Dramatic changes in the sea ice characteristics in the Barents Sea have potential consequences for the weather and climate systems of mid-latitude continents, Arctic ecosystems, and fisheries, as well as Arctic maritime navigation. Simulations and projections of winter sea ice in the Barents Sea based on the latest 41 climate models from the Coupled Model Intercomparison Project Phase 6 (CMIP6) are investigated in this study. Results show that most CMIP6 models overestimate winter sea ice in the Barents Sea and underestimate its decreasing trend. The discrepancy is mainly attributed to the simulation bias towards an overly weak ocean heat transport through the Barents Sea Opening and the underestimation of its increasing trend. The methods of observation-based model selection and emergent constraint were used to project future winter sea ice changes in the Barents Sea. Projections indicate that sea ice in the Barents Sea will continue to decline in a warming climate and that a winter ice-free Barents Sea will occur for the first time during 2042–2089 under the Shared Socioeconomic Pathway 585 (SSP5-8.5). Even in the observation-based selected models, the sensitivity of winter sea ice in the Barents Sea to global warming is weaker than observed, indicating that a winter ice-free Barents Sea might occur earlier than projected by the CMIP6 simulations. Climate change in the Arctic has substantial impacts on human life and ecosystems both within and beyond the Arctic. Our analysis of CMIP6 simulations shows that some climate models project much larger Arctic climate change than other models, including changes in sea ice, ocean mixed layer, air‐sea heat flux, and surface air temperature in wintertime. In particular, dramatic enhancement of Arctic Ocean convection down to a few hundred meters is projected in these models but not in others. Interestingly, these models employ the same ocean model family (NEMO) while the choice of models for the atmosphere and sea ice varies. The magnitude of Arctic climate change is proportional to the strength of the increase in poleward ocean heat transport, which is considerably higher in this group of models. Establishing the plausibility of this group of models with high Arctic climate sensitivity to anthropogenic forcing is imperative given the implied ramifications. Plain Language Summary: Arctic climate is projected to change dramatically in the future based on the latest climate models. However, there is a large discrepancy between these climate models. We find that simulated Arctic climate change tends to be much larger in models using the community ocean model "NEMO" as their ocean component. Future Arctic climate change in winter strongly depends on the strength of the increase in poleward ocean heat transport, which is considerably higher in the NEMO‐family climate models. There are some indications that this group of models can better reproduce the observed winter sea ice decline and mixed layer depth in the Barents Sea in their historical simulations. We suggest that investigating the model physics in the NEMO‐family climate models in comparison with other models may ultimately help to reduce the overall uncertainty in climate change projections. Daphne genkwa , as a traditional medicine, is widely distributed in China, Korea and Vietnam. In China, the dried flower buds of this plant are named "Yuanhua". It has the ability to effectively promote urination, eliminate phlegm and alleviate cough, eliminate parasites and cure of scabies, with a broad spectrum of pharmacological effects and considerable clinical efficacy. This paper provides a summary and classification of the main chemical constituents of D. genkwa based on a review of relevant domestic and foreign literature. It also outlines the current research status of traditional clinical usage, pharmacological effects, and toxicity of D. genkwa. Drug target discovery is an essential step to reveal the mechanism of action (MoA) underlying drug therapeutic effects and/or side effects. Most of the approaches are usually labor-intensive while unable to identify the tissue-specific interacting targets, especially the targets with weaker drug binding affinity. In this work, we proposed an integrated pipeline, FL-DTD, to predict the drug interacting targets of novel compounds in a tissue-specific manner. This method was built based on a hypothesis that cells under a status of homeostasis would take responses to drug perturbation by activating feedback loops. Therefore, the drug interacting targets can be predicted by analyzing the network responses after drug perturbation. We evaluated this method using the expression data of estrogen stimulation, gene manipulation and drug perturbation and validated its good performance to identify the annotated drug targets. Using STAT3 as a target protein, we applied this method to drug perturbation data of 500 natural compounds and predicted five compounds with STAT3 interacting activities. Experimental assay validated the STAT3-interacting activities of four compounds. Overall, our evaluation suggests that FL-DTD predicts the drug interacting targets with good accuracy and can be used for drug target discovery. [ABSTRACT FROM AUTHOR]

Conventional photoelectrochemical (PEC) analysis mostly utilizes photoactive material modified planar indium tin oxides (ITOs) to obtain photocurrent responses for the measurement of analytes in solution. In this work, a CdS quantum dot (QD) modified nanopipette was prepared for the PEC analysis of the alkaline phosphatase (ALP) activity in single MCF-7 cells. The nanopipette was filled with ascorbic acid 2-phosphate (AAP) that was egressed outside the nanopipette by electrochemical pumping. Next, AAP was catalyzed by ALP to generate ascorbic acid (AA), which is an efficient electron donor for CdS QDs under illumination. Based on the result that the nanopipette showed a linear photocurrent response to AA, a nearly linear correlation between the photocurrent and the activity of ALP was established. Accordingly, using these CdS QD modified nanopipettes, the ALP activity in single MCF-7 cells was determined to be 0.12 U mL−1 by PEC analysis. Background: Left-right (LR) asymmetry is an essential feature of bilateral animals. Studies in vertebrates show that LR asymmetry formation comprises three major steps: symmetry breaking, asymmetric gene expression, and LR morphogenesis. Although much progress has been made in the first two events, mechanisms underlying asymmetric morphogenesis remain largely unknown due to the complex developmental processes deployed by vertebrate organs. Results: We here addressed this question by studying Pitx gene function in the basal chordate amphioxus whose asymmetric organogenesis, unlike that in vertebrates, occurs essentially in situ and does not rely on cell migration. Pitx null mutation in amphioxus causes loss of all left-sided organs and incomplete ectopic formation of all right-sided organs on the left side, whereas Pitx partial loss-of-function leads to milder phenotypes with only some LR organs lost or ectopically formed. At the N1 to N3 stages, Pitx expression is gradually expanded from the dorsal anterior domain to surrounding regions. This leads to activation of genes like Lhx3 and/or Prop1 and Pit, which are essential for left-side organs, and downregulation of genes like Hex and/or Nkx2.1 and FoxE4, which are required for right-side organs to form ectopically on the left side. In Pitx mutants, the left-side expressed genes are not activated, while the right-side genes fail to decrease expression on the left side. In contrast, in embryos overexpressing Pitx genes, the left-side genes are induced ectopically on the right side, and the right-side genes are inhibited. Several Pitx binding sites are identified in the upstream sequences of the left-side and right-side genes which are essential for activation of the former and repression of the latter by Pitx. Conclusions: Our results demonstrate that (1) Pitx is a major (although not the only) determinant of asymmetric morphogenesis in amphioxus, (2) the development of different LR organs have distinct requirements for Pitx activity, and (3) Pitx controls amphioxus LR morphogenesis probably through inducing left-side organs and inhibiting right-side organs directly. These findings show much more dependence of LR organogenesis on Pitx in amphioxus than in vertebrates. They also provide insight into the molecular developmental mechanism of some vertebrate LR organs like the lungs and atria, since they show a right-isomerism phenotype in Pitx2 knockout mice like right-sided organs in Pitx mutant amphioxus. The molecular profiling of single axons and dendrites in living neurons could provide important information for the better understanding of neuron function. Here, electrosyringe-assisted electrospray mass spectrometry (MS) is established for the first time to achieve intracellular sampling from one axon or dendrite in living neurons for mass spectrometric analysis. The key is the insertion of a ∼130 nm capillary tip into one axon or dendrite to load the cytosol through electro-osmotic flow. The ionization efficiency from the nano-capillary is enhanced to guarantee mass spectrometric analysis of multiple components from the axon and dendrite. Higher levels of pyroglutamic acid and glutamic acid are revealed in the axon compared to in the body and dendrite. This uneven distribution is in accordance with the accumulation of neurotransmitters in the axon for information delivery. The achievement of electrosyringe-assisted electrospray MS is to unveil the molecular distribution in the whole living neuron, which offers the feasibility to deeply investigate molecular communication between the axon/dendrite and the body inside neurons. Ononin (ON) is an isoflavone with numerous reported bioactivities, including anti-oxidative, anti-inflammatory and neuroprotective effects. Autophagy is a critical homeostatic process in the body that has been reported to closely associate with the apoptotic processes of cardiomyocytes. Using flow cytometry, western blotting, echocardiography and Masson's staining, the present study investigated the effects of ON on H2O2-induced cardiomyocyte apoptosis and myocardial infarction, in addition to any potential underlying molecular mechanisms. H2O2 treatment reliably induced apoptosis in H9C2 cells. The anti-apoptotic effects of ON were revealed by flow cytometry results and by the downregulation of cleaved-caspase 3. Further investigations indicated that ON may alleviate apoptosis by enhancing autophagy, as evidenced by increased microtubule-associated proteins 1A/1B light chain 3B expression and p62 degradation. Activation of the 5' AMP-activated protein kinase (AMPK)/mTOR pathway was observed after ON administration following H2O2-induced cardiomyocyte injury. However, these anti-apoptotic effects mediated by ON were lost after autophagy inhibition by chloroquine or AMPK inhibition by Compound C. Finally, the protective effects of ON on cardiomyocytes in vitro could also be observed in vivo. A myocardial infarction model was established by ligating the left anterior descending branch of the rat heart. Using echocardiography and Masson's staining, ON was shown to increase the ejection fraction and decrease cardiac fibrosis in rats with myocardial infarction. The hydrazine oxidation‐assisted H2 evolution method promises low‐input and input‐free hydrogen production. However, developing high‐performance catalysts for hydrazine oxidation (HzOR) and hydrogen evolution (HER) is challenging. Here, we introduce a bifunctional electrocatalyst α‐MoC/N−C/RuNSA, merging ruthenium (Ru) nanoclusters (NCs) and single atoms (SA) into cubic α‐MoC nanoparticles‐decorated N‐doped carbon (α‐MoC/N−C) nanowires, through electrodeposition. The composite showcases exceptional activity for both HzOR and HER, requiring −80 mV and −9 mV respectively to reach 10 mA cm−2. Theoretical and experimental insights confirm the importance of two Ru species for bifunctionality: NCs enhance the conductivity, and its coexistence with SA balances the H ad/desorption for HER and facilitates the initial dehydrogenation during the HzOR. In the overall hydrazine splitting (OHzS) system, α‐MoC/N−C/RuNSA excels as both anode and cathode materials, achieving 10 mA cm−2 at just 64 mV. The zinc hydrazine (Zn−Hz) battery assembled with α‐MoC/N−C/RuNSA cathode and Zn foil anode can exhibit 97.3 % energy efficiency, as well as temporary separation of hydrogen gas during the discharge process. Therefore, integrating Zn−Hz with OHzS system enables self‐powered H2 evolution, even in hydrazine sewage. Due to high efficiency of click chemistry, a new nanoelectrode‐based pipette architecture with dibenzocyclooctyne at the tip is designed to realize fast conjugation with azide group‐containing triphenylphosphine, which targets mitochondrial membranes. The covalent binding of one mitochondrion at the tip of the nanopipette allows a small region of the membrane to be isolated on the Pt surface inside the nanopipette. Therefore, the release of reactive oxygen species (ROS) from the mitochondrion is monitored, which is not interfered by the species present in the cytosol. The dynamic tracking of ROS release from one mitochondrion reveals the distinctive "ROS‐induced ROS release" within the mitochondria. Further study of RSL3‐induced ferroptosis using nanopipettes provides direct evidence for supporting the noninvolvement of glutathione peroxidase 4 in the mitochondria during RSL3‐induced ROS generation, which has not previously been observed at the single‐mitochondrion level. • The dynamic fracture development of shale are described, and the geometric parameters, topological and fractal characteristics of the fracture network are evaluated. • The dissipation law of rock failure energy is investigated, which reflects the essential characteristics of rock failure under impact load. • Combined with section scanning, stress simulation and energy consumption calculation, the dynamic load rock breaking mechanism is systematically analyzed. Shale fracture and energy dissipation are the basis of reservoir fracturing effect evaluation. The damage and fracture mechanism of shale under dynamic impact load is the key scientific concern of reservoir fracturing. To study the law of fracture, fracture propagation and energy dissipation of shale under dynamic impact, dynamic tests of water-based impact load under different impact velocities were carried out using impact rock breaking test platform based on air cannon. Topological and fractal theory, Weibull distribution model, space vector geometry method and Griffith fracture mechanics theory were used to systematically study the effects of impact velocity on fracture development and expansion, fragment mass and size distribution, damage degree of abutment residual and energy dissipation transformation. Scanning electron microscopy (SEM) was used to observe the micromorphology of the impact fracture surface. The results demonstrated that an increase in impact velocity led to the formation of a network fracture featuring main fractures, cavity fissure, bedding fractures, and crushing zones, thereby enhancing the fracture's capability to penetrate bedding. The apparent fracture network connectivity, topology structure and fractal dimension were positively correlated with impact velocity. The residual height of the shale decreased, resulting in an escalation of the damage degree from mild (0.43) to severe (0.75), while the average penetration angle decreases from 81.00° to 63.52°. The area of new surface experiences rapid initial growth followed by a more gradual increase, with dissipative energy showing a more pronounced increase compared to crushing energy. The micro-fracture mode underwent an evolution from brittleness to toughness and a transition from low energy consumption to high energy consumption. The propagation, reflection, and superposition of stress waves within the rock mass gave rise to a complex stress zone, ultimately causing the rock mass to break. These findings lay the groundwork for developing a stress-damage-energy-failure combined model for shale subjected to dynamic impact loads, thereby enhancing the development of intricate networks in shale reservoir and improving the effective extraction of shale gas. Glucose and platelet are two easily obtained clinical indicators; the present research aimed to demonstrate their association with hemorrhagic transformation (HT) in acute ischemic stroke (AIS) patients without thrombolytic or thrombectomy therapy. This was a single-center retrospective study. Patients who were diagnosed with HT after AIS were included in the HT group. Meanwhile, using the propensity score matching (PSM) approach, with a ratio of 1:2, matched patients without HT were included in the non-HT group. Serum G/P levels were measured on the first morning after admission (at least eight hours after the last meal). Characteristics were compared between the two groups. Multivariate logistic regression was used to determine the independent relationship between G/P and HT after AIS, with G/P being divided into quartiles. From January 2013 to March 2022, we consecutively included 643 AIS patients with HT (426/643 [66.25%] with HI and 217/643 [33.75%] with PH), and 1282 AIS patients without HT, at the First Affiliated Hospital of Wenzhou Medical University. The HT group had higher G/P levels than the non-HT group (0.04 ± 0.02 vs. 0.03 ± 0.02, p < 0.001). However, there was no difference in G/P levels between HI and PH subgroups (0.04 ± 0.02 vs. 0.04 ± 0.02, p > 0.05). Moreover, the G/P levels were divided into quartiles (Q1 ≤ 0.022; Q2 = 0.023–0.028; Q3 = 0.029–0.039; Q4 ≥ 0.040), with Q1 being settled as the reference layer. After controlling the confounders, multivariate regression analyses showed that the Q4 layer (Q4: G/P ≥ 0.040) was independently associated with elevated HT risk (odds ratio [OR] = 1.85, 95% CI = 1.31–2.63, p < 0.001). G/P levels on admission were independently associated with HT risk in AIS patients. In clinical practice, adequate attention should be paid to AIS patients with elevated G/P levels (G/P ≥ 0.040). Abstract: Lime neutralization and coprecipitation of arsenate with iron is widely practiced for the removal and immobilization of arsenic from mineral processing effluents. However, the stability of the generated iron-arsenate coprecipitate is still of concern. In this work, we developed a two-step coprecipitation process involving the use of iron and aluminum and tested the stability of the resultant coprecipitates. The two-step Fe–As–Fe or Fe–As–Al coprecipitation process involved an initial Fe/As = 2 coprecipitation at pH4 to remove arsenic from water down to 0.25 mg/L, followed by introduction of iron or aluminum (Fe/As = 2, Al/As = 1.5 or 2). The two-step coprecipitates showed higher stability than traditional Fe/As = 4 coprecipitate under both oxic and anoxic conditions. Leaching stability was enhanced when aluminum was applied in the second step. The use of aluminum in the second step also inhibited microbial mediated arsenate reduction and arsenic remobilization. The results suggest that the two-step coprecipitation process is superior to conventional coprecipitation methods with respect to the stability of the generated arsenic-bearing solid waste. The use of Al in the second step is better than Fe to enhance the stability. This work may have important implications to the development of new technologies for efficient arsenic removal from hydrometallurgical solutions and safe disposal in both oxic and anoxic environment. The catecholamines, mainly dopamine (DA), are present in the cellular cytosol with low abundance, while, play key roles in various neurodegenerative disorders. Here, platinized nanocavity carbon electrodes are employed to analyze cytosolic catecholamines in a single living PC12 cell, which is not easily quantified using the classic electrodes. The confined structure and excellent conductivity in the platinized nanocavity accelerate the electron transfer of the DA, resulting in a low detection limit down to 50 nM. The sensitivity of DA detection is improved to be 10.73 pA mM−1 nm−1 in the response range of 50 nM–100 μM, which guarantees quantitative analysis of cytosolic catecholamines with low abundance. Eventually, the platinized nanocavity electrode is employed to detect cytosolic catecholamines in a single PC12 cell without an obvious interruption of cellular catecholamine level. The cytosolic catecholamines in a single PC12 cell is measured in situ to be 0.1 μM, which is achieved for the first time at the single cell level using the electrochemical method. The results demonstrate that the nanocavity electrode with a high sensitivity could offer a promising means to dynamically track catecholamines in a single cell. The low abundant cytosolic catecholamines in a single PC12 cell are successfully quantified with high sensitivity using a platinized nanocavity carbon electrodes with the confined structure and excellent conductivity. In 2018, the two-dimensional (2D) PdSeO3 monolayer was predicted to be a highly promising photocatalyst for direct overall water splitting in the light of density functional theory (DFT) computations. Herein, we present the first report on the synthesis of 2D PdSeO3 nanosheets by using the quaternary ammonium intercalation-assisted electrochemical exfoliation method. The resulting atomically thin PdSeO3 nanosheets possess a moderate band gap with suitable band edge alignments for water splitting, and display excellent activity and stability in photocatalytic H2 evolution. Cardiac fibrosis is closely associated with various heart diseases and is an important pathological feature of cardiac remodeling. However, detailed mechanisms underlying cardiac fibrosis remain largely unknown. Long noncoding RNAs (lncRNAs) are reported to serve significant roles in the development of cardiac fibrosis. The present study aimed to identify the role of a novel lncRNA, homeobox A11 antisense (HOXA11-AS), in cardiac fibrosis. Overexpression of HOXA11-AS in mouse cardiac fibroblasts (CFs) increased the expression of transforming growth factor β1 (TGFβ1) and its downstream molecules, while knockdown of HOXA11-AS inhibited the TGFβ1 signaling pathway. Furthermore, as determined by colony formation and MTT assays, HOXA11-AS overexpression promoted colony formation and viability in mouse CFs, while HOXA11-AS knockdown had the opposite effect. In addition, overexpression of HOXA11-AS increased cell migration and invasion in the Transwell assays, whereas expression knockdown decreased the metastatic ability of cells. In order to explore the detailed mechanism, co-transfection of HOXA11-AS expression plasmid and siTGFβ1 into CFs resulted in increased cell proliferative rate and cell metastasis through the TGFβ1 signaling pathway. Taken together, the present study suggested that the lncRNA HOXA11-AS may be a potential therapeutic target against cardiac fibrosis, and provided a novel insight into the diagnosis and treatment of clinical cardiac fibrosis. Alpha-toxin produced by Clostridium perfringens is an important virulence factor, causing food poisoning and gas gangrene in humans. As such, it is considered a potential bioterrorism threat. To date, there is still no human effective therapeutic drug against alpha-toxin. In this study, a human single chain antibody against alpha-toxin was produced from synthetic (Tomlinson I + J) naive phage display libraries, and its preventive and therapeutic efficacy in mice was examined. To prove the neutralizing potential of the scFv, alpha-toxin was preincubated with scFv and subsequently tested for its lecithinase and hemolytic activity, as well as its lethal effect in mice following intravenous administration. The equilibrium association constant between scFv and CPA was 2.02 × 10 10 (1/M), as analyzed by SPR. The scFv could inhibit lecithinase and hemolytic activity, and provided effective protection against alpha-toxin when mice were challenged 1-h post scFv injection. In addition, the survival rate reached 80% for mice treated with scFv within 30 min of being challenged with a 2 × LD50 dose of alpha-toxin. These results confirmed that we successfully prepared a human scFv against C. perfringens type A alpha-toxin, which can be used in the prevention and treatment of alpha-toxin-related illness. Abstract: Sediment bound arsenic usually undergoes phase transformation processes when it is transported and buried in deeper settings. This work investigated anaerobic microbial mediated speciation change of the arsenic in offshore sediment and monitored the transformation process of oxyhydroxide associated arsenate to sulfide associated forms. The fate of arsenic and possible pathways of transformation were discussed based on quantitative analysis of aqueous and solid arsenic and iron, and qualitative characterization using X-ray absorption near edge spectroscopy (XANES). Arsenic was released and reduced upon development of anoxic conditions but was resequestered by authigenic minerals later. Most of the arsenic in the sediment was converted to orpiment-like material. Sulfide may have played double roles in arsenic redistribution process, i.e. promoting arsenic release from host oxyhydroxides in early stage and removal of arsenite from solution in the form of arsenic sulfide in later stage. The findings have implications about the pathways of arsenic transformation when arsenate is transported and buried below redox boundaries in offshore sediment. [Copyright &y& Elsevier]
