Background: Chemotherapy-induced nausea and vomiting (CINV) is a common and distressing side effect. We conducted this clinical trial to compare the effectiveness of true acupuncture vs. sham acupuncture in controlling chemotherapy-induced nausea and vomiting (CINV) among patients with advanced cancer. Methods: A total of 134 participants were randomly allocated into true acupuncture (TA) (n = 68) and sham acupuncture (SA) (n = 66) groups. Participants in both groups received acupuncture session twice on the first day of chemotherapy, and once consecutively on the following 4 days. The primary outcome was using the Common Terminology Criteria for Adverse Events (CTCAE) to assess CINV. The secondary outcome measures were the Eastern Cooperative Oncology Group score (ECOG), Simplified Nutritional Appetite Questionnaire (SNAQ), and Hospital Anxiety and Depression scale (HADS). Results: Compared to the SA group, the TA group didn't show significant improvement in complete response rates of chemotherapy-induced nausea and vomiting (all P > 0.05). However, the TA group could modestly reduce the severity of nausea (from day-3 to day-21, P < 0.05) or vomiting (from day-4 to day-21, P < 0.05), which is notably superior to the control group. Besides, TA promoted the nutritional status of patients with a significantly higher score comparing to the SA group on day 14 (21.82 vs.20.12, P = 0.003) and day 21 (22.39 vs. 20.43, P = 0.001). No apparent differences were found in anxiety and depression assessment between these groups. Participants in both groups were well tolerant of acupuncture therapy. There was no adverse event occurs in our study. Conclusion: Acupuncture as an adjunctive approach could alleviate the severity of chemotherapy-induced nausea and vomiting compared to the sham control, even though the effect of acupuncture in preventing CINV occurring is relatively modest. Fusarium head blight (FHB) is a serious wheat disease all over the world. In this study, the relationships between plant height (PH) and FHB were investigated across the whole wheat genome by QTL meta-analysis from fifty-six experiments. Coincident meta-QTL (MQTL) for PH and FHB were found on chromosomes 2D, 3A, 4B, 4D and 7A. Rht-B1, Rht-D1, Rht8, MQTLs P7 and P26 were consistent with FHB MQTLs. The meta-analysis results confirmed the negative associations of Rht-B1, Rht-D1, and Rht8 with FHB resistance. The associations of PH and FHB resistance on chromosomes 3A and 7A have not been reported and need further investigation. These regions should be given attention in breeding programs. MQTLs derived from several resistance sources were also observed. Some FHB MQTLs for different types of resistance overlapped. Sequence polymorphisms and phylogenetic relationships from different genomes of 25 diploid species in Triticeae (Poaceae) were evaluated by using the sequences of y-type high-molecular-weight glutenin promoter (y-HGP). The length of the amplified y-HGP sequences ranged from 845 to 915 base pairs (bp) in the 25 species of Triticeae. Multiple sequence alignment showed conserved and variable parts in the y-HGP sequences. Higher sequence conservation was detected in the regulatory elements of y-HGP. An 85-bp deletion was found in eight species of Triticum, Aegilops, and Hordeum. Several species-specific indels were identified in the y-HGP from Psathyrostachys, Hordeum, and Pseudoroegneria. Maximum parsimony (MP) and Bayesian analyses defined an Aegilops/Triticum group consisting of closely related species. A close relationship between Pseudoroegneria and the clade of Australopyrum, Dasypyrum, and Agropyron was also strongly supported in the topologies of MP and Bayesian trees. As y-HGP has sufficient amounts of genetic variation and is a single-copy region in diploid Triticeae, it is useful in phylogenetic analyses of this group. [ABSTRACT FROM AUTHOR]

Background: Various enzyme inhibitors act on key insect gut digestive hydrolases, including alpha-amylases and proteinases. Alpha-amylase inhibitors have been widely investigated for their possible use in strengthening a plant's defense against insects that are highly dependent on starch as an energy source. We attempted to unravel the diversity of monomeric alpha-amylase inhibitor genes of Israeli and Golan Heights' wild emmer wheat with different ecological factors (e.g., geography, water, and temperature). Population methods that analyze the nature and frequency of allele diversity within a species and the codon analysis method (comparing patterns of synonymous and non-synonymous changes in protein coding sequences) were used to detect natural selection. Results: Three hundred and forty-eight sequences encoding monomeric alpha-amylase inhibitors (WMAI) were obtained from 14 populations of wild emmer wheat. The frequency of SNPs in WMAI genes was 1 out of 16.3 bases, where 28 SNPs were detected in the coding sequence. The results of purifying and the positive selection hypothesis (p < 0.05) showed that the sequences of WMAI were contributed by both natural selection and coevolution, which ensured conservation of protein function and inhibition against diverse insect amylases. The majority of amino acid substitutions occurred at the C-terminal (positive selection domain), which ensured the stability of WMAI. SNPs in this gene could be classified into several categories associated with water, temperature, and geographic factors, respectively. Conclusions: Great diversity at the WMAI locus, both between and within populations, was detected in the populations of wild emmer wheat. It was revealed that WMAI were naturally selected for across populations by a ratio of dN/dS as expected. Ecological factors, singly or in combination, explained a significant proportion of the variations in the SNPs. A sharp genetic divergence over very short geographic distances compared to a small genetic divergence between large geographic distances also suggested that the SNPs were subjected to natural selection, and ecological factors had an important evolutionary role in polymorphisms at this locus. Background: High-molecular-weight glutenin subunits (HMW-GSs) have been considered as most important seed storage proteins for wheat flour quality. 1Ay subunits are of great interest because they are always silent in common wheat. The presence of expressed 1Ay subunits in diploid and tetraploid wheat genotypes makes it possible to investigate molecular information of active 1Ay genes. Results: We identified 1Ay subunits with different electrophoretic mobility from 141 accessions of diploid and tetraploid wheats, and obtained the complete ORFs and 5' flanking sequences of 1Ay genes including 6 active and 3 inactive ones. Furthermore, the 5' flanking sequences were characterized from 23 wild diploid species of Triticeae. All 6 active 1Ay possess a typical HMW-GS primary structure and some novel characteristics. The conserved cysteine residue within the repetitive domain of y-type subunits was replaced by phenylalanine residue in subunits of 1Ay (Tue1), 1Ay (Tu-e2), 1Ay (Ta-e2) and 1Ay (Td-e). Particularly, 1Ay (Ta-e3) has an unusual large molecular weight of 2202 bp and was one of the known largest y-type HMW-GSs. The translations of 1Ay (Tu-s), 1Ay (Ta-s) and 1Ay (Td-s) were disrupted by premature stop codons in their coding regions. The 5' flanking sequences of active and inactive 1Ay genes differ in a few base substitutions and insertions or deletions. The 85 bp deletions have been found in promoter regions of all 1Ay genes and the corresponding positions of 6 species from Aegilops and Hordeum. Conclusion: The possession of larger molecular weight and fewer conserved cysteine residues are unique structural features of 1Ay genes; it would be interested to express them in bread wheat and further to examine their impact to processing quality of wheat. The 1Ay genes from T. urartu are closer to the genes from T. turgidum dicoccon and T. aestivum, than those from T. monococcum aegilopoides. The 85 bp deletion and some variations in the 5'flanking region, have not interrupted expression of 1Ay genes, whereas the defects in the coding regions could be responsible to the silence of the 1Ay genes. Background: The unique properties of wheat flour primarily depend on gluten, which is the most important source of protein for human being. γ-Gliadins have been considered to be the most ancient of the wheat gluten family. The complex family structure of γ-gliadins complicates the determination of their function. Moreover, γ-gliadins contain several sets of celiac disease epitopes. However, no systematic research has been conducted yet. Results: A total of 170 γ-gliadin genes were isolated from common wheat and its closely related species, among which 138 sequences are putatively functional. The ORF lengths of these sequences range from 678 to 1089 bp, and the repetitive region is mainly responsible for the size heterogeneity of γ-gliadins. The repeat motif P(Q/L/S/T/I/V/R/A)F(S/Y/V/Q/I/C/L)P(R/L/S/T/H/C/ Y)Q1-2(P(S/L/T/A/F/H)QQ)1-2 is repeated from 7 to 22 times. Sequence polymorphism and linkage disequilibrium analyses show that γ-gliadins are highly diverse. Phylogenic analyses indicate that there is no obvious discrimination between Sitopsis and Ae. tauschii at the Gli-1 loci, compared with diploid wheat. According to the number and placement of cysteine residues, we defined nine cysteine patterns and 17 subgroups. Alternatively, we classified γ-gliadins into two types based on the length of repetitive domain. Amino acid composition analyses indicate that there is a wide range of essential amino acids in γ-gliadins, and those γ-gliadins from subgroup SG-10 and SG-12 and γ-gliadins with a short repetitive domain are more nutritional. A screening of toxic epitopes shows that γ-gliadins with a pattern of C9 and γ-gliadins with a short repetitive domain almost lack any epitopes. Conclusion: γ-Gliadin sequences in wheat and closely related Aegilops species are diverse. Each group/subgroup contributes differently to nutritional quality and epitope content. It is suggested that the genes with a short repetitive domain are more nutritional and valuable. The molecular diversity of the rDNA sequences (5S rDNA units) in 71 accessions from 26 taxa of Avena was evaluated. The analyses, based on 553 sequenced clones, indicated that there were 6 unit classes, named according to the haplomes (genomes) they putatively represent, namely the long A1, long B1, long M1, short C1, short D1, and short M1 unit classes. The long and short M1 unit classes were found in the tetraploid A. macrostachya, the only perennial species. The long M1 unit class was closely related to the short C1 unit class, while the short M1 unit class was closely related to the long A1 and long B1 unit classes. However, the short D1 unit class was more divergent from the other unit classes. There was only one unit class per haplome in Avena, whereas haplomes in the Triticeae often have two. Most of the sequences captured belonged to the long A1 unit class. Sequences identified as the long B1 unit class were found in the tetraploids A. abyssinica and A. vaviloviana and the diploids A. atlantica and A. longiglumis. The short C1 unit class was found in the diploid species carrying the C genome, i.e., A. clauda, A. eriantha, and A. ventricosa, and also in the diploid A. longiglumis, the tetraploids A. insularis and A. maroccana, and all the hexaploid species. The short D1 unit class was found in all the hexaploid species and two clones of A. clauda. It is noteworthy that in previous studies the B genome was found only in tetraploid species and the D genome only in hexaploid species. Unexpectedly, we found that various diploid Avena species contained the B1 and D1 units. The long B1 unit class was found in 3 accessions of the diploid A. atlantica (CN25849, CN25864, and CN25887) collected in Morocco and in 2 accessions of A. longiglumis (CIav9087 and CIav9089) collected in Algeria and Libya, respectively, whereas only 1 clone of A. clauda (CN21378) had the short D1 unit. Thus there might be a clue as to where to search for diploids carrying the B and D genomes. Avena longiglumis was found to be the most diverse species, possibly harboring the A, B, and C haplomes. The long M1 and short M1 are the unit classes typical of A. macrostachya. These results could explain the roles of A. clauda, A. longiglumis, and A. atlantica in the evolution of the genus Avena. Furthermore, one clone of the tetraploid A. murphyi was found to have sequences belonging to the short D1 unit class, which could indicate that A. murphyi might have been the progenitor of hexaploid oats and not, as postulated earlier, A. insularis. The evolution of Avena did not follow the molecular clock. The path inferred is that the C genome is more ancient than the A and B genomes and closer to the genome of A. macrostachya, the only existing perennial, which is presumed to be the most ancestral species in the genus. Background: α-Amylase inhibitors are attractive candidates for the control of seed weevils, as these insects are highly dependent on starch as an energy source. In this study, we aimed to reveal the structure and diversity of dimeric α-amylase inhibitor genes in wild emmer wheat from Israel and to elucidate the relationship between the emmer wheat genes and ecological factors using single nucleotide polymorphism (SNP) markers. Another objective of this study was to find out whether there were any correlations between SNPs in functional protein-coding genes and the environment. Results: The influence of ecological factors on the genetic structure of dimeric a-amylase inhibitor genes was evaluated by specific SNP markers. A total of 244 dimeric α-amylase inhibitor genes were obtained from 13 accessions in 10 populations. Seventy-five polymorphic positions and 74 haplotypes were defined by sequence analysis. Sixteen out of the 75 SNP markers were designed to detect SNP variations in wild emmer wheat accessions from different populations in Israel. The proportion of polymorphic loci P (5%), the expected heterozygosity He, and Shannon's information index in the 16 populations were 0.887, 0.404, and 0.589, respectively. The populations of wild emmer wheat showed great diversity in gene loci both between and within populations. Based on the SNP marker data, the genetic distance of pairwise comparisons of the 16 populations displayed a sharp genetic differentiation over long geographic distances. The values of P, He, and Shannon's information index were negatively correlated with three climatic moisture factors, whereas the same values were positively correlated by Spearman rank correlation coefficients' analysis with some of the other ecological factors. Conclusion: The populations of wild emmer wheat showed a wide range of diversity in dimeric α-amylase inhibitors, both between and within populations. We suggested that SNP markers are useful for the estimation of genetic diversity of functional genes in wild emmer wheat. These results show significant correlations between SNPs in the α-amylase inhibitor genes and ecological factors affecting diversity. Single nucleotide polymorphisms (SNPs) identified in EST sequences can be used to map expressed genes. Through SNPs are useful markers for genetic mapping. SNP mapping of genes in common wheat is challenging because the genetic complement of wheat consists of three closely refitted genomes (designated A, B. and D), and most genes are present in triplicate sets. Mapping multi-gene family members is further complicated by the fact that it is difficult to distinguish SNP differences between the various paralogs from those between the different genomes. We have developed a PCR-based method for assigning wheat EST sequences to their proper genetic loci by first identifying and mapping SNPs that distinguish the three genomes. To develop this method, we focused on EST sequences encoding the dimeric α-amylase inhibilors (WDAI). The WDAI coding regions of hexaploid wheal were aligned. The sequences were classified into three groups based on nucleotide variations. Twenty two SNPs were identified that distinguish the three groups. Group-specific primers based on these SNPs were designed to permit selective amplification of each group. The chromosomal location of each group was then determined using Group 3 ditelosomic lines of Chinese Spring. Groups 1 and 2 were assigned to chromosome locations 3DS and 3BS, respectively, whereas no sequence could be assigned to 3AS. A remarkable feature of this method is the ability to discriminate the location of homoeologous multigenes in the three genomes o1 wheat. On the basis of sequence analysis, 69 known low-molecular-weight glutenin subunit (LMW-GS) genes were experimentally classified into nine groups by the deduced amino acid sequence of the highly conserved N-terminal domain. To clarify the chromosomal locations of these groups, 11 specific primer sets were designed to carry out polymerase chain reactions (PCR) with the genomic DNA of group 1 ditelosomic lines of Chinese Spring, among which nine primer sets proved to be LMW-GS group-specific. Each group of LMW-GS genes was specifically assigned on a single chromosome arm and hence to a specific locus. Therefore, these results provided the possibility to predict the chromosome location of a new LMW-GS gene based on its deduced N-terminal sequence. The validity of the classification was confirmed by the amplifications in 27 diploid wheat and Aegilops accessions. The length polymorphisms of LMW-GS genes of groups 1 and 2, and groups 3 and 4.1 were detected in diploid A-genome and S-genome accessions, respectively. The diploid wheat and Aegilops species could be used as valuable resources of novel allele variations of LMW-GS gene in the improvement of wheat quality. To answer the question whether Hordeum spontaneum K. Koch. and Hordeum vulgare L. are conspecific taxonomic entities or not”, we carried out an investigation with amplified restriction fragment length polymorphism (AFLP™) using H. vulgare cultivars, H. spontaneum from the Fertile Crescent, and material from China, primarily Xizang (Tibet), containing landraces and accessions identified morphologically as Hordeum agriocrithon Åberg. and H. spontaneum. Population genetic analyses and discriminant analyses were based on three groupings, H. vulgare, H. spontaneum, and H. agriocrithon, determined from morphology. To independently find groups, cluster analyses were carried out followed by population-genetic analyses and discriminant analyses of the clusters. The results of population-genetic analyses of morphologically based groups were not very different from those of comparable analyses of groups inferred from clustering. Based on 795 AFLP bands scored on 506 individuals, genetic diversity was 30% higher within H. spontaneum than within H. agriocrithon or H. vulgare. The variation among the three entities was 27.3% (FCT = 0.27) and among populations within the three was 40.4% (FSC = 0.56) leaving 32.3% (FST = 0.68) within populations/accessions. Three entities were clearly defined by clustering and supported by discriminant analyses, and then nomenclaturally formulated, two as species and one as an interspecific hybrid. The first, H. spontaneum, comprises populations in the Near East, the second, H. vulgare, comprises the cultivars, and the third, H. ×agriocrithon, comprises all barley landraces and hybrids from China as well as hybrids from the Near East. We discuss evolutionary perspectives of speciation and conclude that barley cultivars form a species distinct from the wild progenitor, H. spontaneum. This analysis may highlight the dilemma regarding other cultivated plants and domesticated animals, a still unresolved evolutionary–taxonomic problem. [ABSTRACT FROM AUTHOR]

Salicylic acid (SA), a vital endogenous hormone, plays a crucial role in plant growth and the response to abiotic and biotic stress. Isochorismate synthase (ICS) and phenylalanine ammonia lyase (PAL) are critical rate-limiting enzymes for SA synthesis. Fusarium head blight (FHB) seriously threatens the safety of wheat production, but increasing the content of SA can enhance FHB resistance. However, the pathway of SA synthesis and regulation in wheat remains unknown. In this study, three wheat ICS (TaICSA, TaICSB, and TaICSD) were identified, and their functions were validated in vitro for isomerizing chorismate to isochorismate. The mutation of one or two homoeoalleles of TaICSA, TaICSB, and TaICSD in the wheat variety 'Cadenza' reduced SA levels under ultraviolet treatment and Fusarium graminearum infection, further enhancing sensitivity to FHB. Overexpression of TaICSA can significantly enhance SA levels and resistance to FHB. To further study SA synthesis pathways in wheat and avoid interference with pathogenicity related genes, the leaves of wild-type Cadenza and different TaICS mutant lines were subjected to ultraviolet treatment for transcriptomic analysis. The results showed that 37 PALs might be involved in endogenous SA synthesis, and 82 WRKY and MYB family transcription factors may regulate the expression of ICS and PAL. These results were further confirmed by RT-PCR. In conclusion, this study expands our knowledge of SA biosynthesis and identifies TaICSA, as well as several additional candidate genes that encode transcription factors for regulating endogenous SA levels, as part of an efficient strategy for enhancing FHB resistance in wheat. Background: High molecular weight glutenin subunits (HMW-GSs), encoded by the genes at Glu-1 loci in wheat and its related species, are significant in the determination of grain processing quality. However, the diversity and variations of HMW-GSs are relatively low in bread wheat. More interests are now focused on wheat wild relatives in Triticeae. The genus Aegilops represents an important germplasm for novel HWM-GSs and other useful genes for wheat genetic improvement. Results: Six novel Glu-1 alleles and HMW-GSs were identified and characterized from three species of Aegilops section Sitopsis (S genome). Both open reading frames (ORFs) and promoter regions of these Glu-1 alleles were sequenced and characterized. The ORFs of Sitopsis Glu-1 genes are approximately 2.9 kb and 2.3 kb for x-type and y-type subunits, respectively. Although the primary structures of Sitopsis HMW-GSs are similar to those of previously reported ones, all six x-type or y-type subunits have the large fragment insertions. Our comparative analyses of the deduced amino acid sequences verified that Aegilops section Sitopsis species encode novel HMW-GSs with their molecular weights larger than almost all other known HMW-GSs. The Glu-1 promoter sequences share the high homology among S genome. Our phylogenetic analyses by both network and NJ tree indicated that there is a close phylogenetic evolutionary relationship of x-type and y-type subunit between S and D genome. Conclusions: The large molecular weight of HMW-GSs from S genome is a unique feature identified in this study. Such large subunits are resulted from the duplications of repetitive domains in Sitopsis HMW-GSs. The unequal crossover events are the most likely mechanism of variations in glutenin subunits. The S genome-encoded subunits, 1Dx2.2 and 1Dx2.2* have independent origins, although they share similar evolutionary mechanism. Fifty-six sequences encoding the pina protein were characterized from three species or subspecies of einkorn wheat. These sequences contained 1,595 nucleotides, including 1,270 conserved sites, 21 single nucleotide polymorphisms (SNPs), and 16 indels. The average frequency of SNPs and indels was one out of 76.1 and 99.9 bases, respectively. Five SNPs and no indels were found in the translated sequences. Fourteen haplotypes were defined, and the accessions in each haplotype ranged from 1 to 18. There were nine haplotypes in Triticum monococcum ssp. aegilopoides, eight in T. monococcum ssp. monococcum, and two in T. urartu. Phylogenetic analysis showed that pina genes from different species or subspecies could be clearly differentiated based on the open reading frame. Genes from T. urartu grouped together, whereas genes from T. monococcum ssp. aegilopoides and T. monococcum ssp. monococcum were shared by three and two clusters, respectively. Both the haplotype and phylogenetic analyses indicated that T. monococcum ssp. aegilopoides was more diverse. A number of murine models are used to mimic the pathology of breast cancer. Tissue inoculation and cell inoculation using orthotopic implantation (OS) and subcutaneous implantation (SQ) are commonly used to generate murine models to investigate cancer. However, limited information is available in regard to the variations of these methods. The present study compared growth, metastasis, survival and histopathology of tumors produced using OS and SQ to characterize features of the tumors produced by the two distinct methods. Additionally, the present study aimed at providing increased options for investigators when designing experiments. 4T1-luc2 cell suspension or 4T1-luc2 tissue suspension was inoculated using either OS or SQ into BALB/c mice. Tumor growth and metastasis were detected using an in vivo imaging system and calipers. Excised tumors and lung were assessed by tissue staining with hematoxylin and eosin, and the vessel marker cluster of differentiation 31. The results of the present study revealed that the cell suspension generated breast tumors of increased size, which was visualized and determined, following inoculation, using calipers at an earlier time point compared with tumors produced by tissue suspension. The increasing bioluminescent trend of OS tumors was more marked compared with that of SQ tumors. The volume of OS tumor was increased with decreased variation, compared with that of SQ tumors. In addition, the OS tumor exhibited increased microvessel density. Bioluminescent signals and histological results in regard to metastasis were consistent: OS implantation produced increased lung metastasis compared with that of SQ implantation, although they exhibited similar survival times. The results of the present study indicated that the inocula from distinct sources (tissue or cell) affected tumor growth. Furthermore, breast tumor progression and histopathological characteristics were distinct between OS and SQ, whereas OS exhibited increased malignant behavior. Understanding the characteristics of murine breast cancer models established by diverse methods may aid investigators to select appropriate animal models, according to the requirements of the study. Spike density and processing quality are important traits in modern wheat production and are controlled by multiple gene loci. The associated genes have been intensively studied and new discoveries have been constantly reported during the past few decades. However, no gene playing a significant role in the development of these two traits has been identified. In the current study, a common wheat mutant with extremely compact spikes and good processing quality was isolated and characterized. A new allele (Qc1) of the Q gene (an important domestication gene) responsible for the mutant phenotype was cloned, and the molecular mechanism for the mutant phenotype was studied. Results revealed that Qc1 originated from a point mutation that interferes with the miRNA172-directed cleavage of Q transcripts, leading to its overexpression. It also reduces the longitudinal cell size of rachises, resulting in an increased spike density. Furthermore, Qc1 increases the number of vascular bundles, which suggests a higher efficiency in the transportation of assimilates in the spikes of the mutant than that of wild type. This accounts for the improved processing quality. The effects of Qc1 on spike density and wheat processing quality were confirmed by analyzing nine common wheat mutants possessing four different Qc alleles. ADP-glucose pyrophosphorylase (AGP) consists of two large (AGP-L) and two small (AGP-S) subunits; it regulates the limiting step in the biosynthesis of starch. Here, we isolated the full-length cDNA sequence of the AGP-S gene (designed as Agp1) from wheat and whole genomic sequences from relatives of wheat. The coding region of Agp1 ranged from 5948 bp to 7666 bp and contained 9 exons and 8 introns. The full-length cDNA sequence of Agp1 encoded a polypeptide containing 473 amino acids without transit peptides. The sequence alignment and phylogenetic tree analyses indicated that the Agp1 cDNA sequence was highly conserved (99.7%) across the diploid Triticeae species examined in this study. Variations in introns caused the Agp1 genomic sequences from wheat relatives to be less similar (79.5%), and Agp1 sequences from the B genome donor species were divergent. In developing grains, rapid increase in Agp1 expression at prophase was closely followed by the accumulation of starch, and a significant (P < 0.001) correlation was observed between them. The heterogeneous mixtures of small and large subunits from different wheat species restarted glycogen synthesis in the Escherichia coli AC70R1-504 cells, which are deficient in endogenous AGP activity (glg C-/-). Background: MEK inhibitors are a group of drugs that have shown reliable effects in the treatment of metastatic melanoma and non-small-cell lung cancer. Peripheral edema is an adverse event associated with MEK inhibitors; however, there has been no systematic attempt to evaluate peripheral edema data observed with these agents. This meta-analysis aimed to determine the risk of peripheral edema in cancer patients treated with MEK inhibitors.Materials and Methods: The authors searched PubMed, the Cochrane Library, EMBASE, and Clinical Trials.gov without language restriction. The final search was conducted on January 9, 2017. Risk ratios (RR) with 95% confidence intervals (CI) were calculated for dichotomous data. Heterogeneity was calculated and reported via Tau2, Chi2, and I2 analyses.Results: A total of 13 eligible studies were obtained. Patients treated with MEK inhibitors (Trametinib and Selumetinib) had an increased risk overall of peripheral edema (RR = 3.05, 95% CI = 1.98-4.70; p < .00001), but the MEK inhibitors (Trametinib and Selumetinib) did not increase the risk of high grade edema (RR = 1.88, 95% CI = 0.66-5.35; p = .24). Sub-group analysis, based on cancer type (melanoma vs non-melanoma), found that the peripheral edema risk in melanoma patients is higher than that in non-melanoma patients (p = .03). However, no significant difference was observed in terms of high-grade edema and other sub-groups (trametinib vs selumetinib; monotherapy vs combination). Due to the absence of cobimetinib data, the result about cobimetinib was not involved.Conclusion: This meta-analysis reveals that the use of MEK inhibitors is associated with an increased risk of peripheral edema in cancer patients. Background: Chemotherapy-induced nausea and vomiting (CINV) is one of the most common and distressing side effects in patients with cancer. The introduction and development of antiemetic drugs have significantly improved the ability of clinicians to control CINV, but it is not easy to translate to practical application, owing to financial issues, provider-related barriers, and patient factors. Nondrug therapies are needed to alleviate the symptoms of CINV. Acupuncture is an appropriate adjunctive treatment for CINV, but additional evidence is needed.Methods/design: This study is a multicenter, randomized, sham-controlled prospective clinical trial. A total of 136 participants will be randomly allocated into the intervention group (verum acupuncture) or the control group (sham acupuncture) in a 1:1 ratio. All treatment will be given for 5 days. Participants in both groups will receive acupuncture sessions twice on the first day of chemotherapy and once consecutively on the following 4 days. Each session takes approximately 30 minutes. The primary outcome measure will be the Common Terminology Criteria for Adverse Events to assess CINV. The secondary outcome measures will be the Eastern Cooperative Oncology Group score, Simplified Nutritional Appetite Questionnaire, and Hospital Anxiety and Depression scale. We demonstrate the utility of optical second harmonic generation (SHG) polarimetry to perform structural characterization of self-assembled zinc-blende/wurtzite III–V nanowire heterostructures. By analyzing four anisotropic SHG polarimetric patterns, we distinguish between wurtzite (WZ), zinc-blende (ZB) and ZB/WZ mixing III–V semiconducting crystal structures in nanowire systems. By neglecting the surface contributions and treating the bulk crystal within the quasi-static approximation, we can well explain the optical SHG polarimetry from the NWs with diameter from 200–600 nm. We show that the optical in-coupling and out-coupling coefficients arising from depolarization field can determine the polarization of the SHG. We also demonstrate micro-photoluminescence of GaAs quantum dots in related ZB and ZB/WZ mixing sections of core–shell NW structure, in agreement with the SHG polarimetry results. The ability to perform in situ SHG-based crystallographic study of semiconducting single and multi-crystalline nanowire heterostructures will be useful in displaying structure-property relationships of nanodevices. Tibetan semiwild wheat (Triticum aestivum ssp. tibetanum Shao) is a primitive hexaploid wheat resource found in Tibet. It is characterized by tolerance to nutrition deficiency and strong seed dormancy and has potential to be useful in wheat breeding programs. To tap the advantages of Tibetan semiwild wheat in wheat breeding, we investigated nine agronomic traits including heading date (HD), anthesis date (AD), plant height (PHT), tiller number (TN), spike length (SL), spikelet number per spike (SNS), spikelet density (DS), grain weight per spike (GWS), and 1000-grain weight (TGW) in 186 recombinant inbred lines from a cross between Tibetan semiwild wheat 'Q1028' and common wheat 'Zhengmai 9023' (ZM 9023) across three growing seasons. Forty-five qualitative trait loci (QTLs) on 12 chromosomes were detected. The phenotypic variation explained by each of these QTL ranged from 4.7 to 29.7%. Positive alleles for 28 of these QTLs were derived from Q1028. Of these QTLs, 25 (56%) were detected in at least two growing seasons. Fifteen stable QTLs that were significant across all three growing seasons were identified. Novel QTLs derived from Q1028 were identified, such as QSd.sau-7A for spikelet density, QTgw.sau-2B for TGW and QSns.sau-3D for SNS. Eleven QTL clusters were detected, including one on chromosome 5A flanked by the markers wPt-9094 and wPt-9513. This cluster consists of QTLs controlling HD, AN, PHT, SL, and spikelet density and explained 6.6 to 12.6% of the phenotypic variation in these traits. Yellow or stripe rust, caused by Puccinia striiformis f. sp. tritici (Pst), is a devastating foliar disease that affects common wheat (Triticum aestivum L.) around the world. In China, common wheat landraces are potential sources of disease and abiotic stress resistance genes for wheat improvement. Yilongtuomai (YL), a wheat landrace from Yilong County, Sichuan Province, shows high levels of resistance against most Chinese Pst races. In this study, the resistance of YL to stripe rust disease was examined in detail. Parent strains, YL and Taichung 29, a variety susceptible to Pst race CYR32, and their F1', F2', and F2:3 offspring, were inoculated with CYR32 during the seedling stage in the field or adult-plant stage in the greenhouse. Results indicated that resistance to CYR32 in YL is conferred by a single dominant gene, designated YrYL. The segregating F2 population (352 plants), was analyzed in terms of its resistance locus using simple sequence repeats (SSRs), resistance gene analog polymorphisms (RGAPs), and sequence- related amplified polymorphism (SRAP). A linkage group of 6 SSRs, 2 RGAPs, and 1 SRAP was constructed for the YrYL gene. Using the identified SSRs associated with physical mapping of RGAP using Chinese Spring nullisomic-tetrasomic stocks, the YrYL gene was localized to the short arm of chromosome 7D. The gene was lanked by 1 SSR marker, Xbarc92, and 1 RGAP marker, CLRRfor/Ptokin4, at genetic distances of 5.35 and 9.86 cM, respectively. The YrYL gene was compared to other stripe rust resistance genes reported on chromosome 7D by evaluating its reaction patterns to CYR32 and its pedigree relationship. The ditelocentric addition line CS-7EL of the spring wheat (Triticum aestivum L.) cultivar Chinese Spring (CS) contains the long arm of the chromosome 7E from Thinopyrum elongatum (CS-7EL), which confers high resistance to Fusarium head blight. It is of great interest to breeders to integrate the resistance locus (loci) from Th. elongatum into commercial wheat varieties. The objectives of this study were to identify candidate genes expressed from the 7EL chromosome of CS-7EL, to develop 7EL-specific molecular markers, and to validate their usefulness for characterizing recombination between one of the group 7 chromosomes of wheat and Th. elongatum. High-throughput sequencing of Fusarium graminearum-infected and control CS and CS-7EL cDNA libraries was performed using RNA-Seq. A stepwise bioinformatics strategy was applied to assemble the sequences obtained from RNA-Seq and to create a conservative list of candidate genes expressed from the foreign chromosome 7EL. Polymerase chain reaction primer pairs were designed and tested for 135 candidate genes. A total of 48 expressed molecular markers specific for the chromosome 7EL were successfully developed. Screening of progenies from two BC1F2 families from the cross CS-7E(7D) × 2*CSph1b showed that these markers are useful for characterizing recombination events between the chromosomes 7D from wheat and 7E from Th. elongatum. Starch-branching enzyme I (SBE I) belongs to a member of SBEs isoforms, which plays an important role in amylopectin biosynthesis. In this study, we measured starch content, determined SBE activity within endosperms, analyzed expression patterns of SBE I by real-time quantitative PCR(RT-qPCR), and characterized sequences of SBE I from two accessions (Y59 and Y63) of Triticum monococcum with different starch content. Our results indicated that contents of total starch and amylopectin during grain development were significantly different between the two investigated accessions. Analysis of enzyme activity revealed that SBE activity in endospermof Y59 was obviously higher than that in Y63 at early developmental stage. Accumulation rates of both amylopectin and starch were significantly correlated to the SBE activities in the two accessions. Expression analysis showed thatSBEI geneswere highly expressed atmiddle stage of development. Further, the expression of SBE I in Y59 was higher than that in Y63 from 5 days after pollination (DAP) to 23 DAP. Coding sequences of SBE I in both Y59 and Y63 included a 2490-bp opening reading frame (ORF) encoding 829 amino acids. The genomic DNAs of SBE I were about 5.5 kb in length, contained 14 exons and 13 introns, and shared a similar structure with wheat. Our results may provide some bases to research amylopectin accumulation of T. monococcum, further provided relevant theoretical foundation for exploring the amylopectin synthesis in wheat. Background: Wheat (Triticum aestivum) is one of the most important cereal crops, providing food for humans and feed for other animals. However, its productivity is challenged by various biotic and abiotic stresses such as fungal diseases, insects, drought, salinity, and cold. Transcription factors (TFs) regulate gene expression in different tissues and at various developmental stages in plants and animals, and they can be identified and classified into families according to their structural and specialized DNA-binding domains (DBDs). Transcription factors are important regulatory components of the genome, and are the main targets for engineering stress tolerance. Results: In total, 2407 putative TFs were identified from wheat expressed sequence tags, and then classified into 63 families by using Hmm searches against hidden Markov model (HMM) profiles. In this study, 2407 TFs represented approximately 2.22% of all genes in the wheat genome, a smaller proportion than those reported for other cereals in PlantTFDB V3.0 (3.33%-5.86%) and PlnTFDB (4.30%-6.46%). We assembled information from the various databases for individual TFs, including annotations and details of their developmental stage- and tissue-specific expression patterns. Based on this information, we identified 1257 developmental stage-specific TFs and 1104 tissue-specific TFs, accounting for 52.22% and 45.87% of the 2407 wheat TFs, respectively. We identified 338, 269, 262, 175, 49, and 18 tissue-specific TFs in the flower, seed, root, leaf, stem, and crown, respectively. There were 100, 6, 342, 141, 390, and 278 TFs specifically expressed at the dormant seed, germinating seed, reproductive, ripening, seedling, and vegetative stages, respectively. We constructed a comprehensive database of wheat TFs, designated as WheatTFDB (http://xms.sicau.edu.cn/wheatTFDB/). Conclusions: Approximately 2.22% (2407 genes) of all genes in the wheat genome were identified as TFs, and were clustered into 63 TF families. We identified 1257 developmental stage-specific TFs and 1104 tissue-specific TFs, based on information about their developmental- and tissue-specific expression patterns obtained from publicly available gene expression databases. The 2407 wheat TFs and their annotations are summarized in our database, WheatTFDB. This study investigated the influence of lipids on amylose, amylopectin and total starch content in barley. Comparisons of various methods of single-wavelength colorimetric procedure (SWC) for amylose, dual-wavelength colorimetric method (DWC) for amylose, amylopectin and total starch, and polarimetric method by calcium chloride dissolution (PCC) for total starch content were conducted as well in barley. The results showed that defatted samples measured by SWC had higher amylose than non-defatted ones (P<0.05). Several samples exhibited significant difference for amylose content when defatted and non-defatted samples were analyzed by DWC (P<0.05). However, for amylopectin, the defatted samples determined by DWC were significantly lower than that of non-defatted ones (P<0.05). The total starch of defatted samples measured by DWC was much lower than those with fat (P<0.05). According to various methods, the amylose contents of defatted samples measured by SWC were higher than those measured by DWC (P<0.05). The non-defatted samples measured by PCC had lower starch than those measured by DWC, but had higher starch than the defatted samples measured by DWC (P<0.05). Collectively, defatting should not be omitted in determining starch content, and DWC is a suitable method for the measuring of barley starch. [ABSTRACT FROM AUTHOR]

The γ-prolamins are important components of seed storage proteins in wheat and other Triticeae species. Here, the γ-prolamin genes from the diploid Triticeae species were systemically characterized. Most of the γ-prolamins (except 75 K γ-secalins) characterized were defined as γ-gliadin-like γ-prolamins, since they shared same characteristic model structure with γ- gliadins. Over one-third of these putatively functional γ-prolamin peptides contained different number of cysteine residues as compared to the eight residues present in γ-gliadins. Sequence polymorphism and linkage disequilibrium analyses showed the conservation of γ-prolamin genes in Triticeae species under evolutionary selection. Phylogenetic analyses indicated that these γ-prolamin genes can not be clearly separated according to their genomic origins, reflecting the conservation of γ-gliadinlike γ-prolamin genes after the divergence of Triticeae species. A screening of coeliac disease (CD) toxic epitopes shows that the γ-prolamins from some other genomes contain much fewer epitopes than those from the A, S (B) and D genomes of wheat. According to the two distal and conserved regions of known α-gliadin genes, gene-specific primers for α-gliadin were designed, and the coding regions of four gliadin genes (i.e. GliTd-1, GliTd-2, GliTd-3 and GliTd-4) with the length of about 800 bp were isolated from the genomic DNA of wild emmer wheat (Triticum dicoccoides). No introns were observed. Sequence comparison indicated that these genes should be classified as α-gliadins. GliTd-3 (GenBank accession No.DQ140351) and GliTd-4 (DQ140352) were potentially functional, whereas GliTd-1 (DQ140349) and GliTd-2 (DQ140350) were both pseudogenes by the definition of in-frame stop codons and frameshifts. Six conserved cysteine residues were observed. Sequence analysis suggested that the motif units of repetitive domain for the four newly detected genes were different from the known genes, and the QQQP sequence before the position 60 was more toxic to coeliac patients. Codons for proline were strongly biased. Codons (CAG and CAA) for glutamine were clustered into the specific regions, and the high percentage of pseudogenes resulted from the mutation of CAG → TAG. Background: Aromatase inhibitors (AIs) are widely used as an adjuvant endocrine treatment in postmenopausal women with early-stage breast cancer. One of the main adverse effects of AIs is musculoskeletal symptoms, which leads to a lower quality of life and poor adherence to AI treatment. To date, no effective management of aromatase inhibitor-associated musculoskeletal symptoms (AIMSS) has been developed.Methods/design: To determine whether the traditional Chinese medicine Yi Shen Jian Gu granules could effectively manage AIMSS we will conduct a multicenter, randomized, double-blind, placebo-controlled clinical trial. Patients experiencing musculoskeletal symptoms after taking AIs will be enrolled and treated with traditional Chinese medicine or placebo for 12 weeks.
