Summary: Background: Quantitative gait analysis is rapidly gaining ground in equine practice, and pros and cons are regularly discussed within the scientific literature. However, no data exist on the appreciation of the technique by equine clinicians, their motivation to use it or not, and their perception of its value in daily practice. Objectives: To make a first inventory of opinions, expectances and experiences of equine veterinarians concerning the use of quantitative gait analysis in their daily general practice. Study design: Survey. Methods: A questionnaire was sent out to a group of equine orthopaedic clinicians working in an equine clinic or practice. Respondents were classified as users (having clinical experience with quantitative gait analysis) or not (nonusers). Data were analysed using descriptive statistics. Results: Within the sample population, users were more positive about the usefulness of quantitative gait analysis than nonusers. Veterinarians who purchased a system were motivated by better objectivity, transparency, documentation and client service. Main reasons not to purchase a system were costs and complexity of data interpretation. A minority of both users and nonusers deemed quantitative gait analysis also suitable for equine professionals other than veterinarians. Main limitations: Users (n = 40) outnumbered nonusers (n = 32), sample size was limited (n = 72) and insufficient to allow for generalisation of results. Conclusions: Users of quantitative gait analysis were more positive about the technology than nonusers. More data are needed to allow for generalisation of the results. Regularly repeating this survey may help in monitoring, and eventually guiding the process of integration of gait analysis technology within equine clinical practice by providing valuable information for individual clinics, educational institutions and the industry producing this technology. Saddle-fit is recognised as an important factor in the pathogenesis of back problems in horses and is empirically being evaluated by pressure measurements in clinical practice, although not much is known about the validity, reliability and usability of these devices in the equine field. This study was conducted to assess critically a pressure measurement system marketed for evaluating saddle fit. Validity was tested by calculating the correlation coefficient between total measured pressure and the weight of 28 different riders. Reliability and discriminative power with respect to different saddle fitting methods were evaluated in a highly standardised, paired measurement set-up in which saddle-fit was quantified by air-pressure values inside the panels of the saddle. Total pressures under the saddle correlated well with riders' weight. A large increase in over-day sensor variation was found. Within trial intra-class correlation coefficients (ICCs) were excellent, but the between trial ICCs varied from poor to excellent and the variation in total pressure was high. In saddles in which the fit was adjusted to individual asymmetries of the horse, the pressure measurement device was able to detect correctly air-pressure differences between the two panels in the back area of the saddle, but not in the front area. The device yielded valid results, but was only reliable in highly standardised conditions. The results question the indiscriminate use of current saddle pressure measurement devices for the quantitative assessment of saddle-fit under practical conditions and suggest that further technical improvement may be necessary. Summary In order to objectively quantify the effect of manipulation on back-related locomotion anomalies in the horse, a recently developed kinematic measuring technique for the objective quantification of thoracolumbar motion in the horse was applied in a dressage horse that was suffering from a back problem. In this horse, clinically, a right-convex bending (scoliosis) from the 10th thoracic vertebra to the second lumbar vertebra was diagnosed. As a result, there was a marked asymmetric movement of the thoracolumbar spine. Functionally, there was severe loss of performance. Thoracolumbar motion was measured in terms of ventrodorsal flexion, lateral flexion, and axial rotation using an automated gait analysis system. Measurements were repeated before and 2 days after treatment, before the second treatment 3 weeks later, and at 4 weeks and 8 months after the second treatment to assess long-term effect. At the same time, performance of the horse was assessed subjectively by the trainer as well. Symmetry of movement improved dramatically after the first treatment. After this, there was a slight decrease in symmetry, but 8 months after the last treatment the symmetry indexes for the various joints were still considerably better than during the first (pre-treatment) measuring session. Subjectively, the trainer did not notice improvement until after measurement session 4. Between sessions 4 and 5 (at 4 weeks and 8 months after the second treatment) there was a change of trainer. The new trainer did not report any back problem, and succeeded in bringing the horse back to its former level in competition. It is concluded that manipulation had a measurable influence on the kinematics of the thoracolumbar spine. However, it is recognized that an improvement in symmetry of motion is not equivalent to clinical improvement and that other measures, such as changes in management, may be more decisive. The flexion test of the distal limb is a commonly used clinical tool in both lame and sound horses. In the latter use it is given some predictive value. In recent studies it has been shown that examiner-related factors (force, time) may strongly influence the outcome of the test. In the present study, the possible influences of a number of horse-related factors and short- and long-term repeatability were investigated. Flexion tests were performed by the same researcher in 100 clinically sound horses under standardized conditions. The outcome of the test was scored on a 9-point semiquantitative scale. The maximum flexion angles of the fetlock joints were measured and the range of motion (ROM) of the fetlock joint was calculated. In the second part of the study, flexion tests were repeated, at intervals of 10 min, 30 min, 48 h and 6 months in 23 horses to assess repeatability. Over 60% of the 100 sound horses had a positive flexion test. Of these, about 50% showed a slight lameness, 35% a mild lameness, and 15% a distinct lameness. There was no influence of weight, height or ROM on the score of the flexion test. The outcome of the flexion test increased significantly with age and was significantly higher in mares than in geldings. When repeating the flexion test with short intervals of 10 and 30 min, the score increased significantly after the second test. Repeated flexion after 48 h did not result in a significantly different outcome. Over a 6-month period, the outcome of the test decreased significantly and the ROM increased significantly. It is concluded that most clinically sound horses have a (slightly) positive flexion test of the distal limb. This and the lack of long-term consistency of the test cast doubt on the presumption that a positive flexion test may be an indication for subclinical joint disorders and question the possible value of the test as a predictor of future joint-related problems. Vertical ground reaction force (GRFz) measurements are the best tool for assessing horses' weight-bearing lameness. However, collection of these data is often impractical for clinical use. This study evaluates GRFz predicted using data from body-mounted IMUs and long short-term memory recurrent neural networks (LSTM-RNN). Twenty-four clinically sound horses, equipped with IMUs on the upper-body (UB) and each limb, walked and trotted on a GRFz measuring treadmill (TiF). Both systems were time-synchronised. Data from randomly selected 16, 4, and 4 horses formed training, validation, and test datasets, respectively. LSTM-RNN with different input sets (All, Limbs, UB, Sacrum, or Withers) were trained to predict GRFz curves or peak-GRFz. Our models could predict GRFz shapes at both gaits with RMSE below 0.40 N.kg−1. The best peak-GRFz values were obtained when extracted from the predicted curves by the all dataset. For both GRFz curves and peak-GRFz values, predictions made with the All or UB datasets were systematically better than with the Limbs dataset, showing the importance of including upper-body kinematic information for kinetic parameters predictions. More data should be gathered to confirm the usability of LSTM-RNN for GRFz predictions, as they highly depend on factors like speed, gait, and the presence of weight-bearing lameness. OBJECTIVETo verify the existence of intertransverse joints (ITJs) in young foals.ANIMALS11 warmblood foals.PROCEDURESPostmortem examination of the lumbar area in foals < 200 days old using CT, MRI, dissection, and histomorphology. Data were analyzed with descriptive statistics.RESULTSAge of foals varied between 1 and 200 days (median, 11 days). Ten foals had 6 lumbar (L) vertebrae, and 1 foal had 5. All 11 foals, irrespective of age, had ITJs between the first sacral and last lumbar vertebrae and between the last and second-to-last lumbar vertebrae. In 6 foals (all with 6 L vertebrae), ITJs also existed between the fourth and fifth L vertebra. One foal, also with 6 L vertebrae, additionally had a unilateral (right) ITJ between the transverse processes of the third and fourth L vertebra. Based on CT, width of ITJs was seemingly greater in young (< 1 month old) foals because of the incomplete ossification of the transverse processes. The ITJs were confirmed and further characterized by MRI, dissection, and histomorphology.CLINICAL RELEVANCEITJs already exist in very young warmblood foals and are present at birth. During the first months of life, these juvenile ITJs develop similarly to other synovial joints with increasing ossification and concomitant decrease of thickness of the cartilage layer. Knowledge of the presence of these ITJs in young animals is clinically relevant, as they should be recognized as nonpathologic when for instance a young foal is presented for presumed arthropathy and examined with advanced imaging techniques. The aim of this study was to test whether the nitric oxide (NO) donor sodium nitroprusside (SNP) has an effect on mineralization in ATDC5 cells. Mineralization in ATDC5 cell culture was induced by addition of β-glycerophosphate or inorganic phosphate, visualized by staining precipitated calcium with an alizarin red stain, and quantified using atomic absorption spectrometry. SNP was shown to inhibit the mineralization of ADTC5 cells. This inhibition was not affected by inhibitors of guanylyl cyclase nor mimicked by a cyclic guanosine monophosphate (cGMP) analog. Furthermore, SNP did not inhibit phosphate uptake or inhibit apoptosis in ATDC5 cells. These findings indicate that SNP can specifically inhibit matrix mineralization via a cGMP-independent pathway and that the effect is not mediated by inhibition of phosphate transport or apoptosis. A population of 805 horses (70 per cent dressage, 20 per cent show jumpers and 10 per cent trotters) with orthopaedic problems was examined for signs of lameness and back problems, irrespective of their original complaints. In the horses with a back problem the prevalence of lameness was 74 per cent and back problems were diagnosed in 32 per cent of the lame horses. These percentages were significantly higher than those recorded in a control population of 399 horses, of which 20 per cent were lame and 12 per cent had back problems. In the group of horses with orthopaedic problems there was a strong association between lameness and back problems and, in particular, there was a high prevalence of lameness among the horses with back problems. We studied the effects of exercise without or with a subsequent period on pasture on Ca2+ ATPase concentration in foal skeletal muscle, and compared the results with those previously reported on Na+, K+ ATPase. Ca2+ ATPase was measured in homogenates as Ca2+-dependent steady-state phosphorylation from [ γ-32P]ATP. From day 7 after birth, 24 foals were divided into three groups: (i) staying in a box stall (Box); (ii) staying in a box stall with an exercise programme of an increasing number of sprints per day (Exercise); and (iii) staying on pasture (Pasture). Half of the foals (12 with four in each treatment group) were killed after 5 months. The remaining foals stayed on pasture until 11 months. In the 5-month Pasture group, Ca2+ ATPase concentration was 29.4 ± 4.3 nmol/g wet weight (wt) ( n = 4) in gluteus medius muscle, 25.2 ± 3.3 nmol/g wet wt ( n = 4) in semitendinosus muscle (both mixed fibre type), and 4.1 ± 1.7 nmol/g wet wt ( n = 3) in the slow masseter muscle. These values were not altered by exercise or by box rest. This was in contrast to the Na+, K+ ATPase concentration which was not different between the three muscles, but showed a 20% rise in gluteus medius and semitendinosus muscle after exercise. In the period from 5 to 11 months on pasture, there was no change in Ca2+ ATPase in any group. In conclusion, the Ca2+ ATPase concentration in foal muscle is around 6-fold higher in mixed fibres than in slow fibres. Summary Second-intention healing of limb wounds in horses is often problematic. Solcoseryl® is a protein-free, standardized dialysate/ultrafiltrate (HD) derived from calf blood, which has been shown to improve healing in both animals and humans. The efficacy of HD in the healing of deep wounds in horses and ponies was investigated. Deep wounds of 20 by 35 mm were created on both metatarsi (skin, subcutis, periosteum) and on both femoral biceps muscles (skin, subcutis, muscle) of five horses and five ponies. The wounds on one side were treated with HD, four times a week during the period that the wounds were bandaged and once daily thereafter. The wounds on the other side were left untreated. In the first 4 weeks of the healing period HD stimulated healing but inhibited healing thereafter. This pattern was significant for all wound groups (P < 0.001). Because of this change in effect, the overall effect on wound healing over the entire period was not significant (P = 0.77). HD stimulated healing initially by provoking a greater initial inflammatory response, faster contraction and faster formation of granulation tissue. Subsequently, HD inhibited healing because it significantly delayed epithelialization and caused protracted inflammation. The effects of HD were most pronounced in the horses. Because this study distinguished between contraction and epithelialization, it could be shown that HD stimulated contraction but inhibited epithelialization. Therefore, HD is useful in horses for the treatment of deep wounds during the initial phase of healing by second intention, i.e. during the first weeks when wound contraction can be expected. Work on curved tracks, e.g. on circles, is commonplace within all forms of horse training. Horse movements in circles are naturally asymmetric, including the load distribution between inner and outer limbs. Within equestrian dressage the horse is expected to bend the back laterally to follow the circle, but this has never been studied scientifically. In the current study 12 horses were measured (optical motion capture, 100 Hz) trotting on left and right circles and on the straight without rider (soft surface). Data from markers placed along the spine indicated increased lateral bending to the inside (e.g. left bending on the left circle) of the thoracolumbar back (difference left circle vs. straight − 3.75°; right circle + 3.61°) and the neck (left − 5.23°; right + 4.80° vs. straight). Lateral bending ROM increased on the circle (+ 0.87° and + 0.62°). Individual variation in straight-circle differences was evident, but each horse was generally consistent over multiple trials. Differences in back movements between circle and straight were generally small and may or may not be visible, but accompanying changes in muscle activity and limb movements may add to the visual impression. For centuries humans have been fascinated by the natural beauty of horses in motion and their different gaits. Gait classification (GC) is commonly performed through visual assessment and reliable, automated methods for real-time objective GC in horses are warranted. In this study, we used a full body network of wireless, high sampling-rate sensors combined with machine learning to fully automatically classify gait. Using data from 120 horses of four different domestic breeds, equipped with seven motion sensors, we included 7576 strides from eight different gaits. GC was trained using several machine-learning approaches, both from feature-extracted data and from raw sensor data. Our best GC model achieved 97% accuracy. Our technique facilitated accurate, GC that enables in-depth biomechanical studies and allows for highly accurate phenotyping of gait for genetic research and breeding. Clinical assessment of spinal motion in horses is part of many routine clinical exams but remains highly subjective. A prerequisite for the quantification of spinal motion is the assessment of the expected normal range of motion and variability of back kinematics. The aim of this study was to objectively quantify spinal kinematics and between -measurement, -surface and -day variation in owner-sound horses. In an observational study, twelve owner-sound horses were trotted 12 times on four different paths (hard/soft straight line, soft lunge left and right). Measurements were divided over three days, with five repetitions on day one and two, and two repetitions on day three (recheck) which occurred 28–55 days later. Optical motion capture was used to collect kinematic data. Elements of the outcome were: 1) Ranges of Motion (ROM) with confidence intervals per path and surface, 2) a variability model to calculate between-measurement variation and test the effect of time, surface and path, 3) intraclass correlation coefficients (ICC) to determine repeatability. ROM was lowest on the hard straight line. Cervical lateral bending was doubled on the left compared to the right lunge. Mean variation for the flexion-extension and lateral bending of the whole back were 0.8 and 1 degrees. Pelvic motion showed a variation of 1.0 (pitch), 0.7 (yaw) and 1.3 (roll) degrees. For these five parameters, a tendency for more variation on the hard surface and reduced variation with increased repetitions was observed. More variation was seen on the recheck (p<0.001). ICC values for pelvic rotations were between 0.76 and 0.93, for the whole back flexion-extension and lateral bending between 0.51 and 0.91. Between-horse variation was substantially higher than within-horse variation. In conclusion, ROM and variation in spinal biomechanics are horse-specific and small, necessitating individual analysis and making subjective and objective clinical assessment of spinal kinematics challenging. Summary: Horses are precocial animals and able to stand and walk within hours after birth. To cope with associated loading, intrauterine bone development has shown to be anticipative. This study provides further insight into the post‐natal development of structurally important features of trabecular and subchondral bone of the talus and sagittal ridge of the tibia of warm‐blooded horses. In all areas studied, the average bone volume fraction showed a gradual increase over time, which was the result of a significant increase in trabecular thickness, without significant changes in the degree of anisotropy. Similar to the mineralised part of the bone, collagen content, measured as average retardation using polarised light microscopy, increased significantly, but the degree of anisotropy of the collagen type I network did not. At birth, the subchondral bone layer had a more trabecular aspect, gradually changing to an even surface with only a few vascular canals at an age of 2 months. Presented results indicate the necessity for a stronger structure, but not for a different structural design after birth, providing further evidence for anticipatory bone development in the horse. The debate on proper head and neck positions (HNP) in horse training is lively, but little is known about the biomechanical effects of various HNPs in horses ridden at walk. The aim was to quantify the influence of different HNPs on the kinematics of horses ridden at walk. The standard competition position (HNP2) was compared to a free, unrestrained position (HNP1), more flexed positions (HNP3, HNP4), a raised extended position (HNP5) and a forward-downward extended position (HNP6). An experimental study in seven high-level dressage horses ridden at walk on a treadmill was designed. Kinetic and kinematic measurements were obtained with different HNPs. HNP2 was used as a speed-matched reference. Kinematics were measured from skin-fixed markers recorded by high-speed video cameras. The kinetics of the limbs were measured by the force-measuring instrumentation of the treadmill. In HNP1, compared to HNP2, the lumbar back and the pelvis were more horizontally positioned (more extended), and fore- and hindlimb pro- and retraction increased, with increased caudal rotation of the femur during the second half of hindlimb stance. HNP6 induced similar changes as HNP1, but caused larger increases in forelimb pro- and retraction. In HNP3, HNP4 and HNP5 the pelvis was more angled (less extended) compared to HNP2 at hindlimb midstance, and in HNP3 and HNP4 also in early hindlimb stance. All three HNPs caused increased maximum flexion of the tarsus, stifle and metatarsophalangeal joint during the swing phase. HNP3 and HNP5, but not HNP4, had a decreasing influence on fore- and hindlimb pro- and retraction, and decreased caudal rotation of the femur during the second half of hindlimb stance. The main limitation was that horses were not ridden overground and the number of horses was small. Osteochondrosis (OC) is a degenerative joint condition developing in a short time frame in young growing gilts that may cause lameness at an older age, affecting welfare and leading to premature culling of breeding sows. Causes of OC are multifactorial including both genetic and environmental factors. Floor type has been suggested to affect OC prevalence and effects might be age dependent during the rearing period. The aim of this study was to investigate possible age-dependent effects of floor type, conventional concrete partially slatted versus wood shavings as deep bedding, on OC prevalence in gilts (Dutch Large White x Dutch Landrace) at slaughter (24 wk of age; 106.5 [14.7 SD] kg of BW). At weaning (4 wk of age; 6.9 [1.3 SD] kg of BW), 212 gilts were subjected to 1 of 4 flooring regimens. Gilts were either subjected to a conventional floor from weaning until slaughter (CC), wood shavings as bedding from weaning until slaughter (WW), a conventional floor from weaning until 10 wk of age after which gilts were switched to wood shavings as bedding (CW), or wood shavings as bedding from weaning until 10 wk of age after which gilts were switched to a conventional floor (WC). After slaughter the elbow, hock, and knee joints were macroscopically examined for OC and scored on a 5 point scale where 0 indicates no OC and 4 indicates the severest form of OC. There was no significant difference (P > 0.4) between treatments on the overall OC prevalence for any joint assessed or at the animal level (all joints combined). At the animal level, however, gilts had greater odds to have OC scores 3 and 4 in the CW treatment (odds ratios [OR] = 2.3; P = 0.05), WC treatment (OR = 2.6; P = 0.02), and WW treatment (OR = 3.7; P < 0.001) compared with gilts in the CC treatment. The results indicate that there are no age-dependent effects of floor types on overall OC prevalence. Although phenylbutazone (PBZ) is commonly used in equine orthopaedic practice, little is known about its in vivo effects on joint inflammation and cartilage turnover. This study investigates the effects of PBZ on inflammatory parameters, matrix metalloproteinase (MMP) activity and cartilage biomarkers in equine joints with acute synovitis. In a two-period cross-over study, transient synovitis was induced at T = 0 h in the middle carpal joint of seven ponies by lipopolysaccharide (LPS) injection. Ponies received PBZ (2 mg/ kg PO twice daily) or placebo for 1 week, starting at T = 2 h. Arthroscopic assessment of the middle carpal joint was performed at T = -504, 48 and 672 h. Synovial fluid (SF) was sampled at T = -504, 0, 8, 24, 48, 168,336 and 672 h and analysed for leukocytes and total protein, substance P, general MMP activity, glycosaminoglycans (GAG), collagen II cleavage marker C2C and synthesis marker CPU. Markers in PBZ- vs. placebo-treated joints were compared over time using a linear mixed model. LPS injection caused marked transient synovitis without visible cartilage changes. Substance P and general MMP activity were not significantly reduced by PBZ treatment, nor were SF GAG or C2C concentrations at any time point. Concentration of CPU was significantly lower at T = 24 and 168 h in PBZ treated joints compared to placebo. Although PBZ is clinically effective in treating acute synovitis, it does not limit inflammation-induced cartilage catabolism and may transiently reduce collagen anabolism as evidenced by SF markers. Osteochondrosis (OC) is one of the main causes of leg weakness causing premature culling in breeding sows and develops in a short time frame in young growing gilts. Dietary restriction may have different effects on OC prevalence depending on the age of the gilts. The aim of this study is to investigate age-dependent effects of dietary restriction, ad libitum vs. restricted (80% of ad libitum), on the occurrence of OC in gilts at slaughter (26 wk of age). At weaning (4 wk of age), 211 gilts were subjected to one of 4 treatments of a feeding regime. Gilts were administered either ad libitum feeding from weaning until slaughter (AA); restricted feeding from weaning until slaughter (RR); ad libitum feeding from weaning until 10 wk of age, after which gilts were switched to restricted feeding (AR); or restricted feeding from weaning until 10 wk of age, after which gilts were switched to ad libitum feeding (RA). At slaughter, the elbow, hock, and knee joints were harvested. Joints were scored macroscopically for articular surface deformations indicative of OC. Gilts in the RA treaUnent had significantly higher odds of being affected by OC than gilts in the RR and AR treatments in the hock joint (OR = 3.3, P = 0.04 and OR = 8.5, P = 0.002, respectively) and at animal level (OR = 2.5, P = 0.001 and OR = 1.9, P = 0.01, respectively). Gilts in the AA treatment had higher odds of being affected by OC than gilts in the AR treatment in the hock joint (OR = 5.3, P = 0.01). The results indicate a possible pathway to reduce the prevalence of OC in breeding gilts that will have to last several parities. Arthroscopy enables direct inspection of the articular surface, but provides no information on deeper cartilage layers. Optical coherence tomography (OCT), based on measurement of reflection and backscatter-ing of light, is a diagnostic technique used in cardiovascular surgery and ophthalmology. It provides cross-sectional images at resolutions comparable to that of low-power microscopy. The aim of this study was to determine if OCT is feasible for advanced clinical assessment of lesions in equine articular cartilage during diagnostic arthroscopy. Diagnostic arthroscopy of 36 metacarpophalangeal joints was carried out ex vivo. Of these, 18 joints with varying degrees of cartilage damage were selected, wherein OCT arthroscopy was conducted using an OCT catheter (diameter 0.9 mm) inserted through standard instrument portals. Five sites of interest, occasionally supplemented with other locations where defects were encountered, were arthroscopically graded according to the International Cartilage Repair Society (ICRS) classification system. The same sites were evaluated qualitatively (ICRS classification and morphological description of the lesions) and quantitatively (measurement of cartilage thickness) on OCT images. OCT provided high resolution images of cartilage enabling determination of cartilage thickness. Comparing ICRS grades determined by both arthroscopy and OCT revealed poor agreement. Furthermore, OCT visualised a spectrum of lesions, including cavitation, fibrillation, superficial and deep clefts, erosion, ulceration and fragmentation. In addition, with OCT the arthroscopically inaccessible area between the dorsal MC3 and P1 was reachable in some cases. Arthroscopically-guided OCT provided more detailed and quantitative information on the morphology of articular cartilage lesions than conventional arthroscopy. This manuscript describes a new classification of the various joint-related lesions that can be seen in the young, growing horse based on their anatomical and functional aetiopathogenesis. Juvenile osteochondral conditions (JOCC) is a term that brings together specific disorders according to their location in the joint and their biomechanical origin. When a biomechanical insult affects the process of endochondral ossification different types of osteochondrosis (OC) lesions may occur, including osteochondral fragmentation of the articular surface or of the periarticular margins, or the formation of juvenile subchondral bone cysts. In severe cases, osteochondral collapse of the articular surface or the epiphysis or even an entire small bone may occur. Tension on ligament attachments may cause avulsion fractures of epiphyseal (or metaphyseal) ossifying bone, which are classified as JOCC, but do not result from a disturbance of the process of endochondral ossification and are not therefore classified as a form of OC. Intra-articular injection of opioids provides analgesia in painful equine joints and u.-opioid receptors (MORs) have been demonstrated in equine synovial membranes. The aim of this study was to determine whether acute inflammatory conditions will lead to up-regulation of MOR in equine synovial membranes and whether anti-inflammatory treatment can prevent any such upregulation. In a two-period, blinded, placebo-controlled randomised cross-over design, lipopolysaccharide (LPS, 1.0 ng) was injected into the left or right middle carpal joint of seven healthy ponies. Arthroscopy and synovial membrane biopsy was performed under general anaesthesia at baseline, 48 h (T48) and 672 h (T672) after LPS injection, with ponies assigned to receive either phenylbutazone (PBZ 2.2 mg/kg PO BID) or placebo from 2 h post-LPS. Ponies were scored for pain and lameness. Repeated synovial fluid samples were obtained and the degree of synovitis scored both macroscopically and microscopically. The density and staining pattern of MOR-like protein in synovial membrane biopsies over the course of the synovitis with or without PBZ treatment was evaluated using immunohistochemical techniques. LPS injection consistently induced a severe transient synovitis. Pain and lameness were significantly attenuated by treatment with PBZ. Up-regulation of MOR-like protein in the inflamed equine synovial membrane could be demonstrated in the placebo treated animals, but not in the PBZ-treated animals overall, although there were no significant differences at any individual time-point between the two groups. Records of lame horses (2004-2007) with (peri-)synovial pain of the metacarpophalangeal joint (MCPJ, n=53), digital flexor tendon sheath (DFTS, n=20), and distal interphalangeal joint (DIPJ, n=31) were evaluated (n=104). Cases were grouped according to the following treatments: (1) systemic administration of NSAIDs (n=40); (2) intrasynovial administration of glucocorticosteroids (n=30); (3) combination treatment (n=34). Outcome was defined as successful when the owner reported that the horse had returned to its previous level of performance within a period of up to six months after treatment. Statistical evaluation was performed using Fisher's Exact tests and multivariable logistic regression analyses at P < 0.05. Successful outcome was significantly higher for horses in treatment group 3 (19/34, 55.9 per cent, P=0.021) than in treatment groups 1 (11/40, 27.5 per cent, OR 0.21, 95 per cent CI 0.06 to 0.71) and 2 (8/30, 26.6 per cent, OR 0.18, 95 per cent CI 0.06 to 0.56). A significant association was only found with regard to the affected synovial structure (P=0.025); relative to the DIPJ, involvement of the MCPJ and DFTS was four to five times more likely to provide a successful outcome (OR 4.18 and 5.59, 95 per cent CI 1.24 to 14.08 and 1.42-22.22, respectively). Summary Squamous cell carcinoma (SCC) is the most common tumour of the penis and prepuce of the horse and is found mainly in older animals. Reports on breed predilection for penile tumour formation are equivocal but castration, coat colour, poor genital hygiene and various infectious agents have been suggested to predispose to the development of SCC. Careful assessment of the primary tumour is an important first step in the design of an optimal treatment protocol. Tumour size, invasiveness, differentiation grade and presence of metastases are relevant to the decision to pursue additional diagnostic procedures or specific treatment options. Complete elimination of the neoplasm and, therefore, risk of recurrence is highly dependent on the type of therapy chosen. However, the size and histopathological features of the primary tumour are also important factors with respect to prognosis. The repair process of tendon injuries, which are common in both human and equine athletes, is slow and the quality of the repair tissue is often inferior to the original tendon tissue, which frequently leads to re‐injury. The relatively poor vascularization of tendons is considered to be one of the reasons for their limited healing potential. Recently, platelet‐rich plasma (PRP), an autologous concentrate of platelets, rich in growth factors, has been shown to enhance the repair process of injured tendons. This effect has been ascribed to the high levels of growth factors in PRP, several of which are known to be involved in tendon repair. Among many other growth factors, the vascular endothelial growth factor, a powerful stimulator of angiogenesis, is abundantly present in PRP, suggesting that enhancement of neovascularization might be one of the working mechanisms. In this study, the effect of PRP on neovascularization was studied in experimentally induced tendon injuries using color Doppler ultrasonography and immunological staining of Factor VIII. PRP induced significantly more neovascularization than the placebo treatment until at least 23 weeks after treatment, as detected by both Doppler ultrasonography and Factor VIII staining. Osteochondrosis (OC) is an important orthopedic developmental disorder in many horse populations. A review of the literature revealed widely variable heritability estimates for the disorder. We estimated the genetic variables (heritabilities and genetic correlations) of various manifestations of OC. Femoropatellar, tarsocrural, and metacarpophalangeal and metatarsophalangeal joints of 811 randomly selected yearlings from the Royal Warmblood Studbook of the Netherlands descending from 32 representative stallions, were scored for OC at 28 predilection sites. At each site, OC was scored in 5 categories, distinguishing between flattened bone contours and fragments. At the animal level, the overall heritability of OC was 0.23, the heritability of flattened bone contours was 0.08, and the heritability of fragments was 0.22. At the joint level, heritability was greatest in the tarsocrural joints, intermediate in the metacarpophalangeal and metatarsophalangeal joints, and least in the femoropatellar joints. The heritability estimates for the coritralateral joint homologs were very similar. The genetic correlation between the tarsocrural arid femoropatellar joint was strong, whereas correlations between the metacarpophalangeal and metatarsophalangeal and other joints were moderate. The genetic correlation between flattened bone contours and fragments at the animal level was 0.80. Scoring OC on a 5-point categorical scale resulted in greater heritability on the observed scale than when analyzing OC as a binary trait. Our results suggest that selection against OC could best be performed by taking into account the OC status of all 4 joints, the femoropatellar, the tarsocrural, and the metacarpophalangeal and metatarsophalangeal joints, and discerning between flattened bone contours and fragments. The objectives of the present study were to test the hypotheses that: (1) the composition of the extracellular matrix of the equine superficial digital flexor tendon (SDFT) shows great functional similarities with articular cartilage, i.e. that significant differences exist in biochemical composition of differently loaded areas (which in the case of tendons may be more apparent as tendon shows more obvious differences than cartilage); and (2) that, as in articular cartilage, no substantial alterations in biochemical composition take place during ageing once adulthood has been attained. Tendon samples were taken from 60 adult slaughter horses from a central area at cross-section in the mid-metacarpal region and at the height of the proximal sesamoid bones (sesamoid region) of the SDFT. Contents of collagenous and non-collagenous components were determined. None of the parameters were correlated with age in either region, except for a significant increase in pentosidine cross-links with age in the sesamoid region. Between the two anatomical regions, there were significant differences in all variables, except for hydroxylysylpyridinoline cross-links. It was concluded that in the equine SDFT, similar to articular cartilage, most molecular parameters are not influenced by age in mature horses, indicating a low remodelling rate. A recently developed indenter system that aims at determination of local structural properties of the cartilage surface was evaluated for suitability in the horse. To this aim, maximum indenter force was measured of the articular surface and related to biochemical characteristics of the cartilage at different sites of the distal metacarpal bone (MC). Significant topographical variation exists in structural properties of the articular surface of the MC. The dorsal margin showed a significantly lower maximum indenter force than more centrally located areas, indicating an increased compliance under load. A high maximum indenter force correlated positively with high glycosaminoglycan levels. No correlation was found between measured indenter force and collagen content of the cartilage. It was concluded that the instrument is able to quantify differences in structural properties of the cartilage surface which reflect glycosaminoglycan content and hence is capable of giving some indication of cartilage quality.
