Green synthesis is an easy, safe, and environmentally beneficial nanoparticle creation method. It is a great challenge to simultaneously improve the capping and stabilizing agent carrier separation efficiency of photocatalysts. Herein, Zn-doped Titanium dioxide (TiO2) nanoparticles with high exposure of 360 nm using a UV/visible spectrophotometer were prepared via a one-step hydrothermal decomposition method. A detailed analysis reveals that the electronic structures were modulated by Zn doping; thus, the responsive wavelength was extended to 600 nm, which effectively improved the visible light absorption of TiO2. We have optimized the different parameters like concentration, time, and temperature. The peak for TiO2 is located at 600 cm-1 in FTIR. A scanning electron microscope revealed that TiO2 has a definite shape and morphology. The synthesized Zn-doped TiO2NPs were applied against various pathogens to study their anti-bacterial potentials. The anti-bacterial activity of Zn-doped TiO2 has shown robust against two gram-ve bacteria (Salmonella and Escherichia coli) and two gram + ve bacteria (Staphylococcus epidermidis and Staphylococcus aureus). Synthesized Zn-doped TiO2 has demonstrated strong antifungal efficacy against a variety of fungi. Moreover, doping TiO2 nanoparticles with metal oxide greatly improves their characteristics; as a result, doped metal oxide nanoparticles perform better than doped and un-doped metal oxide nanoparticles. Plants are subjected to various biotic and abiotic stresses that significantly impact their growth and productivity. To achieve balanced crop growth and yield, including for leafy vegetables, the continuous application of micronutrient is crucial. This study investigates the effects of different concentrations of copper sulphate (0, 75, 125, and 175 ppm) on the morphological and biochemical features of Spinacia oleracea and Avena sativa. Morphological parameters such as plant height, leaf area, root length, and fresh and dry weights were optimized at a concentration of 75 ppm copper sulfate. At this concentration, chlorophyll a & b levels increased significantly in Spinacia oleracea (462.9 and 249.8 휇푔/푔), and Avena sativa (404.7 and 437.63휇푔/푔). However, carotenoid content and sugar levels in Spinacia oleracea were negatively affected, while sugar content in Avena sativa increased at 125 ppm (941.6 µg/ml). Protein content increased in Spinacia oleracea (75 ppm, 180.3 µg/ml) but decreased in Avena sativa. Phenol content peaked in both plants at 75 ppm (362.2 and 244.5 µg/ml). Higher concentrations (175 ppm) of copper sulfate reduced plant productivity and health. Plants exposed to control and optimal concentrations (75 and 125 ppm) of copper sulpate exhibited the best health and growth compared to those subjected to higher concentrations. Maximum plant height, leaf area, root length, fresh and dry weights were observed at lower concentrations (75 and 125 ppm) of copper sulfate, while higher concentrations caused toxicity. Optimal copper sulfate levels enhanced chlorophyll a, chlorophyll b, total chlorophyll, protein, and phenol contents but inhibited sugar and carotenoid contents in both Spinacia oleracea and Avena sativa. Edible mushrooms are an important food source with high nutritional and medicinal value. They are a useful source for studying phylogenetic evolution and species divergence. The exploration of the evolutionary relationships among these species conventionally involves analyzing sequence variations within their complete mitochondrial genomes, which range from 31,854 bp (Cordyceps militaris) to 197,486 bp (Grifolia frondosa). The study of the complete mitochondrial genomes of edible mushrooms has emerged as a critical field of research, providing important insights into fungal genetic makeup, evolution, and phylogenetic relationships. This review explores the mitochondrial genome structures of various edible mushroom species, highlighting their unique features and evolutionary adaptations. By analyzing these genomes, robust phylogenetic frameworks are constructed to elucidate mushrooms lineage relationships. Furthermore, the exploration of different variations of mitochondrial DNA presents novel opportunities for enhancing mushroom cultivation biotechnology and medicinal applications. The mitochondrial genomic features are essential for improving agricultural practices and ensuring food security through improved crop productivity, disease resistance, and nutritional qualities. The current knowledge about the mitochondrial genomes of edible mushrooms is summarized in this review, emphasising their significance in both scientific research and practical applications in bioinformatics and medicine. The pollution of aquatic environments by industrial wastewater is one of the major environmental problems in the world. Due to factors like simplicity in application, cost-effective, rapid and sensitive response to pollutants, algae can be used as early indicator of pollution in aquatic environments. Euglena gracilis has been widely used as a model organism in assessment of various water pollutants with its different parameters as endpoints. Wastewater has been frequently reported to adversely affect aquatic organisms, but its effect on photosynthesis of phytoplankton, particularly in long-term exposure, has been addressed rarely. In the present study, long-term effects of wastewater samples from ten industries on photosynthesis of E. gracilis were assessed using a pulse-amplitude-modulated (PAM) fluorometer. Euglena cultures were exposed to different concentrations of wastewaters, and the effect on quantum yield of photosystem II ( F/ F), and relative electron transport rate (rETR) was assayed after 7 days of exposure. Five samples out of the total ten were found to adversely affect photosynthesis in Euglena. One sample was found to have a stimulatory effect on photosynthesis in Euglena. Unfiltered solutions were observed to affect rETR more severely than filtered ones of the same sample at the same concentration. By comparing the results with previous reports of short-term effects of wastewater on photosynthesis, it is concluded that wastewater can be a risk to photosynthesis in aquatic organisms in long-term exposure. Wastewater is a big source of water pollution in the world. Selected wastewater samples from Dera Ismail Khan (D.I. Khan), a city in Pakistan, were analyzed for physicochemical and ecotoxicological properties. Samples from the Indus River before and after the municipal wastewater is discharged into it were tested to determine the effects of municipal wastewaters from D.I. Khan on the river Indus. Different parameters of the freshwater flagellate Euglena gracilis such as motility, swimming velocity, gravitactic orientation, cell shape, and photosynthetic efficiency were used as end points. Gravitactic orientation and cell shape were found to be significantly impaired by the wastewater samples. Swimming velocity and relative electron transport rate of cells were positively affected by all water samples. Quantum yield of photosystem II ( F/ F) was not affected, except by ghee (oil) industry effluent where it was significantly increased. Comparison of upstream and downstream samples from the Indus River showed that waste effluents from D.I. Khan affect the physicochemical and ecological properties of the river Indus. Background: The Indigenous knowledge of plants is scientifically and culturally very significant. This paper elucidates the empirical findings of an ethnobotanical survey of Banda Daud Shah, District Karak, Pakistan. Methods: Data collection was carried out from October 2011 to September 2012. Total twelve survey trips were made, three in each season. About 100 respondents were interviewed; most of them were aged people between 60-70 years. Interviews were conducted using structured questionnaire composed of variety of questions regarding ethnomedicinal uses of plants of the study area. Direct matrix ranking (DMR), informant citations and market survey of multipurpose plants were also carried out. Results: The local community was using 58 plant species belonging to 52 genera and 34 families for different purposes. A total of 25 plant species were herbs followed by 18 shrubs. Leaf (45%) was the most commonly used plant part followed by the whole plants (23%). In total, 40 plant species were medicinally used to treat variety of diseases, of which highest number of species being used for gastro-intestinal problems (19 spp.), expectorant (3 spp.) and antipyretic (3 spp.). Beside medicinal values, 25 species were used for fuel and 18 for fodder purposes. Informant consensus showed that gastrointestinal and respiratory infections were ranked highest (FIC = 0.75) among all ailments. According to DMR output, Dalbergia sisso ranked first due to high multipurpose uses among all species and was found most threatened with higher market value. Conclusion: The investigated area is rural in nature and the inhabitants are highly dependent on the native plants for their health care needs and other requirements like fuel wood and fodder due to financial constraints and unavailability of resources. Medicinal plants for high ranked diseases may be phtyochemicaly and pharmacologically investigated to prove their efficacy. The local medicinal flora is facing overexploitation, overgrazing and improper way of collection. Proper conservation strategies such as controlled grazing, reforestation and rangeland management among many others may be adopted to promote the sustainable use of medicinal plants. Due to the presence of various potential pollutants industrial wastewaters pose considerable threats to natural waters and make it unfit for the aquatic biota. Analysis of wastewaters by chemical methods is a common practice; however, it does not reflect the toxic effects on living organism. Therefore, bioassessment is necessary for monitoring of wastewater quality. In the present study, the toxic effects of wastewater samples from different industries were evaluated using Euglena gracilis as a biotest organism. Various parameters of the freshwater flagellate E. gracilis like motility, swimming velocity, cell shape, gravitactic orientation (using the automatic biotest ECOTOX) and photosynthetic efficiency (using a pulse amplitude modulated fluorometer) were used as end points for toxicity assessment. In addition, the samples were analysed for some ecologically important physicochemical properties. With some exceptions, most of the physicochemical properties of the tested samples were within the acceptable range of national environmental quality standards for municipal and industrial effluents. However, all the water samples adversely affected different parameters in E. gracilis. This study led to the conclusion that different toxic substances present in wastewater, even at low concentrations, can be a possible threat to aquatic biota. The results of this study prove that ECOTOX is a sensitive, easy, and fast bioassay for monitoring of water and wastewater quality. Diammonium phosphate (DAP) and urea are commonly used fertilizers throughout the world. The effects of these fertilizers on the freshwater flagellate Euglena gracilis was studied after 7 days of growth using morphological, physiological and biochemical parameters as end points. NOEC and EC values for various parameters like cell density, motility, velocity, cell shape, gravitaxis, chlorophyll a, b and total carotenoids were calculated. NOEC and EC values of DAP varied from 0.5 to 2.5 g L and 3.14 to 5.96 g L, respectively, for different parameters. NOEC and EC values for urea ranged from 5 to 25 g L and 28 to 44.05 g L, respectively, for various parameters. Photosynthetic pigments were found to be more sensitive to both fertilizers as compared to other measured end points. The NOEC and EC values obtained for DAP were much lower than those for urea; i.e., DAP showed a stronger inhibitory effect as compared to urea. Application of DAP resulted in an increased concentration of ammonia in Euglena cultures but urea did not. The stronger inhibitory effect of DAP is attributed to release of free ammonia in the culture due to DAP decompostion. Abstract: The effect of two commonly used fertilizers, DAP (diammonium phosphate) and urea was studied on the freshwater flagellate Euglena gracilis using the automatic biotest ECOTOX. NOEC and EC50 values for various parameters like motility, velocity, cell shape and gravitaxis were calculated. The NOEC and EC50 values obtained for DAP were much lower than those for urea; i.e. DAP showed a stronger inhibitory effect as compared to urea. The inhibition caused by DAP increased with increasing exposure time over 24h but urea showed no augmentation with increasing exposure time. Application of DAP resulted in an increased pH and high concentrations of ammonia but urea did neither affect the pH nor affect the ammonia concentration. Recovery experiments in deionized water after urea application showed a reconstitution of motility after 72h. After an application of 1.35gL−1 (24h EC50 for motility) DAP motility recovered after 72h but motility did not recover when the concentration was doubled (2.7gL−1). The EC50 values obtained were compared with the EC50/LC50 values reported for other aquatic organisms and were found to be comparable with the reported values. [Copyright &y& Elsevier]

Abstract: Synthetic detergents are among the commonly used chemicals in everyday life. Detergents, reaching aquatic environments through domestic and municipal wastewater, can cause many different effects in aquatic organisms. The present study was aimed at the toxicity evaluation of a commonly used laundry detergent, Ariel, using the freshwater flagellate Euglena gracilis as a biotest organism. Different parameters of the flagellate like motility, swimming velocity, cell shape, gravitactic orientation, photosynthesis and concentration of light harvesting pigments were used as end points for the toxicity assessment. No Observed Effect Concentration (NOEC) and EC50 values were calculated for the end point parameters at four different incubation times, i.e. 0, 6, 24 and 72h. After 72h incubation, swimming velocity of the cells was found to be the most sensitive parameter giving NOEC and EC50 values of 10.8 and 34mgL−1, respectively. After 72h exposure to the detergent, chlorophyll a and total carotenoids were significantly decreased in cultures treated with Ariel at concentrations of 50mgL−1 and above while chlorophyll b significantly decreased at concentrations above 750mgL−1. The maximum inhibitory effect on the quantum yield of photosystem II was observed after 24h exposure and thereafter a recovery trend was observed. Motility, gravitaxis and cell shape were strongly impaired immediately upon exposure to the detergent, but with increasing exposure time these parameters showed acclimatization to the stress and thus the NOEC values obtained after 72h were higher than those immediately after exposure. Abstract: Water pollution is one of the major threats to public health in Pakistan. Drinking water quality is poorly managed and monitored. Pakistan ranks at number 80 among 122 nations regarding drinking water quality. Drinking water sources, both surface and groundwater are contaminated with coliforms, toxic metals and pesticides throughout the country. Various drinking water quality parameters set by WHO are frequently violated. Human activities like improper disposal of municipal and industrial effluents and indiscriminate applications of agrochemicals in agriculture are the main factors contributing to the deterioration of water quality. Microbial and chemical pollutants are the main factors responsible exclusively or in combination for various public health problems. This review discusses a detailed layout of drinking water quality in Pakistan with special emphasis on major pollutants, sources of pollution and the consequent health problems. The data presented in this review are extracted from various studies published in national and international journals. This study aims to examine the heavy metal phytofiltration potential of Hydrocotyle umbellata from Nullah Lai wastewater and its environmental risk. Wastewater was initially analyzed for heavy metals and physicochemical parameters and compared with irrigation water standards. The phytofiltration potential of H. umbellata was determined by periodically monitoring heavy metal concentrations at time points T1 (day 05), T2 (day10), T3 (day 15), and T4 (day 20). This study shows that some water parameters reached the permissible limits after treatment with H. umbellata. Results of the risk analysis reveal a significant discharge (kg d−y) of heavy metals to arable land. Phytofiltration efficiency of H. umbellata for water quality parameters was in order of Ni (98.75%), Cr (98.11%), Cd (95.84%), Pb (94.90%), Cu (94.10%), Zn (85.34%), BOD (53.67%), TDS (29.28%), EC (27.31%), Cl (6.65%), and SAR (6.34%). The growth of H. umbellata in wastewater resulted in heavy metal bioaccumulation in both roots and shoots of the plant. Less than 01 translocation factor (TF) values for cadmium, copper, lead, chromium, and zinc showed the metal tolerance ability of the tested plant. These results demonstrated that treated water could be used in water-deficient peri-urban areas as supplementary irrigation. Cancer is the leading cause of mortality in the world. The major therapies for cancer treatment are chemotherapy, surgery, and radiation therapy. All these therapies expensive, toxic and show resistance. The plant‐derived compounds are considered safe, cost‐effective and target cancer through different pathways. In these pathways include oxidative stress, mitochondrial dependent and independent, STAT3, NF‐kB, MAPKs, cell cycle, and autophagy pathways. One of the new plants derived compounds is Polyphyllin VII (PPVII), which target cancer through different molecular mechanisms. Nanostructure gold nanoparticles (Au NPs) are well-known biological active materials, synthesised under different environment-friendly approaches that has gained significant interest in the field of biomedicine. This study investigated a novel, fast, easy, cost-effective and the eco-friendly method to synthesise Au NPs from mediated Viscum album Linn plant extract, where the plant metabolites act as stabilising and reducing agents. The synthesised Au NPs were analysed by UV/Vis spectroscopy that gave strong signals and a sharp absorption peak at 545 nm due to the presence of surface plasmon resonance (SPR) bands. In addition, energy dispersive X-ray spectroscopy (EDX) showed that strong signals of Au NPs appeared at 9.7 and 2.3 keV, as the rays of light passed. X-ray diffraction recognised the crystalline material and provided information on the cell unit that the synthesised Au NPs are face-centreed cubic in structure. The diffraction of X-ray spectra showed intense peaks at 38.44°, 44.7°, 44.9° and 77.8°. The mediated V. album plant extracts and synthesised Au NPs were screened against gram-positive and gram-negative (Enterobacter , Salmonella typhi , Escheria coli and Bacillus subtilis) bacterial strains, confirming their antibacterial potential. Au NPs showed strong antibacterial activity due to its unique steric configuration. Au NPs damaged bacterial cell membrane leading to the leakage of the cytoplasm and death of the cell. Wastewater not only provides nutrients to the plant community but may cause abiotic stress conditions due to the presence of toxic substances. The present study was designed to explore wastewater-induced changes in maize seedlings using four different concentrations (0, 10, 50, and 100%) collected from three different sources, i.e., municipal wastewater (designated as MW), woolen mill wastewater (designated as WW) and polyvinylchloride wastewater (designated as PW) of the city of Bannu situated in Khyber Pakhtunkhwa, Pakistan. All physiological parameters such as germination, biomass, and length of maize seedlings gradually slowed down with the increase in wastewater concentration. Similarly, the photosynthetic pigments decreased, while an increase was calculated in Na+ and Ca++ and K+ ions in our present experiment. The malondialdehyde (MDA) and hydrogen peroxide (H2O2) contents as well as antioxidative enzymes such superoxide dismutase (SOD), peroxidase (POD), ascorbate peroxidase (APX), and catalase (CAT) upregulated in all treated samples, while total soluble proteins increased. Various wastewater sources also caused genotoxic effects as revealed by the appearance and disappearance of various bands at DNA and protein levels. The present study reveals that different sources of wastewater caused differential stresses in maize seedlings, which might be due to the presence of different stressful agents in them. The present study was conducted to investigate the effect of sub-lethal doses of Cypermethrin (CYP) on the survival and morphological and biochemical aspects of rohu ( Labeo rohita ) during early developmental stages. Newly fertilized eggs were incubated in the presence of sub-lethal concentration of CYP (20% of LC 50 i.e., 8.43 μg L −1 ) in a well designed flow through system. Increased mortality was found with increase in exposure time. Deformities like eroded yolk and margins, elongated yolk sac, larvae with short tail, no eyes or larvae with zigzag movement and paralyzed larvae were observed in CYP treated group from blastula to fry stage. The activities of antioxidant like Catalase (CAT), Peroxidase (POD) and Lactoperoxidase (LPO) were decreased from 24 to 96 h in the control group whereas increased in CYP treated group. The Glutathione reducatse (GR) activity was also increased with time in both the control and treated groups; however, the activity was significantly higher in CYP treated group. Similarly, the whole body cortisol level showed an increasing trend with time in both control and treated groups. However, in CYP exposed group the cortisol level was considerably higher after 24 h exposure but statistically comparable to the control group after 96 h. It can be concluded from the present results that CYP even at very low concentration adversely affects the early development of L. rohita and enhances mortality. The dramatic increase in healthcare costs has become a significant burden to this era. Many patients are unable to access medication because of the high price of drugs. Genetic engineering has made advances to increase the yield, titer, and productivity in the bio-based production of chemicals, materials of interest, and identification of innovative targets for drug discovery. Currently, the production of homoglutamate (α-Aminoadipic acid) involves petrochemical routes that are costly with low yield and often not suitable for industrial production. Here, we established the development of NADH-dependent homoglutamate by engineering NADH-dependent phenylalanine dehydrogenase (PDH) from Thermoactinomyces intermedius, which provides a novel tool for in-vivo metabolic engineering and in-vitro catalysis. Based on computational insight into the structure, we proposed the site-specific directed mutagenesis of the two important residues of PDH through docking simulations by AutoDock Vina which elucidated the binding mode of PDH with α-Ketoadipic acid and ligands. Our results demonstrated that the catalytic efficiency Km/Kcat of the final mutant Ala135Arg showed a 3-fold increase amination activity towards the ketoadipic acid as compared to the other mutant Gly114Arg, a double mutant Gly114Arg/Ala135Arg, and wild type TiPDH. Furthermore, we have introduced formate dehydrogenase as a cofactor regenerative system in this study which further made this study economically viable. Pinus is one of the most economical and ecological important conifers, model specie for studying sequence divergence and molecular phylogeney of gymnosperms. The less availability of information for genome resources enable researchers to conduct evolutionary studies of Pinus species. To improve understanding, we firstly reported, previously released chloroplast genome of 72 Pinus species, the sequence variations, phylogenetic relationships and genome divergence among Pinus species. The results displayed 7 divergent hotspot regions (trnD-GUC, trnY-GUA, trnH-GUG, ycf1, trnL-CAA, trnK-UUU and trnV-GAC) in studied Pinus species, which holds potential to utilized as molecular genetic markers for future phylogenetic studies in Pinnus species. In addition, 3 types of repeats (tandem, palindromic and dispersed) were also studied in Pinus species under investigation. The outcome showed P. nelsonii had the highest, 76 numbers of repeats, while P. sabiniana had the lowest, 13 13 numbers of repeats. It was also observed, constructed phylogenetic tree displayed division into two significant diverged clades: single needle (soft pine) and double-needle (hard pine). Theoutcome of present investigation, based on the whole chloroplast genomes provided novel insights into the molecular based phylogeny of the genus Pinus which holds potential for its utilization in future studies focusing genetic diversity in Pinnus species. In this research, eight local mung bean (Vigna radiata) varieties were analyzed for their performance against two levels of CdCl2 solution (0.3 and 0.5 mM) alone and priming with gibberellic acid (GA3) (100 μM), salicylic acid (SA) (50 μM) and proline (5 mM) solution prior to Cd exposure. Mung bean seedlings were analyzed for disturbance in cytological, morphological, biochemical and enzymatic parameters under cadmium stress. For cytological studies, 48 h grown mung bean seedlings root tips were used to prepare slides and studied for percent mitotic index (MI%) and to calculate percent C-mitosis, laggard, sticky and fragmented chromosomes, pictures were captured by a Nikon camera (DS-Fi 1 Japan) attached with a microscope. One-week grown mung seedlings were studied for growth traits, malondialdehyde (MDA), protein, proline and antioxidant enzymes. ANOVA and DMR test of this research revealed that all the tested mung bean varieties and treatments were significantly different regarding mitotic index and number of chromosomal aberrations. Both the Cd treatments exhibited increased total chromosomal aberrations with different types and a maximum decrease in MI%. In pretreated samples, GA3, SA and proline serve as mitigating agents that reduce mutagenic effects of Cd in mung bean by increasing MI% and decreasing chromosomal aberrations as compared to non-pretreated samples. Both the Cd treatments showed a decrease in all growth traits. Total proteins were also found to be significantly reduced in a dose-dependent manner in all genotypes. Cd treatment increased the activities of all antioxidant enzymes tested. Cd caused oxidative damage as indicated by elevated levels of MDA content in treated samples in comparison to control. Proline content levels were also high in Cd treated seedlings indicating stress. Results demonstrated that pretreatment with phytohormones and proline before Cd were found to improve all morphological parameters, by altering antioxidant enzymes activities along with a decrease in MDA and proline contents as well. It was further noticed that the performance of GA3 was better at 0.3 mM Cd treatment while SA was found to be a good mitigating agent at 0.5 mM Cd stress in all tested mung bean varieties. This research concluded less deleterious effects of Cd on AZRI-2006 while more sensitivity to NM-51 towards Cd. Priming with phytohormones and proline is a user-friendly, economical, and simple mitigation strategy to reduce Cd toxicity in plants and get better yield from contaminated lands. Background & aim: Hepatitis B and C infections are global issues that are associated with a massive financial burden in developing countries where vertical transmission is the major mode and remains high. This cross-sectional study was designed to investigate the seroepidemiology and associated risk factors of hepatitis B virus (HBV) and hepatitis C virus (HCV) infections among 375 pregnant women attending antenatal care health facilities at Bacha Khan Medical Complex (BKMC) Shahmansoor and District Head Quarter (DHQ) Hospital Swabi, Khyber Pakhtunkhwa, Pakistan. Methodology: From a total of 375 pregnant women selected using systematic random sampling from both hospitals, 10 ml of blood samples were collected and alienated serum was examined for indicators identification through the Immuno-Chromatographic Test (ICT) and 3rd Generation Enzyme-Linked Immunosorbent Assay (ELISA). A pre-structured questionnaire was used to collect the socio-demographic data and possible risk factors. The data was analyzed via SPSS 23.0 statistical software. A chi-square analysis was performed to determine the association between variables. P-value < 0.05 was set statistically significant. Results: The overall frequency of HBV and HCV among 375 pregnant women involved in the study was 3.7% and 2.1% respectively. None of the pregnant women were co-infected with HBV and HCV. Dental extraction (P = 0.001) and blood transfusion (P = 0.0005) were significantly allied with HBV infection while surgical procedure (P = 0.0001) was significantly associated with HCV infection. Moreover the sociodemographic characteristics: residential status (P = 0.017) and educational level (P = 0.048) were found significant risk factors of HBsAg and maternal age (P = 0.033) of anti-HCV, respectively. Conclusion & recommendation: HBV and HCV infections are intermediary endemic in the study area. A higher prevalence of HBV was detected among pregnant mothers with a history of dental extraction, history of blood transfusion, resident to the urban area and low educational level. The age and surgical procedures were the potential risk factors found significantly associated with HCV positivity among pregnant mothers in our setup. Future negotiations to control vertical transmission should include routine antenatal screening for these infections early in pregnancy and the requirement of efficient preventive tools including the birth dose of the hepatitis B vaccine in combination with hepatitis B immune globulins to the neonate. Health assumptions to the population due to the utilization of contaminated vegetables have been a great concern all over the world. In this study, an investigation has been conducted to ascertain metal concentrations in the wastewater, soil and commonly consumed vegetables from the vicinity of Gadoon Industrial Estate Swabi, Khyber Pakhtunkhwa Pakistan. Physicochemical parameters such as pH, electrical conductivity (EC), total dissolved solids (TDS), total suspended solids (TSS) and total solids (TS) and heavy metals such as Pb, Cr, Cd, Ni, Zn, Cu, Fe, Mn were determined using Atomic Absorption Spectrophotometer (AAS). Moreover, possible health risks due to the consumption of vegetables have also been estimated. pH and TSS in wastewater were found to be higher than the permissible limit set by WHO (1996). These results revealed that Cr concentration in the wastewater was above the permissible limits of United States Environmental Protection Agency (USEPA) which may lead to a detrimental effect on soil quality deterioration, ultimately leading to food contamination. ANOVA analysis demonstrated a significant difference in soil samples for Pb, Cr, Cd, Ni, Zn and Cu at p ≤ 0.001, for Mn at p ≤ 0.05 while no significant difference was observed for Fe respectively. ANOVA analysis also exhibited the highest mean value for Pb, Cr, Cd and Zn in vegetables. A substantial positive correlation was found among the soil and vegetable contamination. The transfer factor for Cr, Pb, Zn, Mn, Ni, Cd and Cu was greater than 0.5 due to contamination caused by domestic discharges and industrial effluents. Health assessment via consumption of dietary vegetables revealed a higher level than the permissible limit (HRI > 1) for Pb and Cd in children and adults. Enrichment factor (EF) due to consumption of vegetables was found higher for Pb and Cr respectively. Based on the findings of this study, there would be a significant risk to the consumers associated with consumptions of vegetables being cultivated in Gadoon Industrial Estate area of district Swabi. Pesticides are usually used to eliminate weeds and insects to improve crop quality and yield. The present study was undertaken to explore pesticides (Lambda cyhalotherin (LC) and Emamectin benzoate (EB)) related physiological, biochemical and ultrastructural changes in leaves of maize seedlings at different concentrations of LC and EB both singly (LC100, LC500, LC800, EB100, EB300, EB600 mg/L) and jointly (LC500 + EB300 mg/L) along with control. Germination percentage, root stem and leaf lengths increased at lower concentrations of both pesticides and significantly decreased with the increase in the external application of pesticides. At higher concentrations of LC and EB (i.e., LC800 and EB600 mg/L) the mean values of growth and biomass of maize leaves were lower than control. Also, decreases in photosynthetic pigments and ion concentration of Na1+, Ca2+ and K1+ were significant when the concentration of both pesticides increased. The melondialdehyde (MDA) contents decreased, while the amount of hydrogen peroxide (H2O2) production increased at higher doses in cases of single and joint applications. With the increase in their concentrations, the activities of antioxidant enzymes such as superoxide dismutase (SOD), peroxidase (POD), and catalase (CAT) were found to be pronouncedly enhanced as compared to control. Ultrastructural alterations in mesophyll cells of maize leaf were mostly found in chloroplast and nucleus. The present study revealed that short-term exposure of maize seedlings to pesticides (LC and EB) caused less stressful effects on maize due to the presence of active antioxidative metabolism, which indicates the resistant nature of maize toward these pesticides. Arsenic (As) is a serious pollutant of water bodies. Its presence in water can cause severe health problems in humans and may also induce serious toxicological effects in aquatic organisms. Euglena gracilis, a unicellular freshwater flagellate, is considered very sensitive to environmental pollutants. The aim of the present study was to evaluate the responses of different parameters in E. gracilis toward arsenite (As-III) toxicity and to assess As-III removal potential of E. gracilis from water by calculating bio-concentration factor (BCF). Under the optimized experimental conditions (0.1 to 10 mg L−1 of As-III, (pH 6.8) 7-day exposure), various parameters of E. gracilis like cell growth, motility, cell velocity (speed), cell shape, gravitactic orientation, biochemical parameters, and oxidative-stress markers were measured as endpoints. The experimental results showed that cell growth and photosynthetic pigments (chlorophyll a, b, and total carotenoids) were significantly affected at higher concentrations of As-III, while slight stimulation was observed in motility, orientation, and cell compactness of E. gracilis. Similarly, an increase was observed in total soluble protein and sugar content which can be regarded as a protective strategy in response of As-III stress. The adverse effects of As-III in E. gracilis can be attributed to oxidative stress as revealed by the results for oxidative markers. E. gracilis removed 13.3% of As-III from the medium with a calculated 0.27 mg As-III g−1 DW of E. gracilis. The calculated BCF, an index of the potential of accumulating metal/metalloids, in this study was 27 showing E. gracilis as accumulator but not hyper-accumulator of As-III. It is concluded that cell growth and photosynthetic pigments in E. gracilis were adversely affected by As-III and can be used as indicators of the adverse effects of As-III to aquatic autotrophs. The hexavalent form of chromium [Cr(VI)] causes a major reduction in yield and quality of crops worldwide. The root is the first plant organ that interacts with Cr(VI) toxicity, which inhibits primary root elongation, but the underlying mechanisms of this inhibition remain elusive. In this study, we investigate the possibility that Cr(VI) reduces primary root growth of Arabidopsis by modulating the cell cycle-related genes and that ethylene signalling contributes to this process. We show that Cr(VI)-mediated inhibition of primary root elongation was alleviated by the ethylene perception and biosynthesis antagonists silver and cobalt, respectively. Furthermore, the ethylene signalling defective mutants (ein2-1 and etr1-3) were insensitive, whereas the overproducer mutant (eto1-1) was hypersensitive to Cr(VI). We also report that high levels of Cr(VI) significantly induce the distribution and accumulation of auxin in the primary root tips, but this increase was significantly suppressed in seedlings exposed to silver or cobalt. In addition, genetic and physiological investigations show that AUXIN-RESISTANT1 (AUX1) participates in Cr(VI)-induced inhibition of primary root growth. Taken together, our results indicate that ethylene mediates Cr(VI)-induced inhibition of primary root elongation by increasing auxin accumulation and polar transport by stimulating the expression of AUX1. Bisphenol A (BPA) is an emerging environmental pollutant with potentially toxic effects on living organisms. The present study was undertaken to analyze the effects of BPA on the leaves of Arabidopsis thialina by determining the levels of photosynthetic pigments, reactive oxygen species (ROS), membrane lipid peroxidation, and ultrastructural malformation. The obtained results revealed that while a low dose of BPA (10μM) did not alter the test indices significantly, it did cause significant changes in all test indices at higher concentrations. Upon exposure to 40 μM BPA, chlorophyll a and chlorophyll b content showed a decrease of 33% and 30%, respectively. It significantly increased ROS contents and lipid peroxidation at 40 μM BPA exposure. Biochemical and gene expression analysis revealed that the antioxidant system was activated and mounted a defense against BPA-induced ROS. In the case of superoxide dismutase (SOD), 40 μM of BPA caused an increase of 151%. However, the malfunctioning of ascorbate peroxidase (APX) and catalase (CAT) at the highest dose of BPA (40 μM) resulted in incomplete activation of the antioxidant defensive system. BPA stress significantly altered the ultrastructure of cells as evidenced by the reduced number of starch grains, damaged chloroplast and mitochondria, and altered leaf epidermal surface, guard cells, and stomata. Pesticides are one of the most potentially harmful chemicals introduced into the environment, and their adverse impacts on non-target organisms can be significant. The present study was conducted to shed light on effects of locally used insecticides chlorpyrifos (CPF) and lambda cyhalothrin (LCT) on oxidative stress biomarkers in human erythrocytes. The activity of catalase (CAT), superoxide dismutase (SOD), and protein contents as well as the levels of malondialdehyde (MDA) and osmotic fragility (OF) were measured in human erythrocytes exposed to CPF at concentrations of 0, 100, 500, 1000, and 2000 ppm and LCT at concentrations of 0, 100, 300, 600, and 800 ppm for 1 h and 3 h at 37°C. MDA levels and OF of erythrocytes were significantly higher in erythrocytes incubated with CPF and LCT at increasing concentrations of both insecticides and increased incubation time. However, erythrocyte CAT and SOD activities were decreased at all concentrations of CPF and LCT tested. Protein oxidation products were decreased at lower doses of CPF (100 and 500 ppm); at higher doses (1000 and 2000 ppm), total protein content was increased compared with control. In contrast LCT was associated with decreased in protein contents at all the concentrations. In this study, the load of microorganisms and concentration and daily intake of toxic metals (Cu, Zn, Cd, Pb) in drinking water sources (wells) of three towns, Mian Khail (MK), Mer Ahmad Khail (MAK) and Gari Banurian (GB), in Kohat city of Pakistan were investigated. Multifactorial analysis was conducted to determine the microbial and toxic metals load in drinking water of the investigated towns. The obtained results revealed that the fecal coliform bacteria (FCB),Salmonella,ShigellaandStaphylococciwere the highest in MAK followed by MK and GB. The toxic metals concentrations and their intake load in the study area was in the order: Cd > Pb > Cu > Zn. However, the concentrations of toxic elements were within the permissible limits set by the Pakistan Environmental Protection Agency (PAK EPA). The study area had a higher load of enteric pathogens such as theShigellasp., fecal coliform andStaphylococciin the potable water wells. The redundancy analysis (RDA) illustrated that total viable count TVB, Cu,Staphylococciand FCB were higher in MAK, fungi andShigella sp.in GB and the Pb concentration was higher in MK. It is concluded that water in the study area is fecally contaminated and can cause health hazards. It is recommended that there should be a proper monitoring of water quality in the area to improve the existing quality and prevent further contamination of drinking water in the study area. Environmental pollution with heavy metals is a serious issue worldwide posing threats to humans, animals and plants and to the stability of overall ecosystem. Chromium (Cr) is one of most hazardous heavy metals with a high carcinogenic and recalcitrant nature. Aim of the present study was to select low-cost biosorbent using wheat straw and Eupatorium adenophorum through simple carbonization process, capable of removing Cr (VI) efficiently from wastewater. From studied plants a low cost adsorbent was prepared for removing Cr (VI) from aqueous solution following very simple carbonization method excluding activation process. Several factors such as pH, contact time, sorbent dosage and temperature were investigated for attaining ideal condition. For analysis of adsorption equilibrium isotherm data, Langmuir, Freundlich and Temkin models were used while pseudo-first-order, pseudo-second-order, external diffusion and intra-particle diffusion models were used for the analysis of kinetic data. The obtained results revealed that 99.9% of Cr (VI) removal was observed in the solution with a pH of 1.0. Among all the tested models Langmuir model fitted more closely according to the data obtained. Increase in adsorption capacity was observed with increasing temperature revealing endothermic nature of Cr (VI). The maximum Cr (VI) adsorption potential of E. adenophorum and wheat straw was 89.22 mg per 1 gram adsorbent at 308K. Kinetic data of absorption precisely followed pseudo-second-order model. The effect of copper and silver ions on photosynthetic parameters of the flagellate Euglena gracilis was determined. The photosystem II quantum yield (Y (PSII) ) and steady state fluorescence (F t ) were measured with a PAM-fluorometer, while delayed fluorescence (DF) was determined by means of a sensitive single-photon camera attached to an image analysis device, which counted the relative number of photons released from the algae in darkness after previous light-stimulation. We found that after 3 h of incubation photosynthesis was affected only in the presence of relatively high concentrations of the used noble metals. The obtained EC 50 values for Cu 2+ were 505.8 mg L −1 (Y (PSII) ), 286.5 mg L −1 (DF), and 491.0 mg L −1 (F t ). The EC 50 values for silver ions were 51.9 mg L −1 (Y (PSII) ), 51.8 mg L −1 (DF) (values for F t not determinable). Photosynthesis is far less sensitive against toxic noble metals compared to motility parameters as reported in earlier studies, in part because the organelles of photosynthesis, the chloroplasts, are well protected from external substances within the cell, while flagella, reaching into the surrounding of the cell, are more easily influenced. As chloroplasts are easily accessible by means of non-invasive methods e.g. PAM- fluorometry, they are probably interesting model systems in order to detect effects of substances deep inside the cell. Influence on the chloroplasts is a measure that a germicide entered the cells, diffused through the cytoplasm and the membranous systems and reached compartmentalized enzyme complexes. Ethnopharmacological relevance Substantially, plants produce chemicals such as primary and secondary metabolites, which have significant applications in modern therapy. Indigenous people mostly rely on traditional medicines derived from medicinal plants. These plants have the capacity to absorb a variety of toxic elements. The ingestion of such plants for medicinal purpose can have imperative side effects. Hence, with regard to the toxicological consideration of medicinal plants, an effort has been made to review the elemental contents of ethno medicinally important plants of Pakistan and to highlight the existing gaps in knowledge of the safety and efficacy of traditional herbal medications. Materials and methods Literature related to the elemental contents of ethno medicinal plants was acquired by utilizing electronic databases. We reviewed only macro-elemental and trace elemental contents of 69 medicinal plant taxa, which are traditionally used in Pakistan for the treatment of sundry ailments, including anemia, jaundice, cancer, piles, diarrhea, dysentery, headache, diabetes, asthma, blood purification, sedative and ulcer. Results A majority of plants showed elemental contents above the permissible levels as recommended by the World health organization (WHO). As an example, the concentrations of Cadmium (Cd) and Lead (Pb) were reportedly found higher than the WHO permissible levels in 43 and 42 medicinal plants, respectively. More specifically, the concentrations of Pb (54 ppm: Silybum marianum ) and Cd (5.25 ppm: Artemisia herba-alba ) were found highest in the Asteraceae family. Conclusions The reported medicinal plants contain a higher amount of trace and toxic elements. Intake of these plants as traditional medicines may trigger the accumulation of trace and toxic elements in human bodies, which can cause different types of diseases. Thus, a clear understanding about the nature of toxic substances and factors affecting their concentrations in traditional medicines are essential prerequisites for efficacious herbal therapeutics with lesser or no side effects. The present study was conducted to determine the effect of wood ash application on different parameters of Brassica napus L. including seed germination, seedling growth, fresh and dry biomass, water content in seedlings, photosynthetic pigments, soluble sugars, total protein and cell viability. In addition, the effect of wood ash on soil microflora and accumulation of trace elements in seedlings were determined. The seeds of B. napus were grown at different doses of wood ash (0, 1, 10, 25, 50 and 100 g (wood ash)/kg (soil)) and the effect on various parameters was determined. Wood ash significantly inhibited seed germination at doses above 25 g/kg and there was no germination at 100 g/kg of wood ash. At lower concentrations of wood ash, most of the growth parameters of seedlings were stimulated, but at higher concentrations of wood ash most of the studied parameters were adversely affected. Wood ash was found to be very detrimental to B. napus when applied above 25 g/kg. Wood ash application resulted in an increased bioaccumulation of trace elements in seedlings of B. napus . Almost all trace elements were significantly higher in seedlings grown in wood ash above 10 g/kg as compared to the control. An increase in total microbial count was observed with wood ash treatment which was statistically significant at 1 and 10 g/kg of wood ash. Background: North-West of Pakistan is bestowed with medicinal plant resources due to diverse geographical and habitat conditions. The traditional use of plants for curing various diseases forms an important part of the region's cultural heritage. The study was carried out to document medicinal plants used in Frontier Region (FR) Bannu, an area affected by the "War on Terror". Methods: Fieldwork was carried out in four different seasons (spring, autumn, summer and winter) from March 2012 to February 2013. Data on medicinal plants was collected using structured and semi-structured questionnaires from 250 respondents. The voucher specimens were collected, processed and identified following standard methods. Results: Of the 107 species of ethnomedicinal plants reported, fifty percent species are herbaceous. The majority of the reported species were wild (55%) but a substantial proportion are cultivated (29%). For most of the plant species (34%), leaves are the most commonly used part in the preparation of ethnomedicines. The most common use of species is for carminative purposes (14 species), with the next most common use being for blood purification (11 species). The main methods used in the preparation of ethnomedicinal recipes involves grinding and boiling, and nearly all the remedies are taken orally along with ingredients such as water, milk or honey for ease of ingestion. Traditional healers prepare plant remedies using one or more plants. There was a significant correlation (r² = 0.95) between the age of local people and the number of plants known to them, which indicates that in the coming 20 years, an approximate decrease of 75% in the indigenous knowledge may be expected. Conclusion: Traditional medicines are important to the livelihoods of rural communities in the region affected by the Global war on Terrorism. The medicinal recipes are indigenous; however, there is a threat to their future use on account of rapid modernization and terrorist activities. Documentation of medicinal plants and recipes may help in the conservation of the regional indigenous medicinal knowledge for future generations and to provide a baseline for further studies.
