TeO2 single crystal has drawn much attention in recent years due to its excellent acousto‐optic (AO) performance in the field of spectral imaging and optical processing. The optical properties of TeO2 single crystal are critical for the fabrication of AO devices. To improve the optical properties of the TeO2 single crystal, Mn4+ ions were doped into the TeO2 lattice. Herein, bulk TeO2 single crystal and TeO2:0.02Mn4+ single crystal have been successfully grown by a Bridgman method. Compared with the TeO2 single crystal, a 30% transmittance increase was observed in the TeO2:0.02Mn4+ single crystal. Furthermore, the TeO2:0.02Mn4+ single crystal shows a smaller extinction coefficient (≤0.01) in the Vis–NIR region, indicating less optical loss. The bandgap energy of TeO2:0.02Mn4+ was broadened from 3.360 to 3.635 eV. In addition, TeO2:0.02Mn4+ single crystal exhibits a higher refractive index (about 2.244), which indicates that TeO2:0.02Mn4+ single crystal is a potential medium for high‐efficiency AO devices. Among many unexpected phenomena of active matter is the recently observed superfluid-like thinning (viscosity drop) behavior of bacteria suspensions. Understanding this peculiar self-propelled thinning by active matter is of theoretical and practical importance. Here, we find that, although distinct in driving mechanisms, active matter and shear flows exhibit similar thinning behaviors upon the increase of self-propulsion and shear forces, respectively. Our structural characterizations reveal that they actually share the same cluster-breaking mechanism of thinning. How fast and how shattered the cluster is broken determines the (dis)continuity of the thinning. This explains why adding active particles to Newtonian fluids can cause thinning, in which rotation of active particles play a key role in breaking clusters. The fermionic hierarchical equations of motion (HEOM) approach has found wide application in the exploration of open quantum systems, and extensive efforts have been committed to improving its efficiency and accuracy in practical calculations. In this work, by scrutinizing the stationary-state and dynamic properties of Kondo-correlated quantum impurity systems, we show that the strength of Kondo correlation induced by the system-environment entanglement primarily determines the converged hierarchical truncation tier of the HEOM method. This complements the rule of thumb regarding the positive correlation between the height of hierarchy and system-environment coupling strength. Objective: To analyze effects of tectorigenin on improving cognitive deficits in rats with vascular dementia (VD) by regulating Toll-like receptor 4 (TLR4)/myeloid differentiation factor 88 (MyD88)/nuclear factor-κB (NF-κB) signaling pathway. Methods: A total of 72 rats were randomly divided into sham operation group, model group, low, medium and high doses [25, 50, 100 mg/(kg⋅d)] tectorigenin groups and positive control group [piracetam 324 mg/(kg⋅d], with 12 rats in each group. Except for sham operation group, VD models were replicated in other groups. After successful modeling, different doses tectorigenin groups and positive control group were administered intragastrically with different doses of tectorigenin and piracetam, while other groups were administered intragastrically with same volume of normal saline for 28 d. Spatial learning and memory ability were detected by Morris water maze. Neurotransmitter levels in hippocampus interstitial fluid were detected by high performance liquid chromatography-electro-chemical. Brain-derived neurotrophic factor (BDNF) and tyrosine kinase receptor b (TrkB) expressions in hippocampus were detected by RT-qPCR and Western blot. TLR4/MyD88/NF-κB pathway-related proteins in hippocampus were detected by Western blot. Results: Compared with sham operation group, escape latency was longer, while stay time in target area and times of crossing platform were lower in model group (P<0.05). Compared with model group, escape latency was shorter, while stay time in target area and times of crossing platform were higher in medium and high doses tectorigenin groups (P<0.05). NE, DA, 5-HT and 5-HIAA levels in model group were lower than those in sham operation group (P<0.05), which were higher in medium and high doses tectorigenin groups than model group (P<0.05). Compared with sham operation group, BDNF and TrkB mRNA and proteins levels were lower, while TLR4, MyD88 and p-NF-κB p65/NF-κB p65 proteins levels were higher in model group (P<0.05). Compared with model group, BDNF and TrkB mRNA and proteins levels were higher, while TLR4, MyD88 and p-NF-κB p65/NF-κB p65 proteins levels were lower in medium and high doses tectorigenin groups (P<0.05). The TESS Survey has released a large number of high-precision light curves of contact binaries. However, using the Phoebe program and Markov chain Monte Carlo (MCMC) algorithm to obtain the posterior distribution of contact binary parameters is a time-consuming process. In order to obtain the contact binary parameters from the TESS survey, we build neural network (NN) models and combine them with the MCMC algorithm to obtain the contact binary parameters and parameter errors quickly. NN model is used in place of the physical model, which can generate a light curve with a precision of less than a millimagnitude. The NN model is capable of generating light curves at a speed that is four orders of magnitude faster than Phoebe running on the same computing platform. In this study, we have determined the parameters of 318 contact binary systems exhibiting relatively symmetric light curves. Subsequently, a statistical analysis was conducted on the derived parameters of these 318 targets. The coefficient of determination (R 2) for 318 contact binaries between the light curves generated by Phoebe using these parameters obtained by the NN model and MCMC as inputs and the original light curves is greater than 0.99. In order to improve the mechanical properties and deformation characteristics of permeable concrete, glass fiber was added to this type of concrete. Based on an unconfined compressive strength test, non-contact full-field strain measurement system, and scanning electron microscopy test, the effects of aggregate particle composition, shaking time, fly ash content, fiber length, and fiber content on the strength and permeability of permeable concrete were studied. The results show that the strength and water permeability of permeable concrete are negatively correlated with an increase in shaking time. When the aggregate particle size is 5–10 mm, the permeable concrete has both good strength and permeability. Proper incorporation of fly ash improves the compactness inside the structure. The influence of different lengths of glass fiber on the strength of permeable concrete first increases and then decreases, and the permeable property decreases. With the same fiber length, the strength increases first and then decreases with an increase in the content, while the porosity and water permeability coefficient decrease. Under the test conditions, when the length of glass fiber is 6 mm, and the dosage is 2 kg/m3, the strength performance of permeable concrete is the best, and the permeability effect is good at the same time. As data relay satellites (DRSs) play an increasingly important supporting role in remote sensing missions, efficient coordination across space–ground multiresources becomes a significant problem. Owing to the implementation problem of the centralized coordinate methods, this paper studies a distributed coordinate resource scheduling method which is realizable in the current space network structure. To be specific, we first formulate the multiple resource coordination problem into an MILP problem based on a modified time-expanded graph. Then, the problem is transferred and decomposed into subproblems for remote sensing satellite (RSS) systems and DRS systems to solve distributedly. Afterwards, we propose a distributed iterative scheme for the RSS systems and DRS systems based on alternating direction method of multipliers (ADMM), in which only the schedule information of the inter-satellite links are required to exchange between RSS systems and DRS systems. Herein, reaction of 4,4′,4′′‐nitrilo‐tribenzhydrazide with tris(μ2‐4‐carboxaldehyde‐pyrazolato‐N,N′)‐tricopper (Cu3Py3) generates the crystalline copper organic framework USTB‐11(Cu). Post‐modification with divalent nickel ions affords the heterometallic framework USTB‐11(Cu,Ni). Powder X‐ray diffraction and theoretical simulations reveal their two‐dimensional hexagonal structure geometry. A series of advanced spectroscopic techniques disclose the mixed CuI/CuII state nature of Cu3Py3 in USTB‐11(Cu,Ni) with a uniform bistable Cu34+(CuI2CuII) : Cu35+(CuICuII2) (ca. 1 : 3) oxidation state, resulting in a significantly improved formation efficiency of the charge‐separation state. Cell phone call operations during driving can lead to distraction and cause potential safety hazards. In order to quantitatively characterize the influence of cell phone call mode on the driver's visual behavior in different traffic conditions and to analyze the risk level of distracted driving from the visual level, a distracted driving simulation test was carried out based on a driving simulator and eye tracker. The eye movement data of drivers during normal driving, hands-free call, and video call under two typical traffic conditions of free flow and congested flow on the urban expressway were collected. Firstly, four visual characteristics indicators that were highly sensitive to traffic conditions and driving states were selected in terms of visual field range, visual recognition, visual search, and visual load, which were the information entropy of fixation area (IEFA), saccade amplitude, peak-to-average ratio of saccade velocity (PARSV), and relative change intensity of pupil area (RCPA). Then, based on the improved CRITIC method, the visual stability coefficient (VSC) was constructed as a new indicator to comprehensively assess the risk level of the driving state, and the assessment criteria were divided. Finally, the grey correlation analysis method was introduced to verify the assessment effect of VSC. The results show that different cell phone call modes increased driving risk in both traffic conditions. Among them, the negative influence of video calls on driving safety was significantly higher than that of hands-free calls, with a significant decrease in VSC, and the drivers' VSC in the free flow scenario was more sensitive to the impact of cell phone call operation, and the driving risk increased significantly during distracted driving. The VSC can quantitatively assess driving risk from the perspective of visual psychological safety and contributes to the development of corresponding early warning and control measures. The present longitudinal study examined the role of performance monitoring in the relationships between stress (both stress change and chronic stress) and negative affect to a real-life stressor-final exam among 60 undergraduates. Participants firstly completed a Go/No-Go task in the laboratory with electroencephalogram recordings. At T1 (31 days before the final exam), participants reported their chronic perceived stress. Then, with the daily dairy method, their daily stress level and negative affect were collected for three consecutive days. At T2 (three days before the final exam), the 3-day daily dairy was repeated. Results showed that performance monitoring, as measured by behavioral adjustments and electrophysiological correlates, moderated the effects of stress change as well as chronic perceived stress on the negative emotional response to the final exam. More specifically, as the stress change from baseline to exam increases, individuals with shorter PES, lower PEAD or larger Pe amplitudes experienced less negative affect increases in response to exam. Additionally, individuals with shorter PES or larger Pe amplitudes showed no significant relationship between chronic stress and negative affect, whereas individuals with longer PES or smaller Pe amplitudes showed significant positive relationship between chronic stress and negative affect increases in response to exam. The results demonstrated that efficient performance monitoring is a protective factor for stress. • The study showed that performance monitoring moderated the effects of stress change as well as chronic perceived stress on the negative emotional responses to the final exam. • Taken the present study and previous studies together, it may be advisable to enhance error-related cognitive control among healthy adults to attenuate the stressors' negative effects. To explore the effect of snow-melting agents on the glass fiber-reinforced cemented soil under freezing-thawing cycles, three widely used snow-melting agents, including potassium acetate, magnesium chloride, and sodium sulfate, were used in this article. The effects of snow-melting agent types on the apparent damage, mass loss, and mechanical properties of fiber-reinforced cemented soil under freezing-thawing cycles were analyzed through salt freezing and unconfined compressive strength tests. The results show that the snow-melting effect of potassium acetate is the best, the snow-melting effect of magnesium chloride is the second, and the snow-melting effect of sodium sulfate is the worst. Notably, as the number of freezing-thawing cycles increases, the strength of the test block decreases to varying degrees. After the fifth freezing-thawing cycle, the strength of the block without fiber decreased by 61.30%, 70.22%, and 81.58% in clear water, potassium acetate, and magnesium chloride solution, respectively, while the test block in sodium sulfate solution lost its bearing capacity. A series of studies proved that the snow-melting agent with sodium sulfate as the main component has the most apparent erosion effect on the cemented soil, followed by magnesium chloride, and the erosion effect of potassium acetate is the weakest. The incorporation of glass fiber can effectively improve the resistance of the cemented soil under the action of various salt solution erosion and freezing-thawing coupling and has a significant effect on slowing the development of surface cracks, improving peak strength, and reducing the mass loss rate. Timely diagnosis of bearing faults can effectively predict initial faults and avoid severe accidents. Ordinary neural networks have achieved relatively high accuracy for bearing failure classification. However, for the convolution process in neural networks, convolution kernels of fixed size are used across the whole image acquired by the time-frequency transformation, which is prone to overlook the intrinsic local time-frequency features of data. So as to solve this problem that the kernel may not reflect the local time-frequency characteristics of the non-stationary signal, a new method for planning convolutional kernels is proposed in this paper. We leverage variable kernels to capture the time-frequency resolution altering nature within the non-stationary signals. Firstly, starting from the time-frequency characteristics of non-stationary signals, the theoretical basis of adopting variable convolution kernels is analyzed, and the impact of Heisenberg's measurement inaccuracy principle on the design of the learning framework is analyzed in-depth with wavelet analysis as an example. Secondly, since the performance of different wavelet basis functions on time-frequency resolution varies dramatically, after comprehensive study of the mutual relationship between these resolutions, a proper criteria is deduced to deliver the measurement of performances, and the Gabor wavelet basis is chosen according to this principle. This design can be obtained by using wavelet analysis parameters before learning and training. This method is orthogonal to the model training process and can be plug-and-play with other deep learning frameworks. Finally, it is shown through datasets and practical experiments that the proposed method outperforms other newly proposed classification methods in terms of achieving higher accuracy in less time. Histone deacetylase (HDAC)‐targeted probes and prodrugs are crucial for cancer theranostics. We developed a self‐immolative design that enables in vivo activatable near‐infrared fluorescence (NIRF) and photoacoustic (PA) imaging and prodrug release in response to HDAC. This design comprises a phenyl ester linker with tunable reactivity, facilitating efficient release of caged fluorophores/drugs upon deacetylation. We engineered a new fluorophore using a spirocyclic xanthene scaffold with ring‐open property, affording NIRF/PA detection with high contrast. We showed that a nitro‐substituted self‐immolative linker allows sensitive NIRF/PA in vivo imaging of HDAC with minimal interference. A highly efficient prodrug system was further developed for targeted therapy in HDAC‐overexpressed triple negative breast tumors in mice. The functions of covalent organic frameworks (COFs) can be tailored by covalently reticulating advanced molecular modules into well‐defined porous ordered materials. Herein, four COFs, USTB‐7–USTB‐10, are prepared from the solvothermal reaction of photoactive tetraaldehydes, 5,5″‐(benzo[c]‐[1,2,5]thiadiazole‐4,7‐diyl)diisophthalaldehyde and 5,5″‐(naphtho[2,3‐c][1,2,5]thiadiazole‐4,9‐diyl)diisophthalaldehyde, with p‐phenylenediamine and benzidine, respectively. Comprehensive studies of powder X‐ray diffraction, theoretical simulation, and pore size distribution disclose their isoreticular 2D dual porous structures. In contrast to benzo[c][1,2,5]thiadiazole‐based chromophore, employment of naphtho[2,3‐c][1,2,5]thiadiazole‐based tetraaldehyde enables enlarged conjugation systems for USTB‐9 and USTB‐10, rather than USTB‐7 and USTB‐8. This, in combination with the longer benzidine unit, endows USTB‐10 with a porous structure with bigger pore size than that of USTB‐9, resulting in the highest photocatalytic hydrogen production rate of 21.8 mmol g−1 h−1 with the help of a Pt cocatalyst. Experimental and theoretical studies reveal the outstanding photocatalytic activity for USTB‐10 among the four COFs associated with the narrowed bandgap and increased charge‐carrier separation efficiency. Sciatic nerve block is under investigation as a possible therapeutic strategy for neonatal injury-induced exaggeration of pain responses to reinjury. Spinal microglial priming, brain-derived neurotrophic factor (BDNF) and Src homology-2 domain-containing protein tyrosine phosphatase-2 (SHP2) participate in exaggerated incisional pain induced by neonatal incision. However, effects of sciatic nerve block on exacerbated incisional pain and underlying mechanisms remain unclear. Here, we demonstrated that sciatic nerve block alleviates pain hypersensitivity and microglial activation in rats subjected to neonatal incision and adult incision (nIN-IN). Chemogenetic activation or inhibition of spinal microglia attenuates or mimics effects of sciatic nerve block on pain hypersensitivity, respectively. Moreover, α-amino-3-hydroxy- 5-methy- 4-isoxazole propionate (AMPA) receptor subunit GluA1 contributes to the exaggeration of incisional pain. The inhibition of BDNF or SHP2 blocks upregulations of downstream molecules in nIN-IN rats. Knockdown of SHP2 attenuates the increase of GluA1 induced by injection of BDNF in adult rats with only neonatal incision. The inhibition of microglia or ablation of microglial BDNF attenuates upregulations of SHP2 and GluA1. Additionally, sciatic nerve block downregulates the expression of these three molecules. Upregulation of BDNF, SHP2 or AMPA receptor attenuates sciatic nerve block-induced reductions of downstream molecules and pain hypersensitivity. Microglial activation abrogates reductions of these three molecules induced by sciatic nerve block. These results suggest that decreased activation of spinal microglia contributes to beneficial effects of sciatic nerve block on the neonatal incision-induced exaggeration of incisional pain via downregulating BDNF/SHP2/GluA1-containing AMPA receptor signaling. Background: There is a growing interest in exploring the neurocognitive mechanisms that may underlie psychological resilience. However, how the bottom-up automatic information processing relates to trait resilience has received less attention. We aimed to explore the relationship between trait resilience and trait-like automatic information processing in healthy adults.Methods: Eighty-four healthy adults were recruited to explore whether and how resilience was related to sensory sensitivity by event-related potentials (ERPs). Resilience was measured by Connor-Davidson Resilience Scale (CD-RISC). Sensory sensitivity, more specifically, sensitivity of automatic mismatch detection was measured by two ERPs components, i.e., the mismatch negativity (MMN) with a passive auditory oddball paradigm and the error-related negativity (ERN) with an auditory Go/NoGo task. Using the multiple linear regression analyses, the relationship between self-reported resilience and the sensitivity of automatic mismatch detection (MMN/ERN amplitude/latency) was explored.Results: The results showed that psychological resilience was positively correlated with both MMN and ERN latencies, i.e., higher resilience scores were associated with delayed MMN and ERN latencies. However, resilience was not significantly correlated with MMN and ERN amplitudes.Conclusions: Our results suggested that relatively higher resilience might link with less sensory sensitivity as reflected by slower automatic detection to mismatch information in the environment. This study aimed to investigate the role of JMJD2A in radiotherapy tolerance of esophageal squamous cell carcinoma (ESCC). The levels of H3K9me3 modification were analyzed in anti-PD-1 therapy non-responder or responder patients, and the expression differences of H3K9me3-related modifying enzymes were assessed in TCGA-ESCC and ICGC cohorts. Subsequently, JMJD2A was knocked down in ESCC cells using CRISPR-Cas9 or lentivirus-mediated shRNA, and changes in malignant behavior of ESCC cells were observed. RNA-seq, ATAC-seq, and ChIP-seq analyses were then conducted to investigate the genes and downstream signaling pathways regulated by JMJD2A, and functional validation experiments were performed to analyze the role of downstream regulated genes and pathways in ESCC malignant behavior and immune evasion. JMJD2A was significantly overexpressed in ESCC and anti-PD-1 therapy non-responders. Knockdown or deletion of JMJD2A significantly promoted the malignant behavior and immune evasion of ESCC. JMJD2A facilitated the structural changes in chromatin and promoted the binding of SMARCA4 to super-enhancers, thereby inducing the expression of GPX4. This resulted in the inhibition of radiation-induced DNA damage and cell ferroptosis, ultimately promoting the malignant behavior and immune evasion of ESCC cells. JMJD2A plays an indispensable role in the malignant behavior and immune evasion of ESCC. It regulates the binding of SMARCA4 to super-enhancers and affects the chromatin's epigenetic landscape, thereby promoting the expression of GPX4 and attenuating iron-mediated cell death caused by radiotherapy. The morphology of lithium deposition plays a crucial factor in determining the performance of lithium metal batteries. In order to understand the lithium plating and stripping process, the in-situ cell suitable for soft X-ray microscopy was designed and fabricated. We investigated the plating modes of mossy lithium and the interactions among surrounding grains. Furthermore, we established correlations between the lithium stripping process and the morphology of lithium grains, and thus summarized four stripping modes. The surface stripping and top stripping allow for complete stripping, while the side stripping and root stripping result in the formation of "dead lithium". In this study, the effectiveness and use of bone cement are thoroughly elaborated, and the role of bone cement on percutaneous vertebroplasty (PVP) fixed joints and its distribution on PVP are also elucidated. The aim of this study was to investigate the effect of unilateral and bilateral bone cement distribution on the clinical efficacy of PVP in the treatment of osteoporotic vertebral compression fractures (OVCF) of the thoracolumbar spine. A total of 60 patients with thoracolumbar OVCF (T11-L2) hospitalized in our hospital from January 2020 to January 2021 were studied. All patients had thoracolumbar OVCF. Under the guidance of the C-arm machine, unilateral PVP was performed. According to the distribution of bone cement across the midline, the patients were divided into two groups: the unilateral group (37 cases): bone cement was distributed on one side of the midline of the vertebral body, and the bilateral group (23 cases): bone cement was distributed on both sides of the midline. Visual analogue scale (VAS), vertebral height recovery values, and preoperative and postoperative Cobb's angle were recorded at 3 days, 1 month, 3 months, and 6 months. The differences between the two groups were compared and analyzed to evaluate the clinical efficacy. There was a statistically significant difference in VAS scores between the two groups before and after surgery (P < 0.05), but there was no statistically significant difference in VAS scores between the two groups at 3 days, 1 month, 3 months, and 6 months after surgery (P > 0.05). Purpose: Dysregulation of long non-coding RNAs (lncRNAs) is being found to have relevance to human cancers, including breast cancer (BC). The aim of this study was to further explore the functional role and molecular mechanisms of small nucleolar RNA host gene 14 (SNHG14) on BC progression.Methods: The expression levels of SNHG14, miR-543, and krüppel-like factor 7 (KLF7) mRNA were determined by quantitative real-time PCR. Western blot analysis was used to evaluate KLF7 protein level. Cell proliferation, apoptosis, and migration and invasion abilities were detected by Cell Counting kit-8 assay, flow cytometry, and transwell assay, respectively. The direct interactions between miR-543 and SNHG14 or KLF7 were confirmed using dual-luciferase reporter assays.Results: Our data indicated that SNHG14 expression was increased in BC tissues and cells, and SNHG14 knockdown mitigated the proliferation, migration, and invasion and facilitated apoptosis of BC cells. SNHG14 directly interacted with miR-543. MiR-543 mediated the regulatory effects of SNHG14 silencing on BC cell behaviors. Moreover, KLF7 was a direct target of miR-543. Overexpressed miR-543-mediated anti-proliferation, anti-migration, anti-invasion, and pro-apoptosis effects were mediated by KLF7. Furthermore, SNHG14 modulated KFL7 expression through acting as a competing endogenous RNA (ceRNA) of miR-543 in BC cells.Conclusion: Our study suggested that SNHG14 knockdown hindered BC progression in vitro at least partly through acting as a ceRNA of miR-543 and modulating KLF7 expression, providing evidence for SNHG14 as a potential target for BC therapy. This paper presents a fiber-level finite element model (FE) based on the digital element approach (DEA) for simulating the braiding process and predicting the geometry of tubular braided ropes. The braided yarn is modeled as a bundle of virtual fibers, using chains of truss (rod) elements. In the simulation of the braiding process, a penalty stiffness optimization method is proposed to solve the fiber penetration. The optimization studies show that fiber penetration caused by the combination of insufficient standard penalty stiffness and different number of fibers in the discretized yarn are effectively eliminated until the penetration rate dropped close to 1.85 %. The mesoscopic geometry predictions of models with four kinds of pitch lengths (24 mm, 36 mm, 48 mm, and 60 mm) are validated by quantitative comparison with micro-computed tomography (micro-CT) scans of braided polyarylate fiber ropes. It is shown that geometric convergence of the models can be achieved when each yarn contains 30 fibers. Corrosion of metallic materials can have a detrimental impact on their performance, leading to a shortened service life of engineering equipment and posing risks to both people and the environment. This paper presents a novel epoxy resin (EP)-based composite coating (PFANI-GO/EP) that offers triple corrosion protection functions: barrier, passivation, and corrosion inhibition. Perfluorooctanoic acid-doped polyaniline modified graphene oxide (PFANI-GO) demonstrates superhydrophobicity, effectively enhancing the coating's barrier ability against corrosive substances. The combination of PANI and perfluorooctanoic acid (PFOA) in composite materials aids in the formation of a passivation film and hydrophobic layer, which prevent metal corrosion. After a 15-day immersion in brine, the PFANI-GO/EP coating demonstrates the highest impedance modulus of 1 × 109 Ω·cm2 at 0.01 Hz, compared to pure EP coating, GO/EP coating, and PFANI/EP coating. An open cluster is an ideal region to study the evolution of stars. In this work, we use Gaia Early Data Release 3 (Gaia EDR3) to derive the fundamental parameters of 30 faint open clusters listed in the catalogue given by Cantat-Gaudin et al. (2018, A&A, 618, A93), but the G magnitude of all of the member stars of that catalogue is brighter than ∼18 mag. This catalogue does not provide isochrone fitting parameters and spatial structure parameters. We acquired the member stars of 30 open clusters using the Density-Based Spatial Clustering of Applications with Noise algorithm in Gaia EDR3. The G magnitude of the member stars using our method can be found down to ∼21 mag. The G -band, G BP-band, and G RP-band data of the member stars construct a good color–magnitude diagram, which can further ensure the precision of isochrone fitting. Federated learning (FL), celebrated for its privacy‐preserving features, has been revealed by recent studies to harbour security vulnerabilities that jeopardize client privacy, particularly through data reconstruction attacks that enable adversaries to recover original client data. This study introduces a client‐level handwriting forgery attack method for FL based on generative adversarial networks (GANs), which reveals security vulnerabilities existing in FL systems. It should be stressed that this research is purely for academic purposes, aiming to raise concerns about privacy protection and data security, and does not encourage illegal activities. Our novel methodology assumes an adversarial scenario wherein adversaries intercept a fraction of parameter updates via victim clients’ wireless communication channels, then use this information to train GAN for data recovery. Finally, the purpose of handwriting imitation is achieved. To rigorously assess and validate our methodology, experiments were conducted using a bespoke Chinese digit dataset, facilitating in‐depth analysis and robust verification of results. Our experimental findings demonstrated enhanced data recovery effectiveness, a client‐level attack and greater versatility compared to prior art. Notably, our method maintained high attack performance even with a streamlined GAN design, yielding increased precision and significantly faster execution times compared to standard methods. Specifically, our experimental numerical results revealed a substantial boost in reconstruction accuracy by 16.7%, coupled with a 51.9% decrease in computational time compared to the latest similar techniques. Furthermore, tests on a simplified version of our GAN exhibited an average 10% enhancement in accuracy, alongside a remarkable 70% reduction in time consumption. By surmounting the limitations of previous work, this study fills crucial gaps and affirms the effectiveness of our approach in achieving high‐accuracy client‐level data reconstruction within the FL context, thereby stimulating further exploration into FL security measures. Contact binary stars are important research objects in astrophysics. The calculation speed of deriving the parameters of contact binaries with the Wilson–Devinney program and the Phoebe with Markov chain Monte Carlo (MCMC) program is relatively slow. It is unrealistic to derive the parameters in batches with the program for sky survey data. We obtain a neural network model of supervised learning with the training of synthetic light curves with Phoebe. We calculate the parameters of eight special targets from the simulated data and the Kepler data. Then, we generate the new light curve to fit the light curve of the special target base on these parameters. The correlation index R 2 of the fitting result is more than 0.98. The method can be used to fit the target which has orbital inclinations greater than 50°. By fitting the Kepler data and the observed data on the ground, the method has a good generalization ability to these targets, which have some noise and some starspots. The calculation speed of one light curve with this method is less than seconds. We can derive the parameters quickly in batches to undertake some statistical work for sky survey data with the method. Background: The purpose of this study was to investigate the evolution and latest trends in the application of different polytrauma definitions in the past ten years of clinical research, by collecting published polytrauma study data. Method: We conducted Pubmed, Embase and Ovid searches of literature on polytrauma, published between January 2008 and December 2018. The title, author name, definition of polytrauma, publishing date, country, type of article, and the name and impact factor of journal were recorded and analysed. Results: Based on different definition of polytrauma, the selected articles were grouped into four categories: (1) definition of polytrauma based on ISS, (2) definition of polytrauma based on NISS, (3) definition of polytrauma based on AIS, (4) descriptive definition of polytrauma. The 321 selected articles were published in 56 journals, predominantly in European and American medical journals, led by Injury (n = 41), followed by Journal of Trauma and Acute Care Surgery (n = 27) and European Journal of Trauma and Emergency Surgery (n = 17). Germany had the largest number of publications, with 88 articles. All articles were clinical research. There were 88 multicentre studies and 233 single-centre studies. Conclusions: The definition of polytrauma in clinical research was still mainly based on ISS. The appearance of the "New Berlin Definition" provides a new direction for its development. The detection of plasma tau and its phosphorylation is technically challenging due to the relatively low sensitivity. However, in Alzheimer's disease and other tauopathies, it is hypothesized that tau in the biofluid may serve as a biomarker. In recent years, several ultrasensitive assays have been developed, which can successfully detect tau and its phosphorylation in various biofluids, and collectively demonstrated the prognostic and diagnostic value of plasma tau/phosphorylated tau. Here we have summarized the principle of four ultrasensitive assays newly developed suitable for plasma tau detection, namely single-molecule array, immunomagnetic reduction assay, enhanced immunoassay using multi-arrayed fiber optics, and meso scale discovery assay, with their advantages and applications. We have also compared these assays with traditional enzyme-linked-immunosorbent serologic assay, hoping to facilitate future tau-based biomarker discovery for Alzheimer's disease and other neurodegenerative diseases. Mammary glands infection via Gram-negative bacteria may cause infertility or reduced ovarian function. In the current study, a potential treatment for LPS-induced ovarian inflammation was developed. Propolis was loaded into chitosan nanoparticles and co-administered with menstrual blood stem cells (MenSCs) in mice infused with LPS. Various properties of propolis-loaded chitosan nanoparticles were evaluated using scanning electron microscopy, drug release assay, antibacterial assay, and radical scavenging assay. In vitro studies showed biocompatibility, anti-oxidative, and antibacterial properties of the developed propolis nanoformulation. In vivo study showed that mice treated with co-administration of propolis-loaded chitosan nanoparticles and MenSCs significantly increased the total ovarian follicle reserve in mice infused with LPS. Percentage of mature follicles in co-administration method was around 13.89 ± 1.72 %. Gene expression studies showed that the expression levels of inflammation related cytokines including IL6, IL8, IL-1β, and TNF-α were downregulated in this group compared with other groups. However, the expression levels of PTEN, AKT, FOXO3 did not show a significant difference between groups. The developed treatment may potentially considered as an approach for treating ovarian infection with gram-negative bacteria. OBJECTIVES: To examine pain and spirituality, demographic and clinical factors associated with pain and spirituality, the contribution of spirituality to experiences of pain over time, and how pain and spirituality relate to engagement with a caregiverdelivered intervention. SAMPLE & SETTING: Women with advanced breast cancer (N = 256) enrolled in a home-based randomized controlled trial of foot reflexology. METHODS & VARIABLES: Secondary analyses were conducted with baseline and postintervention data. Stepwise model building, linear mixed-effects modeling, and negative binomial regression were used. RESULTS: Participants who were younger, not married or partnered, not employed, or receiving hormonal therapy had increased odds of higher pain levels. Those who were older, non-White, or Christian had increased odds of higher spirituality. Spirituality's contribution to pain was not significant over time. IMPLICATIONS FOR NURSING: Women in this sample experienced moderate pain, on average, at baseline. Objective: To present the first live birth after uterine transplantation (UTx) in the People's Republic of China.Design: Case study.Setting: University hospital.Patient(s): A 22-year-old woman with Mayer-Rokitansky-Kuster-Hauser syndrome and previous surgery for vaginal reconstruction and UTx.Intervention(s): Endometrial preparation, frozen embryo transfer, pregnancy follow-up, and cesarean section.Main Outcome Measure(s): Results of in vitro fertilization, frozen embryo transfer, ultrasound measurements during pregnancy, rejection diagnosis and treatment, delivery, live birth, and histology of uterus.Results(s): Frozen embryo (cleavage stage) transfer started 1.5 years after UTx. The first embryo transfer (n = 2) resulted in a biochemical pregnancy. The second, third, and fourth embryo transfer (n = 2, 2, 3) did not result in pregnancy. The fifth embryo transfer (n = 3) resulted in pregnancy with two gestational sacs, but with spontaneous disappearance of one in early pregnancy. During early pregnancy three episodes of vaginal bleedings occurred (gestational weeks 6 + 2, 13 + 1, and 16 + 3), with a spontaneously resorbing subchorionic hematoma diagnosed at the last bleeding episode. Bleeding episodes were treated with corticosteroids and tacrolimus. During pregnancy, blood flow velocity waveforms and fetal growth parameters were normal. A subacute cesarean section was performed at gestational week 33 + 6 due to uterine contraction pattern suggesting imminent labor. A healthy boy (2,000 g) with Apgar scores of 10, 10, 10 was delivered. The uterus was kept for a possible second pregnancy.Conclusion(s): The first live birth after UTx in the People's Republic of China is reported and this occurred after a robotic-assisted laparoscopic uterus retrieval from the mother. Background: MicroRNAs (miRNAs) play a role in regulating osteogenic differentiation (OD) of mesenchymal stem cells by inhibiting mRNAs translation under cyclic strain. miR-503-3p was downregulated in OD of human adipose-derived stem cells (hASCs) in vivo under cyclic strain in our previous study, while it might target the Wnt/β-catenin (W-β) pathway. In this study, we explored miR-503-3p's role in OD of hASCs under cyclic strain. Methods: OD of hASCs was induced by cyclic strain. Bioinformatic and dual luciferase analyses were used to confirm the relationship between Wnt2/Wnt7b and miR-503-3p. Immunofluorescence was used to detect the effect of miR-503-3p on Wnt2/Wnt7b and β-catenin in hASCs transfected with miR-503-3p mimic and inhibitor. Mimic, inhibitor, and small interfering RNA (siRNA) transfected in hASCs to against Wnt2 and Wnt7b. Quantitative real-time PCR (RT-PCR) and western blot were used to examine the OD and W-β pathway at the mRNA and protein levels, respectively. Immunofluorescence was performed to locate β-catenin. ALP activity and calcium were detected by colorimetric assay. Results: Results of immunophenotypes by flow cytometry and multi-lineage potential confirmed that the cultured cells were hASCs. Results of luciferase reporter assay indicated that miR-503-3p could regulate the expression levels of Wnt2 and Wnt7b by targeting their respective 3′-untranslated region (UTR). Under cyclic strain, gain- or loss-function of miR-503-3p studies by mimic and inhibitor revealed that decreasing expression of miR-503-3p could significantly bring about promotion of OD of hASCs, whereas increased expression of miR-503-3p inhibited OD. Furthermore, miR-503-3p high-expression reduced the activity of the W-β pathway, as indicated by lowering expression of Wnt2 and Wnt7b, inactive β-catenin in miR-503-3p-treated hASCs. By contrast, miR-503-3p inhibition activated the W-β pathway. Conclusions: Collectively, our findings indicate that miR-503-3p is a negative factor in regulating W-β pathway by Wnt2 and Wnt7b, which inhibit the OD of hASCs under cyclic strain. Osteoprotegerin (OPG), a secreted decoy receptor for receptor activator of nuclear factor B ligand (RANKL), plays an essential role in regulating bone resorption. While much is known about the function of the N-terminal domains of OPG, which is responsible for binding to RANKL, the exact biological functions of the three C-terminal domains of OPG remain uncertain. We have previously shown that one likely function of the C-terminal domains of OPG is to bind cell surface heparan sulfate (HS), but the in vivo evidence was lacking. To investigate the biological significance of OPG-HS interaction in bone remodeling, we created OPG knock-in mice (opgAAA). The mutated OPG is incapable of binding to HS but binds RANKL normally. Surprisingly, opgAAA/AAA mice displayed a severe osteoporotic phenotype that is very similar to opg-null mice, suggesting that the antiresorption activity of OPG requires HS. Mechanistically, we propose that the HS immobilizes secreted OPG at the surface of osteoblasts lineage cells, which facilitates binding of OPG to membrane-anchored RANKL. To further support this model, we altered the structure of osteoblast HS genetically to make it incapable of binding to OPG. Interestingly, osteocalcin- Cre;Hs2stf/f mice also displayed osteoporotic phenotype with similar severity to opgAAA/AAA mice. Background: Opsoclonus-myoclonus syndrome (OMS) is a rare neurological disease. Some children with OMS also have neuroblastoma (NB). We and others have previously documented that serum IgG from children with OMS and NB induces neuronal cytolysis and activates several signaling pathways. However, the mechanisms underlying OMS remain unclear. Here, we investigated whether nitric oxide (NO) from activated microglias and its cascade contribute to neuronal cytolysis in pediatric OMS.Methods: The activation of cultured cerebral cortical and cerebellar microglias incubated with sera or IgG isolated from sera of children with OMS and NB was measured by the expression of the activation marker, cytokines, and NO. Neuronal cytolysis was determined after exposing to IgG-treated microglia-conditioned media. Using inhibitors and activators, the effects of NO synthesis and its intracellular cascade, namely soluble guanylyl cyclase (sGC) and protein kinase G (PKG), on neuronal cytolysis were evaluated.Results: Incubation with sera or IgG from children with OMS and NB increased the activation of cerebral cortical and cerebellar microglias, but not the activation of astrocytes or the cytolysis of glial cells. Moreover, the cytolysis of neurons was elevated by conditioned media from microglias incubated with IgG from children with OMS and NB. Furthermore, the expression of NO, sGC, and PKG was increased. Neuronal cytolysis was relieved by the inhibitors of NO signaling, while neuronal cytolysis was exacerbated by the activators of NO signaling but not proinflammatory cytokines. The cytolysis of neurons was suppressed by pretreatment with the microglial inhibitor minocycline, a clinically tested drug. Finally, increased microglial activation did not depend on the Fab fragment of serum IgG.Conclusions: Serum IgG from children with OMS and NB potentiates microglial activation, which induces neuronal cytolysis through the NO/sGC/PKG pathway, suggesting an applicability of microglial inhibitor as a therapeutic candidate. The aim of this study was to examine the mediating role of stress in the association between people's perceived haze and negative emotions in daily life. Using ecological momentary assessment, 95 college students reported their perceived haze, stress, and negative emotions twice a day over the course of 2 weeks. The results showed a positive relationship between perceived haze and negative emotions. More importantly, this association was significantly mediated by levels of stress. Findings suggested that people who perceived more severe haze may report higher stress levels, which in turn may lead to increases in negative emotions. [ABSTRACT FROM AUTHOR]

Recent research suggests negative emotions result from haze. Yet there has been limited empirical work exploring the potential protective factors which positively influence emotional responses to perceived haze. Moreover, other research suggests dispositional mindfulness play an important role in reducing reactivity to threatening stimuli. The current study aimed to investigate the relation between perceived haze and negative emotions, and explored the moderation of dispositional mindfulness in this relation. Using Ambulatory Assessment (AA), 95 college students at the age of 21 monitored their perceived haze and negative emotions (e.g. depression, anxiety, anger) twice a day over 2 weeks. Dispositional mindfulness was assessed by Mindful Attention Awareness Scale (MAAS). Results showed that people who perceived more serious haze would have more negative emotions. Furthermore, dispositional mindfulness significantly moderated this association. The current study revealed that individuals with higher dispositional mindfulness may feel less depressed to haze, compared with individuals with lower dispositional mindfulness. While the majority of studies have focused on dispositional mindfulness and the efficacy of mindfulness meditation, there has been limited study exploring the effect of state mindfulness on emotions and its potential mechanism. Therefore, we conducted an ambulatory assessment study to monitor state mindfulness and emotions in daily life and examine the mediating effect of rumination in a natural context. One hundred Chinese university students rated their state mindfulness, rumination, and emotions (e.g., anxiety, depression, happiness) five times per day for a week. Using hierarchical linear models, we and found that at the within‐person level, higher current state mindfulness was associated with more subsequent positive emotions and less subsequent negative emotions. Current rumination partially mediated the relation between current state mindfulness and subsequent positive emotions (happiness, relaxation, contentment, interest) and negative emotions (depression, anger, boredom, sadness). At the between‐person level, higher state mindfulness was associated with more positive emotions and less negative emotions. Individuals' overall rumination partially mediated the relation between state mindfulness and emotions (depression, anger, boredom, sadness, relaxation). We concluded that higher state mindfulness was associated with lower rumination, which in turn resulted in more positive emotions and less negative emotions. Although forkhead box protein M1 (FOXM1) is markedly upregulated in human premalignant and oral squamous cell carcinoma (OSCC) tissues and cultured cells, the association of FOXM1 expression with OSCC prognosis is not well understood. The present study investigated the possible association of FOXM1 expression in patients with OSCC with their clinicopathological characteristics and clinical outcomes. The expression of FOXM1 protein in OSCC tissues from 119 patients was evaluated by immunohistochemistry, and the results demonstrated that FOXM1 overexpression in patients with OSCC was associated with tumour recurrence and poor prognosis. To study the in vitro effects of FOXM1, its expression was decreased by small interfering RNA (siRNA) in OSCC cell lines, and FOXM1 knockdown decreased the proliferative, migratory and invasive capacities of cells. FOXM1 inhibition by siRNA gave rise to reduced expression of vimentin and increased expression of E-cadherin. The present study reported FOXM1 as a novel predictor of tumour recurrence in patients with OSCC and its potential involvement in epithelial-mesenchymal transition in OSCC cells. Purpose: To study the effects of oxycontin combined with amitriptyline on patients with severe cancer pain and depression. Methods: Patients with severe cancer pain and depression (n = 46) were randomly divided into two groups. The study group was given oxycontin combined with amitriptyline, while the control group received only oxycontin, each over a period of 4 weeks. Analgesic effect and depression relief were evaluated using FPS-R (Faces Pain Scale-Revised) and HAMD17 (Hamilton Depression Rating Scale 17-Item). In addition, the dose of medication and adverse drug reactions were monitored. Results: Pain in patients in the two groups were significantly relieved after treatment (p < 0.01). Symptoms in the study group were significantly improved, when compared with the situation before treatment, while HAMD17 scores significantly decreased (p < 0.01). The mean dosage of oxycontin in the study group in the fourth week after treatment was significantly lower than that of the control group (p < 0.01). There were no obvious differences in adverse reactions between patients in the two groups. Conclusion: Oxycontin combined with amitriptyline effectively controls severe cancer pain, and ameliorates depression and improves the quality of life of patients with advanced cancer. A mutant L206F/P207F/L259F of Talaromyces thermophilus lipase (TTL) exhibited high hydrolytic activity towards 2-carboxyethyl-3-cyano-5-methylhexanoic acid ethyl ester (CNDE) for synthesis of (S)-2-carboxyethyl-3-cyano-5-methylhexanoic acid (S-CCMA), a key chiral intermediate of pregabalin. However, low conversion at high CNDE concentration and unreusability of the free TTL mutant restricted its industrial applications. In this study, the TTL mutant was immobilized onto epoxy resin and its catalytic properties for kinetic resolution of CNDE were investigated. Under the optimized conditions, the immobilized lipase exhibited an increased catalytic efficiency even at a CNDE concentration of 3 M with 49.7% conversion and 95% eep. The conversion retained higher than 46.3% even after 10 times repeated use of the immobilized lipase in n-heptane-water biphasic system. In this letter, molybdenum nitride (MoN) gate AlGaN/GaN HEMT has been investigated with MoN developed in various physical vapour deposition (PVD) conditions. The Schottky gate is studied through forward and reverse characteristics with different processed MoN, which is compared with the standard Ni gate. Neonatal injury-induced exaggeration of pain hypersensitivity after adult trauma is a significant clinical challenge. However, the underlying mechanisms remain poorly understood. Growing evidence shows that spinal Src homology-2 domain-containing protein tyrosine phosphatase-2 (SHP2) contributes to chronic pain in adult rodents. Here we demonstrated that the phosphorylation and expression of SHP2 in synaptosomal fraction of the spinal dorsal horn are elevated in adult rats subjected to neonatal and adult incisions (nIN-IN), and the upregulation of SHP2 is highly correlated with pain hypersensitivity. Intrathecal blockade of SHP2 phosphorylation using a SHP2 protein tyrosine phosphatase inhibitor NSC-87877, or knockdown of SHP2 by intrathecal delivery of small interfering RNA (siRNA), ameliorates mechanical allodynia and heat hyperalgesia in nIN-IN rats. Moreover, the expression of phosphatidylinositol 3-kinase (PI3K) in the spinal dorsal horn is significantly increased in nIN-IN rats. Intrathecal application of PI3K inhibitor, LY294002 or wortmannin, alleviates pain hypersensitivity in nIN-IN rats. Additionally, intrathecal administration of NSC-87877 or SHP2 siRNA attenuates the upregulation of PI3K. Finally, no alternation of SHP2 phosphorylation in the dorsal root ganglion and dorsal root of nIN-IN rats as well as PI3K expression in the dorsal root of nIN-IN rats intrathecally treated with NSC-87877 or SHP2 siRNA is observed. These results suggest that the phosphorylation and expression of SHP2 in the spinal dorsal horn play vital roles in neonatal incision-induced exaggeration of adult incisional pain via PI3K. Neonatal surgical injury exacerbates spinal microglial reactivity, modifies spinal synaptic function, leading to exaggerated pain hypersensitivity after adult repeated incision. Whether and how the alteration in microglial reactivity and synaptic plasticity are functionally related remain unclear. Previously, we and others have documented that spinal brain-derived neurotrophic factor (BDNF), secreted from microglia, contributes to long-term potentiation (LTP) in adult rodents with neuropathic pain. Here, we demonstrated that the mRNA and protein expression of spinal BDNF are significantly upregulated in adult rats subjected to neonatal incision and adult repeated incision (nIN-IN). Neonatal incision facilitates spinal LTP induced by BDNF or high frequency electrical stimulation after adult incision, including a decreased induction threshold and an increased magnitude of LTP. Coincidently, inhibition of spinal BDNF abrogates the LTP facilitation, alleviates the mechanical allodynia and thermal hyperalgesia in nIN-IN rats. By contrast, spinal application of exogenous BDNF in the adult rats with a single neonatal incision mimics the LTP facilitation and pain hypersensitivity, which have been found in nIN-IN rats. Exogenous BDNF-induced exacerbation of pain hypersensitivity could be blocked by BDNF inhibitor. In addition, blockade of microglial reactivity by intrathecal application of minocycline attenuates the elevation of BDNF and the LTP facilitation, and also, alleviates pain hypersensitivity in nIN-IN rats. In conclusion, spinal BDNF, at least partly derived from microglia, contributes to the neonatal incision-induced facilitation of spinal LTP and to the exacerbation of incisional pain in adult rats. This study explored whether dispositional mindfulness moderated the relationship between depression and posttraumatic growth. A total of 435 Chinese adolescents who experienced a tornado in Yancheng city reported their level of depressive symptoms (The Center for Epidemiologic Studies Depression Scale for Children; CES-DC), posttraumatic growth (Posttraumatic Growth Inventory; PTGI) and dispositional mindfulness (Mindfulness Attention Awareness Scale; MAAS). The current study found that for individuals with higher dispositional mindfulness, their depression was associated with more posttraumatic growth, while the individuals with lower dispositional mindfulness showed no such association. These findings suggest that individuals high in dispositional mindfulness possess the potential to benefit from the process of adaptive coping in response to depression following a traumatic experience. The scissile fatty acid binding site of lipases is divided into different sub-groups and plays an important role in the catalytic properties of the enzymes. In this study, the Talaromyces thermophilus lipase was engineered by altering its crevice-like binding site for efficient synthesis of chiral intermediate of Pregablin through kinetic resolution of 2-carboxyethyl-3-cyano-5-methylhexanoic acid ethyl ester (CNDE). The substitution of residues located at the crevice-like binding site with phenylalanine (Phe) resulted in significantly increased hydrolysis activity. The variant L206F/P207F/L259F exhibited a 37.23-fold and 47.02-fold improvement in the specific activity and turnover number ( k cat ) toward CNDE, respectively. Simultaneously, the optimum temperature and substrate preference were both altered in the variants. The study herein successfully engineered the TTL with improved catalytic properties for efficient biosynthesis of Pregablin intermediate. Toll-like receptors (TLRs) are one of the most important innate immune receptors, which recognize various pathogen-associated molecular patterns and activate the downstream immune response. Mouse TLR13 has been found to recognize a highly conserved sequence from bacterial or viral RNA and activate the myeloid differentiation primary response gene 88-dependent signaling response. The function of teleost tlr13 is still not fully understood, especially its relationship with bacterial RNA. In our study, we identified and characterized a tlr13 from Epinephelus coioides (orange-spotted grouper). The full-length cDNA of Eco. tlr13 contained a 2844 bp open reading frame, encoding 947 amino acids. The polypeptide was constitutive of a signal peptide, 13 leucine-rich repeats domains, a C-terminal leucine-rich repeats, a transmembrane domain and a conserved Toll/interleukin (IL)-1 receptor domain, indicating that Eco. Tlr13 exhibited a typical TLR structure. Multiple alignments showed that the Toll/IL-1 receptor domain of Eco. Tlr13 was identical with other homologues, and the phylogenetic tree suggested that Eco. Tlr13 was clustered with other TLR13s and had the closest relationship with predicted Lates calcarifer (sea bass) Tlr13. Subcellular localization analysis revealed that Eco. Tlr13 colocalized with the endoplasmic reticulum and early endosome. Moreover, Eco. tlr13 was broadly observed in all tested tissues with the relatively high expressions in the brain and immune-related tissues. After challenged with 19-mer Staphylococcus aureus 23S ribosomal RNA-derived oligoribonucleotide (ORN Sa19), the expression of Eco. tlr13 was significantly up-regulated in grouper spleen cells. Also, the luciferase assay further revealed that with the overexpression of Eco. Tlr13 in human embryonic kidney 293T cells, ORN Sa19 activated the promoter activity of interferon-β in a dose-dependent pattern. TLR22, a typical member of the fish-specific TLRs, is a crucial sensor in virally triggered innate immune signalling retained from natural selection. To elucidate the role of the TLR22-specific signalling cascade mechanism, we provide evidence that the double-stranded (ds) RNA-sensor TLR22 positively regulates the ERK pathway and negatively regulates the JNK, p38 MAP kinase and NF-κB pathway. Here, we show that TLR22 restrains NF-κB activation and IFN (interferon) β and AP-1 (activator protein-1) promoter binding (impairing “primary response” genes (TNF and IL-1)), induces “secondary response” genes (IL-12 and IL-6) and mediates the irregular expression of inflammatory genes. Therefore, TLR22 promotes ERK phosphorylation but impairs the JNK and p38 MAP kinases and IκB phosphorylation. Additionally, TLR22 controls the excessive generation of reactive oxygen species (ROS) to avoid damaging the organism. The specific kinetics of TLR22 depends on its distinct cellular localization. We demonstrate that TLR22 is an intracellular receptor localized in the endosome, and the TLR22-TIR domain is the functional structure inducing the signalling cascade post-viral replication in the body. As mentioned above, our data reveal a novel mechanism whereby TLR22-induced positive adjustment and negative regulation evolved independently to avoid harmful and inappropriate inflammatory responses.
