In this work, we present data from the Milky Way Imaging Scroll Painting project for the Maddalena giant molecular cloud (GMC). We decompose the 13CO emission datacube of the observed region into hierarchical substructures using a modified dendrogram algorithm. We investigate the statistical properties of these substructures and examine the role that self-gravity plays on various spatial scales. The statistics of the mass (M), radius (R), velocity dispersion (σ v ), virial parameter (α vir), and sonic Mach number of the substructures are presented. The radius and mass distributions and the σ v –R scaling relationship of the substructures resemble those reported in previous studies that use nonhierarchical algorithms to identify the entities. We find that for the hierarchical substructures α vir decreases as the radius or mass of the substructures increases. The majority of the substructures in the quiescent region of the Maddalena GMC are not gravitationally bound (α vir > 2), while most of the substructures in the star-forming regions are gravitationally bound (α vir < 2). Furthermore, we find that self-gravity plays an important role on scales of 0.8–4 pc in the IRAS 06453 star-forming region, while it is not an important factor on scales below 5 pc in the non-star-forming region. • An ultra-broadband multi-channel OAM mode converter using a pair of helical long period gratings (HLPGs) is proposed. • The maximum channel number of OAM mode generator reaches 46, which is, to the best of our knowledge, the highest number of channels among OAM mode converters. • The maximum wavelength ranges of the channels with coupling efficiency greater than 90 % are measured to be 379 nm experimentally. We demonstrate an ultra-broadband multi-channel orbital angular momentum (OAM) mode generator using a helical long-period grating (HLPG) pair in a few-mode fiber. The numbers of the channel of the OAM mode generator can be flexibly adjusted by designing the cascaded length of the HLPG pair. When the cascaded length is 50 cm, the maximum channel number of OAM mode generator reaches 46, and the mode convert efficiencies of the channels are more than 90 %, which is, to the best of our knowledge, the highest number of channels among OAM mode converters. By reducing grating length and enhancing the refractive index modulation depth, the 3 dB bandwidth of the HLPG is significantly increased, and thus the wavelength range of the HLPG pair can be expanded. The maximum wavelength ranges of the channels with coupling efficiency greater than 90 % are measured to be 379 nm experimentally. By adjusting the spiral direction of the helical refractive index modulation, we can achieve multi-channel ± 1 order OAM mode conversion for each channel. We demonstrated the fabrication of long-period fiber gratings (LPFGs) in the tapered single-mode fiber by CO 2 laser. The phase-matching curves of the LPFGs inscribed in the tapered fiber with different waist diameters have been studied theoretically and experimentally. When the waist diameter of the tapered fiber decreased from 90 μm to 22 μm, the slope of the phase-matching curve changed dramatically. The refractive index sensitivity can be increased by inscribing the LPFGs in the tapered fiber around the dispersion turning point. A maximum refractive index sensitivity of 21702.78 nm/RIU has been achieved in the range of 1.445–1.457 for the LPFG inscribed in the tapered fiber with a waist diameter of 50 μm. This kind of LPFGs inscribing in the tapered fiber may be a good candidate for the development of refractive index based chemical and biological sensors. • The phase-matching curves of the LPFGs inscribed in the tapered fiber with different waist diameters from 90 μm to 22 μm have been studied. • The maximum SRI sensitivity of 21702.78 nm/RIU in the range of 1.445–1.457 was achieved by the LPFG with a DTP. • The waist length of the tapered fiber can reach 60 mm, enabling the fabrication of tapered fiber LPFGs with high repeatability. We demonstrate the inscription of long-period gratings (LPGs) in a fluorine-doped double-cladding fiber (DCF) with a low refractive index trench. The grating has a single resonance dip in the wide wavelength range from 1200 nm to 2400 nm. The transmission spectra and cladding mode evolution of the DCF-LPG during the cladding etching process are investigated theoretically and experimentally. The mode order of the cladding modes decreases with one order of every 20 μ m diameter etching, while the resonance wavelength of the DCF-LPG barely shifts when the fiber diameter decreases gradually. Only LP 11 mode can be excited in the etched DCF-LPG when the fiber diameter is less than 40 μ m. The characteristics of the DCF-LPGs are investigated experimentally. The torsion sensitivity was measured to be −0.154 nm/(rad/m) and −0.271 nm/(rad/m) at 1. 55 μ m and 2. 0 μ m wavebands, respectively, which are one order of magnitude higher than that of the conventional LPG. The proposed DCF-LPG has potential application as a tunable band-rejection filter in broad wavebands. • Only one resonance dip of the DCF-LPG can be observed in the wavelength range from 1200 nm to 2400 nm. • The mode order of the cladding modes decrease with diameter etching, while the resonance wavelength barely shifts. • The twist sensitivity of the DCF-LPGs at 1. 55 μ m and 2 μ m wavebands was one order of magnitude higher than that of the conventional LPG. In view of the status of the present railway wagon number mainly adopts the RFID technology, the paper presents a railway wagon number positioning method based on template matching and morphological operations. This method should first pretreatment and morphological filtering for the carriage image, then using the projection method to locate the license plate number region contains railway logo, finally, through the method of establishing the railway logo templates and removing the minimum connected domain area, to position precisely of the license plate number area. The experiment results show that, the method is not affected by the train modles, train colours, the railway logo spraying position and the license plate number spraying interval, make it the high locating accuracy and good adaptability. The thermal stability of uniform fiber Bragg gratings (FBGs) inscribed in single mode double cladding fiber (DCF), standard single mode fiber (SMF-28) and dispersion shifted fiber (DSF) by 193 nm excimer laser was experimentally investigated. When the temperature rises from 30 °C to 800 °C, the grating contrast of FBGs with the saturated refractive index modulation drops slowly, and the corresponding 3 dB bandwidth gradually decrease After annealing at 400 °C for 2000 h, it is found that the resonance wavelength shift of FBGs mainly occur during the first 200 h. It is also observed that the FBG inscribed in DCF requires a longer time to stabilize wavelength than the FBG written in SMF-28, about 1500 h, which suggests that long-term pre-annealing is required to ensure accurate wavelength measurement. Abstract: A modified electroless silver-plating process has been devised for the preparation of monodispersed, polystyrene/silver (PS/Ag) composite microspheres with tunable shell thickness. Tailoring was achieved by altering the concentration of the silver precursor in the plating bath. PS/Ag composite microspheres were characterized by field-emission scanning electron microscopy, ultraviolet–visible absorption, X-ray diffraction and thermogravimetric analysis. The results showed that a dense, stable and uniform silver nanoshell was formed on the surface of PS microspheres in the presence of poly(vinylpyrrolidone) and glucose. The bulk conductivity of the PS/Ag composites increased from 1.16S/m to 3.57×104 S/m, corresponding to a shell thickness of 35–198nm. The PS/Ag composite microspheres with diameters of ca. 3μm might have great potential to be used as fillers in anisotropic conductive films because of the uniform diameter, low density and good conductivity of the microspheres. Cashmere goats possess two types of hair follicles, with the secondary hair follicles producing valuable cashmere fiber used for textiles. The growth of cashmere exhibits a seasonal pattern arising from photoperiod change. Transcription factors play crucial roles during this process. The transcription factor, cold-shock domain, containing C2 (Csdc2) plays a crucial role in modulating cell proliferation and differentiation. Our preceding research indicated that the expression of Csdc2 changes periodically during anagen to telogen. However, the mechanisms of Csdc2 in regulating SHF growth remain unclear. Here, we found that the knockdown of Csdc2 inhibits the proliferation of dermal papilla cells. ChIP-Seq analysis showed that Csdc2 had a unique DNA binding motif in SHFs. Through conjoint analysis of ChIP-Seq and RNA-Seq, we revealed a total of 25 candidate target genes of Csdc2. Notably, we discovered a putative Csdc2 binding site within roundabout guidance receptor 2 (Robo2) on chromosome 1 of the goat genome. Furthermore, qRT-PCR and dual-luciferase reporter assay confirmed Csdc2's positive regulatory influence on Robo2. These findings expand the research field of hair follicle transcriptional regulatory networks, offering insights into molecular breeding strategies to enhance cashmere production in goats. Simple Summary: Cashmere goats are valuable genetic resources which are famous for their high-quality fiber. Runs of homozygosity (ROHs) refer to uninterrupted sequences of homozygous genotypes present within the DNA sequence of an individual. To enhance the preservation, improvement and enduring utilization of this valuable genetic resource, we performed a comprehensive study on the genetic variance, degree of inbreeding, patterns of ROHs and selected genes situated in ROH islands which are associated with economic attributes in goats via whole-genome sequencing analysis. Our research showed that the Inner Mongolia cashmere goat possesses a notably high level of genetic diversity and presented the lowest inbreeding coefficient. On the contrary, the highest F r o h and lowest levels of diversity were observed in wild goats compared to domesticated goats. The analysis of selection signal through ROHs identified some genes related to meat, fiber and milk production; fertility, disease and cold resistance and adaptability; and body size and growth. Cashmere goats are valuable genetic resources which are famous worldwide for their high-quality fiber. Runs of homozygosity (ROHs) have been identified as an efficient tool to assess inbreeding level and identify related genes under selection. However, there is limited research on ROHs in cashmere goats. Therefore, we investigated the ROH pattern, assessed genomic inbreeding levels and examined the candidate genes associated with the cashmere trait using whole-genome resequencing data from 123 goats. Herein, the Inner Mongolia cashmere goat presented the lowest inbreeding coefficient of 0.0263. In total, we identified 57,224 ROHs. Seventy-four ROH islands containing 50 genes were detected. Certain identified genes were related to meat, fiber and milk production (FGF1, PTPRM, RERE, GRID2, RARA); fertility (BIRC6, ECE2, CDH23, PAK1); disease or cold resistance and adaptability (PDCD1LG2, SVIL, PRDM16, RFX4, SH3BP2); and body size and growth (TMEM63C, SYN3, SDC1, STRBP, SMG6). 135 consensus ROHs were identified, and we found candidate genes (FGF5, DVL3, NRAS, KIT) were associated with fiber length or color. These findings enhance our comprehension of inbreeding levels in cashmere goats and the genetic foundations of traits influenced by selective breeding. We demonstrate an optically tunable mode-locked fiber laser using long-period fiber grating (LPFG) coated with multi-walled carbon nanotubes (CNTs). The multi-walled CNTs can absorb light to convert it into thermal energy, and the resonance wavelength of the grating can be easily turned by varying the external modulated light power. This multi-walled CNT coated LPFG-based all-optical fast and efficient spectrum tunable filter enables continuous tuning of the central wavelength of the laser by manipulating the loss of the mode-locked laser, ensuring the stability of the mode-locking state. In the absence of modulated light on multi-walled CNTs, the soliton laser could generate 890 fs pulses at 1546.7 nm with a spectrum bandwidth of 3.26 nm and a signal-to-noise ratio of 73.1 dB. Through adjustment of the pump power of the modulation light on multi-walled CNTs, the mode-locked fiber laser can be continuously tuned from 1546.71 to 1563.15 nm. The response time of the optically tunable system was measured to be in the order of hundreds of milliseconds. Fine-wool sheep are the most common breed used by the wool industry worldwide. Fine-wool sheep have over a three-fold higher follicle density and a 50% smaller fiber diameter than coarse-wool sheep. This study aims to clarify the underlying genetic basis for the denser and finer wool phenotype in fine-wool breeds. Whole-genome sequences of 140 samples, Ovine HD630K SNP array data of 385 samples, including fine, semi-fine, and coarse wool sheep, as well as skin transcriptomes of nine samples were integrated for genomic selection signature analysis. Two loci at keratin 74 (KRT74) and ectodysplasin receptor (EDAR) were revealed. Fine-scale analysis in 250 fine/semi-fine and 198 coarse wool sheep narrowed this association to one C/A missense variant of KRT74 (OAR3:133,486,008, P = 1.02E-67) and one T/C SNP in the regulatory region upstream of EDAR (OAR3:61,927,840, P = 2.50E-43). Cellular over-expression and ovine skin section staining assays confirmed that C-KRT74 activated the KRT74 protein and specifically enlarged cell size at the Huxley's layer of the inner root sheath (P < 0.01). This structure enhancement shapes the growing hair shaft into the finer wool than the wild type. Luciferase assays validated that the C-to-T mutation upregulated EDAR mRNA expression via a newly created SOX2 binding site and potentially led to the formation of more hair placodes. Two functional mutations driving finer and denser wool production were characterized and offered new targets for genetic breeding during wool sheep selection. Runs of Homozygosity (ROH) are continuous homozygous DNA segments in diploid genomes, which have been used to estimate the genetic diversity, inbreeding levels, and genes associated with specific traits in livestock. In this study, we analyzed the resequencing data from 10 local goat breeds in Yunnan province of China and five additional goat populations obtained from a public database. The ROH analysis revealed 21,029 ROH segments across the 15 populations, with an average length of 1.27 Mb, a pattern of ROH, and the assessment of the inbreeding coefficient indicating genetic diversity and varying levels of inbreeding. iHS (integrated haplotype score) was used to analyze high-frequency Single-Nucleotide Polymorphisms (SNPs) in ROH regions, specific genes related to economic traits such as coat color and weight variation. These candidate genes include OCA2 (OCA2 melanosomal transmembrane protein) and MLPH (melanophilin) associated with coat color, EPHA6 (EPH receptor A6) involved in litter size, CDKAL1 (CDK5 regulatory subunit associated protein 1 like 1) and POMC (proopiomelanocortin) linked to weight variation and some putative genes associated with high-altitude adaptability and immune. Simple Summary: We performed a genome-wide association study of 100 sheep genomes to investigate the hereditary basis of the four-horn phenotype (FHP) in sheep. The results revealed that a total of 3 InDels (e.g., CHR2: g.133,742,709delA and g.133,743,215insC) and 14 SNPs (e.g., CHR2: g.133,727,513C > T and CHR16: g.40,351,378G > A) in CHR2 and CHR16 were significantly associated with the four-horn trait. Furthermore, HOXD1 and ADAMTS12 annotated from both CHR sequences were the key candidate gene loci contributing to FHP in sheep. This study provided a novel understanding of the Mendelian genetic basis for four horns in sheep. Horns are an important breeding trait for sheep. However, no widely recognized viewpoint on the regulatory genes and mechanisms of horns is available, and the genetic basis of the four-horn phenotype (FHP) is unclear. This work conducted a genome-wide association study with 100 sheep genomes from multiple breeds to investigate the genetic basis of the FHP. The results revealed three significant associations (corrected as p < 1.64 × 10−8) of the InDels (CHR2: g.133,742,709delA, g.133,743,215insC, and g.133,743,940delT) for FHP in the intergenic sequence (IGS) between the MTX2 and the LOC105609047 of CHR2. Moreover, 14 significant associations (corrected as p < 1.42 × 10−9) of SNPs with the FHP phenotype were identified in CHR2 and CHR16, including five (e.g., CHR16: g.40,351,378G > A and g.40,352,577G > A) located in the intron of the ADAMTS12 gene, eight (e.g., CHR2: g.133,727,513C > T and g.133,732,145T > G) in the IGS between MTX2 and LOC105609047, and only one (CHR2: g.133,930,761A > G) in the IGS between HOXD1 and MTX2. Obvious divergence was also observed in genotype patterns between the FHP and others (two horns and hornless) in the HOXD1 and ADAMTS12 gene regions. An extremely significant linkage also occurred between Loci I and Loci II within 100 individuals (LD = −156.02186, p < 0.00001). In summary, our study indicated that the genomic sequences from CHR2 and CHR16 contributed to the FHP in sheep, specifically the key candidate genes HOXD1 and ADAMTS12. Simple Summary: In our research, candidate genes related to sheep-milk production were revealed by a genome-resequencing analysis and a genome-signal-selection analysis, and a RT-qPCR experiment was performed to prove the expression levels of these candidate genes, the results showed that the FCGR3A gene's expression level had a significant negative relationship with sheep-milk production. Natural selection and domestication have shaped modern sheep populations into a vast range of phenotypically diverse breeds. Among these breeds, dairy sheep have a smaller population than meat sheep and wool sheep, and less research is performed on them, but the lactation mechanism in dairy sheep is critically important for improving animal-production methods. In this study, whole-genome sequences were generated from 10 sheep breeds, including 57 high-milk-yield sheep and 44 low-milk-yield sheep, to investigate the genetic signatures of milk production in dairy sheep, and 59,864,820 valid SNPs (Single Nucleotide Polymorphisms) were kept after quality control to perform population-genetic-structure analyses, gene-detection analyses, and gene-function-validation analyses. For the population-genetic-structure analyses, we carried out PCA (Principal Component Analysis), as well as neighbor-joining tree and structure analyses to classify different sheep populations. The sheep used in our study were well distributed in ten groups, with the high-milk-yield-group populations close to each other and the low-milk-yield-group populations showing similar classifications. To perform an exact signal-selection analysis, we used three different methods to find SNPs to perform gene-annotation analyses within the 995 common regions derived from the fixation index (FST), nucleotide diversity (Ɵπ), and heterozygosity rate (ZHp) results. In total, we found 553 genes that were located in these regions. These genes mainly participate in the protein-binding pathway and the nucleoplasm-interaction pathway, as revealed by the GO- and KEGG-function-enrichment analyses. After the gene selection and function analyses, we found that FCGR3A, CTSK, CTSS, ARNT, GHR, SLC29A4, ROR1, and TNRC18 were potentially related to sheep-milk-production traits. We chose the strongly selected genes, FCGR3A, CTSK, CTSS, and ARNT during the signal-selection analysis to perform a RT-qPCR (Reale time Quantitative Polymerase Chain Reaction) experiment to validate their expression-level relationship with milk production, and the results showed that FCGR3A has a significant negative relationship with sheep-milk production, while other three genes did not show any positive or negative relations. In this study, it was discovered and proven that the candidate gene FCGR3A potentially contributes to the milk production of dairy sheep and a basis was laid for the further study of the genetic mechanism underlying the strong milk-production traits of sheep. As large-scale genomic studies have progressed, it has been revealed that a single reference genome pattern cannot represent genetic diversity at the species level. While domestic animals tend to have complex routes of origin and migration, suggesting a possible omission of some population-specific sequences in the current reference genome. Conversely, the pangenome is a collection of all DNA sequences of a species that contains sequences shared by all individuals (core genome) and is also able to display sequence information unique to each individual (variable genome). The progress of pangenome research in humans, plants and domestic animals has proved that the missing genetic components and the identification of large structural variants (SVs) can be explored through pangenomic studies. Many individual specific sequences have been shown to be related to biological adaptability, phenotype and important economic traits. The maturity of technologies and methods such as third-generation sequencing, Telomere-to-telomere genomes, graphic genomes, and reference-free assembly will further promote the development of pangenome. In the future, pangenome combined with long-read data and multi-omics will help to resolve large SVs and their relationship with the main economic traits of interest in domesticated animals, providing better insights into animal domestication, evolution and breeding. In this review, we mainly discuss how pangenome analysis reveals genetic variations in domestic animals (sheep, cattle, pigs, chickens) and their impacts on phenotypes and how this can contribute to the understanding of species diversity. Simple Summary: A unique structure of ruminants, the horn trait is not only closely related to natural and sexual selection but is also an important trait for polled sheep breeding. RXFP2 may be a crucial gene in regulating sheep horn. However, the underlying genetic mechanisms of sheep horn development remain largely unknown. In this study, we investigated the gene expression profile of the horn buds in sheep fetuses using RNA-seq technology. We identified 68 differentially expressed genes in the horn buds of 105-day-old Altay sheep fetuses, including RXFP2, FOXL2, and TNN. Further, we found that the Wnt signaling pathway may be responsible for horn development. Our study provides new possible marker genes for horn development, which may promote our understanding of the genetic mechanisms of sheep horn formation. Horns, also known as headgear, are a unique structure of ruminants. As ruminants are globally distributed, the study of horn formation is critical not only for increasing our understanding of natural and sexual selection but also for the breeding of polled sheep breeds to facilitate modern sheep farming. Despite this, a significant number of the underlying genetic pathways in sheep horn remain unclear. In this study, to clarify the gene expression profile of horn buds and investigate the key genes in horn bud formation, RNA-sequencing (RNA-seq) technology was utilized to investigate differential gene expression in the horn buds and adjacent forehead skin of Altay sheep fetuses. There were only 68 differentially expressed genes (DEGs) identified, consisting of 58 up-regulated genes and 10 down-regulated genes. RXFP2 was differentially up-regulated in the horn buds and had the highest significance (p-value = 7.42 × 10−14). In addition, 32 DEGs were horn-related genes identified in previous studies, such as RXFP2, FOXL2, SFRP4, SFRP2, KRT1, KRT10, WNT7B, and WNT3. Further, Gene Ontology (GO) analysis showed that the DEGs were mainly enriched with regard to growth, development, and cell differentiation. Pathway analysis revealed that the Wnt signaling pathway may be responsible for horn development. Further, through combining the protein–protein interaction networks of the DEGs, it was found that the top five hub genes, namely, ACAN, SFRP2, SFRP4, WNT3, and WNT7B, were also associated with horn development. Our results suggest that only a few key genes, including RXFP2, are involved in bud formation. This study not only validates the expression of candidate genes identified at the transcriptome level in previous studies but also provides new possible marker genes for horn development, which may promote our understanding of the genetic mechanisms of horn formation. Transposable elements (TEs) are diverse, abundant, and complicated in genomes. They not only can drive the genome evolution process but can also act as special resources for adaptation. However, little is known about the evolutionary processes that shaped horses. In this work, 126 horse assemblages involved in most horse breeds in China were used to investigate the patterns of TE variation for the first time. By using RepeatMasker and melt software, we found that the horse‐specific short interspersed repetitive elements family, equine repetitive elements (ERE1), exhibited polymorphisms in horse genomes. Phylogenetic analysis based on these ERE1 loci (minor allele frequency ≥0.05) revealed three major horse groups, namely, those in northern China, southern China, and Qinghai‐Tibetan, which mirrors the result determined by SNPs to some extent. The present ERE1 family emerged ~0.26 to 1.77 Mya ago, with an activity peak at ~0.49 Mya, which matches the early stage of the horse lineage and decreases after the divergence of Equus caballus and Equus ferus przewalskii. To detect the functional ERE1(s) associated with adaptation, locus‐specific branch length, genome‐wide association study, and absolute allele frequency difference analyses were conducted and resulted in two common protein‐coding genes annotated by candidate ERE1s. They were clustered into the vascular smooth muscle contraction (p = 0.01, EDNRA) and apelin signalling pathways (p = 0.02, NRF1). Simple Summary: Skeletal muscle satellite cells (SMSCs) serve as the source of myogenic cells and can afford to differentiate into myotubes as well as act as a model for exploring myogenesis in vitro. In this study, the transcriptional profile of ovine skeletal muscle satellite cells was constructed via the RNA-Seq method. A total of 1954 DEGs, 1479 AS, and 253 TFs were detected during the proliferation and differentiation of SMSCs. In total, 1479 alternative splice events (AS) were also identified during SMSC proliferation and differentiation. Among them, a unique AS event was the major per-mRNA splicing type, and SE was the predominant splicing pattern. Furthermore, transcription factors with AS were scanned during SMSC differentiation such as myocyte enhancer factor-2C (MEF2C) and the nuclear receptor subfamily 4 group A member 2 (NR4A2). Our results imply that MEF2C and NR4A2 can interact, and we speculate that NR4A2 and MEF2C might regulate the myogenesis of ovine SMSCs through interaction. How do dust grains in protoplanetary discs overcome rapid radial drift and grow from micron size particles to planets is not well understood. The key is to search for evidence of dust accumulation and growth as a function of radius in the disc. We investigate the radial profile of grain size in the DS Tau disc by fitting multiband ALMA observations with self-consistent radiative transfer models. The best-fit grain sizes range from centimeters in the inner disc down to |${\sim }\, 30\, \mu {\rm m}$| in the outer regions. Such an inside-out decreasing tendency is consistent with theories of dust evolution. Based on the best-fit model, we find that dust of |${\sim }\, 2$| Jupiter masses has been depleted within the gap. By taking the gas-to-dust mass ratio into account, the lost mass is enough to form the 3.5 Jupiter mass planet inferred by literature hydrodynamic simulations. Moreover, our modelling also indicates that at the interface region between the gap and the ring, the grain size profile shows a discontinuity, with its amplitude dependent on the dust model adopted in the radiative transfer analysis. As relatively new members of the non-coding RNA family, circRNAs play important roles in a variety of biological processes. However, the temporal expression pattern and the function of circRNAs during sheep skeletal muscle development remains unclear. This study aimed to identify circRNAs related to sheep skeletal muscle development and explore their roles in myoblast proliferation. The circRNA expression profiles of longissimus dorsi of sheep from F90, L30, and A3Y were obtained by the RNA-seq method. The function and mechanisms of the novel circCHRNG in muscle satellite cell proliferation were explored using CCK-8 assay, Western blot, qPCR, and dual-luciferase reporter assay. We identified 12,375 circRNAs, including 476, 133, and 233 DEcircRNAs found among three comparative groups. KEGG results showed that DEcircRNAs were enriched in muscle contraction, the regulation of cell proliferation, and the AMPK, insulin, and PI3K-Akt signaling pathways. Notably, a novel circRNA, termed circRNA CHRNG, acts as a miR-133 sponge to promote skeletal muscle satellite cell proliferation. Our study provides a systematic description of circRNAs of ovine skeletal muscle across fetal, lamb, and adult stages. Active star-forming regions are excellent laboratories for studying the origins and evolution of young stellar object (YSO) clustering. The W40–Serpens South region is such a region, and we compile a large near- and mid-infrared catalogue of point sources in it, based on deep near-infrared observations of Canada-France-Hawaii Telescope (CFHT) in combination with Two Micron All Sky Survey (2MASS), UKIRT Infrared Deep Sky Survey (UKIDSS), and Spitzer catalogues. From this catalogue, we identify 832 YSOs, and classify 15, 135, 647, and 35 of them to be deeply embedded sources, Class I YSOs, Class II YSOs, and transition disc sources, respectively. In general, these YSOs are well correlated with the filamentary structures of molecular clouds, especially the deeply embedded sources and the Class I YSOs. The W40 central region is dominated by Class II YSOs, but in the Serpens South region, half of the YSOs are Class I. We further generate a minimum spanning tree (MST) for all the YSOs. Around the W40 cluster, there are eight prominent MST branches that may trace the vestigial molecular gas filaments that once fed gas to the central natal gas clump. Of the eight, only two now include detectable filamentary gas in Herschel data and corresponding Class I YSOs, while the other six are populated exclusively with Class II YSOs. Four MST branches overlap with the Serpens South main filament, and where they intersect, molecular gas 'hubs' and more Class I YSOs are found. Simple Summary: This study investigated the family structure, parent–child pair and inbreeding coefficient of a Tan sheep breeding farm in Ningxia, China. The results can be used as the theoretical basis for selecting excellent breeding stock and making an apposite mating plan; furthermore, it can provide an important scientific basis for the conservation and reproduction of Tan sheep germplasm resources. Tan sheep is a special breed of locally protected sheep in China, one of the best quality meat sheep in the world. Due to the unclear pedigree of the rams on the Ningxia Tan sheep breeding farm, we investigated 74 rams in the field and explored a new method for family division. Genomic DNA was extracted from the blood of breeding rams. Using Plink software, GCTA tools and R language, we analyzed the genetic structure, kinship, and inbreeding coefficient of the breeding sheep, which revealed the genetic relationship between the individuals. The results showed that there was no obvious clustering phenomenon in the PCA, and the genetic background of the samples was similar. The G matrix and IBS distance matrix indicated that most individuals were far away from each other. Paternity testing identified 24 pairs of unknown parent–child pairs, and all the Tan sheep could be divided into 12 families, which provided a reference for sheep breeding. The average inbreeding coefficient based on the ROH of this population was 0.049, so there was a low degree of inbreeding and the rams in the field were able to maintain high genetic diversity. Overall, we explored a more accurate method through paternity and kinship analysis; it provides a scientific basis for pedigree construction, which has an important application value for Tan sheep breeding. The FGF5 gene has been associated with the regulation of fibre length in mammals, including cashmere goats. A deletion variant at ~14 kb downstream of the FGF5 gene showed significant divergence between cashmere and non‐cashmere goats in previous studies. In this study, we designed specific primers to genotype the deletion variant. The results of gel electrophoresis and Sanger sequencing revealed that a 507‐bp deletion mutation is located at 95 454 685–95 455 191 of chromosome 6 in goats. Genotyping data from a large panel of 288 goats showed that the deletion at the FGF5 gene locus appeared to be associated with cashmere length. The deletion variant was close to fixation (frequency 0.97) in cashmere goats. Furthermore, electrophoretic mobility shift assays for evaluating DNA–protein interaction and mRNA expression levels of FGF5 suggested that the deletion variant may serve as a cis‐acting element by specifically binding transcription factors to mediate quantitative changes in FGF5 mRNA expression. Our study illustrates how a structural mutation of the FGF5 gene has contributed to the cashmere growth phenotype in domestic goats. The ZBED6 gene is a transcription factor that regulates the expression of IGF2 and affects muscle growth and development. However, its effect on the growth and development of the heart is still unknown. Emerging evidence suggests that long noncoding RNAs (lncRNAs) can regulate genes at the epigenetic, transcriptional, and posttranscriptional levels and play an important role in the development of eukaryotes. To investigate the function of ZBED6 in the cardiac development of pigs, we constructed the expression profiles of mRNAs and lncRNAs in myocardial tissue obtained from Bama Xiang pigs in the ZBED6 knockout group (ZBED6-KO) and the wild-type group (ZBED6-WT). A total of 248 differentially expressed genes (DEGs) and 209 differentially expressed lncRNAs (DELs) were detected, and 105 potential cis target genes of DELs were identified. The functional annotation analysis based on the Gene Ontology (GO) and Kyoto Encyclopaedia of Genes and Genomes (KEGG) databases revealed two GO items related to muscle development by the cis target genes of DELs. Moreover, IGF2 was the direct target gene of ZBED6 by ChIP-PCR experiment. Summary: The multiple teats trait is common in many species of mammals and is considered related to lactation ability in swine. However, in Hu sheep, related gene research is still relatively limited. In this study, a genome‐wide association study was used to identify genetic markers and genes related to the number of teats in the Hu sheep population, a native Chinese sheep breed. A single marker method and several multi‐locus methods were utilized. A total of 61 SNPs were found to be related to the number of teats. Among these, 11 SNPs and one SNP were consistently detected by two and three multi‐locus models respectively. Four SNPs were concordantly identified between the single marker and multi‐locus methods. We also performed quantitative real‐time PCR testing of these identified candidate genes, identifying three genes with significantly different expression. Our study suggested that the LHFP, DPYSL2, and TDP‐43 genes may be related to the number of teats in sheep. The combination of single and multi‐locus GWAS detected additional SNPs not found with only one model. Our results provide new and important insights into the genetic mechanisms of the mammalian multiparous teat phenotype. We demonstrated a torsion sensor based on an in-fiber Mach-Zehnder interferometer (MZI), which consists of two identical cascaded long-period fiber gratings (CLPFGs) inscribed in two-mode fiber (TMF) by high-frequency CO 2 laser. The transmission and mode coupling performances, as well as the spectral properties have been proved theoretically and experimentally. The proposed TMF-CLPFGs could not only be used as an efficient multi-wavelength LP 01 -LP 11 mode converter, but also achieve the vector torsion measurements. As the twist induced, the proposed TMF-CLPFGs present vector torsion ability through the intensity of interference dips varying. The torsion sensitivity of TMF-CLPFGs with spacing length 1 cm can reach −1.07 dB/(rad/m) in counterclockwise direction and 0.9 dB/(rad/m) in clockwise direction. Moreover, the temperature sensitivity of TMF-CLPFGs was measured to be 0.055 dB/°C, which could be negligible to torsion measurement. Abstract: Antimony doped tin oxide supported on carbon black (ATO/C) has been synthesized using an in situ co-precipitation method, and platinum–ATO/C nanoparticles have been prepared using a consecutive polyol process to enhance the catalyst activity for the methanol oxidation reaction. The Pt–ATO/C electrocatalyst is characterized by X-ray diffraction (XRD), transmission electron microscopy (TEM), scanning electron microcopy (SEM), energy dispersive X-ray spectroscopy (EDS) and cyclic voltammetry. The Pt–ATO/C catalyst exhibits a relatively high activity for the methanol oxidation reaction compared to Pt–SnO2/C or commercial Pt/C catalyst. This activity can be attributed to the high electrical conductivities of the Sb-doped SnO2, which induces the electronic effects with Pt catalysts. ZBED6 (zinc finger BED domain containing protein 6) is a transcription factor unique to placental mammals and its interaction with the IGF2 (insulin-like growth factor 2) locus plays a prominent role in the regulation of postnatal skeletal muscle growth. Here, we generated lean Bama miniature pigs by generating ZBED6-knockout (ZBED6−/−) and investigated the mechanism underlying ZBED6 in growth of muscle and internal organs of placental mammals. ZBED6−/− pigs show markedly higher lean mass, lean mass rate, larger muscle fiber area and heavier internal organs (heart and liver) than wild-type (WT) pigs. The striking phenotypic changes of ZBED6-/- pigs coincided with remarkable upregulation of IGF2 mRNA and protein expression across three tissues (gastrocnemius muscle, longissimus dorsi, heart). Despite a significant increase in liver weight, ZBED6-/- pigs show comparable levels of IGF2 expression to those of WT controls. A mechanistic study revealed that elevated methylation in the liver abrogates ZBED6 binding at the IGF2 locus, explaining the unaltered hepatic IGF2 expression in ZBED6-/- pigs. These results indicate that a ZBED6-IGF2-independent regulatory pathway exists in the liver. Transcriptome analysis and ChIP-PCR revealed new ZBED6 target genes other than IGF2, including cyclin dependent kinase inhibitor 1A (CDKN1A) and tsukushi, small leucine rich proteoglycan (TSKU), that regulates growth of muscle and liver, respectively. Chinese indigenous pig breeds carrying the homozygous IGF2 wild-type allele produce low lean meat. We thus generate a lean pig model in Chinese Bama pig by knocking out ZBED6. Background: The mutation of insulin-like growth factor 2 (IGF2 mutation) that a single-nucleotide substitution (G→A) in the third intron of IGF2 abrogates the interaction with zinc finger BED-type containing 6 (ZBED6) and leads to increased muscle mass in pigs. IGF2 mutation knock-in (IGF2 KI) and ZBED6 knockout (ZBED6 KO) lead to changes in IGF2 expression and increase muscle mass in mice and pigs. Long noncoding RNAs (lncRNAs) may participate in numerous biological processes, including skeletal muscle development. However, the role of the ZBED6-lncRNA axis in skeletal muscle development is poorly characterized. Results: In this study, we assembled transcriptomes using RNA-seq data published in previous studies by our group and identified 11,408 known lncRNAs and 2269 potential lncRNAs in seven tissues, heart, longissimus dorsi, gastrocnemius muscle, liver, spleen, lung and kidney, of ZBED6 KO (lean mass model) and WT Bama pigs. ZBED6 affected the expression of 1570 lncRNAs (differentially expressed lncRNAs [DE-lncRNAs]; log2-fold change ≥ 1, nominal p-value ≤ 0.05) in the seven examined tissues. The expressed lncRNAs (FPKM > 0.1) exhibited tissue-specific patterns in WT pigs. Specifically, 3410 lncRNAs were expressed exclusively in only one tissue. Potential functions of lncRNAs were indirectly predicted by searching their target cis- and trans-regulated protein-coding genes. LncRNAs with tissue-specific expression influence numerous genes related to tissue functions. Weighted gene coexpression network analysis (WGCNA) of 1570 DE-lncRNAs between WT and ZBED6 KO pigs was used to define the following six lncRNA modules specific to different tissues: skeletal muscle, heart, lung, spleen, kidney and liver modules. Furthermore, by conjoint analysis of longissimus dorsi data (tissue-specific expression, muscle module and DE-lncRNAs) and ChIP-PCR revealed NONSUSG002145.1 (adjusted p-values = 0.044), which is coexpressed with the IGF2 gene and binding with ZBED6, may play important roles in ZBED6 KO pig skeletal muscle development. Conclusions: These findings indicate that the identified lncRNAs may play essential roles in tissue function and regulate the mechanism of ZBED6 action in skeletal muscle development in pigs. The identification of genome-wide selection signatures can provide insights on the mechanisms of natural and/or artificial selection and uncover genes related to biological functions and/or phenotypes. Tibetan sheep are an important livestock in Tibet, providing meat and wool for Tibetans who are renown for breeding livestock that adapt well to high altitudes. Using whole-genome sequences with an effective sequencing depth of 5×, we investigated the genomic diversity and structure and, identified selection signatures of White Tibetan, Oula and Poll Dorset sheep. We obtained 30,163,679 Single Nucleotide Polymorphisms (SNPs) and 5,388,372 indels benchmarked against the ovine Oar_v4.0 genome assembly. Next, using FST, ZHp and XP-EHH approaches, we identified selection signatures spanning a set of candidate genes, including HIF1A, CAPN3, PRKAA1, RXFP2, TRHR and HOXA10 that are associated with pathways and GO categories putatively related to hypoxia responses, meat traits and disease resistance. Candidate genes and GO terms associated with coat color were also identified. Finally, quantification of blood physiological parameters, revealed higher levels of mean corpuscular hemoglobin measurement and mean corpuscular hemoglobin concentration in Tibetan sheep compared with Poll Dorset, suggesting a greater oxygen-carrying capacity in the Tibetan sheep and thus better adaptation to high-altitude hypoxia. Coat colour is one of the most important economic traits of sheep and is mainly used for breed identification and characterization. This trait is determined by the biochemical function, availability and distribution of phaeomelanin and eumelanin pigments. In our study, we conducted a genome-wide association study to identify candidate genes and genetic variants associated with coat colour in 75 Chinese Tan sheep using the ovine 600K SNP BeadChip. Accordingly, we identified two significant SNPs (rs409651063 at 14.232 Mb and rs408511664 at 14.228 Mb) associated with coat colour in the MC1R gene on chromosome 14 with −log10(P) = 2.47E-14 and 1.00E-13, respectively. The consequence of rs409651063 was a missense variant (g.14231948 G>A) that caused an amino acid change (Asp105Asn); however, the second SNP (rs408511664) was a synonymous substitution and is an upstream variant (g.14228343G>A). Moreover, our PCR analysis revealed that the genotype of white sheep was exclusively homozygous (GG), whereas the genotypes of black-head sheep were mainly heterozygous (GA). Interestingly, allele-specific expression analysis (using the missense variant for the skin cDNA samples from black-head sheep) revealed that only the G allele was expressed in the skin covered with white hair, while both the G and A alleles were expressed in the skin covered with black hair. This finding indicated that the missense mutation that we identified is probably responsible for white coat colour in Tan sheep. Furthermore, qPCR analysis of MC1R mRNA level in the skin samples was significantly higher in black-head than white sheep and very significantly higher in GA than GG individuals. We present the properties of magnetic fields (B fields) in two clumps (clump 1 and clump 2), located at the waist of the bipolar H ii region Sh 2-201, based on James Clerk Maxwell Telescope SCUBA-2/POL-2 observations of 850 μm polarized dust emission. We find that B fields in the direction of the clumps are bent and compressed, showing bow-like morphologies, which we attribute to the feedback effect of the H ii region on the surface of the clumps. Using the modified Davis–Chandrasekhar–Fermi method, we estimate B-field strengths of 266 and 65 μG for clump 1 and clump 2, respectively. From virial analyses and critical mass ratio estimates, we argue that clump 1 is gravitationally bound and could be undergoing collapse, whereas clump 2 is unbound and stable. We hypothesize that the interplay of the thermal pressure imparted by the H ii region, the B-field morphologies, and the various internal pressures of the clumps (such as magnetic, turbulent, and gas thermal pressures) has the following consequences: (a) formation of clumps at the waist of the H ii region; (b) progressive compression and enhancement of the B fields in the clumps; (c) stronger B fields that will shield the clumps from erosion by the H ii region and cause pressure equilibrium between the clumps and the H ii region, thereby allowing expanding ionization fronts to blow away from the filament ridge, forming bipolar H ii regions; and (d) stronger B fields and turbulence that will be able to stabilize the clumps. Background: Circular RNA (circRNA) is produced during the splicing of mRNA (in addition to linear splicing) and is part of the gene regulatory network. The temporal expression patterns the different developmental stages were inseparable from these molecules' function. Results: Skeletal muscles of Anhui white goat (AWG) across seven fetal to postnatal development stages were sequenced and 21 RNA sequencing libraries were constructed. We thereby identified 9090 circRNAs and analyzed their molecular properties, temporal expression patterns, and potential functions at the different stages. CircRNAs showed complexities and diversity of formation as the same host gene produces multiple isoforms of these nucleic acids with different expression profiles. The differential expression of 2881 circRNAs (DECs, P < 0.05) was identified and four were randomly selected and validated by qPCR. Moreover, 1118 DECs under strict selected (SDECs, |log2FC| > 2 and P-adj value < 0.01) showed 4 expression trends (Clusters 0, 19, 16 and 18). Cluster 0 molecules had increasing expression at all stages with effects on muscle through metabolism, regulation of enzyme activity, and biosynthesis. Cluster 16 circRNAs had high expression in the early and late stages and are involved in "Wnt signaling pathway", "AMPK signaling pathway" and others. Cluster 18 molecules were mainly expressed at F120 and participate in "cytoskeletal protein binding", "Notch signaling pathway" and so on. Cluster 19 circRNAs were down-regulated at all stages and related to muscle structure and development. Lastly, the SDECs divided the period of skeletal muscle development into three transitional stages: stage 1 (F45 to F90), which related to muscle satellite cell proliferation and muscle fiber structure; stage 2 (F90 to B1), in which the attachment of the cytoplasmic surface to the actin cytoskeleton initiates; and stage 3, which involved the "cGMP-PKG signaling pathway". Moreover, the paraffin sections messages also validated that there are three transitional stages of skeletal muscle development. Conclusion: Our current study provides a catalog of goat muscle-related circRNAs that can stratify skeletal muscle development fetus 45 days to newborn 90 days into three developmental stages. A high-sensitivity bending sensor based on dual-dip long period fiber gratings (LPFGs) in a single-mode fiber is proposed. The length of the gratings is very short only 7.2 mm. The asymmetric perturbation in fiber is generated during the fabrication, which makes the dual-dip LPFGs show different bending sensing characteristics at opposite directions. Meanwhile, the highest bending sensitivity of −35.91 nm/m−1 and −20.74 dB/m−1 can be obtained at bending directions of 0° and 180°, respectively. Furthermore, the torsion and temperature sensing characteristics are investigated experimentally. The torsion sensitivities follow sine function showing potential for directional sensing. Adipose-derived stem cells (ADSCs) that are enriched in adipose tissue with multilineage differentiation potential have become an important tool in therapeutic research and tissue engineering. Certain breeds of sheep exhibit a unique fat tail trait such that tail tissue accounts for approximately 10 % of body weight and can provide an excellent source of ADSCs. Here, we describe isolation of primary ADSCs from ovine embryonic fat tail tissues that displayed high self-renewal capacity, multilineage differentiation and excellent adipogenic ability. Through transcriptome analysis covering ADSCs differentiating into adipocytes, 37 transcription factors were involved in early transcriptional events that initiate a regulatory cascade of adipogenesis; the entire adipogenic activity consists of a reduction in proliferation ability and upregulation of genes related to lipid generation and energy metabolism, as well as several genes associated with myogenesis. Furthermore, Comparative transcriptome analysis across species (sheep, human, and mouse) revealed enhanced basal metabolic ability in differentiating ovine ADSCs, which may relate to the excellent adipogenic capability of these cells. We also identified a small evolutionarily conserved gene set, consisting of 21 and 22 genes exhibiting increased and decreased expression, respectively. Almost half (20) of these genes have not previously been reported to regulate adipogenesis in mammals. In this study, we identified important regulators that trigger ovine adipocyte differentiation, main biological pathways involved in adipogenesis as well as the evolutionarily conserved genes governing adipogenic process across species. Our study provides a novel excellent biomaterial and novel genes regulating adipogenesis for cellular transplantation therapy and investigations of fat metabolism. Summary: Mefunidone (MFD), a novel derivation of pirfenidone (PFD), exhibits promising anti‐renal fibrosis activity. To avoid early development failures and attrition in clinical phase 1, an evaluation of the preclinical pharmacokinetic (PK) properties of MFD was conducted in this study. The absolute bioavailability was evaluated in Sprague Dawley rats and cynomolgus monkeys, who received an intragastric or intravenous administration at the dose of 31.25 mg/kg and 10 mg/kg, respectively. Tissue distribution and plasma protein binding studies were performed to evaluate the distribution characteristics of MFD. The enzymes involved in the metabolism of MFD were assayed by calculating the inhibition rate using the in vitro incubation system of human liver microsomes. The metabolic pathway of MFD was also examined in the in vitro incubation system as well as in rats in vivo. The results revealed satisfactory absorption of MFD in vivo, with an absolute bioavailability of MFD in rats and monkeys of approximately 77.2% and 70.0%, respectively. MFD was rapidly distributed to all tissues and was highly concentrated in the kidney, which was the target organ of MFD. The plasma protein binding rates in rat, monkey, and human plasma were 36.40%–41.68%, 30.88%–63.92%, and 37.75%–57.77%, respectively. MFD was primarily metabolized by the enzymes CYP3A4, CYP2C9, and CYP2C8. The metabolic pathway of MFD is nitrogen demethylation of the pyridine ring of the MFD nucleus, hydroxylation of the aromatic ring, and demethylation of the piperazine ring. In conclusion, MFD exhibited good PK properties in the initial preclinical PK study. Abstract As a PAC-1 derivative, SM-1 exhibts a promising antitumour property. To better understand the relationship between the drug concentrations and pharmacological effects, both liquid chromatography coupled with tandem mass spectrometry and high performance liquid chromatography methods were developed and validated in the work. Those methods were then applied to the pharmacokinetics (PK), tissue distribution and plasma protein binding (PPB) studies of SM-1. As a results, the proposed methods were demonstrated to be accurate, precise and stable for the analysis of the SM-1 in plasma and tissue samples. Meanwhile, the PK parameters of SM-1 showed that SM-1 had good PK properties. SM-1 had good absorption in the body, with 59.01% of the absolute bioavailability in rats and 55.63% of that in dogs. SM-1 rapidly distributed to all tissues, with the highest distribution in the lung and less in the brain and muscle. The PPB rates in rat plasma, dog plasma, and human plasma were 91.1%, 91.2%, and 90.7%, respectively. These good PK properties will contribute SM-1 to be a promising anti-tumour candidate. Background: Long intergenic noncoding RNAs (lincRNAs) have been recognized in recent years as key regulators of biological processes. However, lincRNAs in goat remain poorly characterized both across various tissues and during different developmental stages in goat (Capra hircus). Results: We performed the genome-wide discovery of the lincRNAs in goat by combining the RNA-seq dataset that were generated from 28 cashmere goat skin samples and the 12 datasets of goat tissues downloaded from the NCBI database. We identified a total of 5546 potential lincRNA transcripts that overlapped 3641 lincRNA genes. These lincRNAs exhibited a tissue-specific pattern. Specifically, there are 584 lincRNAs expressed exclusively in only one tissue, and 91 were highly expressed in hair follicle (HF). In addition, 2350 protein-coding genes and 492 lincRNAs were differentially expressed in the skin of goat. The majority exhibited the remarkable differential expression during the transition of the goat skin from the May–June to August–October time point, which covered the different seasons. Fundamental biological processes, such as skin development, were significantly enriched in these genes. Furthermore, we identified several lincRNAs highly expressed in the HF, which exhibited not only the co-expression pattern with the key factors to the HF development but also the activated expression in the August to October time point. Intriguingly, one of spatiotemporal lincRNAs, linc-chig1598 could be a potential regulator of distal-less homeobox 3 expression during the secondary hair follicle growth. This work presents a novel rapid and sensitive label-free electrochemical method for the detection of bacteria on surface nanostructures. A simple electrochemical deposition and calcination method is employed to prepare different gold nanostructures on FTO substrate. The sensor based on nanostructure gold exhibits excellent linear relation between E. coli DH5α bacteria and the changes of ΔRct, especially FTO-GEDC-D30, with a correction coefficient R2 = 0.998. Both the spectrophotometric (OD600 methods) and fluorescence-staining methods also verified the reliability of electrochemical impedance spectroscopy (EIS) methods for evaluating the antibacterial activity of the gold nanostructure. Heteroplasmy is the presence of more than one mitochondrial DNA (mtDNA) variant within a cell, tissue, or individual. In this study, sequence variation was investigated in the control region (CR) of mitochondrial DNA (mtDNA) from 135 individuals belonging to five primary domestic Bactrian camel breeds in China and Mongolia. Due to variation of the repeat unit G(T/C)(AC)n, length heteroplasmy was detected within each camel by direct sequencing and fragment analysis. A high occurrence of mtDNA heteroplasmy, up to 100 percentages was observed in five camel populations. Our study provides the first evidence of extensive length heteroplasmy in Bactrian camels. The Siberian tiger, Panthera tigris altaica, is an endangered species, and much more work is needed to protect this species, which is still vulnerable to extinction. Conservation efforts may be supported by the genetic assessment of wild populations, for which highly specific microsatellite markers are required. However, only a limited amount of genetic sequence data is available for this species. To identify the genes involved in the lung transcriptome and to develop additional simple sequence repeat (SSR) markers for the Siberian tiger, we used high-throughput RNA-Seq to characterize the Siberian tiger transcriptome in lung tissue (designated 'PTA-lung') and a pooled tissue sample (designated 'PTA'). Approximately 47.5 % (33,187/69,836) of the lung transcriptome was annotated in four public databases (Nr, Swiss-Prot, KEGG, and COG). The annotated genes formed a potential pool for gene identification in the tiger. An analysis of the genes differentially expressed in the PTA lung, and PTA samples revealed that the tiger may have suffered a series of diseases before death. In total, 1062 non-redundant SSRs were identified in the Siberian tiger transcriptome. Forty-three primer pairs were randomly selected for amplification reactions, and 26 of the 43 pairs were also used to evaluate the levels of genetic polymorphism. Fourteen primer pairs (32.56 %) amplified products that were polymorphic in size in P. tigris altaica. In conclusion, the transcriptome sequences will provide a valuable genomic resource for genetic research, and these new SSR markers comprise a reasonable number of loci for the genetic analysis of wild and captive populations of P. tigris altaica. In this article, a fast and sensitive nonenzymatic glucose sensor is reported utilizing a glassy carbon electrode modified by synthesizing nanocomposites of nickel nanoparticle–attapulgite-reduced graphene oxide (Ni NPs/ATP/RGO). A facile one-step electrochemical co-deposition approach is adopted to synthesize Ni NPs-ATP-RGO nanocomposites via electrochemical reduction of mixed precursor solution containing graphene oxide (GO), attapulgite (ATP) and nickel cations (Ni 2+ ) at the cathode potentials. This strategy results in simultaneous depositions of ATP, cathodic reduction of Ni 2+ into nickel nanoparticles under acidic conditions, and in situ reduction of GO. The as-prepared NiNPs/ATP/RGO-based glucose sensor exhibits outstanding performance for enzymeless glucose sensing with sensitivity (1414.4 μA mM −1 cm −2 ), linear range (1–710 μM) and detection limit (0.37 μM). Copy number variation ( CNV), an essential form of genetic variation, has been increasingly recognized as one promising genetic marker in the analysis of animal genomes. Here, we used the Equine 70K single nucleotide polymorphism genotyping array for the genome-wide detection of CNVs in 96 horses from three diverse Chinese breeds: Debao pony ( DB), Mongolian horse ( MG) and Yili horse ( YL). A total of 287 CNVs were determined and merged into 122 CNV regions ( CNVRs) ranging from 199 bp to 2344 kb in size and distributed in a heterogeneous manner on chromosomes. These CNVRs were integrated with seven existing reports to generate a composite genome-wide dataset of 1558 equine CNVRs, revealing 69 (56.6%) novel CNVRs. The majority (69.7%) of the 122 CNVRs overlapped with 438 genes, whereas 30.3% were located in intergenic regions. Most of these genes were associated with common CNVRs, which were shared by divergent horse breeds. As many as 60, 42 and 91 genes overlapping with the breed-specific ss were identified in DB, MG and YL respectively. Among these genes, FGF11, SPEM1, PPARG, CIDEB, HIVEP1 and GALR may have potential relevance to breed-specific traits. These findings provide valuable information for understanding the equine genome and facilitating association studies of economically important traits with equine CNVRs in the future. Four-horned sheep are an ideal animal model for illuminating the genetic basis of horn development. The objective of this study was to locate the genetic region responsible for the four-horned phenotype and to verify a previously reported polled locus in three Chinese breeds. A genome-wide association study ( GWAS) was performed using 34 two-horned and 32 four-horned sheep from three Chinese indigenous breeds: Altay, Mongolian and Sishui Fur sheep. The top two significant single nucleotide polymorphisms ( SNPs) associated with the four-horned phenotype were both located in a region spanning positions 132.6 to 132.7 Mb on sheep chromosome 2. Similar locations for the four-horned trait were previously identified in Jacob, Navajo-Churro, Damara and Sishui Fur sheep, suggesting a common genetic component underlying the four-horned phenotype. The two identified SNPs were both downstream of the metaxin 2 ( MTX2) gene and the HOXD gene cluster. For the top SNP- OAR2:g.132619300G>A-the strong associations of the AA and AG genotypes with the four-horned phenotype and the GG genotype with the two-horned phenotype indicated the dominant inheritance of the four-horned trait. No significant SNPs for the polled phenotype were identified in the GWAS analysis, and a PCR analysis for the detection of the 1.8-kb insertion associated with polled sheep in other breeds failed to verify the association with polledness in the three Chinese breeds. This study supports the hypothesis that two different loci are responsible for horn existence and number. Melatonin can modulate neural stem cell ( NSC) functions such as proliferation and differentiation into NSC-derived pluripotent stem cells (N- iPS) in brain tissue, but the effect and mechanism underlying this are unclear. Thus, we studied how primary cultured bovine NSCs isolated from the retinal neural layer could transform into N- iPS cell. NSCs were exposed to 0.01, 0.1, 1, 10, or 100 μ m melatonin, and cell viability studies indicated that 10 μ m melatonin can significantly increase cell viability and promote cell proliferation in NSCs in vitro. Thus, 10 μ m melatonin was used to study miR-302/367-mediated cell reprogramming of NSCs. We noted that this concentration of melatonin increased reprogramming efficiency of N- iPS cell generation from primary cultured bovine NSCs and that this was mediated by downregulation of apoptosis-related genes p53 and p21. Then, N- iPS cells were treated with 1, 10, 100, or 500 μ m melatonin, and N- iPS (M-N- iPS) cell proliferation was measured. We noted that 100 μ m melatonin increased proliferation of N- iPS cells via increased phosphorylation of intracellular ERK1/2 via activation of its pathway in M-N- iPS via melatonin receptors 1 ( MT1). Finally, we verified that N- iPS cells and M-N- iPS cells are similar to typical embryonic stem cells including the expression of pluripotency markers (Oct4 and Nanog), the ability to form teratomas in vivo , and the capacity to differentiate into all three embryonic germ layers. Background: Timely diagnosis of meningeal carcinomatosis is often difficult even with the assistant of magnetic resonance imaging examination, cerebrospinal fluid analysis, or both. To the best of our knowledge, gadobutrol-enhanced MRI has not been reported in the diagnosis of meningeal carcinomatosis. Here we present two cases where meningeal carcinomatosis was identified on gadobutrol-enhanced magnetic resonance imaging. Case Presentation: We identified two cases of meningeal carcinomatosis who had been diagnosed with malignant tumors several years ago. Both patients presented with progressive headache and seizures. Gadopentetate dimeglumine-enhanced magnetic resonance imaging of the brain was performed and did not detect any abnormality of meninges. Lumbar puncture was performed repeatedly, but cerebrospinal fluid cytology showed no evidence of malignant cells. Finally the gadobutrol-enhanced magnetic resonance imaging detected the meningeal metastasis, and supported the diagnosis of meningeal carcinomatosis. Conclusion: Gadobutrol provides higher lesion conspicuity and enhances lesion detection in meningeal metastasis compared with gadopentetate dimeglumine. Our observation is a cue to analyze the accuracy in the diagnosis of meningeal carcinomatosis, and presents a choice that may facilitate early diagnosis. [ABSTRACT FROM AUTHOR]

To explore if the regulation at post-transcriptional level of follicular phase (Fols) to luteal phase (Luts) transition occurs in the ovaries of Anhuai goats, the differentially expressed microRNAs (miRNAs) of ovaries in the Fols and Luts were analyzed using Solexa sequencing in the study. In total, 320 known miRNAs were co-expressed in the two phases, 339 and 353 known miRNAs were expressed in the ovary in the Fols and Luts, respectively. In addition, 45 novel miRNAs were co-expressed in the two phases, 70 and 94 novel miRNAs were expressed in the ovary in the Fols and Luts, respectively. Let-7f was the highest expressed significantly different known miRNA in the two phases, and mir-159 was the highest expressed significantly different novel miRNA in the two phases, which may participate in the follicular-luteal transition of Anhuai goats. GO annotation and KEGG pathway analysis were applied to analyze the target genes of differentially expressed miRNAs detected in the two phases.
