Destructive tsunamis are most often generated by large earthquakes occurring at subduction interfaces, but also other "atypical" sources—defined as crustal earthquakes and non-seismic sources altogether—may cause significant tsunami threats. Tsunamis may indeed be generated by different sources, such as earthquakes, submarine or coastal landslides, volcano-related phenomena, and atmospheric perturbations. The consideration of atypical sources is important worldwide, but it is especially prominent in complex tectonic settings such as the Mediterranean, the Caribbean, or the Indonesian archipelago. The recent disasters in Indonesia in 2018, caused by the Palu-Sulawesi magnitude Mw 7.5 crustal earthquake and by the collapse of the Anak-Krakatau volcano, recall the importance of such sources. Dealing with atypical sources represents a scientific, technical, and computational challenge, which depends on the capability of quantifying and managing uncertainty efficiently and of reducing it with accurate physical modelling. Here, we first introduce the general framework in which tsunami threats are treated, and then we review the current status and the expected future development of tsunami hazard quantifications and of the tsunami warning systems in Italy, with a specific focus on the treatment of atypical sources. In Italy, where the memory of historical atypical events like the 1908 Messina earthquake or the relatively recent 2002 Stromboli tsunami is still vivid, specific attention has been indeed dedicated to the progressive development of innovative strategies to deal with such atypical sources. More specifically, we review the (national) hazard analyses and their application for coastal planning, as well as the two operating tsunami warning systems: the national warning system for seismically generated tsunamis (SiAM), whose upstream component—the CAT-INGV—is also a Tsunami Service Provider of the North-eastern Atlantic, the Mediterranean and connected seas Tsunami Warning System (NEAMTWS) coordinated by the Intergovernmental Coordination Group established by the Intergovernmental Oceanographic Commission (IOC) of UNESCO, and the local warning system for tsunamis generated by volcanic slides along the Sciara del Fuoco of Stromboli volcano. Finally, we review the state of knowledge about other potential tsunami sources that may generate significant tsunamis for the Italian coasts, but that are not presently considered in existing tsunami warning systems. We investigate the 24–27 December 2018 eruption of Mount Etna occurred from fissures located on the volcano eastern flank and accompanied by a seismic swarm, which was triggered by the magma intrusion and continued for weeks after the end of the eruption. Moreover, this swarm involved some of the shallow volcano‐tectonic structures located on the Mount Etna flanks and culminated on 26 December with the strongest event (ML 4.8), occurred along the Fiandaca Fault. In this work, we analyze seismological data and Sentinel‐1 Differential Interferometric Synthetic Aperture Radar (DInSAR) measurements, the latter inverted through analytical modeling. Our results suggest that a dike source intruded, promoting the opening of the eruptive fissures fed by a shallower dike. Moreover, our findings indicate that the activation of faults in different sectors of the volcano may be considered as a response to accommodate the deformations induced by the magma volumes injection. This work represents a detailed methodological description of automated stereology dedicated to all brainstem catecholamine nuclei. Each tyrosine-hydroxylase-containing nucleus was analyzed to count the following features: i) nuclear volume; ii) neuron number per nucleus; iii) neuron area per each nucleus. A number of reports described catecholamine-containing neurons within brainstem of a variety of animal species. In a recently published work, we reported a simultaneous quantitative analysis of tyrosine hydroxylase-positive neurons in the whole brainstem. Here we report the detailed step by step stereological procedure which allowed to perform a morphometric assessment of each catecholamine nucleus. This protocol provides the method chance to analyze simultaneously various morphological features in the same experimental setting to avoid variability when single nuclei are analyzed in different experiments. This improves the reliability of comparisons between brainstem catecholamine nuclei within the reticular formation to increase our insight about the key functional roles played by these cells in the mammalian brain. In fact, despite being a discrete number of neurons scattered in a small brain area, these cells provide remarkable axonal collateralization which allows the modulation of neuronal activity in the entire CNS. The step by step description of brainstem stereology provided here is reported in order to share these methods and enhance quantitative studies about these fascinating nuclei. Abstract: The causative source of the first damaging earthquake instrumentally recorded in the Island of Ischia, occurred on 21 August 2017, has been studied through a multiparametric geophysical approach. In order to investigate the source geometry and kinematics we exploit seismological, Global Positioning System, and Sentinel‐1 and COSMO‐SkyMed differential interferometric synthetic aperture radar coseismic measurements. Our results indicate that the retrieved solutions from the geodetic data modeling and the seismological data are plausible; in particular, the best fit solution consists of an E‐W striking, south dipping normal fault, with its center located at a depth of 800 m. Moreover, the retrieved causative fault is consistent with the rheological stratification of the crust in this zone. This study allows us to improve the knowledge of the volcano‐tectonic processes occurring on the Island, which is crucial for a better assessment of the seismic risk in the area. Contents Stallion semen storage for artificial insemination is mainly based on liquid cooled storage. In many stallions this technique maintains sperm quality for an extended period of time (24-72 hr) at 7°C. While this technique is commonly used in the horse industry, there can be a decline in fertility in some stallions, due to an inability of their sperm to tolerate the cool storage process. The aim of the present work was to evaluate the effect of two natural antioxidants (epigallocatechin-3-gallate ( EGCG) at 20, 60 and 120 μ m and green tea polyphenols, and p at .001, .01 and .1 mg/ml) on some sperm parameters (sperm motility, viability/acrosome integrity and DNA quality) in extended semen immediately after its collection (T0) and after 2, 6, 24 and 48 hr of cool storage. Two ejaculates from three trotter stallions were analysed after 48 hr of storage at 4°C. No beneficial effect on the analysed parameters was observed: the two antioxidants were not able to improve sperm quality after 48 hr of storage. These results are in agreement with previous findings on the effect of different antioxidants reported by other researches, who have demonstrated that stallion semen keeps good antioxidant capacity after dilution for 24 hr. Contents In vitro embryo production in the horse is still not as efficient as in other species. Oxidative stress negatively affects oocyte and embryo culture. To attenuate/minimize the oxidative stress, antioxidants such as low-molecular thiol compounds can be added to culture media. Beta-mercaptoethanol (BME) has been shown to improve maturation and embryo development in different species. The aim of this study was to investigate whether the addition to maturation medium of BME at common (0.1 mM) and high (0.7 mM) concentration could improve oocyte maturation also in the horse. Equine oocytes recovered from slaughterhouse ovaries were used. Meiotic configuration after in vitro maturation (IVM) and early embryo production after intracytoplasmic sperm injection (ICSI) were considered as criteria for assessing nuclear and cytoplasmic maturation, respectively. A total of 1,076 oocytes were analysed over two experiments: 848 (control n = 293, BME 0.1 n = 270, BME 0.7 n = 285) were stained with Hoechst 33342 and examined for nuclear stage after 26 hr of IVM, and 228 MII oocytes were fertilized by ICSI (control n = 83, BME 0.1 n = 65, BME 0.7 n = 80). Cleavage rates were determined after 60 hr of culture. Unlike results obtained in other species, the addition of BME did not influence maturation rates (51.9% control vs 55.6% BME 0.1 mM and 55.1% BME 0.7 mM), nor cleavage rates after ICSI (38.6% vs 38.5% and 41.3%, respectively). In conclusion, the addition of BME at 0.1 and 0.7 mM to the maturation medium, in our culture conditions, has no effect on nuclear and cytoplasmic maturation of equine oocytes. Contents Stallion spermatozoa are highly dependent on oxidative phosphorylation for ATP production to achieve normal sperm function and to fuel the motility. The aim of this study was to evaluate the response of equine sperm under capacitating conditions to the inhibition of mitochondrial complex I by rotenone and to test whether epigallocatechin-3-gallate ( EGCG), a natural polyphenol component of green tea, could counteract this effect. After 2-h incubation of stallion spermatozoa in modified Tyrode's medium, rotenone (100 n m, 500 n m and 5 μ m) and EGCG (10, 20 and 60 μ m), alone or in combination, did not induce any significant difference on the percentage of viable cells, live sperm with active mitochondria and spermatozoa with intact acrosome. The inhibition of complex I of mitochondrial respiratory chain of stallion sperm with rotenone exerted a negative effect on heterologous ZP binding ability. EGCG at the concentrations of 10 and 20 μ m (but not of 60 μ m) induced a significant increase in the number of sperm bound to the ZP compared with that for control. Moreover, when stallion sperm were treated with rotenone 100 n m, the presence of EGCG at all the concentrations tested (10, 20 and 60 μ m) significantly increased the number of sperm bound to the ZP up to control levels, suggesting that this green tea polyphenol is able to reduce the toxicity of rotenone. Contents In the last 30 years, encapsulation technology has been applied to different species to minimize the loss of spermatozoa after artificial insemination. In particular, the vehiculation of boar sperm cells in barium alginate membrane has proved a valid strategy to reduce the risk of polyspermy and optimize in vivo fertilizing yields. Controlled release of male gametes into the female genital tract has reduced the minimum fertilizing dose of spermatozoa. Notwithstanding these results, encapsulation has not yet reached commercial application, largely due to the additional costs of production. However, encapsulation could be useful in advanced reproductive technology, such as sex sorting, to store sorted boar semen. The controlled release of flow cytometrically sorted spermatozoa could be a promising strategy to reduce the number of cells necessary for each insemination and hence allow the widescale use of sex sorting in this species. In the last two decades, poly(hydroxyalkanoates) (PHA) were solely produced using heterotrophic bacteria in aerobic cultivation. With respect to the great potential (500 Mt yr-1) of raw industrial CO2 streams and even greater potential of flue gases, the focus on photo-autotrophic biotechnological processes is increasing steadily. Primarily, PHA-gene transfer from heterotrophic bacteria into algae and plant cells was attempted, with the intention to combine the known biosynthesis pathway with autotrophic cultivation. The natural occurrence of PHA in cyanobacteria is known at least since 1966. However, cyanobacteria were never considered for commercial production because the PHA amount based on cell mass and based on volumetric productivity is generally very low. Therefore, strain improvements were suggested, either by gene amplification or by suppression of biochemical pathways competing for the cell's acetate pool. In the late 1990s, the success of genetic modification was confirmed experimentally, elevating the cyanobacteria cell's PHA content. With additional optimization, PHB amounts up to 50% w/w of biomass dry matter or up to about 2.4 g L-1 bioreactor volume could be produced within 11 days. Considering the land use for agriculture and the competition for plant biomass between food, feed, fuel and energy production, the binding of CO2 in a biotechnological process using photo-autotrophic microorganisms may become a promising option. Alkaline phosphatase ( AP) catalyses the detachment of phosphate residues from different substrates. Its activity has been demonstrated in seminal plasma and spermatozoa from porcine and other mammalian species; anyway, the role of AP in male reproduction has not been clarified yet and the aim of this study was to determine AP function in boar sperm capacitation and in vitro fertilization ( IVF). AP activity was assayed in seminal plasma and in uncapacitated and in vitro capacitated ( IVC) spermatozoa; in addition, capacitation was studied in presence of different doses of AP (1.2 and 2.5 IU/mL). The effect of different doses of AP (1.2 and 2.5 IU/mL) on several sperm parameters after IVC (viability, acrosome integrity with FITC- PSA, capacitation status with CTC staining, tyrosine phosphorylation) and on fertilizing ability during IVF were also evaluated. High AP activity was detected in seminal plasma, in particular in sperm-rich fraction; a lower activity was detected in uncapacitated spermatozoa while a significant decrease was evidenced after IVC. Viability was not changed by AP supplementation of the capacitating medium, whereas acrosome integrity and capacitation status were significantly affected by 1.2 and 2.5 doses, with a dose-dependent decrease in acrosome-reacted cells as well as in CTC B pattern displaying cells. As for sperm head protein phosphorylation, a decrease in relative fluorescence was detected in AP 2.5 group, if compared with capacitated one. After IVF, a dose-dependent decrease in penetrated oocytes was recorded, with an increase in monospermic zygote rate. In conclusion, we demonstrated that AP activity decreases under capacitating condition and that addition of AP to spermatozoa during capacitation results in a depression of the capacitating process and IVF. We can infer that AP plays a role in keeping spermatozoa quiescent until they are ejaculated and in modulating the acquisition of the fertilizing ability. [ABSTRACT FROM AUTHOR]

Over the past few years the assessment of the earthquake potential of large continental faults has increasingly relied on field investigations. State-of-the-art seismic hazard models are progressively complementing the information derived from earthquake cat- alogues with geological observations of active faulting. Using these observations, however, requires full understanding of the relationships between seismogenic slip at depth and surface deformation, such that the evidence indicating the presence of a large, potentially seismogenic fault can be singled out effectively and unambiguously. We used observations and models of the 6 April 2009, Mw 6.3, L'Aquila, normal faulting earthquake to explore the relationships between the activity of a large fault at seismogenic depth and its surface evidence. This very well-documented earthquake is representative of mid-size yet damaging earthquakes that are frequent around the Mediterranean Basin, and is somehow paradigmatic of the nature of the associated geologic evidence along with observational difficulties and ambiguities. Thanks to available high-resolution geologic, geodetic and seismological data aided by analogue modeling, we reconstructed the full geometry of the seismogenic source in relation with surface and sub-surface faults.We find that the earthquake was caused by seismogenic slip in the range 3-10 km depth, and that the slip distribution was strongly controlled by inherited discontinuities. We also contend that faulting was expressed at the surface by pseudo-primary breaks resulting from coseismic crustal bending and by sympathetic slip on secondary faults. Based on our results we propose a scheme for hierarchizing normal faults through which all surface occurrences related to faulting at depth can be interpreted in the frame of a single, mechanically coherent model. In the present study, acetylation status of histone H4 and methylation status of the lysine 9 residue of histone H3 (H3K9) were assessed by immunofluorescence in order to determine the effect of vitrification on epigenetic status of pig MII oocytes. Hyperacetylation of H4 and dimethylation of H3K9 were assessed in control oocytes, after cryoprotectant treatment and after vitrification at two time points, immediately after warming and after a post-warming incubation for 2 h. While no changes in the immunopositivity for both the epitopes were recorded after cryoprotectants, the percentage of negative oocytes for dimethyl H3K9 was observed to increase immediately after devitrification. The influence of vitrification was more evident after 2 h post-thaw incubation when acetylation status of H4 significantly increased and a rise in the percentages of both oocytes exhibiting strong positivity and negative oocytes for dimethyl H3K9 was observed. In conclusion, acetylation of H4 and methylation of H3K9 are altered by vitrification procedure that may lead to an aberrant epigenetic presentation of female chromatin to the fertilizing event and may be, at least in part, responsible for the reduction of developmental competence of vitrified pig oocytes. We study the miniaturization of Thermal Lens Spectrometry (TLS) towards Lab-on-chip integration in order to reduce the volume of fluid assays in nuclear process control. TLS is of great interest in this context since it combines the advantages of optical detection methods with an inherent suitability for small-scale samples. After validating the experimental principle in a classical thermal lens crossed-beam setup, we show the integration of a Young-interferometer with a microcapillary on a glass substrate, reducing the necessary sample size to 400 nl. The interferometer translates the photothermally induced refractive index change in the fluid to a phase shift of the fringe pattern, which can then be detected by a camera. Measurements of Co(II) in ethanol yield a detection limit of c = 5 \times 10^-4 M for the crossed-beam setup and c = 6 \times 10^-3 M for the integrated sensor. At an interaction length of 10 \mum, it detects a minimum absorbance of K = 1.2 \times 10^-4 in a probed volume of 14 pl. [ABSTRACT FROM AUTHOR]

Contents Little information is available on the quality of stallion spermatozoa after sex sorting. The objectives of the present study were to assess the quality of sex-sorted stallion spermatozoa and determine its fertilizing ability after hysteroscopic low dose insemination. Ejaculates from four stallions were collected and sorted by a MoFlo SX® flow cytometer/sperm sorter. Before and after sorting, spermatozoa were evaluated for motility by Computer Assisted Sperm Analysis, viability (SYBR 14-propidium iodide), mitochondrial function (JC-1) and acrosomal status (fluorescein isothiocyanate Pisum sativum agglutinin conjugated). A fertility trial was carried out on four mares (seven oestrous cycles) by hysteroscopic insemination, depositing 5 × 106 X-bearing spermatozoa. Sex sorting resulted in a significant decrease (p < 0.001) in all motility characteristics. Sperm viability and percentage of spermatozoa with functional mitochondria were not affected by the sorting process, while the percentage of reacted spermatozoa was higher (p < 0.01) for non-sorted than sorted spermatozoa. Pregnancy rate was 28.6% (2/7) after low dose hysteroscopic insemination. Only one pregnancy was carried to term with the birth of a healthy filly. In conclusion, despite the reduction in sperm motility, sex sorting did not impair stallion sperm viability and mitochondrial activity immediately post-thaw; moreover, the sexed spermatozoa retained the ability to fertilize in vivo. Contents Spermatozoa, as other eukaryotic cells, need hexoses to produce energy to maintain membrane homeostasis, to move along the female genital tract and to carry the male genome to the female gamete. GLUTs are a family of proteins that permit and improve the passive transport of hexoses inside cells. This study was aimed at investigating the presence and localization of GLUTs 1, 2, 3 and 5 in boar, stallion and dog spermatozoa by both immunofluorescence and western blotting. GLUTs exhibited a peculiar distribution along the sperm cell depending on the isoforms considered, the hexose they transport and the different species. The localization of GLUTs after capacitation and acrosome reaction highlighted the possible changes in their distribution because of the different functional moment. Only in dog spermatozoa changes in GLUTs distribution were demonstrated; these changes could be related to the different metabolic needs and modifications occurring in the sperm cell. We present a marine palaeoseismology analysis of a dense network of very high resolution seismic profiles along the Gondola Fault Zone (GFZ), a right-lateral, E–W-striking, active fault system in the Adriatic foreland. This case-study aims to show how time and space variations in the activity of a dominantly right-lateral fault system can be assessed using the vertical component of slip. The GFZ has been investigated for a length of 50 km. It includes two parallel subvertical fault sets and two main anticlines. The late Middle Pleistocene to Holocene vertical component of displacement along the fault is bell-shaped, suggesting that in the long-term the fault zone acts as a single, kinematically coherent structure. Slip rates are 0–0.18 mm a−1 and vary temporally on individual segments. This variability is consistent with a model in which individual fault segments rupture independently during earthquakes with magnitudes up to 6.4 and 1.3–1.8 ka recurrence intervals. Highly virulent strains of Trypanosoma cruzi are frequently used as murine models of Chagas’ disease. However, these strains do not fully represent the spectrum of parasites involved in the human infection. In this paper, we analysed parasitaemia, mortality, tissue pathology and parasite-specific IgG serum levels in immune-deficient mice infected with Sylvio X10/4 parasites, a T. cruzi derived from a chagasic patient that yields very low parasitaemias and in C3H/HePAS mice induces a chronic cardiopathy resembling the human disease. IFN- γ was identified as a crucial element for parasite control as its absence determined a drastic increase in parasitaemia, tissue parasitism, leukocyte infiltrates at the heart and striated muscles and mortality. The lack of IFN- γ or IL-12p40, a molecule shared by IL-12 and IL-23, also resulted in spinal cord lesions and a progressive paralysis syndrome. Whereas IgG2a was the main Ig isotype in infected C57BL/6 mice, IL-12p40-KO mice produced IgG2a and IgG1 and IFN- γ-KO mice produced only IgG1. The IFN- γ-protective effect was not essentially mediated by nitric oxide (NO), inasmuch as infected iNOS-KO mice showed no parasitaemia and low tissue damage. Mice deficient in CD4+ or CD8+ T cells showed an intermediate phenotype with increased mortality and tissue pathology but no parasitaemia. Interestingly, CD28-KO mice were unable to produce anti- T. cruzi IgG antibodies but presented moderate tissue pathology and managed to control the infection. Thus, differently from infections with high virulence parasites, neither IgG, NO nor CD28-mediated signalling are essential for the non-sterile control of Sylvio X10/4 parasites. We report on the realization of high-quality silica optical microcavities on a chip. We show that a precise control of radii of both silica disk and silicon pillar defines the shape of molten microresonators from microtoroids to microspheres. These last ones advantageously combine small mode volumes in structures of very small radii (5 < r < 14 μm) which are fully compatible to wafer-based processing. The optical quality-factor of silica microsphere on a chip is as high as 7.5 × 107 at 1.55 μm. The losses sources in these microspheres are detailed and we show that measured quality-factors are limited by the radiative losses for the smallest ones. [ABSTRACT FROM AUTHOR]

In this letter, the fabrication and characterization of a broadband 980-nm/1550-nm wavelength (de)multiplexer is described. This device is adapted for pump/signal multiplexing in erbium-doped waveguide amplifiers. The structure chosen is a segmented asymmetric Y-junction, which can be fabricated with one masking level using silver-sodium ion-exchange on a silicate glass substrate. The nonresonant behavior of the Y-junction allows a wideband multiplexing with isolations increasing from (9.7plusmn0.1) dB at 1500 nm to (15plusmn0.1) dB at 1630 nm and (26plusmn1) dB at 980 nm. The investigation of the neuronal network in mouse spinal cord models represents the basis for the research on neurodegenerative diseases. In this framework, the quantitative analysis of the single elements in different districts is a crucial task. However, conventional 3D imaging techniques do not have enough spatial resolution and contrast to allow for a quantitative investigation of the neuronal network. Exploiting the high coherence and the high flux of synchrotron sources, X-ray Phase-Contrast multiscale-Tomography allows for the 3D investigation of the neuronal microanatomy without any aggressive sample preparation or sectioning. We investigated healthy-mouse neuronal architecture by imaging the 3D distribution of the neuronal-network with a spatial resolution of 640 nm. The high quality of the obtained images enables a quantitative study of the neuronal structure on a subject-by-subject basis. We developed and applied a spatial statistical analysis on the motor neurons to obtain quantitative information on their 3D arrangement in the healthy-mice spinal cord. Then, we compared the obtained results with a mouse model of multiple sclerosis. Our approach paves the way to the creation of a 'database' for the characterization of the neuronal network main features for a comparative investigation of neurodegenerative diseases and therapies. The realisation and characterisation of periodic segmented waveguides made by Ag+/Na+ ion-exchange on glass is presented. In particular, the averaging effects provided by the segmentation are analysed using the measurement of the cutoff wavelengths of the fundamental mode. Numerical calculations are used to evaluate the equivalent index contrast of a continuous waveguide from cutoff wavelength. It is found that the model predicting the index averaging effect of periodically segmented waveguides is valid if the duty cycle of the segmented waveguide is corrected by a term that takes into account the longitudinal ion diffusion.
