Safеd musli, also known as Chlorophytum borivilianum , is gaining recognition in thе hеrbal industry due to its rich phytochеmical contеnt, extracted from the rhizome. In an attempt to produce these phytochemicals via an alternative method, optimization of proliferation of safed musli hairy roots were conducted. The primary objective was to identify and optimize the conditions that promote the growth of hairy roots in Safеd musli such as the type and concentrations of auxin, carbon sourcеs, and dark-light regimes. Thе hairy roots wеrе cultured in mеdia with varying concеntrations of IBA and NAA (ranging from 0 to 0.8 mg/L), both individually and in combinations. Thе combination of 0.8 mg/L IBA and 0.6 mg/L NAA proved to be most еffеctivе resulting in 100 % branching within 12 days. This combination yiеldеd 25.2 branchеs with an avеragе root length of 16.23 cm, whereas in control no branching occurred. Following this, carbon sources (sucrose and fructose) under light and dark conditions were studied. Significant enhancement on proliferation of hairy roots was obtained in treatment with 3 % sucrose giving a fresh weight of 3.68 g, root length of 242.4 cm, root diameter of 0.82 mm, and higher surface area (49.7 cm2) under light (16 h, 35 μmol m−2 s−1). This study on optimization of auxin, carbon sources and light-dark regimes has demonstrated the potential for enhancing hairy root proliferation, offering a robust and efficient method to produce bioactive compounds which can be upscaled for industrial bioreactor production. Ethylene, the simplest gaseous plant growth regulator (PGR), controls diverse physiological pathways in plants. Under various stress conditions and during different developmental stages, such as root elongation, leaf and flower senescence, seed germination, tissue differentiation and organ abscission, ethylene biosynthesis is significantly increased. In rice, the internal ethylene concentration is rapidly regulated to a genetically, physiologically and morphologically relevant level under various stresses. Regulation of the ethylene signalling pathway under adverse conditions results in up- and/or down-regulation of the expression of stress-related genes in different families. Transcription factors are proteins that influence and control a number of biological processes under both normal and stress conditions. APETALA 2/ethylene response factor (AP2/ERF) is a transcription factor that is considered to function in stress response pathways in rice. To date, many AP2/ERF genes have been functionally characterised in rice. An understanding of the interactions between the AP2/ERF genes and ethylene-dependent mechanisms may provide new insights to facilitate the enhanced adaptation of rice to stress. In the current review, the structure and function of ethylene in rice under normal and stress conditions are described, and then the general functions of the plant AP2/ERF transcription factors are discussed. In addition, the interactions between the AP2/ERF genes and ethylene pathways under abiotic stresses, including submergence, cold, salinity, drought and heavy metal stresses, as well as those under biotic stresses, are summarised. Although the AP2/ERF genes have been identified, information on the physiological mechanisms of this gene family under stress conditions in rice remains limited. Seed priming is a useful technique which improves seed germination parameters. The present investigation was conducted to evaluate some of the most important germination parameters such as total germination, germination rate, mean germination time, average speed, germination percentage (GP), germination vigor index (GVI), relative frequency, plumule fresh and dry weight, radical fresh and dry weight, plumule and radical length, leaf and root proline content of six indica rice (Oryza sativa L.) varieties, including MR219, MR219-4, MR219-9, MR220, MR159, and MR 211 under hydro-, hormonal-and polyethylen glycol (PEG)-priming conditions. The highest germination parameters were achieved under the hydro-priming treatment, at 18 h (100%), 6 h (100%), 18 h (90.3%), 12 h (91.6%), 18 h (86.6%), and 18 h (78.3%) for the genotypes MR219, MR219-4, MR219-9, MR220, MR159, and MR211, respectively. The best germination feedback of the rice varieties under the hormonal priming were observed in 50 mg L-1 abscisic acid (ABA), 10 mg L-1 gibberellic acid-3 (GA3), 50 mg L-1 indole-3-acetic acid (IAA), 50 mg L-1 GA3, 100 mg L-1 ABA, and 10 mg L-1 GA3 for the genotypes MR219, MR219-4, MR219-9, MR220, MR159 and MR211, respectively. The rice varieties showed different responses to various levels of PEG in which MR219, MR219-4 and MR219-9 responded positively to higher PEG levels, while MR220, MR159 and MR211 showed better feedback under lower PEG concentrations. The protoplast system is robust and versatile for plant genetic engineering works. There are various ways to deliver nucleic acid materials into the protoplasts, but polyethylene glycol (PEG)-mediated method offers minimal damage compared to others. Therefore, an established PEG-mediated transformation protocol with optimised parameters is needed to improve the system's reproducibility and reliability. This study utilised oil palm mesophyll protoplasts as transformation cells and the red fluorescent protein (DsRED) as a visual reporter. Based on the transformation efficiency scored after 72 h, a method with the following parameters: 5 min incubation in 35% PEG, 10 min incubation with 50 µg DNA, and 90 s heat-shock treatment at 42 °C was found to be the best for oil palm protoplast PEG-mediated transfection. A significant improvement in transformation efficiency at about 56% was achieved using this method compared to the previous study (5%). This newly developed protocol was then used to evaluate the efficiency in terms of the relative strength of 7 promoters, including two plant constitutive promoters, three oil palm constitutive promoters, and two oil palm tissue-specific promoters in regulating the expression of the transgene in oil palm. Based on the results from three individual replicates, CaMV35S showed to be the most efficient amongst the tested promoters in driving the transgene expression in oil palm mesophyll protoplasts, followed by oil palm translationally control tumour protein (TCTP) and maize-ubiquitin (Ubi1) promoters. • An efficient protocol for protoplasts isolation from in vitro leaf and mesocarp of oil palm has been established. • Enzymes composition effect on the yield and viability of protoplast are significant. • Scraping the waxy layer on oil palm leaf increased the protoplast isolation efficiency. • Vacuum treatment enhanced the penetration of enzymes into tissues. • 10% ficoll in 0.6 M mannitol improved the separation of protoplasts from impurities. Incubation of oil palm leaf sample with an enzyme mixture of cellulase R-10, macerozyme R-10, driselase, and pectolyase Y-23, for 14 h has successfully produced up to 2.5 × 106 protoplasts g-1 fresh weight (FW)-1 with 95% viability. Incubation of oil palm mesocarp tissue with the optimized enzyme mixture for 2 h at static condition has also successfully produced 3.98 × 106 protoplasts g-1 FW-1 with 85% viability. Besides, it was found that increasing the sample's surface area in contact with enzyme solution by slicing the samples into narrow strips and thin layers has improved the penetration of enzymes into the tissues and enhanced the isolation efficiency. In addition, a plasmolysis step before enzymatic treatment has also improved the protoplast viability by minimizing the damage incurred during isolation. The successful isolation of protoplast from oil palm leaf and mesocarp has enabled the study of gene function and the characterization of endogenous tissue-specific promoter being carried out in vivo. • First report on the use of Discosoma red fluorescent protein (DsRED) - as visual reporter marker in oil palm cells. • Seven DsRED gene vectors driven by a different promoter were constructed and available for the scientific community. • The expression of DsRED in oil palm cells exhibited better photostability and brightness compared to GFP. Genetic modification of oil palm is being carried out to explore new avenues for improving and expanding the potential of oil palm. Multiple bottlenecks in delivering the DNA into oil palm cells and generating the transgenic oil palm required us to look into an alternative marker which could help us assess the efficiency of the transformation system, moreover at the early stage considering that oil palm is a slow growing plant. In this study, we have constructed seven transformation vectors carrying a gene encoding DsRED, each driven by a different promoter, i.e. plant constitutive promoter, oil palm constitutive promoter or oil palm tissue-specific promoter. The vectors were transformed into oil palm embryogenic calli via microprojectile bombardment and the red fluorescence signals were observed after 48 h. Based on the observation, the construct driven by the CaMV35S promoter produced the highest number of red fluorescent spots. The oil palm calli bombarded with pUbi-DsRED plasmid suffered the most number of red fluorescent spots losses from day three until day 42, as compared to other constructs. The CRISPR/Cas9 genome editing system has been in the spotlight compared to programmable nucleases such as ZFNs and TALENs due to its simplicity, versatility, and high efficiency. CRISPR/Cas9 has revolutionized plant genetic engineering and is broadly used to edit various plants' genomes, including those transformation-recalcitrant species such as oil palm. This review will comprehensively present the CRISPR-Cas9 system's brief history and underlying mechanisms. We then highlighted the establishment of the CRISPR/Cas9 system in plants with an emphasis on the strategies of highly efficient guide RNA design, the establishment of various CRISPR/Cas9 vector systems, approaches of multiplex editing, methods of transformation for stable and transient techniques, available methods for detecting and analyzing mutations, which have been applied and could be adopted for CRISPR/Cas9 genome editing in oil palm. Simple Summary: Phytoremediation is a plant-based approach to extract, stabilise, eliminate, or render pollutants into less harmful form. The study highlights the use of a native polar microalga as a means of phytoremediation in Antarctica where imported microbes are prohibited. Since 1959, Antarctica has been a protected region to preserve its dynamic ecosystems, but it is increasingly vulnerable to climate change and pollution. One of the anthropogenic disturbances in the continent is diesel spillage. Due to the extreme polar environment, natural attenuation of spilled diesel is severely hindered; hence, the problem calls for an effective and sustainable solution. This laboratory study proved that Antarctic microalga was capable of removing diesel (57.6%) through biodegradation and biosorption in the span of nine days. Meanwhile, mixotrophic cultivation triggered the vacuolar activities and potentially stimulated lipid assimilation in the cells. The microalgal-based process offers a cheap alternative in water decontamination while bearing the economic potential through the secretion of valuable products, such as biolipids. Microalgae are well known for their metal sorption capacities, but their potential in the remediation of hydrophobic organic compounds has received little attention in polar regions. We evaluated in the laboratory the ability of an Antarctic microalga to remediate diesel hydrocarbons and also investigated physiological changes consequent upon diesel exposure. Using a polyphasic taxonomic approach, the microalgal isolate, WCY_AQ5_1, originally sampled from Greenwich Island (South Shetland Islands, maritime Antarctica) was identified as Tritostichococcus sp. (OQ225631), a recently erected lineage within the redefined Stichococcus clade. Over a nine-day experimental incubation, 57.6% of diesel (~3.47 g/L) was removed via biosorption and biodegradation, demonstrating the strain's potential for phytoremediation. Fourier transform infrared spectroscopy confirmed the adsorption of oil in accordance with its hydrophobic characteristics. Overall, degradation predominated over sorption of diesel. Chromatographic analysis confirmed that the strain efficiently metabolised medium-chain length n-alkanes (C-7 to C-21), particularly n-heneicosane. Mixotrophic cultivation using diesel as the organic carbon source under a constant light regime altered the car/chl-a ratio and triggered vacuolar activities. A small number of intracellular lipid droplets were observed on the seventh day of cultivation in transmission electron microscopic imaging. Key message: The oil palm EgPAL1 gene promoter and its regulatory region were functional as a promoter in the heterologous system of Arabidopsis according to the cis-acting elements present in that region. Abstract: Phenylalanine ammonia lyase (PAL, EC 4.3.1.24) is the key enzyme of the phenylpropanoid pathway which plays important roles in plant development and adaptation. To date, there is no report on the study of PAL from oil palm ( Elaeis guineensis), an economically important oil crop. In this study, the 5′ regulatory sequence of a highly divergent oil palm PAL gene ( EgPAL1) was isolated and fused with GUS in Arabidopsis to create two transgenic plants carrying the minimal promoter with (2302 bp) and without its regulatory elements (139 bp). The regulatory sequence contained cis-acting elements known to be important for plant development and stress response including the AC-II element for lignin biosynthesis and several stress responsive elements. The promoter and its regulatory region were fully functional in Arabidopsis. Its activities were characterised by two common fundamental features of PAL which are responsive to plant internal developmental programme and external factors. The promoter was developmentally regulated in certain organs; highly active in young organs but less active or inactive in mature organs. The presence of the AC elements and global activity of the EgPAL1 promoter in all organs resembled the property of lignin-related genes. The existence of the MBS element and enhancement of the promoter activity by PEG reflected the behaviour of drought-responsive genes. Our findings provide a platform for evaluating oil palm gene promoters in the heterologous system of Arabidopsis and give insights into the activities of EgPAL1 promoter in oil palm. Some cyanobacteria species have a high capacity for accumulating phycobiliprotein contents in their cells. However, there is a lack of information on screening tropical freshwater cyanobacteria, particularly phycobiliproteins. In addition, it is unclear which characteristics of cyanobacteria (morphological and/or growth) could affect phycobiliprotein contents. This study aimed to screen and characterise Malaysian indigenous freshwater cyanobacteria for the growth, biomass, and pigment contents and determine the major characteristic that contributed to the variation of phycobiliproteins. The surface/volume (S/V) ratio, specific growth rate, biomass productivity, and pigment contents of the isolated cyanobacteria were analysed. Principal component analysis (PCA) and hierarchical cluster analysis (HCA) were applied to distinguish the factor responsible for phycobiliprotein variations in cyanobacteria. For the phycobiliprotein contents, Arthrospira sp., Pseudanabaena sp., and Synechococcus elongatus showed significantly higher (p < 0.05) amounts of cyanobacterial phycocyanin (C-PC), phycoerythrin (C-PE), and allophycocyanin (C-APC), respectively, than other studied cyanobacteria. This study showed no apparent trend of similarity and difference between the unicellular and filamentous cyanobacteria. In addition, carotenoid contents demonstrated a positive correlation with total phycobiliproteins, C-PC and C-APC. Based on the current findings, Arthrospira sp., Pseudanabaena sp. and Synechococcus elongatus might be the promising candidate to be the C-PC, C-PE and C-APC sources, respectively, for commercial production purposes. The selection of optimal cyanobacterial strain is crucial for efficient phycobiliprotein production. This study underlined the potential of freshwater cyanobacteria in producing respective and total phycobiliproteins. Genetic structure and biodiversity of the medicinal plant Ficus deltoidea have rarely been scrutinized. To fill these lacunae, five varieties, consisting of 30 F. deltoidea accessions were collected across the country and studied on the basis of molecular and morphological data. Molecular analysis of the accessions was performed using nine Inter Simple Sequence Repeat (ISSR) markers, seven of which were detected as polymorphic markers. ISSR-based clustering generated four clusters supporting the geographical distribution of the accessions to some extent. The Jaccard's similarity coefficient implied the existence of low diversity (0.50-0.75) in the studied population. STRUCTURE analysis showed a low differentiation among the sampling sites, while a moderate varietal differentiation was unveiled with two main populations of F. deltoidea. Our observations confirmed the occurrence of gene flow among the accessions; however, the highest degree of this genetic interference was related to the three accessions of FDDJ10, FDTT16 and FDKT25. These three accessions may be the genetic intervarietal fusion points of the plant's population. Principal Components Analysis (PCA) relying on quantitative morphological characteristics resulted in two principal components with Eigenvalue >1 which made up 89.96% of the total variation. The cluster analysis performed by the eight quantitative characteristics led to grouping the accessions into four clusters with a Euclidean distance ranged between 0.06 and 1.10. Similarly, a four-cluster dendrogram was generated using qualitative traits. The qualitative characteristics were found to be more discriminating in the cluster and PCA analyses, while ISSRs were more informative on the evolution and genetic structure of the population. Spathoglottis plicata is highly sought after by florists and orchid growers due to its unique structure and colors. However, this orchid is a slow-growing type and thus the planting materials are acutely limited. Therefore, there is a need to improve its multiplication efficiency and speed up its growth in order to meet the market demand. Benzyladenine purine (BAP) has been widely used to improve plantlet multiplication in plant cultures. Polyamines have also been reported to improve the growth and development of in vitro cultures. The objective of this study was to investigate the effects of BAP and different polyamines on the physical and biochemical changes of protocorm-like bodies (PLBs) of S. plicata. Different concentrations of BAP and polyamines (putrescine, spermine, and spermidine) were used. BAP and polyamines were supplemented singly in half-strength MS medium and PLBs were cultured for 2 weeks. It was observed that 5 µM BAP and 25 µM spermidine resulted in the highest fresh weights of 0.38 g and 0.31 g, respectively. The total soluble protein and carbohydrate content for PLBs treated with 25 µM spermidine was 4.85 mg/g FW and 6.36 mg/g FW. A separate experiment was carried out to investigate the interactive effects of 25 µM spermidine with 5 µM BAP. The production of alkaloids in the intact plant is usually low, and some of them are tissue-specific and growth stage–specific. In this study, the total alkaloid content (TAC) in Polyalthia bullata was determined using different extraction solvents, and the extracts were subjected to LC-MS analysis. The manipulation of alkaloid production in P. bullata callus was further carried out by growing the callus on two basal media (Murashige and Skoog (MS) and Woody Plant Medium (WPM)) containing auxins (2,4-dichlorophenoxyacetic acid (2,4-D), α-naphthaleneacetic acid (NAA), 4-amino-3,5,6-trichloropicolinic acid (picloram), 3,6-dichloro-o-anisic acid (dicamba), indole-3-acetic acid (IAA), and indole-3-butyric acid (IBA)), precursors (L-phenylalanine, L-tyrosine, and L-tryptophan), and elicitors (methyl jasmonate (MeJA), salicylic acid (SA), and chitosan). The methanolic extracts exhibited the highest alkaloid content in leaf, stem, and root tissues (4.61 ± 0.04 to 7.71 ± 0.04 mg/g DW), and nine alkaloids were detected in P. bullata. The callus grown on MS + 30 μM 2,4-D exhibited the highest alkaloid accumulation (31.07 ± 0.05 μg/mg DW) after 6 weeks of culture compared to other media: MS media containing auxins, MS media containing auxins and precursors, and MS media containing auxins and elicitors. CRISPR/Cas9 is a powerful genome editing tool applicable to diverse plant species. However, the application of CRISPR/Cas9-ribonucleoproteins (RNPs) in oil palm (Elaeis guineensis) has not yet been reported. In this paper, the specificity, efficiency, and sensitivity of CRISPR/Cas9-RNPs in oil palm were evaluated by assessing the effect of different amounts of Cas9 and heat stress using E. guineensis phytoene desaturase (EgPDS) as the target gene. Seven gRNAs were designed and combined with Cas9 protein to form three combinations of RNPs complexes; RNP1/RNP2, RNP3/RNP4, and RNP5/RNP6/RNP7. Assessment of optimal amount of Cas9 and heat stress using RNP5/RNP6/RNP7 demonstrated that 40 µg of Cas9 and heat treatment at 39ºC increased the genome editing efficiency up to 95% in oil palm protoplasts. The use of both optimal conditions produced targeted mutations at frequencies of up to 63.6% in protoplasts transformed with RNP1/RNP2 and RNP3/RNP4 and up to 100% with RNP5/RNP6/RNP7 combinations. Multiple site mutagenesis at two to three target sites with fragment removal up to 179 bp, 69 bp, and 816 bp occurred at high frequencies producing editing efficiency and a Knockout Score (KO) of 100%. Altogether, this study demonstrates a promising, highly efficient, and precise transgene-free CRISPR/Cas9-RNPs genome editing platform in oil palm that also holds enormous potential for application in other transformation-recalcitrant species. Basal stem rot (BSR) is a detrimental disease in oil palm (Elaeis guineensis) due to the host's lack of symptoms during the early phase of infection. Early detection of BSR at non-symptomatic phase is feasible via molecular-based approaches and has become the major interest among researchers. Our previous study had identified a differentially expressed gene encoding auxin efflux carrier component 5-like (EgPIN5) at 11 day-post-inculation of Ganoderma boninense (symptomless early stage of infection) on oil palm seedlings. We further evaluated the gene expression on samples collected from an oil palm plantation at different BSR stages (symptomless, mild and severe). Our findings revealed that EgPIN5 was expressed in symptomless group but was not expressed in either mild or severe BSR groups. Next, we developed a sandwich-type aptamer-based lateral flow assay (LFA) against EgPIN5 RNA by adapting the versatile DNA-functionalized gold nanoparticles for colorimetric detection. The LFA strip demonstrated visible colorimetric signal of EgPIN5 RNA at 12.5 ng/μL with limit of detection at 2.45 ng/μL using a scanning device. Based on our consistent results, we presented EgPIN5 as a diagnostic biomarker for symptomless BSR infection in oil palm. Background: Hemibiotrophic pathogen such as the fungal pathogen Ganoderma boninense that is destructive to oil palm, manipulates host defense mechanism by strategically switching from biotrophic to necrotrophic phase. Our previous study revealed two distinguishable expression profiles of oil palm genes that formed the basis in deducing biotrophic phase at early interaction which switched to necrotrophic phase at a later stage of infection. Results: The present report is a continuing study from our previous published transcriptomic profiling of oil palm seedlings against G. boninense. We focused on identifying differentially expressed genes (DEGs) encoding transcription factors (TFs) from the same RNA-seq data; resulting in 106 upregulated and 108 downregulated TFs being identified. The DEGs are involved in four established defense-related pathways responsible for cell wall modification, reactive oxygen species (ROS)-mediated signaling, programmed cell death (PCD) and plant innate immunity. We discovered upregulation of JUNGBRUNNEN 1 (EgJUB1) during the fungal biotrophic phase while Ethylene Responsive Factor 113 (EgERF113) demonstrated prominent upregulation when the palm switches to defense against necrotrophic phase. EgJUB1 was shown to have a binding activity to a 19 bp palindromic SNBE1 element, WNNYBTNNNNNNNAMGNHW found in the promoter region of co-expressing EgHSFC-2b. Further in silico analysis of promoter regions revealed co-expression of EgJUB1 with TFs containing SNBE1 element with single nucleotide change at either the 5th or 18th position. Meanwhile, EgERF113 binds to both GCC and DRE/CRT elements promoting plasticity in upregulating the downstream defense-related genes. Both TFs were proven to be nuclear-localized based on subcellular localization experiment using onion epidermal cells. Conclusion: Our findings demonstrated unprecedented transcriptional reprogramming of specific TFs potentially to enable regulation of a specific set of genes during different infection phases of this hemibiotrophic fungal pathogen. The results propose the intricacy of oil palm defense response in orchestrating EgJUB1 during biotrophic and EgERF113 during the subsequent transition to the necrotrophic phase. Binding of EgJUB1 to SNBE motif instead of NACBS while EgERF113 to GCC-box and DRE/CRT motifs is unconventional and not normally associated with pathogen infection. Any forms of valorization of microorganisms would require accurate identity recognition to ensure repeatability, reproducibility and quality assurance. This study aimed to evaluate the effectiveness of different primers for identifying cultured eukaryotic microalgae using a simple 18S rDNA approach. A total of 34 isolated microalgae and one culture collection were utilized in the search for an effective molecular identification method for microalgae. Ammonium formate was applied to marine microalgae prior to DNA extraction. The microalgal DNA was extracted using a commercial kit and subjected directly to PCR amplification using four different published 18S rDNA primers. The DNA sequences were analysed using Basic Local Alignment Search Tool (BLAST) and phylogenetic trees to determine the microalgae identity. The identity was further validated with conventional morphological taxonomic identification, and the relationship of microalgal morphology and genetic materials was also determined. The microalgal DNA was successfully amplified, including marine species without prior cleaning. In addition, the ss5 + ss3 primer pair was found to be an ideal primer set among the tested primers for identifying microalgae. Overall, molecular identification showed relative matching with morphological identification (82.86%). This study is important because it serves as a platform to develop a standardized eukaryotic microalgae identification method. Exposing the skin to solar UV radiation induces cascades of signaling pathways and biological alterations such as redox imbalance, suppression of antioxidant genes and programmed cell death. Therefore, the aim of this study was to use RNA-Seq to unravel the effects of UV radiation on Normal Human Adult Fibroblast cells (NHDF). Cells were exposed to UV (20 mJ/cm2 for 3 mins) and incubated for 24 h. Total mRNA from the cells generated libraries of 72,080,648 and 40,750,939 raw reads from UV-treated and control cells respectively. Of the differentially expressed genes (DEGs) produced 2,007 were up-regulated and 2,791 were down-regulated (fold change ≥2, p < 0.05). The expression of 4 genes was validated with RT-qPCR. Chemokine signaling pathways in cancer were significantly activated and antioxidant genes were down-regulated. Modification of lipid composition in the mesocarp tissue of oil palm involves genetic manipulation of multiple genes. More than one mesocarp-preferential promoter is necessary for the expression of individual transgenes in the same plant to obviate transcriptional gene silencing. This study aimed to identify genes that are preferentially expressed in the mesocarp tissue and characterize selected candidate mesocarp-preferential promoters. Ten transcripts that were preferentially expressed in the mesocarp tissue were identified from the analysis of 82 transcriptome datasets of 12 different oil palm tissues. The expression of two candidate genes, MSP-C1 and MSP-C6 , was verified to be preferentially expressed in the mesocarp tissues and shown to have a low expression level in non-mesocarp tissues by reverse transcription quantitative real-time PCR (RT-qPCR). MSP-C6 promoter fragments of different lengths were transformed into tomato plants for further characterization. Both unripe and ripe fruits of transgenic tomato plants transformed with a construct harboring the MSP-C6-F1 (2014 bp) promoter were shown to have high beta-glucuronidase (GUS) activities. Proteomic analysis was conducted to identify the rice root proteins induced by exogenous proline and their involvement in root growth. Proteins were extracted from the root tissues grown under two conditions, T1 (control) and T2 (10 mM proline), and profiled by two-dimensional polyacrylamide gel electrophoresis. Seventeen of 30 differentially expressed proteins were identified by mass spectrometry. Proline-treated rice roots showed up-regulation and down-regulation of nine and eight proteins, respectively, when compared to those in the control. Among the differentially expressed proteins, the down-regulation of glutathione reductase and peroxidase could be involved in the regulation of cellular hydrogen peroxide and reactive oxygen species levels that modulate the root cell wall structure. Differentially expressed proteins identified as pathogenesis-related proteins might be related to stress adaptive mechanisms in response to exogenous proline treatment. In addition, differentially expressed protein identified as the fructose-bisphosphate aldolases and cytochrome c oxidase might be associated with energy metabolism, which is needed during root developmental process. This is the first attempt to study the changes in rice root proteome treated with proline. Curcuma longa L. is a rhizome plant often used as traditional medicinal preparations in Southeast Asia. The dried powder is commonly known as cure-all herbal medicine with a wider spectrum of pharmaceutical activities. In spite of the widely reported therapeutic applications of C. longa, research on its safety and teratogenic effects on zebrafish embryos and larvae is still limited. Hence, this research aimed to assess the toxicity of C. longa extract on zebrafish. Using a reflux flask, methanol extract of C. longa was extracted and the identification and quantification of total flavonoids were carried out with HPLC. Twelve fertilized embryos were selected to test the embryotoxicity and teratogenicity at different concentration points. The embryos were exposed to the extract in the E3M medium while the control was only exposed to E3M and different developmental endpoints were recorded with the therapeutic index calculated using the ratio of LC50/EC50. C. longa extract was detected to be highly rich in flavonoids with catechin, epicatechin, and naringenin as the 3 most abundant with concentrations of 3,531.34, 688.70, and 523.83μg/mL, respectively. The toxicity effects were discovered to be dose-dependent at dosage above 62.50μg/mL, while, at 125.0μg/mL, mortality of embryos was observed and physical body deformities of larvae were recorded among the hatched embryos at higher concentrations. Teratogenic effect of the extract was severe at higher concentrations producing physical body deformities such as kink tail, bend trunk, and enlarged yolk sac edema. Finally, the therapeutic index (TI) values calculated were approximately the same for different concentration points tested. Overall, the result revealed that plants having therapeutic potential could also pose threats when consumed at higher doses especially on the embryos. Key message: TAAAAT and a novel motif, GCTTCA found in the oil palm stearoyl-ACP desaturase (SAD1) promoter are involved in regulating mesocarp-specific expression.Abstract: Two key fatty acid biosynthetic genes, stearoyl-ACP desaturase (SAD1), and acyl-carrier protein (ACP3) in Elaeis guineensis (oil palm) showed high level of expression during the period of oil synthesis in the mesocarp [12-19 weeks after anthesis (w.a.a.)] and kernel (12-15 w.a.a.). Both genes are expressed in spear leaves at much lower levels and the expression increased by 1.5-fold to 2.5-fold following treatments with ethylene and abscisic acid (ABA). Both SAD1 and ACP3 promoters contain phytohormone-responsive, light-responsive, abiotic factors/wounding-responsive, endosperm specificity and fruit maturation/ripening regulatory motifs. The activities of the full length and six 5′ deletion fragments of the SAD1 promoter were analyzed in transiently transformed oil palm tissues by quantitative β-glucuronidase (GUS) fluorometric assay. The highest SAD1 promoter activity was observed in the mesocarp followed by kernel and the least in the leaves. GUS activity in the D3 deletion construct (− 486 to + 108) was the highest, while the D2 (− 535 to + 108) gave the lowest suggesting the presence of negative cis-acting regulatory element(s) in the deleted − 535 to − 486 (49 bp). It was found that the 49-bp region binds to the nuclear protein extract from mesocarp but not from leaves in electrophoretic mobility shift assay (EMSA). Further fine-tuned analysis of this 49-bp region using truncated DNA led to the identification of GCTTCA as a novel motif in the SAD1 promoter. Interestingly, another known fruit ripening-related motif, LECPLEACS2 (TAAAAT) was found to be required for effective binding of the novel motif to the mesocarp nuclear protein extract. Artemisia vulgaris is one of the important medicinal plant species of the genus Artemisia , which is usually known for its volatile oils. The genus Artemisia has become the subject of great interest due to its chemical and biological diversity as well as the discovery and isolation of promising anti-malarial drug artemisinin. A. vulgaris has a long history in treatment of human ailments by medicinal plants in various parts of the world. This medicinal plant possesses a broad spectrum of therapeutic properties including: anti-malarial, anti-inflammatory, anti-hypertensive, anti-oxidant, anti-tumoral, immunomodulatory, hepatoprotective, anti-spasmodic and anti-septic. These activities are mainly attributed to the presence of various classes of secondary metabolites, including flavonoids, sesquiterpene lactones, coumarins, acetylenes, phenolic acids, organic acids, mono- and sesquiterpenes. Studies related to A. vulgaris morphology, anatomy and phytochemistry has gained a significant interest for better understanding of production and accumulation of therapeutic compounds in this species. Recently, phytochemical and pharmacological investigations have corroborated the therapeutic potential of bioactive compounds of A. vulgaris . These findings provided further evidence for gaining deeper insight into the identification and isolation of novel compounds, which act as alternative sources of anti-malarial drugs in a cost-effective manner. Oil palm is subjected to various diseases including Basal Stem Rot (BSR) which is mainly caused by the basidiomycetes fungi, Ganoderma. However, studies on the fungal infection mechanism especially at molecular level and the biological processes involved are still very limited. Fungal cyclophilin (CYP) has been reported to be involved in fungal pathogenicity but involvement of CYP in the pathogenicity of Ganoderma boninense has not been revealed yet. The main objective of this study is to isolate cDNAs encoding CYP and to profile the expression level of these transcripts during infection stages of G. boninense. In this study, five full-length cDNAs encoding CYP were successfully isolated from G. boninense . They were classified as different family members of CYP because significant differences could be observed on their coding, 5′ or 3′ un-translated regions (UTRs). The expression of these five CYP transcripts in different type of G. boninense tissues and the infecting mycelia tissues was studied using real time quantitative PCR (qPCR). Based on the expression results and analysis, the potential functions of these CYP transcripts were predicted to be involved in the development of fruiting body (GbCYP201) and fungal stress response or pathogenicity (GbCYP203 and GbCYP205). Inflammation and oxidative stress are believed to contribute to the pathology of several chronic diseases including hypercholesterolemia (elevated levels of cholesterol in blood) and atherosclerosis. HMG-CoA reductase inhibitors of plant origin are needed as synthetic drugs, such as statins, which are known to cause adverse effects on the liver and muscles. Amaranthus viridis (A. viridis) has been used from ancient times for its supposedly medically beneficial properties. In the current study, different parts of A. viridis (leaf, stem, and seed) were evaluated for potential anti-HMG-CoA reductase, antioxidant, and anti-inflammatory activities. The putative HMG-CoA reductase inhibitory activity of A. viridis extracts at different concentrations was determined spectrophotometrically by NADPH oxidation, using HMG-CoA as substrate. A. viridis leaf extract revealed the highest HMG-CoA reductase inhibitory effect at about 71%, with noncompetitive inhibition in Lineweaver-Burk plot analysis. The leaf extract showed good inhibition of hydroperoxides, 2,2-diphenyl-1-picrylhydrazyl (DPPH), nitric oxide (NO), and ferric ion radicals in various concentrations. A. viridis leaf extract was proven to be an effective inhibitor of hyaluronidase, lipoxygenase, and xanthine oxidase enzymes. The experimental data suggest that A. viridis leaf extract is a source of potent antioxidant and anti-inflammatory agent and may modulate cholesterol metabolism by inhibition of HMG-CoA reductase. Phytocystatin, a type of protease inhibitor (PI), plays major roles in plant defense mechanisms and has been reported to show antipathogenic properties and plant stress tolerance. Recombinant plant PIs are gaining popularity as potential candidates in engineering of crop protection and in synthesizing medicine. It is therefore crucial to identify PI from novel sources like Curcuma longa as it is more effective in combating against pathogens due to its novelty. In this study, a novel cDNA fragment encoding phytocystatin was isolated using degenerate PCR primers, designed from consensus regions of phytocystatin from other plant species. A full-length cDNA of the phytocystatin gene, designated CypCl, was acquired using 5/3 rapid amplification of cDNA ends method and it has been deposited in NCBI database (accession number KF545954.1). It has a 687 bp long open reading frame (ORF) which encodes 228 amino acids. BLAST result indicated that CypCl is similar to cystatin protease inhibitor from Cucumis sativus with 74% max identity. Sequence analysis showed that CypCl contains most of the motifs found in a cystatin, including a G residue, LARFAV-, QxVxG sequence, PW dipeptide, and SNSL sequence at C-terminal extension. Agricultural sorbents have received attention for their effectiveness in oil removal. In Malaysia, oil palm's empty fruit bunch (EFB) spikelets are an abundant agricultural waste that provides a non-toxic, renewable resource of cellulosic materials. In this study, the effectiveness of EFB spikelets to remove oil spills from seawater pollution in a filter system was investigated and the best optimisation approach for filtering conditions was determined. Experiments for oil spill clean-up were performed using a filter-based oil sorption system with a series of conditions such as temperature, time, packing density, and oil concentration to evaluate sorption capacity, oil and water absorbed efficiency. Fourier transform infrared spectroscopy (FTIR) was used to characterise the physicochemical properties of untreated and treated EFB fibres. Based on one-factor-at -a-time (OFAT) analysis conducted at 160 °C for 30 min on 0.1 g/cm3 of packing density containing 25% diesel, 8.667 mL of oil and 5 mL of water was absorbed. In response surface methodology (RSM), the three parameters of temperature, packing density and diesel concentration were observed as significant. From RSM fitting model analysis, the predicted value obtained for both oil and water absorbed were 8.805 and 5.213 mL, respectively. The experimental RSM values of 9 and 5 mL of oil and water absorbed were obtained. The result demonstrated the validity of the model as the experimental RSM values were close to the RSM model's prediction. As compared to OFAT, the RSM method is more efficient in oil removal. Pesticide treatment dramatically reduces crop loss and enhances agricultural productivity, promoting global food security and economic growth. However, owing to high accrual and persistent tendency, pesticides could create significant ecological consequences when used often. Lately, the perspective has transitioned to implementing biological material, environmentally sustainable, and economical strategies via bioremediation approaches to eradicate pesticides contaminations. Microalgae were regarded as a prominent option for the detoxification of such hazardous contaminants. Sustainable application and remediation strategies of pesticides pollutants in the agriculture system by microalgae from the past studies, and recent advancements were integrated into this review. Bibliometric strategies to enhance the research advancements in pesticide bioremediation by microalgae between 2010 and 2020 were implemented through critical comparative analysis of documents from Scopus and PubMed databases. As a result, this study identified a growing annual research trend from 1994 to 2020 (nScopus > nPubMed). Global production of pesticide remediation by microalgae demonstrated significant contributions from India (23.8%) and China (16.7%). The author's keyword clustering was visualized using bibliometric software (VOSviewer), which revealed the strongest network formed by "microalgae", "bioremediation", "biodegradation", "cyanobacteria", "wastewater", and "pesticide" as significant to the research topic. Hence, this bibliometric review will facilitate the future roadmap for many scholars and authors who were drawing attention to the burgeoning research on bioremediation of pesticides to counteract environmental impacts while maintaining food sustainability. Since the heroic age of Antarctic exploration, the continent has been pressurized by multiple anthropogenic activities, today including research and tourism, which have led to the emergence of phenol pollution. Natural attenuation rates are very slow in this region due to the harsh environmental conditions; hence, biodegradation of phenol using native bacterial strains is recognized as a sustainable remediation approach. The aim of this study was to analyze the effectiveness of phenol degradation by a binary consortium of Antarctic soil bacteria, Arthrobacter sp. strain AQ5-06, and Arthrobacter sp. strain AQ5-15. Phenol degradation by this co-culture was statistically optimized using response surface methodology (RSM) and tolerance of exposure to different heavy metals was investigated under optimized conditions. Analysis of variance of central composite design (CCD) identified temperature as the most significant factor that affects phenol degradation by this consortium, with the optimum temperature ranging from 12.50 to 13.75 °C. This co-culture was able to degrade up to 1.7 g/L of phenol within seven days and tolerated phenol concentration as high as 1.9 g/L. Investigation of heavy metal tolerance revealed phenol biodegradation by this co-culture was completed in the presence of arsenic (As), aluminum (Al), copper (Cu), zinc (Zn), lead (Pb), cobalt (Co), chromium (Cr), and nickel (Ni) at concentrations of 1.0 ppm, but was inhibited by cadmium (Cd), silver (Ag), and mercury (Hg). Imperata cylindrica, often known as cogon grass, is a low-cost and useful sorbent for absorbing oil and optimising processes. The effects of temperature, time, packing density and oil concentration on oil absorption efficiency were investigated and optimised utilising one-factor-at-a-time (OFAT) and response surface methodology (RSM) approaches. Temperature and oil concentration are two important variables in the oil absorption process. Fourier transform infrared spectroscopy (FTIR) and scanning electron microscopy (SEM) analysis were used to characterise cogon grass. After treatment and oil absorption, the FTIR method indicated new formation and deformation of functional groups, while SEM revealed changes in the surface and texture of cogon grass, including a roughened and jagged surface. Validation of the RSM model yielded 93.54% efficiency with 22.45 mL oil absorbed at 128 °C temperature and 36 (v/v)% oil concentration while keeping packing density and time constant at 30 min and 0.20 g/cm3, respectively. This review highlights the background of penguins in Antarctica, the anthropogenic pollution and cases, as well as the toxic effects of diesel, heavy metals and microplastics toward penguins. A bibliometric analysis is also included. Antarctica is a relatively pristine continent that attracts scientists and tourists alike. However, the risk of environmental pollution in Antarctica is increasing with the increase in the number of visitors. Recently, there has been a surge in interest regarding diesel, heavy metals and microplastics pollution. Contamination from these pollutants poses risks to the environment and the health of organisms inhabiting the continent. Penguins are one of the most prominent and widely distributed animals in Antarctica and are at major risk due to pollution. Even on a small scale, the impacts of pollution toward penguin populations are extensive. This review discusses the background of penguins in Antarctica, the anthropogenic pollution and cases, as well as the impacts of diesel, heavy metals and microplastics toxicities on penguins. The trends of the literature for the emerging risks of these pollutants are also reviewed through a bibliometric approach and network mapping analysis. A sum of 27 articles are analyzed on the effects of varying pollutants on penguins in Antarctica from 2000 to 2020 using the VOSviewer bibliometric software, Microsoft Excel and Tableau Public. Research articles collected from the Scopus database are evaluated for the most applicable research themes according to the bibliometric indicators (articles, geography distribution, annual production, integrated subject areas, key source journals and keyword or term interactions). Although bibliometric studies on the present research theme are not frequent, our results are sub-optimal due to the small number of search query matches from the Scopus database. As a result, our findings offer only a fragmentary comprehension of the topics in question. Simple Summary: Microalgae are increasingly recognized as a source of valuable biomass with numerous health benefits. Cleaning of marine microalgal biomass is very crucial for microalgal studies as the salt on the microalgae cells will lead to overestimation of biomass determination. Incomplete washing of salt from microalgae could also interfere with the nutritional analyses. The biomass, especially dry weight, has been utilized for nutritional or compositional evaluation. Although standard methods of marine microalgal dry weight determination are available, these methods did not provide comprehensive details, and the parameters vary among themselves. Without a standard method, a comparison of results among previous studies can be misleading and unreliable. Therefore, the current study aimed to investigate and determine the ideal setting of several parameters in the marine microalgal dry weight determination for laboratory-scale culture. The present findings could assist in developing a standardized protocol to ensure a high quality of biomass for microalgal studies. Microalgal biomass is one of the crucial criteria in microalgal studies. Many reported methods, even the well-established protocol on microalgal dry weight (DW) determination, vary greatly, and reliable comparative assessment amongst published results could be problematic. This study aimed to determine the best condition of critical parameters in marine microalgal DW determination for laboratory-scale culture using four different marine microalgal species. These parameters included the washing process, grades of glass microfiber filter (GMF), GMF pretreatment conditions, washing agent (ammonium formate) concentrations, culture: washing agent ratios (v:v) and washing cycles. GMF grade GF/A with precombustion at 450 °C provided the most satisfactory DW and the highest ash-free dry weight (AFDW)/DW ratio. Furthermore, 0.05 M ammonium formate with 1:2 culture: washing agent ratio and a minimum of two washing cycles appeared to be the best settings of microalgal DW determination. The present treatment increased the AFDW/DW ratio of the four respective microalgae by a minimum of 19%. The findings of this study could serve as a pivotal reference in developing a standardized protocol of marine microalgal DW determination to obtain veracious and reliable marine microalgal DW. Study of the potential of Antarctic microorganisms for use in bioremediation is of increasing interest due to their adaptations to harsh environmental conditions and their metabolic potential in removing a wide variety of organic pollutants at low temperature. In this study, the psychrotolerant bacterium Rhodococcus sp. strain AQ5-07, originally isolated from soil from King George Island (South Shetland Islands, maritime Antarctic), was found to be capable of utilizing phenol as sole carbon and energy source. The bacterium achieved 92.91% degradation of 0.5 g/L phenol under conditions predicted by response surface methodology (RSM) within 84 h at 14.8 °C, pH 7.05, and 0.41 g/L ammonium sulphate. The assembled draft genome sequence (6.75 Mbp) of strain AQ5-07 was obtained through whole genome sequencing (WGS) using the Illumina Hiseq platform. The genome analysis identified a complete gene cluster containing catA, catB, catC, catR, pheR, pheA2, and pheA1. The genome harbours the complete enzyme systems required for phenol and catechol degradation while suggesting phenol degradation occurs via the β-ketoadipate pathway. Enzymatic assay using cell-free crude extract revealed catechol 1,2-dioxygenase activity while no catechol 2,3-dioxygenase activity was detected, supporting this suggestion. The genomic sequence data provide information on gene candidates responsible for phenol and catechol degradation by indigenous Antarctic bacteria and contribute to knowledge of microbial aromatic metabolism and genetic biodiversity in Antarctica.
