Zanthoxylum is a versatile economic tree species utilized for its spice, seasoning, oil, medicinal, and industrial raw material applications, and it has a lengthy history of cultivation and domestication in China. This has led to the development of numerous cultivars. However, the phenomenon of mixed cultivars and confusing names has significantly obstructed the effective utilization of Zanthoxylum resources and industrial development. Consequently, conducting genetic diversity studies and cultivar identification on Zanthoxylum are crucial. This research analyzed the genetic traits of 80 Zanthoxylum cultivars using simple sequence repeat (SSR) and inter-Primer Binding Site (iPBS) molecular markers, leading to the creation of a DNA fingerprint. This study identified 206 and 127 alleles with 32 SSR markers and 10 iPBS markers, respectively, yielding an average of 6.4 and 12.7 alleles (Na) per marker. The average polymorphism information content (PIC) for the SSR and iPBS markers was 0.710 and 0.281, respectively. The genetic similarity coefficients for the 80 Zanthoxylum accessions ranged from 0.0947 to 0.9868 and from 0.2206 to 1.0000, with mean values of 0.3864 and 0.5215, respectively, indicating substantial genetic diversity. Cluster analysis, corroborated by principal coordinate analysis (PCoA), categorized these accessions into three primary groups. Analysis of the genetic differentiation among the three Zanthoxylum (Z. bungeanum, Z. armatum, and Z. piperitum) populations using SSR markers revealed a mean genetic differentiation coefficient (Fst) of 0.335 and a gene flow (Nm) of 0.629, suggesting significant genetic divergence among the populations. Molecular variance analysis (AMOVA) indicated that 65% of the genetic variation occurred within individuals, while 35% occurred among populations. Bayesian model-based analysis of population genetic structure divided all materials into two groups. The combined PI and PIsibs value of the 32 SSR markers were 4.265 × 10− 27 and 1.282 × 10− 11, respectively, showing strong fingerprinting power. DNA fingerprints of the 80 cultivars were established using eight pairs of SSR primers, each assigned a unique numerical code. In summary, while both markers were effective at assessing the genetic diversity and relationships of Zanthoxylum species, SSR markers demonstrated superior polymorphism and cultivar discrimination compared to iPBS markers. To better understand the fruit flesh coloration mechanism of peach (Prunus persica), the composition and accumulation of carotenoids were compared, the expression profile of key genes involved in carotenoid biosynthetic and catabolic pathways was performed, and the differentially expressed genes were identified using "Piqiutao" (white fruit flesh) and its mutant yellow "Piqiutao" at different fruit development stages. The results showed that the total carotenoid content in yellow "Piqiutao" was remarkably higher than that of "Piqiutao," and the accumulation of β-cryptoxanthin, α-carotene, and β-carotene was significantly different, which was most likely caused by the differential expression of CCD4. Therefore, CCD4 may be an essential gene that causes the yellow fruit flesh of yellow "Piqiutao." However, the coding region sequence of CCD4 was entirely identical, and the intron was inserted by a retrotransposon in "Piqiutao" and its mutant, indicating that the expression difference was not caused by the sequence mutation and retrotransposon insertion. In this study, third-generation full-length (FL) transcriptome sequencing was performed of loquat using single-molecule real-time(SMRT) sequencing from the pooled cDNA of embryos of young loquat fruit under different low temperatures (three biological replicates for treatments of 1°C, -1°C, and -3°C, for 12 h or 24 h) and the control group(three biological replicates for treatments of room temperature), Illumina sequencing was used to correct FL transcriptome sequences. A total of 3 PacBio Iso-Seq libraries (1–2 kb, 2–3 kb and 3–6 kb) and 21 Illumina transcriptome libraries were constructed, a total of 13.41 Gb of clean reads were generated, which included 215,636 reads of insert (ROIs) and 121,654 FL, non-chimaric (FLNC) reads. Transcript clustering analysis of the FLNC reads revealed 76,586 consensus isoforms, and a total of 12,520 high-quality transcript sequences corrected with non-FL sequences were used for subsequent analysis. After the redundant reads were removed, 38,435 transcripts were obtained. A total of 27,905 coding DNA sequences (CDSs) were identified, and 407 long non-coding RNAs (lncRNAs) were ultimately predicted. Additionally, 24,832 simple sequence repeats (SSRs) were identified, and a total of 1,295 alternative splicing (AS) events were predicted. Furthermore, 37,993 transcripts were annotated in eight functional databases. This is the first study to perform SMRT sequencing of the FL transcriptome of loquat. The obtained transcriptomic data are conducive for further exploration of the mechanism of loquat freezing injury and thus serve as an important theoretical basis for generating new loquat material and for identifying new ways to improve loquat cold resistance. Graphite negative electrodes are unbeaten hitherto in lithium-ion batteries (LiBs) due to their unique chemical and physical properties. Thus, the increasing scarcity of graphite resources makes smart recycling or repurposing of discarded graphite particularly imperative. However, the current recycling techniques still need to be improved upon with urgency. Herein a facile and efficient hydrometallurgical process is reported to effectively regenerate aged (39.5 %, 75 % state-of-health, SOH) scrapped graphite (SG) from end-of-life lithium-ion batteries. Ultimately, the first cycle reversible capacity of SG1 (SOH = 39.5 %) improved from 266 mAh/g to 337 mAh/g while 330 mAh/g (98 %) remain after 100 cycles at 0.5 C. The reversible capacity for the first cycle of SG2 (SOH = 75 %) boosted from 335 mAh/g to 366 mAh/g with the capacity retention of 99.3 % after 100 cycles at 0.5 C, which is comparable with the benchmark commercial graphite. The regenerated graphites RG1 and RG2 exhibit excellent output characteristics even increasing the rate up to 4 C. This is the best rate level reported in the literature to date. Finally, the diffusion coefficient of Li ions during deintercalation and intercalation in the regenerated graphites have been measured by galvanostatic intermittent titration technique (GITT), determining values 2 orders-of-magnitude higher than that of the spent counterparts. Taking advantage of the synergistic effect of acid leaching and heat treatment, this strategy provides a simple and up-scalable method to recycle graphitic anodes.
