Inflammation is a pivotal pathological factor in colorectal cancer (CRC) initiation and progression, and modulating this inflammatory state has the potential to ameliorate disease progression. NR4A receptors have emerged as key regulators of inflammatory pathways that are important in CRC. Here, we have examined the effect of NR4A agonist, Cytosporone B (CsnB), on colorectal tissue integrity and its effect on the inflammatory profile in CRC tissue ex vivo. Here, we demonstrate concentrations up 100 μM CsnB did not adversely affect tissue integrity as measured using transepithelial electrical resistance, histology and crypt height. Subsequently, we reveal through the use of a cytokine/chemokine array, ELISA and qRT-PCR analysis that multiple pro-inflammatory mediators were significantly increased in CRC tissue compared to control tissue, which were then attenuated with the addition of CsnB (such as IL-1β, IL-8 and TNFα). Lastly, stratification of the data revealed that CsnB especially alters the inflammatory profile of tumours derived from males who had not undergone chemoradiotherapy. The contribution of non-timber forest products (NTFPs) to community sustainability remains an understudied topic of academic inquiry. Focusing on southern and central Ontario, Canada, the ways in which NTFPs may contribute to sustainable livelihoods are explored, along with how a fragmented sociopolitical landscape and the potential impacts of climate change might impede the development of the sector. Following interviews with twenty NTFP producers, four key themes highlighting the major challenges and opportunities are discussed. This paper contends that for an economically viable, socially just and environmentally sustainable NTFP industry to emerge, political-ecological power relations informed by critical social and environmental justice thinking must be understood and addressed. Climate change, as a wicked problem tied to stories and ceremonies, provides us with the opportunity to raise essential questions about our values and power structures and to think creatively about our future. The work of literary scholars, such as J. Edward Chamberlin, allows us to examine the nature of these climate change narratives by imaginatively (re) interpreting the common ground between Them and Us, and by transcending our localism, while also recognizing the validity of traditional place-based knowledges. In his text, "If This Is Your Land, Where Are Your Stories?: Finding Common Ground" (2004), Chamberlin's notions of "believe it and not" (160), finding common ground through stories and ceremonies, "babblers and doodlers" (8), and the barbaric versus the civilized demonstrate the problematic nature of current climate change approaches, as proposed by several Western academics. In this essay, the authors question and extend Chamberlin's contention that how we believe is less problematic than what we believe, as well as the dichotomy implied by the "Them and Us" labels (8). The authors contend that imaginative, transdisciplinary approaches, contextualized by our lived realities, grounded in environmental and social justice, and focussed on processes rather than outcomes are the principles that should underpin climate change action plans. Spurred by the literature on climate change and its calls for undertaking holistic research that more fully integrates the work of biophysical and social scientists, this article responds to the question: To what extent has climate change research in Canada embraced and been guided by the theories and tenets associated with interdisciplinarity and to what extent have integrated approaches been sensitive to cross-cultural perspectives? It provides an overview of some of the epistemological issues raised in the interdisciplinarity literature that particularly impact research development and design. Furthermore, since much of the climate change literature that claims to be integrated or interdisciplinary draws from Indigenous Knowledge (IK), additional insights are provided from this perspective. The article develops a framework that can be used to undertake and/or evaluate research in a way that acknowledges 'upstream' epistemological issues. The framework is then used to evaluate a comprehensive database (n = 282) of Canadian climate change articles. It is argued that an interdisciplinary approach adds a critical voice to the literature on integrated climate change research and is valuable because of its focus on epistemology and methodology. The article advocates the creation of a space for inter-epistemological acknowledgement in which the academy develops an ethos of self-reflection, while simultaneously respecting and integrating parallel knowledge frameworks, such as IK. It examines the role of imagination and the imaginative process in the artistic creation of the play, also focusing on the influence of cultural imagery on Williams. Williams' appreciation for the book "Á la recherche du temps perdu" by Marcel Proust, allusions to the book "Don Quixote" by Miguel de Cervantes, and the influence of American poetry on the play are also addressed.

Abstract: Yersinia enterocolitica (Y. enterocolitica) is a known zoonotic pathogen and is often found in pig tonsils as the primary site of colonisation. In this study we investigated whether or not Y. enterocolitica could be recovered from canine tonsils. During a study on the prevalence of Y. enterocolitica in animal populations in Ireland, 144 canine tonsils and 72 canine rectal swabs were procured over a ten-month period and subjected to microbiological examination for the presence of this human pathogen. Molecular methods were used to determine virulence and all strains were negative for the chromosomally mediated virulence factor (ail) and plasmid-encoded adhesion molecule (pYad). Y. enterocolitica was recovered from 25 of 216 (12%) samples. Twenty-four strains were from tonsils along with one from a rectal swab. All were biotype 1A. Antimicrobial resistance profiling showed two of 25 (8%) were resistant to cephalothin and the remaining strains were resistant to ampicillin and cephalothin with six of these additionally resistant to streptomycin. Our evidence that a human pathogen may be harboured in the oral cavity of dogs’ adds a new dimension to the epidemiology of this organism, identifying a potential public health risk following exposure to dogs. Abstract: The presence of microbiological hazards in foodstuffs including, Salmonella, form a major source of food-borne diseases in humans. In-line milk filters from 97 liquid milk production holdings in Cork, the largest dairy region in Ireland, were surveyed for the presence of Salmonella species at herd level over a 2-year period (September 2001–September 2003). Each dairy farm was visited 6 times at 4 monthly intervals (denoted by cycles A–F). Six of the 97 herds (6%) were positive. Ten isolates were detected based on culture methods. These included five (5%) Salmonella Typhimurium DT104, 4 (4%) Salmonella Dublin, and 1 (1%) Salmonella Agona from a total of 556 filters. During cycle C, in addition to the milk filters, a bulk tank milk (BTM) sample was procured from each dairy holding and analysed but no Salmonella were isolated. For comparison purposes a further 26 temporal veterinary clinical isolates (21 S. Typhimurium of varying phage type, and 5 S. Dublin) were procured from the Cork Veterinary Clinical Diagnostic Laboratory, Cork. The study collection showed resistance to one or more antimicrobial agents. During the study, Salmonella spp. were isolated from five of the herds prior to any clinical signs in the farm animals. Pulsed-field gel electrophoresis (PFGE) profiles indicated clonality among the isolates pre- and post-clinical illness. In this study, antibiotic resistance profiles, and the presence of class 1 integrons were determined for 108 Salmonella isolates comprising 37 serotypes cultured from a variety of sources between 1953 and 2004. Antibiogram analyses showed that all isolates were resistant to streptomycin/spectinomycin. Molecular analysis revealed that 50% of the collection contained an integrase-encoding gene ( int1) and 25% contained class 1 integrons. A Salmonella Wien isolate possessing a complete class 1 integron with a dfrA5-ereA2 gene arrangement within the variable region was characterized. [ABSTRACT FROM AUTHOR]

Abstract: Escherichia coli O157 is a major etiological agent of food-borne illness. Bovine animals are recognized reservoirs for this organism and represent a significant source from where these pathogens can enter the food chain. Food products derived from these animals are convenient vehicles, and are often the focal point(s) of infection. As a useful strategy to provide herd-level surveillance and to investigate for the presence of this pathogen in a population of Irish dairy cattle, milk filters from 97 farms were analysed by conventional culture and other methods. Five hundred and thirty-six milk filters were evaluated over a 2-year period. Filters from 12 of the 97 farms (12%) were found to contain E. coli O157, based on culture methods. Sixteen verocytotoxigenic E. coli O157 organisms were recovered and characterized in detail. The farm families in each case were consuming raw milk from their respective herds. The potential risk to public health associated with the detection of E. coli O157 and the local consumption of raw milk are discussed. [Copyright &y& Elsevier]

Photovoice aims to enable people to record and reflect their community's strengths and challenges, to encourage group dialogue and knowledge about important issues through group discussions and to inform policymaking. While primarily utilized in the health field, an emerging area of focus is to use photovoice in an emergency management or climate change context. Through work conducted in two rural areas recovering from natural disasters in Ontario, Canada, this research, focused on critical infrastructure disaster recovery, underscores the value of undertaking a comparative case-study approach and offers a detailed reporting of the fieldwork methodology. We argue that photovoice has the potential to solicit poorly understood rural and Indigenous community member perspectives, thereby augmenting locally relevant, place-based information and, ideally, empowering voices that are often under-represented in municipal and provincial decision-making processes. We offer lessons learned related to the project's processes and outcomes, and outline the applicability of photovoice for emergency management and climate change research. [ABSTRACT FROM AUTHOR]

Orphan nuclear receptor subfamily 4A (NR4A) are key regulators of inflammatory responses, largely by their interactions with NF-κB. Over the last decade, several NR4A modulators have been developed, and they are showing potential as therapeutics, although their widespread use in laboratory settings is limited. Here, we have examined, using myeloid cell line THP-1, whether the NR4A modulator 3-[(4-Chlorophenyl)-(1H-indol-3-yl)methyl]-1H-indole (C-DIM12) can alter the inflammatory outcome of six inflammatory ligands: lipopolysaccharide (LPS), tumour necrosis factor alpha (TNFα), interleukin-1 beta (IL-1β), flagellin (FL), lipoteichoic acid (LTA), and zymosan (ZY). We demonstrate that C-DIM12 (10 µM) selectively alters the secretion of inflammatory chemokine MCP-1 following exposure to distinct inflammatory ligands in a concentration-dependent manner. Furthermore, data obtained from THP-1 Lucia cell experiments show that 10 µM C-DIM12, and not 1 µM C-DIM12, can significantly attenuate the increased NF-κB transcriptional activity observed following the exposure to several inflammatory ligands (LPS, FL, TNFα, LTA, and ZY). Lastly, experimental analysis confirms that the cellular action(s) of C-DIM12 is independent of changes in metabolic parameters. Thus, these data contribute to the understanding of how the NR4A modulator C-DIM12 alters inflammatory responses in a myeloid cell following exposure to multiple ligands. [ABSTRACT FROM AUTHOR]

Purpose: Transanal minimally invasive surgery (TAMIS) is a surgical alternative to transanal endoscopic microsurgery (TEM), transanal excision and proctectomy in the management of benign rectal polyps and early rectal cancers. Low anterior resection syndrome (LARS) describes the constellation of symptoms which result from and are common after distal colorectal resection. Symptoms include incontinence, frequency, urgency and evacuatory dysfunction. The aim of the current study was to prospectively evaluate pre- and post-operative LARS in patients who undergo TAMIS. Methods: We conducted a prospective analysis of a consecutive series of patients who underwent TAMIS at our institution between January 2021 and February 2022. A LARS questionnaire was undertaken preoperatively, at 1 month and at 6 months post-operatively. Results: Twenty patients were recruited to this pilot study. The mean age was 63 ± 12 years, 11 of the patients were male, mean pre-operative BMI was 29 ± 6 kg/m2, and 30% (n = 6) of patients underwent TAMIS for an invasive rectal cancer, with all patients receiving an R0 resection. Mean distance from the anal verge was 5.7 ± 3.2 cm, and mean lesion diameter was 46 ± 20.5 mm. A statistically significant interval reduction was observed between preoperative (20.3 ± 12.9) and 6-month post-operative (12.6 ± 9.7) LARS scores (p = 0.02) and also between 1-month (18.2 ± 10.6) and 6-month post-operative scores (p = 0.01). Conclusions: We noted a high prevalence of LARS across our cohort preoperatively, and this had improved significantly at 6-month review post-TAMIS. Canada is the world's largest producer of maple syrup. Syrup production depends on weather and climatic conditions of the sugarbush. However, forest ecosystems are highly sensitive to climate change. The effect of rapidly changing precipitation and temperature patterns on tree species is of concern as these long-lived organisms cannot quickly adapt to the new environmental conditions in which they find themselves. As temperatures increase it is expected that there will be a change in species' ranges poleward. This study uses Multi-Criteria Decision Making (MCDM) and Geographic Information System (GIS) weighted sum analysis to project near future (2050) and distant future (2100) suitability maps of sugar maple (Acer saccharum) habitat in Ontario associated with three different Intergovernmental Panel on Climate Change (IPCC) Fourth Assessment Report (AR4) scenarios. Our maps project an overall decrease in the amount of suitable habitat within the current sugar maple range under the scenarios modelled, which intensifies in the later time period. Furthermore there is a projected shift in central and southern Ontario from a region dominated by suitable habitat to one dominated by unsuitable habitat. [ABSTRACT FROM AUTHOR]

Octreotide is approved as a one-month injectable for treatment of acromegaly and neuroendocrine tumours. Oral delivery of the octapeptide is a challenge due mainly to low intestinal epithelial permeability. The intestinal permeation enhancer (PE) salcaprozate sodium (SNAC) has Generally Regarded As Safe (GRAS) status and is a component of an approved oral peptide formulation. The purpose of the study was to examine the capacity of salcaprozate sodium (SNAC), to increase its permeability across isolated rat intestinal mucosae from five regions and across human colonic mucosae mounted in Ussing chambers. Apical-side buffers were Kreb's-Henseleit (KH), fasted simulated intestinal fluid (FaSSIF-V2), rat simulated intestinal fluid (rSIF), and colonic simulated intestinal fluid (FaSSCoF). The basal apparent permeability coefficient (P app) of [3H]-octreotide was equally low across rat intestinal regional mucosae in KH, rSIF, and FaSSIF-V2. Apical addition of 20 mM SNAC increased the P app across rat tissue in KH: colon (by 3.2-fold) > ileum (3.4-fold) > upper jejunum (2.3-fold) > duodenum (1.4-fold) > stomach (1.4-fold). 20 mM and 40 mM SNAC also increased the P app by 1.5-fold and 2.1-fold respectively across human colonic mucosae in KH. Transepithelial electrical resistance (TEER) values were reduced in the presence in SNAC especially in colonic regions. LC-MS/MS analysis of permeated unlabelled octreotide across human colonic mucosae in the presence of SNAC indicated that [3H]-octreotide remained intact. No gross damage was caused to rat or human mucosae by SNAC. Attenuation of the effects of SNAC was seen in rat jejunal mucosae incubated with FaSSIF-V2 and rSIF, and also to some extent in human colonic mucosae using FaSSCoF, suggesting interaction between SNAC with buffer components. In conclusion, SNAC showed potential as an intestinal permeation enhancer for octreotide, but in vivo efficacy may be attenuated by interactions with GI luminal fluid contents. The goal of this article is to introduce activity systems as a methodological tool in narrative inquiry to gain a holistic understanding of socially shared experiences from an examination of documents. The research question was how can qualitative researchers use activity systems as a tool for engaging in narrative inquiry of socially shared experiences to uncover new meanings by constructing a story? In this article, we share a sample analysis of our experience relying on documents and media as a form of narrative to begin to understand the socially shared human activity associated with net neutrality and its potential impact on U.S. residents.
