A confiscated package of street drugs was characterized by the usual mass spectral (MS) and FT–IR analyses. The confiscated powder material was highly crystalline and was found to consist of two very different species, accidentally of sizes convenient for X-ray diffraction. Thus, one each was selected and redundant complete sets of data were collected at 100 K using Cu Kα radiation. The selected crystals contained: (a) 1,2-diphenyl-2-(pyrrolidin-1-yl)ethanone hydrochloride hemihydrate or 1-(2-oxo-1,2-diphenylethyl)pyrrolidin-1-ium chloride hemihydrate, C18H20NO+ ·Cl-·0.5H2O, (I), a synthetic cathinone called ‘α-D2PV’, and (b) the sugar myo-inositol, C6H12O6, (II), probably the only instance in which the drug and its diluent have been fully characterized from a single confiscated sample. Moreover, the structural details of both are rather attractive showing: (i) interesting hydrogen bonding observed in pairwise interactions by the drug molecules, mediated by the chloride counter-anions and the waters of crystallization, and (ii) π–π interactions in the case of the phenyl rings of the drug which are of two different types, namely, π–π stacking and edge-to-π. Recent shifts in political support to populist parties worldwide have been linked to the changing preferences of "left behind communities." Based on apparently growing "left behind" support for populists, some commentators have argued for policy changes including tightened immigration rules coupled with increasing investment in economically deprived areas, particularly in health care. However, left behind communities' policy preferences are unclear from existing research due to a series of methodological challenges associated with researching polarization and stigmatization. We complement existing research with an innovative photo elicitation methodology covering five field sites in the United Kingdom during 2019, focusing on left behind communities' policy preferences concerning Brexit. Photo elicitation overcomes methodological challenges associated with emotional attachment and stigmatization. Drawing on 418 interviews with 489 participants, we find that interviewees rejected elite framings suggesting a logical link between Brexit and health care investment, instead articulating policy preferences for health care investment drawing on personal experiences. Diffuse gliomas are a heterogeneous category of primary central nervous system tumors. Due to their infiltrative growth precluding complete surgical resection, most diffuse high-grade gliomas are treated with adjuvant chemotherapy and radiation. Recurrent/progressive diffuse gliomas may show genetic differences when compared to the primary tumors, giving insight into their molecular evolution and mechanisms of treatment resistance. In adult-type diffuse gliomas with or without isocitrate dehydrogenase gene mutations, tumor recurrence/progression can be associated with mutations in genes encoding DNA mismatch repair proteins, leading to a dramatic increase in tumor mutation burden. This phenomenon is closely linked to treatment with the DNA alkylating agent temozolomide, a mainstay of adult diffuse glioma chemotherapeutic management. Post-treatment mismatch repair deficiency and acquired high tumor mutation burden is relatively unexplored in pediatric patients who have recurrent high-grade gliomas. Here, we report a molecular and histological analysis of an institutional cohort of eleven pediatric patients with paired initial and recurrent high-grade astrocytoma samples with intervening temozolomide treatment. We identified three cases with evidence for increased tumor mutation burden at recurrence, including two cases of diffuse hemispheric glioma H3 G34-mutant (one previously reported). We also show that molecular analysis by next-generation DNA sequencing and DNA methylation-based profiling enabled an integrated diagnosis per 2021 World Health Organization criteria in 10 of 11 cases (91%). Our findings indicate that increased tumor mutation burden at post-treatment recurrence is relevant in pediatric-type diffuse high-grade gliomas. Background: Periodic updates to the World Health Organization (WHO) classification system for central nervous system (CNS) tumors reflect advances in the pathological diagnosis, categorization, and molecular underpinnings of primary brain, spinal cord, and peripheral nerve tumors. The 5th edition of the WHO Classification of CNS Tumors was published in 2021. This review discusses the guiding principles of the revision, introduces the more common new diagnostic entities, and describes tumor classification and nomenclature changes that are relevant for pediatric neurological surgeons. Summary: Revisions to the WHO CNS tumor classification system introduced new diagnostic entities, restructured and renamed other entities with particular impact in the diffuse gliomas and CNS embryonal tumors, and expanded the requirements for incorporating both molecular and histological features of CNS tumors into a unified integrated diagnosis. Many of the new diagnostic entities occur at least occasionally in pediatric patients and will thus be encountered by pediatric neurosurgeons. New nomenclature impacts the terminology that is applied in communication between pathologists, surgeons, clinicians, and patients. Requirements for molecular information in tumor diagnosis are expected to refine diagnostic categories while also introducing practical considerations for intraoperative consultation, preliminary histological evaluation, and triaging of neurosurgical tissue samples for histology, molecular testing, and clinical trial requirements. Key Messages: Pediatric brain tumor diagnosis and clinical management are a multidisciplinary effort that is rapidly advancing in the molecular era. Interdisciplinary collaboration is critical for providing the best care for pediatric CNS tumor patients. Simple Summary: Robotic surgery is reported to have benefits for the surgical management of patients with a high BMI. However, there is a lack of information on patient-reported outcomes and recovery following robotic-assisted hysterectomy (RH). A study collecting information on participant characteristics, intra- and post-operative events was conducted. Telephone questionnaires at 2, 4, 6, and 12 weeks were used to collect patient-reported recovery using the QoR-40 quality-of-recovery questionnaire. Of the 53 individuals recruited, 50 underwent RH and three cases were converted to open surgery. Patient BMI had a small impact on operative time (p = 0.04) but not on length of stay (p = 0.62). Overall quality-of-life (QOL) scores were consistently high post-operatively, indicating a high quality of recovery, and were not impacted by patient BMI. A longitudinal, descriptive, prospective, and prolective study of individuals with endometrial or cervical cancer/pre-cancer diagnoses and high BMI (over 35 kg/m2) undergoing RH was conducted. Of the 53 participants recruited, 3 (6%) were converted to open surgery. The 50 RH participants had median BMI 42 kg/m2 (range 35 to 60): the range 35–39.9 kg/m2 had 17 cases; the range 40–44.9 kg/m2 had 15 cases; 45–49.9 kg/m2 8 cases; and those ≥50 kg/m2 comprised 10 cases. The mean RH operating time was 128.1 min (SD 25.3) and the median length of hospital stay was 2 days (range 1–14 days). Increased BMI was associated with small, but statistically significant, increases in operating time and anaesthetic time, 65 additional seconds and 37 seconds, respectively, for each unit increase in BMI. The median self-reported time for individuals who underwent RH to return to their pre-operative activity levels was 4 weeks (range 2 to >12 weeks). There was a significant improvement in pain and physical independence scores over time (p = 0.001 and p < 0.001, respectively) and no significant difference in scores for overall QOL, pain, or physical independence scores was found between the BMI groups. Background: Bats are increasingly being recognized as important hosts for viruses, some of which are zoonotic and carry the potential for spillover within human and livestock populations. Biosurveillance studies focused on assessing the risk of pathogen transmission, however, have largely focused on the virological component and have not always considered the ecological implications of different species as viral hosts. The movements of known viral hosts are an important component for disease risk assessments as they can potentially identify regions of higher risk of contact and spillover. As such, this study aimed to synthesize data from both virological and ecological fields to provide a more holistic assessment of the risk of pathogen transmission from bats to people. Results: Using radiotelemetry, we tracked the small-scale movements of Rousettus aegyptiacus, a species of bat known to host Marburg virus and other viruses with zoonotic potential, in a rural settlement in Limpopo Province, South Africa. The tracked bats exhibited seasonal variations in their movement patterns including variable usage of residential areas which could translate to contact between bats and humans and may facilitate spillover. We identified a trend for increased usage of residential areas during the winter months with July specifically experiencing the highest levels of bat activity within residential areas. July has previously been identified as a key period for increased spillover risk for viruses associated with R. aegyptiacus from this colony and paired with the increased activity levels, illustrates the risk for spillover to human populations. Conclusion: This study emphasizes the importance of incorporating ecological data such as movement patterns with virological data to provide a better understanding of the risk of pathogen spillover and transmission. When verifying the correctness of single‐digit multiplication problems, children and adults show a robust ERP correctness effect thought to reflect similar cognitive processes across groups. Recent studies suggest that this effect is instead a modulation of the negative‐going N400 component in children, reflecting access to semantic memory, and the positive‐going P300 component in adults, reflecting stimulus categorization. However, the relative difference in ERP amplitude is the same for both components, more positive for correct than incorrect solutions, presenting a challenge to ascertaining the appropriate interpretation. Time‐frequency analysis (TFA) of the N400/P300 window provides an objective approach to dissociating these effects. TFA measured from solution onset during single‐digit multiplication verification revealed significant modulations of event‐related as theta power (3–6 Hz) in both groups. Correct trials elicit less power in children (9–12 years) and more power in adults relative to incorrect trials. These findings are consistent with modulations of the N400 and P300, respectively, where opposite effects were predicted for spectral power. The ERP results further support a reinterpretation of the multiplication correctness effect. In contrast, TFA of the N400 effect elicited to a word‐picture verification task revealed the same event‐related theta effect in both groups, with increased power for mismatched than matched pictures. Together, these findings provide evidence for a developmental shift in cognitive processing specific to the multiplication task. Models of arithmetic should account for this overlooked difference in cognitive processing between children and adults. Although previous research proposed that adults and children use the same cognitive processes to verify multiplication problems, the current results suggest otherwise. Children elicit a robust N400 effect, whereas adults show a P300 effect. Given that these components are difficult to dissociate using conventional analysis, we used time‐frequency analysis to quantify the amplitude of the ERP oscillations. What is the value of including vulnerable people in risk regulation decision-making in the European Union (EU)? This article examines a distinctive approach employed by the European Medicines Agency (EMA): public hearings integrated within safety reviews of medicinal products. The article presents findings from a case study of the EMA's public hearing on Quinolone antibiotics, which was included by the EMA as part of a review process that led to significant tightening of regulatory restrictions on the prescribing of this class of antibiotics. The article argues that the public hearing enabled a group of patients who had been victims of a debilitating toxicity syndrome associated with Quinolone antibiotics to criticise the existing scientific evidence base around the safety of Quinolone. Deploying the quantitative Discourse Quality Index and an interpretive analytical approach, the article shows how patients challenged the evidence base in a manner that was efficacious in advancing knowledge in this area of risk regulation. When physically staged alongside interventions by professional experts, the article argues that patients facilitated a process of "negotiation" of expertise, leading professional representatives to propose methods of coordination in order to integrate the patients' qualitative evidence of their suffering with the toxicity syndrome. In 2017, the European Medicines Agency staged the first effort at democratic innovation within transnational European Union institutions directly influencing the transnational regulation of medicines. Alongside its public consultation on epilepsy drug Valproate, European Medicines Agency included a public hearing involving representatives of patients and testimony from those directly affected by the medicines. Using this critical case study, the article argues from a deliberative democratic perspective that although the hearing in many ways exhibited the traditional shortcomings of elite-driven deliberation, it also demonstrated unexpected and surprising deliberative qualities. Based on new quantitative analysis of the hearing using the Discourse Quality Index, and qualitative observation of over 4 hours of footage, the article argues European Medicines Agency's hearing facilitated equitable access and influence of patients and members of the public who had previously been excluded from decision making on drug distribution. This article opens a Special Issue on Democratic Stress. Against the background of recent debates on 'democratic crisis', we argue that the concept of 'democratic stress' provides a useful way to understand the diversity of pressures that representative democracy faces in the contemporary context, as well as a valuable 'organising perspective' for developing more nuanced analyses. Anti-politics has emerged as an important concept for analysing the effects of distrust on liberal democratic politics. However, it is unclear why democrats should trust individuals who distrust politics to help them in renewing democracy. This article addresses this puzzle by defining four types of anti-politics: technocratic, elitist, populist and participatory. It then compares the political thought of four democratic thinkers associated with each type, to discern the extent to which they are 'productive' or 'unproductive' for representative democracy. The article argues that participatory and technocratic types of anti-politics, illustrated by the thought of Carole Pateman and, to a lesser extent, Friedrich Hayek, are productive for representative democracy because they prompt reflexivity in how representative institutions work. By contrast, populist and elitist types of anti-politics, illustrated by the thought of Ernesto Laclau and Joseph Schumpeter, are less productive. The article concludes that scholars need to carefully discern the logic underlying populist and technocratic 'solutions' to our contemporary democratic crisis because those solutions can themselves be advocated by 'false friends' who are unreflexive about what should be considered ideal sources of 'expert knowledge' or 'popular will'. Water is vital for mammals. Yet, as ephemeral sources can be difficult to find, it raises the question, how do mammals locate water? Elephants (Loxodonta africana) are water-dependent herbivores that possess exceptional olfactory capabilities, and it has been suggested that they may locate water via smell. However, there is no evidence to support this claim. To explore this, we performed two olfactory choice experiments with semi-tame elephants. In the first, we tested whether elephants could locate water using olfactory cues alone. For this, we used water from two natural dams and a drinking trough utilised by the elephants. Distilled water acted as a control. In the second, we explored whether elephants could detect three key volatile organic compounds (VOCs) commonly associated with water (geosmin, 2-methylisoborneol, and dimethyl sulphide). We found that the elephants could locate water olfactorily, but not the distilled water. Moreover, they were also able to detect the three VOCs associated with water. However, these VOCs were not in the odour profiles of the water sources in our experiments. This suggests that the elephants were either able to detect the unique odour profiles of the different water sources or used other VOCs that they associate with water. Ultimately, our findings indicate that elephants can locate water olfactorily at small spatial scales, but the extent to which they, and other mammals, can detect water over larger scales (e.g. km) remains unclear. HIV-infected infants are at an increased risk of progressing rapidly to AIDS in the first weeks of life. Here, we evaluated immunological and virological parameters in 25 SIV-infected infant rhesus macaques to understand the factors influencing a rapid disease outcome. Infant macaques were infected with SIVmac251 and monitored for 10 to 17 weeks post-infection. SIV-infected infants were divided into either typical (TypP) or rapid (RP) progressor groups based on levels of plasma anti-SIV antibody and viral load, with RP infants having low SIV-specific antibodies and high viral loads. Following SIV infection, 11 out of 25 infant macaques exhibited an RP phenotype. Interestingly, TypP had lower levels of total CD4 T cells, similar reductions in CD4/CD8 ratios and elevated activation of CD8 T cells, as measured by the levels of HLA-DR, compared to RP. Differences between the two groups were identified in other immune cell populations, including a failure to expand activated memory (CD21-CD27+) B cells in peripheral blood in RP infant macaques, as well as reduced levels of germinal center (GC) B cells and T follicular helper (Tfh) cells in spleens (4- and 10-weeks post-SIV). Reduced B cell proliferation in splenic germinal GCs was associated with increased SIV+ cell density and follicular type 1 interferon (IFN)-induced immune activation. Further analyses determined that at 2-weeks post SIV infection TypP infants exhibited elevated levels of the GC-inducing chemokine CXCL13 in plasma, as well as significantly lower levels of viral envelope diversity compared to RP infants. Our findings provide evidence that early viral and immunologic events following SIV infection contributes to impairment of B cells, Tfh cells and germinal center formation, ultimately impeding the development of SIV-specific antibody responses in rapidly progressing infant macaques. Author summary: Despite significant reductions in vertical HIV transmission, nearly 100,000 children succumb to AIDS-related illnesses each year. Indeed, infants face a disproportionately higher risk of progressing to AIDS, with roughly half of HIV+ infants exhibiting a rapid progression to AIDS-associated morbidity and mortality. This case supports that H3.3 p.G34-mutant gliomas should be included in studies relating to hypermutation in central nervous system tumours, and that re-sequencing of recurrent gliomas can uncover unexpected mechanisms of treatment resistance. This case of post-treatment hypermutation in a glioma with H3F3A p.G34 mutation suggests that acquired somatic hypermutation and clonal selection can contribute to therapeutic resistance and tumour progression in this glioma subtype. TAR DNA-binding protein 43 (TDP-43; also known as TARDBP) is an RNA-binding protein whose aggregation is a hallmark of the neurodegenerative disorders amyotrophic lateral sclerosis and frontotemporal dementia. TDP-43 loss increases DNA damage and compromises cell viability, but the actual function of TDP-43 in preventing genome instability remains unclear. Here, we show that loss of TDP-43 increases R-loop formation in a transcriptiondependent manner and results in DNA replication stress. TDP-43 nucleic-acid-binding and self-assembly activities are important in inhibiting R-loop accumulation and preserving normal DNA replication. We also found that TDP-43 cytoplasmic aggregation impairs TDP-43 function in R-loop regulation. Furthermore, increased R-loop accumulation and DNA damage is observed in neurons upon loss of TDP-43. Together, our findings indicate that TDP-43 function and normal protein homeostasis are crucial in maintaining genomic stability through a co-transcriptional process that prevents aberrant R-loop accumulation. We propose that the increased R-loop formation and genomic instability associated with TDP-43 loss are linked to the pathogenesis of TDP-43 proteinopathies. [ABSTRACT FROM AUTHOR]

BCG vaccination has been demonstrated to increase levels of activated CD4+ T cells, thus potentially influencing mother-to-child transmission of human immunodeficiency virus (HIV). To assess the risk of BCG vaccination in HIV infection, we randomly assigned newborn rhesus macaques to receive BCG vaccine or remain unvaccinated and then undergo oral simian immunodeficiency virus (SIV) challenges 3 weeks later. We observed elevated levels of activated peripheral CD4+ T cells (ie, HLA-DR+CD38+CCR5+ CD4+ T cells) by week 3 after vaccination. BCG was also associated with an altered immune gene expression profile, as well as with monocyte activation in both peripheral blood and the draining axillary lymph node, indicating significant BCG vaccine–induced immune activation. Despite these effects, BCG vaccination did not increase the rate of SIV oral transmission or disease progression. Our findings therefore identify patterns of T-cell and monocyte activation that occur after BCG vaccination but do not support the hypothesis that BCG vaccination is a risk factor for postnatal HIV transmission or increased pathogenesis in infants. Political scientists are wary of engaging with 'the public' on mainstream and social media because they fear those mediums fail to get across the deep and nuanced argument they develop in their own research. This article suggests a way of justifying public engagement that begins not with debates about the ethical and political concerns of doing this in practice (of which there are many), but how we as political scientists justify public media engagement to ourselves on the basis of the ethical and political process of 'doing' political science. As such, this article identifies the disciplinary basis upon which we may justify media-driven public engagement as an integral part of political science as an academic enterprise. Drawing on current epistemological debates in political science, the article characterises moments of political research as impressionistic exercises, which require public engagement. This means making the public aware of the deep and valuable insights of political science, in a way that sketches out how the discipline can shed light on important social and political phenomena, thereby informing our own scholarly thinking, and that of those we engage with. This study investigates the relationship between an organization's religious values, as espoused by the founder or in media messaging, and applicant intentions to pursue a job. Drawing on person-organization fit theory, we also explored interactions between an organization's espoused religious values and characteristics of the individual applicant. We tested our predictions via two conjoint analysis experiments, one with 191 employed adults collectively making 2292 employment pursuit decisions and a second with 120 employed adults making 1080 employment pursuit decisions. Espousing religious values as a founder or in media messaging yielded lower intentions to pursue a job than when an organization espouses non-religious values. However, when there was fit based on religious values, these relationships were mitigated. The results expand our understanding of person-organization fit by demonstrating the potential influence of espousing religious values on attracting organizational members. [ABSTRACT FROM AUTHOR]

• Fluoride-responsive degradable unit can be incorporated into PUs with a range of adhesive properties. • The mechanical properties can be tuned by varying structure and density of H-bonding monomers. • Introducing crystallinity into the adhesives can lower the bonding temperature to 50 °C. • The adhesives undergo depolymerisation to facilitate debonding on addition of fluoride ions. This paper reports the solvent-free synthesis of a series of six fluoride responsive debond-on-demand polyurethane (PU) adhesives that contain a silyl functionalised degradable unit (DU). To optimise the adhesion strength and debonding nature of the adhesives, the chemical composition of the PUs was varied according to the structure of the polyol or the diisocyanate component in the polymer mainchain. 1H NMR spectroscopy was used to study the depolymerisation behaviour in solution state. It showed that tetra -butylammonium fluoride (TBAF) triggered the breakdown of the DU unit without fragmenting the polyol mainchain indiscriminately. On exposure to fluoride ions, the PUs underwent depolymerisation with reductions in M n ranging from 64 to 90% as measured by GPC analysis. The morphology and thermal properties of the PUs were characterised by differential scanning calorimetry (DSC), rheology and variable temperature (VT) SAXS/WAXS analysis. Each technique demonstrated the reversibility of the supramolecular polymer network under thermal stimuli. PUs containing poly(butadiene) soft segments were amorphous with glass transition and viscoelastic transition temperatures dependent on the nature of the soft segment and diisocyanate starting materials. The PU containing a polyester soft segment exhibited a defined melting point at 49 °C. Mechanical stress-strain analysis of the series of PUs showed each exhibited greater than 70% reduction in toughness after treatment with TBAF for 30 min as a consequence of the chemo-responsive degradation of the polymer mainchain. The material featuring an ester-based polyol demonstrated excellent adhesion at bonding temperatures as low as 60 °C. Moreover, this material could be thermally rebonded if broken by force without loss in adhesion strength over three debond-rebond cycles. Lap shear adhesion tests showed a reduction in adhesive strength of approximately 40% (from 11.4 MPa to 7.3 MPa) on exposure to fluoride ions. [ABSTRACT FROM AUTHOR]

Periprosthetic acetabular fractures are rare but potentially devastating complications of total hip arthroplasty. As the number of total hip arthroplasties performed annually increases, so has the incidence of periprosthetic fractures, with the topic being spotlighted more frequently in the orthopaedic community. There is a particular sparsity of literature regarding periprosthetic acetabular fractures, with periprosthetic femoral fractures after total hip arthroplasty being traditionally far more commonly reported. This article aims to provide an up-to-date review of the epidemiology, risk factors, diagnostic challenges, classifications, and management strategies for periprosthetic acetabular fractures after total hip arthroplasty. Purpose: T-regulatory cells (Tregs) are a sub-population of lymphocytes that act to suppress aberrant immune responses. We investigated changes in the numbers of naïve and terminally differentiated Tregs in the peripheral blood to establish their role in the immuno-suppressive response to prolonged exercise.Methods: Blood was drawn from seventeen experienced runners (age 40 ± 12 years; height 1.75 ± 0.08 m; mass 71.4 ± 10.8 kg) before, ~1 h after (POST-1h), and on the day following the marathon (POST-1d). Tregs (CD3+CD4+Foxp3+CD25++CD127-) were analysed in peripheral blood mononuclear cells using flow cytometry. The markers CD45RA and HLA-DR were included to define naïve and terminally differentiated Tregs, respectively.Results: The absolute number of Tregs decreased (27%) POST-1h marathon (P < 0.001) but increased (21%) at POST-1d (P < 0.01). Naïve CD45RA+ Tregs fell by 39% POST-1h (P < 0.01) but were unaffected POST-1d (P > 0.05). In contrast, an increased number of Tregs expressing HLA-DR was observed at POST-1d (P < 0.01). Interleukin (IL)-1β, IL-6, IL-8 and IL-10 levels in the serum all increased POST-1h (P > 0.05) but returned to pre-exercise levels POST-1d. The suppressive cytokine, transforming growth factor-beta, was unaffected by the marathon (P > 0.05).Conclusions: These results suggest that Tregs do not play a major role in immune suppression in the early hours of recovery from a marathon. However, terminally differentiated HLA-DR+ Tregs are mobilized the following day, which could represent a compensatory attempt by the host to restore immune homeostasis and limit excessive cell damage. The aim of this article is to explore the link between different notions of co‐production. It seeks to emphasise the underlying politics of co‐production in the sense of who defines co‐production, especially in relation to initial decisions concerning which specific policy areas are deemed suitable for codesigning, cocreating, or codelivering with services users or local communities. We argue that the rejection of co‐production by government may inflame political resentment and reconfirm negative pre‐existing attitudes about "the establishment." This is particularly problematic when politicians have promoted the rhetoric of "inclusive governance," "sharing power," or "delegating power" but then reject the co‐productive claims emerging from such statements. The study contributes to existing work by analysing what happens when co‐productive structures are terminated or when public protests demand the reinstitutionalisation of those relationships. We make this contribution by presenting findings from an ethnographic case study involving street trees in a large English city. We suggest these specific findings have a broader relevance. Arguments about the legitimate role of expert bodies in Europe often centre on the following question: Does their independence help to make policies credible or should they be made democratically accountable to principals and stakeholders? This article claims this is a false dichotomy. It does so by arguing theoretically that credibility can be achieved through accountability processes. Then, drawing on exemplary case studies, this article identifies distinctive accountability processes for ensuring credibility: revisable competencies, deliberation over institutional design, and engagement in public justification. Credibility and accountability are thus not conflicting, but coconstitutive aims of delegation to expert bodies. This study sought to develop knowledge about understandings of sexuality and appearance by using a story completion task combined with an innovative visual methodology. Fifty-four (mainly female) participants were randomly assigned to complete a story about a fictional bisexual, lesbian, or heterosexual character who is going on a date, and then asked to build a Bitstrips online avatar of their character. Our social constructionist thematic analysis identified that looking good was a common feature of the stories and that this was understood in largely heteronormative terms. This imperative was also clear in stories of the date, which relied on traditional notions of gender and heterosexuality. Finally, individuality and authenticity were rewarded in participants' stories, yet this was simultaneously constrained by (dominant) heteronormative narratives. We conclude that combining story completion with visual methods enabled deeper insights into representations of appearance, dating, and relationships, than a textual method alone. Abstract Background One potential treatment strategy to enhance axon regeneration is transplanting Schwann Cells (SCs) that overexpress glial cell line-derived neurotrophic factor (GDNF). Unfortunately, constitutive GDNF overexpression in vivo can result in failure of regenerating axons to extend beyond the GDNF source, a phenomenon termed the "candy-store" effect. Little is known about the mechanism of this axon entrapment in vivo. New Method We present a reproducible in vitro culture platform using a microfluidic device to model axon entrapment and investigate mechanisms by which GDNF causes axon entrapment. The device is comprised of three culture chambers connected by two sets of microchannels, which prevent cell soma from moving between chambers but allow neurites to grow between chambers. Neurons from dorsal root ganglia were seeded in one end chamber while the effect of different conditions in the other two chambers was used to study neurite entrapment. Results The results showed that GDNF-overexpressing SCs (G-SCs) can induce axon entrapment in vitro. We also found that while physiological levels of GDNF (100 ng/mL) promoted neurite extension, supra-physiological levels of GDNF (700 ng/mL) induced axon entrapment. Comparison with Existing Method All previous work related to the "candy-store" effect were done in vivo. Here, we report the first in vitro platform that can recapitulate the axonal entrapment and investigate the mechanism of the phenomenon. Strength-based assessment has achieved acceptance from educational, mental health, and social service professionals as a means to measuring emotional and behavioral strengths of children. Several standardized, norm-referenced tests have been developed to assess these strengths; however, the primary mode of assessment is via informal interviews of parents and caregivers. In many cases, these interviews lack psychometric support, including basic validity and reliability. The purpose of this study was to document the multistep process used to establish the content validity of a newly developed instrument: Behavioral and Emotional Rating Scale: Strength-Based Interview. Specifically, the results of a survey of 40 U.S. professional mental health staff and educators are described where respondents rated the importance of interview questions to the construct of strength-based assessment. The findings of the survey as well as the 18 items deemed most important are reported. Abstract: Astroblastoma is a rare and controversial glioma with variable clinical behavior. The diagnosis currently rests on histologic findings of a circumscribed glioma with astroblastomatous pseudorosettes and vascular hyalinization. Immunohistochemical studies have suggested different oncogenic drivers, such as BRAF p.V600E, but very few cases have been studied using genome‐wide methodologies. Recent genomic profiling identified a subset of CNS embryonal tumors with astroblastoma‐like morphology that harbored MN1 gene fusions, termed “CNS high‐grade neuroepithelial tumors with MN1 alteration” (CNS‐HGNET‐MN1). To further characterize the genetic alterations that drive astroblastomas, we performed targeted next‐generation sequencing (NGS) of 500 cancer‐associated genes in a series of eight cases. We correlated these findings with break‐apart fluorescence in situ hybridization (FISH) analysis of the MN1 locus and genome‐wide DNA methylation profiling. Four cases showed MN1 alteration by FISH, including two pediatric cases that lacked other pathogenic alterations, and two adult cases that harbored other cancer‐associated gene mutations or copy number alterations (eg, CDKN2A/B homozygous deletion, TP53, ATM and TERT promoter mutations). Three of these cases grouped with the CNS‐HGNET‐MN1 entity by methylation profiling. Two of four MN1 intact cases by FISH showed genetic features of either anaplastic pleomorphic xanthoastrocytoma (BRAF p.V600E mutation, CDKN2A/B homozygous deletion and TERT promoter mutation) or IDH‐wildtype glioblastoma (trisomy 7, monosomy 10, CDK4 amplification and TP53, NRAS and TERT promoter mutations) and these cases had an aggressive clinical course. Two clinically indolent cases remained unclassifiable despite multimodal molecular analysis. We conclude that astroblastoma histology is not specific for any entity including CNS‐HGNET‐MN1, and that additional genetic characterization should be considered for astroblastomas, as a number of these tumors likely contain a methylation profile or genetic alterations that suggest classification as other tumor entities. Antisense oligonucleotides (ASOs) were first discovered to influence RNA processing and modulate protein expression over two decades ago; however, progress translating these agents into the clinic has been hampered by inadequate target engagement, insufficient biological activity, and off-target toxic effects. Over the years, novel chemical modifications of ASOs have been employed to address these issues. These modifications, in combination with elucidation of the mechanism of action of ASOs and improved clinical trial design, have provided momentum for the translation of ASO-based strategies into therapies. Many neurological conditions lack an effective treatment; however, as research progressively disentangles the pathogenic mechanisms of these diseases, they provide an ideal platform to test ASO-based strategies. This steady progress reached a pinnacle in the past few years with approvals of ASOs for the treatment of spinal muscular atrophy and Duchenne muscular dystrophy, which represent landmarks in a field in which disease-modifying therapies were virtually non-existent. With the rapid development of improved next-generation ASOs toward clinical application, this technology now holds the potential to have a dramatic effect on the treatment of many neurological conditions in the near future. miRNAs are small, noncoding RNAs that not only regulate gene expression within cells, but might also constitute promising extracellular biomarkers for a variety of pathologies, including the progressive muscle-wasting disorder Duchenne Muscular Dystrophy (DMD). A set of muscle-enriched miRNAs, the myomiRs (miR-1, miR-133, and miR-206) are highly elevated in the serum of patients with DMD and in dystrophin-deficient animal models. Furthermore, circulating myomiRs might be used as pharmacodynamic biomarkers, given that their levels can be restored towards wild-type levels following exon skipping therapy in dystrophic mice. The relationship between muscle pathology and extracellular myomiR release is complex, and incompletely understood. Here, we discuss current progress leading towards the clinical utility of extracellular miRNAs as putative DMD biomarkers, and their possible contribution to muscle physiology. The structures of the hydrochloride hydrates of pentylone and dibutylone and the hydrochloride salt of ephylone, three new clandestinely-manufactured designer cathinones are described. Pentylone is (±)-1-(1,3-benzodioxol-5-yl)-2-(methylamino)pentan-1-one, dibutylone is (±)-1-(1,3-benzodioxol-5-yl)-2-(dimethylamino)butan-1-one, and ephylone is (±)-1-(1,3-benzodioxol-5-yl)-2-(ethylamino)pentan-1-one. These three drugs have recently appeared in law enforcement seizures of illicit drug materials and are referred to as NPS (new psychoactive substances). The free base pentylone crystallizes as the hydrochloride monohydrate salt with H bonds extending from the protonated amine to the Cl anion very close to a center of symmetry, thence to a water of hydration, resulting in an interesting six-sided species of approximately chair conformation whose components are linked by hydrogen bonds. An interesting feature of the dibutylone free base is that it crystallizes as the HCl sesquihydrate salt. Most of the street drugs crystallize as their hydrates; but, unlike most of them, dibutylone HCl 1.5HO crystallizes as a kryptoracemate in space group P222. A kryptoracemic crystal is one whose asymmetric unit contains a racemic pair, but still belongs in a Sohncke space group. The phenomenon was recognized relatively recently and, to date, there are very few drugs identified as crystallizing thus (for more on this topic, see below). In this structure, both independent amine cations have a chloride counter-anion, and they share three water molecules of crystallization, with the chloride anions and the waters forming an interesting H-bonded cyclic array. The free base ephylone crystallizes as the HCl salt and is a brand new substance, not appearing until about November 2015. The packing of all three of these cathinones contains 'sandwich' π-π stacking interactions where the phenyl rings of the cations are stacked face-to-face with each other, with distances ranging from 3.531 to 3.862 Å. These compounds are among the most recent examples of cathinone-based stimulants designed to circumvent existing drug laws, and they represent a new danger to recreational drug users. Navigating large-scale environments involves dynamic interactions between the physical world and individuals’ knowledge, goals, and strategies. Time pressure can result from self-imposed goals or relatively dynamic situational factors that induce varied constraints. While time pressure is ubiquitous in daily life and has been shown to influence affective states, cost-benefit analyses, and strategy selection, its influence on navigation behaviour is unknown. The present study examined how introducing varied time constraints during virtual urban navigation would influence spatial strategies and impact the efficiency and effectiveness of goal-directed wayfinding. Participants learned a large-scale urban virtual environment by wayfinding between a series of 20 successive landmark goals (e.g.,You have reached the Theater. Now find the Bank.). A day later, they again performed the same task, but landmark-to-landmark trials were characterized by conditions of low-, moderate-, or high-pressure time limits as quantified by a pilot experiment. As time pressure increased, participants more likely navigated along previously experienced paths and less likely travelled in the global direction of the destination. Results suggest strategy shifts under time constraints that increase reliance on egocentric, route-based strategies and decrease reliance on global configural knowledge, probably in an attempt to reduce cognitive demands and support performance under pressure. Declining voter turnout has been highlighted as problematic for a number of western democracies. However, in this article we argue that whether an election is seen as ‘legitimate’ or not depends crucially upon interpretations of the levels of turnout by elite actors. Through comparing two recent democratic ballots in the UK we demonstrate how elections with lower turnouts can come to be seen as holding more legitimacy than those with higher turnouts. The cases demonstrate, we argue, a distinction between actual legitimacy, defined as a binary concept, and the process of legitimization – a process through which the authority of an institution is discursively constructed and conferred. This suggests a new research agenda which extends beyond the current literature to focus upon how the legitimacy of a ballot is socially constructed in a broader context of unequal socioeconomic power relations. Spinal muscular atrophy (SMA) is a genetic disorder with severity ranging from premature death in infants to restricted motor function in adult life. Despite the genetic cause of this disease being known for over twenty years, only recently has a therapy been approved to treat the most severe form of this disease. Here we discuss the genetic basis of SMA and the subsequent studies that led to the utilization of splice switching oligonucleotides to enhance production of SMN protein, which is absent in patients, through a mechanism of exon inclusion into the mature mRNA. Whilst approval of oligonucleotide-based therapies for SMA should be celebrated, we also discuss some of the limitations of this approach and alternate genetic strategies that are currently underway in clinical trials. Objective: This study examines the influence of weight bias and health consciousness on eating and dietary behavior; it also investigates the role of normative influences.Design and Methods: A sample of adults living in South-East England (N=498) completed anonymous questionnaires about their diet, attitudes and beliefs towards obese people, health consciousness and normative influences. The survey included validated measures of anti-fat attitudes, and beliefs about obese people.Results: The findings demonstrate anti-fat attitudes are positively related to self-perceived dietary behavior. Surprisingly, self-perceived dietary behavior is negatively related to health consciousness and activities designed to enable healthy eating, for example meal planning. Significant differences exist between people with, or without, obese family members.Conclusions: Attempts to improve eating behavior by raising health consciousness and offering related support activities may fail; promoting health eating may also contribute to weight bias in society. The concepts of 'politicisation' and 'depoliticisation' have become increasingly used by political scientists over the last decade, in light of the presumed 'triumph' of neoliberal ideology in the 1990s, and its subsequent challenge ('re-politicisation') by the 2008 financial crisis. Debate has emerged in the literature, however, over what conceptual strategy is best for critically analysing depoliticisation - 'broad' or 'narrow'. This article argues for attempting to find common ground among 'competing' definitions, by viewing politicisation and depoliticisation as 'multilevel' concepts, with theoretical, analytical and empirical purchase. The uniting theme, it is argued, is a critical analysis of the dynamics of neoliberalism as a paradigm of political thought, and how that paradigm allegedly leads to the rise of 'anti-politics'. Thinking about (de)politicisation in this way helps avoid facile conceptual reinvention and stimulates a coherent yet pluralistic research agenda focused on translating insights and criticism across macro, meso and micro levels. Existing research on depoliticisation emphasises how blame is shifted away from governments due to tactics employed by politicians. The argument of this article is that the analytical utility of this approach can be significantly enhanced by also examining the conduct of non-state actors, and broader contextual factors. Using a detailed case study of record flooding in the UK in 2007, the article shows how, despite significant failures in flood preparation, the agenda-setting and preference-shaping actions of politicians and the media, and the 'path-shaping' effects of a benign political context and volatile and unstable policy area, the government was largely absolved of blame in the eyes of the public. In particular, the article highlights a paradox of drivers that appear simultaneously politicising and depoliticising, but lead ultimately to depoliticisation. Somatic mutations of the H3F3A and HIST1H3B genes encoding the histone H3 variants, H3.3 and H3.1, were recently identified in high-grade gliomas arising in the thalamus, pons and spinal cord of children and young adults. However, the complete range of patients and locations in which these tumors arise, as well as the morphologic spectrum and associated genetic alterations remain undefined. Here, we describe a series of 47 diffuse midline gliomas with histone H3-K27M mutation. The 25 male and 22 female patients ranged in age from 2 to 65 years (median = 14). Tumors were centered not only in the pons, thalamus, and spinal cord, but also in the third ventricle, hypothalamus, pineal region and cerebellum. Patients with pontine tumors were younger (median = 7 years) than those with thalamic (median = 24 years) or spinal (median = 25 years) tumors. A wide morphologic spectrum was encountered including gliomas with giant cells, epithelioid and rhabdoid cells, primitive neuroectodermal tumor (PNET)-like foci, neuropil-like islands, pilomyxoid features, ependymal-like areas, sarcomatous transformation, ganglionic differentiation and pleomorphic xanthoastrocytoma (PXA)-like areas. In this series, histone H3-K27M mutation was mutually exclusive with IDH1 mutation and EGFR amplification, rarely co-occurred with BRAF-V600E mutation, and was commonly associated with p53 overexpression, ATRX loss (except in pontine gliomas), and monosomy 10. An ongoing study of oils and Paleozoic source rock extracts from the Anadarko Basin, Oklahoma, revealed the presence of multiple series of novel tricyclic terpanes in addition to the commonly observed series of 13β(H),14α(H) tricyclic terpanes. The precise structures of these novel tricyclic compounds are not known but several proposed spectral and chromatographic similarities with known compounds suggests the tentatively identified analytes are rearranged and structural isomers of the 13β(H),14α(H) tricyclic terpanes. The occurrences and relative abundances of these compounds are predominantly lower in more carbonate-rich formations, which implies a potential role of clay minerals in their formation. The presence and distributions of these unique tricyclic terpane configurations are seemingly unaffected by thermal maturation, although relative concentrations may increase as a result of preferential thermal degradation of the less thermally-stable hopanes. Many of the samples bearing the novel rearranged tricyclic terpanes also contain varying proportions of four previously reported series of rearranged hopanes, which are formed via rearrangement reactions catalyzed by clay minerals. The rearranged hopanes include the commonly observed Ts and C 30 diahopanes in addition to 9,15-dimethyl rearranged hopanes and 21-methyl-28,30-dinorhopanes (28-norspergulanes) that have been previously reported in a number of basins worldwide but not the Anadarko Basin. No apparent relationship was identified between the presence of the rearranged tricyclic terpanes and the rearranged hopanes. Both groups require the presence of clay minerals, as determined from empirical relationships with source rock lithology, and are observed to co-occur in many of the samples studied. Still, the lack of a clear correlation between rearranged tricyclic terpanes and rearranged hopanes suggests additional controls, beyond lithology, on their genesis and preservation in the geologic record. In a supposedly 'anti-political' age, the scholarly literature on celebrity politicians argues that politicians gain popularity by adopting strategies from within the world of entertainment. This article offers the findings of a research project that has detected a marked shift in the interplay between celebrity culture and the presentational strategies adopted by politicians. At the heart of this shift is an increased focus on the concept of 'normality' as politicians increasingly attempt to shake-off the negative connotations associated with 'professional politicians' and instead attempt to appear 'just like us'. As such, this article offers an original approach by distinguishing between 'superstar' celebrity politicians and 'everyday' celebrity politicians before identifying three aspects of each strategy (i.e. media platform, marketing technique and performative role). It offers numerous empirical examples that serve to underpin this distinction before using the example of Boris Johnson as a case study in the attempted shift from 'superstar' to 'everyday' celebrity. This focus on normality offers a fresh entry-point into the analysis of contemporary political statecraft while also posing distinctive questions about the tension between political popularity and credibility in an anti-political age.
