Background: Hashimoto's thyroiditis (HT) is an independent risk factor for papillary thyroid carcinoma (PTC), but the underlying mechanism remains unknown. The incidence of PTC in patients with HT is significantly elevated, and the presence of both HT and PTC contributes to a higher rate of misdiagnosis. Materials and Methods: Gene ontology (GO) enrichment analysis and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis were performed on the thyroid nodule gene chip dataset from GEO Datasets. Serum and clinical data from 191 patients with thyroid nodules at the affiliated hospital were collected for analysis. Experimental techniques, including real-time quantitative PCR, ELISA, immunohistochemistry (IHC), and enzyme activity detection, were used to measure the level of dipeptidyl peptidase 4 (DPP4) in thyroid nodule tissues and serum. Results: Thyroid nodules in patients with HT and PTC exhibit high levels of DPP4, along with elevated concentrations of soluble DPP4 in the serum. These findings demonstrate the potential predictive value of soluble DPP4 for PTC diagnosis. Conclusions: The concentration and enzymatic activity of soluble DPP4 in serum can serve as diagnostic biomarkers for patients with HT-associated PTC. [ABSTRACT FROM AUTHOR]

The ontogenetic sequence of an early Cambrian redlichiid trilobite Bathynotus kueichouensis is presented on the basis of numerous articulated specimens from Guizhou, China. The body regionalization of B. kueichouensis witnessed a wide variation in the morphology of trunk segments between different thoracic regions, referred to as the ‘thoracic region 1-4’. A greater size variation of these long pleural spines is recognized from the earlier meraspid degrees in a gradual appearance or disappearance until the holaspid period. The thoroughly inverse variation of these pleural spines in size and form is seen for the first time during the process of trilobite segment formation, which offers an opportunity to trace the evolutionary path of trilobite body structures, associated with their functional morphology and behavior. The ontogenetic strategy of B. kueichouensis is broadly similar to those of other redlichioid representatives where the full ontogenetic sequence exhibited a balanced rate in segment expression and liberation. Such a developmental mode may be the reason why most redlichiids possess a micropygous body patterning during ontogeny and provides insights into the controls of early arthropod developmental segmentation during evolutionary process. High-quality epitaxial films of a ferroelectric oxide Sr2Nb2O 7 were successfully fabricated by solid phase epitaxy (SPE) on (110) LaAlO3 single crystal substrates. In the SPE method, amorphous Sr–Nb–O films are first deposited by pulsed laser deposition at room temperature and then annealed in vacuum at elevated temperatures, resulting in the crystallization of Sr2Nb2O 7 with highly ordered atomic arrangement and an atomically flat surface. The refractive index of the resultant film was 2.1, indicating that the dielectric permittivity of the film was in between 20 and 80, which corresponds well with that of single crystal Sr2Nb2O7, demonstrating the effectiveness of the SPE method for the fabrication of high-quality epitaxial films of Sr2Nb2O7. Disulfide bonds play essential roles in the folding of secretory and plasma membrane proteins in the endoplasmic reticulum (ER). In eukaryotes, protein disulfide isomerase (PDI) is an enzyme catalyzing the disulfide bond formation and isomerization in substrates. The Arabidopsis (Arabidopsis thaliana) genome encodes diverse PDIs including structurally distinct subgroups PDI-L and PDI-M/S. It remains unclear how these AtPDIs function to catalyze the correct disulfide formation. We found that one Arabidopsis ER oxidoreductin-1 (Ero1), AtERO1, can interact with multiple PDIs. PDI-L members AtPDI2/5/6 mainly serve as an isomerase, while PDI-M/S members AtPDI9/10/11 are more efficient in accepting oxidizing equivalents from AtERO1 and catalyzing disulfide bond formation. Accordingly, the pdi9/10/11 triple mutant exhibited much stronger inhibition than pdi1/2/5/6 quadruple mutant under dithiothreitol treatment, which caused disruption of disulfide bonds in plant proteins. Furthermore, AtPDI2/5 work synergistically with PDI-M/S members in relaying disulfide bonds from AtERO1 to substrates. Cretaceous-Cenozoic basins developed in the NE Tibetan Plateau contain key archives to unravel the growth history of the plateau in response to the India-Eurasia collision. Here we present magnetostratigraphic results of a Late Cretaceous to Paleogene succession of the Zhongba section outcropping at the southern margin of the eastern Xining basin. This succession consists of three lithological units punctuated by two stratigraphic unconformities, which best recorded the deformation history of this foreland basin. Detailed magnetostratigraphic investigation show that the lower terrestrial sedimentary rock unit, the Minhe Group, was deposited in latest Cretaceous in the time span of ca. 74.5-69.2 Ma; the middle unit was deposited in Paleogene in the time span of ca. 49.3-22 Ma; and the upper conglomeratic unit, not dated, possibly was deposited in early Miocene. Accordingly, the Cretaceous-Paleogene unconformity, widely observed in the foreland basins of NE Tibet, represents a sedimentary hiatus duration of ~19.9 m.y., from ca. 69.2 Ma to ca. 49.3 Ma, which possibly recorded the far-field response to the tectonic transition from Neo-Tethys oceanic plate subduction to the India-Eurasia collision in southern Tibet. Changes in provenance, sedimentary accumulation rate, and mean susceptibility value at ca. 33-30 Ma, and the total prolate anisotropy of magnetic susceptibility (AMS) ellipsoids and provenance shifting since ca. 23-19 Ma, point to the pulsed growth of West Qinling, and rapid uplift of Laji Shan, respectively, indicating an enhanced effect of the India-Eurasia collision in Oligocene and early Miocene. Guanylate-binding protein 7 (GBP7) belongs to the GBP family, which plays key roles in mediating innate immune responses to intracellular pathogens. Thus far, GBP7 has been reported to be a critical cellular factor against bacterial infection. However, the relationship between GBP7 and influenza A virus (IAV) replication is unknown. Here, we showed that GBP7 expression was significantly upregulated in the lungs of mice, human peripheral blood mononuclear cells (PBMCs), and A549 cells during IAV infection. Using the CRISPR-Cas9 system and overexpression approaches, it was found that GBP7 knockout inhibited IAV replication by enhancing the expression of IAV-induced type I interferon (IFN), type III IFN, and proinflammatory cytokines. Conversely, overexpression of GBP7 facilitated IAV replication by suppressing the expression of those factors. Furthermore, GBP7 knockout enhanced IAV-induced nuclear factor-κB (NF-κB) activation and phosphorylation of stat1 and stat2; overexpression of GBP7 had the opposite effect. Our data indicated that GBP7 suppresses innate immune responses to IAV infection via NF-κB and JAK-STAT signaling pathways. Taken together, upon IAV infection, the induced GBP7 facilitated IAV replication by suppressing innate immune responses to IAV infection, which suggested that GBP7 serves as a therapeutic target for controlling IAV infection. Hydrogen is becoming a more and more crucial resource for chemical industries. Hydrogen network integration is an effective tool in hydrogen resource conservation. Hydrogen sources flow in and flow out from different processing units where consumption, generation, mixing, and separation occur. Consequently, there is a dependence on both hydrogen and contaminant species among those streams, especially when throughputs of processing units fluctuate. Hydrogen network integration should account for such a dependence so that real properties including the flow rate and concentration can be obtained to perform reliable network integration. This article proposes the processing unit mass balance to describe the dependence of hydrogen streams and establishes a corresponding superstructure as problem illustration. A mixed integer nonlinear programing (MINLP) model is built to synthesize hydrogen networks for fresh hydrogen minimization. This method considers the tolerable disturbance of the processing unit and thus a reliable fresh hydrogen target and internal sources profile can be obtained. In this research, a new triterpenoid, tirucalla-8,24-diene-3β,11β-diol-7-one (1), and eupha-8,24-diene-3β,11β-diol-7-one (2), which was isolated from Euphorbia kansui for the first time, together with twelve other known compounds (3-14), were isolated from the ethyl acetate extract of Euphorbia kansui. Their structures were elucidated based on High resolution electrospray ionization mass spectrometry (HR-ESI-MS), Infrared Spectroscopy (IR), 1D and 2D Nuclear Magnetic Resonance (NMR) data. The first metal-free, three-component radical aminofluorination of styrene derivatives has been developed, which employs Selectfluor as a fluorine source and azole derivatives as a nitrogen source. This transformation proceeds with high regioselectivity, providing various 1,2-aminofluorination compounds under mild conditions. Biotransformation of trans-resveratrol and synthetic (±)-ε-viniferin in aqueous acetone using horseradish peroxidase and hydrogen peroxide as oxidants resulted in the isolation of two new resveratrol trimers (3 and 4), one new resveratrol derivative (5) with a dihydrobenzofuran skeleton, together with two known stilbene trimers (6 and 7), and six known stilbene dimers (8–13). Their structures and relative configurations were identified through spectral analysis and possible formation mechanisms were also discussed. Among these oligomers, trimers 6 and 7 were obtained for the first time through direct transformation from resveratrol. UHRF2 has been implicated as a novel regulator for both DNA methylation (5mC) and hydroxymethylation (5hmC), but its physiological function and role in DNA methylation/ hydroxymethylation are unknown. Here we show that in mice, UHRF2 is more abundantly expressed in the brain and a few other tissues. Uhrf2 knock-out mice are viable and fertile and exhibit no gross defect. Although there is no significant change of DNA methylation, the Uhrf2 null mice exhibit a reduction of 5hmC in the brain, including the cortex and hippocampus. Furthermore, the Uhrf2 null mice exhibit a partial impairment in spatial memory acquisition and retention. Consistent with the phenotype, gene expression profiling uncovers a role for UHRF2 in regulating neuron-related gene expression. Finally, we provide evidence that UHRF2 binds 5hmC in cells but does not appear to affect the TET1 enzymatic activity. Anterior cervical discectomy and fusion (ACDF) and cervical disc arthroplasty (CDA) are the most commonly used procedures in cervical spondylosis. However, only a few published studies exist in the literature comparing these two operation types, particularly its mid-term efficacy and safety. Furthermore, in those studies, even large sample trials, when compared, have elicited controversial results, making it inconvenient for clinicians to refer to them. The aim of the present study was to clarify the advantages and shortcomings of the two procedures. Articles indexed in the PubMed, Web of Science, Cochrane Library, EMBASE, China Biological Medicine and China National Knowledge Infrastructure (CNKI) databases, as of March 2016, that met our criteria were searched. A total of 18 trials involving 3,040 patients were included in our final analysis. The most important results drawn from the present analysis were as follows: Insignificant differences were identified in the blood loss [weighted mean difference (WMD)=6.23; 95% confidence intervals (CI), -0.85 to 13.32; P=0.08], surgical time [standardized mean difference (SMD)=0.40; 95% CI, -0.01 to 0.82; P=0.06], the time of hospital stay (SMD=0.05; 95% CI, -0.28 to 0.37; P=0.77) and the total complications rate [odds ratio (OR)=0.86; 95% CI, 0.66 to 1.13l; P=0.28] on a comparison of the two operation methods. By contrast, comparing CDA with ACDF, the CDA had higher Short Form survey (SF-36) scores (WMD=1.65; 95% CI, 0.61 to 2.69; P=0.002), a larger range of motion in the operation level (SMD=6.53; 95% CI, 3.89 to 9.17; P<0.0001), a higher rate of neurological improvement following the operation (OR=1.80; 95% CI, 1.29 to 2.52; P=0.0006), a lower Visual Analog Scale (VAS) score of neck pain (WMD= 0.16; 95% CI, -0.28 to 0.05; P=0.006) and arm pain (WMD= 0.12; 95% CI, -0.24 to - 0.01; P=0.04). In addition, in the mid-term following the surgery, CDA had a lower Neck Disability Index (NDI; SMD=0.18; 95% CI, -0.28 to - 0.07; P=0.001) and a lower reoperation rate of adjacent levels (OR=0.54; 95% CI, 0.35 to 0.85; P=0.007) compared with ACDF. Taken together, these results suggested that CDA and ACDF are efficient and safe methods for dealing with cervical spondylosis. Hydrogen is a crucial and expensive resource for refineries. Hydrogen network integration is an effective way to conserve fresh hydrogen. On the basis of relative concentration analysis, a mixed integer linear programming (MILP) mathematical model is established to synthesize refinery multi-impurity hydrogen networks. By correlating the hydrogen consumption of a sink with its oil processing throughputs, this model reveals the fresh hydrogen variation tendency caused by fluctuation of each capacity, and determines the key sink(s) of a hydrogen network with multiple impurities on the premise of minimum fresh hydrogen consumption. With a structure matrix variable, practical hydrogen networks can be analyzed through the proposed model. The results show that the novel method in this paper is superior to traditional absolute concentration-based methods in fresh hydrogen conservation under cases of static, fluctuant throughputs, and fluctuant throughputs with restricted network structure. Endothelin-1 (ET-1) autocrine and paracrine signaling modulate cell proliferation of tumor cells by activating its receptors, endothelin A receptor (ETAR) and endothelin B receptor (ETBR). Dysregulation of ETAR activation promotes tumor development and progression. The potential of ETAR antagonists and the dual-ETAR and ETBR antagonists as therapeutic approaches are under preclinical and clinical studies. Salvianolic acid A (Sal A) is a hydrophilic polyphenolic derivative isolated from Salvia miltiorrhiza Bunge (Danshen), which has been reported as an anti-cancer and cardio-protective herbal medicine. In this study, we demonstrate that Sal A inhibits ETAR activation induced by ET-1 in both recombinant and endogenous ETAR expression cell lines. The IC50 values were determined as 5.7 µM in the HEK293/ETAR cell line and 3.14 µM in HeLa cells, respectively. Furthermore, our results showed that Sal A suppressed cell proliferation and extended the doubling times of multiple cancer cells, including HeLa, DU145, H1975, and A549 cell lines. In addition, Sal A inhibited proliferation of DU145 cell lines stimulated by exogenous ET-1 treatment. Moreover, the cytotoxicity and cardio-toxicity of Sal A were assessed in human umbilical vein endothelial cells (HUVEC) and Human-induced pluripotent stem cell-derived cardiomyocytes (hiPS-CMs), which proved that Sal A demonstrates no cytotoxicity or cardiotoxicity. To accurately, safely, and efficiently evaluate the bitterness of Traditional Chinese Medicines (TCMs), a robust predictor was developed using robust partial least squares (RPLS) regression method based on data obtained from an electronic tongue (e-tongue) system. The data quality was verified by the Grubb's test. Moreover, potential outliers were detected based on both the standardized residual and score distance calculated for each sample. The performance of RPLS on the dataset before and after outlier detection was compared to other state-of-the-art methods including multivariate linear regression, least squares support vector machine, and the plain partial least squares regression. Both R² and root-mean-squares error (RMSE) of cross-validation (CV) were recorded for each model. With four latent variables, a robust RMSECV value of 0.3916 with bitterness values ranging from 0.63 to 4.78 were obtained for the RPLS model that was constructed based on the dataset including outliers. Meanwhile, the RMSECV, which was calculated using the models constructed by other methods, was larger than that of the RPLS model. After six outliers were excluded, the performance of all benchmark methods markedly improved, but the difference between the RPLS model constructed before and after outlier exclusion was negligible. Purpose: To demonstrate the effect of resveratrol on Notch signaling in MG-63 and U2OS osteosarcoma cell lines. Methods: Cell Counting Kit 8 reagent was used to analyze cell proliferation while TRIzol° reagent was employed for the extraction of total RNA. High Capacity cDNA reverse transcription chain reaction kit was used to transcribe 2 μg RNA. Western blot analysis was performed to examine Hes1 and Hey1 expression. Results: The results revealed that resveratrol treatment exhibited dual mechanisms of action on the activation of Notch signaling in osteosarcoma cells. The osteosarcoma cell lines, MG-63 and U2OS, when exposed to 20 μM concentration of resveratrol for 48 h showed significant toxicity compared to untreated cells. However, 30 μM concentration of resveratrol induced higher toxicity which was lethal to cell growth. The results from RT qPCR and Western blot data revealed a concentration-dependent effect of resveratrol on the expression of Notch signaling genes including Hes1, Hes5, Hey1, Hey2 and HeyL in U2OS cells. Treatment of U2OS cells with 20 μM concentration of resveratrol for 48 h induced a marked increase in the expression of Hes1, Hes5, Hey1, Hey2 and HeyL mRNA compared to the untreated cells. However, at a concentration of 30 μM, resveratrol inhibited the activation of Notch signaling pathway. This was evident by a decrease in the expression of Hes1, Hes5, Hey1, Hey2 and HeyL, Notch signaling target genes. Due to the wide deployment of wireless local area networks (WLAN), received signal strength (RSS)-based indoor WLAN localization has attracted considerable attention in both academia and industry. In this paper, we propose a novel page rank-based indoor mapping and localization (PRIMAL) by using the gene-sequenced unlabeled WLAN RSS for simultaneous localization and mapping (SLAM). Specifically, first of all, based on the observation of the motion patterns of the people in the target environment, we use the Allen logic to construct the mobility graph to characterize the connectivity among different areas of interest. Second, the concept of gene sequencing is utilized to assemble the sporadically-collected RSS sequences into a signal graph based on the transition relations among different RSS sequences. Third, we apply the graph drawing approach to exhibit both the mobility graph and signal graph in a more readable manner. Finally, the page rank (PR) algorithm is proposed to construct the mapping from the signal graph into the mobility graph. The experimental results show that the proposed approach achieves satisfactory localization accuracy and meanwhile avoids the intensive time and labor cost involved in the conventional location fingerprinting-based indoor WLAN localization. Laser-induced breakdown spectroscopy (LIBS) was applied to perform a rapid elemental analysis and provenance study of Blumea balsamifera DC. Principal component analysis (PCA) and partial least squares discriminant analysis (PLS-DA) were implemented to exploit the multivariate nature of the LIBS data. Scores and loadings of computed principal components visually illustrated the differing spectral data. The PLS-DA algorithm showed good classification performance. The PLS-DA model using complete spectra as input variables had similar discrimination performance to using selected spectral lines as input variables. The down-selection of spectral lines was specifically focused on the major elements of B. balsamifera samples. In this work, isothermal circular strand-displacement polymerization amplification assay is developed for highly specific and sensitive detection of adenosine triphosphate (ATP). The amplification process consists of circular common target molecule-displacement polymerization (CCDP) and circular nucleic acid strand-displacement polymerization (CNDP). In the presence of ATP, the complementary strand was released from the aptamer by the target recognition of ATP, and catalyzed the subsequent cycle reaction. With the polymerase and primer, the displaced target triggers the process of CCDP. With the involvement of nicking endonuclease, the released complementary strand triggers the CNDP. Combined CCDP with CNDP, the exponentially produced fluorescence probes are obtained, achieving a detection limit of ATP as low as 2.6×10-10M. Moreover, the proposed strategy exhibits an excellent specificity and is successfully applied in real sample assay which demonstrates potential application in practical samples. Using microalgae for synchronous biodiesel production and wastewater treatment is a promising technology. The growth, lipid accumulation and nutrient uptake characteristics of an oleaginous microalga Chlorella sp. HQ were evaluated at different initial pH from 5.0 to 11.0. The pH values changed towards neutrality and ended in the range 6.0-9.0 without artificial control. The alkalinity change before 8 days was in accordance with pH changing. The alkalinity increase after 8 days might be due to the nitrate consumption, CO2 absorption and the algal release at stationary phases. The algal maximal cell density and population growth rate increased with initial pH values while the specific growth rate kept high without significant difference. After 30 days, the maximal algal lipid yield reaching 167.5 mg ∙ L-1 occurred at initial pH of 7.0 and the triacylglycerols content was significantly enhanced to 63.0% at initial pH of 5.0 but with a peak of 54.4 mg ∙ L-1 at initial pH of 9.0. Furthermore, nutrients were taken up by the alga obviously at all initial pH values. The total nitrogen (TN) and total phosphorus (TP) uptake efficiencies in neutral/alkalic circumstances were larger than that in acid circumstance. Background: Insulin enhancer binding protein-1 (ISL-1), a LIM-homeodomain transcription factor, is essential for the heart, motor neuron and pancreas development. Recently, ISL-1 has been found in some types of human cancers. However, how ISL-1 exerts the role in tumor development is not clear. Methods and results: The expression of ISL-1 was assessed in 211 human lymphoma samples and 23 normal lymph node samples. Immunohistochemistry results demonstrated a markedly higher expression of ISL-1 in 75% of non-Hodgkin lymphoma (NHL) samples compared with that in normal lymph nodes or Hodgkin lymphoma (HL) samples. CCK-8 analysis, cell cycle assay and xenograft model were performed to characterize the association between ISL-1 expression level and biological functions in NHL. The results showed that ISL-1 overexpression obviously promoted NHL cells proliferation, changed the cell cycle distribution in vitro and significantly enhanced xenografted lymphoma development in vivo. Real-time PCR, Western blot, luciferase assay and ChIP assay were used to explore the potential regulatory targets of ISL-1 and the results demonstrated that ISL-1 activated the c-Myc expression in NHL by direct binding to a conserved binding site on the c-Myc enhancer. Further results revealed that ISL-1 could be positively regulated by the c-Jun N-terminal kinase (JNK) and the Janus kinase/signal transducer and activator of transcription (JAK/STAT) pathways. Both the JNK and JAK/STAT signaling inhibitors could significantly suppressed the growth of NHL cells through the down-regulation of ISL-1 as demonstrated by CCK-8 and Western blot assays. Bioinformatic analysis and luciferase assay exhibited that ISL-1 was a novel target of p-STAT3 and p-c-jun. ChIP, Co-IP and ChIP-re-IP analysis revealed that ISL-1 could participate with p-STAT3 and p-c-Jun to form a p-STAT3/p-c-Jun/ISL-1 transcriptional complex that binds directly on the ISL-1 promoter, demonstrating a positive feedback regulatory mechanism for ISL-1 expression in NHL. Conclusions: Our results provide the first evidence that ISL-1 is tightly linked to NHL proliferation and development by promoting c-Myc transcription, and its aberrant expression was regulated by p-STAT3/p-c-Jun/ISL-1 complex activation. The aim of this study was to establish phosphorus fertilization recommendation index of different vegetables (leafy, gourd and leguminous vegetables) by soil test P (Olsen-P and Brayl-P) with the "3414" experiments of 121 sites during 2007-2010 in 30 counties of Guangdong Province, China. The results showed that relationship between vegetable relative yield and soil test P was linear with plateau and logarithmic equation model. The optimal amount of P fertilizer was established with soil test P by logarithmic equation mathematical models. The soil P content was calculated by the relative yield of 50, 75, 90 and 95%, to obtain the soil P abundance and deficiency indicators. Taking plentiful-lack of soil-test P as an index, soil Olsen-P and Bray l-P were classified into five classes corresponding to relative yield of 50, 75, 90 and 95%. The formation of a ferric hydroperoxide species from [Fe(bbpc)(MeCN)2]2+ (bbpc = N,N'-dibenzyl-N,N'-bis(2-pyridylmethyl)-1,2-cydohexanediamine) and its subsequent decomposition were analyzed kinetically. The rate of decay is not strongly influenced by the presence of either water or substrate, suggesting that the ferric hydroperoxide degrades through O--O bond homolysis and is not the relevant metal-based oxidant in the observed catalysis of C-H activation. The ten-eleven translocation (TET) family of dioxygenases (TET1/2/3) converts 5-methylcytosine to 5-hydroxymethylcytosine and provides a vital mechanism for DNA demethylation. However, how TET proteins are regulated is largely unknown. Here we report that the O-linked β-GlcNAc (O-GlcNAc) transferase (OGT) is not only a major TET3-interacting protein but also regulates TET3 subcellular localization and enzymatic activity. OGT catalyzes the O-GlcNAcylation of TET3, promotes TET3 nuclear export, and, consequently, inhibits the formation of 5-hydroxymethylcytosine catalyzed by TET3. Although TET1 and TET2 also interact with and can be O-GlcNAcylated by OGT, neither their subcellular localization nor their enzymatic activity are affected by OGT. Furthermore, we show that the nuclear localization and O-GlcNAcylation of TET3 are regulated by glucose metabolism. The growth, lipid accumulation and nutrient removal characteristics of an oleaginous microalga Chlorella sp. HQ in two types of secondary effluents (named as X and Q) before/after sterilization were evaluated. The results show that the algal growth rates under sterilization were higher than those under non-sterilization. However, sterilization caused a significant decrease in algal lipid and triacylglycerol (TAG) contents in both X and Q. And the lipid and TAG yields in non-sterile X were as much as 2.7 and 7.7 times higher than those in sterile X, reaching up to 51.3 and 16.1 mg L-1, respectively. However, the sterilization caused algal biomass increase in sample Q. Sterilization or not had almost no effect on the total phosphorus (TP) removal ability of Chlorella sp. HQ and it was found to have similar abilities to remove almost 100.0% TP from samples X and Q. While the total nitrogen (TN) removal efficiencies were promoted slightly under non-sterilization, increasing from 88.5 to 89.7% in X and from 13.3 to 17.2% in Q. Ribosome biogenesis drives cell growth and proliferation, but mechanisms that modulate this process within specific lineages remain poorly understood. Here, we identify a Drosophila RNA polymerase I (Pol I) regulatory complex composed of Under-developed (Udd), TAF1B, and a TAF1C-like factor. Disruption of udd or TAF1B results in reduced ovarian germline stem cell (GSC) proliferation. Female GSCs display high levels of ribosomal RNA (rRNA) transcription, and Udd becomes enriched in GSCs relative to their differentiating daughters. Increasing Pol I transcription delays differentiation, whereas reducing rRNA production induces both morphological changes that accompany multicellular cyst formation and specific decreased expression of the bone morphogenetic protein (BMP) pathway component Mad. The compound N,N'-dineopentyl-N,N'-bis(2-pyridylmethyl)-1,2-ethanediamine (dnbpn) and its ferrous complex [Fe(dnbpn)(OTf)2] were synthesized. The Fe(II) complex was used to catalyze the oxidation of hydrocarbons by H2O2 and O2. Although the catalyzed alkane oxidation by H2O2 displays a higher preference for secondary over tertiary carbons than those associated with most previously reported nonheme iron catalysts, the catalytic activity is markedly inferior. In addition to directing the catalyzed oxidation toward the less sterically congested C-H bonds of the substrates, the neopentyl groups destabilize the metal-based oxidants generated from H2O2 and the Fe(II) complex. The presence of benzylic substrates with weak C-H bonds stabilizes an intermediate which we have tentatively assigned as a high-spin ferric hydroperoxide species. The oxidant generated from O2 reacts with allylic and benzylic C-H bonds in the absence of a sacrificial reductant; less substrate dehydrogenation is observed than with related previously described systems that use O2 as a terminal oxidant. Background: Subarachnoid hemorrhage is a common and dangerous disease with an unfavorable prognosis. Patients with poor-grade subarachnoid hemorrhage (Hunt & Hess Grades 4-5) are unconscious on admission. Because of the high mortality and disability rate associated with poor-grade subarachnoid hemorrhage, it is often treated conservatively. Timing of surgery for poor-grade aneurysmal subarachnoid hemorrhage is still controversial, therefore this study aims to identify the optimal time to operate on patients admitted in poor clinical condition. Methods/design: Ninety-nine patients meeting the inclusion criteria were randomly assigned into three treatment groups. The early surgery group received operation within 3 days after onset of subarachnoid hemorrhage (day of SAH = day 1); the intermediate surgery group received operation from days 4 to 7, and surgery was performed on the late surgery group after day 7. Follow-up was performed 1, 3, and 6 months after aneurysm clipping. Primary indicators of outcome included the Extended Glasgow Outcome Scale and the Modified Rankin Scale, while secondary indicators of outcome were assessed using the Barthel Index and mortality. Discussion: This is the first prospective, single-center, observer-blinded, randomized controlled trial to elucidate optimal timing for surgery in poor-grade subarachnoid hemorrhage patients. Germination of Bacillus subtilis spores can be triggered by the binding of specific nutrients, called germinants, to germinant receptors (GRs) in the spore's inner membrane. This interaction eventually initiates, with variable time delays, the release of dipicolinic acid and cations from the spore core--a key step in spore germination. The kinetics of this process are highly heterogeneous for individual spores. In this work, we sought to investigate how the germination heterogeneity was controlled. In particular, we tested whether the rates of germination were determined by GR levels, which vary from spore to spore due to stochastic gene expression. Both the expression levels of GRs and the germination rate were measured in single spores, and the experimental results were compared to theoretical predictions. Our results indicated that the variation in the expression levels of GRs was not the primary factor that controls spore germination heterogeneity. In this work, we reported a simple rapid and point-of-care magnetic immunofluorescence assay for avian influenza virus (AIV) and developed a portable experimental setup equipped with an optical fiber spectrometer and a microfluidic device. We achieved the integration of immunomagnetic target capture, concentration, and fluorescence detection in the microfluidic chip. By optimizing flow rate and incubation time, we could get a limit of detection low up to 3.7 × 104 copy/μL with a sample consumption of 2 μL and a total assay time of less than 55 min. This approach had proved to possess high portability, fast analysis, high specificity, high precision, and reproducibility with an intra-assay variability of 2.87% and an interassay variability of 4.36%. As a whole, this microfluidic system may provide a powerful platform for the rapid detection of AIV and may be extended for detection of other viral pathogens; in addition, this portable experimental setup enables the development of point-of-care diagnostic systems while retaining adequate sensitivity. Abstract: Integrated layout optimization of multi-component structures under static loads and random excitations is studied in this paper. Dynamic responses are obtained by using the mode superposition method, and the prestress effect introduced by the static loads is taken into account in the analyses of dynamic responses. Locations of embedded components and pseudo-densities related to supporting structure are chosen as design variables. Design sensitivities of the dynamic responses with respect to the pseudo-densities are analytically derived. The optimization algorithm GCMMA (Global Convergent Method of Moving Asymptotes algorithm) is used to minimize simultaneously the static strain energy and dynamic responses. The ligand N,N′-bis(2-pyridylmethyl)-bis(ethylacetate)-1,2-ethanediamine (debpn) coordinates divalent transition metal ions in either a pentadentate or hexadentate fashion. The coordination number correlates with the ionic radius of the metal ion, with larger cations being heptacoordinate as assessed by solid-state analysis. With Mn(II), the debpn ligand is hexadentate and remains bound to the oxophilic metal ion, even when dissolved in water. The ligand's incomplete coordination of the manganous ion allows water molecules to coordinate to the metal center. These two properties, coupled with the high paramagnetism associated with the S = 5/2 metal center, enable [Mn(debpn)(H2O)](ClO4)2 to serve as a stable and effective magnetic resonance imaging contrast agent despite the ligand's lack of both a macrocyclic component and an anionic charge. Area-based yield insurance (AYI) is a kind of index insurance that can eliminate asymmetric information problems (adverse selection and moral hazard) and reduce transaction cost. It is important in managing crop risk for developing countries such as China, which has many small farms. However, the biggest challenge in index insurance is basis risk. In this regard, can AYI be used in China? Is AYI competitive with traditional multi-peril crop insurance (MPCI)? What are the principal factors affecting the effectiveness of AYI? Following and extending the approaches suggested by Miranda in 1991, this article compares the effectiveness of AYI and MPCI in terms of risk reduction per premium. An empirical model was established, indicating the relationship between the efficiency of AYI and the explanatory variables, using data from a survey of 108 wheat farmers from two counties in Hebei Province, China. We report the case of a 46-year-old man with von Hippel-Lindau (VHL) disease, manifesting disseminated leptomeningeal hemangioblastomatosis. The patient initially presented with a solitary hemangioblastoma in the right cerebellum. Later, he was diagnosed with VHL disease and underwent several surgical procedures in the following 14 years. But the prognosis was poor. Recently, the hemangioblastomatosis disseminated along leptomeninges involving both brain and spine. We aim to analyze the possible reason for the leptomeninges dissemination, discuss the imaging characteristics of this rare disease with ominous manifestation and propose the optimal strategy for treatment. We think the optimal treatment strategy should be surgical biopsy and surgical decompression. A long-term follow-up is inevitable. Based on vortex-acoustic coupling theory, large-eddy simulation with wall-adapting local eddy-viscosity model and finite element method are carried out to study the internal flowfield and acoustic field, respectively, in a tailpipe nozzle solid rocket motor with transition-section grain configuration. The numerical method by means of a mesh sensitivity analysis is proposed for validation. The instantaneous flowfield characteristics in the combustion chamber and tailpipe are analyzed. The excited low frequencies are close to that observed in experiment. The phenomenon in which acoustic signals, superimposed on the vortex-shedding motions, couple with an internal flowfield is proven to be one of the main reasons contributing to oscillation in the motor. According to fast Fourier transform, low frequencies predominate in the combustion chamber; however, high frequencies predominate in the tailpipe. Dozens of cases with different geometrical configurations are presented to investigate parameters that have impact on the low-frequency oscillation characteristics. The results indicate that the oscillation characteristics are mainly influenced by upstream mean velocity, transition-section angle, distance between vortex source and impingement points, tailpipe radius, and convergence angle of the nozzle. Silver nanoplates with an extremely high aspect ratio (up to over 400) and a widely tunable surface plasmon resonance (SPR) band have been successfully synthesized by combining the concepts of selective ligand adhesion and seeded growth. Citrate ligands are used as the sole surfactant to effectively block overgrowth on the basal {111} facets and only allow growth in the lateral direction. By slowing down the reaction rate using Ag−citrate complex as precursor, the thin nature of Ag nanoplates is maintained with the edge length grown up to 4 μm, which ensures the high aspect ratio and the widely tunable SPR band. We also observe a size distribution focusing effect that helps to produce uniform nanoplates as well as narrow SPR bands over a wide range, which is important in many practical applications. The present work describes the plant regeneration via somatic embryogenesis in two wild cotton species belonging to G genome: Gossypium nelsonii Fryx and Gossypium australe F Muell. The role of plant hormones and carbohydrates was also evaluated for somatic embryogenesis and somatic embryo development. Normal plants were obtained from G. nelsonii Fryx; abnormal plants and somatic embryos were obtained from G. australe F Muell. The best medium for callus induction for these G genome wild cotton species was MSB5 supplemented with 0.1 mg L−1 KT and 0.1 mg L−1 2,4-D. For embryogenic callus proliferation, the best medium used was MSB5 supplemented with 0.2 mg L−1 KT and 0.5 mg L−1 IBA. The medium MSB5 supplemented with 0.15 mg L−1 KT and 0.5 mg L−1 NAA was used successfully for root initiation and plant growth. In addition, adding CuSO4 and AgNO3 in the callus-inducing and proliferation medium resulted in a number of somatic embryos. Glucose and maltose, the carbon sources in somatic culture, were used for callus induction, but maltose worked even better than glucose for proliferation of embryogenic callus and development of somatic embryos. To determine the expression of nucleostemin (NS), epidermal growth factor (EGF) and epidermal growth factor receptor (EGFR) mRNA in human esophageal squamous cell carcinoma (ESCC) tissues and their association in a human ESCC cell line. The expression of NS, EGF and EGFR mRNA was determined in paired normal esophageal and ESCC tissues of 62 patients using in situ hybridization. The association between NS and EGF or EGFR was examined using immunoblotting and real time polymerase chain reaction in a human ESCC cell line transfected with NS siRNA or treated with a selective EGFR inhibitor. In normal esophageal and ESCC tissues, the positive detection rates were 21.0% (13/62) and 69.4% (43/62) for NS mRNA staining, 40.3% (25/62) and 77.4% (48/62) for EGF mRNA staining, and 30.6% (19/62) and 75.8% (41/62) for EGFR mRNA staining, respectively. These results indicated that NS, EGF and EGFR mRNA expression was upregulated mostly in ESCC tissues. Moreover, the expression of NS, EGF and EGFR mRNA was positively correlated with tumor grade, invasion and lymphatic metastasis of ESCC cells. NS mRNA was co-expressed with EGF and EGFR mRNA in ESCC tissues. The in vitro studies using a human ESCC cell line showed that knockdown of NS with NS siRNA significantly reduced EGF and EGFR expression. However, inhibition of the EGFR kinase activity with a specific EGFR kinase inhibitor had minimal effect on NS expression. The upregulation of NS, EGF and EGFR mRNA frequently occurs in ESCC tissues and is associated with malignancy of human esophageal squamous tumors. NS is required for EGF and EGFR expression. In this article, the finite-circle method is introduced for 2D packing optimization. Each component is approximated with a group of circles and the non-overlapping constraints between components are converted into simple constraints between circles. Three new algorithms—the bisection algorithm, the three-step algorithm, and the improved three-step algorithm with gap—are developed to automatically generate fewer circles approximating the components. The approximation accuracy, the circle number, and the computing time are analyzed in detail. Considering the fact that packing optimization is an NP-hard problem, both genetic and gradient-based algorithms are integrated in the finite-circle method to solve the problem. A mixed approach is proposed when the number of components is relatively large. Various tests are carried out to validate the proposed algorithms and design approach. Electric field induced, spectacular reduction of longitudinal resistivity of two dimensional electrons placed in strong magnetic field is studied in broad range of temperatures. The data are in good agreement with theory, considering the strong nonlinearity of the resistivity as result of non-uniform spectral diffusion of 2D electrons induced by the electric field. Comparison with the theory gives inelastic scattering time τin of the 2D electrons. In temperature range T = 2 - 20 K for overlapping Landau levels, the inelastic scattering rate 1/τin is found to be proportional to T2, indicating dominant contribution of the electron-electron interaction to the inelastic electron relaxation. At strong magnetic field, at which Landau levels are well separated, the inelastic scattering rate is proportional to T3 at high temperatures. We suggest the electron-phonon scattering as the dominant mechanism of the inelastic electron relaxation in this regime. We describe a “surface-protected etching” strategy that allows convenient conversion of sol−gel derived silica into porous structures. Poly(vinyl pyrrolidone) is used to protect the near surface layer, and NaOH is used to selectively etch the interior of the silica spheres. Etching initially yields porous structures and eventually removes the core to leave behind hollow silica spheres with porous shells. This strategy is useful for constructing core−shell systems where active nanomaterials are embedded in silica shell for enhanced stability against aggregation. We experimentally demonstrate use of the surface-protected etching approach to create openings on silica shells; these openings allow dissolved chemical species to reach embedded catalytic particles to be chemically transformed while the porous shells continue to act as effective barriers against aggregation and loss of activity of the core particles. Here, we established a system for displaying heterologous protein to the C terminus of the peptidoglycan- binding domain (CA domain) of AcmA (a major autolysin from Lactococcus lactis). Western blot and flow cytometric analyses revealed that the fusion proteins (cA-AmyA) of the cA domain and α-amylase from Streptococcus bovis 148 (AmyA) are efficiently expressed and successfully displayed on the surfaces of L. lactis cells. AmyA was also displayed on the cell surface while retaining its activity. Moreover, with an increase in the number of cA domains, the quantity of cA-AmyA fusion proteins displayed on the cell surface increased. When three repeats of the cA domain were used as an anchor protein, 82% of α-amylase activity was detected on the cells. The raw starch-degrading activity of AmyA was significantly higher when AmyA was fused to the C terminus of the cA domain than when it was fused to the N terminus. Nearly monodisperse PbWO4and CaWO4microspheres have been synthesized in large scale by a surfactant-assisted solution route, in which either sodium dodecyl benzenesulfonate (SDS) or cetyltrimethyl ammonium bromide (CTAB) is used. By controlling the solution reaction conditions, such as temperature, surfactant, and pH value, we can synthesize nearly monodisperse microspheres of tetragonal tungstate (MWO4, M Pb, Ca) composed of subunits with different shapes. The diameters of these microspheres have been found to be sensitive to the reaction conditions and could be tuned by controlling the reaction conditions. The growth process of these nearly monodisperse microspheres has been examined. The optical properties of the tungstate monodisperse microspheres in the temperature range of 20−270 K were studied. A balanced bandpass filter with high common-mode (CM) suppression using isosceles right-angled triangular (RAT) resonator is proposed in this Letter. The two high order orthogonal modes (TM210Z and TM200Z) are generated in the two overlapped isosceles RAT resonators under differential-mode (DM) and CM excitation, respectively. In this structure, TM210Z mode will be transmitted and TM200Z mode will be blocked. Furthermore, four via-holes are introduced to disturb TM210Z mode while TM200 mode is unaffected, in order to shift the DM passband to the position of CM suppression. The designed balanced bandpass filter operating at 6.12 GHz with 3 dB fractional bandwidth of 5% is fabricated and measured. Objectives: To investigate the efficacy and postoperative complications of lattice carbon dioxide laser in the treatment of postmenopausal patients with mild to moderate stress urinary incontinence. Methods: A total of 30 postmenopausal female patients with mild to moderate stress urinary incontinence, recruited from the Affiliated Hospital of Hebei University from September to November 2019, were selected as the study subjects and treated with lattice carbon dioxide laser therapy. Treatment was given at intervals of one month. The degree of urinary incontinence, the urinary incontinence questionnaire (ICI-Q-SF) score, and the urinary incontinence quality of life scale (I-QOL)) Score, surgical satisfaction, one hour pad test and postoperative complications before treatment and after each treatment of all patients were respectively recorded and compared. Results: Compared with those before treatment, the grade of urinary incontinence and ICI-Q-SF scores of these 30 patients after each treatment were lower, and their I-QOL scores were higher. The difference of one hour urine pad test was statistically significant (P<0.05), but the follow-up data of three months after the third treatment was close to that of one month after the first treatment. The satisfaction rate of these 30 patients was 76.67% (23/30). After treatment, only one patient presented vaginal itching discomfort on the first day after surgery and the symptoms disappeared three days later. No obvious complications occurred in the other 29 patients.
