Given rapid global urban development, increases to impervious surfaces, urban population growth, building construction, and energy consumption result in the urban heat island (UHI) phenomenon. However, the spatial extent of UHIs is not clearly mapped in many UHI studies based on a remote sensing approach. Therefore, we developed a method to extract the spatial extent of the UHI during the period from 2000 to 2021 in Nanjing, China, and explored the impact of urban two- and three-dimensional expansion on UHI spatial extent and UHI intensity. After cropland effects (i.e., bare soil) were eliminated, our proposed method combines the Getis-Ord-Gi* and the standard deviation of the normalized difference vegetation index (NDVI STD) to extract the UHI area from Landsat 5 and Landsat 8 images using land surface temperature (LST) spatial autocorrelation characteristics and the seasonal variation of vegetation. Our results show the following: (1) Bare farmland has a large influence on the extraction results of UHI—combined with the seasonal variation characteristics of NDVI STD, the impact of bare soil on UHI extraction was highly reduced, strongly improving the accuracy of UHI extraction. (2) The dynamics of the UHI area are consistent with the changes in the built-up area in Nanjing at both spatial and temporal scales, but with the increase of the urban green ratio, the UHI area of mature urban areas trends to decrease due to the cooling effect of green space. (3) The accumulation of population and GDP promote the vertical expansion of urban buildings. Facing the huge energy consumption of buildings, it is highly practical to study every aspect of energy-saving technologies for heating and cooling devices. Solar thermal technology and air/water source heat pump are two popular energy-saving technologies, which could be combined into a composite heating system with good energy-saving effect and efficiency. This paper constructs a solar-air source composite heating system, analyzes its thermodynamic features, and builds up the corresponding thermodynamic model. In addition, the authors modeled the daytime and nighttime heat balances, as well as the monthly cumulative heat supply for the constructed system. Finally, a dual-tank composite heating system was designed with independent heat supply/storage. The proposed models and system were proved valid and scientific through experiments. Structural health monitoring plays a crucial role in maintaining the long lifespan of large-scale infrastructure. In this paper, a new type of sensing technique for localized structural damage is proposed. This technology can be adopted to measure cracks by using a transparent capacitive sensor. The capacitive sensor is fabricated using a dielectric elastomer with attached stretchable transparent electrodes. The electrodes are ionic conductors. The materials of the sensor are transparent and inexpensive, which make the sensory sheets a feasible application for large surfaces without impeding optical signals. The performance transparency of the sensor is beneficial to visually detect the expansion of cracks through other non-destructive testing technologies. An experiment was conducted to determine the robustness of the capacitive sensor. Tests on perspex sheet and over-reinforced concrete beam demonstrated the ability of the sensor to detect strains and cracks over the surface that it covered. [ABSTRACT FROM AUTHOR]

Background: Subjective memory impairment (SMI) is common in older people. The aim of this study was to investigate the factors influencing SMI among older people in China, with specific focus on the interaction effect of midday napping duration and depressive symptoms on the risk of SMI. Methods: Using a dataset representative of the Chinese population from a longitudinal study of health and retirement in China, subjects with SMI were screened using the question "how do you feel about your memory now?" and the Mini-Mental State Examination. A logistic regression model was applied to explore the factors affecting SMI. Additive and multiplicative models were used to analyze the interaction effect of midday napping duration and depressive symptoms on the risk of SMI. Results: We enrolled 8,254 subjects included and the incidence of SMI was 63.9%. Depressive symptoms, nap time, and physical activity were influencing factors of SMI. Midday napping duration and depressive symptoms had positive additive interaction effects on the risk of SMI. When extended-length naps and depressive symptoms coexisted, the risk of SMI was 1.06 times greater than that for either alone (RERI, relative excess risk due to interaction = 0.27, 95% CI = 0.07–0.43; AP, attributable proportion = 0.14, 95% CI = 0.01–0.23; S, synergy index = 1.06, 95% CI = 0.57–1.62). When short naps and depressive symptoms coexisted, the risk of SMI was 1.2 times higher than that for either alone (RERI = 0.12, 95% CI=-0.14–0.39; AP = 0.13, 95% CI=-0.07–0.22; S = 1.20, 95% CI = 0.79–1.82). Limitations: Since this was a cross-sectional study, the cause-and-effect relationships between the associated variables cannot be inferred. Conclusions: The interaction effect that exists between nap time and depressive symptoms in older people is important for the identification and early intervention of people at risk for SMI. Ginseng, the root of Panax ginseng C.A. Meyer, has been used as a tonic to enhance bodily functions against various ailments for hundreds of years in Far Eastern countries without apparent adverse effects. Ginsenoside Rb1, one of the most active ingredients of ginseng, has been shown to possess various pharmacological activities. Here we report that Rb1 exhibits potent neuroprotective effects against oxidative injury induced by tert-butylhydroperoxide (t-BHP). Lactate dehydrogenase (LDH) assay demonstrated that incubation with 300 μm t-BHP for 2.5 h led to a significant cell loss of cultured rat embryonic cortex-derived neural progenitor cells (NPCs) and the cell viability was pronouncedly increased by 24 h pretreatment of 10 μm Rb1. TUNEL staining further confirmed that pretreatment of Rb1 significantly reduced the cell apoptosis in t-BHP-induced oxidative injury. Real time PCR revealed that pretreatment with Rb1 activated Nrf2 pathway in cultured NPCs and led to an elevated expression of HO-1. R4375 --> 698.12 --> Background: A number of preclinical advances have indicated the cellular and molecular mechanisms underlying renal fibrosis, such as the signaling of angiotensin II (Ang II), which is a bioactive peptide in renin‐angiotensin system. As one of kidney‐tonifying herbs, Fructus Ligustri Lucidi (FLL), the fruit of Ligustrum lucidum Ait. (Oleaceae) recorded as a tonic and dietary supplement, is a commonly prescribed component in traditional kidney‐tonifying herbal formula. This study aimed to clarify the polysaccharide component in FLL and investigate its potential role in preservation against renal fibrosis. Methods: The crude polysaccharide of FLL was prepared and deproteinized by Sevag reagent. After lyophilization, the DEAE‐cellulose DE52 column and the Sephacryl S‐200 HR column were used for further isolation and purification of homogenous polysaccharide. The mice with unilateral ureteral obstruction (UUO) underwent daily intraperitoneal injection with FLL polysaccharide starting 7 days before UUO surgery until day 7 after surgery. The murine mesangial SV40‐MES13 cells were treated with vehicle, Ang II (10‐7 M) either with the absence and presence of angiotensin type I receptor blocker (ARB) (10‐6M), or with low dose (50 μg/mL, Poly‐L) and high dose (200 μg/mL, Poly‐H) of FLL polysaccharide. Results: The purified homogenous polysaccharide (Poly) was explored from FLL and the carbohydrate content was 97.45%. A hypothetical structure for Poly was proposed considering the comprehensive data of methylation, monosaccharides, and NMR analyses. Treatment of UUO mice with Poly decreased kidney index and ameliorated glomerular sclerosis and interstitial fibrosis, as well as down‐regulated the expression of profibrogenic factor VEGF and matrix fibrotic protein fibronectin (FN). Furthermore, as the same effect to ARB, the pre‐incubation with Poly alleviated the Ang II‐induced up‐regulation of FN in mesangial cells by immunofluoresence staining. The protein expressions of FN and proinflammatory factors like MCP‐1 and Rantes were significantly lower in Poly‐treated cell group than those in Ang II treatment group. Conclusion: The polysaccharide of Ligustrum lucidum Ait. was accurately explored with structure clarification. Background: Human immunodeficiency virus (HIV-1) preferentially selects tRNALys,3 as the primer for reverse transcription. HIV-1 can be forced to select alternative tRNAs through mutation in the primer-binding site (PBS) and a region upstream of the PBS designated as the A-loop. Alteration of the PBS and A-loop to be complementary to the 3′ terminal nucleotides and anticodon of tRNAHis results in HIV-1 that can stably utilize this tRNA for replication. Results: In the current study, we have investigated the effect that mutations within the A-loop have on the stability of HIV-1 with a PBS complementary to tRNAHis. For these studies, we have altered the A-loop to be complementary to tRNAMet, tRNAGln, tRNAIle, tRNAThr and tRNASer. All substitutions of the A-loops with the PBS complementary to tRNAHis resulted in a reduction of infectious virus obtained following transfection of proviral genomes in the 293T cells. Virus replication in SupT1 cells was also impaired as a result of the alteration of the A-loop. Viruses with the A-loop complementary to tRNALys,3 and tRNASer reverted to utilize tRNALys,3 following in vitro replication. In contrast, viruses with the A-loop complementary to the other tRNAs remained stable and continued to use tRNAHis. RNA modeling of the stem-loop structure revealed that nucleotides were displayed on the loop region that could potentially interact with the anticodon of tRNAHis. To further explore the effects of the A-loop mutations on virus replication, the A-loops complementary to tRNASer or tRNAHis were cloned into the wild type genome with the PBS complementary to tRNALys,3. Transfection of proviral genomes which contained the wild type PBS and A-loops complementary to tRNASer or tRNAHis into 293 T cells did not impact on the production of viruses as measured by p24 antigen ELISA. However, viruses with the A-loop complementary to tRNAHis had greatly reduced infectivity and replicated poorly in SupT1 compared to the wild type or viruses with the A-loop complementary to tRNASer. Conclusion: These studies demonstrate that complementarity of A-loop region with the anticodon of tRNAHis has a pronounced effect on the capacity of HIV-1 to utilize tRNAHis as the primer for reverse transcription. Bitter acids (BA) are main component of Humulus lupulus L. (hops). They are known for beer brewing and have various biological and pharmacological properties, especially the bone-protective effect confirmed by our previous in vivo study. Here we aimed to elucidate the anti-senior osteoporosis (SOP) effect of BA on osteoblasts and explore its underlying mechanism. In vitro SOP model was established by D-galactose (D-gal) injured osteoblasts, and the bone formation markers and apoptosis level were measured. mCherry-EGFP-LC3 adenovirus infection and autophagic markers including beclin1 and LC3 proteins were detected to investigate the autophagy level in osteoblasts. To further verify whether BA play the bone-protective role through regulating autophagy, the autophagy inhibitor 3-MA was used, and the cell proliferation, ALP activity, bone mineralization, apoptosis rate and SA-b-gal staining areas were measured. Finally, the protein expressions of AKT/mTOR signaling pathway were detected by Western blotting, and AKT agonist SC79 and mTOR agonist MHY1485 were used to further study the mechanism of BA on AKT/mTOR-mediated autophagy. The results showed that BA stimulated osteoblastic differentiation and inhibited apoptosis proteins Bcl-2/Bax in D-gal-treated osteoblasts. BA also increased the expression of autophagic markers beclin1 and LC3-II/LC3-I in D-galtreated osteoblasts. mCherry-EGFP-LC3 autophagic double fluorescent adenovirus showed BA promoted the generation of autolysosomes and autophagosomes in D-gal-injured osteoblasts, indicating that BA might prevent osteoblastic bone loss through activating autophagy. Autophagy inhibitor 3-MA was used to further verify whether BA played the boneprotective role via regulating autophagy. The results revealed the promotion effects of BA on proliferation, ALP activity, and mineralized nodule formation in D-gal-injured osteoblasts were eliminated after autophagy blocking with 3-MA, and the inhibitory effects of BA on apoptosis rate and SA-β-gal staining areas were also eliminated. Moreover, BA reduced the phosphorylation levels of AKT, mTOR, p70S6K, and 4EBP in AKT/mTOR pathway, and the promotion of BA on the autophagic markers was blocked after the activation of AKT and mTOR by SC79 and MHY1485. In conclusion, it was the first time to demonstrate that BA improved cell activities and bone formation in aging osteoblasts, and revealed the mechanism of BA against SOP in osteoblasts was activating AKT/mTOR-mediated autophagy. HIV-1 packages two copies of RNA into one particle, and the dimerization initiation signal (DIS) in the viral RNA plays an important role in selecting the copackaged RNA partner. We analyzed the DIS sequences of the circulating HIV-1 isolates in the GenBank database and observed that, in addition to the prevalent GCGCGC, GTGCAC, and GTGCGC sequences, there are many other minor variants. To better understand the requirements for the DIS to carry out its function, we generated a plasmid library containing a subtype B HIV-1 genome with a randomized DIS, infected cells with viruses derived from the library, and monitored the emergence of variants at different time points until 100 days postinfection. We observed rapid loss of viral diversity and found that the selected variants contained palindromes in the DIS. The "wild-type" GCGCGC-containing virus was a major variant, whereas GTGCAC- and GTGCGC-containing viruses were present at low frequencies. Additionally, other 6-nucleotide (nt) palindromic sequences were selected; a major category of the selected variants contained two GC dyads in the center of the palindrome, flanked by a non-GC dyad. Surprisingly, variants with GC-rich 4-nt palindromes were sustained throughout the selection period at significant frequencies (~12 to 38%); of these, variants containing the CGCGC sequence were observed frequently, suggesting that this sequence has a selection advantage. These results revealed that multiple sequences can fulfill the function of the HIV-1 DIS. A common feature of the selected DIS sequence is a 4- or 6-nt GC-rich palindrome, although not all sequences with these characteristics were selected, suggesting the presence of other unidentified interactions. Xanthohumol, a natural isoflavone from Humulus lupulus L., possesses biological activities. However, the biological fate of xanthohumol in vivo remains unclear. The aim of this study was to investigate the absorption and metabolism of xanthohumol in rats through UPLC‐MS/MS. The plasma, urine and fecal samples were collected after oral administration of xanthohumol (25, 50, 100 mg/kg) in SD rats. The contents of xanthohumol and its metabolites were determined by UPLC‐MS/MS. A total of 6 metabolites of xanthohumol were identified in rats, including methylated, glucuronidated, acid‐catalyzed cyclization and oxidation, indicating xanthohumol underwent phase I and II metabolism. Besides, isoxanthohumol was the major metabolites of xanthohumol. Xanthohumol was rapidly absorbed, metabolized, and eliminated in rats. The pharmacokinetics results showed the Tmax of xanthohumol and isoxanthohumol were 3 and 2.33 h, respectively. The AUC0−t of xanthohumol and isoxanthohumol were 138.83 ± 6.03 and 38.77 ± 4.46 ng/ml·h, respectively. Furthermore, xanthohumol was mainly excreted in the form of prototype through feces and a small amount of xanthohumol was excreted through urine. We previously reported that shear stress induces phosphorylation and disassembly of keratin intermediate filaments (IFs). Shear stress also induces a time- and strain-dependent degradation of keratin IFs, and the current study examines the mechanisms involved in degradation of keratin proteins in human A549 cells exposed to 0-24 h of shear stress (7.5-30 dynes/cm²). Ubiquitin was found to be covalently associated with keratin proteins immunoprecipitated from shear-stressed cells, and pretreatment with the proteasomal inhibitor MG132 prevented the degradation of the keratin IF network. Importantly, phosphorylation of K8 Ser-73 is required for the shear stress-mediated ubiquitination, disassembly, and degradation of the keratin IF network. Immunofluorescence microscopy revealed that shear stress caused the thin array of keratin fibrils observed in control cells to be reorganized into a perinuclear aggregate, known as an aggresome, and that ubiquitin was also associated with this structure. Finally, the E2 enzymes, UbcH5b, -c, and Ubc3, but not E2-25K are required for the shear stress-mediated ubiquitin-proteasomal degradation of keratin proteins. Background: Previous studies have shown that infection with human immunodeficiency virus type 1 (HIV-1) causes acceleration of the synthesis of glutamine tRNA (tRNAGln) in infected cells. To investigate whether this might influence HIV-1 to utilize tRNAGln as a primer for initiation of reverse transcription, we have constructed HIV-1 proviral genomes in which the PBS and the Aloop region upstream of the PBS have been made complementary to either the anticodon region of tRNAGln,1 or tRNAGln,3 and 3' terminal 18 nucleotides of each isoacceptor of tRNAGln. Results: Viruses in which the PBS was altered to be complementary to tRNAGln,1 or tRNAGln,3 with or without the A-loop all exhibited a lower infectivity than the wild type virus. Viruses with only the PBS complementary to tRNAGln,1 or tRNAGln,3 reverted to wild type following culture in SupT1 cells. Surprisingly, viruses in which the PBS and A-loop were complementary to tRNAGln,1 did not grow in SupT1 cells, while viruses in which the PBS and A-loop were made complementary to tRNAGln,3 grew slowly in SupT1 cells. Analysis of the PBS of this virus revealed that it had reverted to select tRNAThr as the primer, which shares complementarity in 15 of 18 nucleotides with the PBS complementary to tRNAGln,3. Objectives Xanthohumol (XAN) is a unique component of Humulus lupulus L. and is known for its diverse biological activities. In this study, we investigated whether Xanthohumol could ameliorate memory impairment of APP/PS1 mice, and explored its potential mechanism of action. Methods APP/PS1 mice were used for in vivo test and were treated with N-acetylcysteine and Xanthohumol for 2 months. Learning and memory levels were evaluated by the Morris water maze. Inflammatory and oxidative markers in serum and hippocampus and the deposition of Aß in the hippocampus were determined. Moreover, the expression of autophagy and apoptosis proteins was also evaluated by western blot. Key findings Xanthohumol significantly reduced the latency and increased the residence time of mice in the target quadrant. Additionally, Xanthohumol increased superoxide dismutase level and reduced Interleukin-6 and Interleukin-1ß levels both in serum and hippocampus. Xanthohumol also significantly reduced Aß deposition in the hippocampus and activated autophagy and anti-apoptotic signals. Conclusions Xanthohumol effectively ameliorates memory impairment of APP/PS1 mice by activating mTOR/LC3? and Bax/Bcl-2 signalling pathways, which provides new insight into the neuroprotective effects of Xanthohumol. Fructus Ligustri Lucidi (FLL), the fruit of Ligustrum lucidum Ait., is a traditional Chinese medicine that has been used for tonifying the kidney and liver for decades. This study aimed to explore and identify polysaccharides from FLL and elucidate its protective effect against renal fibrosis. Polysaccharides were extracted and isolated from FLL. The purified fraction was identified by serial phytochemical work, such as gel-permeation chromatography, ion chromatography, gas chromatography-mass spectrometry, and nuclear magnetic resonance. Mice with unilateral ureteral obstruction (UUO) were applied as a renal fibrosis model. The male UUO mice were pretreated with heteropolysaccharide (Poly) 1 week prior to surgery and continuously treated for 7 days after the operation. Renal fibrosis was assessed by Periodic Acid-Schiff (PAS) staining and Masson's trichrome staining in paraffin-embedded slides. The murine mesangial cells SV40-MES13 upon angiotensin II (Ang II) treatment were developed as an in vitro fibrotic model. The cells were treated by Poly in the presence of Ang II. Molecular expression was detected by RT-PCR, immunoblotting, and immunofluorescence staining. We identified a heteropolysaccharide composed of arabinose and galactose (molar ratio, 0.73:0.27) with a predicted chemical structure characterized by a backbone composed of 1,5-α-Ara f , 1,3,5-α-Ara f , 1,6-α-Gal p , and 1,3,6-β-Gal p and side chains comprised of T-α-Ara f , T-α-Ara p , and 1,3-α-Ara f. Pretreatment of UUO mice with Poly effectively alleviated glomerulosclerosis and tubulointerstitial fibrosis. Moreover, Poly pretreatment down-regulated the expression of extracellular matrix (ECM) protein fibronectin (FN), profibrotic factor VEGF, proinflammatory cytokines MCP-1 and Rantes in the obstructed kidney. Similarly, the incubation of SV40-MES13 cells with Poly significantly inhibited Ang II-induced elevation in accumulation and expression level of FN and attenuated Ang II-evoked up-regulation in protein expression of MCP-1 and Rantes. Our study isolated and identified a naturally occurring heteropolysaccharide in FLL and revealed its potential in protecting the kidneys from fibrosis. Background: The rapid determination of pathogenic agent is very important to clinician for guiding their clinical medication. However, current diagnostic methods are of limitation in many aspects, such as detecting range, time-consuming, specificity and sensitivity. In this report, we apply our new-developing pathogen detection method to clarify that Propionibacterium acnes is the causative agent of a two-year-old boy with juvenile myelomonocytic leukemia presenting clinical symptoms including serious rash and hyperpyrexia while traditional clinical methods of diagnosis fail to detect the pathogenic agent and multiple antimicrobial drugs are almost ineffective Propionibacterium acnes is confirmed to be the infectious agent by quantitative real-time polymerase chain reaction.Case Presentation: After haploidentical hematopoietic stem cell transplantation, a two-year-old boy with juvenile myelomonocytic leukemia presented to a pediatrist in a medical facility with hyperpyrexia and red skin rash which later changed to black skin rash all over his body. Traditional diagnostic assays were unrevealing, and several routine antimicrobial treatments were ineffective, including the vancomycin, meropenem, tobramycin, cefepime and rifampin. In this case, pediatrist resorted to the next-generation sequencing technology for uncovering potential pathogens so as to direct their use of specific drugs against pathogenic bacteria. Therefore, based on the BGISEQ100 (Ion Proton System) which performed sequencing-by-synthesis, with electrochemical detection of synthesis, and each such reaction coupled to its own sensor, which are in turn organized into a massively parallel sensor array on a complementary metal-oxidesemiconductor chip, we detect and identify the potential pathogens. As a result, we detected a significantly higher abundance of skin bacteria Propionibacterium acnes in patient's blood than controls. It had been reported that patients infected by Propionibacterium acnes almost always had history of immunodeficiency, trauma or surgery. Considering this possible cause, antimicrobial treatment was adjusted to target this rare opportunistic pathogen. Fever and black skin rashes were rapidly reduced after administrating specific drugs against Propionibacterium acnes.Conclusion: This case showed our new-developing pathogen detection method was a powerful tool in assisting clinical diagnosis and treatment. The Permian mafic dyke swarms are widely distributed in the Tarim Block, East Tianshan and the western Junggar Block in northwestern China. These mafic dyke swarms, together with coeval basalts, mafic-ultramafic intrusions and A-type granitic plutons, constitute the Permian Tarim large igneous province (LIP), which is considered to be mantle plume-derived. In this study, multi-source high-resolution remote sensing images are used to identify the geometry and emplacement mode of the mafic dykes, sills and flood basalts in Keping and its adjacent areas including Bachu (northwestern Tarim Block), Beishan and Kuluketag (northeastern Tarim Block) and Kelamyi (western Junggar Block). Structural analysis and paleomagnetic results are used to restore the primary geometry of the mafic dykes. The results indicate that the lengths and thicknesses among a total of 117 extracted mafic dykes in Keping follow negative exponent size distributions. The mafic dykes have a mean thickness of 3.8 m with a maximum of 21.4 m. The length of unconnected mafic dyke segments ranges from 0.127 km to 17.1 km, with an arithmetic length of 1.8 km. The restoration of geometry of the mafic dykes indicates that the Permian mafic dyke swarms in Keping may have extended as far as 61-69 km along a primary orientation of about N320°W, and the flood basalts of the Tarim LIP may have extended to Keping so that the areal extent of the Tarim LIP could be enlarged for about 12,000 km² compared with that originally reported (about 250,000 km²). The geometric features of the mafic dyke swarms in Keping and its adjacent areas are different from those of the giant radiating dyke swarms due to radial fractures associated with domal uplift. The mafic dyke swarms and sills in the Tarim LIP make up the plumbing system of the mantle plume. Accompanied by the upwelling of mantle plume, many eruptive centers and regional dykes are generated in Beishan, East Tianshan and Tarim basin. The flood basalts in Keping are fissure-type, which may have been fed by magma conduits from the plume center; whereas in Bachu, the central volcanic eruption is dominant. [ABSTRACT FROM AUTHOR]

Abstract: A series of acidic task-specific ionic liquids 1-R2-2-R1benzimidazolium tetrafluoroborate (R1 =Me, R2 =Me, Et, Pr-n, Bu-n, Pen-n) were prepared by simple acid-base neutralization of the corresponding 1-alkylbenzimidazole and tetrafluoroboric acid. The compounds were characterized by FTIR spectra, elemental analysis, 1HNMR spectra and thermogravimetric analysis. These new ionic liquids are nonvolatile, and have potential use as alternatives to conventional organic solvents due to their solubility and thermal stability. These novel tetrafluoroborate salts show good catalytic activity to esterification of carboxylic acids with alcohols under mild reaction conditions, which could maintain good catalytic performance after recycling at least six times. Furthermore, a crystal of one compound, [H-bmBim]BF4, was prepared, with the crystal structure determined by X-ray diffraction analysis. The molecular structure is formed by weak π-π interactions and intermolecular hydrogen bonds between the benzimidazole rings, yielding a three-dimensional net-like supramolecule. A long-standing question in retrovirus biology is how RNA genomes are distributed among virions. In the studies presented in this report, we addressed this issue by directly examining HIV-1 RNAs in virions using a modified HIV-1 genome that contained recognition sites for BglG, an antitermination protein in the Escherichia coli bgl operon, which was coexpressed with a fragment of BglG RNA binding protein fused to a fluorescent protein. Our results demonstrate that the majority of virions (>90%) contain viral RNAs. We also coexpressed HIV-1 genomes containing binding sites for BglG or the bacteriophage MS2 coat protein along with 2 fluorescent protein-tagged RNA binding proteins. This method allows simultaneously labeling and discrimination of 2 different RNAs at single-RNA-detection sensitivity. Using this strategy, we obtained physical evidence that virions contain RNAs derived from different parental viruses (heterozygous virion) at ratios expected from a random distribution, and we found that this ratio can be altered by changing the dimerization sequences. Our studies of heterozygous virions also support a generally accepted but unproven assumption that most particles contain 1 dimer. This study provides answers to long-standing questions in HIV-1 biology and illustrates the power and sensitivity of the 2-RNA labeling method, which can also be adapted to analyze various issues of RNA biogenesis including the detection of different RNAs in live cell imaging. While extensive research has been carried out in the field of structural polymer-based nanocomposites much less investigations have been concerned with polymer nanocomposites for functional applications. Among the functional nanomaterials, nanocomposites consisting of metal nanoparticles dispersed in a dielectric matrix are of particular interest due to their novel functional properties offering hosts of new applications. Here, polymers are attractive as matrix, and several approaches have been reported to incorporate metal nanoparticles into polymers. The present review is concerned with the preparation of polymer-based nanocomposites by vapor phase co-and tandem deposition and the resulting functional properties. The techniques involve evaporation and sputtering, respectively, of metallic and organic components and inter alia allow the preparation of composites which contain alloy clusters of well defined composition. Emphasis is placed on soft-magnetic high frequency materials with cut-off frequencies well above 1 GHz and on optical composites with tuned plasmon resonances suitable for ultra thin color filters, Bragg reflectors, and other devices. In addition, antibacterial coatings and sensors for organic vapors are addressed. The latter take advantage of the steep drop of the electrical resistivity at the percolation threshold. First results are also reported on the incorporation of photo-switchable molecules into nanocomposites near the percolation threshold. The article discusses study on infection by and dissemination of New Delhi Metallo-beta-lactamase (NDM)-5-producing Escherichia coli in China. It mentions the study conducted on four patients in Chinese cities revealed their resistant to most β-lactams, except for aztreonam. It adds the spread of NDM-5 across five cities in China highlight the need for surveillance and to develop appropriate interventions to control its dissemination.

Oxidative stress is a crucial pathogenic factor in osteoporosis. Autophagy is a cellular self-digestion process that can selectively remove damaged organelles under oxidative stress, and thus presents a potential therapeutic target against osteoporosis. Monotropein is an iridoid glycoside which can increase osteoblastic bone formation and be applied for medicinal purpose in China. The aim of this work is to investigate whether autophagy participates the protection effects of monotropein in osteoblasts under oxidative stress and the possible mechanism of such involvement. Here, monotropein was capable of inhibiting the H 2 O 2 -induced reactive oxygen species generation in osteoblasts. Monotropein induced autophagy and protected osteoblasts from cytotoxic effects of H 2 O 2 , as assessed by viability assays, apoptosis and western blotting. Moreover, it significantly attenuated H 2 O 2 -evoked oxidative stress as measured by malondialdehyde, catalase, and superoxide dismutase levels. Importantly, monotropein reduced the phosphorylation of protein kinase B (Akt), mammalian target of rapamycin (mTOR) and its two downstream proteins (p70S6K and 4EBP1). The autophagy level increased in osteoblasts treated with monotropein as represented by an increased in both Beclin1 expression and the LC3-II/LC3-I ratio. However, the Akt activator (SC79) and mTOR activator (MHY1485) suppressed the autophagy level induced by monotropein in H 2 O 2 -treated cells. Consequently, the antioxidant effects of monotropein were mediated, at least in part, by enhancing autophagy through the Akt/mTOR pathway.
