The elderly account for the majority of medical spending in many countries, raising concerns about potentially unnecessary spending, especially during the final months of life. Using a well-defined starting point (hospitalization for an initial acute myocardial infarction) with evidence-based postevent treatments, we examined age trends in treatments in the US and Norway, two countries with high levels of per capita medical spending. After accounting for comorbidities, we found marked decreases within both countries in the use of invasive treatments with age (for example, less use of percutaneous coronary interventions and surgery) and the use of relatively inexpensive medications (for example, less use of anticholesterol [statin] drugs for which generic versions are widely available). The treatment decreases with age were larger in Norway compared with those in the US. The less frequent treatment of the oldest of the old, without even use of basic medications, suggests potential age-related bias and a disconnect with the evidence on treatment value. Health-related quality of life (HRQoL) was measured with the 16D instrument, and quality-adjusted life-years (QALYs) were derived; total costs included the costs of therapy, and last we calculated the incremental cost-effectiveness ratio (ratio of differences in costs and QALYs gained). We performed nonparametric bootstrapping and used the results to draw a cost-effectiveness acceptability curve depicting the probability that TFCBT is cost-effective. HRQoL increased in both treatment groups, whereas no significant differences in QALYs were observed. Resource use measured in minutes per session was significantly higher in the TF-CBT group; however, total minutes of therapy and costs were not significantly different between the two groups. In addition, use of resources, such as psychological counseling services, welfare services, and medication, was lower in the TFCBT group posttreatment. The likelihood of TF-CBT being cost-effective varied from 91% to 96%. Background: Very preterm (VPT) children, with a birth weight below 1500 g or delivered before 32 weeks of gestational age, are at increased risk of poorer long-term health outcomes and higher rates of hospitalization in childhood. However, considerable variation exists in the need for in-hospital care within this population. We assessed the utilization and distribution of hospital-based care from ages 1 through 9 years for a nationwide population.Methods: This was a population-based cohort of VPT children born in the period 2001-2009. We evaluated their utilization of hospital care in 2008-2010, when aged 1-9 years old. Outcomes were the incidence of hospital admissions and outpatient visits. We used Poisson regression models with multiple imputation of missing data.Results: Children born VPT had more hospital admissions compared with the general population of children aged 1-9 years. The rates of hospital admissions and outpatient visits were strongly related to clinical characteristics of the child at birth and age at admission/outpatient visit but to only a variable and minor degree to characteristics pertaining to maternal health, the sociodemographic factors, and geographical proximity to hospital services.Conclusions: Prior to this study, hospital utilization during the period 5-9 years old has been poorly documented. We found that excess utilization of hospital resources on average declines with increasing age. We also noted substantial differences in the use of hospital care across age groups and clinical factors for VPT children. We study gatekeeping physicians’ referrals of patients to specialty care. We derive theoretical results when competition in the physician market intensifies. First, due to competitive pressure, physicians refer patients to specialty care more often. Second, physicians earn more by treating patients themselves, so refer patients to specialty care less often. We assess empirically the overall effect of competition with data from a 2008–2009 Norwegian survey, National Health Insurance Administration, and Statistics Norway. From the data we construct three measures of competition: the number of open primary physician practices with and without population adjustment, and the Herfindahl–Hirschman index. The empirical results suggest that competition has negligible or small positive effects on referrals overall. Abstract: Patients switching physicians involves costs to the patients because of less continuity of care. From a theoretical model we derive that inferior physician quality as perceived by patients, implies patient shortage for the physician and more patients switching physicians. By means of a unique panel data set covering the entire population of Norwegian general practitioners (GPs) and estimation methods that adjust for potential endogeneities, we find that the occurrence of patient shortage increases the proportion of patients switching physicians by 50%. A competing hypothesis that GPs with a shortage of patients experience less switching because of superior access is rejected by data. To assist patients in making informed decisions, we suggest that the number of switches a physician experiences should be made public. [Copyright &y& Elsevier]

Abstract: A central theme in the international debate on genetic testing concerns the extent to which insurance companies should be allowed to use genetic information when offering insurance contracts. We provide a welfare analysis of this issue within a model of an insurance market with asymmetric information, having the following crucial feature: in addition to a state-contingent consumption profile, a person's well-being depends on her attitude towards resolution of future health uncertainty, and this attitude varies across the population. We present stylized facts that motivate this approach. In the formal analysis, we find that both tested high-risks and untested individuals are equally well off whether or not test results can be used by insurers. Individuals who test for being low-risks, on the other hand, are made worse off by not being able to verify this to insurers. This implies that, in terms of welfare, a regulatory regime in which the use of genetic information by insurers is allowed is better than one in which it is not allowed. [Copyright &y& Elsevier]

When the insurer has access to information about test status, genetic insurance can handle the negative effects of genetic testing on insurance coverage and income distribution. Hence, efficient testing is promoted. When information about prevention and test status is private, two types of social inefficiencies may occur; genetic testing may not be done when it is socially efficient and genetic testing may be done although it is socially inefficient. The first type of inefficiency is shown to be likely for consumers with compulsory insurance only, while the second type of inefficiency is more likely for those who have supplemented the compulsory insurance with substantial voluntary insurance. This second type of inefficiency is more important the less effective prevention is. It is therefore a puzzle that many countries have imposed strict regulation on the genetic information insurers have access to. Enzymatic digestion of the cell wall of Brassica napus hypocotyls gave a heterogeneous suspension of protoplasts with the cortical microtubules (CMTs) randomly organised or CMTs organised in parallel. The effect of variable g-influences has been tested on CMT organisation. In contrast to the 1 g-protoplasts, which reorganised the CMTs into parallel arrays during the 96 h test period, the frequency of randomly-oriented CMTs in the protoplasts exposed to simulated weightlessness (0 g) on a 2-D clinostat increased significantly during the same period. The opposite effect was obtained when the protoplasts were exposed to hyper-g (7 or 10 g), where the reorganisation of the CMTs into parallel arrays was accelerated compared to the 1 and 0 g-protoplasts. These results indicate that a unidirectional gravity force is a necessity for the reorganisation of CMTs in protoplasts to parallel arrays and that CMTs act as responding elements that are able to sense different levels of gravity. Besides the inability of the protoplasts to reorganise the CMTs into parallel arrays, the quantity of CMTs in the individual protoplast decreased during 4 days of simulated weightlessness, both compared to the CMTs quantity in the protoplasts immediately after isolation and compared to the 1 g- and hyper-g-protoplasts after 24 and 48 h of g-exposure. The size of the protoplasts was also affected by the g-exposure. Protoplasts exposed to simulated 0 g increased significantly after 24 and 48 h, whereas the 1 g- and 10 g-protoplasts maintained the same size during the 48 h test period. [ABSTRACT FROM AUTHOR]

The pea mutant (Pisum sativum ageotropum) and the normal pea (P. sativum cv. Sabel) were compared in order to see if there were any differences in root anatomy or submorphology which could explain the presumed ageotropic behaviour of the mutant. In both types the root cap consists of a central core (columella) distinct from the peripheral part. The core contains five to six rows of columella cells, each consisting of 10 to 16 storeys of statocytes. The ultrastructure of the columella cells in the two types is very similar; the main difference is confined to the distribution of rough endoplasmic reticulum (ER), which in the mutant statocyres is evenly distributed throughout the cell, while in the normal pea statocytes it is mainly concentrated in the distal part at the "floor" of the cell. Using light micrographs, the movement of amyloplasts and nuclei have been followed in detail during a 40 mm inversion period. The pattern of movement of the amyloplasts is apparently identical in the two types and the distances moved during the inversion period are 39 μm and 44 μm in the normal and mutant statocytes, respectively. The nucleus has not been observed to move in normal pea; a slight rearrangement of the nucleus position can be observed during the period 30 to 40 mm after the start of inversion of the mutant. Based on magnified electron micrographs of the statocytes a morphometrical analysis was made of five cell structures - amyloplasts, nuclei, mitochondria, vacuoles and ER - which appeared to be freely movable or redistributable under the influence of the gravitational force. In an attempt to explain the influence of gravity on the behaviour of ageotropic plant organs, a pea mutant (Pisum sativum ageotropum) and normal pea (Pisum sativum cv. Sabel) were examined. The mutant has a significantly lower germination rate (large seeds: 25%, small seeds: 10%) than normal pea seeds (55%). Removal of testa increased germination dramatically, the values obtained were 63 and 89%, respectively. Immediately after imbibition the mutant from which the testa had been removed, developed more slowly than normal pea seeds; after 28 h the difference in elongation rate between the two types was reversed. When continuously stimulated geotropically in the horizontal position the elongation in the mutant is larger than in the normal pea roots kept in the same position. During a 24 h period starting 48 h after inhibition the mutant root elongated 45.0 mm while the value for the normal pea root was 11.5 mm. The course of the geotropic curvature in roots of the two types has been followed during a period of 24 h. Normal pea roots develop an asymmetry in the extreme root tip region after 30 mm of horizontal stimulation. After prolonged stimulation (exceeding 2 h) the asymmetry has disappeared and the curvature is distributed over the entire growth region. When roots of normal pea are stimulated continuously at various angles, the optimum angle of geotropic response is 90° with decreasing responses in the order 135° (i.e. the root tip is pointing obliquely upward) and 45°. The presumed ageotropic behaviour of the mutant has only to a certain extent been confirmed in the present study. The geotropic development in roots of Norway spruce [(Picea abies (L.)] H. Karst. has been followed by light and electron microscopy and compared with the movement of cell organelles (statoliths) in the root cap cells. The geotropic curvature develops in two phases: (a) an initial curvature in the root cap region, which results in an asymmetry in the extreme root tip and which appears after about 3 h stimulation in the horizontal position: and (b) the geotropic curvature in the basal parts of the root tip, which after 8 h is distributed over the entire elongation zone. A graphic extrapolation, based on measurements of the root curvatures after various stimulation periods, indicates a presentation time in the range of 8 to 10 min. The root anatomy and ultrastructure have been examined in detail in order to obtain information as to which organelles may act as gravity receptors. The root cap consists of a central core (columella) distinct from the peripheral part. The core contains three to four rows of parenchymatic cells each consisting of 15 to 18 storeys of statocyte cells with possibly mobile cell organelles. Amyloplasts and nuclei have been found to be mobile in the root cap cells, and the movement of both types of organelles has been followed after inversion of the seedlings and stimulation in the horizontal position for various periods of time at 4°C and 21°C. Three-dimensional reconstructions of spruce root cap cells based on serial sectioning and electron microscopy have been performed. These demonstrate that the endoplasmic reticulum (ER)-system and the vacuoles occupy a considerable part of the statocyte cell. Seedlings of Norway spruce (Picea abies L.) have been found to synthesize anthocyanins in the root tips as well as in the hypocotyls upon irradiation with white light when kept at 4°C for 6-8 days. In addition, it has also been found that the elongation and the geotropic curvature of spruce roots are dependent on the light conditions. The course of the geotropic curvature in spruce roots containing anthocyanins has been followed during a period of 5 h, in which the seedlings were geotropically stimulated continuously in the horizontal position. When the stimulation was performed in white light and in darkness at 21°C, significantly larger curvatures were observed in the roots pretreated at 4°C in darkness than in the roots containing anthocyanins. The specific curvature (curvature in degrees per mm elongation), however, was approximately the same in both types of roots stimulated in white light. This was due to a retarded elongation of the roots pretreated with light at 4°C and containing anthocyanins. A smaller difference in elongation rate between roots with and without anthocyanins was observed in the dark than in the light, but even in the dark the anthocyanin-containing roots grew more slowly than roots without anthocyanins. In order to find out if it is the anthocyanin content or the illumination which affects the elongation and geotropic curvature in the roots, a series of similar experiments was performed using cress seedlings grown at 4°C in light or darkness. Roots of cress seedlings cultivated under conditions which would induce anthocyanin formation in spruce roots exhibited the highest geotropic responses both in light and darkness as compared to cress seedlings grown at 4°C in darkness. As in the case of spruce roots an increase in elongation was observed in cress roots illuminated during the geotropic stimulation. This article examines the effect of a private sector on the waiting time associated with treatment in a public hospital. Without rationing of waiting-list admissions, a private sector is shown to result in a longer waiting time if the demand for a public treatment is sufficiently elastic with respect to the waiting time. When waiting-list admissions are rationed, the waiting time is shown to increase if the public sector consultants arc permitted to work in the private sector in their spare time. Previously inverted Lepidium roots were placed in a horizontal position and the amyloplasts in the statocytes of the root cap allowed to fall through their entire range of movement across the cell. Under these conditions the amyloplasts first follow a mainly downward course for 6 to 8 min at a speed between 0.5 and 0.8 μm per mm. For the next 10 min they move slightly more slowly in a direction away from the apical end of the cell, still sinking somewhat, but without reaching the plasmalemma along the lower wall. Previous experiments have shown that conditions assumed to allow the amyloplasts to slide parallel to the longitudinal cell walls are those that give rise to the largest geotropic curvatures. Such conditions are for instance (1) stimulation at 135° (root tips pointing obliquely upward) and (2) inversion of roots for 16 min followed by stimulation at 45° Treatments assumed not to permit extensive sliding of the amyloplasts produce smaller geotropic curvatures, namely (3) stimulation at 45° without pre-inversion and (4) inversion followed by stimulation at 135°. The location of the amyloplasts after these four kinds of treatment has now been determined on photomicrographs and the assumptions concerning the paths and extent of sliding of the amyloplasts confirmed. Observations on electron micrographs showed that under all, conditions the amyloplasts are separated from the plasmalemma by other organelles, such as ER, nucleus or vacuoles. In roots rotated for 15 min parallel, to the horizontal axis of the klinostat at 2 rpm, the amyloplasts are not clumped together as densely as in normal, inverted or stimulated. roots, but they are not scattered over the entire cell volume. Roots of cress growing between two agar slices develop an asymmetry in the extreme root tip region after 10 to 20 min of horizontal stimulation. After prolonged stimulation (exceeding 50 min) the asymmetry disappears and after 3 h the curvature is distributed over the entire growing region. The course of the initial stages in the geotropic curvature has been followed by light microscopy and scanning electron microscopy. -- When stimulated at an angle of 135° with the gravitational force, the asymmetry in the root tip is clearly visible after 10 min of stimulation. The asymmetry in the root cap can be explained by a difference in the elongation rate of the epidermal cells on the upper and lower sides of the stimulated root, The disappearance of the asymmetry is followed by a second phase in which there is a differential growth of the cortical cells or the two sides of the elongation zone. The average growth rate of cells in the upper half of the apical region during the first 50 min of continuous stimulation is 1.5 μm per min, while the elongation rate of the entire root is 16.2 μm per min. Only small modifications in the elongation rates were observed when stimulated and unstimulated roots were rotated parallel to the horizontal axis of a klinostat at 2 rpm. The ultimate curvature developed after 50 mm is unaffected by stimulation times exceeding the reaction time which for cress roots has been found to be about 5 min. The movement of auxin in Phaseolus vulgaris roots has been examined after injection of IAA−3H into the basal root/hypocotyl region of intact, dark‐grown seedlings. Only a portion of the applied IAA−3H was transported unchanged to the root tip. The major part of the chromatographed, labelled compounds translocated to the roots was indole‐3‐acetylaspartic acid (IAAsp) and an unidentified compound running near the front in isopropanol, ammonia, water. The velocity of the auxin transport (7.2 mm per hour) was calculated from scintillation countings of methanol extracts from serial sections of the root. An accumulation of radioactive compounds in the extreme root tip, was observed 5 h after the injection of IAA. The influence of exogenous IAA on the geotropical behaviour of the bean seedling roots was examined. Pretreated roots were stimulated for 5 min in the horizontal position and then rotated parallel to the horizontal axis of the klinostat for 60 or 90 min. The resulting geotropic curvature of IAA‐injected and control roots showed significantly different patterns of development. When the stimulation was started 5 h after application of the auxin, the geotropic curvature became larger in roots of the injected plants than in the controls. If, however, the translocation period was extended to 20 h the geotropic curvature was significantly smaller in the roots of the injected plants. The auxin injection did not significally affect the rate of root elongation. The change in geotropical behaviour of the roots is interpreted as a result of the influence of the conversion products of the applied IAA on the geotropical responsiveness. [ABSTRACT FROM AUTHOR]

When roots of cress seedlings (Lepidium sativum L.) are stimulated for 10 min at an angle of 135° (i.e. the root tips are pointing obliquely upward), the resulting geotropic curvatures become larger than after 10 min stimulation at 45°. This well-known behaviour has been explained by the better conditions for statoliths, initially located in the floor end of the statocytes, to slide along the cell walls when root tips are pointing upward at 135° than when pointing downward at 45°. Accepting this explanation, one would predict the optimum angle of stimulation to be near 45° when roots had first been kept inverted long enough for their statoliths to accumulate in the opposite end of each functional statocyte. This prediction has been verified in experiments with cress seedlings which were first kept inverted for 16 min, then stimulated for 10 min at given angles, and subsequently rotated parallel to the horizontal axis of the klinostat at 2 rpm. Under these conditions, roots stimulated at 45° curve faster during a 20 to 30 min period on the klinostat than roots stimulated at 135°, but thereafter they stop curving. Roots stimulated at 135°, on the other hand, although initially curving slower than those at 45°, continue curving for at least a whole hour, and attain larger curvatures than the others after 40 min. The optimum shifts from near 45° to near 135° during the course of the klinostat rotation. The behavior of normal and pre-inverted roots is interpreted as the result of at least two effects: (1) a stimulation due to the movement of amyloplasts, which is enhanced if these are allowed to slide along the cells walls, and (2) a modification of the development of the resulting curvatures by tonic effects, which are inhibitory between stimulation angles 0° and 90°, and absent or enhancing between 90° and 180°. The polar movement of IAA has been examined in 5-mm root segments of Brassica oleracea and Helianthus annuus. The movement was studied partly with IAA-1-14C and partly with IAA-5-³H In both plants a slight acropetal flux of 14C and IAA-³H was found through the segments. The recovered radioactivity in the agar receiver blocks and in the receiver end of the segments increased as a function of time. A large portion of the applied IAA was converted on the cut surfaces and in the tissues of the segments. Chromatographic analysis indicated different destruction products when estimated by scintillation counting and by spraying with indole reagent (DMCA). Chromatograms run in isopropanol : ammonia : water, 8 : 1 : 1, yielded three different substances, one spot near the starting line and one near the front, neither of which has been identified. Finally there was a spot with Rf 0.4-0.6, probably representing IAA.

Background: In 2003, the New Cooperative Medical Scheme (NCMS) was introduced in China to re-establish health insurance for the country's vast rural population. In addition, the coverage of NCMS has been expanding after the new health care reform launched in 2009. This study aims to examine whether the NCMS and its recent expansion have reached the goal of reducing the risk and inequality of catastrophic health spending for rural residents in China.Methods: We conducted a face-to-face household survey in three counties of the Shandong province in 2009 and 2012. Using this unique panel data, we examined the changes in the incidence and intensity of catastrophic health expenditures (CHEs) before and after NCMS reimbursement. We used concentration index (CI) and decomposition method to study the changes in inequality in CHEs.Results: We found that NCMS reimbursement played a role of reducing both the incidence and intensity of CHEs, and that this impact was stronger after the new health care reform was launched. After reimbursement, the concentration indices for CHEs were 0.073 and 0.021 in 2009 and 2012, indicating that the rich had a greater tendency to incur CHEs and there existed less inequality in the incidence of CHEs after reimbursement in 2012 compared with 2009. The decomposition analysis results suggested that changes in CHE inequality between 2009 and 2012 were attributed to changes in economic status and household size rather than reimbursement levels.Conclusions: Our results indicated that inequality was shrinking from 2009 to 2012, which could be a result of fewer rich people having CHEs in 2012 compared with 2009. The impact of NCMS in alleviating the financial burden of rural residents was still limited, especially among the poor. Quality improvements in markets for medical care are key objectives in any health reform. An important question is whether disclosing physicians’ performance can contribute to achieving these goals. Due to the asymmetric information inherent in medical markets, one may argue that changes in the information structure are likely to influence the environment in which health care providers operate. In a laboratory experiment with medical students that mimics a physician decision-making environment we analyze the effect of disclosing performance information to peers. Our results suggest that the information structure does influence the individual physician's supply of medical services. Volatile fractions from the roots of Echinops kebericho Mesfin were obtained by hydrodistillation and solid-phase microextraction (SPME) and analysed by gas chromatography-mass spectrometry (GC-MS); 65 compounds, representing 73.9% of the volatile fraction in the hydrodistillate from the roots, and 63 compounds, representing 78.6% of the volatile compounds in the SPME extract of the roots of E. kebericho, were identified. The main constituent found in hydrodistilled essential oil was eudesm-7(11)-en-4-ol (14.3%), followed by caryophyllene oxide (9.7%) and τ-cadinol (8.3%). High amounts of β-cubebene (22.7%), followed by β-patchoulene (11.1%), longifolene (10.4%), and cyperene (8.2%), were detected by SPME. Using both extraction methods, a total of 83 compounds were detected in E. kebericho essential oil. Monoterpenoid compounds represented 2.1% (seven compounds) and 2.1% (10 compounds) of the essential oil volatiles obtained by hydrodistillation and SPME, respectively, whereas sesquiterpenoids represented 72.5% (43 compounds) and 75.7% (31 compounds), respectively. Abstract: The MULTIGEN-1 ISS experiment has two objectives: a complete life cycle seed-to-seed growth experiment (about 70 days) and simultaneous observations of circumnutations, i.e. plant growth movements often describing circular or elliptical paths. MULTIGEN-1 is carried out in the European Modular Cultivation System (EMCS) hardware. The subject for this study is the shoot system of the plant Arabidopsis thaliana. The experiment provides the first long-term observations of circumnutations in microgravity. Preparatory experiments in the experiment reference model and under controlled laboratory conditions are performed. These evaluate 1-g growth behaviour under similar growth conditions as in the EMCS on the ISS. The rapid movements observed on Earth for hypocotyls, leaves and flower stalks require frequent image capture over several weeks. The circumnutations are 3D movements, but with only one video camera available per growth chamber, only two dimensions can easily be extracted. Triangulation of different plant parts using one video source is discussed. Limited bandwidth for downlink of images and limited onboard local storage combined with the image frequency required for recording fast moving circumnutations is also a strain on the system. The dynamics of root growth was studied in weightlessness. In the absence of the gravitropic reference direction during weightlessness, root movements could be controlled by spontaneous growth processes, without any corrective growth induced by the gravitropic system. If truly random of nature, the bending behavior should follow socalled ‘random walk’ mathematics during weightlessness. Predictions from this hypothesis were critically tested. In a Spacelab ESA-experiment, denoted RANDOM and carried out during the IML-2 Shuttle flight in July 1994, the growth of garden cress (Lepidium sativum) roots was followed by time lapse photography at 1-h intervals. The growth pattern was recorded for about 20 h. Root growth was significantly smaller in weightlessness as compared to gravity (control) conditions. It was found that the roots performed spontaneous movements in weightlessness. The average direction of deviation of the plants consistently stayed equal to zero, despite these spontaneous movements. The average squared deviation increased linearly with time as predicted theoretically (but only for 8–10 h). Autocorrelation calculations showed that bendings of the roots, as determined from the 1-h photographs, were uncorrelated after about a 2-h interval. It is concluded that random processes play an important role in root growth. Predictions from a random walk hypothesis as to the growth dynamics could explain parts of the growth patterns recorded. An experiment using plant protoplasts has been accepted for the IML-1 mission to be flown on a space shuttle in 1991. Preparatory experiments include studies of cell wall formation, cell division, the effect of simulated weightlessness using fast and slow rotating clinostats, and the development and testing of hardware for the IML-I mission. After 24 h at 25°C, protoplasts isolated from hypocotyls or leaves of rapeseed seedlings, or from carrot suspension cells, show 60, 20 and 15% cell wall formation, respectively. The time course of formation of the cell wall and cell division could be delayed by treatment at low temperatures or immobilization in alginate or agarose. This aspect is of importance in connection with problems of late access to the space shuttle before launch. At 4°C only 18% of the rapeseed hypocotyl protoplasts had formed cell walls after 24 h. Protoplasts immobilised in agarose or alginate gradually regain their cell division capacity and after 72 h the frequencies are 51 and 26%, respectively, compared to non-immobilised control protoplasts. A significant decrease in cell division activity is observed after rotation for 6 h on the slow clinostat. A similar effect is not observed on the fast clinostat. Growth and gravitropism have been studied in three mutant strains of Arabidopsis thaliana L. that are resistant to auxin-herbicide. Two of the mutations are allelic and recessive (aux-1 and aux-2) and are unlinked to a dominant mutation. Dwf, which confers a very high level of auxin-resistance and is apparently lethal when homozygous. The aux-1 and Dwf strains have altered response to gravity whereas aux-2 appears to he gravitropically normal. Alter 96 h in the normal, vertical position only minor differences in elongation were observed between roots of wild-type, aux-1 and aux-2, but the hypocotyls of aux-1 were significantly retarded compared with the gravitropically normal aux-2 and wild-type. In the progeny of selfed Dwf plants, where both normal (dwf) and agravitropic (Dwf) seedlings arc present, the Dwf seedlings had much longer roots and shorter hypocotyls than dwf. During 22 h of continuous stimulation the optimum angle for gravitropism in wild-type roots and hypocotyls was 135° (i.e. the organ points obliquely upwards), with decreasing responses in the order 90° and 45°. The agravitropic nature of the roots of aux-1 was confirmed as no significant response was obtained at any of the stimulation angles. In marked contrast, the negative gravitropic response of aux-1 hypocotyls was greater than the wild-type response in terms of the final angle attained at 22 h, but between 6 and 22 h the elongation rate was lower in aux-1. Alter varying stimulation periods in the horizontal position, the curvature which had developed, decreased rapidly and almost disappeared during ensuing rotation on clinostats (2 and 4 rpm). Essential oil from the seeds and husks of Aframomum corrorima (Braun) P.C.M. Jansen were analysed by gas chromatography (GC-MS), and 55 compounds were identified. The main constituents found in the seed oil were the monoterpenes 1,8-cineole (44.3%) and sabinene (17.3%), whereas sesquiterpenic structures (≥2%) such as ( E)-nerolidol (17.2%), β-caryophyllene (9.7%) and caryophyllene oxide (6.9%), dominated in the husk oil. Background In the elderly, kidney transplantation is associated with increased survival and improved health-related quality of life compared with dialysis treatment. We aimed to study the short-term health economic effects of transplantation in a population of elderly kidney transplant candidates. Methods Self-perceived health, quality-adjusted life years (QALYs) and costs were evaluated and compared 1 year before and 1 year after kidney transplantation in patients included in a single-centre prospective study of 289 transplant candidates ≥65 years of age. Results Self-perceived health and QALYs both significantly improved after transplantation. At 1 year, the costs per QALY were substantially higher for transplantation (€88 100 versus €76 495), but preliminary analyses suggest a favourable long-term health economic effect. Conclusions Kidney transplantation in older kidney transplant recipients is associated with improved health but also with increased costs the first year after engraftment when compared with remaining on the waiting list. Abstract: Gene expression analysis using microarrays has proved to be an important method in life science. The opportunity to grow higher plants on the International Space Station (ISS) opens up the possibility for gene expression profiling of plants grown in microgravity. The work presented focuses on how to meet the scientific requirements of plant growth and the sample preservation, given the technical and operational constraints associated with space research. The growth chamber (Multigen-2 Science Testing Unit) and a protocol suggested to be used in the European Modular Cultivation System (EMCS) Multigen-2 experiment on the ISS to grow and later preserve Arabidopsis seedlings, were tested on ground. The results showed that most of the plants developed normally. In order to avoid high population stress the number of seedlings per growth area should be reduced. The RNAlater preservation method to be used in the space experiment was compared with a quick freeze in Liquid Nitrogen (LN2). The RNA from samples preserved in RNAlater at room temperature for 24h was slightly more degraded than the RNA from the LN2 preserved samples (RNA integrity number, RIN: 7.7 and 8.6, respectively). However, the RNA quality and quantity was satisfactory for microarray analysis. Of the genes analysed, 74 genes (0.28%) were significantly differentially expressed, most of them showing moderate to low regulation. Among the genes induced in the RNAlater preserved samples, three salt inducible transcription factors (ZAT10, SZF1 and SZF2) were identified, suggesting that the high salt concentration in RNAlater causes salt stress before the transcription stopped. In conclusion, the Multigen-2 preservation protocol tested here will allow for the genes regulated by microgravity in the space experiment to be revealed. The results do indicate that not all the biological processes are stopped instantly by the RNAlater. The limited diffusion indirectly caused by the microgravity may potentially result in a different degree of salt stress in the test compared to the 1×g control during the space experiment. This has to be accounted for during the evaluation of the results. Since slightly degraded RNA was observed, further optimalisation of the preservation protocol will be performed. Polygalacturonase-inhibiting proteins (PGIPs) have been demonstrated to play a role in host defence in several plants. The PGIP now cloned from strawberry (Fragaria x ananassa) showed a high degree of homology to other fruit PGIPs. The gene expression of strawberry PGIP was monitored in healthy leaves, flowers and fruit at different maturity stages. PGIP transcript levels were also analysed following fruit inoculation with the fungal pathogen Botrytis cinerea in strawberry cultivars displaying variation in susceptibility. Healthy mature berries showed the highest constitutive PGIP gene expression levels compared with leaves, flowers and immature fruit, indicating that the gene is developmentally regulated. Among the cultivars studied ('Elsanta', 'Korona', 'Polka', 'Senga sengana', 'Tenira'), 'Polka' had the highest constitutive expression level of PGIP. After inoculation with B. cinerea, all five cultivars displayed a significant induction of PGIP gene expression, but the differences between them were not statistically significant. Production of varieties of the medicinal plants Mentha × piperita (peppermint) and Mentha sachalinensis (sachalinmint) was investigated over a 2-year trial period (1995–1996) with regard to harvesting date, oil content and terpene composition. Oil accumulation of peppermint (clones "Hardanger" and "Tøyen") and sachalinmint (var. "Mentolcsepp") increased throughout the season. Oil accumulation in both peppermint and sachalinmint was higher in plants grown in southern than in northern parts of Norway. An attempt was made to correlate the dependence of peppermint oil accumulation on the plant's age with the development of the peltate and capitate glands during the morphological development of the leaves. Changes in menthone–menthol metabolism were related to plant maturation for peppermint, with increasing menthol and reduced menthone content from the vegetative stage to full bloom. In contrast, the menthol–menthone content in sachalinmint was unchanged during plant maturation in 1995, while the results from 1996 showed increasing menthol content throughout the season. The growth and development of protoplasts of rapeseed (Brassica napus L. cv Line) and carrot (Daucus carota L. cv. Navona) were studied onboard the Space Shuttle 'Discovery' during an 8 day International Microgravity Laboratory (IML1) mission in January 1992. The flight experiments were carried out in 'Biorack', a fully controlled cell biological experimental facility. under microgravity conditions and in a 1-g centrifuge. Parallel experiments were performed in a 'Biorack' module on the ground. After retrieval, some samples were subcultured on appropriate media and analysed for callus growth and regeneration to intact plants. The remainder were used for biochemical analysis. Samples fixed on board the Space Shuttle were kept in 1% glutaraldehyde fixative at 4° C for 3-7 days for microscopy analysis after retrieval. Protoplasts exposed to microgravity conditions showed a delay in cell wall synthesis. Cells were swollen in appearance and formed cell aggregates with only few cells. Callus were obtained from protoplasts cultured under microgravity (F0g), on the 1-g centrifuge on board the shuttle (F1g), under normal 1g conditions on the ground (Glg), and on a centrifuge on the ground giving 1.4 g (G1.4g). Regeneration of intact rapeseed plants was obtained from F1g, G1g and G1.4g. However, no plants were regenerated from protoplasts exposed to microgravity (F0g). Biochemical analysis indicated that the microgravity samples (F0g) displayed a reduced packed cell volume, an increased concentration of soluble proteins per cell, and a reduced specific activity of peroxidase in the cytoplasm. Morphometric analysis of fixed samples demonstrated that 3 day old protoplasts under microgravity conditions were significantly larger than protoplasts kept on the 1g centrifuge in space. Ultrastructural analysis by transmission electron microscopy showed that protoplasts exposed to microgravity conditions for 3 days had larger vacuoles and a slightly reduced starch content compared to F1g cells. The root anatomy and ultrastructure of the agravitropic Arabidopsis thaliana L. mutants Dwf and aux-1 were compared with the gravitropic mutant aux-2 and the wild type (WT) in an attempt to find an explanation for the hick of response to gravity. No differences were found in the organization of the root cap. The central part of the cap (columella) Contains 5 storeys of developing, functioning and degenerating statocytes. Their ultrastructure is very similar in all four types of plant. Particular attention was paid to the distribution of rough endoplasmic reticulum (ER). Both in the WT and the mutants the ER is concentrated in the distal part at the "floor" of the cell. Light micrographs were used to compare the sedimentation rates of movable cell structures in normal and agravitropic root statocytes. A longitudinal movement of amyloplasts and nuclei was observed when the roots were inverted. In WT and aux-2 the rates were on average 6.3 μm h-1 (amyloplasts) and 2.1 μm h-1 (nucleus) In aux-1 the sedimentation rates were significantly lower: 2.4 and 0.6 μm h-1. respectively. Based on magnified electron micrographs of normal and inverted statocytes a morphometrical analysis of the distribution and redistribution of amyloplasts, nuclei, mitochondria, vacuoles and ER was made.
