Aim: Contemporary species' distributions are shaped by both geography and historical events, such as extinction, diversification in specific areas and long‐distance dispersals. In the most diverse family of lichen‐forming fungi, Parmeliaceae, the Hypogymnioid clade, is an example of an evolutionary lineage comprised of species occurring in temperate to subpolar regions in both hemispheres. Here, we elucidate the timing of diversification events and the impact of historical events on the species distribution in this lineage. Location: Worldwide. Taxon: Genera Arctoparmelia, Brodoa, Hypogymnia and Pseudevernia (Parmeliaceae). Methods: Our sampling focused on the most diverse genus of the clade, Hypogymnia, including c. 70% of the described species. We reconstructed phylogenetic relationships using a multi‐locus data set, estimated divergence times, and inferred ancestral distributions. Results: Our analyses suggest that the ancestor of the Hypogymnioid clade occurred in the Holarctic. In each of the four genera, all recovered as monophyletic here, diversification have occurred largely during the Miocene and Pliocene. A number of currently accepted species did not form monophyletic groups, especially in cases where specimens were collected from distinct geographic areas, with multiple, distinct clades corresponding to the geographic region of origin. Our results suggest that only a very few species in the Hypogymnioid clade have cosmopolitan distributions, all of which reproduces using vegetative propagules including both symbiotic partners. Main conclusions: While the diversification occurred predominantly in the Northern Hemisphere during the Miocene, a long‐distance dispersal event from the Northern to the Southern Hemisphere, resulted in diversification of a clade of species largely restricted to the Southern Hemisphere. Strigula has traditionally been circumscribed based on morphology, but species delimitation in the genus generally lacks comprehensive analyses. Molecular approach has now been applied to foliicolous material of the genus from tropical areas in China. On the basis of combined phenotype and genotype, Strigula univelbiserialis is described as a new species and Strigula delicata , S. janeirensis are reported as two new records in China. Endolichenic fungi within 17 lichen species in the area near Ny-Ålesund (Svalbard, High Arctic) were studied by a culture-based method. The 247 fungal isolates were obtained from 2712 lichen thallus segments. The colonization rate of endolichenic fungi ranged from 1.6 to 26.5 %, respectively. These isolates were identified to 40 fungal taxa, including 35 Ascomycota (10 orders), 4 Basidiomycota (3 orders), and 1 unidentified fungus. Thelebolales was the most abundant order, while Sordariales were the most diverse order. The common fungal taxa shared by more than 3 lichen species were Thelebolus microsporus (93 isolates), Coniochaeta hoffmannii (7 isolates), Sarocladium kiliense (33 isolates), Coniochaeta sp. 1 (5 isolates), Coniochaeta sp. 4 (28 isolates), and Coniochaeta sp. 2 (5 isolates). Low Sorenson's similarity coefficients were observed among different lichen species, indicating that host-related factor may shape the endolichenic fungal communities in this region. In addition, no endolichenic fungal taxa were previously found in the Antarctica and Austrian Alps, suggesting endolichenic fungal communities in this region might be also shaped by the Arctic climate. The results demonstrate the existence of specific cultured endolichenic fungal species, which may be suitable objects for further study of their possible functional roles in the lichen thalli. The genus Hypogymnia is morphologically characterized by swollen lobes, a hollow medulla, perforations in the lower surface, and the absence of rhizines. In our studies on Hypogymnia spp. we found rhizines and hapters were occasionally present on the lower surface of 35 species. Morphology and anatomy of the appendage-organs were studied using dissecting, compound, and scanning electron microscopy and compared with rhizines found in the related genus Arctoparmelia. Two types of rhizine-like structures were found in Hypogymnia: slender and elongated rhizines, and thick and short hapters, both of which were few in number and sparsely distributed on the lower surface. There are no obvious differences in the morphology of rhizines between Arctoparmelia and Hypogymnia, except that rhizines are abundant in Arctoparmelia. Within Parmeliaceae, Hypogymnia belongs to the hypogymnioid clade together with the genera Arctoparmelia, Brodoa, and Pseudevernia, most of which are considered to lack rhizines. Allocetraria yunnanensis R. F. Wang, X. L. Wei & J. C. Wei is described as a new species from the Yunnan Province of China, and is characterized by having a shiny upper surface, strongly wrinkled lower surface, and marginal pseudocyphellae present on the lower side in the form of a white continuous line or spot. Six pruinose species of Hypogymnia are reported in this paper, including one new species Hypogymniapruinoidea. The type of Hypogymnia pseudopruinosa was found to be a mixture with H. laccata. Hypogymnia pseudopruinosa is therefore typified with a lectotype, and the description of H. pseudopruinosa is revised. Distributions of the six pruinose species are given and discussed. Comments on differences and similarities between pruinose species of Hypogymnia are made. The development of novel photonic devices which incorporate biological materials is strongly tied to the development of thin film forming processes. Solution-based (“wet”) processes when used with biomaterials in device fabrication suffer from dissolution of underlying layers, incompatibility with clean environment, inconsistent film properties, etc. We have investigated ultra-high-vacuum molecular beam deposition of surfactant-modified deoxyribonucleic acid (DNA). Two new species of the lichenized genus Lasioloma are described from Asia: Lasioloma longiramosum W. C. Wang & A. Abas (collected from Malaysia), is characterized by a distinct woolly prothallus between dispersed thallus patches, comparatively small, muriform ascospores, long filiform conidia (main branch 22–28 μm in length, the other three branches 65–80 μm) and a foliicolous habitat; L. verrucosum W. C. Wang & X. L. Wei (collected from China), is characterized by a warted thallus, filiform conidia (main branch 22–32 μm in length, the other three branches 50–65 μm) and a corticolous habitat. The placement of both new species was confirmed by a molecular phylogenetic approach based on combined ITS, mtSSU and mtLSU sequences, and both are compared in detail to other similar species of the genus. Our study also revealed that the length of the conidial branches, which has not been explored in previous studies, should be regarded as an important feature for species delimitation in Lasioloma. Lichen as mutualistic symbiosis is the dominant organism in various extreme terrestrial environment on Earth, however, the mechanisms of their adaptation to extreme habitats have not been fully elucidated. In this study, we chose the Antarctic dominant lichen species Usnea aurantiacoatra to generate a high-quality genome, carried out phylogenetic analysis using maximum likelihood and identify genes under positive selection. We performed functional enrichment analysis on the positively selected genes (PSGs) and found that most of the PSGs focused on transmembrane transporter activity and vacuole components. This suggest that the genes related to energy storage and transport in Antarctic U. aurantiacoatra were affected by environmental pressure. Inside of the 86 PSGs screened, two protein interaction networks were identified, which were RNA helicase related proteins and regulator of G-protein signaling related proteins. The regulator of the G-protein signaling gene (UaRGS1) was chosen to perform further verification by the lichen genetic manipulation system Umbilicaria muhlenbergii. Given that the absence of UmRgs1 resulted in elevated lethality to cold shock, the role for UaRgs1 in Antarctic U. aurantiacoatra resistance to cold can be inferred. The Gram-negative bacterium Xanthomonas oryzae pv. oryzicola (Xoc) is the causal agent of rice bacterial leaf streak (BLS), one of the most destructive diseases of rice (Oryza sativa L.) that is the important staple crop. Xoc can invade host leaves via stomata and wounds and its type three secretion system (T3SS) is pivotal to its pathogenic lifestyle. In this study, using a novel dual RNA-seq approach, we examined transcriptomes of rice and Xoc in samples inoculated with wild type Xoc GX01 and its T3SS defective strain (T3SD), to investigate the global transcriptional changes in both organisms. Compared with T3SD strain, rice inoculated with wild type Xoc GX01 resulted in significant expression changes of a series of plant defence related genes, including ones altered in plant signalling pathway, and downregulated in phenylalanine metabolism, flavonoid and momilactone biosynthesis, suggesting repression of plant defence response and reduction in both callose deposition and phytoalexin accumulation. Also, some known transcription activator-like effector (TALE) targets were induced by Xoc GX01, e.g. OsSultr3;6 which contributes to rice susceptibility. Some cell elongation related genes, including several expansin genes, were induced by GX01 too, suggesting that Xoc may exploit this pathway to weaken cell wall strength, beneficial for bacterial infection. On the other hand, compared with wild type, the T3SD strain transcriptome in planta was characterized by downregulation of ATP, protein and polysaccharide synthesis, and upregulation of antioxidation and detoxification related genes, revealing that T3SD strain faced serious starvation and oxidation stresses in planta without a functional T3SS. In addition, comparative global transcript profiles of Xoc in planta and in medium revealed an upregulation of virulence factor synthesis and secretion in planta in favour of bacterial infection. Collectively, this study provides a comprehensive representation of cross talk between the host and bacterial pathogen, revealing insights into the Xoc-rice pathogenic dynamic and reveals novel strategies exploited by this important pathogen to cause disease. The lichenized fungal genus Astrothelium is an important element of crustose lichen communities in tropical to subtropical forests. Morphological and molecular phylogenetic approaches to investigate species diversity of Astrothelium (Trypetheliaceae) from Southern China were carried out in this study. Bayesian and maximum-likelihood (ML) analyses were generated based on the combined data set of internal transcribed spacer (ITS), partial regions of the nuclear ribosomal large subunit (LSU), and the largest subunit of RNA polymerase II gene sequences (RPB1). The morphological comparison with the known Astrothelium taxa and molecular phylogeny support five new species: Astrothelium jiangxiense sp. nov., A. luminothallinum sp. nov., A. pseudocrassum sp. nov., A. subeustominspersum sp. nov., and A. subrufescens sp. nov. Hypogymnia papilliformis McCune, Tchabanenko & X. L.Wei is described as a new species of lichenized fungus from the Primorsky region of Russia and Shaanxi Province in China. It is a relatively rare species from mixed conifer-broadleaved forests in suboceanic climates, in mountainous areas at elevations of 600–1500 m. Most similar to H. delavayi, the species are distinguished by branching pattern and chemistry. Hypogymnia papilliformis is predominantly isotomically branched while H. delavayi usually develops subpinnate branching. The ceiling of the lobe cavity is mid brown to dark brown in H. papilliformis, while the ceiling is often pale brownish or white in H. delavayi. Also, H. papilliformis lacks 3-hydroxyphysodic acid, and thus has a K– medulla, while H. delavayi always contains 3-hydroxyphysodic acid as a major substance and is thus K+ slowly reddish brown. Abstract: Successful feeding is critical to biomass utilization processes such as pyrolysis liquefaction or gasification. The biomass particles in hopper frequently become bridge or adhere to the wall to prevent the feeding system from providing the required uniform and continuous flow of feedstocks. To solve the problem, an automatic shake mechanism was devised which was set in the hopper and driven by the rotation of screw. Experiments were done using the automatic shake mechanism to feed three kinds of sawdust; the total torque requirement of the screw feeder with the automatic shake mechanism in the hopper was predicted and compared with experiment results. The results showed that the biomass particles could be delivered uniformly and the feeding rate had a significant increase with 7% extra power consumption. [ABSTRACT FROM AUTHOR]

Clinical studies have indicated that early brain injury (EBI) following subarachnoid hemorrhage (SAH) is associated with fatal outcomes. Oxidative stress and brain edema are the characteristic pathological events in occurrence EBI following SAH. The present study aimed to examine the effect of 3,4-dihydroxyphenylethanol (DOPET) against SAH-induced EBI, and to demonstrate whether the effect is associated with its potent free radical scavenging property. SAH was induced in rats using an endovascular perforation technique, and 24 h later the rats displayed diminished neurological scores and brain edema. Furthermore, elevated malondialdehyde (an index of lipid peroxidation) and depleted levels of antioxidants were observed in the rat cerebral cortex tissue. Quantitative polymerase chain reaction analysis indicated the upregulated mRNA expression of the apoptotic markers caspase-3 and -9 in the cerebral cortex. Furthermore, the protein expression levels of the proinflammatory cytokines tumor necrosis factor-α, interleukin (IL)-1β and IL-6 were significantly upregulated in SAH-induced rats. By constrast, treatment with DOPET significantly attenuated EBI by reducing brain edema, elevation of antioxidant status, inhibition of apoptosis and inflammation. The biodegradation and toxicity of the purified terephthalic acid (PTA) processing wastewater was researched at NJYZ pilot with the fusant strain Fhhh in the carrier activated sludge process (CASP). Sludge loading rate (SLR) for Fhhh to COD of the wastewater was 1.09 d^{-1} and to PTA in the wastewater was 0.29 d^{-1}. The results of bioassay at the pilot and calculation with software Ebis3 showed that the 48h-LC_{50} (median lethal concentration) to Daphnia magna for the PTA concentration in the wastewater was only 1/10 of that for the chemical PTA. There were 5 kinds of benzoate pollutants and their toxicities existing in the wastewater at least. The toxicity parameter value of the pure chemical PTA cannot be used to predicate the PTA wastewater toxicity. The toxicity of the NJYZ PTA wastewater will be discussed in detail in this paper. We employed a molecular phylogenetic approach using five markers (ITS, nuSSU, nuLSU, TEF1-α, and RPB2) to assess potential cryptic speciation in foliicolous members of Strigula s.lat. (Strigulaceae), including the recently segregated genera Phylloporis, Puiggariella, Raciborskiella, Racoplaca, and Serusiauxiella, from tropical areas in Asia, with selected materials from the Neotropics as reference. On the basis of combined molecular and phenotypic datasets, two new species of Racoplaca and 10 new species of Strigula s.str. are described: Racoplaca macrospora sp. nov., R. maculatoides sp. nov., Strigula guangdongensis sp. nov., S. intermedia sp. nov., S. laevis sp. nov., S. microcarpa sp. nov., S. pseudoantillarum sp. nov., S. pseudosubtilissima sp. nov., S. pycnoradians sp. nov., S. sinoconcreta sp. nov., S. stenoloba sp. nov., and S. subtilissimoides sp. nov. In addition, we propose the new combination Phylloporis palmae comb. nov. (≡ =Manaustrum palmae) and we validate the earlier combination Racoplaca melanobapha comb. nov. (≡ Verrucaria melanobapha; Strigula melanobapha). Our data clearly indicate a considerable degree of cryptic diversification in foliicolous representatives of Strigula s.lat., particularly in the presumably widespread taxa Strigula antillarum, S. concreta, S. nitidula, and S. smaragdula. Given that these phylogenetic revisions are thus far limited to few regions, we predict that our findings only represent the proverbial tip of the iceberg in this group of lichenized fungi. Ratcheting and low-cycle fatigue failure behaviors of the adhesively bonded hollow cylindrical butt-joints has been experimentally investigated. A series of uniaxial cyclic tension experiments were carried out under stress-controlled mode. The effects of stress amplitude, mean stress and cycle time on the uniaxial ratcheting response, fatigue damage variable evolution and fatigue life of the adhesively bonded butt-joints were analyzed. The results show that the ratcheting strain, ratcheting strain rate and fatigue damage variable all increase with the increase of stress amplitude and mean stress. The shorter cycle time results in the increase of fatigue damage variable and the degradation of the stiffness of the adhesive material. It is also found that the increase of stress amplitude and mean stress can reduce the low-cycle fatigue life. Meanwhile, the fatigue life increases with the increase of cycle time for the adhesively bonded butt-joints. [ABSTRACT FROM AUTHOR]

Uniaxial and multiaxial fatigue experiments were conducted on the adhesively bonded hollow cylindrical butt-joint specimen under load-controlled mode, and several prediction criteria were used for multiaxial fatigue life prediction. The experimental results showed that the fatigue life of uniaxial tensile cycle was shorter than that of uniaxial torsional cycle, and the fatigue life of the non-proportional circular loading path was shorter than that of the proportional loading path. In addition, the multiaxial fatigue life prediction results showed that Smith-Watson-Topper criterion based on the maximum principal strain as the critical plane had a conservative life prediction for the non-proportional circular loading path. Fatemi-Socie criterion mainly considered the influence of shear strain which yielded the non-conservative prediction for some data points of proportional loading path. Chen-Xu-Huang criterion combined energy density and critical plane achieved better fatigue life predictions than those from the previous two criteria. Furthermore, considering general failure modes of the adhesively bonded butt-joints, the modified Chen-Xu-Huang model was proposed, and the predicted results are good agreement with the experiments.
