Seepage-induced instabilities pose a challenge in many geotechnical applications. Particle-scale mechanisms govern the initiation of instability. However, current understanding is based on a macro-scale perspective that draws on continuum mechanics. Recent developments in imaging and numerical analysis can provide the particle-scale fundamental perspective needed to develop a comprehensive insight. This contribution demonstrates the value of combining particle-scale experimental and numerical studies. The experiments consider transparent soil samples created using refractive image matching and monitored by particle image velocimetry (PIV). Three-dimensional pore topology is extracted from a series of two-dimensional images and imported into computational fluid dynamics (CFD) simulations. Permeability is estimated by three distinct approaches: using flow rate, PIV- and CFD-generated data. The flow fields obtained from PIV and CFD are in good agreement considering both flow rate contour plots and flow rate distributions; this demonstrates the successful reconstruction of three-dimensional pore structure and flow-field analysis. The comparison also reveals that the side boundary effects in CFD simulations are constrained within a limited region. The multi-plane results characterise the variance of flow velocity with the three-dimensional pore topology. Finally, the fluid–particle interactions obtained from CFD results show a larger variance in the angular particle packings. [ABSTRACT FROM AUTHOR]

The granular temperature is an index of the level of collisional activity in a granular flow, and increasingly important in the verification of extended kinetic theories. The granular temperature is related to the square of the difference between a particle’s velocity and that of the group mean. Image analysis of high-speed video is the most common method to measure granular temperature in experimental flows and depends on correlation of a search mask or a portion of the original image to the next image frame to determine the particle’s movement. This invariably involves some level of estimation of the location at a resolution finer than the pixels that make up the image. However, errors in determining particle movement at the subpixel level can be shown to have a significant impact on granular temperature identification. We show that taking particle movement to be a chain of displacement vectors provides context to the apparent impulses on the particle. Here we propose two novel methods for determining the granular temperature of experimental flows, namely a novel method of initializing Particle Image Velocimetry (PIV) for granular systems where each search subset is centred on a previously determined particle location to reduce bias, and a method of filtering the apparent impulses on a particle on a frequency basis. We term these methods Guided-PIV and Impulse Frequency Filtering (IFF), respectively. In a verification exercise using synthetically generated images, we show Guided-PIV to produce substantially more accurate results than ordinary applications of PIV. The IFF method is shown to greatly reduce the influence of analyzed framerate on granular temperature results. Our results demonstrate practical improvements for granular temperature identification from image analysis, throughout a range of experimental image quality levels, and we anticipate that these improvements will enable experimental assessment towards verification of theorized models of collisional-frictional granular flows. Objectives: The risk of cardiometabolic complications in children with perinatally acquired HIV infection (PHIVs) and in perinatally HIV‐exposed but uninfected children (HEUs) and its relationship to systemic inflammation and markers of gut integrity are not well established. In this current study, we assed insulin resitance in PHIV compared to HEUs and HIV unexposed uninfected children and explored potential association with intestinal damage biomarkers. Methods: This was a cross‐sectional study in PHIVs, HEUs and HIV‐unexposed, uninfected children (HUUs) aged 2–10 years enrolled in Uganda. PHIVs were on stable antiretroviral therapy (ART) with HIV viral load < 400 HIV‐1 RNA copies/mL. Insulin resistance was estimated using the homeostasis model assessment of insulin resistance (HOMA‐IR). We measured markers of systemic inflammation, monocyte activation and gut integrity. Kruskal–Wallis tests were used to compare markers by HIV status; Pearson correlation and multiple linear regressions were used to assess associations of the HOMA‐IR index with biomarkers of intestinal damage and translocation. Results: Overall, 172 participants were enrolled in the study (57 PHIVs, 59 HEUs and 56 HUUs). The median age was 7.8 [interquartile range (IQR) 6.39, 8.84] years, 55% were female and the median body mass index (BMI) was 15 (IQR 14.3, 15.8) kg/m2. Among PHIVs, the median CD4% was 37%, and 93% had viral load ≤ 20 copies/mL. PHIVs had higher waist:hip ratio, high‐density lipoprotein (HDL) cholesterol, triglycerides and HOMA‐IR index than the other groups (P ≤ 0.02). Factors correlated with insulin resistance included higher BMI and HDL cholesterol and lower soluble tumour necrosis factor receptor I (sTNFRI) (P ≤ 0.02). There was no correlation between any of the other inflammatory or gut biomarkers and HOMA‐IR index (P ≥ 0.05). After adjusting for age and sTNFRI, BMI remained independently associated with the HOMA‐IR index (β = 0.16; P < 0.01). Conclusions: Despite viral suppression, Ugandan PHIVs have disturbances in glucose metabolism. A rigid-walled 'transparent soil' permeameter has been developed to study visually the mechanisms occurring during seepage-induced internal erosion in susceptible granular media under upward flow. The experiments use borosilicate glass particles in place of soil, and an optically matched oil mixed with fluorescent dye in place of water. The technique known as plane laser-induced fluorescence (PLIF) enables a two-dimensional plane of particles and fluid to be viewed inside the permeameter, away from the walls. Results of tests have provided close agreement with those of other researchers on soil of comparable particle size grading. Unstable materials showed migration of fine grains under mean hydraulic gradients as low as i = 0·25, whereas stable materials eventually failed by heave at hydraulic gradients close to unity. Internally unstable soils where the loads were predominantly supported by the coarser fraction exhibited suffusion (fines migration without disruption of the load-bearing system); those supported by both coarse and fine particles exhibited suffosion (i.e. volume change during fines migration). Quantitative image analysis conducted on one unstable sample showed areas of open void space migrating through the sample at low hydraulic gradients near critical, as defined by Skempton and Brogan in 1994. This occurred before the externally measured local hydraulic gradients began to diverge significantly from the mean. The testing technique developed shows that optically matched glass and oil behave mechanically similarly to soil and water, and that the PLIF technique coupled with image analysis can provide an additional insight into the mechanisms of internal erosion. The Dongla Bridge, Muli County, China, is a key access road between counties in Sichuan Province and the only traffic facility for an important 500-kV electricity substation which provides power to nearby counties. In June 2011, during the bridge construction, an unknown landslide, subsequently named the Dongla Ancient Landslide (DAL), was encountered. Local reactivation of a portion of the DAL, termed the Dongla Reactivated Landslide (DRL), was indicated by evident ground deformation at the Dongla Bridge. In spite of this, transportation of some heavy electrical equipment to the substation was scheduled on 9 May 2013. So, to guarantee the stability and security of the bridge became a primary emergency task. Detailed field investigations of the landslide occurrence, monitoring and emergency mitigation construction were carried out in the period between 2011 and 2014. This resulted in a temporary stabilization of the revived landslide and ensured the timely transportation of the heavy electrical equipment. It is thought that the revival mechanism of the DRL is creep, translational sliding, and was mainly triggered by artificial slope-cutting, but also influenced by precipitation, river erosion, and man-made activities in the form of small-scale mining operations. Following complete failure of the DRL, the probability of landslide-dam and dam breach is proved to be small by risk analyses. Although microRNAs (miRs) have been implicated in the pathogenesis of various human malignancies, limited information is available regarding mechanisms by which these noncoding RNAs contribute to initiation and progression of tobacco-induced esophageal cancers. In this study, array and quantitative reverse transcriptase-PCR techniques were used to examine miR expression in immortalized esophageal epithelia (IEE) and esophageal adenocarcinoma (EAC) cells cultured in normal media with or without cigarette smoke condensate (CSC). Under relevant exposure conditions, CSC significantly decreased miR-217 expression in these cells. Endogenous levels of miR-217 expression in cultured EAC cells (EACC)/primary EACs were significantly lower than those observed in IEE/ paired normal esophageal tissues. RNA crosslink immunoprecipitation, quantitative reverse transcriptase-PCR (qRT-PCR) and immunoblot experiments demonstrated direct interaction of miR-217 with kallikrein 7 (KLK7), encoding a putative oncogene not previously implicated in EAC. Repression of miR-217 correlated with increased levels of KLK7 in primary EACs, particularly those from smokers. Chromatin and methylated DNA immunoprecipitation experiments demonstrated that CSC-mediated repression of miR-217 coincided with DNMT3b-dependent hypermethylation and decreased occupancy of nuclear factor 1 within the miR-217 genomic locus. Deoxyazacytidine induced miR-217 expression and downregulated KLK7 in EACC; deoxyazacytidine also attenuated CSC-mediated miR-217 repression and upregulation of KLK7 in IEE and EACC. Overexpression of miR-217 significantly decreased, whereas overexpression of KLK7 increased proliferation, invasion and tumorigenicity of EACC. Collectively, these data demonstrate that epigenetic repression of miR-217 contributes to the pathogenesis of EAC via upregulation of KLK7 and suggest that restoration of miR-217 expression may be a novel treatment strategy for these malignancies. In a prospective study the criteria and characteristics associated with the admission of acutely ill psychiatric patients to in-patient or day hospital care were examined. Over a four-month period, 54 patients were admitted to hospital and 43 to a day hospital. There was significantly more schizophrenics in the hospital. Day hospital patients were significantly younger, had shorter psychiatric histories, were considered less severely ill and had more insight into their illness. Hospital patients had poorer employment histories, and perceived their families as less supportive; admission had more often been requested by them or their families. [ABSTRACT FROM AUTHOR]

The dynamic fragmentation of rock within rock avalanches is examined using the fragmentation concepts introduced by Grady and co-workers. The analyses use typical material values for weak chalk and limestone in order to determine theoretical strain rate thresholds for dynamic fragmentation and resulting fragment sizes. These are found to compare favourably with data obtained from field observations of long runout rock avalanches and chalk cliff collapses in spite of the simplicity of the approach used. The results provide insight as to the energy requirements to develop long runout behaviour and hence may help to explain the observed similarities between large rock avalanches and much smaller scale chalk cliff collapses as seen in Europe. Background and purpose: Niemann-Pick disease type C (NPC) is a progressive neurovisceral disorder associated with dystonia, ataxia and a characteristic gaze palsy. Neuropathological studies have demonstrated brainstem atrophy associated with neuronal inclusions and loss, and neurofibrillary tangles, although it is not known whether this pathology can be detected in vivo or how these changes relate to illness variables, particularly ocular-motor changes. Our aim was to utilize a method for brainstem atrophy, validated in progressive supranuclear palsy (PSP), in a group of adult patients with NPC, and explore its relationship to illness variables and ocular-motor functioning. Methods: We calculated the midbrain and pontine area, and pontine-to-midbrain ratio (PMR) from midsagittal images of 10 adult patients with NPC and 27 age- and gender-matched controls. Measures were correlated with illness variables, and measures of horizontal saccadic functioning. Results: Pontine-to-midbrain ratio was 14% higher in the NPC group, but this difference was not significant. However, PMR showed a significant positive correlation with duration of illness and a measure of illness severity. Furthermore, PMR was significantly negatively correlated with saccadic peak velocity and gain, and self-paced saccadic performance. Conclusions: Pontine-to-midbrain ratio was increased in adult patients with NPC compared to controls, although not to the same degree as previously described in PSP, which also presents with significant gaze palsy. These changes were driven predominantly by progressive midbrain atrophy. LKB1/STK11 is a tumor suppressor and a negative regulator of mammalian target of rapamycin signaling. It is inactivated in 30% of lung cancer cell lines but only 5-15% of primary lung adenocarcinomas. There is evidence that homozygous deletion (HD) of chromosome 19p at the LKB locus contributes to the inactivation of the gene in primary human lung cancers. Here, we used several complementary genetic approaches to assess the LKB1 locus in primary non-small cell lung cancers (NSCLCs). We first analyzed 124 NSCLC cases for allelic imbalance using eight microsatellite markers on chromosome 19p, which revealed an overall rate of 65% (80 of 124) loss of heterozygosity (LOH). We next used chromogenic in situ hybridization (CISH) to directly examine the chromosomal status of the LKB1 locus. In all, 65 of 124 LOH tested samples were available for CISH and 58 of those (89%) showed either loss of one copy of chromosome 19p (LOH, 40 of 65 cases, 62%) or both copies (HD 18 of 65 cases, 28%). The occurrence of HD was significantly more frequent in Caucasian (35%) than in African-American patients (6%) (P=0.04). A total of 62 of 124 samples with LOH at one or both markers immediately flanking the LKB1 gene were further analyzed by directly sequencing the complete coding region, which identified 7 of 62 (11%) tumors with somatic mutations in the gene. Jointly, our data identified total inactivation of the LKB1 gene by either HD or LOH with somatic mutation in 39% of tested samples, whereas loss of chromosome 19p region by HD or LOH at the LKB1 region occurred in 90% of NSCLC. We describe the differential presentation of schizophrenia-like psychosis in two siblings with the 'variant' biochemical presentation of adult Niemann-Pick disease type C. The male sibling presented with psychosis at age 16 years and cognitive and motor disturbance at age 25 years, whereas his elder sister, sharing the same mutation but showing less severe biochemical, neuroimaging and ocular motor parameters, presented with a similar schizophrenia-like illness with associated cognitive and motor disturbance at age 31 years. Their illness onset, course and response to treatment mirrors the sex dimorphism seen in schizophrenia, and is suggestive of an interaction between the neurobiology of their metabolic disorder and sex differences in neurodevelopment. Midbrain dopamine neurons are phasically activated by a variety of sensory stimuli. It has been hypothesized that these activations contribute to reward prediction or behavioural switching. To test the latter hypothesis we recorded from 131 single neurons in the ventral tegmental area and retrorubral field of thirsty rats responding during a modified go/no-go task. One-quarter ( n = 33) of these neurons responded to conditioned stimuli in the task, which varied according to the outcome with which they were associated (saccharin or quinine solution) and according to whether they triggered a switch in the ongoing sequence of the animal's behaviour (‘behavioural switching’). Almost half the neurons (45%) responded differentially to saccharin- vs. quinine-conditioned stimuli; the activity of a minority (15%) correlated with an aspect of behavioural switching (mostly exhibiting changes from baseline activity in the absence of a behavioural switch) and one-third (33%) encoded various outcome–switch combinations. The strongest response was excitation to the saccharin-conditioned stimulus. Additionally, a proportion (38%) of neurons responded during outcome delivery, typically exhibiting inhibition during saccharin consumption. The neurons sampled did not fall into distinct clusters on the basis of their electrophysiological characteristics. However, most neurons that responded to the outcome-related properties of conditioned stimuli had long action potentials (> 1.2 ms), a reported characteristic of dopamine neurons. Moreover, responses to saccharin-conditioned stimuli were functionally akin to dopamine responses found in the macaque and rat nucleus accumbens responses observed within the same task. It has been proposed that nucleus accumbens neurons respond to outcome (reward and punishment) and outcome-predictive information. Alternatively, it has been suggested that these neurons respond to salient stimuli, regardless of their outcome-predictive properties, to facilitate a switch in ongoing behavior. We recorded the activity of 82 single-nucleus accumbens neurons in thirsty rats responding within a modified go/no-go task. The task design allowed us to analyze whether neurons responded to conditioned stimuli that predicted rewarding (saccharin) or aversive (quinine) outcomes, and whether the neural responses correlated with behavioral switching. Approximately one third (28/82) of nucleus accumbens neurons exhibited 35 responses to conditioned stimuli. Over 2/3 of these responses encoded the nature of the upcoming rewarding (19/35) or aversive (5/35) outcome. No response was selective solely for the switching of the rat's behavior, although the activity of approximately one third of responses (11/35) predicted the upcoming outcome and was correlated with the presence or absence of a subsequent behavioral switch. Adhesion molecules and chemokine receptors are involved in tissue-specific homing of T cells to the skin and play an important role in the pathophysiology of cutaneous lymphoma. It has recently been reported that the chemokine CCL27 expressed by keratinocytes attracts lymphocytes bearing the chemokine receptor CCR10.To investigate the expression of CCR4, CCR7 and CCR10 on skin-homing CLA+ and CD4+ T cells in the peripheral blood of patients with Sézary syndrome (SS), a rare leukaemic variant of cutaneous T-cell lymphoma.Lymphocytes from five patients with SS, six patients with mycosis fungoides and four healthy volunteers were isolated and analysed using flow cytometry. Additionally, the T-cell receptor (TCR)-Vβ CDR3 regions were cloned and sequenced in two patients.We found that CCR4 is expressed on almost all CLA+ and CD4+ memory T cells. Using monoclonal antibodies specific for single TCR-Vβ chains we identified malignant T cells in four patients with SS. Importantly, we found that most but not all malignant Sézary cells expressed the skin-homing chemokine receptor CCR10. Additionally, we found that a significant proportion of these cells also expressed the lymph node-homing chemokine receptor CCR7.Our results support the concept that chemokine receptors play an important role in the pathophysiology of SS and suggest that the malignant clone may represent an expansion of skin-homing cutaneous‘central’ memory T cells in the peripheral blood of these patients. Rationale. Second-order schedules of reinforcement have been used extensively to model reward-seeking and drug-seeking behaviour. Second-order stimuli within second-order schedules have been shown to enhance response rates during operant responding for natural reinforcers and drug reinforcers. This has led some to view second-order schedules of drug reinforcement as a model maintained of drug-seeking in addicts by drug-associated stimuli. However, the functional role of the second-order stimulus within second-order schedules is complex. Objective. We investigated the role of second-order stimuli within a second-order schedule of reinforcement [FI 4 min (FR10: S)] maintained by sweetened water reinforcement. Methods. Eight rats were trained to press a bar on a second-order schedule of reinforcement and tested in the presence and absence of the second-order stimulus. Results. In contrast to most previous work, overall bar-pressing rates were significantly increased when the second-order stimulus was omitted (second-order stimulus omission: 0.17 Hz (±0.04, 95% CI); second-order stimulus present: 0.13 Hz (±0.04, 95% CI)). However, second-order stimuli also changed the pattern of responding whereby rats would make a bout of bar presses prior to the presentation of the second-order stimulus and then pause briefly after the second-order stimulus. In the absence of second-order stimuli, responding was uniformly high. Control measures, such as the ability of the second-order stimulus to evoke checking for the primary reinforcers, indicated that the second-order stimulus was associated with the primary reinforcer. Conclusions. These results demonstrated that although second-order stimuli maintained responding and caused the rat to check for primary reinforcement, overall response rates were increased when the second-order stimuli were omitted. This has implications for interpreting the results of studies where overall response rates within second-order schedules have been the only measure used to assess the effects of potential anti-addiction drugs. Future studies could be improved by performing a second-order stimulus omission test analysing both the overall response rates and the temporal organization of responding with respect to the second-order stimulus. Objective: To contrast the mortality rates and changes in the causes of death of very preterm infants (23-27 weeks), before and after the introduction of exogenous surfactant in 1991, and to identify any preventable causes of death remaining in the 1990s.Methodology: This was a cohort study on consecutive preterm infants of 23-27 weeks' gestational age born in the Royal Women's Hospital, Melbourne, a level III perinatal centre. The infants were livebirths free of lethal anomalies from two distinct eras, 1983-90, and 1992-96, inclusive. The main outcome measures were mortality during the primary hospitalization and the causes of death before and after the introduction of exogenous surfactant in 1991.Results: In 1983-90, 261 of 508 livebirths (51.4%) of 23-27 weeks' gestational age died, a significantly higher proportion than the 109 of 384 (28.4%) livebirths who died in the period 1992-96. The mortality rate fell significantly with increasing gestational age and was lower at each week of gestational age in 1992-96. More infants who died in 1992-96 were treated intensively in the neonatal intensive care unit (NICU). Of the group of infants who died or who were treated intensively in NICU, respiratory causes of death predominated. However, the causes of death changed over time. In 1992-96 proportionally fewer infants died from respiratory causes (1983-90, 82.5%; 1992-96, 60.0%; odds ratio (OR) 0.31, 95%; confidence interval (CI) 0.16-0.57), but more from septic causes (1983-90, 14.3%; 1992-96, 43.8%; OR 4.9, 95%; CI 2.6-9.2).Conclusions: As the mortality rate has fallen over time, respiratory causes of death have diminished, but septic causes of death have increased. Further advances in the use of exogenous surfactant and respiratory support may reduce respiratory deaths. Effective strategies to reduce nosocomial infections are urgently required. Background: Exposure to environmental tobacco smoke (ETS) is considered to be a major lung cancer risk factor for never smokers. We investigated the hypothesis that never-smoking women who are exposed to ETS and develop lung cancer are a genetically susceptible population.Methods: Archival tumor tissues were analyzed from 106 never-smoking women enrolled in a case-control study of ETS (and other personal and environmental factors) and lung cancer risk. We analyzed germline polymorphisms in genes that have been associated with cancer susceptibility and whose products activate (cytochrome P450 1A1 [CYP1A1]) and detoxify (glutathione S-transferases M1 [GSTM1] and T1 [GSTT1]) chemical carcinogens found in tobacco smoke.Results: When compared with never smokers who had no ETS exposure and developed lung cancer (n = 55), never smokers with exposure to ETS who developed lung cancer (n = 51) were more likely to be deficient in GSTM1 activity (i.e., were GSTM1 null) because of a genetic polymorphism in the GSTM1 gene (odds ratio = 2.6; 95% confidence interval = 1.1-6.1). A statistically significant rising trend in risk occurred with increasing ETS exposure (two-sided P =. 02), reaching a more than sixfold excess risk in those exposed to 55 pack-years of ETS (ETS pack-year = ETS produced by an active smoker, within a confined space such as a room, who smokes one pack of cigarettes a day for a year). No evidence was found of associations between GSTT1 deficiency or the CYP1A1 valine variant and lung cancer risk due to ETS exposure.Conclusions: A common genetic polymorphism divides the population of never smokers into two groups of approximately equal size, one (homozygous carriers of the GSTM1 null allele) that has a statistically significant greater risk of lung cancer from ETS than the other (heterozygous or homozygous carriers of the wild-type GSTM1 allele). This study examined the possibility that lesions of the nucleus accumbens in rats impair the perception of the ”cost of reward”, as defined by the number of operant responses needed to obtain a food pellet. In a first experiment, visual cues indicated the cost of reward under a multiple-ratio schedule of reinforcement. In a second experiment, the number of lever presses required for each reward incremented with each trial in a progressive-ratio schedule of reinforcement. Lesions of the nucleus accumbens altered the behavioral response to the increasing cost of reward when there was an absence of external cues. There was no change in the ability of the lesioned rats to respond to visual cues that indicated reward availability. The results are considered in terms of the traditional idea of the nucleus accumbens as a limbic-motor interface: it is suggested that, if the nucleus accumbens serves such a function, it is limited to only some contexts. There is considerable disagreement on the classification of Asperger's syndrome and its relationship to autism. Unlike autism, Asperger's is not usually recognised before 30 months of age, and speech delay is not typical. However, if a child exhibits all the symptoms of autism apart from these two features, are these sufficient grounds for withholding a diagnosis of autism? This paper describes four boys and their father who, by presenting at various points on the autistic/Asperger spectrum, embody this dilemma for the diagnostician. The article describes two linear programming solutions to the assembly-line balancing problem. The balancing problem arises from a number of operations that must be performed sequentially with partial ordering constraints. The linear constraints are in variables that can be evaluated in linear objective functions. The linear programs developed to solve the balancing problem involve large numbers of constraint equations, variables, special integer variables. This makes complex the computations to develop an algorithm needed to solve the problem.

This paper reports some research, ideas, and theory about managerial decision making. The first research projects dealt with are aggregate production and employment scheduling. From this is developed the idea that management's own (past) decisions can be incorporated into a system of improving their present decisions. Decision rules are developed, with the coefficients in the rules derived from management's past decisions (rather than from a cost or value model).
