Land‐use change and habitat degradation are among the biggest drivers of aboveground biodiversity worldwide but their effects on soil biodiversity are less well known, despite the importance of soil organisms in developing soil structure, nutrient cycling and water drainage. Combining a global compilation of biodiversity data from soil assemblages collated as part of the PREDICTS project with global data on soil characteristics, we modelled how taxon richness and total abundance of soil organisms have responded to land use. We also estimated the global Biodiversity Intactness Index (BII)—the average abundance and compositional similarity of taxa that remain in an area, compared to a minimally impacted baseline, for soil biodiversity. This is the first time the BII has been calculated for soil biodiversity. Relative to undisturbed vegetation, soil organism total abundance and taxon richness were reduced in all land uses except pasture. Soil properties mediated the response of soil biota, but not in a consistent way across land uses. The global soil BII in cropland is, on average, a third of that originally present. However, in grazed sites the decline is less severe. The BII of secondary vegetation depends on age, with sites with younger growth showing a lower BII than mature vegetation. We conclude that land‐use change has reduced local soil biodiversity worldwide, and this further supports the proposition that soil biota should be considered explicitly when using global models to estimate the state of biodiversity. The distribution of earthworm ecological groups in urban areas is not well-known, despite their crucial role in delivering soil ecosystem services such as nutrient cycling and water drainage. Citizen science engages public audiences in the scientific research process and is an excellent tool for collecting biodiversity data in urban areas, where most of the UK population resides. However, a disadvantage is that differing levels of skill and engagement among participants can create statistical challenges. The Earthworm Watch citizen science project used 668 matched-pair surveys to estimate how the abundance and ecological diversity of earthworms respond to land management practices, and soil properties in UK urban habitats. A total of 5170 earthworms were counted during the project with a mean of 8 earthworms per soil pit - equivalent to a density of 198 earthworms per m2. Soil moisture and texture were the largest drivers of total earthworm abundance, with habitat borderline statistically insignificant. Endogeic earthworms were found in 71 % of soil pits, epigeic in 62 % and anecic in 33 %. Fertiliser use also had a significant effect on total abundance, but only when organic fertiliser was used. Earthworm ecological groups demonstrated varied responses to habitat, with endogeic earthworms consistently the most abundant group, showing slight preferences for grasslands and vegetable beds. Anecic earthworms had the lowest abundance across all habitats but were more prevalent in grasslands and vegetable beds. Epigeic earthworms were most abundant beneath shrubs and hedges. These findings align with expected patterns of earthworm ecology, underscoring the potential of well-designed citizen science projects to yield valuable insights into urban earthworms and soil health. • Earthworm Watch used a matched-pair design to reduce variability among participants. • Soil texture and moisture had the largest effect on total numbers, habitat less so. • There were more earthworms when organic, but not inorganic fertiliser was used. • Earthworm ecological groups differed in their response to habitat. The high biodiversity of soil ecosystems is often attributed to their spatial heterogeneity at multiple scales, but studies on the small-scale spatial distribution of soil macrofauna are rare. This case study of an Ash – Field Maple woodland partially converted to conifer plantation investigates differences between species assemblages of soil and litter invertebrates, and plants, using multivariate ordination and indicator species analysis for eleven microhabitats. Microhabitats representing the main body of uniform litter were compared with more localised microhabitats including dead wood and areas of wet soil. Species accumulation curves suggest that for this site it is more efficient to sample from varied microhabitats of limited spatial scale rather than the broad habitat areas when generating a species inventory. For comparative work sampling the main body of uniform litter is more appropriate, given that microhabitats vary from woodland to woodland and would make standardisation problematic. Vegetation showed more distinctive microhabitat-specific species assemblages than soil and leaf litter invertebrates and was strongly associated with environmental variables. Microhabitats with distinct assemblages included dead wood habitats, which had a high proportion of saproxylic species; a highly disturbed microhabitat with distinct plant and soil species characteristic of ruderal habitats and seeps with earthworm species rarely sampled in standard soil biodiversity surveys. The leaf litter in the conifer plantation area was species poor and the biodiversity quantified was considerably enhanced by the sampling from the additional microhabitats - illustrating the importance of small-scale heterogeneity for increasing plant and soil macrofauna biodiversity at this site. Previous studies from our group have demonstrated that bone morphogenetic protein receptor-II (BMPR-II), expressed on pulmonary artery endothelial cells, imparts profound anti-inflammatory effects by regulating the release of proinflammatory cytokines and promoting barrier function by suppressing the transmigration of leukocytes into the pulmonary vessel wall. Here we demonstrate that, in mice with endothelial-specific loss of BMPR-II expression (L1Cre(+);Bmpr2f/f), reduction in barrier function and the resultant pulmonary hypertension observed in vivo are the result of increased leukocyte recruitment through increased CXCR½ signaling. Loss of endothelial expressed BMPR-II leads to elevated plasma levels of a wide range of soluble mediators important in regulating leukocyte migration and extravasation, including the CXCR½ ligand, KC. Treatment of L1Cre(+);Bmpr2f/f mice with the CXCR½ antagonist SCH527123 inhibits leukocyte transmigration into lung and subsequently reverses the pulmonary hypertension. Our data have uncovered a previously unrecognized regulatory function of BMPR-II, which acts to regulate the expression of CXCR2 on endothelial cells, suggesting that increased CXCR2 signaling may also be a feature of the human pathology and that CXCR½ pathway antagonists may represent a novel therapeutic approach for treating pulmonary hypertension because of defects in BMPR-II expression. Abstract: We investigated the migration of human leukocytes through endothelial cells (EC), and particularly their underlying basement membrane (BM). EC were cultured for 20days on 3μm-pore filters or collagen gels to form a distinct BM, and then treated with tumour necrosis factor-α, interleukin-1β or interferon-γ. Neutrophil migration through the cytokine-treated EC and BM was delayed for 20-day compared to 4-day cultures. The BM alone obstructed chemotaxis of neutrophils, and if fresh EC were briefly cultured on stripped BM, there was again a hold-up in migration. In studies with lymphocytes and monocytes, we could detect little hold-up of migration for 20-day versus 4-day cultures, in either the filter- or gel-based models. Direct microscopic observations showed that BM also held-up neutrophil migration under conditions of flow. Treatment of upper and/or lower compartments of filters with antibodies against integrins, showed that neutrophil migration through the endothelial monolayer was dependent on β2-integrins, but not β1- or β3-integrins. Migration from the subendothelial compartment was supported by β1- and β2-integrins for all cultures, but blockade of β3-integrin only inhibited migration effectively for 20-day cultures. Flow cytometry indicated that there was no net increase in expression of β1- or β3-integrins during neutrophil migration, and that their specific subendothelial function was likely dependent on turnover of integrins during migration. These studies show that BM is a distinct barrier to migration of human neutrophils, and that β3-integrins are particularly important in crossing this barrier. The lesser effect of BM on lymphocytes and monocytes supports the concept that crossing the BM is a separate, leukocyte-specific, regulated step in migration. [Copyright &y& Elsevier]

Motivation and aim: Soil biodiversity is central to ecosystem function and services. It represents most of terrestrial biodiversity and at least a quarter of all biodiversity on Earth. Yet, research into broad, generalizable spatial and temporal patterns of soil biota has been limited compared to aboveground systems due to complexities of the soil system. We review the literature and identify key considerations necessary to expand soil macroecology beyond the recent surge of global maps of soil taxa, so that we can gain greater insight into the mechanisms and processes shaping soil biodiversity. We focus primarily on three groups of soil taxa (earthworms, mycorrhizal fungi and soil bacteria) that represent a range of body sizes and ecologies, and, therefore, interact with their environment at different spatial scales. Results: The complexities of soil, including fine‐scale heterogeneity, 3‐D habitat structure, difficulties with taxonomic delimitation, and the wide‐ranging ecologies of its inhabitants, require the classical macroecological toolbox to be expanded to consider novel sampling, molecular identification, functional approaches, environmental variables, and modelling techniques. Main conclusions: Soil provides a complex system within which to apply macroecological research, yet, it is this property that itself makes soil macroecology a field ripe for innovative methodologies and approaches. To achieve this, soil‐specific data, spatio‐temporal, biotic, and abiotic considerations are necessary at all stages of research, from sampling design to statistical analyses. Insights into whole ecosystems and new approaches to link genes, functions and diversity across spatial and temporal scales, alongside methodologies already applied in aboveground macroecology, invasion ecology and aquatic ecology, will facilitate the investigation of macroecological processes in soil biota, which is key to understanding the link between biodiversity and ecosystem functioning in terrestrial ecosystems. Soils are key components of our ecosystems and provide 95%–99% of our food. This importance is reflected by an increase in participatory citizen science projects on soils. Citizen science is a participatory research method that actively involves and engages the public in scientific enquiry to generate new knowledge or understanding. Here, we review past and current citizen science projects on agricultural soils across Europe. We conducted a web‐based survey and described 24 reviewed European citizen science projects in the light of the 10 principles of citizen science and identified success factors for citizen science. Over 66% of the projects generated soil biodiversity data; 54% and 42% of the projects generated data on vegetation cover and soil organic carbon, respectively. Our findings show that soil citizen science projects aligned with the 10 principles of citizen science offer an unexploited resource for European soil health research. We conclude that promoting co‐creation, fostering knowledge‐sharing networks and enabling long‐term communication and commitment with citizens are success factors for further development of citizen science on soils. Abstract: While the effect of ultrasound as an extreme example of low-magnitude high-frequency stimulation has been explored in the response of bone to injury, little is known about its effect on normal bone. This experiment was designed to test the hypothesis that ultrasound exerts a similar influence on bone as mechanical stimulation at a physiological level. Three groups of female Wistar rats were anaesthetised (6 per group). In one group, the left ulna was loaded cyclically in vivo 40 times, repeated on a further 5 occasions on alternate days. In a second group, transcutaneous low-intensity pulsed ultrasound stimulation was applied to the left ulnae for the same duration as the period of loading. In a third group, loading and ultrasound stimulation were applied concurrently. The right ulna served as non-loaded control in each animal. At the end of the experiment after 14 days, both ulnae were removed. Induced bone formation was assessed by measuring the proportion of medial periosteal bone surface with double label (dLS/BS, %) and by calculation of mineral apposition rate (MAR) from the inter-label distance. All three treatments induced a significant periosteal response, increasing dLS/BS values from <10% in control limbs to >80% in treated limbs. Increases in MAR of experimental ulnae versus contralateral control ulnae were 2.9 (±0.9), 8.6 (±2.4) and 8.7μm (±3.2) for the ultrasound only, ultrasound and load, and load only groups, respectively. The effects of loading plus ultrasound were not significantly different from ultrasound alone. These data suggest that ultrasound is able to induce changes in bone that share at least some features with mechanical loading. [Copyright &y& Elsevier]

Abundance and distribution of earthworms in agricultural fields is frequently proposed as a measure of soil quality assuming that observed patterns of abundance are in response to improved or degraded environmental conditions. However, it is not clear that earthworm abundances can be directly related to their edaphic environment, as noted in Darwin's final publication, perhaps limiting or restricting their value as indicators of ecological quality in any given field. We present results from a spatially explicit intensive survey of pastures within United Kingdom farms, looking for the main drivers of earthworm density at a range of scales. When describing spatial variability of both total and ecotype-specific earthworm abundance within any given field, the best predictor was earthworm abundance itself within 20–30 m of the sampling point; there were no consistent environmental correlates with earthworm numbers, suggesting that biological factors (e.g. colonisation rate, competition, predation, parasitism) drive or at least significantly modify earthworm distributions at this spatial level. However, at the national scale, earthworm abundance is well predicted by soil nitrate levels, density, temperature and moisture content, albeit not in a simple linear fashion. This suggests that although land can be managed at the farm scale to promote earthworm abundance and the resulting soil processes that deliver ecosystem services, within a field, earthworm distributions will remain patchy. The use of earthworms as soil quality indicators must therefore be carried out with care, ensuring that sufficient samples are taken within field to take account of variability in earthworm populations that is unrelated to soil chemical and physical properties. GROW Observatory is a project funded under the European Union's Horizon 2020 research and innovation program. Its aim is to establish a large scale (more than 20,000 participants), resilient and integrated 'Citizen Observatory' (CO) and community for environmental monitoring that is self-sustaining beyond the life of the project. This article describes how the initial framework and tools were developed to evolve, bring together and train such a community; raising interest, engaging participants, and educating to support reliable observations, measurements and documentation, and considerations with a special focus on the reliability of the resulting dataset for scientific purposes. The scientific purposes of GROW observatory are to test the data quality and the spatial representativity of a citizen engagement driven spatial distribution as reliably inputs for soil moisture monitoring and to create timely series of gridded soil moisture products based on citizens' observations using low cost soil moisture (SM) sensors, and to provide an extensive dataset of in situ soil moisture observations which can serve as a reference to validate satellite-based SM products and support the Copernicus in situ component. This article aims to showcase the initial steps of setting up such a monitoring network that has been reached at the mid-way point of the project's funded period, focusing mainly on the design and development of the CO monitoring network. The PREDICTS project-Projecting Responses of Ecological Diversity In Changing Terrestrial Systems ()-has collated from published studies a large, reasonably representative database of comparable samples of biodiversity from multiple sites that differ in the nature or intensity of human impacts relating to land use. We have used this evidence base to develop global and regional statistical models of how local biodiversity responds to these measures. We describe and make freely available this 2016 release of the database, containing more than 3.2 million records sampled at over 26,000 locations and representing over 47,000 species. We outline how the database can help in answering a range of questions in ecology and conservation biology. To our knowledge, this is the largest and most geographically and taxonomically representative database of spatial comparisons of biodiversity that has been collated to date; it will be useful to researchers and international efforts wishing to model and understand the global status of biodiversity. Land use and related pressures have reduced local terrestrial biodiversity, but it is unclear how the magnitude of change relates to the recently proposed planetary boundary (“safe limit”). We estimate that land use and related pressures have already reduced local biodiversity intactness—the average proportion of natural biodiversity remaining in local ecosystems—beyond its recently proposed planetary boundary across 58.1%of the world’s land surface, where 71.4% of the human population live. Biodiversity intactness within most biomes (especially grassland biomes), most biodiversity hotspots, and even some wilderness areas is inferred to be beyond the boundary. Such widespread transgression of safe limits suggests that biodiversity loss, if unchecked, will undermine efforts toward long-term sustainable development. [ABSTRACT FROM AUTHOR]
