Methodology for detection of activated benzo[ a]pyrene (B[a]P)-nucleoside adducts by liquid chromatography-tandem mass spectrometry is reported. Adducts of B[a]P-dihydrodiol epoxide (B[a]PDE) with guanosine and adenosine have been detected for the first time by use of precursor ion scan and neutral loss scan. B[a]P was then activated by use of UV irradiation and some of the products obtained have been identified by taking advantage of the information obtained for B[a]PDE. Photoactivation has also been carried out in the presence of hydrogen peroxide; this resulted in a higher yield of products with increased production of BaP diones. The reactivity of these compounds toward nucleosides has been tested. The proposed method was successfully used for detection of one stable guanosine-B[a]P dione adduct. The free fatty acid (FFA) content together with the lipase activity control can be considered as useful indexes of good quality and correct storage of food, especially for milk. The quantitative analysis of FFA in different kinds of milk has been performed by a potentiometric method, using a new extractive methodology outlined herein. The lipase activity has been controlled by a sensitive calorimetric method, previously validated by us, based on the direct measure of the heat quantity involved in the enzymatic reaction. In order to verify the milk quality after the healing treatments and/or during the shelf life, the behaviours of FFA content and of lipase activity have been outlined in function of storage time and pH variations on different typologies of milk. The FFA content in sample of fresh pasteurised milk was found to be quite high after the 5th/6th day of storage at +4 °C, meanwhile the pH values were always constant and only after the 9th day begun to decrease. At the same time the lipase activity, directly measured, was found to be appreciable after the 6th day of storage, giving an exothermic answer at the calorimeter, similar to that of a milk sample where only three international units of standard lipase were added. The coordination chemistry of dioxouranium(VI) with organic ligands may represent an useful model to describe the migration of radionuclides in natural fluids. A new dioxouranium(VI) ternary complex of formula Na4(UO2)2(OH)4(C2O4)2 was precipitated at pH≅8. It was characterized by elemental analysis, ICP and IR spectroscopy, thermogravimetry (TG) and coupled TG–IR evolved gas analysis. Potentiometric measurements in the solution suggest that the equilibrium shifts from the prevalence of the (UO2)2(OH)2(C2O4)46− complex to the [(UO2)2(OH)4(C2O4)2]4− complex before the formation of a solid phase. Le endotossine sono molecole infiammatorie derivate da lisi di batteri gram-negativi, ubiquitari nell'ambiente. Numerosi studi hanno dimostrato la pericolosità dell'esposizione ad endotossine batteriche sia in ambienti di lavoro che di vita. Per determinare i livelli di esposizione all'endotossina ed eventualmente proporre dei limiti di riferimento, è necessario valutarne la presenza con metodi standardizzati. Poiché è stata recentemente accertata l'influenza dei metodi di campionamento, di estrazione e di analisi sul risultato, in questo lavoro abbiamo cercato di valutare quali sia la metodologia migliore per la determinazione della presenza di endotossina, considerando anche l'"effetto matrice", cioè il campionamento dell'aria in ambienti indoor. In questo lavoro cominciano a delinearsi le migliori condizioni per la valutazione delle endotossine aereodisperse, tenendo sempre presente l'estrema variabilità dei campioni biologici, sia in fase di campionamento che di analisi. This work describes a new analytical method for the determination of four cobalamins (adenosylcobalamin (AdoCbl), methylcobalamin (MeCbl), hydroxocobalamin (OHCbl) and cyanocobalamin (CNCbl)) in cow's milk. The extraction procedure is fast and based on dilution/protein precipitation of a milk sample with 50 mM sodium acetate buffer (pH 4.6), followed by solid phase extraction (SPE) of the filtered supernatant. Relative recoveries higher than 60% have been obtained for all the cobalamins by combining two different types of sorbents in the same SPE cartridge: two disks of buckypaper (BP), a nanoporous felt composed of oxidized multiwalled carbon nanotubes (MWCNTs), separated by a Teflon frit from OASIS HLB (500 mg), a hydrophilic–lipophilic balance copolymer. Before its use as sorbent, BP was characterized in terms of porosity, permeability, surface area, specific adsorption capacity and tested for a potential reuse after adequate chemical regeneration. The analysis of the extracts was performed by liquid chromatography (LC) coupled to tandem mass spectrometry (MS/MS) on an analytical C 18 column in less than 10 min. After validation, the method was applied to the determination of the natural content of the four B 12 homologues in cow's milk samples, providing data lacking in the literature. Abstract: It is here described the comparison of extraction efficiency of some solutions (acetate buffer, deionized water, diluted HNO3 and EDTA) frequently adopted in literature for evaluating the elemental solubility in airborne particulate matter. This comparison was performed considering the distribution of As, Ba, Ca, Cd, Co, Cr, Cu, Fe, Mg, Mn, Na, Ni, Pb, S, Si, Sb, Sn, Sr, Ti, V, Zn between the extractable and mineralized residual fractions on the NIST 1648 certified material, PM10 real samples and size-segregated samples, collected by a 13-stage impactor. The extracting solutions were evaluated by comparing extractive efficiencies and robustness towards some factors, such as acidity and concentration of complexing species, that have great environmental variability and that could be able to modify the extractive efficiency. Furthermore, extraction methods application to size-segregated samples allowed estimating the selectivity of extracting solutions towards dimensionally characterized emission sources, as dusts originated from abrasion and road dust re-suspension. On the basis of the obtained results, it was possible to define the main advantages and disadvantages resulting from the use of different extracting solutions, necessary to make possible the comparison of environmental studies carried out in different extractive conditions and to start up a proper study for harmonizing extracting procedures. An analytical procedure for the simultaneous determination in human plasma and oral fluids of several illicit drugs belonging to different chemical and toxicological classes is presented. Amphetamine, methamphetamine, morphine, 6-monoacetylmorphine, methylenedioxyamphetamine, methylenedioxyethylamphetamine, methylenedioxymethamphetamine, cocaine, benzoylecgonine, tetrahydrocannabinol, carboxytetrahydrocannabinol, ketamine, and phencyclidine have been quantified in real samples using a very rapid sample treatment, basically a protein precipitation. The quantitative analysis was performed by liquid chromatography–tandem mass spectrometry and has been fully validated. All the analytes were detected in positive ionization mode using a TurboIonSpray source, except carboxytetrahydrocannabinol, which was detected in negative ionization mode. The use of a diverter valve between the column and the mass spectrometer allows the preservation of the ion source performances for high-throughput analysis. A thermoanalytical study of unusual adrenaline coordination compounds with the divalent transition-metal ions Co(II), Ni(II) and Cu(II) is reported. The precipitated compounds were characterized by elemental analysis and by thermogravimetric (TG) analysis coupled to FTIR spectroscopy to analyze the evolved gases and make consistent the proposed decomposition steps. L'avvento di dispositivi elettronici in ambienti di ufficio non solo ha migliorato l'efficienza di lavoro, ma ha anche cambiato la qualità dell'aria negli ambienti confinati (IAQ). Nello studio si sono caratterizzati e determinati quantitativamente i COV (Composti Organici Volatili), la formaldeide e l'ozono emessi da una stampante laser, in assenza ed in presenza di un sistema filtrante, nella "stanza dei servizi"di un ufficio contenente oltre ad altri dispositivi elettronici una stampante laser. L'inquinamento atmosferico può essere definito come la presenza nell'atmosfera di sostanze che causano un effetto sull'essere umano, sugli animali, sulla vegetazione o sui diversi materiali. Il loro effetto sull'uomo e sull'ambiente dipende da vari fattori, come ad esempio la quantità di inquinante presente nell'aria, la durata dell'esposizione e la pericolosità dell'inquinante stesso. Considerata quindi, l'importanza e la necessità, di controllare e monitorare l'aria che respiriamo nei luoghi dove viviamo e lavoriamo, il laboratorio ambientale del Centro Ricerche di Lamezia Terme (CZ) ha pianificato ed effettuato una campagna di monitoraggio della qualità dell'aria in punti strategici del comune di Lamezia Terme (CZ). I siti presi in considerazione sono due; l'uno posizionato all'interno dell'area industriale del comune, l'altro, nel centro urbano della cittadina calabrese. I parametri chimici monitorati, in conformità ai dettami tecnici definiti nel D.lgs n. 155 del 13/08/2010, sono anidride solforosa (SO2), ossidi di azoto (NO/NO2/NOx), ossido di carbonio (CO), ozono (O3), IPA (idrocarburi policiclici aromatici) e BTX (benzene, toluene, e xilene). Il monitoraggio è stato effettuato in continuo, con gli analizzatori a misura diretta e mediante l'analizzatore di campo DOAS (Differential Optical Absorption Spectroscopy), posizionato sul laboratorio mobile. Allo scopo di effettuare anche un'analisi biologica ambientale sono stati utilizzati i filtri del TEOM per l'estrazione di endotossine batteriche. [ABSTRACT FROM AUTHOR]

L'uso di citostatici antiblastici (CA) per il trattamento del cancro è una pratica comune negli ospedali e l'esposizione professionale a CA costituisce un potenziale pericolo per la salute degli operatori. Inoltre molte pubblicazioni hanno dimostrato che la contaminazione da CA è ancora presente sulle superfici di lavoro dopo le procedure di pulizia. Tra i farmaci citotossici trattati in ambito ospedaliero, abbiamo scelto la ciclofosfamide (IARC, gruppo 1) come indicatore di esposizione professionale. Con l'obiettivo di valutare l'esposizione alla ciclofosfamide del personale ospedaliero addetto alla preparazione e somministrazione dei CA, in questo studio presentiamo tre metodi di screening innovativi e rapidi basati sull'uso dell'interfaccia Desorption Electrospray Ionization (DESI) accoppiata con uno spettrometro di massa a triplo quadrupolo. Dal momento che la contaminazione da ciclofosfamide nei luoghi di lavoro è spesso dovuta all'aerosolizzazione delle sue polveri o liquidi, al contatto diretto con materiali contaminati e ad errate procedure di pulizia, abbiamo messo a punto due metodi per l'analisi ambientale e uno per il monitoraggio biologico. A questo scopo, per il monitoraggio ambientale, abbiamo usato una placca di vetro stampata con PTFE come campionatore passivo e un panno a cattura elettrostatica come wipe, mentre per il monitoraggio biologico abbiamo scelto la matrice urina. Tutti i campioni sono analizzati direttamente senza alcun pre-trattamento. I metodi proposti sono in grado di fornire informazioni qualitative sulla presenza/assenza della ciclofosfamide e consentono analisi rapide, accurate, eco-friendly e, non ultimo, low-cost. Il possibile rischio cancerogeno associato all'esposizione a basse dosi di derivati del petrolio e, in particolare, a vapori di benzina contenenti benzene, è un argomento di rilevante interesse da un punto di vista di sanità pubblica. I benzinai rappresentano una popolazione modello per l'esposizione a basse dosi di benzene (generalmente inferiori ai 3 µg/m3 ovvero <1 ppm) e studi precedenti condotti presso l'ISS hanno mostrato, pur a questi bassi livelli di esposizione, incrementi di alcuni indicatori di danno genotossico. Non è però chiaro se tali incrementi si traducano in danni sulla salute evidenziabili clinicamente o rilevabili attraverso le analisi dei rischi di mortalità per causa. Nel presente lavoro, viene proposto il risultato di un'indagine effettuata sul territorio del Comune di Cosenza, condotta direttamente presso gli operatori dei distributori. L'indagine ha lo scopo di conoscere l'effettiva esposizione a benzene, toluene e xileni da parte dei benzinai. Tutti gli operatori delle stazioni di rifornimento monitorate sono stati sottoposti alla valutazione dell'esposizione ai solventi aerodispersi, mediante campionatori personali posti direttamente all'altezza della zona respiratoria degli addetti alle stazioni di servizio, per la determinazione della concentrazione di benzene, toluene e xileni aerodispersi eseguiti in tre diverse giornate. I valori delle concentrazioni rilevate per i BTX sono stati quindi confrontati con quelli riportati nel D.Lgs. n. 81 del 9 aprile 2008, in particolare nell'Allegato XLIII per quanto concerne il benzene (3,25 mg/m3) e nell'Allegato XXXVIII per quanto attiene agli isomeri o-, m-, p degli xileni (221 mg/m3) ed al toluene (192 mg/m3). I risultati di questa prima fase di indagini evidenziano che l'esposizione media a BTX per i benzinai delle stazioni di rifornimento monitorate durante il turno di lavoro è, in termini numerici, di molto inferiore ai limiti proposti per gli ambienti di lavoro. Per quanto riguarda le tre categorie studiate (Corigliano, Rende, Cosenza), i valori ottenuti mostrano un andamento proporzionale al traffico veicolare delle località stesse e ancora, rispecchiano l'ubicazione delle stazioni di distribuzione, da zone centrali a zone più periferiche. Infine i risultati dell'indagine sull'esposizione a BTX nei distributori di carburante dei Comuni in studio, evidenziano che, nel periodo primaverile quando le condizioni meteoclimatiche favoriscono la diffusione degli inquinanti, le concentrazioni rilevate sono inferiori rispetto al periodo invernale. The identification of dyes from ancient textiles represents a challenging issue requiring high proficiency and versatility, that must be guided by the principles of non-destructiveness. The complexity of the dye/textile system influences both the choice of sampling and extraction procedures and the analytical methodology. For instance, in order to preserve both the fibre and the original chromophores on it, the extraction procedures commonly used for natural dyes have evolved in last few decades towards increasingly mild conditions. Along with this approach, the use of solid substrates for sampling has become prominent in several fields of Cultural Heritage diagnostics.With reference to these two aspects, the aim of this work is to combine gel substrates and a mild basic solution for the extraction of anthraquinone dyes (madder and cochineal) from wool textiles, in order to evaluate the versatility of the extraction procedure for different analytical techniques. Because of their wide use in the field of Conservation, agar and nanorestore gel substrates soaked in an ammonia-EDTA solution were tested. The dye-extracting substrates were then analysed with Raman and SERS spectroscopy, whilst the fraction re-extracted from them was examined with HPLC-HRMS. Finally, scanning electron microscopy was used to monitor the integrity of the wool after the extraction treatment with the ammonia-EDTA solution.HPLC-HRMS results show that the basic solution in both gels is effective for the dye extraction, and no alteration of wool fibres was evidenced by SEM-EDS. The Raman spectra give no information about the dyes, but only substrate signals are obtained. Conversely, SERS spectroscopy makes it possible to distinguish the two dyes, especially using the 633 nm laser on the agar substrate. [ABSTRACT FROM AUTHOR]

Thermogravimetry (TG) and derivative thermogravimetry (DTG) are proposed as a fast and sensitive tool to discriminate the industrial processing of spaghetti pasta by focusing the attention on the thermal profiles of the structured water release. Thirteen different name-brand and store-brand kinds of spaghetti pasta were analysed. The results show different profiles according to the drying procedures.
