phytophagous beetle Chrysomela tremulae (Coleoptera, Chrysomelidae) has been assessed on six month-old greenhouse-grown selected plants in laboratory conditions. Laboratory experiments consisted of feeding tests of fresh detached leaves on C. tremulae at all developmental stages. Our results indicate that the transgenic poplar leaves, expressing a Cry3Aa protein amount in a range of 0.05–0.0025% of total soluble protein, were definitely deleterious for C. tremulae, regardless of the developmental stage. During the last two centuries, thousands of insect species have been transported (largely inadvertently) and established outside of their native ranges worldwide, some with catastrophic ecological and economic impacts. Global variation in numbers of invading species depends on geographic variation in propagule pressure and heterogeneity of environmental resistance to invasions. Elton’s diversity-invasibility hypothesis, proposed over sixty years ago, has been widely explored for plants but little is known on how biodiversity affects insect invasions. Here we use species inventories from 44 land areas, ranging from small oceanic islands to entire continents in various world regions, to show that numbers of established insect species are primarily driven by diversity of plants, with both native and non-native plant species richness being the strongest predictor of insect invasions. We find that at large spatial scales, plant diversity directly explains variation in non-native insect species richness among world regions, while geographic factors such as land area, climate and insularity largely affect insect invasions indirectly via their effects on local plant richness.
