Travelling waves, such as light and sound, can carry angular momentum. Orbital angular momentum (OAM) is one of the components which is determined by the helicity of the phase fronts. The helical waveform is characterised in terms of an integer l and an azimuthal phase term of exp(âˆ'i lθ), but for | l | > 1 the resulting high-order beam structure is unstable to perturbation. In this work, using Fourier transform profilometry and stroboscopic imaging techniques, we demonstrate the real-time visualisation of the OAM-carrying acoustic waveform by imaging the pressure imprint of the acoustic wave on a thin rubber sheet. Furthermore, based on the visualised waveform, we are able to optimise high-order (| l | > 1) OAM states by controlling the individual elements of the acoustic source. Beyond the study of acoustic OAM, the real-time monitoring and optimising methods could be a benefit to other applications requiring acoustic waveform shaping, such as acoustic communications, acoustic holograms, etc. [ABSTRACT FROM AUTHOR]

Cyanobacteria are attractive microbial hosts for production of chemicals using light and CO2. However, their low productivity of chemicals is a major challenge for commercial applications. This is mostly due to their relatively slow growth rate and carbon partitioning toward biomass rather than products. Many cyanobacterial strains synthesize sucrose as an osmoprotectant to cope with salt stress environments. In this study, we harnessed the photosynthetic machinery of the fast-growing cyanobacterium Synechococcus elongatus UTEX 2973 to produce sucrose under salt stress conditions and investigated if the high efficiency of photosynthesis can enhance the productivity of sucrose. By expressing the sucrose transporter CscB, Synechococcus 2973 produced 8 g L−1 of sucrose with a highest productivity of 1.9 g L−1 day−1 under salt stress conditions. The salt stress activated the sucrose biosynthetic pathway mostly via upregulating the sps gene, which encodes the rate-limiting sucrose-phosphate synthase enzyme. To alleviate the demand on high concentrations of salt for sucrose production, we further overexpressed the sucrose synthesis genes in Synechococcus 2973. The engineered strain produced sucrose with a productivity of 1.1 g L−1 day−1 without the need of salt induction. In various nonlinear extended media, increasing driving causes the transition from the ordered state to the turbulent state. For the hydrodynamic flow, the transition starts with spreading and decaying intermittent turbulent puffs in the laminar background, followed by the emergence of one percolating turbulent puff leading to full turbulence. Nevertheless, for nonlinear waves, the transition scenario and the corresponding spatiotemporal waveform dynamics from the ordered wave to wave turbulence still remain elusive. Here, these issues are experimentally explored in three dimensional traveling dust acoustic waves of the dusty plasma system through direct visualization. Turbulent sites (TSs) with a wide instantaneous bandwidth in the 2 + 1D spatiotemporal space are identified through wavelet transform. It is found that the transition from the plane wave to the weakly disordered states starts with a small fraction of emerging and decaying TSs mainly clustering around defect filaments with null wave amplitude, in the ordered wave background. Further transition to turbulence with a smooth rapid increase in the fraction of TSs, which exhibit scale-free cluster size distributions and eventually percolate through the 2 + 1D space, is similar to the percolating turbulent transition in hydrodynamic flows. We experimentally study the spatiotemporal dynamics of self-excited shedding of millimeter-sized water drops acoustically levitated in a single-node standing wave cavity. By decreasing the sound intensity below the threshold, the interplay of drop motion and its perturbed acoustic wave field lead to the transition from stable self-excited drop oscillation to chaotic drop oscillation with growing fluctuations and intermittent droplet shedding. Using azimuthal Fourier transform, the top-view drop shape can be decomposed into zonal and sectoral modes with varying amplitudes. The shedding is led by the increasing amplitudes of the low order sectoral modes (azimuthal mode number m = 2 and 3), which cause the strongest amplitude in the zonal mode (m = 0) in the re-expansion stage after the shrinking of the side lobes in the low order modes. It in turn causes synchronized excitations of high order sectoral modes with m > 3. Their constructive superposition at certain points along the flattened thin edge of the re-expanding drop leads to sharp protrusions, where the surface tension cannot hold the thin rapid expanding jets, and shedding occurs. Background/Objectives: Benign paroxysmal positional vertigo (BPPV) is the most common cause of recurrent vertigo and the most common peripheral vestibular disorder. It is characterized by intense vertigo triggered by head and position changes. This study investigates the risk of subsequent injury in BPPV patients and the effects of treatment. Methods: A population-based retrospective cohort study was conducted using data from the Longitudinal Health Insurance Database 2005 in Taiwan. Patients with and without BPPV were identified between 2000 and 2017. The study outcomes were diagnoses of all-cause injuries. The Kaplan–Meier method determined the cumulative incidence rates of injury in both cohorts, and a log-rank test analyzed the differences. Cox proportional hazard models calculated each cohort's 18-year hazard ratios (HRs). Results: We enrolled 50,675 patients with newly diagnosed BPPV and 202,700 matched individuals without BPPV. During follow-up, 47,636 patients were diagnosed with injuries (13,215 from the BPPV cohort and 34,421 from the non-BPPV cohort). The adjusted HR for injury in BPPV patients was 2.63 (95% CI, 2.49–2.88). Subgroup analysis showed an increased incidence of unintentional and intentional injuries in BPPV patients (aHR 2.86; 95% CI, 2.70–3.13 and 1.10; 95% CI, 1.04–1.21, respectively). A positive dose–response relationship was observed with increasing BPPV diagnoses. Treatment with canalith repositioning therapy (CRT) or medications reduced the risk of injury slightly but not significantly (aHR, 0.78; 95% CI, 0.37–1.29, 0.88; 95% CI, 0.40–1.40, respectively). Conclusions: BPPV is independently associated with an increased risk of injuries. CRT or medications have limited effects on mitigating this risk. The natural compound n-butylidenephthalide (BP), which is isolated from the chloroform extract of Angelica sinensis, has been investigated for its antitumoral effects on glioblastoma multiform (GBM) brain tumors both in vitro and in vivo. To determine the mechanism of BP-induced growth arrest and apoptosis, we examined BP-induced changes in gene expression by microarray screening using human GBM brain tumor cells. This analysis identified several BP-inducible genes, including the nuclear receptors NOR-1, Nurr1, and Nur77. Among these genes, Nur77 is particularly interesting because it plays an important role in the apoptotic processes in various tumor cell lines. BP was able to increase Nur77 mRNA and protein expression in a time-dependent manner. After BP treatment in GBM 8401 cells, Nur77 translocated from the nucleus to the cytoplasm, the cytochrome c was released from the mitochondria, and caspase 3 became activated. Furthermore, using Nur77 promoter-luciferase assay, BP increased Nur77 was AP1 related. Inhibition of BP-induced Nur77 expression by Nur77 short interfering RNA blocked BP-induced apoptosis in GBM 8401 cells, suggesting that the induction of Nur77 negatively affected GBM 8401 cell survival. Osteoarthritis (OA) is a degenerative disease that causes pain, cartilage deformation, and joint inflammation. Mesenchymal stem cells (MSCs) are potential therapeutic agents for OA treatment. However, the 2D culture of MSCs could potentially affect their characteristics and functionality. In this study, calcium-alginate (Ca-Ag) scaffolds were prepared for human adipose-derived stem cell (hADSC) proliferation with a homemade functionally closed process bioreactor system; the feasibility of cultured hADSC spheres in heterologous stem cell therapy for OA treatment was then evaluated. hADSC spheres were collected from Ca-Ag scaffolds by removing calcium ions via ethylenediaminetetraacetic acid (EDTA) chelation. In this study, 2D-cultured individual hADSCs or hADSC spheres were evaluated for treatment efficacy in a monosodium iodoacetate (MIA)-induced OA rat model. The results of gait analysis and histological sectioning showed that hADSC spheres were more effective at relieving arthritis degeneration. The results of serological and blood element analyses of hADSC-treated rats indicated that the hADSC spheres were a safe treatment in vivo. This study demonstrates that hADSC spheres are a promising treatment for OA and can be applied to other stem cell therapies or regenerative medical treatments. [ABSTRACT FROM AUTHOR]

Background: Thoracic ossification of the ligamentum flavum (TOLF), a rare condition more prevalent in East Asia, is managed through open and endoscopic surgical approaches. Determining the superior surgical option remains unclear. This study assesses the safety and clinical outcomes associated with these approaches in TOLF patients.Following Preferred Reporting Items for Systematic Reviews and Meta-Analyses (PRISMA) guidelines, we conducted a systematic literature search up to August 5, 2023, across PubMed, Scopus, EMBASE, Web of Science, Cochrane, and ClinicalTrials.gov. We included randomized controlled trials and cohort studies reporting complication rates, mJOA (modified Japanese Orthopedic Association) scores, JOA scores, VAS (Visual Analog Scale) scores, or hospitalization duration for both open and endoscopic surgeries in TOLF patients.We analyzed 37 studies encompassing 1,646 TOLF patients using a random-effects model. Our findings revealed a significant difference in complication rates (overall complication rates: 0.12; 95% CI: 0.07, 0.19; p < 0.01; I2: 69%; quality of evidence: moderate), with lower complication rates in the endoscopy group. However, no significant differences were observed in JOA scores (overall JOA: 8.35; 95% CI: 7.16, 9.54; p = 0.12; I2: 99%; quality of evidence: very low), VAS scores (overall VAS: 1.31; 95% CI: 1.03, 1.59; p = 0.35; I2: 91%; quality of evidence: very low), or hospitalization duration (hospital stay: 10.83 days; 95% CI: 6.86, 14.80; p = 0.35; I2: 91%; quality of evidence: very low) between the open and endoscopic groups.This meta-analysis reports lower complication rates and improved postoperative mJOA scores for endoscopic surgery in TOLF patients compared to open surgery. It represents the first comprehensive evaluation of clinical outcomes and safety of different surgical approaches for TOLF patients. Abstract: Electromagnetic radiation can be categorized into ionizing and non-ionizing varieties. To determine the mechanism how non-ionizing radiation affects biological cells, we analyzed the difference between its thermal and electromagnetic effects. Two-beam optical tweezers were designed to demonstrate that infrared radiation could enhance the cellular interaction between red blood cells by its electromagnetic nature. An IR spot in the optical tweezers was irradiated on two RBCs to polarize them and induce electromagnetic attraction, while the other focused visible spot was used to quantify the intensity of the intercellular interaction. It was found that 0.1mW/μm2 infrared radiation was adequate to cause pN-scale interaction between a cell pair, which was only 1/1000 of the power density used in a CD-R drive. We then set up a model to describe how non-ionizing radiation affected a cell assembly by deriving electromagnetic micro-stress transverse to its propagation axis. [Copyright &y& Elsevier]

Cognitive impairment is a serious side effect of post-myocardial infarction (MI) course. We have recently demonstrated that human adipose-derived stem cells (hADSCs) ameliorated myocardial injury after MI by attenuating reactive oxygen species (ROS) levels. Here, we studied whether the beneficial effects of intramyocardial hADSC transplantation can extend to the brain and how they may attenuate cognitive dysfunction via modulating ROS after MI. After coronary ligation, male Wistar rats were randomized via an intramyocardial route to receive either vehicle, hADSC transplantation (1 × 106 cells), or the combination of hADSCs and 3-Morpholinosydnonimine (SIN-1, a peroxynitrite donor). Whether hADSCs migrated into the hippocampus was assessed by using human-specific primers in qPCR reactions. Passive avoidance test was used to assess cognitive performance. Postinfarction was associated with increased oxidative stress in the myocardium, circulation, and hippocampus. This was coupled with decreased numbers of dendritic spines as well as a significant downregulation of synaptic plasticity consisting of synaptophysin and PSD95. Step-through latency during passive avoidance test was impaired in vehicle-treated rats after MI. Intramyocardial hADSC injection exerted therapeutic benefits in improving cardiac function and cognitive impairment. None of hADSCs was detected in rat's hippocampus at the 3rd day after intramyocardial injection. The beneficial effects of hADSCs on MI-induced histological and cognitive changes were abolished after adding SIN-1. MI-induced ROS attacked the hippocampus to induce neurodegeneration, resulting in cognitive deficit. The remotely intramyocardial administration of hADSCs has the capacity of improved synaptic neuroplasticity in the hippocampus mediated by ROS, not the cell engraftment, after MI. Abstract: Aims: To investigate the feasibility and mechanism of liver damage repair using human bone marrow mesenchymal stem cells (hBMMSCs), we investigated the potential for hBMMSCs in recovery from liver damage, including fibrotic liver repair, using the CCl4-induced model for liver damage in the rat. Main methods: Rats were injected with 0.5 ml/kg CCl4 to induce liver damage and progressive liver fibrosis. hBMMSCs labeled with GFP were injected into the rats through the portal vein. Key findings: After one day of transplantation, GFP-labeled cells were found around the liver lobules, the hepatic blood vessels, and the edge of the liver lobes. Biochemical and histopathological analyses showed significantly increased recovery from liver damage in the transplanted group. In addition, transplanted hBMMSCs express matrix metalloproteinases (MMP), and liver fibrosis was significantly decreased. The degree of fibrosis reduction paralleled the number of hBMMSCs observed in liver sections. Significance: Our data suggest that hBMMSCs may facilitate recovery from chronic liver damage and may decrease liver fibrosis. Adipose-derived stem cell (ADSC) transplantation is a promising new therapy to improve cardiac function after myocardial infarction. However, its low efficacy of transdifferentiation hampers its usefulness. Glycogen synthase kinase-3β (GSK-3β) signal has been shown to play a role in preconditioning-induced cardioprotection. We assessed whether n- butylidenephthalide (BP) primed ADSCs can attenuate arrhythmias by a GSK-3β-dependent pathway after myocardial infarction. Male Wistar rats after coronary ligation was randomly allocated to receive intramyocardial injection of vehicle, ADSCs, BP-preconditioned ADSCs, (BP + lithium)-preconditioned ADSCs, (BP + SB216763)-preconditioned ADSCs, and (BP + LY294002)-preconditioned ADSCs. ADSCs were primed for 16 h before implantation. After 4 weeks of implantation, ADSCs were retained in myocardium, reduced fibrosis and improved cardiac function. Sympathetic hyperinnervation was blunted after administering ADSCs, assessed by immunofluorescent analysis, and Western blotting and real-time quantitative RT-PCR of nerve growth factor. Arrhythmic scores during programmed stimulation in the ADSC-treated infarcted rats were significantly lower than vehicle. BP-preconditioned ADSCs had superior cardioprotection, greater ADSC engraftment and transdifferentiation, and antiarrhythmic effects compared with ADSCs alone. Simultaneously, BP increased the levels of phospho-Akt and down-regulated GSK-3β activity. The effects of BP against sympathetic hyperinnervation were blocked by LY294002, a PI3K inhibitor. Addition of either lithium or SB216763 did not have additional effects compared with BP alone. Compared with ADSC alone, BP-primed ADSC implantation improved stem cell engraftment and attenuated sympathetic hyperinnervation and arrhythmias through a PI3K/Akt/GSK-3β-dependent pathway, suggesting that a synergic action was achieved between BP pretreatment and ADSCs. Abstract: High levels of arsenic in groundwater and drinking water represent a major health problem worldwide. Drinking arsenic-contaminated groundwater is a likely cause of blackfoot disease (BFD) in Taiwan, but mechanisms controlling the mobilization of arsenic present at elevated concentrations within aquifers remain understudied. Microcosm experiments using sediments from arsenic contaminated shallow alluvial aquifers in the blackfoot disease endemic area showed simultaneous microbial reduction of Fe(III) and As(V). Significant soluble Fe(II) (0.23±0.03mM) in pore waters and mobilization of As(III) (206.7±21.2nM) occurred during the first week. Aqueous Fe(II) and As(III) respectively reached concentrations of 0.27±0.01mM and 571.4±63.3nM after 8 weeks. We also showed that the addition of acetate caused a further increase in aqueous Fe(II) but the dissolved arsenic did not increase. We further isolated an As(V)-reducing bacterium native to aquifer sediments which showed that the direct enzymatic reduction of As(V) to the potentially more-soluble As(III) in pore water is possible in this aquifer. Our results provide evidence that microorganisms can mediate the release of sedimentary arsenic to groundwater in this region and the capacity for arsenic release was not limited by the availability of electron donors in the sediments.
