Fluid-flow within tectonic belts and fault-fluid interaction are fundamental and controlling factors of large thrust belt evolutions, formation of mineral deposits in the upper crust and the ductile shear deformation in the middle crust. In central South China, a large Chuandong-Xiangexi fold-and-thrust belt and long-distance displaced thrust faulting are developed, but their formation mechanism is still in debate. In particular, the evolution of fluid-flow and fault-fluid interactions in this fold-and-thrust belt is still not well constrained. In this study, through detailed structural and microstructural observations together with stable isotope and fluid inclusion analyses, we aim to unravel the deformation characteristics and syn-tectonic fluid-flow along the detachment in the frontal zone of this fold-and-thrust belt. One approximately 30–60 m thick, NW-verging detachment thrust with brittle-ductile deformation and alternating the strongly and weakly deformed zones is identified in the Lower Permian bioclastic and argillaceous limestones. The strongly deformed zone is characterized by tectonic lenses, fault gouge, calcite veins, S–C fabric, and foliated fault rocks/gouge, while the weak deformation zone is marked by scattered fractures without calcite veins and weakly deformed tectonic lenses. The fluids within the thrust faults originate from formation waters (saline fluids or brines) in a closed system, evidenced by the δ18O values, the 87Sr/86Sr ratios, the hydrocarbon-bearing fluid inclusions characterized by moderate salinity (10–14 wt% NaClequiv.) and moderate-high homogenization temperatures (∼160–350 °C). In the hanging wall of the Huangyingshan fault, the fluids involved in the thrusting processes are most likely brines derived from Middle and Lower Triassic carbonate aquifers. While in the footwall, the fluids are likely brines derived from Cambrian evaporitic aquifers. These findings on the detachment thrust fault zone could provide valuable constraints on the conduit and distance of hydrocarbon secondary migration for exploring the potential of large-scale hydrocarbon accumulation in the Chuandong-Xiangexi fold-and-thrust belt. Two new Schiff‐base bismuth (III) complexes were prepared by an equivalent reaction between Schiff‐base ligand and Bi (NO3)3•5H2O with the assistance of Mannitol. The chemical structures of the two complexes were characterized by spectroscopic studies (FT‐IR, NMR, and MS), elemental analysis, and single‐crystal X‐ray diffraction. The ligand‐to‐metal ion ratio was found to be 1:1 in the complexes. During the formation of the complexes, Schiff bases changed from the amidic forms to the iminol forms, and the resulting tautomers could coordinate with bismuth (III) ions to produce dinuclear BiIII complexes(1a and 2a). Structural analyses showed that each Bi (III) ion held a distorted capped octahedron geometry with a seven‐coordinate mode in two complexes. Screening in vitro biological activities revealed that two bismuth (III) complexes exhibited much higher antimicrobial and cytotoxic activity than their parent ligands. The cytotoxic activity of the complex(1a) was close to that of the known anticancer drug (Doxorubicin) by evaluating against SGC7901 cells, with the IC50 value 0.59 μM. The complex(1a) could effectively induce SGC7901 cell apoptosis and its oral acute toxicity for LD50 value was found to be 576 mg kg−1. Background. The spondin-2 (SPON2) gene is overexpressed in multiple malignant tumors and may promote tumor aggressiveness. However, its expression profile and functional roles in clear cell renal cell carcinoma (ccRCC) are still unclear. Methods. SPON2 expression in ccRCC was evaluated using expression data from TCGA and GEO databases, then confirmed by local patient population (94 patients). The clinical significance of SPON2 expression was evaluated. Downregulation of SPON2 was performed using small-interfering RNA (siRNA). The effects of SPON2 silencing on cell proliferation, apoptosis, invasion, and migration in vitro were investigated. Results. SPON2 was overexpressed in the majority of the ccRCC at both mRNA and protein levels. SPON2 expression was significantly correlated with stage, grade, and recurrence (all P < 0.05) in patients with localized ccRCC. The receiver operating characteristic (ROC) curve showed that SPON2 expression could serve as a predictor of recurrence. SPON2 expression was significantly associated with recurrence-free survival (RFS) in patients with localized ccRCC. Knocking down SPON2 resulted in suppressed cell invasion and migration in vitro. Polycyclic aromatic compounds (PACs) are important toxic organic components in fine particulate matter (PM 2.5), whereas the links between PM 2.5 toxicity and associated PACs in ambient air are poorly understood. This study investigated the spatial–temporal variations of PACs in PM 2.5 collected from 11 sampling sites across a Chinese megacity and characterized the reactive oxygen species (ROS) generation and cytotoxicity induced by organic extracts of PM 2.5 based on cellular assays. The extra trees regression model based on machine learning and ridge regression were used to identify the key toxicants among complex PAC mixtures. The total concentrations of these PACs varied from 2.12 to 71.7 ng/m3 across the study city, and polycyclic aromatic hydrocarbons (PAHs) are the main PACs. The spatial variations of the toxicological indicators generally resembled those of the PAC concentrations, and the PM 2.5 related to waste treatment facilities exhibited the strongest toxic potencies. The ROS generation was highly correlated with high molecular weight PAHs (MW302 PAHs), followed by PAHs with MW<302 amu and oxygenated PAHs, but not with nitrated PAHs and the plastics additives. The cell mortality showed weak correlations with these organic constituents. The associations between the biological endpoints and these PM 2.5 -bound contaminants were further confirmed by exposure to authentic chemicals. Four primary sources of PACs were identified, among which coal and biomass combustion sources (30.2% of the total PACs) and industrial sources (31.0%) were predominant. PACs emitted from industrial sources were highly associated with ROS generation in this city. Our findings highlight the potent ROS-generating potential of MW302 PAHs and the importance of industrial sources contributing to PM 2.5 toxicity in this megacity, raising public concerns and further administration. The present study was conducted to characterize the C6orf120 gene, by using C6orf120 gene-deleted rats ( C6orf120 − / − ), to determine its role in the development and severity of autoimmune hepatitis induced by concanavalin A (Con A), as well as the underlying mechanisms. We found that following Con A injection, C6orf120 − / − rats were less susceptible to developing autoimmune hepatitis with low levels of alanine aminotransferase (ALT) and aspartate aminotransferase (AST) post challenge. Additionally, C6orf120 deficiency increased the frequency of cluster of differentiation (CD)4 + CD25 + Forkhead box P3 + regulatory T cells (Tregs) among intrahepatic lymphocytes, splenocytes, peripheral blood mononuclear cells, and CD4 + T in vitro . Moreover, C6orf120 deficiency downregulated interleukin (IL)-1β, IL-6, tumor necrosis factor alpha-α, interferon-γ and IL-17a secretion in the plasma and liver tissues. Our results indicated that the C6orf120 gene-deleted rats were less susceptible to Con A-induced autoimmune hepatitis, which may be partly related to the increased frequency of Tregs and inhibited secretion of inflammatory cytokines. Background: Schistosomiasis is a relatively neglected parasitic disease that afflicts more than 250 million people worldwide, for which the control strategy relies mainly on mass treatment with the only available drug, praziquantel (PZQ). This approach is not sustainable and is a priority for developing novel drug candidates for the treatment and control of schistosomiasis. Methodologys/Principal findings: In our previous study, we found that DW-3-15, a kind of PZQ derivative, could significantly downregulate the expression of the histone acetyltransferase of Schistosoma japonicum (SjHAT). In this study, several commercially available HAT inhibitors, A485, C646 and curcumin were screened in vitro to verify their antischistosomal activities against S. japonicum juveniles and adults. Parasitological studies and scanning electron microscopy were used to study the primary action characteristics of HAT inhibitors in vitro. Quantitative real-time PCR was employed to detect the mRNA level of SjHAT after treatment with different HAT inhibitors. Our results demonstrated that curcumin was the most effective inhibitor against both juveniles and adults of S. japonicum, and its schistosomicidal effects were time- and dose dependent. However, A485 and C646 had limited antischistosomal activity. Scanning electron microscopy demonstrated that in comparison with DW-3-15, curcumin caused similar tegumental changes in male adult worms. Furthermore, both curcumin and DW-3-15 significantly decreased the SjHAT mRNA level, and curcumin dose-dependently reduced the SjHAT expression level in female, male and juvenile worms. Conclusions: Among the three commercially available HATs, curcumin was the most potent against schistosomes. Both curcumin and our patent compound DW-3-15 markedly downregulated the expression of SjHAT, indicating that SjHAT may be a potential therapeutic target for developing novel antischistosomal drug candidates. Author summary: Schistosomiasis is a zoonotic parasitic disease caused by Schistosoma spp. infection. Due to the lack of effective vaccines for schistosomiasis, PZQ is the only available drug chosen for the treatment and control of schistosomiasis in most developing countries. Drug resistance due to long-term, large-scale and single-agent use has been a great concern. Therefore, there is an urgent need for new therapies. In our previous study, we developed the patented compound DW-3-15 (patent no. ZL201110142538.2), which has significant and stable antiparasitic activity, and can significantly decrease the expression of S. japonicum histone acetyltransferase (SjHAT). As we known, HATs are a class of epigenetic enzymes that are crucial for chromatin restructuring and transcriptional regulation in mammals and in parasitic development, and are thus promising targets for antischistosomal therapeutic development. In this study, we assessed three commercial HATs, including two selective p300/CBP inhibitors A485 and C646, and one natural compound, curcumin. Our results demonstrated that curcumin had the most potent worm-killing efficacy against S. japonicum juveniles and adults. In contrast, A485 and C646 displayed no effects on adult worms or mild effects on juveniles. Moreover, curcumin caused severe damage to the tegument of male worms, and the damage was similar to that of DW-3-15. Coincidentally, both curcumin and DW-3-15 significantly downregulated the expression of SjHAT, indicating that SjHAT might be an effective target for the development of novel therapeutic drugs. The inhibiting effect of Norcantharidin (NCTD) on IL-6 (interleukin-6) and STAT3 and the involvement of the IL-6/STAT3 pathway in hepcidin expression prompted us to speculate that NCTD could affect iron metabolism. We examined the effects of NCTD on serum iron (SI) and transferrin (Tf) saturation, iron and ferritin light chain (FTL), transferrin receptor 1 (TfR1), divalent metal transporter 1 (DMT1), ferroportin 1 (Fpn1), iron regulatory protein 1 (IRP1) and hepcidin, as well as IL-6 and STAT3 in the liver, spleen and duodenum of mice treated with lipopolysaccharide (LPS) in vivo, using RT-PCR, Western blotting and immunofluorescence analysis. NCTD could increase SI and Tf saturation and reduce tissue iron and FTL content by affecting expression of cell-iron transport proteins TfR1, DMT1 and Fpn1. The impact of NCTD on TfR1, DMT1 and Fpn1 expression is mediated by up-regulating IRP1 and down-regulating hepcidin expression, while NCTD-induced down-regulation of hepcidin is mediated by the IL-6/STAT3 signalling pathway in LPS-treated mice. NCTD affects iron metabolism by modifying the expression of IL-6/JAK2/STAT3/hepcidin and IRP1 and suggest that the ability of NCTD to reduce tissue iron contents may be a novel mechanism associated with the anti-cancer effects of NCTD. A new chelating resin (CPS-DMA-SABA) was synthesized with the chloromethylated cross-linked polystyrene porous beads (CPS) as raw materials, which were bonded with salicylidene- o-aminobenzoic acid (SABA) on the surface via a quaternary ammonium unit as a linker. The results showed that CPS-DMA-SABA would easily change into a zwitterionic chelating resin (CPS-DMA-SABZ) when washed with distilled water. CPS-DMA-SABA was characterized by elemental analysis, infrared spectra and thermo-gravimetric analysis. The ion sorption capacities of CPS-DMA-SABA were found to be 1916 μg/g for Zn with pH at 4.5, 1620 μg/g for Cu with pH at 6.2, 1291 μg/g for Ni with pH at 6.6 and 780 μg/g for Cr with pH at 5.5, respectively. The experiments showed CPS-DMA-SABA also changed its color when meeting with the metal ion in the aqueous solution. As a consequence, CPS-DMA-SABA can not only be used as a solid phase extractant but also an indicator for confirming the heavy metal ion in solutions. [ABSTRACT FROM AUTHOR]

Abstract: Aloperine has been shown to inhibit 2,4-dinitrofluorobenzene (DNFB) induced allergic contact dermatitis in BALB/c mice. In the present study, we further investigated the effect of aloperine on DNFB-induced atopic dermatitis-like skin lesions in NC/Nga mice. NC/Nga mice elicited atopic dermatitis-like skin lesions after the topical application of DNFB. Aloperine treatment significantly inhibited dermatitis index and ear thickness in DNFB-treated NC/Nga mice in a dose-dependent manner. Eosinophils, mast cells infiltration into the ears and plasma level of immunoglobulin (Ig) E were also suppressed by aloperine treatment. Finally, cytokine (interleukin (IL)-1β, IL-4, IL-6, IL-10, IL-13, tumor necrosis factor (TNF)-α and interferon (IFN)-γ) productions in ear biopsies homogenates were significantly elevated after DNFB challenge. Topical application of aloperine increased the immunosuppressive cytokine IL-10 level, while it reduced other cytokines production in a dose-dependent manner. Taken together, these data suggest that aloperine may be one of the effective therapeutic agents for the treatment of atopic dermatitis. [Copyright &y& Elsevier]

An approach to the chiral separation of racemic mixtures of amino acids by means of micellar electrokinetic chromatography after derivatization with a new triazine spectroscopic reagent, 3-(4,6-dichloro-1,3,5-triazinylamino)-7-dimethylamino-2-methylphenazine (DTDP), has been evaluated. It was found that the derivatives of the aliphatic amino acids such as serine, valine and arginine, could produce a strong UV absorption at 282 nm, whose apparent molar absorptivities are of 104 M−1 cm−1, and thus the concentration of the amino acids down to 3×10−7 M can still give a detectable signal (S/N=3). β-Cyclodextrin (β-CD) added to the buffer system was used as a chiral selector, and separation conditions were optimized. The presence of an organic modifier (2-propanol) was also a prerequisite for the chiral separation. The best results for the chiral separation of DTDP-amino acids were achieved in a mixed sodium dodecylsulfate–β-CD–borate–2-propanol medium at pH 9.0. Compared to some of the commonly used derivatization methods, the present one offers a relatively stable derivative and strong UV absorption for the spectroscopically inert amino acids, thus enabling amino acids to be separated and detected by CE even with a simpler UV detector. Schiff base {(E)‐4‐((2‐hydroxy‐3‐methoxybenzylidene)amino)‐1,5‐dimethyl‐2‐phenyl‐1,2‐dihydro‐3H‐pyrazol‐3‐one} was prepared by the condensation reaction between 2‐hydroxy‐3‐methoxybenzaldehyde and 4‐aminoantipyrine. In the process of forming the title complex, Schiff base was first coordinated with CuII ions to form the mononuclear CuII complex (abbreviated as the complex (I)). Next, Schiff base in the complex (I) removed a phenyl group and then changed from the amide to the iminol. The tautomer further formed the complex through the iminol oxygen, and the phenolic oxygen coordinated with metal ions (CuII and BiIII). The structure of the polynuclear CuIIBiIII complex (abbreviated as the complex (II)) was identified as [Bi2Cu6 L6O3] (L = C13H13N3O3) by elemental analysis, FT‐IR spectra, and single‐crystal X‐ray diffraction analysis. The antibacterial effect of Schiff base and its complex (II) were assessed on four bacterial strains involving gram‐positive strains (S. aureus and B. subtilis) and gram‐negative strains (E. coli and P. aeruginosa). The complex (II) exhibited stronger antibacterial activity than Schiff base. Assessment of cytotoxic activity against SNU‐16 cells (Human gastric cancer) has demonstrated that the complex (II) was more active than Schiff base, and their IC50 values were 0.29 and 1.83 μM, respectively. Evidence regarding the link between long-term ambient ozone (O 3) exposure and childhood sleep disorders is little. This study aims to examine the associations between long-term exposure to O 3 and sleep disorders in children. We conducted a population-based cross-sectional survey, including 185,428 children aged 6–18 years in 173 schools across 14 Chinese cities during 2012 and 2018. Parents or guardians completed a checklist using Sleep Disturbance Scale for Children, and O 3 exposure at residential and school addresses was estimated using a satellite-based spatiotemporal model. We used generalized linear mixed models to test the associations with adjustment for factors including socio-demographic variables, lifestyle, meteorology and multiple pollutants. Mean concentrations of O 3 , particulate matter with diameters ≤2.5 mm (PM 2.5) and nitrogen dioxide (NO 2) were 89.0 μg/m3, 42.5 μg/m3 and 34.4 μg/m3, respectively. O 3 and NO 2 concentrations were similar among provinces, while PM 2.5 concentration varied significantly among provinces. Overall, 19.4% of children had at least one sleep disorder. Long-term exposure to O 3 was positively associated with odds of sleep disorders for all subtypes. For example, each interquartile increment in home-school O 3 concentrations was associated with a higher odds ratio for global sleep disorder, at 1.22 (95% confidence interval: 1.18, 1.26). Similar associations were observed for sleep disorder subtypes. The associations remained similar after adjustment for PM 2.5 and NO 2. Moreover, these associations were heterogeneous regionally, with more prominent associations among children residing in southeast region than in northeast and northwest regions in China. We concluded that long-term exposure to O 3 is positively associated with risks of childhood sleep disorders. These associations varied by geographical region of China. Background: Previous studies have revealed that current secondhand smoke exposure showed highly suggestive evidence for increased risk of simultaneous sleep problems in children. Data on the associations between early-life exposure to SHS with subsequent sleep problems in children were scarce. We aimed to evaluate the associations of early-life SHS exposure with sleep problems in children. Methods: In this cross-sectional study, children were recruited from elementary and middle schools in Liaoning Province, China between April 2012 and January 2013. We assessed early-life SHS exposure (pregnancy and the first 2 years of life) via questionnaires. Sleep problems and different types of sleep-related symptoms were measured based on the validated tool of the Sleep Disturbance Scale for Children (SDSC). Generalized linear mixed models were applied to estimate the associations of early-life SHS exposure with sleep problems. Results: We included a total of 45,562 children (22,657 [49.7%] males; mean [SD] age, 11.0 [2.6] years) and 6167 of them (13.5%) were exposed to early-life SHS during both pregnancy and the first 2 years of life. Compared with unexposed counterparts, children exposed to early-life SHS had higher total T-scores of SDSC (β = 4.32; 95%CI: 4.06, 4.58) and higher odds of increased sleep problems (OR = 2.14; 95%CI: 1.89, 2.42). When considering different sleep-related symptoms, the associations between early-life SHS exposure and symptom of sleep-wake transition disorders (i.e., bruxism) were the strongest in all analyses. Conclusions: Early-life SHS exposure was associated with higher odds of global sleep problems and different sleep-related symptoms in children aged 6–18 years. Anthropogenic heat has been reported to have significant health impacts, but research on its association with childhood adiposity is still lacking. In this study, we matched the 2008–2012 average anthropogenic heat flux, as simulated by a grid estimation model using inventory methods, with questionnaire and measurement data of 49,938 children randomly recruited from seven cities in Northeast China in 2012. After adjusting for social demographic and behavioral factors, we used generalized linear mixed-effect models to assess the association between anthropogenic heat flux and adiposity among children. We also examined the effect modification of various social demographic and behavioral confounders. We found that each 10 W/m2 increase in total anthropogenic heat flux and that from the industry source was associated with an increase of 5.82% (95% CI = 0.84%–11.05%) and 6.62% (95% CI = 0.87%–12.70%) in the odds of childhood adiposity. Similarly, the excess rate of adiposity among children were 5.26% (95% CI = −1.33%-12.29%) and 8.51% (95% CI = 2.24%–15.17%) per 1 W/m2 increase in the anthropogenic heat flux from transportation and buildings, and was 7.94% (95% CI = 2.28%–13.91%) per 0.001 W/m2 increase in the anthropogenic heat flux from human metabolism. We also found generally greater effect estimates among female children and children who were exposed to passive smoking during pregnancy, born by caesarean section, non-breastfed/mixed-fed, or lived within 20 m adjacent to the main road. A convenient, fluorescent dual-channel chemosensor on the basis of bis(3-pyridylmethyl)amine-functionalized carbon quantum dots (BPMA-CQDs) nanoprobe was constructed, and it can discriminatively detect glutathione from its analogues cysteine and homocysteine based on two distinctive strategies. Two distinct fluorescence responses of BPMA-CQDs probe to Cu(II) and Ag(I) were identified and further employed to achieve selective detection of Cu(II) and Ag(I) respectively. Based on the BPMA-CQDs/Cu(II) conjugate, discriminative detection of GSH was achieved in terms of correlation between the amounts of GSH and fluorescence recovery. The addition of GSH into BPMA-CQDs/Cu(II) system induces the reduction of Cu(II) to Cu(I), which could efficiently block PET process resulting in the following fluorescence recovery. Based on the BPMA-CQDs/Ag(I) conjugate, GSH assay could also be established on the basis of fluorescence response to GSH. The introduction of GSH into the preceding system triggers the competitive coordination to Ag(I) between BPMA and GSH, and silver ions are finally taken away by GSH from the probe, where the fluorescence is restored to its original weak state. Both of the detection strategies can achieve discriminative detection of GSH from Cys and Hcy. A lysosome-targeting fluorescent off-on probe has been developed by one-step synthesis for detecting lysosomal nitroreductase and hypoxia. The probe is constructed by incorporating morpholine (a lysosome-targeting unit) into 4-nitro-1,8-naphthalimide (as a fluorochrome and specific substrate for nitroreductase), and the detection mechanism is based on the nitroreductase-catalyzed reduction of the probe to 4-amino-1,8-naphthalimide, accompanied by a large fluorescence enhancement at a wavelength of 543 nm. The probe shows an accurate lysosome-targeting ability with high selectivity and sensitivity to nitroreductase (detection limit: 2.2 ng mL−1). Notably, the probe has been used to image the change of lysosomal nitroreductase in live cells during hypoxia, revealing that the increase of nitroreductase in lysosomes may be smaller than that in the cytoplasm. A new cresyl violet-based fluorescent off-on probe has been developed through a one-step synthesis for the detection of nitroreductase (NTR) and hypoxia. The detection mechanism is based on the NTR-catalyzed reduction of the probe to cresyl violet, accompanied with a large fluorescence enhancement at a long wavelength of 625 nm. The probe can detect NTR in aqueous solution with high selectivity and sensitivity, and the detection limit is 1 ng mL−1 NTR. The objective of this study is to observe effects of acupuncture and electro-acupuncture (EA) on blood perfusion in the stomach, and probe into the application of laser Doppler blood perfusion imaging technique in the study of the effect of acupuncture and moxibustion on the entrails. In the acupuncture group of 20 rats, acupuncture was given at "Zusanli" (ST 36) and in EA group of 18 rats, EA was applied at "Zusanli" (ST 36), with 18 rats without acupuncture used as control group. Changes of blood perfusion and microcirculation distribution in the stomach were investigated with laser Doppler blood perfusion imager (LDPI). The laser Doppler blood perfusion image could clearly display changes of blood flow distribution in the stomach before and after acupuncture. After acupuncture or EA was given at "Zusanli" (ST 36), the blood perfusion in the stomach increased significantly, the blood perfusion in the blood vessels andmicrocirculation of other parts significantly increased, and themaximum increase of the blood perfusion was found at 10 min after acupuncture or EA, with increases of 0.50 0.11 (PU) and 0.66 0.16 (PU), respectively, and the blood perfusion still kept at a higher degree within 10 min after ceasing of the acupuncture or EA. While the blood perfusion in the stomach in the rat of the control group tended to gradual decrease. It has been concluded that both acupuncture and EA can increase blood perfusion in the stomach, the EA having stronger action, and LDPI can display the regulative action of acupuncture on the blood vessel of the stomach by using an image.
