Adaptive survey design refers to using targeted procedures to recruit different sampled cases. This technique strives to reduce bias and variance of survey estimates by trying to recruit a larger and more balanced set of respondents. However, it is not well understood how adaptive design can improve data and survey estimates beyond the well-established post-survey adjustment. This paper reports the results of an experiment that evaluated the additional effect of adaptive design to post-survey adjustments. The experiment was conducted in the Detroit Metro Area Communities Study in 2021. We evaluated the adaptive design in five outcomes: 1) response rates, 2) demographic composition of respondents, 3) bias and variance of key survey estimates, 4) changes in significant results of regression models, and 5) costs. Responsive survey design is a technique aimed at improving the efficiency or quality of surveys by using incoming data from the field to make design changes. The technique was pioneered on large national surveys, but the tools can also be applied on the smaller-scale surveys most commonly used by sociologists. We demonstrate responsive survey design in a small-scale, list-based sample survey of students on the topic of sexual misconduct. We investigate the impact of individual incentive levels and a two-phase responsive design with changes to mode of contact as approaches for limiting the potential of nonresponse bias in data from such surveys. Our analyses demonstrate that a two-phase design introducing telephone and face-to-face reminders to complete the survey can produce stronger change in response rates and characteristics of those who respond than higher incentive levels. Responsive survey design (RSD) aims to increase the efficiency of survey data collection via live monitoring of paradata and the introduction of protocol changes when survey errors and increased costs seem imminent. Daily predictions of response propensity for all active sampled cases are among the most important quantities for live monitoring of data collection outcomes, making sound predictions of these propensities essential for the success of RSD. Because it relies on real-time updates of prior beliefs about key design quantities, such as predicted response propensities, RSD stands to benefit from Bayesian approaches. However, empirical evidence of the merits of these approaches is lacking in the literature, and the derivation of informative prior distributions is required for these approaches to be effective. In this paper, we evaluate the ability of two approaches to deriving prior distributions for the coefficients defining daily response propensity models to improve predictions of daily response propensity in a real data collection employing RSD. The first approach involves analyses of historical data from the same survey, and the second approach involves literature review. We find that Bayesian methods based on these two approaches result in higher-quality predictions of response propensity than more standard approaches ignoring prior information. Survey researchers have carefully modified their data collection operations for various reasons, including the rising costs of data collection and the ongoing Coronavirus disease (COVID-19) pandemic, both of which have made in-person interviewing difficult. For large national surveys that require household (HH) screening to determine survey eligibility, cost-efficient screening methods that do not include in-person visits need additional evaluation and testing. A new study, known as the American Family Health Study (AFHS), recently initiated data collection with a national probability sample, using a sequential mixed-mode mail/web protocol for push-to-web US HH screening (targeting persons aged 18–49 years). To better understand optimal approaches for this type of national screening effort, we embedded two randomized experiments in the AFHS data collection. The first tested the use of bilingual respondent materials where mailed invitations to the screener were sent in both English and Spanish to 50 percent of addresses with a high predicted likelihood of having a Spanish speaker and 10 percent of all other addresses. We found that the bilingual approach did not increase the response rate of high-likelihood Spanish-speaking addresses, but consistent with prior work, it increased the proportion of eligible Hispanic respondents identified among completed screeners, especially among addresses predicted to have a high likelihood of having Spanish speakers. The second tested a form of nonresponse follow-up, where a subsample of active sampled HHs that had not yet responded to the screening invitations was sent a priority mailing with a $5 incentive, adding to the $2 incentive provided for all sampled HHs in the initial screening invitation. Survey costs are an understudied area. However, understanding survey costs is critical for making efficient decisions about cost-error trade-offs, as well as to accurately project future costs. In this article, we examine a measure of survey costs—per interview costs—over time in a repeated cross-sectional survey. We examine both measurement issues and variability in costs. The measurement issues relate to the classification of various costs into the appropriate time period. We explore several issues that make this process of classification difficult. A time series analysis is then utilized to examine the trends and seasonality in per interview costs. Under the assumptions of our model, after removing the trend and seasonality components, the remainder is variation in costs. Survey costs are a major driver of survey design decisions and thought to be related to survey errors. Despite their importance, no common language exists for discussing survey costs, nor are there established criteria for identifying which cost metrics are useful for which purposes. Past efforts to study survey costs may have been hampered by the notion that more reporting is better reporting. This article starts by introducing a typology for survey cost metrics defined by the type of cost (estimated, observed in records, and actually incurred), currency versus non-currency measures, and level of aggregation (total, by components, per unit, relative). We also suggest a set of criteria – errors in costs, generalizability, and the degree to which a cost measure is informative about survey error sources – for evaluating the utility of cost metrics. We illustrate the evaluative criteria with the cost metrics. We argue that clearly articulating types of survey costs and resetting these baseline evaluative criteria for the utility of different types of costs will help us expand research in this critical area. Sorting large libraries of cells for improved small molecule secretion is throughput limited. Here, we combine producer/secretor cell libraries with whole-cell biosensors using a microfluidic-based screening workflow. This approach enables a mix-and-match capability using off-the-shelf biosensors through either coencapsulation or pico-injection. We demonstrate the cell type and library agnostic nature of this workflow by utilizing single-guide RNA, transposon, and ethyl-methyl sulfonate mutagenesis libraries across three distinct microbes (Escherichia coli, Saccharomyces cerevisiae, and Yarrowia lipolytica), biosensors from two organisms (E. coli and S. cerevisiae), and three products (triacetic acid lactone, naringenin, and L-DOPA) to identify targets improving production/secretion. This article presents the results of an adaptive design experiment in the recruitment of households and individuals for a two-stage national probability web or mail mixed-mode survey, the American Family Health Study (AFHS). In the screening stage, we based the adaptive design's subgroup differentiation on Esri Tapestry segmentation. We used tailored invitation materials for a subsample where a high proportion of the population was Hispanic and added a paper questionnaire to the initial mailing for a subsample with rural and older families. In the main-survey stage, the adaptive design targeted the households where a member other than the screening respondent was selected for the survey. The adaptations included emailing and/or texting, an additional prepaid incentive, and seeking screening respondents' help to remind the selected individuals. The main research questions are (i) whether the adaptive design improved survey production outcomes and (ii) whether combining adaptive design and postsurvey weighting adjustments improved survey estimates compared to performing postsurvey adjustments alone. Unfortunately, the adaptive designs did not improve the survey production outcomes. We found that the weighted AFHS estimates closely resemble those of a benchmark national face-to-face survey, the National Survey of Family Growth, although the adaptive design did not additionally change survey estimates beyond the weighting adjustments. Nonetheless, our experiment yields useful insights about the implementation of adaptive design in a self-administered mail-recruit web or mail survey. We were able to identify subgroups with potentially lower response rates and distinctive characteristics, but it was challenging to develop effective protocol adaptations for these subgroups under the constraints of the two primary survey modes and the operational budget of the AFHS. In addition, for self-administered within-household selection, it was difficult to obtain contact information from, reach, and recruit selected household members that did not respond to the screening interview. BACKGROUND: Epidemiological studies support the hypothesis that diabetes alters pulmonary responses to air pollutants like ozone (O3). The mechanism(s) underlying these associations and potential links among diabetes, O3, and lung inflammation and remodeling are currently unknown. OBJECTIVES: The goal was to determine whether pulmonary responses to repetitive ozone exposures are exacerbated in murine strains that are hyperglycemic and insulin resistant. METHODS: Normoglycemic and insulin-sensitive C57BL/6J mice; hyperglycemic, but mildly insulin-resistant, KK mice; and hyperglycemic and markedly insulin-resistant KKAy mice were used for ozone exposure studies. All animals were exposed to filtered air (FA) or repetitive ozone (0.5 ppm O3, 4 h/d, for 13 consecutive weekdays). Tissue analysis was performed 24 h following the final exposure. This analysis included bronchoalveolar lavage (BAL) for cell and fluid analysis, and tissue for pathology, immunohistology, mRNA, and hydroxyproline. RESULTS: Following repetitive O3 exposure, higher bronchoalveolar lavage fluid inflammatory cells were observed in all mice (KKAy>KK>C57BL=6), with a notable influx of neutrophils and eosinophils in KK and KKAy mice. Although the lungs of O3-exposed C57BL/6J and KK mice had minimal centriacinar histological changes without fibrosis, the lungs of O3-exposed KKAy mice contained marked epithelial hyperplasia in proximal alveolar ducts and adjacent alveoli with associated centriacinar fibrosis. Fibrosis in O3-exposed KKAy lungs was confirmed with immunohistochemistry, tissue hydroxyproline content, and tissue mRNA expression of fibrosis-associated genes (Ccl11, Il13, and Mmp12). Immunofluorescence staining and confocal microscopy revealed alterations in the structure and composition of the airway and alveolar epithelium in regions of fibrosis. DISCUSSION: Our results demonstrate that in diabetic animal strains repetitive ambient ozone exposure led to early and exaggerated pulmonary inflammation and remodeling. Changes in distal and interstitial airspaces and the activation of Th2 inflammatory and profibrotic pathways in experimental animals provide a preliminary, mechanistic framework to support the emerging epidemiological associations among air pollution, diabetes, and lung disease. Background: Inhalation exposure to biological particulate matter (BioPM) from livestock farms may provoke exacerbations in subjects suffering from allergy and asthma. The aim of this study was to use a murine model of allergic asthma to determine the effect of BioPM derived from goat farm on airway allergic responses. Methods: Fine (<2.5 μm) BioPM was collected from an indoor goat stable. Female BALB/c mice were ovalbumin (OVA) sensitized and challenged with OVA or saline as control. The OVA and saline groups were divided in sub-groups and exposed intranasally to different concentrations (0, 0.9, 3, or 9 μg) of goat farm BioPM. Bronchoalveolar lavage fluid (BALF), blood and lung tissues were collected. Results: In saline-challenged mice, goat farm BioPM induced 1) a dose-dependent increase in neutrophils in BALF and 2) production of macrophage inflammatory protein-3a. In OVA-challenged mice, BioPM induced 1) inflammatory cells in BALF, 2) OVA-specific Immunoglobulin (Ig)G1, 3) airway mucus secretion-specific gene expression. RNAseq analysis of lungs indicates that neutrophil chemotaxis and oxidation-reduction processes were the representative genomic pathways in saline and OVA-challenged mice, respectively. Conclusions: A single exposure to goat farm BioPM enhanced airway inflammation in both saline and OVA-challenged allergic mice, with neutrophilic response as Th17 disorder and eosinophilic response as Th2 disorder indicative of the severity of allergic responses. Identification of the mode of action by which farm PM interacts with airway allergic pathways will be useful to design potential therapeutic approaches. [ABSTRACT FROM AUTHOR]

Ionic liquids show promise for deconstruction of lignocellulosic biomass prior to fermentation. Yet, imidazolium ionic liquids (IILs) can be toxic to microbes even at concentrations present after recovery. Here, we show that dominant overexpression of an Ilt1p homolog (encoded by YlILT1/YALI0C04884) from the IIL-tolerant yeast Yarrowia lipolytica confers an improvement in 1-ethyl-3-methylimidazolium acetate tolerance in Saccharomyces cerevisiae compared to the endogenous Ilt1p (ScILT1/YDR090C). We subsequently enhance tolerance in S. cerevisiae through directed evolution of YlILT1 using growth-based selection, leading to identification of mutants that grow in up to 3.5% v/v ionic liquid. Lastly, we demonstrate that strains expressing YlILT1 variants demonstrate improved growth rate and ethanol production in the presence of residual IIL. This shows that dominant overexpression of a heterologous protein (wild type or evolved) from an IIL-tolerant yeast can increase tolerance in S. cerevisiae at concentrations relevant to bioethanol production from IIL-treated biomass. Medical errors can involve multiple team members. Few curricula are being developed to provide instruction on disclosing medical errors that include simulation training with interprofessional team disclosure. To explore more objective evidence for the value of an educational activity on team disclosure of errors, faculty developed and assessed the effectiveness of a multimodal educational activity for learning team‐based disclosure of a medical error. This study employed a methodological triangulation research design. Participants (N = 458) included students enrolled in academic programs at three separate institutions. The activity allowed students to practice team communication while: (1) discussing a medical error within the team; (2) planning for the disclosure of the error; and (3) conducting the disclosure. Faculty assessed individual student's change in knowledge and, using a rubric, rated the performance of the student teams during a simulation with a standardized family member (SFM). Students had a high level of preexisting knowledge and demonstrated the greatest knowledge gains in questions regarding the approach to disclosure (P < .001) and timing of an apology (P < .001). Both SFMs and individual students rated the team error disclosure behavior highly (rho = 0.54; P < .001). Most participants (more than 80%) felt the activity was worth their time and that they were more comfortable with disclosing a medical error as a result of having completed the activity. This activity for interprofessional simulation of team‐based disclosure of a medical error was effective for teaching students about and how to perform this type of important disclosure. Researchers are interested in the effectiveness of adaptive and responsive survey designs that monitor and respond to data using tailored or targeted interventions. These designs often require adherence to protocols, which can be difficult when surveys allow in-person interviewers flexibility in managing cases. This article describes examples of interviewer noncompliance and compliance in adaptive design experiments that occurred in two United States decennial census tests. The two studies tested adaptive procedures including having interviewers work prioritized cases and substitute face-to-face attempts with telephone calls. When to perform such procedures was communicated to interviewers via case management systems that necessitated twice-daily transmissions of data. Occupational exposure to respirable crystalline silica (cSiO2) has been linked to lupus development. Previous studies in young lupus-prone mice revealed that intranasal cSiO2 exposure triggered autoimmunity, preventable with docosahexaenoic acid (DHA). This study explores cSiO2 and DHA effects in mature lupus-prone adult mice, more representative of cSiO2-exposed worker age. Female NZBWF1 mice (14-week old) were fed control (CON) or DHA-supplemented diets. After two weeks, mice were intranasally instilled saline (VEH) or 1 mg cSiO2 weekly for four weeks. Cohorts were then analyzed 1- and 5-weeks postinstillation for lung inflammation, cell counts, chemokines, histopathology, B- and T-cell infiltration, autoantibodies, and gene signatures, with results correlated to autoimmune glomerulonephritis onset. VEH/CON mice showed no pathology. cSiO2/CON mice displayed significant ectopic lymphoid tissue formation in lungs at 1 week, increasing by 5 weeks. cSiO2/CON lungs exhibited elevated cellularity, chemokines, CD3+ T-cells, CD45R + B-cells, IgG + plasma cells, gene expression, IgG autoantibodies, and glomerular hypertrophy. DHA supplementation mitigated all these effects. The mature adult NZBWF1 mouse used here represents a life-stage coincident with immunological tolerance breach and one that more appropriately represents the age (20–30 yr) of cSiO2-exposed workers. cSiO2-induced robust pulmonary inflammation, autoantibody responses, and glomerulonephritis in mature adult mice, surpassing effects observed previously in young adults. DHA at a human-equivalent dosage effectively countered cSiO2-induced inflammation/autoimmunity in mature mice, mirroring protective effects in young mice. These results highlight life-stage significance in this preclinical lupus model and underscore omega-3 fatty acids' therapeutic potential against toxicant-triggered autoimmune responses. Evolutionary approaches to strain engineering inherently require the identification of suitable selection techniques for the product and phenotype of interest. In this work, we undertake a comparative analysis of two related but functionally distinct methods of high-throughput screening: traditional single cell fluorescence activated cell sorting (single cell FACS) and microdroplet-enabled FACS (droplet FACS) using water/oil/water (w/o/w) emulsions. To do so, we first engineer and evolve the non-conventional yeast Yarrowia lipolytica for high extracellular production of riboflavin (vitamin B2), an innately fluorescent product. Following mutagenesis and adaptive evolution, a direct parity-matched comparison of these two selection strategies was conducted. Both single cell FACS and droplet FACS led to significant increases in total riboflavin titer (32 and 54 fold relative to the parental PO1f strain, respectively). However, single cell FACS favored intracellular riboflavin accumulation (with only 70% of total riboflavin secreted) compared with droplet FACS that favored extracellular product accumulation (with 90% of total riboflavin secreted). We find that for the test case of riboflavin, the extent of secretion and total production were highly correlated. The resulting differences in production modes and levels clearly demonstrate the significant impact that selection approaches can exert on final evolutionary outcomes in strain engineering. Moreover, we note that these results provide a cautionary tale when intracellular read-outs of product concentration (including signals from biosensors) are used as surrogates for total production of potentially secreted products. In this article, we investigate the relationship between interviewer travel behavior and field outcomes, such as contact rates, response rates, and contact attempts in two studies, the National Survey of Family Growth and the Health and Retirement Study. Using call record paradata that have been aggregated to interviewer-day levels, we examine two important cost drivers as measures of interviewer travel behavior: the distance that interviewers travel to segments and the number of segments visited on an interviewer-day. We explore several predictors of these measures of travel -- the geographic size of the sampled areas, measures of urbanicity, and other sample and interviewer characteristics. We also explore the relationship between travel and field outcomes, such as the number of contact attempts made and response rates.Wefind that the number of segments that are visited on each interviewer-day has a strong association with field outcomes, but the number of miles travelled does not. These findings suggest that survey organizations should routinely monitor the number of segments that interviewers visit, and that more direct measurement of interviewer travel behavior is needed. The timing and nutritional value of a first meal is important for spiderlings, but little is known about what spiderlings specifically consume. For wandering spiders, nectar feeding is a common occurrence thought to be directly beneficial in providing nutrients and serving to fuel energy costly for foraging. Cannibalism is also prevalent among many spiders. We suspect in spiderlings of the cursorial species, Oxyopes salticus Hentz, 1845, nectar feeding could decrease cannibalism by causing satiation, or increase cannibalism by enhancing energy levels and rates of interaction. We conducted laboratory experiments to test the longevity of newly hatched O. salticus in the presence of different nutrient resources and the effect these resources had on cannibalism rates. Spiderlings were housed solitarily or in pairs and given access to different nutrient resources that reflect those available in the wild, including nectar and insect prey items (fructose, protein, and water). In a 14-day period, we recorded the number of days each spiderling was alive to determine survivorship. By the end of the experiment, 60% of spiderlings housed with fructose were still alive and 10% survived when housed with protein or water. Based on survivorship models, the predicted mean age at death differed between treatments (15.9 days for fructose, 11.2 days for protein, and 9.5 days for water). Spiderlings housed in pairs declined more rapidly in survivorship compared to solitary spiderlings, suggesting cannibalism occurred across all treatments. Fructose significantly increased longevity of spiderlings regardless of their housing and reduced cannibalism. [ABSTRACT FROM AUTHOR]

Nonresponse rates have been growing over time leading to concerns about survey data quality. Adaptive designs seek to allocate scarce resources by targeting specific subsets of sampled units for additional effort or a different recruitment protocol. In order to be effective in reducing nonresponse, the identified subsets of the sample need two key features: 1) their probabilities of response can be impacted by changing design features, and 2) once they have responded, this can have an impact on estimates after adjustment. The National Agricultural Statistics Service (NASS) is investigating the use of adaptive design techniques in the Crops Acreage, Production, and Stocks Survey (Crops APS). The Crops APS is a survey of establishments which vary in size and, hence, in their potential impact on estimates. In order to identify subgroups for targeted designs, we conducted a simulation study that used Census of Agriculture (COA) data as proxies for similar survey items. In push-to-web surveys that use postal mail to contact sampled cases, participation is contingent on the mail being opened and the survey invitations being delivered. The design of the mailings is crucial to the success of the survey. We address the question of how to design invitation mailings that can grab potential respondents' attention and sway them to be interested in the survey in a short window of time. In the household screening stage of a national survey, the American Family Health Study, we experimentally tested three mailing design techniques for recruiting respondents: (1) a visible cash incentive in the initial mailing, (2) a second incentive for initial nonrespondents, and (3) use of Priority Mail in the nonresponse follow-up mailing. We evaluated the three techniques' overall effects on response rates as well as how they differentially attracted respondents with different characteristics. We found that all three techniques were useful in increasing the screening response rates, but there was little evidence that they had differential effects on sample subgroups that could help to reduce nonresponse biases. In the evolution of sexual reproduction, the development of asymmetrical gametes (anisogamy) often leads to the characterisation that males invest less in reproduction than females. This view is founded on the fact that male investment in individual gametes (sperm) and in gonadal tissue is often less than that of females. While it may be true that within a single mating interaction the relative reproductive investment by males and females differs, over their respective lifetimes an evolutionary view predicts that total lifetime reproductive investment should be the same for both sexes. We tested this prediction by measuring total reproductive investment in reproducing adult male and female seed beetles. Since seed beetle adults are facultatively aphagous, not needing to feed or drink as adults, we could track the daily loss of biomass in the reproducing adults as a proxy for total lifetime investment in maintenance and reproduction. By the end of their lifetime there was not a significant difference in total reproductive effort between males and females. Both sexes invested 70 ± 1% SE of their initial body mass in maintenance and reproduction. Altering sex ratios, creating male-biased or female-biased treatments, significantly influenced total investment between the sexes, but not in the manner predicted. Ours is one of the few studies that have measured total adult lifetime reproductive investment in an organism, and our results support the theoretical expectation of equal total reproductive investment between the sexes. Over the course of six years and more than two dozen meetings, members of the Presidential Commission for the Study of Bioethical Issues learned so many things: about emerging science; technological challenges; citizen engagement; the public's, experts', and our own understandings and misperceptions; and even the nature of our own most cherished values. Our commission's commitment to democratic deliberation began deliberatively, when we decided (in the summer of 2010) upon basic principles to guide our first report. At the time, Craig Venter had just announced that he had used techniques of synthetic biology to 'create life,' albeit the most elementary form of life, from nonliving building blocks. Headlines alternated between applauding Venter for creating life and accusing him of playing God. It was one of those all-too-frequent instances where sound-bite democracy was headed toward shedding more heat and fear than light and understanding. The commission was asked to take synthetic biology under review and make recommendations to President Obama on how to proceed. Mixed-mode surveys need to determine a number of design parameters that may have a strong influence on costs and errors. In a sequential mixed-mode design with web followed by telephone, one of these decisions is when to switch modes. The web mode is relatively inexpensive but produces lower response rates. The telephone mode complements the web mode in that it is relatively expensive but produces higher response rates. Among the potential negative consequences, delaying the switch from web to telephone may lead to lower response rates if the effectiveness of the prenotification contact materials is reduced by longer time lags, or if the additional e-mail reminders to complete the web survey annoy the sampled person. On the positive side, delaying the switch may decrease the costs of the survey. We evaluate these costs and errors by experimentally testing four different timings (1, 2, 3, or 4 weeks) for the mode switch in a web–telephone survey. This experiment was conducted on the fourth wave of a longitudinal study of the mental health of soldiers in the U.S. Army. We find that the different timings of the switch in the range of 1–4 weeks do not produce differences in final response rates or key estimates but longer delays before switching do lead to lower costs. In an ideal world, transportation networks and services would be adapted to the specific travel needs of each individual and would perfectly fit the corresponding desire lines (direct lines between origin and destination points). However, in practice, networks cannot be designed to accommodate each individual trip. Still, it is possible to optimize transportation systems from a collective demand point of view. To move from an individual to a collective scale, individual demands need to be encapsulated into demand corridors. Although current spatial tools and data mining techniques are able to identify corridors from numerous movements by using linear or non-linear trajectory data, their limitations—from a transportation point of view—include the use of non-intuitive parameters and the application of some aggregation processes that make it difficult to retrace the attributes of individual input data that could benefit the richness of the available data after processing. For that reason, we propose a new algorithm called Trajectory Clustering for Desire Lines (TraClus-DL), which can identify corridors from Origin-Destination (OD) information with simple parameters, such as spatial location, angles between lines, and sampling weights. The functionality of TraClus-DL as a diagnostic tool for transportation supply was assessed and tested using data from the 2013 OD travel survey conducted in the Montreal area. The sensitivity of the results, with respect to parameter settings, was evaluated, and a comparison with an existing algorithm was proposed. The results of this study demonstrate that transportation specialists can benefit from the convenience of using TraClus-DL as a corridor identification tool, which includes its potential to perform deep analyses at the corridor level. In addition, this study provides new insights into the possible uses of demand corridors as relevant tools for transportation planning, and in the decision-making processes in which a neutral reference is needed to evaluate how much the transportation supply differs from the collective travel demand.
