Background: Many educational disciplines, especially in health professions, are required by accrediting bodies to introduce or include health informatics (HI); however, faculty (academics) teaching this content may not be well prepared. Objective: The aim of this project was to explore how a doctoral physical therapy (DPT) program could more accurately represent compliance with HI accreditation requirements by identifying hidden instances of relevant content within the curriculum. Method: This exploratory, mixed methods, action research study utilised document review, questionnaires and interviews in the collection of quantitative and qualitative data to enable assessment of knowledge about HI, to determine if HI content was already incorporated in current courses, and, whether the content was accurately reflected in course student learning objectives (SLOs). Change in faculty understanding of HI as a result of this study was also assessed. Results: Of 16 DPT faculty, 13 participated in the pre-interview questionnaire; 8 (50%) representing 22/39 courses participated in the full study. Most were unfamiliar with HI and had unrecognised HI already incorporated in their courses leading to several SLO revisions and additions. Interview and post-interview questionnaire results documented significant increases in HI understanding among faculty. Conclusion: Physical therapy and HIM collaboration was successful in identifying HI content hiding in plain sight. Results revealed multiple instances of unrecognised HI content across the DPT curriculum. Revised and newly added SLOs, with others likely to follow due to this study, will assist faculty with future reaccreditation and in preparing graduates to more fully utilise HI in today's digital healthcare environments. Purpose: The usefulness of closed suction drains (CSD) after open reduction and internal fixation (ORIF) of tibial plateau fractures is a contested topic. The purpose of this study was to examine the impact of CSD in postoperative outcomes after tibial plateau fracture. Methods: Data were retrospectively collected from patients who underwent primary repair of closed tibial plateau fractures via an anterolateral approach between June 2021 to May 2022 at a single academic center. Fifty-six patients were included and 28 received CSDs at time of surgery. P values less than 0.05 were considered significant. Results: Fifty-six patients were included. There was no significant difference in demographics, pre- and post-op hemoglobin, estimated blood loss during surgery, length of stay, postoperative MMEs and pain at 3 month follow-up, deep vein thrombosis (DVT), compartment syndrome, flexion contracture, use of incisional vac, infection rate, wound drainage, hematoma, neurologic pain, dehiscence, additional surgery, or range of motion at 3 months follow-up. We noted a significant difference in Defense and Veterans Pain Rating Scale (DVPRS) on POD1, demonstrating greater pain in those in the CSD group. Conclusion: Our findings suggest that the use of CSD in ORIF of tibial plateau fractures may not be of significant prophylactic benefit. Summary Gene set scoring (or enrichment) is a common dimension reduction task in bioinformatics that can be focused on the differences between groups or at the single sample level. Gene sets can represent biological functions, molecular pathways, cell identities, and more. Gene set scores are context dependent values that are useful for interpreting biological changes following experiments or perturbations. Single sample scoring produces a set of scores, one for each member of a group, which can be analyzed with statistical models that can include additional clinically important factors such as gender or age. However, the sparsity and technical noise of single-cell expression measures create difficulties for these methods, which were originally designed for bulk expression profiling (microarrays, RNAseq). This can be greatly remedied by first applying a smoothing transformation that shares gene measure information within transcriptomic neighborhoods. In this work, we use the nearest neighbor graph of cells for matrix smoothing to produce high quality gene set scores on a per-cell, per-group, level which is useful for visualization and statistical analysis. Development of a growth factor delivery vehicle providing appropriate temporal-spatial release together with an appropriate preclinical large animal model to evaluate bone formation is critical in the development of delivery strategies for bone tissue regeneration. Smectite nanoclays such as LAPONITE™ possess unique thixotropic and protein retention properties offering promise for use in growth factor delivery in bone repair and regeneration. This study has examined bone formation mediated by a clinically approved growth factor delivery system (InductOs®) in combination with Laponite gel in an aged female ovine femoral condyle defect preclinical model (10 weeks). Two different designs, one containing a low volume of Laponite gel (LLG) in combination with the InductOs® absorbable collagen sponge (ACS), the other in which Laponite gel formed the implant (HLG), were compared against InductOs® alone and an autograft positive control. Thus, five groups: (i) empty defect, (ii) autograft, (iii) BMP2 + ACS, (iv) BMP2 + ACS + LLG and (v) BMP2 + HLG + ACS were examined in 9 mm × 12 mm defects performed bilaterally in the medial femoral condyles of 24 aged (>5 years) sheep. Bone formation within the defect was assessed using micro-computed tomography (micro-CT), digital volume correlation (DVC) for biomechanical characterisation as well as histology. The autograft and InductOs® mediated enhanced bone formation (p < 0001) compared to blank controls, while no significant differences were observed between the Laponite/Collagen/BMP delivery vehicles. However, the current study illustrated the excellent biocompatibility of Laponite and its ability to deliver localised active BMP-2, with the opportunity for improved efficacy with further optimisation. Interestingly, DVC-computed strain distributions indicated that the regenerated bone structure is mechanically adapted to bear external loads from the early remodelling stages of the bone reparation cascade. The current studies of selected nanoclay delivery platforms for BMP, assessed in a clinically relevant large animal model auger well for the development of bone fracture therapeutics for an ageing population. Supplemental Digital Content is Available in the Text. BACKGROUND: Although vitiligo is often treated medically, there is increasing evidence for surgical therapies. Overlap with in-office surgical therapies that are already employed for other dermatologic conditions suggest that there is a significant opportunity to expand dermatologists' therapeutic repertoire for vitiligo. OBJECTIVE: To systematically review the efficacy of nonphototherapy surgical treatments for vitiligo in comparative or placebo-controlled trials. METHODS: A systematic review for surgical treatments for vitiligo was conducted. Primary outcomes were treatment success (>75% repigmentation) and failure (<25% repigmentation) for which meta-analyses were performed. Adverse effects were noted. The Cochrane risk of bias tool was used to assess study quality. RESULTS: Surgical treatments reviewed included platelet-rich plasma, microneedling, ablative therapies, and surgical modalities. Seventy-three studies with 2,911 patients were included. The repigmentation benefits and adverse events are summarized. Meta-analyses suggest benefits for ablative laser therapies or microneedling in combination with narrowband ultraviolet B (NB-UVB) and for suction blister epidermal grafting over punch grafting. CONCLUSION: The addition of microneedling or ablative laser therapy to NB-UVB phototherapy may improve repigmentation with minimal adverse effects. Surgical therapies, such as suction blister grafting and punch grafting, may offer the highest likelihood of repigmentation but have a risk of adverse effects including scarring and hyperpigmentation. Changing environmental conditions are forcing natural resource managers and communities to adapt their strategies to account for global shifts in precipitation, temperature, sea level and more, all of which are occurring in addition to local human impacts. Adapting to threats from climate change requires a fundamental shift in the practice of natural resource management through the development of forward-looking "climate-smart" goals and strategies. Here we present a proof-of-concept application of a decision-support tool to help design climate-smart management actions for the watershed and coral reef management plan for Guánica Bay watershed in southwest Puerto Rico. We also explore the connection between adaptation planning and coral reef resilience, using a recently developed Puerto Rico-wide reef resilience assessment. In the first phase of the study, we used the publicly available Adaptation Design Tool to draft initial climate-smart versions of twelve proposed management actions. In the second phase, two actions (dirt road management on steep slopes, and coral reef restoration) were further refined through consultations with local experts to make more detailed design adjustments; this included the option to use information from the coral reef resilience assessment to inform design improvements. The first phase resulted in moderately detailed assessments that broadly accounted for anticipated direct and indirect effects of climate change on the planned management actions. The second phase resulted in more site-specific technical assessments and additional important design details. The expert panel charged with discussing climate-smart reef restoration around Guánica used the reef resilience assessment to guide discussion of reef restoration, highlighting the importance of having such information available for adaptation planning. This study demonstrates how climate change impacts can be effectively incorporated into a management plan at the most granular level of planning and how a structured, formalized process can be as valuable as the resulting adaptation information. Introduction: Hundreds of children suffer burn injuries each day, yet care guidelines regarding the need for acute inpatient treatment vs outpatient follow-up vs no required follow-up remain nebulous. This gap in the literature is particularly salient for the emergency clinician, who must be able to rapidly determine appropriate disposition. Methods: This was a retrospective review of patients presenting to a Level II pediatric trauma center, January 1, 2017-December 31, 2019, and discharged with an International Classification of Diseases, Rev 10, burn diagnosis. We obtained and analyzed demographics, burn characteristics, and follow-up data using univariate and bivariate analysis as well as logistic regression modeling. Patients were stratified into three outcome groups: group 1-patients who underwent emergent evaluation at a burn center or were admitted at their first follow-up appointment; group 2-patients who followed up at a burn center (as an outpatient) or at the emergency department (and were discharged home); and group 3-patients with no known follow-up. Results: A total of 572 patients were included in this study; 58.9% of patients were 1-5 years of age. Sixty-five patients met group 1 criteria, 189 patients met group 2 criteria, and 318 patients met group 3 criteria. Sixty-five percent of patients met at least one American Burn Association criteria, and 79% of all burns were second-degree burns. Flame and scald burns were associated with increased odds (odds ratio [OR] 1.21, OR 1.12) of group 1 vs group 2 + group 3 (P = 0.02, P < 0.001). Second/third-degree burns and concern for non-accidental trauma were also associated with increased odds of group 1 vs 2 or 3 (OR = 1.11, 1.35, P ≤ 0.001, 0.001, respectively). Scald burns were associated with increased odds of group 2 compared to group 3 (OR 1.11, P = 0.04). Second/third degree burns were also associated with increased odds of group 2 vs 3 (OR 1.19, P ≤ 0.001). Conclusion: There were few statistically significant variables strongly associated with group 1 (emergent treatment/admission) vs group 2 (follow-up/outpatient treatment) vs group 3 (no follow- up). However, one notable finding in this study was the association of scald burns with treatment (admission or follow-up) suggesting that the presence of a scald burn in a child may signify to clinicians that a burn center consult is warranted. The demand for high-resolution imaging, and high sample throughputs in LA-ICP-MS experiments has led to developing rapid-response ablation cells with low dispersion. These cells can achieve short transient signals called single pulse responses (SPRs), whose width is in the single-digit millisecond range at 1% of the maximum. However, coupled with ICP-Q-MS, recording those short signals poses a problem due to the sequential measurement of selected m/z ratios. If more than one m/z ratio is targeted, the time resolution of quadrupole detection systems is insufficient for accurately determining short SPRs. This work focuses on utilizing rapid response ablation cells for the analysis of multiple elements with quadrupole-based detection systems in the SPR mode. To achieve this, an ArF-excimer laser equipped with a rapid-response ablation chamber is coupled to an ICP-MS with a short settling time of 0.2 ms and below. The two naturally occurring Ag-isotopes were analyzed in the NIST SRM 612 to optimize the dwell times for ideal data acquisition. The data shows that the ratio of the dwell time and the settling time, plays a crucial role. With the optimized parameters, the natural ratio of 107Ag and 109Ag within a 10 ms FW0.01M transient signal could be determined. This concept of LA-ICP-MS was then applied for depth profiling analysis of Al-doped SiC, a wide bandgap semiconductor. This paper aims to understand the role of green entrepreneurs in urban sustainability transitions. We propose an analytical framework combining transition approaches and green entrepreneurship from a relational lens. It includes four processes: emergence of green entrepreneurs, multi-scalar interest coordination, empowering through anchoring, and struggling with the regime at the urban scale. This framework is illustrated through an empirical analysis of the role of green entrepreneurs in the development of the solar water heater industry in China's Solar City. Describes a two-part study of the research needs of graduate students at Georgetown University. The first part, conducted in May 2010, consisted of an online survey about students' research practices and opinions about the library. The second part, conducted in May 2011, consisted of focus groups with particular subsets of graduate students. The goals of the study were to: 1) gain insight into students' research habits; 2) determine how they use and perceive the library; 3) determine any differences in responses between students in different disciplines; and 4) uncover distinctions between “scholarly” students (those intent on careers in academia) and “professional” students (those intent on careers outside of academia). Survey responses and focus group transcripts reflected three common broad themes: 1) Infrastructure (library building, furniture, designated space for graduate students); 2) Awareness & Scholarly Support (awareness of specialized library resources and services, tailored orientation and instruction); and fcase3) Access Services (availability of resources on and off campus, interlibrary and consortial lending, borrowing policies). Humanities students were much more familiar with library services and resources than social science students. Background Mechanistic models, when combined with pertinent data, can improve our knowledge regarding important molecular and cellular mechanisms found in cancer. These models make the prediction of tissue-level response to drug treatment possible, which can lead to new therapies and improved patient outcomes. Here we present a data-driven multiscale modeling framework to study molecular interactions between cancer, stromal, and immune cells found in the tumor microenvironment. We also develop methods to use molecular data available in The Cancer Genome Atlas to generate sample-specific models of cancer. Results By combining published models of different cells relevant to pancreatic ductal adenocarcinoma (PDAC), we built an agent-based model of the multicellular pancreatic tumor microenvironment, formally describing cell type–specific molecular interactions and cytokine-mediated cell-cell communications. We used an ensemble-based modeling approach to systematically explore how variations in the tumor microenvironment affect the viability of cancer cells. The results suggest that the autocrine loop involving EGF signaling is a key interaction modulator between pancreatic cancer and stellate cells. EGF is also found to be associated with previously described subtypes of PDAC. Moreover, the model allows a systematic exploration of the effect of possible therapeutic perturbations; our simulations suggest that reducing bFGF secretion by stellate cells will have, on average, a positive impact on cancer apoptosis. Conclusions The developed framework allows model-driven hypotheses to be generated regarding therapeutically relevant PDAC states with potential molecular and cellular drivers indicating specific intervention strategies. Immune cell infiltration of tumors and the tumor microenvironment can be an important component for determining patient outcomes. For example, immune and stromal cell presence inferred by deconvolving patient gene expression data may help identify high risk patients or suggest a course of treatment. One particularly powerful family of deconvolution techniques uses signature matrices of genes that uniquely identify each cell type as determined from single cell type purified gene expression data. Many methods from this family have been recently published, often including new signature matrices appropriate for a single purpose, such as investigating a specific type of tumor. The package ADAPTS helps users make the most of this expanding knowledge base by introducing a framework for cell type deconvolution. ADAPTS implements modular tools for customizing signature matrices for new tissue types by adding custom cell types or building new matrices de novo, including from single cell RNAseq data. It includes a common interface to several popular deconvolution algorithms that use a signature matrix to estimate the proportion of cell types present in heterogenous samples. ADAPTS also implements a novel method for clustering cell types into groups that are difficult to distinguish by deconvolution and then re-splitting those clusters using hierarchical deconvolution. We demonstrate that the techniques implemented in ADAPTS improve the ability to reconstruct the cell types present in a single cell RNAseq data set in a blind predictive analysis. Globally increasing sea surface temperatures threaten coral reefs, both directly and through interactions with local stressors. More resilient reefs have a higher likelihood of returning to a coral-dominated state following a disturbance, such as a mass bleaching event. To advance practical approaches to reef resilience assessments and aid resilience-based management of coral reefs, we conducted a resilience assessment for Puerto Rico's coral reefs, modified from methods used in other U.S. jurisdictions. We calculated relative resilience scores for 103 sites from an existing commonwealth-wide survey using eight resilience indicators—such as coral diversity, macroalgae percent cover, and herbivorous fish biomass—and assessed which indicators most drove resilience. We found that sites of very different relative resilience were generally highly spatially intermixed, underscoring the importance and necessity of decision making and management at fine scales. In combination with information on levels of two localized stressors (fishing pressure and pollution exposure), we used the resilience indicators to assess which of seven potential management actions could be used at each site to maintain or improve resilience. Fishery management was the management action that applied to the most sites. Furthermore, we combined sites' resilience scores with projected ocean warming to assign sites to vulnerability categories. Island-wide or community-level managers can use the actions and vulnerability information as a starting point for resilience-based management of their reefs. This assessment differs from many previous ones because we tested how much information could be yielded by a "desktop" assessment using freely-available, existing data rather than from a customized, resilience-focused field survey. The available data still permitted analyses comparable to previous assessments, demonstrating that desktop resilience assessments can substitute for assessments with field components under some circumstances. Concentration of 25-hydroxyvitamin D3 (25(OH)D3), the main circulating form of vitamin D, is inversely associated with incident, sporadic colorectal adenoma risk. We investigated whether this association differs by 2 functional variants in the vitamin D–binding protein (DBP) gene, group-specific component (GC), that encode for common protein isoforms Gc1s, Gc1f, and Gc2 linked to differences in vitamin D metabolism. We pooled data (418 patients with adenoma and 524 polyp-free control subjects) from 3 colonoscopy-based case-control studies (Minnesota, 1991–1994; North Carolina, 1994–1997; South Carolina, 2002). We estimated 25(OH)D3–adenoma associations, stratified by DBP isoforms, using multivariable logistic regression. Higher 25(OH)D3 concentrations were inversely associated with colorectal adenoma risk among those with the Gc2 isoform (per 10-ng/mL increase in 25(OH)D3, odds ratio = 0.71, 95% confidence interval: 0.56, 0.90), but not among those with only Gc1 isoforms (odds ratio = 1.07, 95% confidence interval: 0.87, 1.32; P for interaction = 0.03). Thus, the vitamin D–incident, sporadic colorectal adenoma association may differ by common DBP isoforms, and patients with the Gc2 isoform may particularly benefit from maintaining higher circulating 25(OH)D3 concentrations for adenoma prevention. This paper outlines a research agenda focused on the development of "experience landscapes" for outdoor leisure practices. It uses the example of trail centres for mountain biking to highlight the importance of examining embodied experience and human-technology hybridity in understanding participation in active outdoor pursuits. We propose that such experience landscapes are being created as part of a broader shift towards an experience economy, as particular spaces are carefully and deliberately imagined and built to host specific activities and attract users. In the case of trail centres, we argue that this landscape is a co-production of actors involved in rural development, professional and volunteer "trail builders", changing bike equipment technologies and mountain bike riders themselves. The embodied and hybrid "experience" of mountain biking is itself transformed by being practised in experience landscapes. We argue that research focusing on understanding the process of co-producing experience landscapes will lead to valuable inputs to the continuing success of both trail centres and rural development strategies. Background: The application of different approaches calculating the anthropogenic carbon net flux from land, leads to estimates that vary considerably. One reason for these variations is the extent to which approaches consider forest land to be "managed" by humans, and thus contributing to the net anthropogenic flux. Global Earth Observation (EO) datasets characterising spatio-temporal changes in land cover and carbon stocks provide an independent and consistent approach to estimate forest carbon fluxes. These can be compared against results reported in National Greenhouse Gas Inventories (NGHGIs) to support accurate and timely measuring, reporting and verification (MRV). Using Brazil as a primary case study, with additional analysis in Indonesia and Malaysia, we compare a Global EO-based dataset of forest carbon fluxes to results reported in NGHGIs. Results: Between 2001 and 2020, the EO-derived estimates of all forest-related emissions and removals indicate that Brazil was a net sink of carbon (− 0.2 GtCO2yr−1), while Brazil's NGHGI reported a net carbon source (+ 0.8 GtCO2yr−1). After adjusting the EO estimate to use the Brazilian NGHGI definition of managed forest and other assumptions used in the inventory's methodology, the EO net flux became a source of + 0.6 GtCO2yr−1, comparable to the NGHGI. Remaining discrepancies are due largely to differing carbon removal factors and forest types applied in the two datasets. In Indonesia, the EO and NGHGI net flux estimates were similar (+ 0.6 GtCO2 yr−1), but in Malaysia, they differed in both magnitude and sign (NGHGI: -0.2 GtCO2 yr−1; Global EO: + 0.2 GtCO2 yr−1). Spatially explicit datasets on forest types were not publicly available for analysis from either NGHGI, limiting the possibility of detailed adjustments. Conclusions: By adjusting the EO dataset to improve comparability with carbon fluxes estimated for managed forests in the Brazilian NGHGI, initially diverging estimates were largely reconciled and remaining differences can be explained. Despite limited spatial data available for Indonesia and Malaysia, our comparison indicated specific aspects where differing approaches may explain divergence, including uncertainties and inaccuracies. Our study highlights the importance of enhanced transparency, as set out by the Paris Agreement, to enable alignment between different approaches for independent measuring and verification. The Influence Maximization Problem (IMP) aims to discover the set of nodes with the greatest influence on network dynamics. The problem has previously been applied in epidemiology and social network analysis. Here, we demonstrate the application to cell cycle regulatory network analysis for Saccharomyces cerevisiae. Fundamentally, gene regulation is linked to the flow of information. Therefore, our implementation of the IMP was framed as an information theoretic problem using network diffusion. Utilizing more than 28,000 regulatory edges from YeastMine, gene expression dynamics were encoded as edge weights using time lagged transfer entropy, a method for quantifying information transfer between variables. By picking a set of source nodes, a diffusion process covers a portion of the network. The size of the network cover relates to the influence of the source nodes. The set of nodes that maximizes influence is the solution to the IMP. By solving the IMP over different numbers of source nodes, an influence ranking on genes was produced. The influence ranking was compared to other metrics of network centrality. Although the top genes from each centrality ranking contained well-known cell cycle regulators, there was little agreement and no clear winner. However, it was found that influential genes tend to directly regulate or sit upstream of genes ranked by other centrality measures. The influential nodes act as critical sources of information flow, potentially having a large impact on the state of the network. Rapid urbanization has altered many ecosystems, causing a decline in many ecosystem services, generating serious ecological crisis. To cope with these challenges, we presented a comprehensive framework comprising five core steps for identifying and managing hotspots of critical ecosystem services in a rapid urbanizing region. This framework was applied in the case study of the Yangtze River Delta (YRD) Region. The study showed that there was large spatial heterogeneity in the hotspots of ecosystem services in the region, hotspots of supporting services and regulating services aggregately distributing in the southwest mountainous areas while hotspots of provisioning services mainly in the northeast plain, and hotspots of cultural services widespread in the waterbodies and southwest mountainous areas. The regionalization of the critical ecosystem services was made through the hotspot analysis. This study provided valuable information for environmental planning and management in a rapid urbanizing region and helped improve China's ecological redlines policy at regional scale. Green entrepreneurs have been seen as key drivers for a transition to a green economy. However, there has been limited in-depth qualitative empirical research with green entrepreneurs to date, focusing instead on typologies categorising certain ‘types’ of green entrepreneur. Moreover, the literature rarely situates such individual activities within broader concepts such as the green economy. In contrast, we suggest that current discourses of the green economy are important in contextualising the ways that green entrepreneurs make sense of themselves and their businesses. Green entrepreneurs are thus negotiating varying tensions between their business activities, environmental philosophies and wider contexts at the intersection between the green economy and the mainstream economy. Drawing on evidence from 55 interviews, we explore the narratives employed by green entrepreneurs to situate themselves within/outwith the wider green economy – the recursive framing of mainstream and niche ‘green’ activities provides a sense of the tensions and politics at play in the development of the green economy. We thus offer a new and more dynamic view of the evolving nature of ‘being’ and ‘becoming’ a green entrepreneur, rather than relying on the fixed categories espoused in previous typologies. We conclude that it is important that policy makers recognise the complex and contentious nature of green entrepreneurship, and that it is essential to view the green economy as a diverse constellation of myriad actors rather than corporate reinventions of business as usual. Motivation: Combining P-values from multiple statistical tests is a common exercise in bioinformatics. However, this procedure is non-trivial for dependent P-values. Here, we discuss an empirical adaptation of Brown's method (an extension of Fisher's method) for combining dependent P-values which is appropriate for the large and correlated datasets found in high-throughput biology. Results: We show that the Empirical Brown's method (EBM) outperforms Fisher's method as well as alternative approaches for combining dependent P-values using both noisy simulated data and gene expression data from The Cancer Genome Atlas. Background: Solar elastosis and neval remnants are histologic markers characteristic of divergent melanoma pathways linked to differences in age at onset, host phenotype, and sun exposure. However, the association between these pathway markers and newly identified low-penetrance melanoma susceptibility loci remains unknown.Methods: In the Genes, Environment and Melanoma (GEM) Study, 2103 Caucasian participants had first primary melanomas that underwent centralized pathology review. For 47 single-nucleotide polymorphisms (SNPs) previously identified as low-penetrant melanoma risk variants, we used multinomial logistic regression to compare melanoma with solar elastosis and melanoma with neval remnants simultaneously to melanoma with neither of these markers, excluding melanomas with both markers. All statistical tests were two-sided.Results: IRF4 rs12203592 was the only SNP to pass the false discovery threshold in baseline models adjusted for age, sex, and study center. rs12203592*T was associated positively with melanoma with solar elastosis (odds ratio [OR] = 1.47, 95% confidence interval [CI] = 1.18 to 1.82) and inversely with melanoma with neval remnants (OR = 0.65, 95% CI = 0.48 to 0.87) compared with melanoma with neither marker (P global = 3.78 x 10(-08)). Adjusting for phenotypic characteristics and total sun exposure hours did not materially affect rs12203592's associations. Distinct early- and late-onset age distributions were observed in patients with IRF4 rs12203592 [CC] and [TT] genotypes, respectively.Conclusions: Our findings suggest a role of IRF4 rs12203592 in pathway-specific risk for melanoma development. ABSTRACT This article is focused upon exploring the development of the green economy in particular locations, with the aim of identifying why some cities and regions have been successful in engendering green growth. To date we have little idea where the green economy is developing, nor much insight, beyond anecdotal evidence, into why certain cities and regions appear to be more successful than others in this regard. We position our analysis within the context of research on socio-technical transitions that has theorized the potential shift to a more sustainable economy. We review the literature on sustainability transitions and the development of the multi-level perspective encompassing niches, regimes and landscapes. However, most research into socio-technical transitions has not given adequate consideration to the influence of places and spatial scale in these transition processes, and we therefore critique the socio-technical transitions literature from a geographical perspective. In this article we are interested in four key questions. What role does the enabling and facilitative state play in these cities and regions? What new institutional forms and governance structures are being developed? How do actors in particular cities and regions construct their green vision, and how do they encourage other actors to buy-in to this vision? How are links across levels and spatial scales developed to connect niches with the regime? We address these through a focus upon the Boston city-region in the USA, drawing upon both primary and secondary research material. Making the transition to a green economy is a major policy driver in the UK and other countries. Entrepreneurs are suggested as being at the forefront of this transition and as a driving force for sustainability. These “green entrepreneurs” may represent a new type of entrepreneurial behaviour combining economic, environmental and social aims. In this paper, we present empirical work conducted with green entrepreneurs in the UK green building sector. Buildings have significant impacts on the environment, both in terms of materials and post-construction energy demands. Drawing on sustainability transitions theory, we examine the role of green entrepreneurs in affecting change and suggest that green building niches are less consensual than previously theorised. In theorising green entrepreneurs, we also point to the need to consider them within wider networks of activity rather than as lone actors and the implications this has for policy. When different life stages have different environmental tolerances, development needs to be regulated so that each life stage experiences environmental conditions that are suitable for it, if fitness is to be maintained. Restricting the timing of developmental transitions to occur under specific combinations of environmental conditions is therefore adaptively important. However, impeding development can itself incur demographic and fitness costs. How do organisms regulate development and physiological processes so that they occur under the broadest range of permissive conditions? Gene duplication offers one solution: Multiple genes contribute to the same downstream process, but do so under distinct combinations of environmental conditions. We present a simple model to examine how environmental sensitivities of genes and how gene duplication influence the distribution of environmental conditions under which an end process will proceed. The model shows that the duplication of genes that retain their downstream function but diverge in environmental sensitivities can allow an end process to proceed under more than one distinct combination of environmental conditions. This paper explores the development of green entrepreneurship and its potential role in transformative change towards a green economy. It achieves this through a study of the green building sector in England and Wales, based on qualitative empirical data from fifty-five semistructured interviews with businesses in the green building sector and with support organisations, including banks, financial sources, and business advice and support. The paper both critiques and synthesises two bodies of literature-- entrepreneurial research and sociotechnical transitions theories, specifically the multilevel perspective (MLP)--to better understand the role of green entrepreneurs in facilitating a shift towards a green economy. This analysis embeds green entrepreneurs in a wider system of actors, rather than reifying the lone entrepreneurial hero, in order to explore how green entrepreneurs facilitate sustainability transitions. The paper challenges the notion that green entrepreneurs are an unproblematic category. We discovered that individuals move between 'green' and 'conventional' business, evolving over time, such that this is a fluid and blurred, rather than static, state. Moreover, while the green economy and the green building sector are often referred to as coherent sectors, with agreed and consistent practices, our evidence suggests that they are far from agreed, that business models vary, and that there are significant contradictions within so-called green building practices. The paper contributes to the development of sociotechnical transitions theory and suggests that the MLP needs to incorporate complexity and multiplicity within niches, that niches may be inherently conflictual rather than consensual, and that the concept of 'protection' for niches is problematic. As urban centres of agglomeration expand and compete for investment, new demands may arise for additional housing, infrastructure, and services. Failure to meet these demands imposes costs on firms and workers, stifles expansion, and potentially compromises the long-run economic competitiveness of the growth area. Drawing on evidence from Germany (Munich), Sweden (Stockholm), and the UK (Cambridge) this paper examines the organisational and political challenges of growth facilitation in the context of post-Keynesian political and economic restructuring. Particular emphasis is placed on tensions arising from changes in the form and function of European state social regulation. These tensions are not simply a matter of neoliberal regulatory deficit but reflect broader societal cleavages in relation to the uneven spatial impact of local economic growth. Deploying the concept of territorial structures of growth facilitation provides a conceptual framework for taking forward research on the relationship between state spatial regulation, state restructuring, and the competitiveness of city-regions. This article explores economy-environment relations in urban areas through a focus upon the reorientation of development in the Boston city-region in Massachusetts around a vision of smart growth. The article draws upon the evolving tradition of critical economy-environment research from a broadly regulationist perspective. However, to date research has tended to mask the politics and mediations of the development agenda by actors locked in struggle to shape their social, economic and environmental landscapes. It is argued that reducing institutions to their stylized forms ignores the real politics associated with institutional formation and outcome. A more fruitful approach, we suggest, is to draw upon recent work on institutions and institutional change. In particular the article utilizes a decentered institutional analysis put forth by political scientists Mark Bevir and Rod Rhodes and investigates empirical data from secondary sources and interviews in the Boston city-region with reference to Bevir and Rhodes' three concepts of dilemmas, traditions and beliefs. The new urban politics (NUP) literature has helped to draw attention to a new generation of entrepreneurial urban regimes involved in the competition to attract investment to cities. Interurban competition often had negative environmental consequences for the urban living place. Yet knowledge of the environment was not very central to understanding the NUP. Entrepreneurial urban regimes today are struggling to deal with climate change and reductions in greenhouse gas emissions. Carbon reduction strategies could have profound implications for interurban competition and the politics of urban development. This paper explores the rise of a distinctive low-carbon urban polity—carbon control—and examines its potential ramifications for a new environmental politics of urban development (NEPUD). The NEPUD signals the growing centrality of carbon control in discourses, strategies and struggles around urban development. Using examples from cities in the US and Europe, the paper examines how these new environmental policy considerations are being mainstreamed in urban development politics. Industrial ecology is often promoted as a practice to close industrial production loops and reduce waste, thereby making better use of resources and preventing the overuse of raw materials. Eco-industrial parks (EIPs) are the spatial concentrations of businesses that use the by-products of one industrial process as inputs to another. Both sets of practices are based on particular tropes involving nature. This paper explores the use of various tropes in the literature of industrial ecology and EIPs. It highlights the need better to understand the pre-theoretical and theoretical bases of industrial ecology and to increase the potential for practical implementation by applying insights from the literatures of urban planning, social science and economic geography. Abstract: This paper investigates whether eco-industrial parks (EIPs) offer possibilities to implement sustainable development policies. EIPs are based upon industrial ecology principles that suggest industrial systems can operate in a similar fashion to natural ecological systems. Drawing upon a survey of all identifiable EIPs in the USA and Europe and in-depth interview surveys of 16 EIPs, the research discovered few examples of networking between firms or processes using wastes and materials recycling. Most EIPs are at a very early stage of development, where linkages are potential rather than real. The problems encountered in developing EIPs from scratch suggests that a more fruitful approach is to build upon existing and potential linkages within a locality, assisted by a pro-active policy to encourage interchanges. Demands for increased levels of protection for species and ecosystems are being translated into new modes of spatial governance at national, regional, and local scales. As part of this, new ways of analysing and representing ecological systems are leading to qualitatively different treatment of the (time) spaces of nature conservation. In this paper we explore what happens when the land-use demands of new paradigms of nature conservation collide with preestablished modes of spatial regulation, taking as our focus the implementation of the European Union Habitats Directive in estuary spaces in the United Kingdom. We provide evidence of the ways in which ecological regulation is reshaping political-economic landscapes and offer insights into the direction of ‘postmodern’ nature conservation practice, including the extent to which theoretical notions of hybrid and nonequilibrium systems have become absorbed into conflicts around ecological regulation. This paper focuses on investigating the role that sustainability entrepreneurship may have in engendering a shift in the practices and operations of contemporary capitalism. Sustainability entrepreneurs are increasingly seen as being in the vanguard of a shift to a new form of capitalist development that can help to address fears over global warming, climate change and their associated negative environmental impacts. Such developments can be set within a wider popular and academic discourse of ecological modernisation, at the heart of which is a relatively optimistic view of the potential for technological change to lead to solutions for environmental problems. This paper focuses on a subset of sustainable entrepreneurs termed 'ecopreneurs' who seek to combine business practice with sustainable development and so transform their business sectors. The paper suggests that work on sustainable entrepreneurship could be substantially improved by an engagement with the literature on transition management in science and technology studies and makes some suggestions as to how such a research agenda could be advanced. Abstract: Although the "new" economic geography has explored links between the subdiscipline's traditional areas of study and cultural, institutional, and political realms, environmental issues remain comparatively underresearched within the subdiscipline. This article contends not only that the environment is of key importance to economic geography, but also that economic geographers can make an important contribution to environmental debates, through providing not just a better analysis and theoretical understanding, but also better policy proscription. Rather than claim new intellectual territory, the intention is to suggest potential creative opportunities for linking economic geography's strengths with those insights from other theoretical perspectives. Abstract: A LC–tandem mass spectrometry method to quantify the quinazoline-based thymidylate synthase inhibitors BGC945 and BGC638 in mouse plasma was developed. BGC945 and BGC638 were extracted from mouse plasma using protein precipitation with acetonitrile. Chromatography was performed on a Fluophase RP 5μm, 100mm×2.0mm i.d. column using a gradient of ammonium acetate and acetonitrile as a mobile phase with a flow rate of 0.2mLmin−1. The injection volume for each sample was 20μL with a total run time of 7.5min. This method was validated in the range 25–4000nM (r 2 =0.99). The analytical assay performance showed that the method was accurate (mean intra- and inter-day assay R.E. were below 12% and 11%, respectively), reproducible (mean intra- and inter-day R.S.D. were less than 13% and 5% for all quality control levels, respectively) and sensitive (lower limit of quantification was 25nM) in the range studied. This validated method has been used to define the first pharmacokinetic report of BGC945 and BGC638 in mice. [Copyright &y& Elsevier]

This article analyzes the way that US foreign policy élites have used pretexts to manage public opinion. Policymakers, it is argued, often seize upon threatening external events, and use these events to create a favorable political climate in which to "sell" policies of militarization and external expansion to the public. The article argues that the Bush administration has used the threat of terrorism as a pretext to implement a wide range of policies that had been decided upon in advance of the 9/11 attacks. It also argues that the recent uses of pretexts by the Bush administration have strong historical precedents: extended case studies of pretexts are presented for the events surrounding the Korea crisis of 1950 and the Afghanistan crisis of 1979-1980, as well as the more recent War on Terrorism. Using an urban regime theory approach, the article aims to investigate the degree to which environmental policy in England is devolved to the local level of the state and integrates with local economic governance. Intensive case study research was undertaken in two local areas of the eastern region of England having divergent economic growth trajectories. Methods included analysis of local media and documentation from local and regional organizations, and 30 primary interviews with strategic local actors. The Cambridge Subregion area is experiencing pressures of economic development, creating conflict around the Green Belt and contested meanings of sustainable development. In Waveney—a declining area in search of growth—local groups have struggled to manage local economic regeneration in an environmentally sustainable manner. Both areas have witnessed new territorial-institutional developments in relation to environmental policy making, with limited evidence of policy integration at the subregional scale. Despite the emphasis on local policy integration through sustainable development, the relationship between the economy and the environment in England is a focus of conflict and new territorial-institutional developments. Urban regime theory needs to broaden its focus to include the full variety of interests in local environmental policy and the mechanisms producing new territorial scales of economic and environmental governance. Recent research on local and regional economic development has focused upon transformations in local governance and institutional capacity. It has been argued that local authorities have ceded power to other actors and institutions involved in economic development and regeneration, and that the success of local and regional economic development is closely related to the strength of ‘institutional capacity’ within an area. In this paper, we examine these claims with reference to the operation of EU Structural Funds in the Humber Sub-region of the UK. Previous research on local governance and institutional capacity has had a limited empirical focus, drawing conclusions from studies of either economically ‘successful’ regions or regions undergoing regulatory and institutional transformation and precluding analysis of the nature and conditions of local governance and institutional capacity in less developed regions. Our case study evidence not only suggests that arguments about the declining influence of the local state are overdrawn, but also indicates a need for more nuanced accounts of the role of institutional capacity in regional development.
