It is known that air pollution is harmful to creatures, though until now most of the human thermal comfort indices that existed were calculated only with meteorological conditions. Therefore, a new index - meteorology and environment comfort (MEC) - was given out in this paper that considers both meteorology and air pollution conditions and presents the comprehensive and synergistic effects of meteorological and air pollution. The meteorology and air pollution data were used to establish the influence function of the five air pollutants (PM 2.5 , PM 10 , O 3 , NO 2 , and SO 2) according to Fechner's law; then, we calculated the somatosensory temperature (ST, a class of human thermal comfort indices) and MEC values of five typical cities (Beijing, Xining, Nanjing, Kunming, and Guangzhou). The results showed average improvements of five cities on MEC as a new comprehensive human comfort index to new ST. In spring, the MEC comfort proportion fell by 29.25%. Besides, the extreme heat discomfort ratio in Nanjing and Kunming has increased over 20%. In summer, the comfort proportion fell 12.54%; the extreme heat discomfort proportion of Beijing increased 37.86% and Kunming increased 24.09%. Air pollution significantly raised discomfort stress in Beijing. In fall, the comfort proportion fell by 20.87%; and the extreme heat discomfort of Nanjing increased 23.67% caused by poor air quality. About winter, the comfort ratio decreased 12.72%, and the cold discomfort proportion of Nanjing increased 30.30%, signifying awful air quality in winter. Air pollution levels significantly affect the comfort levels in all seasons, which is more evident with good weather patterns. MEC can offer early warnings of extreme weather events and provide a basis for the better prevention and control of air pollution to protect human health basing on the predictions of meteorological and environmental impact factors. Organic small‐molecule fluorophores, characterized by flexible chemical structure and adjustable optical performance, have shown tremendous potential in biosensing. However, classical organic fluorophore motifs feature large overlap between excitation and emission spectra, leading to the requirement of advanced optical set up to filter desired signal, which limits their application in scenarios with simple settings. Here, a series of wavelength‐tunable small‐molecule fluorescent dyes (PTs) bearing simple organic moieties have been developed, which exhibit Stokes shift up to 262 nm, molar extinction coefficients ranged 30,000–100,000 M−1 cm−1, with quantum yields up to 54.8 %. Telocytes (TCs) are distinctive interstitial cells due to their characteristic structures and heterogeneity. They are suggested to participate in tissue repair/regeneration. TCs have been identified in many organs of various mammals. However, data on TCs in lower animals are still very limited. In this work, TCs were identified in the myocardium of the bullfrog (Rana catesbeiana) by light and transmission electron microscopy (TEM). The structural relationships between TCs and neighbouring cell types were measured using the ImageJ (FiJi) morphometric software. TCs with slender Tps (telepodes) were located around cardiomyocytes (CMC). TEM revealed TCs with long Tps in the stroma between CMC. The homocellular tight junctions were observed between the Tps. The Tps were also very close to the neighbouring CMC. The distance between Tps and CMC was 0.15 ± 0.08 μm. Notably, Tps were observed to adhere to the periphery of the satellite cells. The Tps and the satellite cells established heterocellular structural connections by tight junctions. Additionally, Tps were frequently observed in close proximity to mast cells (MCs). The distance between the Tps and the MCs was 0.19 ± 0.09 μm. These results confirmed that TCs are present in the myocardium of the bullfrog, and that TCs established structural relationships with neighbouring cell types, including satellite cells and MCs. Over the last 30 years, there have been several methods developed for the synthesis of oligosaccharides, such as combinatorial synthesis, programmable one‐pot glycosylations, pre‐activation‐based chemo‐selective glycosylation strategy, random glycosylation, chemo‐enzymatic processes, and automated platforms. Distinguishing hydroxyl groups in carbohydrate monomers form the crux of oligosaccharides synthesis. The protection of hydroxyl groups stops glycosylation at undesired positions on carbohydrates, but additional steps are required to install and remove the protecting groups. The targeted functionalization of carbohydrates relies mostly on the protection of hydroxyl groups that are not supposed to undergo glycosylation and to leave the hydroxyl group unprotected where the glycosylation should occur. Numerous procedures to avoid or decrease protecting group manipulations have been developed for the synthesis of a single or an oligosaccharide library, such as glycosylation on unprotected or partially protected acceptors, and random glycosylation on unprotected acceptors. In this review, overall approaches for glycosylation on unprotected or partially protected acceptors in the synthesis of oligosaccharides and oligosaccharide libraries are summarized, the glycosylation on unprotected or partially protected carbohydrate acceptors with or without tin or boron mediation, and random glycosylation on unprotected or partially protected acceptors in solution phase or on solid phase are discussed. Fondaparinux (trade name Arixtra) is a safe and efficacious anticoagulant, and it is chemically related to low-molecular-weight heparins such as enoxaparin. Fondaparinux is a synthetic pentasaccharide, and its synthesis is difficult and expensive. The high cost of fondaparinux thwarts its extensive worldwide usage. Over the last two decades, several research groups and pharmaceutical companies have been interested in finding efficient and practical methods for its synthesis. In mammals, oxidative stress-induced apoptosis of granulosa cells is one of the major causes of follicular atresia, affecting ovarian physiological function. Protegrin-1 (PG-1) is an antimicrobial peptide with effective antimicrobial activity, immunomodulatory function, and porcine growth-promoting effects. PG-1 has been detected in porcine ovaries follicles. This study aimed to investigate the effect of PG-1 on oxidative stress-induced apoptosis of porcine ovarian granulosa cells and the underlying molecular mechanism. Granulosa cells were obtained from porcine follicles and treated with H 2 O 2 to establish the oxidative stress model, and then treated with or without PG-1 (10 μg/mL). PG-1 significantly suppressed H 2 O 2 -induced apoptosis in granulosa cells after 24 h of treatment. Furthermore, these results revealed that PG-1 increased the mRNA and protein expression of anti-apoptotic B cell lymphoma/leukemia 2 (BCL2) and the BCL2/Bcl-2-associated X protein (BAX) ratio while decreasing the expression of pro-apoptotic BAX and active caspase-3. Using Western blot analysis, it was found that PG-1 decreased the phosphorylation of RNA-like endoplasmic reticulum kinase (PERK) and the α-subunit of eukaryotic initiation factor 2 (eIF2α) as well as the protein expression level of CCAAT enhancer-binding protein homologous protein (CHOP), all of which were increased by H 2 O 2. Moreover, inhibitors against PERK and phospho-eIF2ɑ both suppressed the H 2 O 2 -induced granulosa cells apoptosis and enhanced the anti-apoptosis effect of PG-1. Taken together, our findings demonstrated that PG-1 inhibited porcine ovarian granulosa cell apoptosis from oxidative stress via the PERK/eIF2α/CHOP signaling pathway in vitro , which suggests the novel regulatory function of the antimicrobial peptide in the ovary. • PG-1 significantly inhibited H 2 O 2 -induced apoptosis in porcine granulosa cells. • PG-1 significantly suppressed PERK/eIF2α/CHOP signaling pathway which was activated by H 2 O 2. • Inhibition of PERK/eIF2α signaling pathway enhanced the anti-apoptosis effect of PG-1 on porcine granulosa cells. Background: Exosomes, internal proteins, lipids, and nucleic acids coated by phospholipid bilayer membranes, are one type of small extracellular vesicles, which can mediate cell-cell communication. In recent years, exosomes have gained considerable scientific interest due to their widely applied prospect in the diagnosis and therapeutics of human and animal diseases. In this study, we describe for the first time a feasible method designed to isolate and characterize exosomes from feline plasma, urine and adipose-derived mesenchymal stem cells. Results: Exosomes from feline plasma, urine and adipose-derived mesenchymal stem cells were successfully isolated by differential centrifugation. Quantification and sizing of exosomes were assessed by transmission electron microscopy, flow nano analysis and western blotting. Detected particles showed the normal size (30–100 nm) and morphology described for exosomes, as well as presence of the transmembrane protein (TSG101, CD9, CD63, and CD81) known as exosomal marker. Conclusions: The results suggest that differential centrifugation is a feasible method for isolation of exosomes from different types of feline samples. Altrenogest (ALT), a synthetic progestogen, serves a critical role in estrus synchronization among animals like gilts and mares. However, its practical application in animal husbandry is hampered due to its poor solubility and limited oral bioavailability. To address this challenge, a solvent evaporation method was employed to create an inclusion complex of ALT with hydroxypropyl-β-cyclodextrin (ALT/HP-β-CD). The formation of this inclusion complex was confirmed by scanning electron microscopy, power X-ray diffraction, differential scanning calorimetry, Fourier transform infrared spectroscopy, nuclear magnetic resonance spectroscopy, and docking calculations. In addition, we further conducted pharmacokinetic investigation involving gilts, comparing ALT/HP-β-CD inclusion complex to an ALT oral solution. The physicochemical characterization results unveiled a transformation of ALT's crystal morphology into an amorphous state, with ALT effectively entering the cavity of HP-β-CD. Compared with ALT, the solubility of ALT/HP-β-CD inclusion complex increased by 1026.51-fold, and its dissolution rate demonstrated significant improvement. Pharmacokinetic assessments further revealed that the oral bioavailability of ALT/HP-β-CD inclusion complex surpassed that of the ALT oral solution, with a relative bioavailability of 114.08 %. Protegrin-1 (PG1) is an antimicrobial peptide (AMP) that has garnered increasing attention due to its potent immune defense activity. Our previous studies demonstrated the ability of PG1 to enhance proliferation and inhibit apoptosis of porcine granulosa cells (GCs) under oxidative stress. GCs play a crucial role in ovary follicular development. However, the specific function and underlying mechanisms of AMP in follicular development still need further elucidation. The present study aimed to comprehensively explore the biological effects of PG1 on porcine GCs using transcriptome profiling by RNA sequencing technology. Isolated GCs were incubated with or without PG1 for 24 h and transcriptome-wide analysis was exerted to identify differentially expressed genes (DEGs). The results of expression analysis revealed 1,235 DEGs, including 242 up-regulated genes and 993 down-regulated genes (|log2 (FoldChange)| > 1; adjusted P -value < 0.05). The expression levels of 7 selected DEGs were validated by quantitative reverse transcription-polymerase chain reaction (RT-qPCR) analysis, which was consistent with the RNA-sequencing data. Among the significant DEGs, several genes associated with GC function and ovarian follicle development were identified, such as estrogen receptor 2 (ESR2), growth and differentiation factor 6 (GDF6), cell division cycle 20 homolog (CDC20), Notch3 , ephrin and Eph receptor system, Egl nine homolog 3 (EGLN3), and BCL2 like 14 (BCL2L14). Gene Ontology (GO) analysis revealed that the top three significant GO terms were inflammatory response, defense response, and granulocyte migration. Additionally, Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis presented that DEGs were mainly enriched in the immune system, infectious disease, signaling molecules and interaction, and immune disease. Furthermore, Ingenuity Pathway Analysis (IPA) predicted that the top activated pathway was Liver X Receptor (LXR)/ Retinoid X Receptor (RXR) Activation which is known to be associated with female reproduction. Predicted protein–protein interactions (PPIs) analysis identified complement C3 (C3) as the top node with the highest degree of network connection and revealed that DEGs in the sub-networks were involved in cytokine-cytokine receptor interaction, neuroactive ligand-receptor interaction, chemokine signaling pathway, and metabolic process. In conclusion, this study expanded the understanding of the effects of PG1 on porcine GCs at the transcriptomic level and provided a theoretical basis for further investigation into the role of PG1 in immune defense and mammalian ovarian follicular development. Background: Mesenchymal stem cells (MSCs) have generated a great amount of interest in recent years as a novel therapeutic application for improving the quality of pet life and helping them free from painful conditions and diseases. It has now become critical to address the challenges related to the safety and efficacy of MSCs expanded in vitro. In this study, we establish a standardized process for manufacture of canine adipose-derived MSCs (AD-MSCs), including tissue sourcing, cell isolation and culture, cryopreservation, thawing and expansion, quality control and testing, and evaluate the safety and efficacy of those cells for clinical applications. Results: After expansion, the viability of AD-MSCs manufactured under our standardized process was above 90 %. Expression of surface markers and differentiation potential was consistent with ISCT standards. Sterility, mycoplasma, and endotoxin tests were consistently negative. AD-MSCs presented normal karyotype, and did not form in vivo tumors. No adverse events were noted in the case treated with intravenously AD-MSCs. Enrofloxacin, a third-generation fluoroquinolone, is a broad-spectrum antimicrobial drug against a lot of veterinary bacterial diseases. However, bactericidal activity of enrofloxacin is concentration-dependent and its poor aqueous solubility and bitter taste limit its development and application. Meanwhile, 2-hydroxypropyl-β-cyclodextrin (HP-β-CD), a widely used cyclodextrin analog, is a safe and an effective drug carrier. It forms inclusion complexes with its drug substrates and improves their physiochemical and pharmacokinetic properties. Enrofloxacin was also found to form a stable inclusion complex with HP-β-CD and different research groups have shown improved solubility for enrofloxacin by 32.5%, 9.25 and 165-fold. Our own efforts in this direction resulted in manifold improvement (916-fold) in its solubility compared to the previous studies. The Weather Research and Forecasting (WRF) model was employed to simulate the water and energy process over the near surface in the Yellow River source region from July to September 2015. The Rapid Update Cycle (RUC), Noah, Noah-Multi-Physics (Noah-MP), and Community Land Model (CLM) version 4 land surface schemes were selected and evaluated in our experiments. The simulated results were also compared with the Global Land Data Assimilation System (GLDAS) outputs, the ground-based automatic weather stations (AWSs), and the Zoige plateau wetlands ecosystem research flux station data. There was substantial agreement between the results of the simulations and observations in terms of 2-m temperature and relative humidity. However, the simulated values of the 2-m temperature were mostly lower than observations, while the values of relative humidity were higher than the observations. The correlation coefficients (R) between the simulations and observations of air temperature in four experiments were all higher than 0.94, and simulated results of the WRF-RUC experiment had the minimum bias compared with other experiments. The root mean square error (RMSE) and bias of 2-m relative humidity between the WRF-CLM4 simulations and observations were lower than other experiments. Moreover, the simulation of the 10-cm soil temperature was lower than the observations. WRF-CLM4 simulations agreed better with the observations, and WRF-Noah-MP simulations had the largest bias in all of the experiments. The soil moisture increased rapidly when precipitation was occurring and decreased slowly after the precipitation. The four experiments overestimate precipitation leading to higher soil moisture compared with the real situations. The peak value of net radiation was appropriately 850 W/m2 during the summertime in the Zoige plateau area. The simulated net radiation agreed well with the observation except in the WRF-RUC scheme. During the summertime, vegetation grew rapidly, and the latent heat flux transport increased and played a dominant role in surface energy transport in the Zoige area. The RMSE between the simulation and observation of latent heat flux had the minimum value of 98.0 W/m2 in the WRF-CLM4 scheme, and the bias of sensible heat flux had the minimum value of 15.4 W/m2 in the WRF-Noah scheme. This study used the WRF model to explore the applicability of the different land surface schemes in the Zoige plateau area. The research related to this study can be used to understand the water-energy cycle process over the Qinghai-Tibet Plateau wetland and can also provide references for the application of regional meteorological models over this area. To study the land surface and atmospheric meteorological characteristics of non-uniform underlying surfaces in the semi-arid area of Northeast China, we use a “High-Resolution Assimilation Dataset of the water-energy cycle in China (HRADC)”. The grid points of three different underlying surfaces were selected, and their meteorological elements were averaged for each type (i.e., mixed forest, grassland, and cropland). For 2009, we compared and analyzed the different components of leaf area index (LAI), soil temperature and moisture, surface albedo, precipitation, and surface energy for various underlying surfaces in Northeast China. The results indicated that the LAI of mixed forest and cropland during the summer is greater than 5 m2 m−2 and below 2.5 m2 m−2 for grassland; in the winter and spring seasons, the Green Vegetation Fraction (GVF) is below 30%. The soil temperature and moisture both vary greatly. Throughout the year, the mixed forest is dominated by latent heat evaporation; in grasslands and croplands, the sensible heat flux and the latent heat flux are approximately equal, and the GVF contributed more to latent heat flux than sensible heat flux in the summer. This study compares meteorological characteristics between three different underlying surfaces of the semi-arid area of Northeast China and makes up for the insufficiency of purely using observations for the study. We investigated the anti-inflammatory effect of trilobatin, the flavonoid isolated from the leaves of Lithocarpus polystachyus Rehd, as well as the underlying molecular mechanisms. Treatment with trilobatin (0.005-5μM) dose-dependently inhibited the lipopolysaccharide (LPS)-induced mRNA expression and secretion of pro-inflammatory cytokines, including tumor necrosis factor α (TNFα), interleukin-1β (IL-1β) and interleukin-6 (IL-6), in RAW 264.7 macrophages. However, no further inhibition was detected when the concentration of trilobatin was increased to 50μM. Western blot analysis confirmed that the mechanism of the anti-inflammatory effect was correlated with the inhibition of LPS-induced inhibitor of nuclear factor-kappa B α (IκBα) degradation and nuclear factor-kappa B (NF-κB) p65 phosphorylation. In addition, trilobatin also showed a significant inhibition of LPS-induced TNFα and IL-6 at both the mRNA and protein levels in a mouse model. It's urgent to clarify the quantitative relationship between declining fine−particle (PM 2.5) concentrations and increasing ozone (O 3) levels as ozone pollution is increasingly severe in China. Here, we analyze the ground–based data of aerosol optical properties, particulates, and gaseous pollutants collected in a simultaneous observation in Kunming, locate in the Yunnan–Guizhou Plateau, from June 2014 to May 2017. The data exhibit slightly rising levels of PM 2.5 , O 3 , and carbon monoxid (CO), declining AOD (aerosol optical depth) but increasing AE (Angstrom exponent). Both the seasonal PM 2.5 and CO concentrations were low in the wet season and high in the dry season. The seasonal ozone levels and AE almost increased monotonically while the seasonal AOD decreased wavelike. Overall, the maximum daily 8 h average (MDA8) O 3 concentrations positively related to PM 2.5 concentrations with temperature>22 °C, RH<45%, and AOD<1.0 in the wet season. Generally, MDA8 O 3 concentrations didn't fluctuate with PM 2.5 concentrations in the dry season. High SSA (single scattering albedo) corresponded to high MDA8 O 3 concentrations and low SSA corresponded to low MDA8 O 3 concentrations in the wet season. However, lower SSA values in the dry season were observable. Most high MDA8 O 3 concentrations (>100 μg m−3) in the wet season occurred with AE at 1.0–1.5. High MDA8 O 3 concentrations (>100 μg m−3) in dry season appeared with an AE range of ~0.75–1.75. Low MDA8 O 3 concentrations occurred with AOD>2.0 and RH>75%, despite T>20 °C. The biomass−burning/urban aerosols were dominant (71.11% in the wet season and 74.40% in the dry season). The PSCF (Potential Source Contribution Function) results revealed the air pollutants from inside and outside China will have great impacts on the pollutant levels in Kunming besides the local emissions. Particularly, the severe pollution in India may aggravate Kunming's air pollution. • High SSA corresponded to high MDA8 O 3 concentrations and positive MDA8 O 3 − PM 2.5 relations were observable with SSA>0.95. • Most high MDA8 O 3 concentrations (>100 μg m−3) in the wet season occurred with AE at 1.0–1.5. • Biomass−burning/urban aerosols were dominant, accounting for over 70% of the whole aerosol types. • The air pollutants from outside China may have an important impact on the pollutant levels in Kunming. Cholic acid and galactose or lactose dual conjugated chitosan derivatives were designed and synthesized as potential anti liver cancer drug carriers, their structures were characterized through proton NMR spectra, elemental analysis, size distribution, zeta potential, and scanning electron microscope image studies. The ability of the dual conjugates to enhance the aqueous solubility of the cancer drug sorafenib was evaluated. The entrapment efficiency (EE%) and drug content (DC%) of sorafenib in the inclusion complexes were measured. The chitosan dual conjugate with cholic acid and galactose was found to be best in enhancing the aqueous solubility of sorafenib. Precipitation microphysics over the Tibetan Plateau (TP) remain insufficiently understood, due to the lack of observations and studies. This paper presents a comprehensive investigation of the raindrop size distribution (DSD) for rainfall that happened on the eastern slope of TP in summer. DSD differences between different rain types and under different rain rates are investigated. Confidential empirical relationships between the gamma shape and slope parameters, and between reflectivity and rain rate are proposed. DSD properties in this area are also compared with those in other areas. The results indicate that the stratiform and convective rains contribute to different rain duration and amount, with diverse rainfall macro- and microphysical properties. The rain spectra of two rain types can become broader with higher concentrations as the rain rate increases. DSDs in this area are different to those in other areas. The stratiform DSD is narrower than that in the non-plateau area. The two rain types of this area both have higher number concentrations for 0.437–1.625 mm raindrops than those of the mid-TP. The relationships of shape–slope parameters and reflectivity–rain rate in this area are also different from those in other areas. The rain spectra in this area can produce a larger slope parameter under the same shape parameter than in the mid-TP. The convective rain can produce a smaller rain rate under the same reflectivity. The accuracy proposed reflectivity–rain rate relationship in application to quantitative rainfall estimation is also discussed.
