We demonstrate directed translocation of ClO4− anions from cationic to neutral binding site along the synthetized BPym‐OH dye molecule that exhibits coupled excited‐state intramolecular proton‐transfer (ESIPT) and charge‐transfer (CT) reaction (PCCT). The results of steady‐state and time‐resolved spectroscopy together with computer simulation and modeling show that in low polar toluene the excited‐state redistribution of electronic charge enhanced by ESIPT generates the driving force, which is much stronger than by CT reaction itself and provides more informative gigantic shifts of fluorescence spectra signaling on ultrafast ion motion. The associated with ion translocation red‐shifted fluorescence band (at 750 nm, extending to near‐IR region) appears at the time ~83 ps as a result of electrochromic modulation of PCCT reaction. It occurs at substantial delay to PCCT that displayed fluorescence band at 640 nm and risetime of <200 fs. The dielectric materials utilized in high-performance barium titanate-based base-metal electrode multilayer ceramic capacitors (BME MLCCs) must satisfy stringent criteria such as high dielectric constant, low dielectric loss, high insulation resistivity, and reliability. In this investigation, the effects of raw materials, BaTiO 3 (BT), and Ba 0.95 Ca 0.05 TiO 3 (BCT) on densification, microstructure, and dielectric behavior were explored by introducing acceptor and rare earth elements. The findings revealed that BCT could attain densification at a lower sintering temperature of 1275 °C than BT. Moreover, BCT samples displayed relatively smaller grain sizes, resulting in a greater number of grain boundaries, which in turn increased the insulation resistivity. BCT had a lower oxygen vacancy concentration, thus resulting in lower leakage currents at high temperatures than BT. Substituting BCT for BT resulted in higher activation energies for conductivity in the core, shell, and grain boundary. Highly pathogenic avian influenza (HPAI) clade 2.3.4.4 viruses have been reported to be the source of infections in several outbreaks in the past decades. In a previous study, we screened out a broad-spectrum virus strain, H5N6-Sichuan subtype, by using a lentiviral pseudovirus system. In this project, we aimed to investigate the potential of H5N6 virus-like particles (VLPs) serving as a broad-spectrum vaccine candidate against H5Nx viruses. We cloned the full-length M1 gene and H5, N6 genes derived from the H5N6-Sichuan into pFASTBac vector and generated the VLPs using the baculovirus-insect cell system. H5N6 VLPs were purified by sucrose gradient centrifugation, and the presence of H5, N6 and M1 proteins was verified by Western blot and SDS-PAGE. The hemagglutination titer of H5N6 VLPs after purification reached 5120 and the particle structure remained as viewed by electron microscopy. The H5N6 VLPs and 293T mammalian cell-expressed H5+N6 proteins were sent for mice immunization. Antisera against the H5+N6 protein showed 80 to 320 neutralizing antibody titers to various H5Nx pseudoviruses. In contrast, H5N6 VLPs not only elicited higher neutralizing antibody titers, ranging from 640 to 1280, but also induced higher IL-2, IL-4, IL-5, IFN-γ and TNF production, thus indicating that H5N6 VLPs may be a potential vaccine candidate for broad-spectrum H5Nx avian influenza vaccines. The importance of pancreatic endocrine cell activity modulation by autonomic innervation has been debated. To investigate this question, we established an in vivo imaging model that also allows chronic and acute neuromodulation with genetic and optogenetic tools. Using the GCaMP6s biosensor together with endocrine cell fluorescent reporters, we imaged calcium dynamics simultaneously in multiple pancreatic islet cell types in live animals in control states and upon changes in innervation. We find that by 4 days post fertilization in zebrafish, a stage when islet architecture is reminiscent of that in adult rodents, prominent activity coupling between beta cells is present in basal glucose conditions. Furthermore, we show that both chronic and acute loss of nerve activity result in diminished beta-beta and alpha-beta activity coupling. Pancreatic nerves are in contact with all islet cell types, but predominantly with beta and delta cells. Surprisingly, a subset of delta cells with detectable peri-islet neural activity coupling had significantly higher homotypic coupling with other delta cells suggesting that some delta cells receive innervation that coordinates their output. Overall, these data show that innervation plays a vital role in the maintenance of homotypic and heterotypic cellular connectivity in pancreatic islets, a process critical for islet function. Lately, carbazole‐based self‐assembled monolayers (SAMs) are widely employed as effective hole‐selective layers (HSLs) in inverted perovskite solar cells (PSCs). Nevertheless, these SAMs tend to aggregate in solvents due to their amphiphilic nature, hindering the formation of a monolayer on the ITO substrate and impeding effective passivation of deep defects in the perovskites. In this study, a series of new SAMs including DPA‐B‐PY, CBZ‐B‐PY, POZ‐B‐PY, POZ‐PY, POZ‐T‐PY, and POZ‐BT‐PY are synthesized, which are employed as interfacial repairers and coated atop CNph SAM to form a robust CNph SAM@pseudo‐planar monolayer as HSL in efficient inverted PSCs. The CNph SAM@pseudo‐planar monolayer strategy enables a well‐aligned interface with perovskites, synergistically promoting perovskite crystal growth, improving charge extraction/transport, and minimizing nonradiative interfacial recombination loss. As a result, the POZ‐BT‐PY‐modified PSC realizes an impressively enhanced solar efficiency of up to 24.45% together with a fill factor of 82.63%. Furthermore, a wide bandgap PSC achieving over 19% efficiency. Upon treatment with the CNph SAM@pseudo‐planar monolayer, also demonstrates a non‐fullerene organic photovoltaics (OPVs) based on the PM6:BTP‐eC9 blend, which achieves an efficiency of 17.07%. MALT1 controls several receptors-mediated signaling to nuclear factor κB (NF-κB) through both its scaffold and protease function. MALT1 protease activity is shown to inactivate several negative regulators of NF-κB signaling and augment NF-κB activation ability. In this study, MALT1 was demonstrated to autoprocess itself in the presence of oligomerization-competent BCL10. Cleavage occurred after Arginine 781 located in the C-terminus of MALT1. Shortened MALT1 cleavage products showed attenuated binding ability with TRAF6. Its NF-κB activation ability was also weakened. Various MALT1 constructs including wild type, catalytically-inactive (MALT1_C464A), cleavage-defective (MALT1_R781L), or truncated (MALT1_1–781) form of MALT1 was introduced into MALT1-knocked-down-Jurkat T cells. Cleavage-defective MALT1_R781L retained its proteolytic and initial IκBα phosphorylation activity as MALT1. Truncated MALT1_1–781 mutant showed weakness in IκBα phosphorylation and the expression of NF-κB targets IL-2 and IFN-γ. Cleavage at R781 was detectable but marginal after activation with TPA/ionomycin or anti-CD3 antibody in lymphocytes. However, cleavage at R781 was evident in ABC-DLBCL cells such as OCI-Ly3, HBL-1. HBL-1 cells with induced expression of catalytically-inactive MALT1_C464A or cleavage-defective MALT1_R781L exhibited characteristic of retarded-growth. This study investigated the effects of calcination treatment on the densification, microstructure, and dielectric behavior of Ba 0.95 Ca 0.05 TiO 3 (BCT) doped with acceptor and rare earth elements. The calcination process promoted sintering densification and enabled the densification of the green body at lower sintering temperatures. The electrical properties of the calcined samples showed higher dielectric constant, lower losses, and higher resistivity than the uncalcined samples. The increased conduction activation energies observed in the core and shell of the calcined BCT samples were attributed to the comparatively lower concentrations of Ti3+ and oxygen vacancy. The calcined BCT samples exhibited higher resistivity at room temperature and lower leakage currents at high temperatures, indicating superior reliability. Patients with end-stage kidney disease (ESKD) rely on dialysis to remove toxins and stay alive. However, hemodialysis alone is insufficient to completely remove all/major uremic toxins, resulting in the accumulation of specific toxins over time. The complexity of uremic toxins and their varying clearance rates across different dialysis modalities poses significant challenges, and innovative approaches such as microfluidics, biomarker discovery, and point-of-care testing are being investigated. This review explores recent advances in the qualitative and quantitative analysis of uremic toxins and highlights the use of innovative methods, particularly label-mediated and label-free surface-enhanced Raman spectroscopy, primarily for qualitative detection. The ability to analyze uremic toxins can optimize hemodialysis settings for more efficient toxin removal. Integration of multiple omics disciplines will also help identify biomarkers and understand the pathogenesis of ESKD, provide deeper understanding of uremic toxin profiling, and offer insights for improving hemodialysis programs. Distance estimation based on the received signal strength (RSS) of wireless radio has been extensively studied; however, the estimation accuracy is susceptible to radio propagation conditions, particularly during periods of precipitation. This study performed experiments on the 1.8-GHz band of Global System for Mobile Communications (GSM) networks at Yuan Ze University, Taoyuan, Taiwan, sensing actual radio power signals using mobile phones and recording transmission conditions during various weather events. The aim was to compare RSS performance under dry and wet weather conditions and study the dynamics of distance estimation from theoretical and empirical perspectives. Traditional rain attenuation models have largely discounted the impact of precipitation on GSM signals due to the relatively low frequency band; however, on-site experimental results demonstrated that dynamic weather significantly degrades accuracy in distance estimation. This study employed a typical rain attenuation model and a radio propagation model to evaluate errors in distance estimation under various rain conditions, i.e., heavy rain, extremely heavy rain, torrential rain, and extremely torrential rain, in accordance with the standards set out by the Central Weather Bureau, Taiwan. This study aims to use power ultrasound-assisted extraction (UAE) and maceration (ME) to valorize lemon juice processing residues (flavedo+albedo). Also, the effects of maturity stage (mature vs. immature) and drying method (freeze-drying-FD vs. oven drying-OD) on antioxidant activity, digestive enzymes inhibition, and bioactive compounds (phenolics and flavonoids) were analyzed. Besides. effects of independent parameters were elaborated through principal component analysis (PCA). According to the results, immature lemons exhibited a higher inhibition of α-amylase, α-glucosidase, and lipase by 10, 5, and 46%. Also, replacing OD with FD enhanced the antioxidant activity. HPLC identified hesperidin and sinapic acid as the most abundant bioactive compounds in all extracts and UAE enhanced their concentration by 2.3 and 1.2 times, respectively. Besides, sonication enhanced antioxidant activity and anti-diabetic effects by 1.1–5.6 times. Therefore, replacing mature peels, OD, and ME with immature peels, FD, and UAE can boost the potential health benefits of extracts of lemon processing by-product. In this paper, The advantage of ternary metal sulfides for the detection of various gas at extremely low concentrations has been explored. Ultrafine TiO 2 /PbSnS based thin film has been fabricated by the Successive Ionic Layer Adsorption and Reaction (SILAR) method and employed as an effective CO and NO 2 sensor. The advanced material characterization technique such as HRTEM and XRD revealed that high-quality PbSnS nanoparticle with a diameter of 19.7 nm and minimum crystallite size of 19.34 nm were obtained. The bandgap of the synthesizing material was estimated by using data of UV visible spectroscopy and reduces from 3.3 eV to 2.3 eV by adding ternary metal sulfide (PbSnS) in TiO 2. The sensor also showed the best sensing performance with sensor response 8.3 at 60 ppm of CO and 0.24 at 100 ppb of NO 2 at room temperature. The minimum response and recovery time was calculated at 25 ppm and found to be 198 s and 36 s for CO and 16.03 s and 27 s for NO 2 23 ppb respectively and the limit of detection (LOD) is extremely low i. e. 3.89 ppm of carbon monoxide and 3.51 ppb for NO 2 at room temperature. Density Function Theory has been used to explain the possible sensing mechanism for the sensing of carbon mono oxide and NO 2 using PbSnS nanomaterial. In this study, a bamboo stick board with rotary-cut bamboo veneers was successfully fabricated. Additionally, vacuum heat (VH) treatment, which is a popular thermal modification method, was used to modify bamboo sticks. Therefore, the effects of different VH treatment temperatures on the dimensional stability and flexural properties of bamboo stick boards with and without bamboo veneers were investigated. For all boards, as the temperature increased to 220 °C, the thickness change rate and equilibrium moisture content decreased, and the flexural properties increased. The results exhibited that VH treatment improved the dimensional stability and flexural properties of the boards. Furthermore, the board with veneers had lower flexural properties and higher thickness swelling after water absorption than the board without veneers (BSB). The results indicated that bamboo veneer caused low flexural properties and high thickness swelling of the board compared to the BSB. Fresh-cut vegetables have attracted consumer attention due to their convenience; however, found to be less shelf-life. Currently, few studies attempted to extend shelf-life of produces by the application of novel technologies. Thus, we employed modified atmosphere packaging (MAP) and high-voltage electrostatic field (HVEF, kV/m × time) to extend the shelf-life of fresh-cut cabbage for 60 days & baby corn for 48 days and evaluated their quality and functional properties. Samples were treated with CO 2 and O 2 ratio of 1:1 (MAP1) and 1:0.70 (MAP2) for fresh-cut cabbage; 2:1 (MAP3) and 1:0.80 (MAP4) for baby corn, followed by HVEF (strengths 6000 (HVEF6) for fresh-cut cabbage and 4500 (HVEF45) for baby corn) treatment at 4 °C. As storage time progressed, fresh-cut cabbage treated with MAP1 + HVEF6 and baby corn treated with MAP3 + HVEF45 showed good quality (no color change and lower electrolyte leakage) and functional properties (SOD-like activity). Sensory evaluation of samples treated with MAP1 + HVEF6 and MAP3 + HVEF45 received higher rating for overall acceptability. The synergies of MAP and HVEF could prolong the shelf-life of fresh-cut cabbage up to 60 days and baby corn until 48 days. Atmospheric polychlorinated-dibenzo-dioxins/dibenzo-furans (PCDD/Fs) remains an important environmental health concern. Although the total emission inventories of PCDD/Fs in Taiwan decreased from 320 to 52.1 g-I-TEQ/year during 2002–2016, the resulting concentrations of atmospheric PCDD/F and distributions in Taiwan are unknown. We, therefore, conducted a comprehensive investigation of spatial and seasonal variations and apportioned potential sources of ambient PCDD/F concentrations in Taiwan-based on 11-year observation data. A total of 1,008 atmospheric PCDD/F samples were collected from 25 air monitoring stations (from seven areas) and 1 background station for 2006–2016. Linear regression was used to model changes in PCDD/F concentrations. Principal component analysis (PCA) and positive matrix factorization (PMF) were used to identify potential contributors. PCDD/F concentrations in the ambient air gradually decreased during the study period, with a median concentration of 28.2 fg I-TEQ/m3 over 11 years. The highest median PCDD/F concentrations were found in the highly industrialized regions of western Taiwan (38.0–43.4 fg I-TEQ/m3). Lower concentrations were found in eastern Taiwan (∼10 fg I-TEQ/m3). Background stations reported the lowest concentrations of PCDD/Fs, with a median concentration of 1.47 fg I-TEQ/m3. Overall, the concentrations of atmospheric PCDD/Fs in Taiwan were higher in winter (13.4–86.7 fg I-TEQ/m3) than in summer (9.65–27.2 fg I-TEQ/m3). The PCA results indicated that PCDD/F profiles varied by both region (industrialized, urbanized, and background areas) and season. The PMF model for the overall data revealed that the major sources of PCDD/Fs were industrial activities (71.2%). However, in less industrialized areas, traffic activities, long-range transport, and open burning were dominant. • Atmospheric PCDD/Fs widely ranged from 0.97 to 255 fg I-TEQ/m3. • Annual median of ambient PCDD/Fs dropped from 43.9 to 18.2 fg I-TEQ/m3 in 11 years. • Highest level of PCDD/Fs were in western Taiwan spatially and in winter seasonally. • PMF showed major sources to be industry (71.2%), LRT (28.6%), and traffic (2.2%). (1) Background/aims: Intragastric botulinum toxin A injection (IGBI) combined with diet control is a new and effective weight loss method for grade 2 obese patients. However, the application of IGIB on overweight or obese adults still needs further research to confirm its efficacy. (2) Methods: We retrospectively collected medical data from 1 July 2021 to 1 January 2022 from a total of 71 patients without diabetes who participated in the bariatric clinic with a body mass index (BMI) > 25 kg/m2. Forty-nine participants opted for intragastric botulinum injection (IGBI) using 300 units of botulinum injected into the antrum, body, and fundus, followed with a low-calorie high-protein diet course. Another 22 people participated only in the low-calorie high-protein diet course as a placebo group. This study analyzes the weight loss percentage of the two groups. Adverse events after IGBI are also reported in a safety assessment. (3) Results: In terms of the characteristics of the two groups, the mean BMI was 29.3 kg/m2 in the IGBI group and 28.0 kg/m in the placebo group (p = 0.63 without significant difference). Comparing the percent weight loss from baseline in the two groups after 12 weeks, the IGBI group lost 11.5% of their body weight and the placebo group lost 1.8%. In terms of group analysis, the percentages of participants with a weight reduction of at least 5% for the IGBI and placebo groups were 95% and 4%, respectively. For weight reduction of at least 10%, these values for the IGBI and placebo groups were 63% and 4%, respectively. In terms of adverse events after IGBI for 12 weeks, 12 participants (24.4%) had constipation, which was the main side effect. No serious adverse events were observed during the study period. (4) Conclusion: The combination of a low-calorie high-protein diet and IGBI is an effective and safe procedure in overweight or obese adults for weight reduction, but further larger studies are needed. In this study, hydrogen-rich water (HRW) was obtained by reacting hybrid magnesium–carbon hydrogen storage materials with water. Then, the effects of intake of HRW on the activities of xenobiotic-metabolizing enzymes, membrane transporters, and oxidative stress in rats were investigated. Rats were given HRW ad libitum for four weeks. The results showed that intake of HRW had no significant effect on the activities of various cytochrome P450 (CYP) enzymes (CYP1A1, 1A2, 2B, 2C, 2D, 2E1, 3A, and 4A), glutathione-S-transferase, and Uridine 5′-diphospho (UDP)-glucuronosyltransferase. Except for a mild lower plasma glucose concentration, intake of HRW had no effect on other plasma biochemical parameters in rats. p-Glycoprotein and multidrug resistance-associated protein (Mrp) 2 protein expressions in liver were elevated after intake of HRW. However, HRW had no significant effects on glutathione, glutathione peroxidase, or lipid peroxidation in liver. The results from this study suggest that consumption of HRW may not affect xenobiotic metabolism or oxidative stress in liver. 6‐Cyano‐7‐aminoquinoline (6CN−7AQ) and 3‐cyano‐7‐aminoquinoline (3CN−7AQ) were synthesized and found to exhibit intense emission with quantum yield as high as 63 % and 85 %, respectively, in water. Conversely, their derivatives 6‐cyano‐7‐azidoquinoline (6CN−7N3Q) and 3‐cyano‐7‐azidoquinoline (3CN−7N3Q) show virtually no emission, which makes them suitable to be used as recognition agents in azide reactions based on fluorescence recovery. Moreover, conjugation of 6CN−7AQ with a hydrophobic biomembrane‐penetration peptide PFVYLI renders a nearly non‐emissive 6CN−7AQ‐PFVYLI composite, which can be digested by proteinase K, recovering the highly emissive 6CN−7AQ with ∼200‐fold enhancement. Quantum dots (QDs) thin films based on binary metal sulfides (PbS) and ternary metal sulfides (PbSbS) were fabricated utilizing the Successive Ionic Layer Adsorption and Reaction (SILAR) method. Using the Tauc plot, 2.32 eV and 3.22 eV optical bandgaps were calculated for TiO 2 /Pb 5 Sb 8 S 17 (TiO 2 /PbSbS) and TiO 2 /PbS, respectively. The sensing performance of TiO 2 /PbSbS and TiO 2 /PbS QDs has been investigated for the detection of carbon monoxide (CO) gas at room temperature (300 K). At 25 ppm, ∼2.5 times higher CO gas response was measured for TiO 2 /PbSbS (0.63) than for TiO 2 /PbS (0.2469) QDs with response/recovery time (t res. / t rec) 14.8/10.21 s. Furthermore, TiO 2 /PbSbS QDs showed a good response of 0.247 with t res. / t rec 20 /9 s at 5 ppm. These results provide a simple and highly scalable approach to developing high-performance gas sensors. To understand the gas sensing phenomena of TiO 2 /PbSbS and TiO 2 /PbS, a theoretical model has been developed. Background Intradialytic hypotension (IDH) is a common hemodialysis complication causing adverse outcomes. Despite the well-documented associations of ambient temperatures with fluid removal and pre-dialysis blood pressure (BP), the relationship between ambient temperature and IDH has not been adequately studied. Methods We conducted a cohort study at a tertiary hospital in southern Taiwan between 1 January 2016 and 31 October 2021. The 24-h pre-hemodialysis mean ambient temperature was determined using hourly readings from the weather station closest to each patient's residence. IDH was defined using Fall40 [systolic BP (SBP) drop of ≥40 mmHg] or Nadir90/100 (SBP <100 if pre-dialysis SBP was ≥160, or SBP <90 mmHg). Multivariate logistic regression with generalizing estimating equations and mediation analysis were utilized. Results The study examined 110 400 hemodialysis sessions from 182 patients, finding an IDH prevalence of 11.8% and 10.4% as per the Fall40 and Nadir90/100 criteria, respectively. It revealed a reverse J-shaped relationship between ambient temperature and IDH, with a turning point around 27°C. For temperatures under 27°C, a 4°C drop significantly increased the odds ratio of IDH to 1.292 [95% confidence interval (CI) 1.228 to 1.358] and 1.207 (95% CI 1.149 to 1.268) under the Fall40 and Nadir90/100 definitions, respectively. Lower ambient temperatures correlated with higher ultrafiltration, accounting for about 23% of the increased IDH risk. Stratified seasonal analysis indicated that this relationship was consistent in spring, autumn and winter. Conclusion Lower ambient temperature is significantly associated with an increased risk of IDH below the threshold of 27°C, irrespective of the IDH definition. In vehicular ad hoc networks (VANETs), network topology and communication links frequently change due to the high mobility of vehicles. Key challenges include how to shorten transmission delays and increase the stability of transmissions. When establishing routing paths, most research focuses on detecting traffic and selecting roads with higher vehicle densities in order to transmit packets, thus avoiding carry-and-forward scenarios and decreasing transmission delays; however, such approaches may not obtain accurate real-time traffic densities by periodically monitoring each road because vehicle densities change so rapidly. In this paper, we propose a novel routing information system called the machine learning-assisted route selection (MARS) system to estimate necessary information for routing protocols. In MARS, road information is maintained in roadside units with the help of machine learning. We use machine learning to predict the moves of vehicles and then choose some suitable routing paths with better transmission capacity to transmit packets. Further, MARS can help to decide the forwarding direction between two RSUs according to the predicted location of the destination and the estimated transmission delays in both forwarding directions. ER stress and apoptosis contribute to the loss of pancreatic β-cells under pro-diabetic conditions of glucolipotoxicity. Although activation of canonical intrinsic apoptosis is known to require pro-apoptotic Bcl-2 family proteins Bax and Bak, their individual and combined involvement in glucolipotoxic β-cell death are not known. It has also remained an open question if Bax and Bak in β-cells have non-apoptotic roles in mitochondrial function and ER stress signaling, as suggested in other cell types. Using mice with individual or combined β-cell deletion of Bax and Bak, we demonstrated that glucolipotoxic β-cell death in vitro occurs by both non-apoptotic and apoptotic mechanisms, and the apoptosis could be triggered by either Bax or Bak alone. In contrast, they had non-redundant roles in mediating staurosporine-induced apoptosis. We further established that Bax and Bak do not affect normal glucose-stimulated β-cell Ca2+ responses, insulin secretion, or in vivo glucose tolerance. Finally, our experiments revealed that combined deletion of Bax and Bak amplified the unfolded protein response in islets during the early stages of chemical- or glucolipotoxicity-induced ER stress. 7‐Aminoquinoline (7AQ) and various amino derivatives thereof (‐NHR) have been strategically designed and synthesized to study their excited‐state proton‐transfer (ESPT) properties. Due to the large separation between the proton donor ‐NHR and the acceptor ‐N‐ site, ESPT in 7AQ derivatives, if available, should proceed under protic solvent catalysis. ESPT is found to be influenced by the acidity of ‐NHR and the basicity of the proton‐acceptor ‐N‐ in the quinoline moiety. The latter is varied by the resonance effect at the quinoline ‐N‐ site induced by the ‐NHR substituent. For those 7AQ derivatives undergoing ESPT, increased quinoline basicity results in a faster rate of ESPT, implying that proton donation from methanol to the quinoline moiety may serve as a key step in the process. Our studies also indicate the existence of an equilibrium between cis and trans arrangements of ‐NHR in terms of its hydrogen‐bond (H‐bond) configuration with methanol, whereby only the cis‐H‐bonded form undergoes methanol‐assisted ESPT. With one exception, the interconversion between cis and trans configurations is much faster than the rate of ESPT, yielding amino‐type (normal form) and imine‐type (proton‐transfer tautomer) emissions with distinct relaxation dynamics. It is generally established that PCDD/Fs is harmful to human health and therefore extensive field research is necessary. This study is the first to use a novel geospatial-artificial intelligence (Geo-AI) based ensemble mixed spatial model (EMSM) that integrates multiple machine learning algorithms and geographic predictor variables selected using SHapley Additive exPlanations (SHAP) values to predict spatial-temporal fluctuations in PCDD/Fs concentrations across the entire island of Taiwan. Daily PCDD/F I-TEQ levels from 2006 to 2016 were used for model construction, while external data was used for validating model dependability. We utilized Geo-AI, incorporating kriging, five machine learning, and ensemble methods (combinations of the aforementioned five models) to develop EMSMs. The EMSMs were used to estimate long-term spatiotemporal variations in PCDD/F I-TEQ levels, considering in-situ measurements, meteorological factors, geospatial predictors, social and seasonal influences over a 10-year period. The findings demonstrated that the EMSM was superior to all other models, with an increase in explanatory power reaching 87 %. The results of spatial-temporal resolution show that the temporal fluctuation of PCDD/F concentrations can be a result of weather circumstances, while geographical variance can be the result of urbanization and industrialization. These results provide accurate estimates that support pollution control measures and epidemiological studies. Aryl hydrocarbon receptor (AHR), a ligand-activated transcription factor, has been studied extensively in carcinogenesis through the genomic pathway. In recent years, AHR has also been reported to exert positive or negative effects on epithelial-mesenchymal transition (EMT), the crucial step in tumor malignant progression. However, the detailed mechanism remains controversial. Analysis of AHR-expression levels in non-small cell lung cancer cell lines and lung cancer tissues revealed an inverse correlation between AHR protein levels and tumor cell invasion and metastasis. Overexpression of wild-type AHR in H1299 cells (AHR poorly expressed, potently invasive) not only accelerated mesenchymal vimentin degradation, but also prevented cell invasion in vitro and in vivo. In the absence of AHR agonists, the overexpressed AHR protein was predominantly localized in the cytoplasm, where it interacted with vimentin and functioned as an E3 ubiquitin ligase. A 6-h incubation with the proteasome inhibitor MG-132 fully rescued vimentin from AHR-mediated proteasomal degradation. In AHR-overexpressing H1299 cells, either vimentin degradation or invasive suppression could be reversed when glycogen synthase kinase 3 beta (GSK3β) was inactivated by CHIR-99021 treatment. In contrast, silencing of AHR in A549 cells (AHR highly expressed, weakly invasive) resulted in the downregulation of epithelial biomarkers (E-cadherin and claudin-1), augmentation of mesenchymal vimentin level, and GSK3β Ser-9 hyper-phosphorylation, which led to enhanced invasiveness. This work demonstrates that cytoplasmic, resting AHR protein may act as an EMT suppressor via a non-genomic pathway. Conventional cytogenetics can categorize patients with acute myeloid leukemia ( AML) into favorable, intermediate, and unfavorable-risk groups; however, patients with intermediate-risk cytogenetics represent the major population with variable outcomes. Because molecular profiling can assist with AML prognosis and next-generation sequencing allows simultaneous sequencing of many target genes, we analyzed 260 genes in 112 patients with de novo AML who received standard treatment. Multivariate analysis showed that karyotypes and mutation status of TET2, PHF6, KIT, and NPM1mutation/ FLT3- internal tandem duplication ( ITD)negative were independent prognostic factors for the entire cohort. Among patients with intermediate-risk cytogenetics, patients with mutations in CEBPAdouble mutation, IDH2, and NPM1 in the absence of FLT3- ITD were associated with improved Overall survival ( OS), similar to those with favorable-risk cytogenetics; patients with mutations in TET2, RUNX1, ASXL1, and DNMT3A were associated with reduced OS, similar to those with unfavorable-risk cytogenetics. We concluded that integration of cytogenetic and molecular profiling improves prognostic stratification of patients into three groups with more distinct prognoses ( P < 0.001) and significantly reduces the number of patients classified as intermediate risk. In addition, our study demonstrates that next-generation sequencing ( NGS)-based multi-gene sequencing is clinically applicable in establishing an accurate risk stratification system for guiding therapeutic decisions. Background: Pancreatic adenocarcinoma is one of the most lethal cancers, yet it remains understudied and poorly understood. Hyperinsulinemia has been reported to be a risk factor of pancreatic cancer, and the rapid rise of hyperinsulinemia associated with obesity and type 2 diabetes foreshadows a rise in cancer incidence. However, the actions of insulin at the various stages of pancreatic cancer progression remain poorly defined. Methods: Here, we examined the effects of a range of insulin doses on signalling, proliferation and survival in three human cell models meant to represent three stages in pancreatic cancer progression: primary pancreatic duct cells, the HPDE immortalized pancreatic ductal cell line, and the PANC1 metastatic pancreatic cancer cell line. Cells were treated with a range of insulin doses, and their proliferation/viability were tracked via live cell imaging and XTT assays. Signal transduction was assessed through the AKT and ERK signalling pathways via immunoblotting. Inhibitors of AKT and ERK signalling were used to determine the relative contribution of these pathways to the survival of each cell model. Results: While all three cell types responded to insulin, as indicated by phosphorylation of AKT and ERK, we found that there were stark differences in insulin-dependent proliferation, cell viability and cell survival among the cell types. High concentrations of insulin increased PANC1 and HPDE cell number, but did not alter primary duct cell proliferation in vitro. Cell survival was enhanced by insulin in both primary duct cells and HPDE cells. Moreover, we found that primary cells were more dependent on AKT signalling, while HPDE cells and PANC1 cells were more dependent on RAF/ERK signalling. Conclusions: Our data suggest that excessive insulin signalling may contribute to proliferation and survival in human immortalized pancreatic ductal cells and metastatic pancreatic cancer cells, but not in normal adult human pancreatic ductal cells. Transplantation of pancreatic progenitors or insulin-secreting cells derived from human embryonic stem cells (hESCs) has been proposed as a therapy for diabetes. We describe a seven-stage protocol that efficiently converts hESCs into insulin-producing cells. Stage (S) 7 cells expressed key markers of mature pancreatic beta cells, including MAFA, and displayed glucose-stimulated insulin secretion similar to that of human islets during static incubations in vitro. Additional characterization using single-cell imaging and dynamic glucose stimulation assays revealed similarities but also notable differences between S7 insulin-secreting cells and primary human beta cells. Nevertheless, S7 cells rapidly reversed diabetes in mice within 40 days, roughly four times faster than pancreatic progenitors. Therefore, although S7 cells are not fully equivalent to mature beta cells, their capacity for glucose-responsive insulin secretion and rapid reversal of diabetes in vivo makes them a promising alternative to pancreatic progenitor cells or cadaveric islets for the treatment of diabetes. Polyhydroxyalkanoates (PHAs) are renewable and biodegradable polyesters which can be synthesized either by numerous of microorganisms in vivo or synthase in vitro. The synthesis of PHAs in vitro requires an efficient separation for high yield of purified enzyme. The recombinant Escherichia coli harboring phaC gene derived from Ralstonia eutropha H16 was cultivated in the chemically defined medium for overexpression of synthase in the present work. The purification and characteristics of PHA synthase from clarified feedstock by using aqueous two-phase systems (ATPS) was investigated. The optimized concentration of ATPS for partitioning PHA synthase contained polyethylene glycol 6000 (30%, w/w) and potassium phosphate (8%, w/w) with 3.25 volume ratio in the absence of NaCl at pH 8.7 and 4°C. The results showed that the partition coefficient of enzyme activity and protein content are 6.07 and 0.22, respectively. The specific activity, selectivity, purification fold and recovery of phaCRe achieved 1.76 U mg−1, 29.05, 16.23 and 95.32%, respectively. Several metal ions demonstrated a significant effect on activity of purified enzyme. The purified enzyme displayed maximum relative activity as operating condition at pH value of 7.5 and 37°C. As compared to conventional purification processes, ATPS can be a promising technique applied for rapid recovery of PHA synthase and preparation of large quantity of PHA synthase on synthesis of P(3HB) in vitro. Diabetes is a devastating disease that is ultimately caused by the malfunction or loss of insulin-producing pancreatic betacells. Drugs capable of inducing the development of new beta-cells or improving the function or survival of existing betacells could conceivably cure this disease. We report a novel high-throughput screening platform that exploits multiparameter high-content analysis to determine the effect of compounds on beta-cell survival, as well as the promoter activity of two key beta-cell genes, insulin and pdx1. Dispersed human pancreatic islets and MIN6 beta-cells were infected with a dual reporter lentivirus containing both eGFP driven by the insulin promoter and mRFP driven by the pdx1 promoter. Bscore statistical transformation was used to correct systemic row and column biases. Using this approach and 5 replicate screens, we identified 7 extracts that reproducibly changed insulin and/or pdx1 promoter activity from a library of 1319 marine invertebrate extracts. The ability of compounds purified from these extracts to significantly modulate insulin mRNA levels was confirmed with real-time PCR. Insulin secretion was analyzed by RIA. Follow-up studies focused on two lead compounds, one that stimulates insulin gene expression and one that inhibits insulin gene expression. Thus, we demonstrate that multi-parameter, high-content screening can identify novel regulators of beta-cell gene expression, such as bivittoside D. This work represents an important step towards the development of drugs to increase insulin expression in diabetes and during in vitro differentiation of beta-cell replacements.
