Background and Aims: Botryosphaeriaceae species are recognised as important pathogens of grapevines both in Australia and overseas. The identity, prevalence and distribution of Botryosphaeriaceae species in vineyards throughout the major winegrowing regions of New South Wales (NSW) and South Australia (SA) was determined to provide a foundation for improved disease prevention and management. Methods and Results: Field surveys from 91 vineyards across NSW and SA resulted in the collection of 2239 diseased wood samples and subsequent isolation of 1258 Botryosphaeriaceae isolates. Morphological identification along with phylogenetic analysis of ribosomal DNA internal transcribed spacer regions (ITS1-5.8S-ITS2) and partial sequences of the translation elongation factor 1-α gene (EF1-α) showed that eight Botryosphaeriaceae species from four phylogenetic lineages occur on grapevines in eastern Australia, including Diplodia seriata, Diplodia mutila, Lasiodiplodia theobromae, Neofusicoccum parvum, Neofusicoccum australe, Botryosphaeria dothidea, Dothiorella viticola and Dothiorella iberica. Conclusions: The prevalence of individual species varied according to geography and climate. Species of Diplodia and Dothiorella, characterised by thick-walled, pigmented conidia were the most prevalent and were distributed widely throughout both NSW and SA. Species with hyaline conidia, such as Neofusicoccum and Fusicoccum, were isolated less frequently and displayed more limited geographic ranges, whilst only a single isolate of Lasiodiplodia was recovered, this being from the northern most region of NSW. Significance of the Study: The identification of eight taxa within the Botryosphaeriaceae, and their distributions throughout south-eastern Australia was established and discussed in context with climate, reported optimum growth temperatures, and more recent taxonomic and nomenclatural revisions. Context: Omega-3 in pasture-fed beef and lamb is related to its availability in the forage species grazed. The variation in omega-3 content of several forages has been examined in the USA and UK, but not in south-eastern Australia across different stages of maturity. Aims: This study aimed to determine the change in omega-3, at different stages of plant development, in four species of forage commonly grazed by ruminants in south-eastern Australia. Methods: Four species, oats (Avena sativa L. cv. Eurabbie), annual ryegrass (Lolium perenne L. cv. Rambo), phalaris (Phalaris aquatica L. cv HoldfastGT) and subterranean clover (Trifolium subterrraneum L. cv. Coolamon) were grown in a replicated pot trial and harvested at seven stages of maturity corresponding to early vegetative, late vegetative, stem elongation, boot, anthesis, soft dough and ripening for oats, phalaris and ryegrass, or relative to day of flowering in clover. Proximate analysis and the concentration of fatty acids including C18:3n-3 and C18:2n-6 as well as the ratio of n-6:n-3 was determined. Key results: The mean (±s.e.) proportion of C18:3n-3 was highest in clover (50.6 ± 0.8), and higher in ryegrass (45.1 ± 0.5) and phalaris (44.0 ± 0.6) compared with oats (39.4 ± 0.5). Omega-3 proportion decreased with increasing maturity for oats (13.9 ± 0.86 vs 64.3 ± 0.68), ryegrass (17.3 ± 0.86 vs 68.2 ± 0.65) and phalaris (21.6 ± 1.63 vs 65.1 ± 0.79) but not to the same extent for sub clover (41.5 ± 1.38 vs 57.4 ± 0.79). The proportion of omega-3 was positively correlated with crude protein content. Conclusions: Omega-3 levels decreased in forages during development and was positively related to crude protein content, which is likely associated with total leaf chloroplasts. Although crude protein content remained higher for clover compared with other species, this did not translate to a higher proportion of omega-3 for any crude protein level. Implications: Grazing forages at earlier stages of maturity and maximising the amount of crude protein available for growth will increase the amount of omega-3 in plant material. The availability of this omega-3 for incorporation into meat and milk should be examined. Abstract Objective: The objective of this study was to assess and validate the EDIS (Emergency Department Information System) as a means of acquiring Level 2 injury surveillance data as defined by the National Data Standards for Injury Surveillance in public hospital emergency departments in Queensland. Methods: This validation was based on interviews with patients presenting to selected emergency departments in Queensland for treatment of an injury which was then compared to data obtained from emergency department staff completing the routine medical record for these patients using EDIS. To assess the level of coding variability and set baseline levels of agreement the data were separately coded by three experienced coders and the level of agreement evaluated. At the time of the validation emergency department staff were also asked to complete a questionnaire relating to their attitudes towards EDIS and to injury surveillance. Results: A total of 572 cases of injury were collected during the period of the validation of which 509 (89.0%) corresponded to an injury recorded in EDIS. Demographic details collected in the emergency department data were coded correctly in almost all cases. The exception was Indigenous status where the emergency department systems recorded only 38% of the Indigenous cases correctly. Injury data fields were better than 85% complete for two sites but only 65% complete for the third. Coding agreement between Emergency Department Information System and the validation study for items relating to the injury event varied from 95% for Intent to 50% for Activity when injured. The interrater agreement analysis revealed that the highest agreement between the coders was for intent (94–96%) while the lowest was for main injury factor (63–78%). The results of the emergency department staff satisfaction survey showed a high acceptance of EDIS and injury surveillance in general. Conclusions: This analysis demonstrates that injury... Bacterial pathogens resistant to antibiotics have become a serious health threat. Those species which have developed resistance against multiple drugs such as the carbapenems, are more lethal as these are last line therapy antibiotics. Current diagnostic tests for these resistance traits are based on singleplex target amplification techniques which can be time consuming and prone to errors. Here, we demonstrate a chip based optofluidic system with single molecule sensitivity for amplification-free, multiplexed detection of plasmids with genes corresponding to antibiotic resistance, within one hour. Rotating disks and microfluidic chips with functionalized polymer monoliths provided the upstream sample preparation steps to selectively extract these plasmids from blood spiked with E. coli DH5α cells. Waveguide-based spatial multiplexing using a multi-mode interference waveguide on an optofluidic chip was used for parallel detection of three different carbapenem resistance genes. Invasive vertebrates are a leading cause of extinction on islands and rats ( Rattus spp.) are one of the most damaging to island ecosystems. Methods to eradicate rats from islands are well established and there have been over 580 successful eradications to date. Increasingly, rat eradications are being implemented on tropical islands, a reflection of the need to protect the threatened biodiversity in the tropics. Yet rat eradications on tropical islands fail more frequently than those in temperate climates. In an effort to identify the main reasons for the lower success rate on tropical islands and possible solutions, a workshop was convened with 34 experts in rat eradication, tropical rodent and island ecology and toxicology. The workshop focused on projects using aerial broadcast of brodifacoum, a 2nd generation anticoagulant, because this approach has provided the highest success rate for eradicating rodents from islands. The workshop participants reviewed previously identified challenges to successful rat eradications on tropical islands including increased insect and crab densities resulting in competition for bait, year round or unpredictable timing of breeding rats and increased or unpredictable availability of alternative, natural foods. They also identified a number of new, likely reasons for the lower success rate on tropical islands and provided recommendations for how to address these risks in the planning and implementation of rat eradications. While the success rate of aerial broadcast rat eradications in tropical environments is quite high at 89% ( n = 47), it is hoped that by following the recommended best practices provided in this paper, future success rates will be closer to the 96.5% ( n = 116) demonstrated for aerial broadcast rat eradications on temperate islands. Paediatric emergency research is hampered by a number of barriers that can be overcome by a multicentre approach. In 2004, an Australia and New Zealand-based paediatric emergency research network was formed, the Paediatric Research in Emergency Departments International Collaborative (PREDICT). The founding sites include all major tertiary children's hospital EDs in Australia and New Zealand and a major mixed ED in Australia. PREDICT aims to provide leadership and infrastructure for multicentre research at the highest standard, facilitate collaboration between institutions, health-care providers and researchers and ultimately improve patient outcome. The problem of acoustic feedback in hearing aids could be solved potentially by applying a compliant hydrogel to the outer surface that would conform to the ear canal and block feedback. With this objective, several formulations of hydrogels were developed and their swelling and mechanical properties investigated. Hydrogel formulations were polymerized from hydroxyethyl methacrylate (HEMA) and N-vinyl-pyrrolidone (NVP), with various photo-initiators, crosslinkers, and swelling agents. The hydrogel that swelled most rapidly and yet remained undissolved in water had a monomer composition of 40 mol% HEMA, 60 mol% NVP, with 1 wt% polyethylene glycol dimethacrylate as a crosslinker, and 0.5 wt% 2,2-dimethoxy-2-phenyl-acetophenone as the photo-initiator. The tensile modulus, strength, hardness, and durability of the dry hydrogels were not a strong function of composition. In the swollen state, the mechanical properties were much reduced. The aim of this study is to investigate whether pulsed ultrasound (US) in combination with gentamicin yields a decreased viability of bacteria in biofilms on bone cements in vivo. Bacterial survival on bone cement in the presence and absence of ultrasound was compared in a rabbit model. Two bone cement samples with an Escherichia coli ATCC 10798 biofilm were implanted in a total of nine rabbits. In two groups bone cement discs loaded with gentamicin, freshly prepared and aged were used, and in one group unloaded bone cement discs in combination with systemically administered gentamicin. Pulsed ultrasound with a frequency of 28·48 kHz and a maximum acoustic intensity of 500 mW cm−2 was applied continuously from 24 h till 72 h postsurgery on one of the two implanted discs. After euthanization and removal of the bacteria from the discs, the number of viable bacteria were quantified and skin samples were analysed for histopathological examination. Application of ultrasound, combined with gentamicin, reduced the viability of the biofilms in all three groups varying between 58 and 69% compared with the negative control. Histopathological examinations showed no skin lesions. Ultrasound resulted in a tendency of improved efficacy of gentamicin, either applied locally or systemically. Usage of ultrasound in this model proved to be safe. This study implies that ultrasound could improve the prevention of infection immediately after surgery, especially because the biomaterials, gentamicin and ultrasound used in this model are all in clinical usage, but not yet combined in clinical practice. Infection of implanted medical devices by Gram-positive organisms such as Staphylococcus ssp. is a serious concern in the biomaterial community. In this research the application of low frequency ultrasound to enhance the activity of vancomycin against implanted Staphylococcus epidermidis biofilms was examined. Polyethylene disks covered with a biofilm of S. epidermidis were implanted subcutaneously in rabbits on both sides of their spine. The rabbits received systemic vancomycin for the duration of the experiment. Following 24 h of recovery, one disk was insonated for 24 or 48 h while the other was a control. Disks were removed and viable bacteria counted. At 24 h of insonation, there was no difference in viable counts between control and insonated biofilms, while at 48 h of insonation there were statistically fewer viable bacteria in the insonated biofilm. The S. epidermidis biofilms responded favorably to combinations of ultrasound and vancomycin, but longer treatment times are required for this Gram-positive organism than was observed previously for a Gram-negative species. Objective: To determine the presentation rates for paediatric poisoning by ingestion and the determinants of hospital admission.Methodology: Cross-sectional survey using an injury surveillance database from emergency departments in South Brisbane, Mackay and Mt Isa, Queensland, from January 1998 to December 1999. There were 1516 children aged 0-14 years who presented following ingestional poisoning.Results: The presentation rates for poisoning were 690, 40 and 67 per 100000 population aged 0-4, 5-9 and 10-14 years, respectively. The admission rates to hospital for poisoning were 144, 14 and 22 per 100000 population aged 0-4, 5-9 and 10-14 years, respectively. Although presentation rates for poisoning were higher in the rural centres the admission rates were disproportionately high for the 0-4 years age group. The agents most frequently ingested were paracetamol, Dimetapp, rodenticides and essential oils.Conclusion: There is a need to design and implement interventions aimed at reducing poison exposures and unnecessary hospital admissions in the 0-4 years age group.
