Wheat powdery mildew, which is mainly caused by Blumeria graminis f. sp. tritici ( Bgt ), seriously damages wheat production. The wheat- Thinopyrum intermedium alien addition disomic line germplasm SN6306, being one of the important sources of genes for wheat resistance, is highly resistant to Bgt E09 and to many other powdery mildew physiological races. However, knowledge on the resistance mechanism of SN6306 remains limited. Our study employed high-throughput RNA sequencing based on next-generation sequencing technology (Illumina) to obtain an overview of the transcriptome characteristics of SN6306 and its parent wheat Yannong 15 (YN15) during Bgt infection. The sequencing generated 104,773 unigenes, 9909 of which showed varied expression levels. Among the 9909 unigenes, 1678 unigenes showed 0 reads in YN15. The expression levels in Bgt -inoculated SN6306 and YN15 of exactly 39 unigenes that showed 0 or considerably low reads in YN15 were validated to identify the genes involved in Bgt resistance. Among the 39 unigenes, 12 unigenes were upregulated in SN6306 by 3–45 times. These unigenes mainly encoded kinase, synthase, proteases, and signal transduction proteins, which may play an important role in the resistance against Bgt . To confirm whether the unigenes that showed 0 reads in YN15 are really unique to SN6306, 8 unigenes were cloned and sequenced. Results showed that the selected unigenes are more similar to SN6306 and Th. intermedium than to the wheat cultivar YN15. The sequencing results further confirmed that the unigenes showing 0 reads in YN15 are unique to SN6306 and are most likely derived from Th. intermedium (Host) Nevski. Wheat powdery mildew, caused by the fungal pathogen Blumeria graminis f. sp. tritici ( Bgt), is a serious wheat disease both in China and worldwide. Finding and cloning the key resistance genes to Bgt are important for wheat breeding. In this study, 23 differentially expressed genes were isolated by GeneFishing from the hybrid wheat alien disomic addition line germplasm SN6306, which is highly resistant to powdery mildew. SN6306 originated from the intergeneric hybridization between wheat Yannong 15 and Elytrigia intermedium (Host) Nevski. Among the 23 genes, a gene encoding for auxin-repressed protein ( TaARP1) was selected for further study, and its full-length complementary DNA (cDNA) and DNA sequences were obtained. The function of TaARP1 was investigated by real-time quantitative reverse transcription PCR and Barley Stripe Mosaic Virus-mediated gene silencing. Results showed that, after inoculation with Bgt, the expression of TaARP1 was upregulated more than ten times compared with non-inoculated controls. Ten days after inoculation with powdery mildew, BSMV: TaARP1 leaves clearly showed higher susceptibility to mildew infections than BSMV:00 leaves. Powdery mildew colonies and mycelia development were analyzed by microscopic observation. These data suggested that TaARP1 may be involved in the response to Bgt and may be a potential resistance gene to wheat powdery mildew. [ABSTRACT FROM AUTHOR]

Background: Rodents, such as mice, are vulnerable targets, and potential intermediate hosts, of SARS-CoV-2 variants of concern, including Alpha, Beta, Gamma, and Omicron. N501Y in the receptor-binding domain (RBD) of Spike protein is the key mutation dictating the mouse infectivity, on which the neighboring mutations within RBD have profound impacts. However, the impacts of mutations outside RBD on N501Y-mediated mouse infectivity remain to be explored. Results: Herein, we report that two non-RBD mutations derived from mouse-adapted strain, Ins215KLRS in the N-terminal domain (NTD) and H655Y in the subdomain linking S1 to S2, enhance mouse infectivity in the presence of N501Y mutation, either alone or together. This is associated with increased interaction of Spike with mouse ACE2 and mutations-induced local conformation changes in Spike protein. Mechanistically, the H655Y mutation disrupts interaction with N657, resulting in a less tight loop that wraps the furin-cleavage finger; and the insertion of 215KLRS in NTD increases its intramolecular interaction with a peptide chain that interfaced with the RBD-proximal region of the neighboring protomer, leading to a more flexible RBD that facilitates receptor binding. Moreover, the Omicron Spike that contains Ins214EPE and H655Y mutations confer mouse infectivity > 50 times over the N501Y mutant, which could be effectively suppressed by mutating them back to wild type. Conclusions: Collectively, our study sheds light on the cooperation between distant Spike mutations in promoting virus infectivity, which may undermine the high infectiousness of Omicron variants towards mice. YUCCA protein participates in a key rate-limiting step in the tryptophan-dependent pathway for auxin biosynthesis and is involved in numerous processes during plant development. In this study, the genomic and cDNA sequences of three TaYUC10 homoeologous genes were isolated. These sequences showed a very high conservation in coding region and the exon/intron structure, whereas their intron lengths were different. The cDNA and polypeptide chains of the three TaYUC10 genes were highly similar. These genes were most homologous to BdYUC10 . Location analysis showed that TaYUC10.1 was present in chromosome 5BL. TaYUC10.3 was expressed in all parts of the wheat, but was predominant in the reproductive organs of mature wheat, such as flowering spikelets or fertilized embryos. In the fertilized embryos 28 d post-anthesis, expression of TaYUC10.3 was clearly increased with the development of seeds. This indicates that TaYUC genes may play a vital role in seed development. TaYUC10.3 overexpressed in Arabidopsis had a typical phenotype, excessive auxin accumulation also seen in higher plants, and showed increased spacing of silique and downward curling of the blade margin. Sterility was observed in adult transgenic plants, becoming more severe in late development. The floral structures of sterile plants were not integrated. Summary: The biotrophic fungal pathogen Blumeria graminis f. sp. tritici (Bgt) is a crucial factor causing reduction in global wheat production. Wild wheat relatives, for example Thinopyrum intermedium, is one of the wild‐used parents in wheat disease‐resistant breeding. From T. intermedium line, we identified the aspartic protease gene, TiAP1, which is involved in resistance against Bgt. TiAP1 is a secreted protein that accumulates in large amounts at the infection sites of Bgt and extends to the intercellular space. Yeast two‐hybrid, luciferase complementation imaging and bimolecular florescent complimentary analysis showed that TiAP1 interacted with the chitin deacetylase (BgtCDA1) of Bgt. The yeast expression, purification and in vitro test confirmed the chitin deacetylase activity of BgtCDA1. The bombardment and VIGS‐mediated host‐induced gene silencing showed that BgtCDA1 promotes the invasion of Bgt. Transcriptome analysis showed the cell wall xylan metabolism, lignin biosynthesis‐related and defence genes involved in the signal transduction were up‐regulated in the transgenic TiAP1 wheat induced by Bgt. The TiAP1 in wheat may inactivate the deacetylation function of BgtCDA1, cause chitin oligomers expose to wheat chitin receptor, then trigger the wheat immune response to inhibit the growth and penetration of Bgt, and thereby enhance the resistance of wheat to pathogens. Cell-in-cell structure is prevalent in human cancer, and associated with several specific pathophysiological phenomena. Although cell membrane adhesion molecules were found critical for cell-in-cell formation, the roles of other membrane components, such as lipids, remain to be explored. In this study, we attempted to investigate the effects of cholesterol and phospholipids on the formation of cell-in-cell structures by utilizing liposome as a vector. We found that Lipofectamine-2000, the reagent commonly used for routine transfection, could significantly reduce entotic cell-in-cell formation in a cell-specific manner, which is correlated with suppressed actomyosin contraction as indicated by reduced β-actin expression and myosin light chain phosphorylation. The influence on cell-in-cell formation was likely dictated by specific liposome components as some liposomes affected cell-in-cell formation while some others didn't. Screening on a limited number of lipids, the major components of liposome, identified phosphatidylethanolamine (PE), stearamide (SA), lysophosphatidic acid (LPA) and cholesterol (CHOL) as the inhibitors of cell-in-cell formation. Importantly, cholesterol treatment significantly inhibited myosin light chain phosphorylation, which resembles the effect of Lipofectamine-2000, suggesting cholesterol might be partially responsible for liposomes' effects on cell-in-cell formation. Together, our findings supporting a role of membrane lipids and cholesterol in cell-in-cell formation probably via regulating actomyosin contraction. [ABSTRACT FROM AUTHOR]
