The role of Wnt signaling in neural crest fate was examined using RCAS retroviral infections to target gene expression to neural crest cells from transgenic mice (DCT-TVA) that express the virus receptor, TVA, under the control of a melanoblast promoter. While 90% of DCT-TVA+ cells expressed early melanoblast markers, only 5% of descendents were melanocytes while the majority were smooth muscle cells. The fate of DCT-TVA+ descendents can be biased to melanocytes by inducing the canonical WNT signaling pathway through intrinsic over-expression of _CAT or WNT3a or by treatment with soluble WNT3a. WNT1 acts in a distinct mode of action to expand melanocytes by paracrine signaling on melanoblast precursors, not by expansion of DCT-TVA+ cells or biasing DCT-TVA+ cells to melanocytes.
