The rhizome of Zingiber roseum has been used as traditional medicines for the treatment of gastric ulcer, skin diseases, and liver infections. Zerumbone is an essential sesquiterpenoid previously isolated from different species of Zingiberaceae and possesses numerous pharmacological properties. Based on previous reports, the aim of the current study was to investigate the antimicrobial activity of extract, fractions, and isolation of zerumbone from Zingiber roseum. The antimicrobial activity was determined using disc diffusion method. The serial tube dilution technique was used to determine Minimum Inhibitory Concentration (MIC) of the samples. Vacuum Liquid Chromatography (VLC) and open column chromatography were used for separation. The methanol extract exhibited maximum antimicrobial activity against Escherichia coli, Saccharomyces cerevisiae, and Aspergillus niger in both methods. VLC of the extract yielded five fractions (Fr. 1 to Fr. 5). Fr. 1 exhibited maximum antimicrobial activity compared to the other fractions. Column chromatography of Fr.1 generated 47 fractions. Fraction 1 to 9 were combined and crystallized in n-hexane to isolate zerumbone as the most abundant source (yield 6.5 % from dried MeOH extract). Zerumbone showed good activity against all tested bacteria. The results of our study might validate the ethnobotanical use of Zingiber roseum rhizomes in the treatment of infectious diseases. Moreover, Zingiber roseum could be one of the most abundant sources for the isolation of natural zerumbone to study different pharmacological studies in the field of new drug discovery. Plant extracts are considered as a promising alternative to chemical pest control agents. The chemistry of plant extracts varies with several environmental as well as geographical factors. Zingiber roseum (Roxb.) Roscoe also known as "Rosy Ginger" and " Jangli Adrak" is one of the most important aromatic and medicinal plant of family Zingiberaceae. The present study aimed to investigate the impact of altitude on the chemical composition, nematicidal and herbicidal activity of Z. roseum rhizome and seed extracts. The plants were collected from three different altitudes of Kumaun, Uttarakhand representing different environmental conditions. The extracts were prepared in acetone using cold percolation method. The compositional variation in chemical constituents of the extracts was determined by gas chromatography-mass spectrometry (GC–MS) analysis. The yield and chemical composition of extracts showed a significant altitudinal impact. Nematicidal activity of extracts was performed against Meloidogyne incognita. Results showed that the extracts exhibited significant nematode mortality irrespective of the altitudinal gradients. A significant correlation of altitudinal gradient was observed for the nematode egg hatchability reduction for the rhizome extracts. It was observed that plants growing at higher altitudes had better egg hatchability reduction (86.42 ± 2.30% at 1000 μg/mL after 96 h), compared with those growing at lower elevations. The herbicidal activity of rhizome and seed extracts was performed against radish and chicory seeds and the extracts showed different level of intensity of germination percentage, root and shoot length inhibition. In silico molecular docking was conducted to screen the nematicidal activity of major and potent compounds with the crystalline structure of Glutathione S-transferases (GSTs) of Caenorhabditis elegans and Wuchereria bancrofti , and Glutathione S-transferase-1 (GST-1) of M. incognita. Based on the current study, extract of Z. roseum may be regarded as potential nematicidal and herbicidal agents and may have potential application as botanicals. The present study focussed on phytochemical analysis of Zingiber roseum through GC & GC-MS, antifungal and antioxidant activities of methanolic extracts of seed, rhizome and perianth of the plant. GC-MS analysis was done by using GC MS-QP 2010 Plus equipment revealed diverse chemical make-up of the constituents. Among identified constituents ascorbic acid 2, 6-dihexadecanoate, 2-(4-hydroxy-3-methoxyphenyl)-3,7-dimethoxy-4h-chromen-4-one, hexadecanoic acid metyl ester, 9,12-octadecadienoic acid (Z,Z) and β-citronellal were found as major compounds in methanolic extracts from seeds, rhizome and petals respectively. The antifungal activity was done by disc diffusion methods for which extracts were tested against Fusarium accuminata. Among them the extracts were found to be remarkable fungicide according to its inhibition action against Fusarium accuminata. The in vitro antioxidant activity of the methanol extracts were evaluated by DPPH radical scavenging, FRAP assay and metal chelating activity. The antioxidant assay revealed good to moderate antioxidant potential with different IC50 values with respect to Fe3+ reducing power (200.34-438.79 μg/ml), Fe2+ metal chelating ability (310.80-469.69 μg/ml) and DPPH anion scavenging activities (215.99-884.40 μg/ml) in comparison to known standards. Abstract: A combination of GC and GC–MS revealed the presence of 60 compounds out of which 47 compounds (96.2% of the oil) were identified in the essential oil of seeds of Zingiber roseum Rosc., the major compounds being α-pinene, β-pinene, limonene, p-cymene, α-terpineol and verticiole. The unique feature is the dominant presence of mono- and sesquiterpene hydrocarbons which make about 82% of the oil. The oil showed myorelaxant activity on isolated rat duodenal smooth muscle. The Zingiber roseum seed essential oil (ZRSEO) relaxed both cabachol- and KCl-induced contractile responses with IC50 of non-significant difference. The results confirm a common mechanism in both the types of action of ZRSEO and suggests a probable inhibitory effect of ZRSEO on influx of Ca2+ through cell membrane of rat duodenal smooth muscle. Zingiber roseum is native to Bangladesh and widely used in folk medicine. This present study was designed to assess the ameliorative potential of Zingiber roseum rhizome extract in carbon tetrachloride (CCl 4) induced hepatotoxicity in mice model. Seven phenolic compounds were identified and quantified by HPLC analysis in the plant extract, including quercetin, myricetin, catechin hydrate, trans-ferulic acid, trans-cinnamic acid, (-) epicatechin, and rosmarinic acid. Hepatotoxicity was induced by administrating a single intraperitoneal injection of CCl 4 (10 mL/kg) on 7th day of treatment. The results revealed that plant extract at all doses (100, 200 and 400 mg/kg) significantly reduced (p < 0.05) the elevated serum aspartate aminotransferase (AST), alanine aminotransferase (ALT) and alkaline phosphatase (ALP) concentrations, and these effects were comparable to that of standard drug silymarin. Histopathological examination also revealed the evidence of recovery from CCL 4 induced cellular damage when pretreated with Z. roseum rhizome extract. The in-vivo hepatoprotective effects were further investigated by the in-silico study of the aforementioned compounds with liver-protective enzymes such as superoxide dismutase (SOD), peroxiredoxin, and catalase. The strong binding affinities (ranging from −7.3359 to −9.111 KCal/mol) between the phenolic compounds (except trans-cinnamic acid) and oxidative stress enzymes inhibit ROS production during metabolism. The compounds were also found non-toxic in computational prediction, and a series of biological activities like antioxidant, anticarcinogen, cardio-protectant, hepato-protectant have been detected.
