Marbofloxacin (MBF) was once widely used as a veterinary drug to control diseases in animals. MBF residues in animal food endanger human health. In the present study, an immunochromatographic strip assay (ICSA) utilizing a competitive principle was developed to rapidly detect MBF in beef samples. The 50% inhibitory concentration (IC50) and the limit of detection (LOD) of the ICSAs were 2.5 ng/mL and 0.5 ng/mL, respectively. The cross-reactivity (CR) of the MBF ICSAs to Ofloxacin (OFL), enrofloxacin (ENR), norfloxacin (NOR), and Ciprofloxacin (CIP) were 60.98%, 32.05%, 22.94%, and 23.58%, respectively. The CR for difloxacin (DIF) and sarafloxacin (SAR) was less than 0.1%. The recovery rates of MBF in spiked beef samples ranged from 82.0% to 90.4%. The intra-assay and interassay coefficients of variation (CVs) were below 10%. In addition, when the same authentic beef samples were detected in a side-by-side comparison between the ICSAs and HPLC‒MS, no statistically significant difference was observed. Purpose: Mowing may exert profound influences on nutrient acquisition strategies of herbaceous species due to the nutrient removal along with the mown plants. Nutrient uptake by roots and nutrient resorption from senescent leaves, are two main nutrient acquisition strategies of perennial herbaceous species. However, few studies have considered both nutrient acquisition strategies in exploring the effects of long-term mowing on the nutrient acquisition of perennial herbs. Methods: We measured root traits of absorptive roots and leaf nitrogen resorption efficiency (NRE) of seven herbaceous species with different plant height on a long-term mowing experimental platform (2003-2020) in a temperate steppe. Results: We found that root traits of tall and short species had little response to long-term mowing. While, mowing exerted opposite effects on leaf NRE of the two plant groups, i.e., mowing significantly decreased the NRE of tall species, but increased that of short species (except for Potentilla acaulis). Furthermore, root absorption capacity (indicated by root traits) was positively correlated with leaf NRE for tall species in control plots, but mowing eliminated this relationship. In contrast, for short species, root traits and leaf NRE had no correlation in control plots, while, mowing led to synergic relationship between root absorption capacity and leaf resorption rates. Conclusions: Our results suggested that perennial herbs changed the relationships between alternative resource acquisition strategies in response to long-term mowing in temperate steppe. Soil, which exhibits difference in nutrient contents and aggregate sizes, provides spatially distinct habitats for biota. Cover crops influence the compositions of soil organism communities, playing an indispensable role in regulation of underground food webs and ecosystem functions. However, the effect of cover crops on soil microbes and nematodes distribution within different aggregate sizes remains unknown. Thus, a field experiment in a kiwifruit orchard with cover crops was conducted to estimate the distribution of soil nematodes and bacteria with different soil aggregate sizes (mega-aggregate (>2 mm, LMA), macro-aggregate (0.25–2 mm, SMA), and micro-aggregate (<0.25 mm, MA)) and cover crop treatments (four cover crop species (CC) and no cover crop as control (CK)). The results showed that bacterial compositions varied with both aggregate sizes and cover crop treatments. The composition of bacterial community was significantly different between mega-aggregate and micro-aggregate, and bacterial community diversity was significantly higher in micro-aggregate compared with mega-aggregate. Moreover, cover crop treatment dramatically changed the compositions of bacterial communities. However, the nematode communities were mainly impacted by soil aggregate sizes. Larger aggregates (mega- and macro-aggregates) contained higher abundance of omnivores/predators and lower abundance of fungivores. In contrast to bacterial community, the richness of nematode community was lower in micro-aggregates compared with larger aggregates (mega- and macro-aggregates). Redundancy analysis (RDA) and structural equation model (SEM) showed soil organic carbon (SOC) was the main soil factor that directly and indirectly affected both bacterial and nematode communities. Iron is one of the essential mineral elements for the human body and this nutrient deficiency is a worldwide public health problem. Iron is essential in oxygen transport, participates in many enzyme systems in the body, and is an important trace element in maintaining basic cellular life activities. Iron also plays an important role in collagen synthesis and vitamin D metabolism. Therefore, decrease in intracellular iron can lead to disturbance in the activity and function of osteoblasts and osteoclasts, resulting in imbalance in bone homeostasis and ultimately bone loss. Indeed, iron deficiency, with or without anemia, leads to osteopenia or osteoporosis, which has been revealed by numerous clinical observations and animal studies. This review presents current knowledge on iron metabolism under iron deficiency states and the diagnosis and prevention of iron deficiency and iron deficiency anemia (IDA). With emphasis, studies related to iron deficiency and bone loss are discussed, and the potential mechanisms of iron deficiency leading to bone loss are analyzed. To investigate the clinical effect of lymphatic preserving varicocelectomy (LPV) versus lymphatic non-preserving varicocelectomy (nLPV). Literature retrieval was performed for literatures published until January 2021 using the terms "varicoceles" and "lymphatic" through databases including Embase, PubMed, Cochrane Review database, Wanfang, and China National Knowledge Infrastructure (CNKI). Randomized controlled trials (RCTs) comparing LPV and nLPV were included. A meta-analysis was conducted using risk ratios (RR) for dichotomous variables. A total of 8 RCTs involving 419 cases with LPV and 434 cases with nLPV met inclusion criteria. LPV was with lower incidence of hydrocele compared with nLPV (RR = 0.099, 95% CI 0.046, 0.212; p = 0.000). In addition, no significant difference was found in recurrence rate (RR = 0.883, 95% CI 0.468, 1.665; p = 0.700) or testicular atrophy rate (RR = 0.750, 95% CI 0.170, 3.311; p = 0.704). This meta-analysis provided evidence that LPV was with lower incidence of hydrocele compared with nLPV, LPV may be preferable to nLPV in the treatment of varicoceles. [ABSTRACT FROM AUTHOR]

With the rapid development of global satellite constellation clusters and the popularity of ground-based intelligent terminals, the interconnection between large-scale low Earth orbit satellites and ground base stations has become more frequent. The calculation of satellite position requires a large number of floating point square calculations and coordinate system conversions, with high computational complexity and long computation time. Based on the grid encoding method of GeoSOT, we propose a GeoSOT-based grid calculation for a satellite-ground position (GCSGP) model. We use binary bit operations instead of complex topological operations and floating point calculations, and use grid distance calculations instead of existing angle calculations. Thus, the relative positions of satellites to the ground can be quickly calculated and thus connected to more suitable satellites quickly. The results of the grid calculation were verified by simulation experiments. The calculation speed was improved by about 82% compared with the traditional method. The test accuracy error of the 10-level grid at a terminal 45° inclination was less than 1% at all heights, and the maximum error of the 10-level grid within a 10–45° inclination was 7.2% at 10,000 km altitude, which provides theoretical support for the subsequent grid space calculation. In order to cope with the rapid growth of spatiotemporal big data, data organization models based on discrete global grid systems have developed rapidly in recent years. Due to the differences in model construction methods, grid level subdivision and coding rules, it is difficult for discrete global grid systems to integrate, share and exchange data between different models. Aiming at the problem of information fusion between a GeoSOT grid and global hexagonal equal area grid system, this paper proposes the GeoSOT equivalent aggregation model (the GEA model). We establish a spatial correlation index method between GeoSOT grids and global hexagonal equal area grids, and based on the spatial correlation index, we propose an interoperable transformation method for grid attributes information. We select the POI (points of interest) data of Beijing bus and subway stations and carry out the transformation experiment of hexagonal grid to GeoSOT grid information so as to verify the effectiveness of the GEA model. The experimental results show that when the 17th-level GeoSOT grid is selected as the particle grid to fit the hexagonal grid, the accuracy and efficiency can be well balanced. The fitting accuracy is 95.51%, and the time consumption is 30.9 ms. We establish the associated index of the GeoSOT grid and the hexagonal grid and finally realized the exchange of information. • Exendin-4 intervention led to a significant reduction in aortic plaques and body weight in ApoE-/- mice compared to the model group. • Exendin-4 significantly increased the proportion of myeloid-derived suppressor cells (MDSCs) in bone marrow and peripheral blood, while reducing pro-inflammatory cytokines (TNF-α, IFN-γ, IL-6) and increasing anti-inflammatory cytokines (TGF-β, IL-10) in both spleen and plasma. • Exendin-4 regulated lipid levels by decreasing triglycerides (TG), total cholesterol (TC), and low-density lipoprotein cholesterol (LDL-C), while increasing high-density lipoprotein cholesterol (HDL-C) in ApoE-/- mice. To investigate the impact of the glucagon-like peptide-1 (GLP-1) receptor agonist Exendin-4 on the proportion of myeloid-derived suppressor cells (MDSCs) in male ApoE-/- mice, and investigate alterations in the concentrations of inflammatory factors in plasma and spleen tissues and assess their correlation with MDSCs. Thirty male ApoE-/- mice were randomly divided into five groups (n = 6 per group): control group (CON), model group (MOD), Exendin-4 intervention group (MOD/Ex-4), Exendin-9-39 intervention group (MOD/Ex-9-39), and Exendin-4 + Exendin-9-39 combined intervention group (MOD/Ex-4 + Ex-9-39). After 4 weeks of drug intervention, changes in aortic plaque were observed using Oil Red O staining and H&E staining. Flow cytometry was employed to detect the content of myeloid-derived suppressor cells (MDSCs) in bone marrow and peripheral blood. ELISA was utilized to measure the concentrations of inflammatory factors in mouse peripheral blood plasma, while RT-qPCR was employed to quantify the expression levels of inflammatory factors in the spleen. Pearson correlation analysis was conducted to assess the relationship between MDSCs and inflammatory factors. Mice in the MOD group had significantly higher body weight compared to the CON group, with a statistically significant difference (P<0.05). Following Exendin-4 intervention, body weight was reduced compared to the MOD group (P<0.05). Additionally, Exendin-4 treatment led to a significant reduction in atherosclerotic plaque compared to the MOD group (P<0.001). After Exendin-4 intervention, the proportion of MDSCs in the bone marrow was higher than in the MOD group (P<0.001), and the proportion of MDSCs in peripheral blood was significantly higher than in the MOD group (P<0.05). Further investigation revealed that Exendin-4 could regulate lipid levels in mice, decreasing concentrations of TG (P<0.01), TC (P<0.0001), and LDL-C (P<0.0001), while increasing HDL-C concentrations (P<0.01). Moreover, after Exendin-4 treatment, the level of the cytokine IL-6 in peripheral plasma was significantly lower compared to the MOD group (P<0.0001), while levels of IL-10 and TGF-β were significantly higher compared to the MOD group (P<0.0001). In the spleen, levels of the cytokines IL-10 (P<0.0001) and TGF-β (P<0.001) were significantly increased compared to the MOD group. Pearson correlation analysis showed that the proportion of MDSCs in peripheral blood was positively correlated with IL-10 and TGF-β levels in both the spleen and peripheral blood. Additionally, the proportion of MDSCs in the bone marrow was positively correlated with IL-10 and TGF-β levels in the spleen and peripheral blood. Exendin-4 alleviates the severity of atherosclerosis. This process may be achieved by promoting the secretion of myeloid-derived suppressor cells (MDSCs) in the bone marrow and peripheral blood of atherosclerotic ApoE-/- mice, regulating the ratio of inflammatory factors in the body, reducing mouse body weight, and lowering blood lipids. Freeze–thaw (FT) damage is one of the main factors affecting the durability of concrete structures. This paper focused on the effects of the coarse aggregate volume fraction (CAVF) on FT damage to concrete in different salt solution concentrations (SSCs). The dynamic elastic modulus (DEM) and cumulative scaling quantity (CSQ) were obtained by rapid FT tests to investigate the CAVF's influence at different SSCs. Both the dynamic elastic modulus and cumulative scaling quantity became worse with the increasing FT cycles. The dynamic elastic modulus increased with an increasing coarse aggregate volume fraction, but the cumulative scaling quantity decreased significantly. Meanwhile, the salt solution concentration had little effects on the DEM and CSQ. Furthermore, an empirical prediction model for evaluating the relative dynamic elastic modulus (RDEM) considering the CAVF was established via regression analysis and validated by experiments. A prediction model for assessing the relative cumulative scaling quantity taking into account the effects of the loss rates of the RDEM and CAVF was developed, and it correlated well with the experimental results. The COVID-19 pandemic is currently spreading widely around the world, causing huge threats to public safety and global society. This study analyzes the spatiotemporal pattern of the COVID-19 pandemic in China, reveals China's epicenters of the pandemic through spatial clustering, and delineates the substantial effect of distance to Wuhan on the pandemic spread. The results show that the daily new COVID-19 cases mostly occurred in and around Wuhan before March 6, and then moved to the Grand Bay Area (Shenzhen, Hong Kong and Macau). The total COVID-19 cases in China were mainly distributed in the east of the Huhuanyong Line, where the epicenters accounted for more than 60% of the country's total in/on 24 January and 7 February, half in/on 31 January, and more than 70% from 14 February. The total cases finally stabilized at approximately 84,000, and the inflection point for Wuhan was on 14 February, one week later than those of Hubei (outside Wuhan) and China (outside Hubei). The generalized additive model-based analysis shows that population density and distance to provincial cities were significantly associated with the total number of the cases, while distances to prefecture cities and intercity traffic stations, and population inflow from Wuhan after 24 January, had no strong relationships with the total number of cases. The results and findings should provide valuable insights for understanding the changes in the COVID-19 transmission as well as implications for controlling the global COVID-19 pandemic spread. To study the efficacy of a polyoxometalate, Cs2K4Na[SiW9Nb3O40]·H2O, as an antiviral treatment in HBV transgenic mice. HBV transgenic mice were treated with Cs2K4Na[SiW9Nb3O40]·H2O by intragastric administration. Adefovir and distilled water were administered as controls. Serum HBV DNA, liver HBV RNA levels were measured by quantitative RT‐PCR. Serum HBsAg levels were measured by ELISA. The hepatitis B virus surface antigen (HBsAg) in liver cells was detected by immunohistochemistry (IHC). Pathological changes in the liver tissues were also observed by light and electron microscopy. Cs2K4Na[SiW9Nb3O40]·H2O significantly decreased serum HBsAg and HBV DNA levels. Cs2K4Na[SiW9Nb3O40]·H2O resulted in a 98% decrease in serum HBV DNA at 28 days, from 4.3 log10 copies/ml at baseline to 2.5 log10 copies/ml after treatment, and the inhibition rate of HBV DNA was higher than ADV at the same dose. The HBV replication levels in each group slightly increased at 7 days after withdrawal, but rebounded slightly more in the Cs2K4Na[SiW9Nb3O40]·H2O treatment group compared to the H2O control group (p <.05). There were no differences in HBV RNA levels. No significant differences were observed in the pathology, but there were decreased HBsAg levels in the Cs2K4Na[SiW9Nb3O40]·H2O‐treated group compared to the control group. Resistance to radiotherapy accounts for most therapeutic failures in cervical cancer patients who undergo radical radiation therapy. To indicate the possible molecular mechanism of radioresistance and improve the 5-year survival rate, we focused on how SET domain protein 3 (SETD3) regulated radioresistance in human cervical cancer cells in this study. Our results indicated that SETD3 over-expression markedly increased the radiosensitivity of cervical cancer cells with radioresistance, as evidenced by the further reduced cell viability, proliferation, DNA damage and cell death. In addition, we found that SETD3 down-regulated the expression of kinesin light chain 4 (KLC4), contributing to the radiosensitivity of cervical cancer cells, and the regulatory role of SETD3 could be abolished by KLC4 over-expression. Moreover, nitric oxide (NO) production was significantly reduced by SETD3 over-expression through repressing the expression of inducible NO synthase (iNOS) and endothelial NO synthase (eNOS) in cervical cancer cells. In vivo studies using xenograft animal models also demonstrated that SETD3 over-expression combined with irradiation treatment markedly inhibited tumor growth and induced apoptosis. Following acute infection of mucosal surfaces by bovine herpesvirus 1 (BoHV-1), sensory neurons are a primary site for lifelong latency. Stress, as mimicked by the synthetic corticosteroid dexamethasone, consistently induces reactivation from latency. Two viral regulatory proteins (VP16 and bICP0) are expressed within 1 h after calves latently infected with BoHV-1 are treated with dexamethasone. Since the immediate early transcription unit 1 (IEtu1) promoter regulates both BoHV-1 infected cell protein 0 (bICP0) and bICP4 expressions, we hypothesized that the bICP4 protein is also expressed during early stages of reactivation from latency. In this study, we tested whether bICP4 and bICP22, the only other BoHV-1 protein known to be encoded by an immediate early gene, were expressed during reactivation from latency by generating peptide-specific antiserum to each protein. bICP4 and bICP22 protein expression were detected in trigeminal ganglionic (TG) neurons during early phases of dexamethasone-induced reactivation from latency, operationally defined as the escape from latency. Conversely, bICP4 and bICP22 were not readily detected in TG neurons of latently infected calves. In summary, it seems clear that all proteins encoded by known BoHV-1 IE genes (bICP4, bICP22, and bICP0) were expressed during early stages of dexamethasone-induced reactivation from latency. Abstract Cervical cancer is one of the most lethal types of cancer among female. Microfibrillar-associated protein 5 (MFAP5) is an extracellular matrix (ECM) glycoprotein, and is confirmed to be involved in cell signaling during microfibril assembly, elastinogenesis and cell survival. However, the role of MFAP5 in cervical cancer development and progression remains poorly understood. In the study, MFAP5 was over-expressed in human cervical cancers, and in different cervical cancer cell lines. Patients suffering from cervical cancer with low MFAP5 expression exhibited better survival rate. Suppressing MFAP5 in cervical cancer cells markedly reduced the cell proliferation, migration and invasion by modulating epithelial-mesenchymal transition (EMT)-related signaling pathway. In addition, MFAP5 knockdown induced large number of cells distributed in G2/M phase, along with reduced Cyclin B1, Cyclin D1 and cyclin-dependent kinase 4 (CDK4) expressions, and enhanced p21 and p53 levels. Moreover, apoptosis was highly induced by MFAP5 silence through reducing Bcl-xl and Bcl-2 expressions, and promoting Bax, cleaved Caspase-3 and poly (ADP-Ribose) polymerase (PARP) expressions in cervical cancer cells. Reactive oxygen species (ROS) production levels were also higher in MFAP5-knockdown cells, along with Jun-N-terminal kinase (JNK) activation. Importantly, we found that MFAP5 knockdown-inhibited cervical cancer cell growth was dependent on ROS production. Finally, the depletion of MFAP5 prevented cervical cancer progression in vivo. In summary, our study identified a critical role played by MFAP5 in the progression of cervical cancer and the potential mechanisms by which exerted its effects, indicating that targeting MFAP5-related pathways could be conducive to the therapies for cervical cancer. Objective: The aim of this study was to explore the tolerance, variability, and pharmacokinetics (PK) of bevacizumab biosimilars (MIL60, BAT1706, IBI305) in Chinese healthy male subjects.Methods: This randomized, double-blind, two-arm, parallel studies included three separate investigations, which were conducted by three sponsors to investigate the bioequivalence of bevacizumab biosimilars (MIL60, BAT1706, IBI305) with that of bevacizumab-EU as a reference drug. Subjects received a single-dose of 1 or 3 mg/kg of the bevacizumab biosimilars or bevacizumab-EU and were followed up for 70-99 days. Serum concentrations of bevacizumab, antidrug antibody (ADA), and neutralizing antibody (NAb) were measured using electrochemiluminescence. In addition, the PK parameters were determined using non-compartmental methods. The safety assessments included adverse events, hematology tests, and biochemistry tests.Results: The three bevacizumab biosimilars exhibited similar PK properties to that of bevacizumab-EU. Bevacizumab demonstrated linear PK properties and a concentration-dependent disposition. When comparing the three biosimilars with bevacizumab-EU, the 90% CIs of the ratios for Cmax, AUC0-t, and AUC0-∞ were within 80-125%. The inter-CV ranged from 12.6 to 23.3%. Three subjects in the biosimilar groups and bevacizumab-EU were positive for the ADA and negative for the NAb. Treatment-related mild or moderate adverse events were reported in 56-80 and 36-80% of subjects in the biosimilar and bevacizumab treatment arms, respectively.Conclusions: The bevacizumab biosimilars exhibit similar PK characteristics to that of the reference product bevacizumab-EU. Lateral flow immunoassay (LFIA) is widely used as a rapid point-of-care testing (POCT) technique in food safety, veterinary and clinical detection on account of the accessible, fast and low-cost characteristics. After the outbreak of the coronavirus disease 2019 (COVID-19), different types of LFIAs have attracted considerable interest because of their ability of providing immediate diagnosis directly to users, thereby effectively controlling the outbreak. Based on the introduction of the principles and key components of LFIAs, this review focuses on the major detection formats of LFIAs for antigens, antibodies and haptens. With the rapid innovation of detection technologies, new trends of novel labels, multiplex and digital assays are increasingly integrated with LFIAs. Myeloid-derived suppressor cells (MDSCs) are a heterogeneous cell population found in the bone marrow, peripheral blood, and tumor tissue. Their role is mainly to inhibit the monitoring function of innate and adaptive immune cells, which leads to the escape of tumor cells and promotes tumor development and metastasis. Moreover, recent studies have found that MDSCs are therapeutic in several autoimmune disorders due to their strong immunosuppressive ability. Abstract: We investigated the resonant transmission of planar metamaterials structured in symmetric and asymmetric double-split rings through simulations and experiments. With increasing of the arm lengths the transmission dips coming from the LC resonance red-shift by 50–100GHz. In addition to the LC resonance, dipole resonance occurs in the asymmetric structure which is formed by displacing the two splits away from the center in the same direction. Sensing characteristics by changes in the media surrounding the structures were demonstrated by simulations and experiments. By changing the surrounding medium from air to oil, we obtained resonant frequency red-shifts of 30–80GHz. This study may lead to the application of these terahertz devices as modulators and sensors. Increasing concerns over climate change have renewed interest in temperature effects on aquatic organisms. The effects of temperature, food concentration, population den-sity, and photoperiod on Daphnia carinata fitness were examined using life-table experiments in a full factorial design. The high temperature enhanced fitness at the high food concentration, but depressed it at the low food concentration. The tempera-ture-food interactions were significant in the four combinations of two population densities and two photoperiods. The temperature-population density interactions were significant in the two combinations of two photoperiods and the high food concentra-tion, in which the high population density masked the temperature effects. The temper-ature-photoperiod interaction was only significant in one combination with the low population density and the low food concentration, in which the long day length weak-ened the temperature effects. Such complicated interactions suggest that multiple eco-logical variables need to be taken into account simultaneously when assessing temperature effects on Zooplankton in nature. Abstract: Trunk kidney is a vital organ for excretion in teleosts. There have been sporadic reports of processing pathogens for the immune function in trunk kidney. However, molecular processes of pathogen recognition receptors (PRRs) responding to virus and viral/bacterial pathogen-associated molecular patterns (PAMPs) are poorly elucidated in trunk kidney. In the present study, we investigated transcriptional profiles of twelve representative immune-related genes (TLRs (TLR3, TLR7 and TLR22); RLRs (RIG-I, MDA5 and LGP2); NLRs (NOD1 and NOD2); adapter molecules (MyD88 and IPS-1); effector molecule type I interferon (IFN-I) and immunoglobulin M (IgM)) in trunk kidney tissue of grass carp (Ctenopharyngodon idella) (designated as Ci) injection of grass carp reovirus (GCRV) utilizing quantitative real-time RT-PCR (qRT-PCR). Furthermore, mRNA expression patterns of these genes (IgM excepted) were examined post GCRV infection and polyinosine-polycytidylic acid (poly(I:C)), lipopolysaccharide (LPS) or peptidoglycan (PGN) stimulation in primary trunk kidney cells of grass carp. The relative values of CiTLR3, CiTLR22 and CiMyD88 were increased post GCRV challenge and viral/bacterial PAMPs stimulation. The mRNA transcriptions of CiTLR7 were obviously activated with GCRV challenge. Remarkably, the mRNA expressions of CiRIG-I, CiMDA5, CiLGP2 and CiIPS-1 were largely up-regulated with GCRV challenge and viral/bacterial PAMPs stimulation. Interestingly, the expression tendencies of CiNOD1 and CiNOD2 were differential not only in GCRV challenge and poly(I:C) stimulation, but also in LPS and PGN stimulation. It was demonstrated that CiIFN-I induced powerful anti-viral and anti-bacterial effects in trunk kidney. In addition, the expression of CiIgM was induced at 72 h post GCRV injection in vivo. Collectively, these results suggest that trunk kidney of grass carp serves as an important immune organ, and plays crucial roles in triggering anti-viral and anti-bacterial immune responses both in vivo and in vitro. Static magnetic fields (SMFs), magnetic fields with constant intensity and orientation, have been extensively studied in the field of bone biology both fundamentally and clinically as a non-invasive physical factor. A large number of animal experiments and clinical studies have shown that SMFs have effective therapeutic effects on bone-related diseases such as non-healing fractures, bone non-union of bone implants, osteoporosis and osteoarthritis. The maintenance of bone health in adults depends on the basic functions of bone cells, such as bone formation by osteoblasts and bone resorption by osteoclasts. Numerous studies have revealed that SMFs can regulate the proliferation, differentiation, and function of bone tissue cells, including bone marrow mesenchymal stem cells (BMSCs), osteoblasts, bone marrow monocytes (BMMs), osteoclasts, and osteocytes. In this paper, the effects of SMFs on bone-related diseases and bone tissue cells are reviewed from both in vivo studies and in vitro studies, and the possible mechanisms are analyzed. This paper proposed a new remote sensing observation capability evaluation model (RSOCE) based on analytic hierarchy process to quantitatively evaluate the capability of multi-satellite cooperative remote sensing observation. The analytic hierarchical process model is a combination of qualitative and quantitative analysis of systematic decision analysis method. According to the objective of the remote sensing cooperative observation mission, we decompose the complex problem into several levels and a number of factors, compare and calculate various factors in pairs, and obtain the combination weights of different schemes. The model can be used to evaluate the observation capability of resource satellites. Taking the optical remote sensing satellites, such as China's resource satellite series and GF-4, as examples, this paper verifies and evaluates the model for three typical tasks: point target observation, regional target observation, and moving target continuous observation. The results show that the model can provide quantitative reference and model support for comprehensive evaluation of the collaborative observation capability of remote sensing satellites. An abrupt decrease in egg production and quality in laying hens can be brought on by the avian adenovirus known as the egg drop syndrome virus (EDSV). So far, there are no efficient commercial diagnostic approaches for EDSV. In the present study, we first purified two monoclonal antibodies (mAbs 5G4 and 6G6) specifically against the EDSV fibre protein. The mAb 5G4 was then employed as capture antibody. Additionally, the detection antibody 6G6 (HRP-6G6) was conjugated with horseradish peroxidase. Consequently, based on these two mAbs, we developed a sandwich enzyme-linked immunosorbent assay (ELISA) for EDSV detection. Specificity evaluation demonstrated that only EDSV gave a positive result while other avian viruses showed negative results. Sensitivity investigation showed that the limit of detection of EDSV was 102.9 TCID50/ml and the limit of detection of the purified His-Fiber was 5 ng/ml. Furthermore, the screening of samples from infected chickens showed that this method was very effective and could be applied to the detection of EDSV in infected samples. In summary, the sandwich enzyme-linked immunosorbent assay developed here provides an efficient, sensitive, and large-scale method for the specific diagnosis of EDSV. A sandwich ELISA was developed to detect EDSV using the mAbs 5G4 and HRP-6G6. The sandwich ELISA maintained high specificity and sensitivity. Background: H7N9 avian influenza virus (AIV) including highly and low pathogenic viruses have been detected in China since 2013. H7N9 AIV has a high mortality rate after infection in humans, and most human cases have close contacted with poultry in the live poultry market. Therefore, it is necessary to develop a rapid point-of-care testing (POCT) technique for H7N9 AIV detection. Methods: The H7N9 AIV was inactivated and purified, and was used as the antigen to immunize BALB/c. Twelve H7-HA specific monoclonal antibodies (McAbs) were produced through the hybridoma technique. The McAb 10A8 was conjugated with colloid gold as detecting antibody; McAb 9B6 was dispensed on the nitrocellulose membran as the capture test line and the Goat-anti mouse IgG antibody was dispensed as control line respectively. The immunochromatographic strip was prepared. Results: The analysis of ELISA and virus neutralization test showed that the obtained McAbs specifically recognized H7 HA. Based on the prepared strip, the detection of H7 AIV was achieved within 10 min. No cross-reaction occurred between H7 AIVs and other tested viruses. The detection limit of the strip for H7 was 2.4 log10EID50/0.1 mL for chicken swab samples. Conclusion: The McAbs were specific for H7 and the immunochromatographic strip developed in this study was convenient, rapid and reliable for the detection of H7 AIV. Summary: Pestiviruses, including classical swine fever virus, remain a concern for global animal health and are responsible for major economic losses of livestock worldwide. Despite high levels of vaccination, currently available commercial vaccines are limited by safety concerns, moderate efficacy, and required high doses. The development of new vaccines is therefore essential. Vaccine efforts should focus on optimizing antigen presentation to enhance immune responses. Here, we describe a simple herringbone‐dimer strategy for efficient vaccine design, using the classical swine fever virus E2 expressed in a rice endosperm as an example. The expression of rE2 protein was identified, with the rE2 antigen accumulating to 480 mg/kg. Immunological assays in mice, rabbits, and pigs showed high antigenicity of rE2. Two immunizations with 284 ng of the rE2 vaccine or one shot with 5.12 μg provided effective protection in pigs without interference from pre‐existing antibodies. Crystal structure and small‐angle X‐ray scattering results confirmed the stable herringbone dimeric conformation, which had two fully exposed duplex receptor binding domains. Our results demonstrated that rice endosperm is a promising platform for precise vaccine design, and this strategy can be universally applied to other Flaviviridae virus vaccines. Summary: Root anatomy plays important roles in the control of leaf water relations. However, few studies have evaluated whether and how anatomical traits of absorptive roots influence leaf physiology of herbaceous species in a temperate grassland.We measured absorptive root anatomical traits and leaf physiological traits of 15 herbaceous species in a temperate steppe and monitored their responses to increased precipitation in a field stimulating experiment.Root anatomical and leaf physiological traits differed among monocotyledonous grasses, monocotyledonous liliaceous species and dicotyledonous forbs. The species with higher stele: root diameter, lower root diameter and cortex thickness exhibited higher transpiration rates and stomatal conductance, but lower intrinsic water‐use efficiency. Increased precipitation enhanced transpiration and stomatal conductance of forbs and lilies, but it enhanced photosynthesis in lilies exclusively. The sensitive response of lilies to precipitation may be related to their large root diameter and cortex thickness.In summary, we observed distinct differences in anatomical traits of absorptive roots among plant groups in temperate steppes. These differences drove variations in leaf physiological traits and their diverse responses to precipitation change. Suppressor of cytokine signaling 3 (SOCS3) is a negative regulator of TBK1 and interferon pathway and the expression of SOCS3 is closely correlated with symptoms of influenza patients. However, whether deletion of Socs3 in the lung epithelial cells would affect influenza lung replication and inflammation in vivo is unknown. To test this, we approached the influenza infected Socs3f/f and SpcCre.Socs3f/f mice. We first found that knockdown of Socs3 in lung epithelial cells reduced influenza replication. However, in the in vivo study, there was a reduction of SOCS3 in the influenza-infected neutrophils coincided with an increase of SOCS3 in the CD45−CD326+ lung epithelial cells in PR8-infected SpcCre.Socs3f/f mice. SOCS3-deficient neutrophils expressed higher levels of IL-17 that enhanced chemokine expression in the lung epithelial cells. Lung SOCS3-dificient epithelial cells increased expression of GM-CSF and PGE 2 which promoted SpcCre.Socs3f/f neutrophils to yield SOCS3. SpcCre.Socs3f/f lung epithelial cells internalized SOCS3 released from GM-CSF + PGE 2 -stimulated SpcCre.Socs3f/f neutrophils, which could boost influenza replication in the lung epithelial cells. Thus, in the in vivo study, deletion of SOCS3 from lung epithelium could be nullified by the uptake from SOCS3 from infiltrated neutrophils. A strip test is described for the optical determination of influenza virus H3 subtype. It utilizes gold nanoparticle (AuNP) coated polystyrene latex microspheres (PS) as the label and a sandwich format. The AuNP and PS particles were linked using monoclonal antibodies against influenza virus as the bridge. Under the optimal conditions, the visual detection limit of the AuNP-PS-based strip test was as low as 1/16 hemagglutination unit (HAU). It was 64 times higher than that of 10 nm (4 HAU) AuNP-based strip tests. Quantitative analysis showed that the detection limit of the AuNP-PS-based strip is 0.016 HAU. Purpose: Mowing is an important management practice in the Inner Mongolia grassland and can significantly regulate plant and soil microbial communities. However, impact of long-term mowing on soil microbiota has been rarely evaluated in grassland ecosystems, and the effects of plant community on belowground microbiota under mowing were mainly focused on plant α-diversity, while the role of plant β-diversity is largely unknown. Methods: We examined the soil bacterial, fungal and protistan communities under the influence of a 17-year mowing treatment in a natural grassland of Inner Mongolia, and studied the relationship between the soil microbial communities and plant/soil parameters, and the correlations between β-diversity of the soil microbial and plant communities. We also constructed co-occurrence networks to investigate the legacy of mowing on the interactions between plant community and the three microbial groups (bacteria, fungi, protists). Results: We found that long-term mowing had significant effects on the community structures of plants and the three microbial groups. The microbial community structures mainly correlated to the plant parameters including aboveground biomass, plant litter, root carbon-to-nitrogen ratio, and plant Shannon index rather than to soil properties. Moreover, β-diversity of the soil microbial communities was significantly regulated by plant community β-diversity. Furthermore, mowing enhanced plant-microbe interactions in the networks under long-term mowing. Conclusions: Long-term mowing significantly alters plant and soil microbial community compositions and can reinforce their connections. The induced shifts of plant community attributes by long-term mowing rather than soil properties may be more effective factors in shaping soil microbial communities in grasslands. A multiplex reverse transcription polymerase chain rection (mRT-PCR) was developed for simultaneous detection of four RNA viruses in swine. The conserved target sequences directed to classical swine fever virus (CSFV), porcine reproductive and respiratory syndrome virus (PRRSV), porcine epidemic diarrhea virus (PEDV) and transmissible gastroenteritis coronavirus (TGEV) were selected based on alignments of genomic sequences and then specific primers were designed. The mRT-PCR assay was developed and evaluated for its specificity and sensitivity. The expected product from the single viral template was amplified by mRT-PCR and no spurious PCR amplification occurred from the genomic RNA or DNA of other pathogens. For single virus or different combinations of two viruses the detection limit of mRT-PCR was consistent with a single RT-PCR wtith 1 × 103 copies. For different combinations of the three viruses or four viruses, sensitivity of PEDV detection partially decreased. All of positive clinical specimens by the mRT-PCR were identically confirmed using Taqman RT-qPCR. Abstract Reinforcement bar corrosion induced by chloride ions ingress is one of the dominating causes for the deterioration of reinforced concrete structures in salty environment. Thus, it is particularly important to accurately predict chloride concentration in concretes and evaluate the structural service life. This paper presents the results of an indoor experimental investigation on chloride diffusion in plain and reinforced mortar specimens exposed to tidal zone, which is simulated by a self-design marine environment automatic simulation device. Concrete covering-layer was divided into major influence area (MIA) and secondary influence area (SIA) to accurately describe the chloride ion concentration distribution characteristics. MIA is the region affected by direct and indirect blocking effect of reinforcement bar, while SIA is the area only influenced by the indirect one. Also, the relative influence coefficient C re was defined to measure the impact of reinforcement bar on the chloride ion ingress. The results indicate that indirect blocking effect of rebar resulting from changes of material properties is the major cause for the chloride ion concentration distribution in SIA and it is only relative to the diffusion depth. Whereas, direct blocking effect of bar caused by physical obstruction to accumulate chloride ions contributes most to the chloride increase in MIA and it is correlated to the diffusion depth, exposure time and diameter of bar. Finally, an empirical prediction model of chloride concentration in concrete covering-layer considering the effect of reinforcement presence is proposed. Abstract: Objective: Yimitasvir phosphate, an inhibitor of nonstructural protein 5A (NS5A) replication complex of hepatitis C virus (HCV), was evaluated in a double‐blind, placebo‐controlled, parallel, multiple‐dose study. Methods: Twenty‐four patients with chronic HCV genotype 1 infection were randomized to receive a 7‐day course of yimitasvir phosphate at daily doses of 30, 100 or 200 mg or placebo. Antiviral efficacy, resistance profile, pharmacokinetics (PK), safety and tolerability were assessed. Key findings: The maximal reduction in HCV RNA from baseline was 5.17 log10 IU/ml. However, most patients experienced viral rebound on or before day 3 after yimitasvir treatment was initiated. The PK profile revealed median peak plasma concentrations at 4–12 h postdose and a mean terminal half‐life of 14.47–17.09 h, the basis for daily dosing. Steady drug state was achieved following 5 days of daily dosing. The accumulation rate was low (1.29–1.73). There were no significant alterations in vital signs and laboratory findings among all participants. Conclusions: This study shows that yimitasvir phosphate was well tolerated, and the PK profile supported daily dosing regimens. Many aquatic organisms respond phenotypically, through morphological, behavioral, and physiological plasticity, to environmental changes. The small-size cladoceran Bosmina longirostris, a dominant zooplankter in eutrophic waters, displayed reduced growth rates in response to the presence of a toxic cyanobacterium, Microcystis aeruginosa, in their diets. The magnitude of growth reduction differed among 15 clones recently isolated from a single population. A significant interaction between clone and food type indicated a genetic basis for the difference in growth plasticity. The variation in phenotypic plasticity was visualized by plotting reaction norms with two diets. The resistance of each clone to dietary cyanobacteria was measured as the relative change in growth rates on the “poor” diet compared with the “good” diet. The enhanced resistance to M. aeruginosa in B. longirostris was derived from both the reduced slope of reaction norms and the increased mean growth rates with two diets. The large clonal variation within a B. longirostris population may contribute to local adaptation to toxic cyanobacteria and influence ecosystem function via clonal succession. BACKGROUND Fumonisins are a kind of toxic and carcinogenic mycotoxin. A rapid immunochromatographic test strip has been developed for simultaneous detection of fumonisin B1, B2 and B3 ( FB1, FB2 and FB3) in maize based on colloidal gold-labelled monoclonal antibody ( McAb) against FB1 probe. RESULTS The anti- FB1 McAb ( 2E11-H3) was produced through immunisation and cell fusion, and identified as high affinity, specificity and sensitivity. The cross-reaction ratios with fumonisin B2 and B3 were accordingly 385% and 72.4%, while none with other analogues. The colloid gold-labelled anti- FB1 McAb probe was successfully prepared and used for establishing the immunochromatographic strip. The test strip showed high sensitivity and specificity, the IC50 for FB1 was 58.08 ng mL−1, LOD was 11.24 ng mL−1, calculated from standard curve. Moreover, the test strip exhibited high cross-reactivity with FB2 and FB3, and could be applied to the simultaneous detection of FBs ( FB1: FB2: FB3 = 12:4:1) in maize sample with high accuracy and precision. The average recoveries of FBs in maize ranged from 90.42% to 95.29%, and CVs were 1.25-3.77%. The results of the test strip for FBs samples showed good correlation with high-performance liquid chromatography analysis. In a high-level radioactive waste (HLW) repository, pH has an impact on the solubility, migration, and adsorption of radionuclides. Thus, understanding the effects of pH on the diffusion of radionuclides is essential for long-term disposal of HLW. In this work, the diffusion behaviors of Re(VII) and Se(IV) in compacted Gaomiaozi (GMZ) bentonite at different pH have been investigated by a through-diffusion method. The effective diffusion coefficient, i.e., D e values of Re(VII) and Se(IV) were in the range of (1.0–2.4) × 10 −11 m 2 /s at pH 3.0–10.0 and (0.38–2.3) × 10 −11 m 2 /s at pH 3.0–9.0. In the case of Re(VII), the D e values remained almost unchanged probably because ReO 4 − was the dominant species in the pH range of 3.0–10.0. In the case of Se(IV), whose predominant species were HSeO 3 − at pH < 9.0 and SeO 3 2− at pH ≥ 9.0, the D e values decreased by a factor of 3–6 at pH 9.0, i.e., D e (pH < 9.0)/ D e (pH 9.0) ≈ 3–6, implying that the species with a higher valence state had a stronger anion exclusion effect. The decrease in D e values can be explained by the diffusion species of Se(IV). Additionally, the rock capacity factor α decreased with the increase of pH. HSeO 3 − was absorbed on GMZ bentonite with distribution coefficient K d values in the range of (1.0–2.5) × 10 −4 m 3 /kg at pH ≤ 8.0, whereas SeO 3 2− was negligibly sorbed at pH > 8.0. In order to understand the diffusion properties of anionic radionuclides (RN) in bentonite in the presence of organic compounds with chelating and reducing properties, the effect of humic acid (HA), citrate, oxalate and formate on Re(VII) diffusion was investigated by a through-diffusion method under different pH (pH 7.0 and 10.0) and temperature conditions (10–65 °C). When the molar ratio of oxalate to Re(VII) was higher than 1:1, the accumulated mass and flux were observed to decrease drastically as a function of time. It can be explained by photocatalytic impact of Fe(III) on the reduction of Re(VII) to insoluble Re(IV) by oxalate. Thus, the diffusion of Re(VII) was also observed to decrease in the presence of minor citrate, oxalate and formate. Effective diffusion coefficient (D e ) values decreased from 8.7 × 10 − 11 m 2 /s to (5.1–5.9) × 10 − 11 m 2 /s. In contrast, HA had no significant impact on Re(VII) diffusion. The D e values of 1.54 × 10 − 11 and 6.5 × 10 − 12 m 2 /s were obtained at pH 7.0 and 10.0, respectively. HA thus neither has an effect on the speciation of Re(VII) nor does it change the charge of the surface. The latter is probably due to the fact that HA preferentially interacts with the aluminol sites of the montmorillonite edges without changing the zeta potential of the surface. The dependence of D e of Re(VII) on temperature in the absence and presence of citrate or formate was described by the Arrhenius equation. Iron overload is a risk factor for osteoporosis due to its oxidative toxicity. Previous studies have demonstrated that an excessive amount of iron increases osteocyte apoptosis and receptor activator of nuclear factor κ-B ligand (RANKL) production, which stimulates osteoclast differentiation in vitro. However, the effects of exogenous iron supplementation-induced iron overload on osteocytes in vivo and its role in iron overload-induced bone loss are unknown. This work aimed to develop an iron overloaded murine model of C57BL/6 mice by intraperitoneal administration of iron dextran for two months. The iron levels in various organs, bone, and serum, as well as the microstructure and strength of bone, apoptosis of osteocytes, oxidative stress in bone tissue, and bone formation and resorption, were assessed. The results showed that 2 months of exogenous iron supplementation significantly increased iron levels in the liver, spleen, kidney, bone tissue, and serum. Iron overload negatively affected bone microstructure and strength. Osteocyte apoptosis and empty lacunae rate were elevated by exogenous iron. Iron overload upregulated RANKL expression but had no significant impact on osteoprotegerin (OPG) and sclerostin levels. Static and dynamic histologic analyses and serum biochemical assay showed that iron overload increased bone resorption without significantly affecting bone formation. Exogenous iron promoted oxidative stress in osteocytes in vivo and in vitro. Additional supplementation of iron chelator (deferoxamine) or N-acetyl- l -cysteine (NAC) partially alleviated bone loss, osteocyte apoptosis, osteoclast formation, and oxidative stress due to iron overload. The mixed infection of duck hepatitis A virus 3 (DHAV-3) and novel duck reovirus (NDRV) has caused significant losses to the global duck farming industry. On-site point-of-care testing of viruses plays a crucial role in the early diagnosis, prevention, and disease control. Here, we proposed an RPA-CRISPR Cas12a/Cas13a one-pot strategy (DRCFS) for rapid and simultaneous detection of DHAV-3 and NDRV. This method integrated the reaction of RPA and CRISPR Cas12a/Cas13a in a single tube, eliminating the need to open the lid during the intermediate processes and thereby avoiding aerosol contamination. On this basis, we proposed a dual RPA-CRISPR strategy coupled with a lateral flow analysis platform (DRC-LFA). This circumvented the necessity for complex instruments, enabling direct visual interpretation of results, making the test more accessible and user-friendly. Our findings demonstrated that the DRCFS method could detect DHAV-3 and NDRV at concentrations as low as 100 copy/μL, while DRC-LFA achieved limit of 101 copies/μL within 35 min. Furthermore, when DRCFS, DRC-LFA, and qPCR were employed collectively for clinical samples analysis, all three methods yielded consistent results. Background: Classical swine fever (CSF) virus is the causative agent of an economically important, highly contagious disease of pigs. CSFV is genetically and serologically related to bovine viral diarrhea virus (BVDV). BVDV infection in pigs can mimic CSF clinical signs, which cause difficulty in differentiation. Serological test for detection of virus specific antibodies is a valuable tool for diagnosis and surveillance of CSFV and BVDV infections in animals. The aim of this study was to develop the CSFV Erns and BVDV tE2 -based ELISAs to distinguishably test specific antibodies against CSFV and BVDV. Methods: The CSFV Erns and truncated E2 (tE2, residues 690–865) of BVDV were expressed in E. coli and purified by Ni–NTA affinity chromatography, respectively. Employing Erns or tE2 protein as diagnostic antigen, indirect ELISAs were developed to distinguishably test specific antibodies against CSFV and BVDV. The specificity and sensitivity of ELISAs were evaluated using a panel of virus specific sera of pigs, immunized rabbits and immunized mice. A total 150 clinical serum samples from farm pigs were measured by the developed ELISAs and compared with virus neutralizing test (VNT). Results: Indirect ELISA was established based on recombinant CSFV Erns or BVDV tE2 protein, respectively. No serological cross-reaction between antibodies against CSFV and BVDV was observed in sera of immunized rabbits, immunized mice or farm pigs by detections of the Erns and tE2 -based ELISAs. Compared to VNT, the CSFV Erns -based ELISA displayed a high sensitivity (93.3%), specificity (92.0%) and agreement rate (92.7%), and the sensitivity, specificity and agreement rate of BVDV tE2 -based ELISA was 92.3%, 95.2% and 94.7%, respectively. African swine fever (ASF) caused by African swine fever virus (ASFV) is becoming a serious threat to the swine industry worldwide. CD2v is a key pathogenic factor of ASFV and the protective antigen with low immunogenicity, whereas viral protein-based nanoparticles have advantages of precise assembly and high immunogenicity. In this study, the CD2v protein fused with Norovirus (NoV) P particle assembled into nanoparticle for improved immunogenicity. Then, CD2v protein nanoparticle and monomer CD2v protein were expressed in HEK293F cells. The former induced higher levels of antibodies, and thus highly potent monoclonal antibodies (mAbs) were generated and characterized. The highest antibody titration of mAb 10A3 reached 1:2048000, and mAb 2E9 had the highest inhibition percent of 84% when competed with ASFV positive serum. Meanwhile, all mAbs reacted specifically with the denatured CD2v protein, and the linear epitope with the location of amino acids 28th to 51st of CD2v extracellular domain sequence was identified. In summary, this study produced a highly immunogenic CD2v protein and generated high-titer mAbs, the precise location of linear epitope on the CD2v was further determined. Summary: Reduced substrates produced by the serpentinization reaction under hydration of olivine may have fuelled biological processes on early Earth. To understand the adaptive strategies and carbon metabolism of the microbes in the serpentinizing ecosystems, we reconstructed 18 draft genomes representing dominant species of Omnitrophicaeota, Gammaproteobacteria and Methanobacteria from the Manleluag serpentinizing spring in Zambales, Philippines (hyperalkaline and rich in methane and hydrogen). Phylogenomics revealed that two genomes were affiliated with a candidate phylum NPL‐UPA2 and the references of all our genomes were derived from ground waters, hot springs and the deep biosphere. C1 metabolism appears to be widespread as most of the genomes code for methanogenesis, CO oxidation and CO2 fixation. However, likely due to the low CO2 concentration and election acceptors, the biomass in the spring was extremely low (<103 cell/ml). Various Na+ and K+ transporters and Na+‐driving ATPases appear to be encoded by these genomes, suggesting that nutrient acquisition, bioenergetics and normal cytoplasmic pH were dependent on Na+ and K+ pumps. There are numerous studies that investigate the effects of static magnetic fields (SMFs) on osteoblasts and osteoclasts. However, although osteocytes are the most abundant cell type in bone tissue, there are few studies on the biological effects of osteocytes under magnetic fields. Iron is a necessary microelement that is involved in numerous life activities in cells. Studies have shown that high static magnetic fields (HiSMF) can regulate cellular iron metabolism. To illustrate the effect of HiSMF on activities of osteocytes, and whether iron is involved in this process, HiSMF of 16 tesla (T) was used, and the changes in cellular morphology, cytoskeleton, function-related protein expression, secretion of various cytokines, and iron metabolism in osteocytes under HiSMF were studied. In addition, the biological effects of HiSMF combined with iron preparation and iron chelator on osteocytes were also investigated. The results showed that HiSMF promoted cellular viability, decreased apoptosis, increased the fractal dimension of the cytoskeleton, altered the secretion of cytokines, and increased iron levels in osteocytes. Moreover, it was found that the biological effects of osteocytes under HiSMF are attenuated or enhanced by treatment with a certain concentration of iron. These data suggest that HiSMF-regulated cellular iron metabolism may be involved in altering the biological effects of osteocytes under HiSMF exposure. [ABSTRACT FROM AUTHOR]

Middle East respiratory syndrome coronavirus (MERS-CoV) is a coronavirus which can cause severe human respiratory diseases with a fatality rate of almost 36%. In this study, we report the generation, characterization and epitope mapping of several monoclonal antibodies against the spike receptor-binding domain (RBD) of MERS-CoV. Two monoclonal antibodies (4C7 and 6E8) that can react with linearized RBD have been selected for subsequent identification of RBD mAb-binding epitopes. Two distinct novel linear epitopes, 423FTCSQIS429 and 546SPLEGGGWL554,were precisely located at the outermost surface of RBD by dot-blot hybridization and ELISAs. Multiple sequence alignment analysis showed that these two peptides were highly conserved. Alanine (A)-scanning mutagenesis demonstrated that residues 423F, 428I, and 429S are the crucial residues for the linear epitope 423FTCSQIS429 while residues 548L, 550G, 553W, 554L for epitope 546SPLEGGGWL554. Abstract: ADAR (adenosine deaminase acting on RNA) is an RNA editing enzyme that targets both coding and noncoding dsRNAs (double stranded RNAs) and converts adenosine to inosine, which is read by translation machinery and by polymerases during RNA-dependent RNA replication as if it is guanosine. This editing is a widespread post-transcriptional modification event in animals. In this study, we identified the full-length cDNA sequence of Ctenopharyngodon idella ADAR1 (designated as CiADAR1) and detected the mRNA expression patterns in response to dsRNA (polyinosinic-polycytidylic acid sodium salt, poly(I:C)) and grass carp reovirus (GCRV). CiADAR1 is a large gene in size, consisting of 4833 nucleotides encoding a protein of 1392 amino acids. The deduced amino acid sequence contains seven putative domains: one proline-rich region (Pro-R), two Z-DNA-binding domains (Zalpha), three dsRNA binding motifs (DSRM) and one tRNA-specific and dsRNA adenosine deaminase domain (ADEAMc). It is most homologous to Danio rerio ADAR (E-value = 0.0, identities = 80% (1110/1395)), also close homology to Homo sapiens ADAR1 (E-value = 0.0, identities = (47%) 530/1122). CiADAR1 mRNA was investigated in fifteen tissues, and was low expressed in muscle and head kidney tissues, high in blood and spleen tissues by quantitative real-time RT-PCR (qRT-PCR). mRNA expressions of CiADAR1 were significantly up-regulated and reached peak at 24 h post GCRV challenge in vivo and in vitro (P < 0.05). After poly(I:C) stimulation at different concentrations, CiADAR1 transcripts were rapidly and significantly up-regulated and recovered in dose-dependent and time-dependent manners (P < 0.05). The results indicate CiADAR1 was implicated in the antiviral immune response and laid the foundation for further studies on functions and mechanisms of RNA editing in fishes. Platelet dysfunction plays a critical role in vascular complications of type 2 diabetes mellitus (T2DM). But the relationship between platelet hyperactivity and its energy metabolic process remains unclear. This study was designed to explore alterations of platelet mitochondrial ATP production and the possible mechanism. A total of 39 T2DM patients without macrovascular and microvascular complications and 32 normal controls were fasting sampled. Platelet ATP content was measured by a high performance liquid chromatograph (HPLC). The flow cytometry technique was adopted to evaluate mitochondrial membrane potential (ΔΨm), the stored force for platelet ATP production. Consequently, T2DM patients exhibited obvious hyperglycemia, hyperlipidemia and hypertension, but normal platelet morphology. Platelet ATP content was significantly higher in T2DM (0.032 ± 0.010 µmol/109 platelets versus 0.017 ± 0.006 µmol/109 platelets, p < 0.001) than in the control group. Interestingly, ΔΨm was markedly decreased in T2DM patients (0.79 ± 0.18 versus 2.70 ± 1.03, p < 0.001) compared with normal controls. For whole subjects, a stepwise regression showed that plasma glycated hemoglobin A1c (HbA1c) level positively correlated to platelet ATP content (β = 0.552, 95% CI = 0.072–1.451), and fasting plasma glucose (FPG) level was negatively correlated to ΔΨm (β = −0.372, 95% CI = −0.471 to −0.089). These data support that hyperglycemia of T2DM promotes platelet mitochondria to generate more ATP, but decreases platelet mitochondrial potential. Receptors for the Fc region (FcγRs) of immunoglobulin G (IgG) play a crucial role in the immune system and host protection against infection. In this study, we describe the cloning, sequencing, and expression of the high-affinity IgG receptor from pig. By screening a translated Expressed Sequence Tags database with the human FcγRI (CD64) protein sequence, we identified a putative porcine homologue. Subsequent polymerase chain reaction amplification confirmed that the identified full-length cDNA was expressed in porcine cells. Rosetting analysis shows that COS-7 cells transfected with a plasmid containing the cloned cDNA were able to bind chicken erythrocytes sensitized with porcine IgG. Scatchard analysis indicated that monomeric IgG bound to transiently transfected cells with an affinity of approximately 4×107 M−1. The porcine FcγRI cDNA is 1,038 nucleotides long and is predicted to encode a 346-amino-acid transmembrane glycoprotein composed of three Ig-like domains, a transmembrane region, and a short cytoplasmic tail. The overall identity of the porcine FcγRI to its human and mouse counterparts at the level of the amino acid sequence was 75% and 57%, respectively. Classical swine fever virus (CSFV) is a member of the genus Pestivirus, which causes serious economic losses. The re-emergence of the disease in Japan in 2018 has increased awareness of CSFV. In this study, Balb/c mice were immunized with plant-derived E2 protein, and four monoclonal antibodies (mAbs) 4B11, 7B3, 11A5 and 6F3 were generated. Two of these mAbs, 4B11 and 7B3, effectively blocked CSFV infection of PK-15 cells. Both mAbs recognized a novel linear epitope, 256CLIGNTTVKVHASDER271. The neutralizing ability of anti-CSFV serum decreased 63%, when pre-incubated with the linear peptide at 200 μg/mL. Structural analysis showed that this linear epitope is present at the border of Domain C and Domain D on the surface of the E2 protein. Alignment of amino acid sequences showed that the epitope was conserved in different subgroups of CSFV but not in other members of the Pestivirus genus. Consistently with the analysis above, this epitope distinguished antibodies against CSFV from those against bovine viral diarrhea virus (BVDV). Our study provides an ideal candidate peptide for new vaccine design and differential diagnosis of CSFV. This study developed somatic embryogenesis protocols for Picea pungens (Engelm), an important ornamental species, including initiation, proliferation, maturation, germination, and acclimation. Somatic embryogenic tissues were induced from mature zygotic embryos of five families, with a frequency of ≥ 22% for each. Embryogenic tissues (ET) from 13 clones of three families were proliferated for one week, achieving an average rate of 179.1%. The ET of 38 clones of three families were cultured in maturation medium for six weeks; 188 mature embryos on average were counted per gram ET cultured, of which ≥ 81.1% appeared normal, and each clone developed at least 28 normally matured embryos. A total of 69.9% or more of cotyledonary somatic embryos germinated normally and developed into normal emblings. The experiment of transplanting the emblings into a greenhouse had an average survival rate of 68.5%. Considerable variation among and within families during initiation and proliferation was observed, but this variation decreased in the maturation and germination. Changing the concentration of plant growth regulator of the initiation medium did not significantly change the initiation frequency. We recommend incorporating these protocols into the current Picea pungens practical programs, although further research is essential to increase efficiencies and reduce cost. [ABSTRACT FROM AUTHOR]
