Spinal cord stimulation (SCS) offers an alternative treatment for refractory pain resulting from various etiologies. Generally, SCS electrodes are inserted in an anterograde fashion, moving from caudal to rostral direction. However, there are instances where anterograde placement is unfeasible due to technical limitations. We present the use of retrograde surgical electrode placement in SCS for a patient with extensive epidural fibrosis at the site intended for electrode insertion. A 48-year-old female suffering from refractory neuropathic pain caused from injuries to the conus medullaris and cauda equina opted for SCS. During the SCS trial procedure, challenges emerged when attempting percutaneous electrode insertion at the site of a prior T12 laminectomy. However, the trial stimulation resulted in significant pain relief. For the permanent placement of the stimulator, utilizing a surgical electrode centered at T11 vertebral level, a considerable amount of epidural fibrosis was encountered at the entry of the spine, particularly at the T12 vertebral level. To avoid dural injury and ensure accurate electrode positioning, a retrograde technique for surgical electrode was employed via partial laminectomies at the T9-T10 level. The final electrode positioning was in accordance with the preoperative plan, well-centered at the T11 vertebral level. The patient experienced sustained relief from neuropathic pain over the long term. Retrograde epidural SCS is a suitable option for cases characterized by extensive epidural fibrosis resulting from a previous spinal surgery or when the anterograde placement of the electrode is unattainable due to aberrant vertebral anatomy. Deep ocean water (DOW) is obtained from 600 m below the sea surface. In recent years, DOW has been applied in the development of fermentation biotechnologies and functional foods. DOW is rich in trace minerals, comprises multiple physiological and health functions, and is able to promote microbe growth; therefore, the application of DOW directly benefits the development of the fermentation industry and functional foods. This study integrated the current health functions and applications of DOW with the latest results from studies related to fermentation biotechnology. Subsequently, the influence of applying DOW in fermented functional food development and the effects in health function improvements were summarized. According to the previous studies, the main reasons for the increased effect of fermented functional foods through the application of DOW are increased generation of functional metabolite contents in the microbes, intrinsic health functions of DOW, and the microbial use of mechanisms of converting the absorbed inorganic ions into highly bioavailable organic ions for the human body. Although researchers have published many studies on computer literacy and anxiety related to computer self-efficacy, there are two gaps in relevant literature. First, the effects of computer literacy and computer anxiety on computer self-efficacy are considered separately, yet their interaction effect is neglected. Second, the role of individual gender characteristics in the relationships between computer literacy and anxiety on computer self-efficacy is far from clear. To address these two concerns, this study empirically investigates the interaction effect between computer literacy and computer anxiety, and the moderating role of gender. This study tests hypotheses using survey data from people who have experience using computers in Taiwan, and uses hierarchical regression to analyze the models. Results indicate that computer literacy can help form positive computer self-efficacy more effectively for males than for females, and computer anxiety can lead to more negative computer self-efficacy for females than for males. A three-way interaction also exists among computer literacy, computer anxiety, and gender. Red mold rice (RMR) is a traditional food and folk medicine to Asian people and has recently become a popular health supplement. RMR has been shown to have some anticancer activities, although the mechanism for inducing cell death of human breast cancer cells is still not fully understood. In this study, bioactive extracts of RMR fermented by Monascus purpureus NTU 803 were analyzed for effects on apoptosis induction in human breast cancer cells. The RMR ethanol extract and ethyl acetate extract contain monacolin K, total phenols, and flavonoids, the three components that have been reported to have anticancer activity. Red mold rice extracts (RMRE) exhibited selective cytotoxic effect on MCF-7 cells. RMRE treatment induced apoptosis and cell cycle arrest at G2/M phase. Apoptosis was confirmed by annexin V-fluorescein isothiocyanate (FITC)/propidium iodide staining, the observation of characteristic chromatin condensation, nuclear DNA fragmentation, and poly(ADP-ribose) polymerase cleavage. Furthermore, the RMRE-induced apoptosis in MCF-7 cells may occur through a mitochondria-dependent pathway while triggering an appropriate balance of bax/bcl-2 and activation of caspase-9 and caspase-3 in a time-dependent manner. Monascus species has been used as the traditional food fungus in Eastern Asia for several centuries. Monascus-fermented products are gradually developed as the popular functional food for the prevention of cardiovascular disease, but we know that culture condition affects the hypolipidemic effect of Monascus-fermented product. In the past, the cholesterol-lowering agent-monacolin K-is regarded as the most important hypolipidemic agent. Two natural yellow pigments-monascin and ankaflavin-are also proven as novel hypolipidemic agents in recent years. However, the hypolipidemic effect of Monascus-fermented product should contribute from monacolin K, monascin, ankaflavin, and other unknown functional ingredients. In addition to hypolipidemic effect, the safety concern of Monascus-fermented product is involved in the levels of mycotoxin-citrinin. The hypolipidemic effect and the production of these functional metabolites or mycotoxin are influenced by many factors such as the choice of culture substrates, carbon and nitrogen source, pH value, extra nutrients, and so on. Therefore, this review focused on the effect of various culture conditions and nutrients on the functional metabolites production, hypolipidemic effect as well as citrinin concentration, and further organized the fermentation technologies used by previous studies for the promotion of hypolipidemic effect and safety. Ever since the concept of metabolism was extended from biological science by social scientists to analyze human systems, socio-economic metabolism has been extensively applied to explore resource consumption, asset accumulation, waste emissions, and complex processes of land use change in a socio-economic system. Current research in socio-economic metabolism and land use change has used accounting approaches for macroscopic comparisons of countries and regions. However, socio-economic metabolism has seldom been applied to the analysis of land use change. To simulate the spatial-temporal dynamics of socio-economic metabolism and land use change, this study adopts a spatial system modeling method to develop a Socio-Economic Metabolism and Land Use Change (SEMLUC) model for the Taipei Metropolitan Region. The simulation results illustrate that the Taipei Metropolitan Region is highly dependent on inflows of non-renewable energy and exhibits a spatial hierarchy of non-renewable energy consumption centering on Taipei's Main station. Additionally, urban assets provide feedback to natural and agricultural systems to extract additional resource inflows which, driven by the maximum power principle, accelerate the convergence of energy flows toward urban assets. Accumulating urban assets also facilitates inflows of non-renewable material to nearby cells thereby enhancing land use conversion to urban areas. Alzheimer's disease (AD) is the most common neurodegenerative disease, with symptoms appearing in the cerebral cortex and hippocampus. amyloid β peptide (Aβ) has been shown to deposit in the brain, causing oxidative stress and inflammation, leading to impaired memory and learning. Lactocaseibacillus fermentation can produce deglycosylated isoflavones with high physiological activity, which can scavenge free radicals, enhance total antioxidant capacity and inhibit oxidative inflammatory responses. Therefore, in this study, Lactocaseibacillus paracasei subsp. paracasei NTU101 (NTU101) fermented soybean milk and its extracts were used as test substances, and AD model rats were established by infusion of Aβ40 in the brain for 28 days, and the preventive and ameliorating effects of NTU 101 fermented soymilk were discussed. Effects of soymilk and unfermented soymilk on AD, and explore its effects on AD. Main functional ingredients. The results showed that deglycosylated isoflavones in NTU101 fermented soybean milk improved AD symptoms. Mechanisms of actions include the inhibition of oxidative inflammation; reduction in the expression of risk factors for tau protein and apo E protein production, the deposition of Aβ40 around the hippocampus, and the expression of TLR-2 and RAGE proteins in astrocytes and microglia; and improvement in the memory and learning ability. Amyloid β-peptide (Aβ), a risk of Alzheimer's disease (AD), causes cell death by inflammation and oxidative stress. Red mold rice (RMR) fermented by Monascus species is regarded as cholesterol-lowering functional food in virtue of the metabolite monacolin K identified as lovastatin. In addition, RMR is also demonstrated to express antioxidation because of multiple antioxidants. Therefore, this study focuses on the synergism of RMR against Aβ neurotoxicity and compares the effect between lovastatin and RMR including monacolin K and other functional metabolites. In this study, RE 568, an ethanol extract of RMR produced by strain Monascus purpureus NTU 568, is used to protect PC12 cell against Aβ40 neurotoxicity. All tests contain the treatments with lovastatin or RE 568 including equal monacolin K levels in order to compare the effect and investigate whether other metabolites of RE 568 provide potent assistance against Aβ40 neurotoxicity. In the results, monacolin K represses Aβ40 neurotoxicity via repressing small G-protein-mediated inflammation, and other metabolites of RE 568 also exhibit potent antioxidative ability against Aβ-induced oxidative stress. Importantly, stronger effects on repressing the Aβ40-induced cell death, inflammation, and oxidative stress are performed by RE 568 than that by the equal levels of lovastatin, which results from a potent synergism made up of monacolin K, antioxidants, and anti-inflammatory agents. The present study is the first report to demonstrate the potent synergistic protection of RMR against Aβ40 neurotoxicity, which would cause RMR to be developed as potential and novel functional food for the prophylaxis of AD pathogenesis. Monacolin K, an inhibitor for cholesterol synthesis, is the secondary metabolite of Monascus species. The formation of the secondary metabolites of the Monascus species is affected by cultivation environment and method. This research uses sweet potato ( Ipomoea batatas), potato ( Solanum tuberosum), casava ( Manihot esculenta), and dioscorea ( Dioscorea batatas) as the substrates and discusses the best substrate to produce monacolin K. The results show that Monascus purpureus NTU 301, with dioscorea as the substrate, can produce monacolin K at 2,584 mg kg−1, which is 5.37 times to that resulted when rice is used as the substrate. In addition, more amount of yellow pigment can be found in Monascus-fermented dioscorea than in Monascus-fermented rice. The certain composition of yellow pigment is identified as monascin, which has been shown as an antiinflammation agent exhibiting potent inhibitory effects on 12- O-tetradecanoylphorbol-13-acetate (TPA)-induced inflammation in mice in previous studies. Cordyceps cicadae is a well-known traditional Chinese medicine for treating palpitations and eye diseases. It contains several bioactive compounds such as adenosine, N6-(2-hydroxyethyl)-adenosine (HEA), and polysaccharide. Those bioactive compounds have been reported to perform anti-oxidation and anti-inflammatory properties and provide renal protection. In this study, we researched different fermentation conditions in order to enhance the biomass, adenosine, HEA, and polysaccharide productions of C. cicadae NTTU 868. Solid fermentation was carried out with different grain substrates (barley, oat, rice and wheat). Various submerged fermentation scales were used to produce the C. cicadae NTTU 868 mycelium. The results of solid fermentation revealed that C. cicadae NTTU 868 produced higher adenosine and HEA concentrations in oat rather than in other substrates. C. cicadae NTTU 868 mycelium had obtained the highest concentrations of adenosine and HEA on Day 2 as using the small-scale submerged fermentation. Furthermore, potato dextrose broth with extra 0.2% of yeast extract was able to result in higher HEA concentration. In conclusion, using submerged fermentation to culture C. cicadae NTTU 868 resulted in more efficient adenosine, HEA, and polysaccharide productions than using solid-fermentation, especially when 0.2% of yeast extract was used in the PDB. Antrodia camphorata is a fungus that is endemic to Taiwan, and its fruiting body has been used as a folk medicine for the prevention or treatment of diverse diseases. The present study is aimed at investigating the antimelanogenesis and antioxidation effect of the ethanolic extract of Antrodia camphorata fruiting body (EE-AC), as well as its antiproliferation effects in B16-F0 melanoma cells. Regarding antimelanogenic effects, EE-AC had effective cupric ions reducing capacity and expressed more potent inhibitory effect than kojic acid on mushroom tyrosinase activity. Moreover, EE-AC significantly inhibited cellular tyrosinase activity and the melanin content in B16-F0 cells at 12.5 μg/mL concentration without cell toxicities. Regarding antioxidant effects, EE-AC exhibited potent DPPH radical- and SOD-like-scavenging activities. Regarding antiproliferative effects, EE-AC exhibited a selective cytotoxic effect and markedly inhibited the migration ability of B16-F0 cells. EE-AC increased the population of B16-F0 cells at sub-G1 phase of the cell cycle. EE-AC also caused the increase of early apoptotic cells and chromatin condensation, which indicated the apoptotic effects in B16-F0 cells. We demonstrated that EE-AC possessed antimelanogenic, antioxidant and anti-skin cancer actions. This study assessed the anti-obesity effects of Lactobacillus paracasei subsp. paracasei NTU 101 (NTU 101) both in vitro and in vivo. Initially, the cytotoxicity and lipid accumulation inhibitory effects of NTU 101 on 3T3-L1 cells were evaluated using the MTT assay and oil red O assay, respectively. Subsequently, the anti-obesity effects of NTU 101 were investigated in high-fat diet-induced obese rat. Moreover, western blotting was performed to measure the obesity-related protein expression of PPARα, PPARβ, PPARγ, C/EBPα, C/EBPβ, ATGL, p -p38 MAPK, p -ERK1/2, p -AMPK and CPT-1 in both 3T3-L1 adipocytes and adipose and liver tissues. Treatment with 16 × 108 CFU/mL NTU 101 reduced lipid accumulation in 3T3-L1 adipocytes by more than 50 %. Oral administration of NTU 101 significantly attenuated body weight gain, as well as adipose tissue weight. NTU 101 administration enhanced fatty acid oxidation increasing expression levels of PPARα, CPT-1, and p -AMPK proteins in liver tissue, while simultaneously inhibited adipogenesis by reducing PPARγ and C/EBPα proteins in adipose tissue. Furthermore, NTU 101 supplementation positively modulated the composition of gut microbiota, notably increasing the abundance of Akkermansiaceae. Viewing urbanization as a change in the source and amount of energy flows from rural background to urban core provides a biophysical perspective of urban development. In this paper we begin by applying energetic principles to an urban setting and relating them to spatial hierarchy. Based on past research in urban energy theory and system modeling, we developed a spatial model to simulate the evolving spatial hierarchy of an urban system due to changing energy flows. Using an energy systems diagram, the spatial unit model consists of three interacting subsystems – natural area, agricultural area and urban area – representing a simplified entity or unit model of each grid element within a city-region. The Taipei metropolitan region is used as an example and is divided into grids of 1km×1km to reveal the spatial heterogeneity of the urban landscape system. The spatial simulation was performed using geographical information system (GIS) and the model results show an increase in the urban energy hierarchy and reveal a pattern of spatial convergence. Safety design and probabilistic optimization are fields that are widely subject to uncertainty, thus making traditional deterministic methods highly unreliable for these two fields. Popular design optimizations methods widely used for safety design and probabilistic optimization are the performance measure approach (PMA) and the performance measure approach (RIA). In a problem where the analysis is performed from an infeasible design space, a modified reliability index approach (MRIA) is employed to address some inefficiency of the traditional RIA to be able to find the optimal solutions. The PMA uses an inverse reliability analysis, which is more computationally efficient at finding the most probable design points but has been reported to have numerical instabilities on some cases. In this paper, three benchmark examples were thoroughly studied with various initial points to examine the stability and efficiency of MRIA and PMA. A hybrid reliability approach (HRA) was then presented after determining a selection factor from the optimum conditions. Antrodia camphorata is a unique fungus in Taiwan; the submerged fermentation product is used as the functional food for liver protection. Deep ocean water (DOW) containing rich metals and trace elements is proven to stimulate the production of functional metabolites and health function of functional fungus product in our previous study. Therefore, A. camphorata-fermented product cultured in DOW (DOW-AC) or reverse osmosis water (ROW-AC) as culture water was daily fed thioacetamide (TAA)-induced fibrosis rat for 8 weeks in order to investigate whether DOW promoted the effect of A. camphorata-fermented product on the prevention against TAA-induced liver damage and fibrosis. In the results, feeding one dose of DOW-AC prevented from TAA-induced weight loss and had more effect on inhibiting lipid peroxidation, reactive oxygen species, iNOS, and TNF-α expression than one dose of ROW-AC. Furthermore, DOW-AC also had more potent effect on protection against TAA-induced liver damage and fibrosis according to the results of H&E stain and collagen stain. However, higher liver protection of DOW-AC should be due to the fact that DOW not only increased the production of A. camphorata-fermented functional metabolites including triterpenoids, polysaccharides, flavonoids, and polyphenols but also contributed to protection against TAA-induced damage and fibrosis. Antrodia camphorata is a particular and precious medicinal mushroom, and its fruiting body was found to provide more efficient protection from oxidative stress and inflammation than its mycelium because of its higher content of triterpenoids, total phenols, and so on . In the previous in vitro studies, the mycelium of A. camphorata is proven to provide strong neuroprotection in neuron cells and suggested to have the potential of protection against neurotoxicity of amyloid β-protein (Aβ) known as the risk factor toward Alzheimer's disease (AD) development. However, the in vivo study and the comparison study with the fruiting body have not yet been investigated. This study compared the effect of the fruiting body and mycelium of A. camphorata on alleviating the Aβ40-induced neurocytotoxicity in the in vitro Aβ-damaged neuron cell model (PC-12 cell treated with Aβ40) and memory impairment in the in vivo AD animal model induced with a continuous brain infusion of Aβ40. In the results of in vitro and in vivo studies, the fruiting body possessed stronger anti-oxidative and anti-inflammatory abilities for inhibiting neurocytotoxicity in Aβ40-treated PC-12 cells and Aβ40 accumulation in Aβ40-infused brain than mycelium. Moreover, hyperphosphorylated tau ( p-tau) protein expression, known as an important AD risk factor, was suppressed by the treatment of fruiting body rather than that of mycelium in the in vitro and in vivo studies. In search for novel anti-tumour agents targeting non-homologous end joining pathway, we conducted a virtual screening using traditional Chinese medicine (TCM) on Ku heterodimer which plays an important role on DNA break repair system. Docking results gave glycyrrhizic acid and macedonoside C as potential TCM candidates. Both glycyrrhizic acid and macedonoside C were docked onto the Ku heterodimer with high-predicted binding affinities as evidenced in their Dock Score. These two ligands also interact with key residues which have been previously shown to influence Ku DNA-binding affinity. Both compounds show consistent hydrogen bonding interactions with key residues throughout the 10- ns dynamics simulation run. Furthermore, glycyrrhizic acid and macedonoside C are able to form additional hydrogen bond interactions with positively charged Ku heterodimer surface. Such interactions strengthen the binding interaction of the top two TCM compounds with Ku heterodimer. Glycyrrhizic acid and macedonoside C are products of licorice (Glycyrrhiza glabra), which is a commonly used herb in TCM formulations. Microsomal prostaglandin E synthase-1 (mPGES-1) has been regarded as an attractive drug for inflammation-related diseases. In search of new mPGES-1 inhibitors, we performed virtual screening using our traditional Chinese medicine and natural products database (http://tcm.cmu.edu.tw/) and constructed comparative molecular field analysis (CoMFA) and comparative molecular similarity indices analysis (CoMSIA) using a training set of 30 experimentally tested mPGES-1 inhibitors. The CoMFA and CoMSIA models derived were statistically significant with cross-validated coefficient values of 0.808 for CoMFA and 0.829 for CoMSIA and non-cross-validated coefficient values of 0.829 for CoMFA and 0.980 for CoMSIA. Docking and de novo evolution design gave three top derivatives, 2-O-caffeoyl tartaric acid-Evo_2, glucogallin-Evo_1 and 3-O-feruloylquinic acid-Evo_7 that have higher binding affinities than the control, glutathione. These three derivatives have interactions with Arg70, Arg73, Arg110, Arg126 and Arg38, which all are mPGES-1 key active site residues. In addition, these derivatives fit well into the CoMFA and CoMSIA models, with hydrophobic, hydrophilic and electropositive substructures mapped onto corresponding contour plots. Alcohol is metabolized in liver. Chronic alcohol abuse results in alcohol-induced fatty liver and liver injury. Red quinoa (Chenopodium formosanum) was a traditional staple food for Taiwanese aborigines. Red quinoa bran (RQB) included strong anti-oxidative and anti-inflammatory polyphenolic compounds, but it was usually regarded as the agricultural waste. Therefore, this study is to investigate the effect of water and ethanol extraction products of RQB on the prevention of liquid alcoholic diet-induced acute liver injury in mice. The mice were given whole grain powder of red quinoa (RQ-P), RQB ethanol extract (RQB-E), RQB water extract (RQB-W), and rutin orally for 6 weeks, respectively. The results indicated that RQB-E, RQB-W, and rutin decreased alcoholic diet-induced activities of aspartate aminotransferase and alanine aminotransferase, and the levels of serum triglyceride, total cholesterol, and hepatic triglyceride. Hematoxylin and eosin staining of liver tissues showed that RQB-E and RQB-W reduced lipid droplet accumulation and liver injury. However, ethanol extraction process can gain high rutin and antioxidative agents contents from red quinoa, that showed strong effects in preventing alcoholic fatty liver disease and liver injury via increasing superoxide dismutase/catalase antioxidative system and repressing the expressions of fatty acid synthesis enzyme acetyl-CoA carboxylase. Alcohol metabolism causes an excessive accumulation of liver lipids and inflammation, resulting in liver damage. The yellow pigments monascin (MS) and ankaflavin (AK) of Monascus purpureus-fermented rice were proven to regulate ethanol-induced damage in HepG2 cells, but the complete anti-inflammatory and anti-fatty liver mechanisms in the animal model are still unclear. This study explored the roles of MS and AK in improving alcoholic liver injury. MS and AK were simultaneously fed to evaluate their effects and mechanisms in C57BL/6J mice fed the Lieber–DeCarli liquid alcohol diet for 6 weeks. The results indicated that MS and AK significantly reduced the serum aspartate aminotransferase and alanine aminotransferase activity, as well as the total liver cholesterol and triglyceride levels. The histopathological results indicated that MS and AK prevented lipid accumulation in the liver. MS and AK effectively enhanced the activity of antioxidant enzymes and reduced the degree of lipid peroxidation; AK was particularly effective and exhibited a superior preventive effect against alcoholic liver injury and fatty liver. In addition to inhibiting the phosphorylation of the MAPK family, MS and AK directly reduced TNF-α, IL-6, and IL-1β levels, thereby reducing NF-κB and its downstream iNOS and COX-2 expressions, as well as increasing PPAR-γ, Nrf-2, and HO-1 expressions to prevent liver damage.
