Background: The complete mitochondrial respiratory chain is a precondition for maintaining cellular energy supply, development, and metabolic balance. Due to the evolutionary differentiation of complexes and the semi-autonomy of mitochondria, respiratory chain subunits have become critical targets for crop improvement and fungal control. In fungi, mitochondrial complex I mediates growth and metabolism. However, the role of this complex in the pathogenesis of phytopathogenic fungi is largely unknown. Results: In this study, we identified the NADH: ubiquinone oxidoreductase 24-kDa subunit (VdNuo1) of complex in vascular wilt pathogen, Verticillium dahliae, and examined its functional conservation in phytopathogenic fungi. Based on the treatments with respiratory chain inhibitors, the mitochondria-localized VdNuo1 was confirmed to regulate mitochondrial morphogenesis and homeostasis. VdNuo1 was induced during the different developmental stages in V. dahliae, including hyphal growth, conidiation, and melanized microsclerotia development. The VdNuo1 mutants displayed variable sensitivity to stress factors and decreased pathogenicity in multiple hosts, indicating that VdNuo1 is necessary in stress tolerance and full virulence. Comparative transcriptome analysis demonstrated that VdNuo1 mediates global transcriptional effects, including oxidation and reduction processes, fatty acid, sugar, and energy metabolism. These defects are partly attributed to impairments of mitochondrial morphological integrity, complex assembly, and related functions. Its homologue (CgNuo1) functions in the vegetative growth, melanin biosynthesis, and pathogenicity of Colletotrichum gloeosporioides; however, CgNuo1 does not restore the VdNuo1 mutant to normal phenotypes. Conclusions: Our results revealed that VdNuo1 plays important roles in growth, metabolism, microsclerotia development, stress tolerance, and virulence of V. dahliae, sharing novel insight into the function of complex I and a potential fungicide target for pathogenic fungi. Constructing highly efficient catalysts for the degradation of organic pollutants driven by solar light in aquatic environments is a promising and green strategy. In this study, a novel hexagonal sheet-like Pt/SnS2 heterojunction photocatalyst is successfully designed and fabricated using a hydrothermal method and photodeposition process for photocatalytic tetracycline (TC) degradation. The optimal Pt/SnS2 hybrid behaves with excellent photocatalytic performance, with a degradation efficiency of 91.27% after 120 min, a reaction rate constant of 0.0187 min−1, and durability, which can be attributed to (i) the formation of a metal/semiconductor interface field caused by loading Pt nanoparticles (NPs) on the surface of SnS2, facilitating the separation of photo-induced charge carriers; (ii) the local surface plasmon resonance (LSPR) effect of Pt NPs, extending the light absorption range; and (iii) the sheet-like structure of SnS2, which can shorten the transmission distance of charge carriers, thereby allowing more electrons (e−) and holes (h+) to transfer to the surface of the catalyst. This work provides new insights with the utilization of sheet-like structured materials for highly active photocatalytic TC degradation in wastewater treatment and environmental remediation. Verticillium dahliae is a soil-borne fungal pathogen that can cause severe vascular wilt in many plant species. Kelch repeat proteins are essential for fungal growth, resistance, and virulence. However, the function of the Kelch repeat protein family in V. dahliae is unclear. In this study, a Kelch repeat domain-containing protein DK185_4252 (VdLs.17 VDAG_08647) included in the conserved VdPKS9 gene cluster was identified and named VdKeR1. Phylogenetic analysis demonstrated a high degree of evolutionary conservation of VdKeR1 and its homologs among fungi. The experimental results showed that the absence of VdKeR1 impaired vegetative growth, microsclerotia development, and pathogenicity of V. dahliae. Osmotic and cell wall stress analyses suggested that VdKeR1-deleted mutants were more tolerant to NaCl, sorbitol, CR, and CFW, while more sensitive to H2O2 and SDS. In addition, analyses of the relative expression level of sqe and the content of squalene and ergosterol showed that VdKeR1 mediates the synthesis of squalene and ergosterol by positively regulating the activity of squalene epoxidase. In conclusion, these results indicated that VdKeR1 was involved in the growth, stress resistance, pathogenicity, and ergosterol metabolism of V. dahliae. The quality of the queen is crucial for the survival and growth of honey bee colonies. There have been studies related to raising queens with excellent performance since the invention of artificial queen rearing technologies. However, relatively few studies have been undertaken that evaluate the relationship between queen quality and queen morphology and the expression level of reproduction-related genes using different-aged brood in artificial queen rearing practices of Eastern honey bees (Apis cerana). Therefore, we transferred eggs, 1-day-old larvae, 2-day-old larvae, and 3-day-old larvae to queen cells to rear queens until the queen emerged. We measured 12 indicators (the acceptance rate, birth weight, length, caliber and maximum diameter of queen cells, thorax length, width and weight, head width, wing length and width, and number of ovarian tubes). Moreover, the relative expression levels of Hexamerin, Vitellogenin, and Transferrin were measured using fat bodies and ovaries dissected from the newly emerged virgin queens. The results showed that queens reared from eggs exhibited the best morphological indexes and occupied the most abundant reproductive related gene expression level. Parameter values for queen from 1-day-old larvae were significantly higher than those for queens from 2-day-old larvae and 3-day-old larvae, and there was no significant difference between values for queens from 2-day-old larvae and 3-day-old larvae in most data, except for the relative gene expression. In conclusion, as the age of the brood increased, the quality of the queen bees declined. Rearing queens from fertilized eggs are far more likely to yield a better outcome for queen performance and colony function. This study provides data support for raising high-quality queens of Apis cerana, which would be benefit for the protection and better utilization of our native honey bee species. Although new-type energetic materials have been investigated extensively, there is a challenge on how to integrate energy density and mechanical properties of energetic materials simultaneously. Herein, a versatile approach was proposed to design energetic materials with high energy density, reactivity, and flexibility based on a bio-inspired strategy. By mimicking the "brick-and-mortar" structure within a natural nacre, the energetic film with alternative layers of polytetrafluoroethylene (PTFE) and boron (B) was successfully fabricated. The nacre-mimetic PTFE/B energetic film exhibited excellent reaction heat (4413.9 J·g-1) and bright combustion flame, which may originate from the exothermic reaction mechanism between fluorine (F) and B. Even more remarkably, such PTFE/B energetic film revealed prominent mechanical flexibility reported for the first time. These findings indicate that the nacre-mimetic strategy provides an effective route to engineer energetic materials with high energy density, reactivity, and flexibility. [ABSTRACT FROM AUTHOR]

Background: Neuroendocrine breast carcinoma (NECB) is a rare, special histologic type of breast cancer. There are some small sample studies on the clinical outcomes of NECB patients, which are worthy of further discussion. Methods: We conducted a retrospective case-control study of clinical characteristics and outcomes among patients with primary NECB versus invasive carcinoma of no special type (NST) between November 2004 and November 2017 in the Peking Union Medical College Hospital, Beijing. NST patients were strictly matched 1:4 during the same period based on the TNM stage. Statistical comparisons were performed to determine the differences in survival between NST and NECB patients and to identify clinical factors that correlate with prognosis. Results: A total of 121 participants affected by primary NECB were included in our analysis from November 2004 to November 2017. Elderly persons (>60 years of age) were more likely to have primary NECB than young persons (p=0.001). In addition, primary NECB patients had significantly higher odds of having tumors 2-5 cm (36.5%) and >5 cm (6.1%) in size than NST patients. Despite a significant difference in tumor size, the proportion of patients with lymph node metastases showed no difference between the two groups (p=0.021). In addition, the rate of patients with ER-negative tumors in the NECB group (4.2%) was significantly lower than that in the primary NST group (29.8%). Significant differences were noted in the PR-negative (13.3% versus 36.6%, P<0.001) and HER2-negative (90.5% versus 76.4%, P=0.001) expression statuses among these patients. Of 121 primary NECB patients, 11 (9.1%) experienced relapses during the follow-up period. We found that tumor size was an independent risk factor for relapse. For hormone receptors on tumor cells, ER-positive breast cancer patients had significantly lower odds of relapse than receptor-negative patients. Conclusions: Our data demonstrate no significant difference in mortality and relapse between the primary NECB and NST groups. The tumor size in the primary NECB group was significantly larger than that in the NST group. In addition, the absence of ER independently increased the relapse rate for breast carcinoma patients. A nonionic waterborne polyurethane(WPU) was synthesized by the self-emulsification method using polyether diol (N220), isophorone diisocyanate (IPDI), trimethylolpropane poly (ethylene glycol monomethyl ether) (N120), 1,4-butanediol (BDO) and trimethylolpropane (TMP) as the main materials. The effects of the NCO/OH ratio on the emulsion and film properties of NWPU were explored. The experimental results show that the NWPU prepared at an NCO/OH ratio of 1.1 has good emulsion stability and easy film formation, and the resultant film was elastic, soft, and transparent. The sample was used for wool finishing and the application performance was evaluated. When the NWPU dosage reached 40 g·L−1, the fabric area felt shrinkage rate reduced from 8.97% to 4.75%, the pilling rating raised from grade 2–3 to grade 4, and the whiteness value only decreased by 3.87%. Xanthoceras sorbifolium (yellowhorn) is a woody oil plant with super stress resistance and excellent oil characteristics. The yellowhorn oil can be used as biofuel and edible oil with high nutritional and medicinal value. However, genetic studies on yellowhorn are just in the beginning, and fundamental biological questions regarding its very long-chain fatty acid (VLCFA) biosynthesis pathway remain largely unknown. In this study, we reconstructed the VLCFA biosynthesis pathway and annotated 137 genes encoding relevant enzymes. We identified four oleosin genes that package triacylglycerols (TAGs) and are specifically expressed in fruits, likely playing key roles in yellowhorn oil production. Especially, by examining time-ordered gene co-expression network (TO-GCN) constructed from fruit and leaf developments, we identified key enzymatic genes and potential regulatory transcription factors involved in VLCFA synthesis. In fruits, we further inferred a hierarchical regulatory network with MYB-related (XS03G0296800) and B3 (XS02G0057600) transcription factors as top-tier regulators, providing clues into factors controlling carbon flux into fatty acids. Our results offer new insights into key genes and transcriptional regulators governing fatty acid production in yellowhorn, laying the foundation for efforts to optimize oil content and fatty acid composition. Moreover, the gene expression patterns and putative regulatory relationships identified here will inform metabolic engineering and molecular breeding approaches tailored to meet biofuel and bioproduct demands. To improve the flame retardancy and solid content of waterborne polyurethane (WPU), AOT, and AOD series of WPU were synthesized and characterized. Carboxylic acid/nonionic composite units were used as hydrophilic groups, and OP550/THPO and OP550/DOPO‐HQ combinations were used as flame retardant modifiers, respectively. The results revealed that the flame retardant‐modified WPU emulsions still had excellent dispersion, stability, and fluidity. The UL‐94 rating of modified WPU could reach V‐0, and the AOD series samples had better flame retardancy than the AOT series, where AO10D3‐WPU containing 3% DOPO‐HQ had an limiting oxygen index value up to 30.5%. Besides, the peak heat release rate and total heat release of AO10D3‐WPU were respectively decreased by 32% and 34%, compared with the control sample in the conical calorimetric test. The TGA, TG‐IR, and SEM‐EDS exhibited that the flame retardants functioned in the condensed phase by promoting carbon formation. The tensile strength of AOT series samples was excellent with a range of 15.84–27.47 MPa, while the elongation at break of AOD series samples was relatively high with a distribution of 850.8%–913.11%, which could be selected to suit the needs of different application scenarios. Introduction: Dupilumab is the first biologic agent used to clinically treat moderate and severe atopic dermatitis (AD) and is currently the only biologic agent used for this condition. Many studies have reported that moderateto-severe AD was significantly improved after dupilumab injection, although head/neck dermatitis occurred with itching, flushing, and scaling. Moreover, because all the symptoms occur after dupilumab treatment, they are called "dupilumab facial redness (DFR)". Aim: To retrospectively analyse the clinical characteristics and treatment of facial erythema in patients with atopic dermatitis treated with dupilumab. Material and methods: The clinical data of patients with moderate-to-severe atopic dermatitis treated with dupilumab (600 mg for the first time, 300 mg every 2 weeks thereafter) in the department of dermatology from July 2020 to May 2022 were obtained. We described their characteristics and analysed their symptomatic treatment measures and efficacy. Results: Twenty-one patients with DFR were included. Most clinical manifestations were erythema and pruritus, which differed from the symptoms of typical moderate-to-severe AD. After treatment, drug withdrawal, and dressing change, the symptoms of 17 patients were effectively controlled or completely improved, while these of 4 did not improve. Conclusions: Although the mechanism of DFR is still unclear, symptomatic treatment is partially effective, and medication discontinuation and switching to Janus kinase inhibitors are acceptable for some patients. As the demand for cultural and recreational services grows, the mismatch between the supply and demand of park services significantly affects residents' well-being. Optimizing the spatial layout of park services is a focal point of urban park and green space research. Taking Hangzhou, Zhejiang Province, as a case study, this research analyzes the spatial patterns and balance of park service supply and demand. Utilizing the Grey Wolf Optimization Model optimized by the K-Nearest Neighbor Model (GWO-KNN), this study proposes construction objectives for optimizing park services. The results indicate the following: (1) significant differences exist in the park service demands of residents in different residential environments; (2) there is a noticeable spatial disparity in park service supply among various residential areas with an overall positive correlation between park service supply levels and resident demands, yet an imbalance exists; (3) this study categorizes spatial types into low-service coordination, high-service coordination, low-service imbalance, and high-service imbalance; (4) the GWO-KNN Model is applied with optimization objectives being the innovative aspect of this study. Strategies for each park category are proposed: emphasizing suburban park construction by utilizing surrounding green resources and adding diverse facilities; introducing facilities friendly to vulnerable groups to meet the needs of diverse populations; enhancing the complementary advantages between "new" and "old" cities by moderately increasing park sizes and improving cultural and facility development levels; optimizing spatial structure with limited land resources to construct an urban park network system. Background: Melanin plays important roles in morphological development, survival, host–pathogen interactions and in the virulence of phytopathogenic fungi. In Verticillum dahliae, increases in melanin are recognized as markers of maturation of microsclerotia which ensures the long-term survival and stress tolerance, while decreases in melanin are correlated with increased hyphal growth in the host. The conserved upstream components of the VdCmr1-regulated pathway controlling melanin production in V. dahliae have been extensively identified, but the direct activators of this pathway are still unclear. Results: We identified two genes encoding conserved C2H2-type zinc finger proteins VdZFP1 and VdZFP2 adjacent to VdPKS9, a gene encoding a negative regulator of both melanin biosynthesis and microsclerotia formation in V. dahliae. Both VdZFP1 and VdZFP2 were induced during microsclerotia development and were involved in melanin deposition. Their localization changed from cytoplasmic to nuclear in response to osmotic pressure. VdZFP1 and VdZFP2 act as modulators of microsclerotia melanization in V. dahliae, as confirmed by melanin biosynthesis inhibition and supplementation with the melanin pathway intermediate scytalone in albino strains. The results indicate that VdZFP1 and VdZFP2 participate in melanin biosynthesis by positively regulating VdCmr1. Based on the results obtained with yeast one- and two-hybrid (Y1H and Y2H) and bimolecular fluorescence complementation (BiFC) systems, we determined the melanin biosynthesis relies on the direct interactions among VdZFP1, VdZFP2 and VdCmr1, and these interactions occur on the cell walls of microsclerotia. Additionally, VdZFP1 and/or VdZFP2 mutants displayed increased sensitivity to stress factors rather than alterations in pathogenicity, reflecting the importance of melanin in stress tolerance of V. dahliae. Conclusions: Our results revealed that VdZFP1 and VdZFP2 positively regulate VdCmr1 to promote melanin deposition during microsclerotia development, providing novel insight into the regulation of melanin biosynthesis in V. dahliae. Terpenes and terpenoids are key natural compounds for plant defense, development, and composition of plant oil. The synthesis and accumulation of a myriad of volatile terpenoid compounds in these plants may dramatically alter the quality and flavor of the oils, which provide great commercial utilization value for oil-producing plants. Terpene synthases (TPSs) are important enzymes responsible for terpenic diversity. Investigating the differentiation of the TPS gene family could provide valuable theoretical support for the genetic improvement of oil-producing plants. While the origin and function of TPS genes have been extensively studied, the exact origin of the initial gene fusion event - it occurred in plants or microbes - remains uncertain. Furthermore, a comprehensive exploration of the TPS gene differentiation is still pending. Here, phylogenetic analysis revealed that the fusion of the TPS gene likely occurred in the ancestor of land plants, following the acquisition of individual C- and Nterminal domains. Potential mutual transfer of TPS genes was observed among microbes and plants. Gene synteny analysis disclosed a differential divergence pattern between TPS-c and TPS-e/f subfamilies involved in primary metabolism and those (TPS-a/b/d/g/h subfamilies) crucial for secondary metabolites. Biosynthetic gene clusters (BGCs) analysis suggested a correlation between lineage divergence and potential natural selection in structuring terpene diversities. Recently, an emerging category green of energetic material ammonium dinitramide (ADN) has exhibited promising application in propellants due to its outstanding merits in energy release and environmental friendliness. It can be considered to substitute traditional oxidizer of ammonium perchlorate (AP) in military systems and aerospace. In this paper, a novel spherical energetic composite ADN/copper alginate (CA) with a microporous structure was designed and prepared by the W/O gel emulsion method, and a desirable porous microsphere structure was obtained. Multiple characterization techniques were used to investigate the structure and properties of ADN/CA composites. The results showed that ADN crystals were homogeneously encapsulated in an alginate-gel matrix. Thermal decomposition temperature was reduced to 151.7 °C compared to ADN, while the activation energy of them was reduced from 129.73 kJ/ mol (ADN) to 107.50 kJ/mol (ADN/CA-4). In addition, as-prepared samples had lower impact and frictional sensitivity than ADN. The mechanism of sensitivity reduction and decomposition are also discussed. Constant-volume combustion tests show that peak pressure of the ADN/CA-4 achieves 253.4 kPa and pressurization rate of 2750.4 kPa/s. Background: Elderly patients with breast cancer are highly heterogeneous, and tumor load and comorbidities affect patient prognosis. Prediction models can help clinicians to implement tailored treatment plans for elderly patients with breast cancer. This study aimed to establish a prediction model for breast cancer, including comorbidities and tumor characteristics, in elderly patients with breast cancer. Methods: All patients were ≥65 years old and admitted to the Peking Union Medical College Hospital. The clinical and pathological characteristics, recurrence, and death were observed. Overall survival (OS) was analyzed using the Kaplan-Meier curve and a prediction model was constructed using Cox proportional hazards model regression. The discriminative ability and calibration of the nomograms for predicting OS were tested using concordance (C)- statistics and calibration plots. Clinical utility was demonstrated using decision curve analysis (DCA). Results: Based on 2,231 patients, the 5- and 10-year OS was 91.3% and 78.4%, respectively. We constructed an OS prediction nomogram for elderly patients with early breast cancer (PEEBC). The C-index for OS in PEEBC in the training and validation cohorts was 0.798 and 0.793, respectively. Calibration of the nomogram revealed a good predictive capability, as indicated by the calibration plot. DCA demonstrated that our model is clinically useful. As a typical energetic composite, polytetrafluoroethylene (PTFE)/aluminum (Al) has been widely applied in explosives, pyrotechnics, and propellants due to its ultra-high energy density and intense exothermic reaction. In this work, the radial gradient (RG) structure of PTFE/Al cylinders with three different PTFE morphologies (200 nm and 5 µm particles and 5 µm fiber) and content changes are prepared by 3D printing technology. The effect of radial gradient structure on the pressure output of PTFE/Al has been studied. Compared with the morphology change of PTFE, the change of component content in the gradient structure has an obvious effect on the pressure output of the PTFE/Al cylinder. Furthermore, the relationships of the morphology, content of PTFE and the combustion reaction of the PTFE/Al cylinder reveal that the cylinder shows a more complex flame propagation process than others. Against the backdrop of rapid urbanization, there is a passive adaptation state shown in urban and rural ecological spaces. Due to the shrinking of ecological patches and the fracture of corridors, problems such as the obstruction of ecological processes, the decline of ecosystem services, and the loss of biodiversity occur. Considering that county ecological space is the key level to undertake provincial ecological security patterns and implement ecological demonstration projects, the construction of a county ecological infrastructure (EI) network is beneficial to the protection of regional ecological security, the improvement of the structures and functions of farmland ecosystems, and the enhancement of the quality of human settlements. In this study, taking Langzhong County in Sichuan Province as an example, a method path for an EI network construction was explored, and an optimization strategy for ecological patterns was proposed. Firstly, morphological spatial pattern analysis (MSPA) and a patch importance index were employed to identify ecological sources. Secondly, by constructing a landscape resistance surface and adopting a minimum cumulative resistance (MCR) model, potential EI corridor paths were extracted. Thirdly, the interaction force values between ecological sources were calculated with a gravity model and important ecological corridors were identified for priority protection and restoration. Finally, an EI corridor network was optimized by combining network structure indexes (α, β, and γ) with the field situation, and stepping stone patches and ecological breakpoints were identified. Based on the analysis results, an ecological protection pattern, which involved three vertical and two horizontal ecological belts, four ecological control zones, and six clusters of EI networks in Langzhong County, was put forward, aiming at protecting 50 ecological sources, repairing 105 ecological corridors of different grades, adding 9 stepping stone patches near long-distance corridors and 15 at the intersection of ecological corridors, and repairing 18 ecological breakpoints. This study has guiding significance for the optimization of county ecological patterns, the construction of farmland forest belts, and site selection of ecological restoration projects. European hornbeam (Carpinus betulus L.) is widely planted in landscaping. In October 2021 and August 2022, leaf spot was observed on C. betulus in Xuzhou, Jiangsu Province, China. To identify the causal agent of anthracnose disease on C. betulus, 23 isolates were obtained from the symptomatic leaves. Based on ITS sequences and colony morphology, these isolates were divided into four Colletotrichum groups. Koch's postulates of four Colletotrichum species showed similar symptoms observed in the field. Combining the morphological characteristics and multi-gene phylogenetic analysis of the concatenated sequences of the internal transcribed spacer (ITS) gene, Apn2-Mat1-2 intergenic spacer (ApMat) gene, the calmodulin (CAL) gene, glyceraldehyde3-phosphate dehydrogenase (GAPDH) gene, Glutamine synthetase (GS) gene, and beta-tubulin 2 (TUB2) genes, the four Colletotrichum groups were identified as C. gloeosporioides, C. fructicola, C. aenigma, and C. siamense. This study is the first report of four Colletotrichum species causing leaf spot on European hornbeam in China, and it provides clear pathogen information for the further evaluation of the disease control strategies. Background: The high relative mortality rate in elderly breast cancer patients is most likely the result of comorbidities rather than the tumor load. Foregoing axillary lymph node dissection or omitting radiotherapy after breast-conserving surgery (BCS) does not affect the prognosis of elderly breast cancer patients. We sought to assess the safety of breast-conserving surgery without axillary lymph node dissection as well as breast and axillary radiotherapy (BCSNR) in elderly patients with early-stage breast cancer. Methods: We retrospectively included 541 consecutive breast cancer patients aged over 70 years with clinically negative axillary lymph nodes in one clinical center. Of these patients, 181 underwent mastectomy plus axillary lymph node dissection (MALND) with negative axillary cleaning and 360 underwent BCSNR. Results: After a median follow-up of 5 years, there was no significant difference between the BCSNR and MALND groups in either distant recurrence-free survival (DRFS) (p=0.990) or breast cancer-specific survival (p=0.076). Ipsilateral axillary disease was found in 11 (3.1%) patients in the BCSNR group and 3 (1.7%) patients in the MALND group; this difference was not significant (p=0.334). We did not observe a significant difference in distant recurrence between the groups (p=0.574), with 25 (6.9%) patients in the BCSNR group experiencing distant recurrence compared to 15 (8.3%) patients in the MALND group. Our findings did show a significant difference in ipsilateral breast cancer recurrence (IBTR), with 31 (8.6%) patients in the BCSNR group experiencing IBTR compared to only 2 (1.1%) patients in the MALND group (p=0.003). Wood decay resistance (WDR) is marking the value of wood utilization. Many trees of the Lauraceae have exceptional WDR, as evidenced by their use in ancient royal palace buildings in China. However, the genetics of WDR remain elusive. Here, through comparative genomics, we revealed the unique characteristics related to the high WDR in Lauraceae trees. We present a 1.27-Gb chromosome-level assembly for Lindera megaphylla (Lauraceae). Comparative genomics integrating major groups of angiosperm revealed Lauraceae species have extensively shared gene microsynteny associated with the biosynthesis of specialized metabolites such as isoquinoline alkaloids, flavonoid, lignins and terpenoid, which play significant roles in WDR. In Lauraceae genomes, tandem and proximal duplications (TD/PD) significantly expanded the coding space of key enzymes of biosynthesis pathways related to WDR, which may enhance the decay resistance of wood by increasing the accumulation of these compounds. Among Lauraceae species, genes of WDRrelated biosynthesis pathways showed remarkable expansion by TD/PD and conveyed unique and conserved motifs in their promoter and protein sequences, suggesting conserved gene collinearity, gene expansion and gene regulation supporting the high WDR. Owing to its capacity to eliminate a long-standing methodological limitation, fiber photometry can assist research gaining novel insight into neural systems. Fiber photometry can reveal artifact-free neural activity under deep brain stimulation (DBS). Although evoking neural potential with DBS is an effective method for mediating neural activity and neural function, the relationship between DBS-evoked neural Ca2+ change and DBS-evoked neural electrophysiology remains unknown. Therefore, in this study, a self-assembled optrode was demonstrated as a DBS stimulator and an optical biosensor capable of concurrently recording Ca2+ fluorescence and electrophysiological signals. Before the in vivo experiment, the volume of tissue activated (VTA) was estimated, and the simulated Ca2+ signals were presented using Monte Carlo (MC) simulation to approach the realistic in vivo environment. When VTA and the simulated Ca2+ signals were combined, the distribution of simulated Ca2+ fluorescence signals matched the VTA region. In addition, the in vivo experiment revealed a correlation between the local field potential (LFP) and the Ca2+ fluorescence signal in the evoked region, revealing the relationship between electrophysiology and the performance of neural Ca2+ concentration behavior. Concurrent with the VTA volume, simulated Ca2+ intensity, and the in vivo experiment, these data suggested that the behavior of neural electrophysiology was consistent with the phenomenon of Ca2+ influx to neurons. Pinus bungeana, an endangered and native coniferous tree species in China, has considerable timber and horticulture value. However, little is known about needle diseases in P. bungeana. A needle blight of P. bungeana has been observed in Hebei Province, China. P. bungeana inoculated with mycelial plugs of fungal isolates presented symptoms similar to those observed under field conditions. Ten virulent fungal isolates were identified as a small-spored Alternaria species based on morphological observations. Maximum likelihood and Bayesian phylogenetic analyses carried out with multilocus sequence typing of eight regions (SSU, LSU, ITS, gapdh, tef1, Alt a 1, endoPG, OPA10-2) assigned the pathogen to Alternaria alternata. Simple Summary: Honey bees collect pollen with different nutrients to meet their physiological metabolism needs and maintain colony development. The protein contents vary among bee pollens, and the nutritional level of pollen is mainly reflected in the amino acid composition of its proteins. In northern China, pear, rape, and apricot plants are monoculture plants grown in a large area, and their pollen is collected by honey bee colonies during the spring. However, honey bees showed a foraging preference for apricot or rape pollen. It is important to understand the differences in the amino acid levels of these three pollen species and the associated physiological metabolic responses in honey bees. In this study, we evaluated the differences in amino acid content and hemolymph metabolism in caged adult bees that were fed three different bee pollen types. The numbers and levels of essential amino acids in pear pollen were significantly higher than those in apricot and rape pollen. Furthermore, these three bee pollens induced different metabolic responses in adult honey bees of different ages. Overall, pear pollen had the highest nutritional value among the three bee pollens in terms of amino acid level, and bee pollen was shown to play an important role in the pathways related to amino acid and lipid metabolism in bees in early adulthood. Pollen is essential to the development of honey bees. The nutrients in bee pollen vary greatly among plant species. Here, we analyzed the differences in the amino acid compositions of pear (Pyrus bretschneideri), rape (Brassica napus), and apricot (Armeniaca sibirica) pollens and investigated the variation in hemolymph metabolites and metabolic pathways through untargeted metabolomics in caged adult bees at days 7 and 14. The results showed that the levels of five essential amino acids (isoleucine, phenylalanine, lysine, methionine, and histidine) were the highest in pear pollen, and the levels of four amino acids (isoleucine: 50.75 ± 1.93 mg/kg, phenylalanine: 87.25 ± 2.66 mg/kg, methionine: 16.00 ± 0.71 mg/kg and histidine: 647.50 ± 24.80 mg/kg) were significantly higher in pear pollen than in the other two kinds of bee pollen (p < 0.05). The number of metabolites in bee hemolymph on day 14 (615) was significantly lower than that on day 7 (1466). The key metabolic pathways of bees, namely, "sphingolipid metabolism (p = 0.0091)", "tryptophan metabolism (p = 0.0245)", and "cysteine and methionine metabolism (p = 0.0277)", were significantly affected on day 7. There was no meaningful pathway enrichment on day 14. In conclusion, pear pollen had higher nutritional value among the three bee pollens in terms of amino acid level, followed by rape and apricot pollen, and the difference in amino acid composition among bee pollens was reflected in the lipid and amino acid metabolism pathways of early adult honey bee hemolymph. Few incidents of ancient allopolyploidization (polyploidization by hybridization or merging diverged genomes) were previously revealed, although there is significant evidence for the accumulation of whole genome duplications (WGD) in plants. Here, we focused on Ericales, one of the largest and most diverse angiosperm orders with significant ornamental and economic value. Through integrating 24 high-quality whole genome data selected from ~ 200 Superasterids genomes/species and an algorithm of topology-based genetree reconciliation, we explored the evolutionary history of in Ericales with ancient complex. We unraveled the allopolyploid origin of Ericales and detected extensive lineage-specific gene loss following the polyploidization. Our study provided a new hypothesis regarding the origin of Ericales and revealed an instructive perspective of gene loss as a pervasive source of genetic variation and adaptive phenotypic diversity in Ericales. Background: Immunotherapy plays an important role in the treatment of triple-negative breast cancer (TNBC). This study aimed to identify immunerelated genes that are associated with the prognosis of patients with TNBC as possible targets of immunotherapy, alongside their related tumor-infiltrating lymphocytes (TILs). Methods: The clinical data and gene expression profiles of patients with breast cancer were extracted from The Cancer Genome Atlas (TCGA) and Gene Expression Omnibus (GEO) databases and divided into training (n = 1,053) and verification (n = 508) groups. CIBERSORT was used to predict the differences in immune cell infiltration in patient subsets that were stratified according to risk. Gene Ontology (GO) enrichment analysis was used to identify pathways associated with immune-related genes in patient subsets that were stratified according to risk. The clinical data and insulin-like growth factor 2 receptor (IGF2R) expression profiles of patients with breast cancer were extracted from METABRIC. The expression of IGF2R and TILs were evaluated in a cohort containing 282 untreated patients with TNBC. The correlations of IGF2R expression, TILs, and clinicopathological parameters with patient prognosis were analyzed in the whole cohort. Results: The prognostic model, which was composed of 26 immune-related gene pairs, significantly distinguished between high- and low-risk patients. Univariate and multivariate analyses indicated that the model was an independent prognostic factor for breast cancer. Among the identified genes, the expression of IGF2R significantly distinguished between high- and low-risk patients in TCGA (P = 0.008) and in METABRIC patients (P < 0.001). The expression of IGF2R was significantly associated with clinical risk factors such as TNBC, estrogen receptor (ER)-negative expression, human epidermal growth factor receptor 2 (HER2)-positive expression, and age =60 years old in METABRIC patients. In addition, the patients with IGF2R-positive expression had lower disease-free survival (DFS) rates than those with IGF2R-negative expression in the TNBC cohort (67.8% vs. 78.5%, P = 0.023). IGF2R expression also was significantly negatively correlated with TILs, particularly with CD8+ TILs and CD19+ TILs in the cohort of patients with TNBC. The pinewood nematode (PWN), Bursaphelenchus xylophilus, is a notorious parasitic nematode of pine trees that causes pine wilt disease (PWD), leading to extensive mortality of different pine species around the world and considerable economic losses, thus posing a threat to healthy pines worldwide. Fast and accurate detection technology is necessary for the management of PWD spread. This study describes the development of a new DNA extraction method and detection technology, enzyme-mediated amplification (EMA), using primers and a newly designed probe according to the rRNA internal transcribed spacer gene ITS2. The detection process can be completed within 40 min, including DNA extraction for 10 min and detection for 30 min, by exploiting the synergistic action of multiple enzymes. This method can detect PWNs from different geographic areas quickly and accurately at all life stages, singly or in a mixture, and can distinguish PWNs from other species of the Bursaphelenchus group, showing that it is not only reliable but also rapid, greatly improving the efficiency and speed of PWN detection. Oxidized sucrose is a non-formaldehyde crosslinking agent with many applications in polymer crosslinking and modification, such as in the preparation of starch films and protein films. However, research on the structure of oxidized sucrose is lacking. In this paper, oxidized sucrose was synthesized through selective oxidation of sodium periodate. By LC-MS, FTIR, TGA, NMR, and HRMS analyses, it was shown that oxidized sucrose existed in the form of a hydrate, and the tetraaldehyde oxidized sucrose could isomerize into the form of two six-membered hemiacetal rings. The structure of oxidized sucrose was also verified by theoretical calculations. Furthermore, the diffusional properties of oxidized sucrose were investigated by the rolling-film method. There are great challenges in the generation of higher-order modes for long period fiber grating (LPFG). The cross-coupling coefficient between the fundamental mode and the higher-order azimuthal modes is extremely small due to the difficulty of angular modulation with traditional methods. Therefore, how to generate higher-order orbital angular momentum (OAM) modes with a high efficiency and low insertion loss is an urgent problem to be solved. In this paper, an efficient all-fiber approach to generate fourth-order OAM mode by employing a preset twisted LPFG (PT-LPFG) in a few-mode fiber (FMF) is presented. The difficulty of generating fourth-order mode utilizing traditional single-side exposure carbon dioxide laser manufacturing method and the advantages of the PT-LPFG in enhancing the coupling coefficient are theoretically analyzed in detail. In experiment, the conversion from the fundamental mode to the fourth-order mode is achieved, and the fourth-order interference patterns demonstrated the successful generation of the OAM±4 modes. The conversion efficiency of the OAM modes is over 99% and the purity is measured to be more than 90%. Cyclocarya paliurus is an endemic Chinese tree species with considerable medicinal, timber, and horticultural value. The anthracnose disease of C. paliurus is caused by the fungal pathogen Colletotrichum fructicola , which results in great losses in yield and quality. Here, resistance evaluation of six cultivars of C. paliurus exhibited varying degrees of resistance to C. fructicola infection, where Wufeng was the most resistant and Jinggangshan was the most susceptive. Physiological measurements and histochemical staining assays showed that the Wufeng cultivar exhibits intense reactive oxygen species accumulation and defense capabilities. A multiomics approach using RNA sequencing and metabolome analyses showed that resistance in C. paliurus (Wufeng) is related to early induction of reprogramming of the flavonoid biosynthesis pathway. In vitro antifungal assays revealed that the flavonoid extracts from resistant cultivars strongly inhibited C. fructicola hyphal growth than susceptible cultivars. Relative gene expression analysis further demonstrated the pivotal antifungal role of C. paliurus flavonoids in targeting Colletotrichum appressorium formation. Together, these results represent a novel resistance mechanism of C. paliurus against anthracnose through the reprogramming of flavonoids, which will lay a foundation for breeding anthracnose-resistant varieties and the application of flavonoid extraction of C. paliurus as a natural antifungal treatment. Background: During the disease cycle, plant pathogenic fungi exhibit a morphological transition between hyphal growth (the phase of active infection) and the production of long-term survival structures that remain dormant during "overwintering." Verticillium dahliae is a major plant pathogen that produces heavily melanized microsclerotia (MS) that survive in the soil for 14 or more years. These MS are multicellular structures produced during the necrotrophic phase of the disease cycle. Polyketide synthases (PKSs) are responsible for catalyzing production of many secondary metabolites including melanin. While MS contribute to long-term survival, hyphal growth is key for infection and virulence, but the signaling mechanisms by which the pathogen maintains hyphal growth are unclear. Results: We analyzed the VdPKSs that contain at least one conserved domain potentially involved in secondary metabolism (SM), and screened the effect of VdPKS deletions in the virulent strain AT13. Among the five VdPKSs whose deletion affected virulence on cotton, we found that VdPKS9 acted epistatically to the VdPKS1-associated melanin pathway to promote hyphal growth. The decreased hyphal growth in VdPKS9 mutants was accompanied by the up-regulation of melanin biosynthesis and MS formation. Overexpression of VdPKS9 transformed melanized hyphal-type (MH-type) into the albinistic hyaline hyphal-type (AH-type), and VdPKS9 was upregulated in the AH-type population, which also exhibited higher virulence than the MH-type. Conclusions: We show that VdPKS9 is a powerful negative regulator of both melanin biosynthesis and MS formation in V. dahliae. These findings provide insight into the mechanism of how plant pathogens promote their virulence by the maintenance of vegetative hyphal growth during infection and colonization of plant hosts, and may provide novel targets for the control of melanin-producing filamentous fungi. Low temperature germination (LTG) is a key agronomic trait in rice (Oryza sativa L.). However, the genetic basis of natural variation for LTG is largely unknown. Here, a genome-wide association study (GWAS) was performed using 276 accessions from the 3,000 Rice Genomes (3K-RG) project with 497 k single nucleotide polymorphisms (SNPs) to uncover potential genes for LTG in rice. In total, 37 quantitative trait loci (QTLs) from the 6th day (D6) to the 10th day (D10) were detected in the full population, overlapping with 12 previously reported QTLs for LTG. One novel QTL, namely qLTG1-2, was found stably on D7 in both 2019 and 2020. Based on two germination-specific transcriptome datasets, 13 seedexpressed genes were isolated within a 200 kb interval of qLTG1-2. Combining with haplotype analysis, a functional uncharacterized gene, LOC_Os01g23580, and a seed germination-associated gene, LOC_Os01g23620 (OsSar1a), as promising candidate genes, both of which were significantly differentially expressed between high and low LTG accessions. Collectively, the candidate genes with favorable alleles may be useful for the future characterization of the LTG mechanism and the improvement of the LTG trait in rice breeding. Buddhist pine (Podocarpus macrophyllus) is a widely cultivated evergreen tree species with high ornamental and economic value. Recently, severe leaf blight on Buddhist pine was detected in Anhui Province, China. Seven single-spore isolates obtained from symptomatic samples were identified as Pestalotiopsis lushanensis on the basis of morphology and multilocus (internal transcribed spacer region, translation elongation factor 1-α and β-tubulin) phylogenetic analysis. Pathogenicity tests of potted Buddhist pine seedlings revealed that regardless of whether a conidial suspension or mycelial disk was used, inoculation of P. lushanensis could result in leaf blight symptoms similar to those observed in the field.. It is the basic characteristic of pure vortex light that there is a phase singularity at the origin. Such a singularity may be multiple degenerate, which determines the order of vortex light. Singularities splitting phenomenon means that singularities no longer concentrate at the origin but distribute around the space, usually occurring in impure vortex light. In this paper, we demonstrate the singularities splitting phenomenon and propose an analysis method, based on which one may rapidly estimate the modal components of impure vortex light. As two common singularity discrimination methods, the spiral and fork wire interference patterns are compared in distinguishing splitting singularities. The most widely used spiral interference pattern is revealed to be the worst form because of the low resolution. Instead, the fork wire interference pattern is with higher and easily adjusted resolution. 1‰ impurity is still able to be distinguished through fork wire interference patterns in the experiment. Verticillium dahliae is a destructive soil-borne pathogen of many economically important dicots. The genetics of pathogenesis in V. dahliae has been extensively studied. Spt-Ada-Gcn5 acetyltransferase complex (SAGA) is an ATP-independent multifunctional chromatin remodeling complex that contributes to diverse transcriptional regulatory functions. As members of the core module in the SAGA complex in Saccharomyces cerevisiae , Ada1, together with Spt7 and Spt20, play an important role in maintaining the integrity of the complex. In this study, we identified homologs of the SAGA complex in V. dahliae and found that deletion of the Ada1 subunit (VdAda1) causes severe defects in the formation of conidia and microsclerotia, and in melanin biosynthesis and virulence. The effect of VdAda1 on histone acetylation in V. dahliae was confirmed by western blot analysis. The deletion of VdAda1 resulted in genome-wide alteration of the V. dahliae transcriptome, including genes encoding transcription factors and secreted proteins, suggesting its prominent role in the regulation of transcription and virulence. Overall, we demonstrated that VdAda1, a member of the SAGA complex, modulates multiple physiological processes by regulating global gene expression that impinge on virulence and survival in V. dahliae. In this study, waste engine oil (WEO) was used as catalyst for co-pyrolysis with waste tires (WT) to increase the heat transfer efficiency between tire particles. The synergetic effects during co-pyrolysis process of WT and WEO were investigated via thermogravimetric analyzer. Co-pyrolysis of different mixing ratios blends were performed in a self-made tube furnace at 500℃ to explore the effect on product distribution. The compositions of products were determined by FTIR, Py-GC/MS, and BET. TG and DTG curves of mixture demonstrate that WEO could change the pyrolysis process of WT significantly. The main pyrolysis temperature is reduced by 50℃. The conversion of pyrolysis oil increased, and the conversion of char decreased with the addition of WEO. The yield of H2 and D-limonene increased by 5.6% and 1.85%, respectively, when 20% WEO was added. We investigated the effects of the transverse components of a tendon core suture on tensile resistance and strength of 4-strand repairs. Forty-four pig flexor tendons were repaired with one of the following four methods: double Tsuge, U-shaped, 4-strand cross and 4-strand rectangular repairs. We recorded the number of the repaired tendons that formed a 2 mm gap between the tendon ends during cyclic loading for 20 cycles, stiffness of the tendon at the 1st and 20th cycle, gap distance at the repair site and ultimate strength of the repair at the 20th cycle. When transverse components were added to the core suture, a greater number of tendons formed a 2 mm gap during cyclic loading. The stiffness gradually decreased, and the repair site's gap distance after cyclic loading increased with the presence of transverse components of the sutures. Background: Kinesio tape (KT) is an elastic therapeutic tape used for treating sports-related injuries and a number of other disorders. To date, the objective evidence to link pathophysiological effects and actual reactions triggered by KT is limited.Purpose: To explore the effect of KT on the lumbar paraspinal muscles by magnetic resonance (MR) elastography.Study Type: Prospective observational study.Population: Sixty-six asymptomatic volunteers with 31 women and 35 men.Field Strength/sequence: 3.0T MRI and elastography with vibration frequency of 120 Hz.Assessment: The 5-cm-width KT with full tension was placed on a single side of the lumbar paraspinal muscle. The taping side and adhering direction were randomly decided. Two rectangular regions of interest (ROIs) of 5- and 2.5-cm-width were positioned at the bilateral paraspinal regions from the L2 to L4 level on the confidence map of MR elastography before and after KT taping. The mean shear stiffness values of the ROIs at the superficial, middle, and deep depths were recorded; then the differences between the taping and reference sides were calculated.Statistical Tests: Paired t-test and Pearson correlations were used to evaluate the stiffness changes after KT application and intraoperator errors of the stiffness measures on the reference side, respectively.Results: A significant decrease in the muscle stiffness value between taping and reference sides (-0.71 kPa ± 0.60 with KT and -0.25 kPa ± 0.78 without KT, P < 0.0001 for 5-cm ROI; -0.67 kPa ± 1.12 with KT and -0.16 kPa ± 1.17 without KT, P = 0.0004 for 2.5-cm ROI) was found in the superficial depth, but no significant differences in the middle and deep depths (P = 0.25 and P = 0.79 for 5-cm ROI; P = 0.09 and P = 0.67 for 2.5-cm ROI, respectively). For many years, the number and losses of modes have restricted the large-scale application of hollow core photonic bandgap fibers (HC-PBFs) in data transmission. Researchers have successfully reduced fiber loss to an extremely low level; however, little attention has been given to the specific characteristics of the higher-order modes (HOMs) in HC-PBF. In this paper, the high-order mode characteristics of a 7-cell HC-PBF are investigated in detail, and the non-degenerate phenomenon of HE21 is observed. Through an in-depth theoretical analysis by discussing the influence of the structural deformation of the fiber near the core on the high-order mode characteristics, the cause of the phenomenon is described. Thus, an optimized 7-cell HC-PBF for supporting 6 modes is developed. Additionally, the bending losses of all the HOMs in the fiber are measured. Marked bending loss is not observed even when the bending radius is as small as 0.5 cm, which is different from a solid core four-mode fiber. Diabetic kidney disease (DKD) is a major complication of diabetes. Expression of members of the microRNA (miRNA) miR-379 cluster is increased in DKD. miR-379, the most upstream 5′-miRNA in the cluster, functions in endoplasmic reticulum (ER) stress by targeting EDEM3. However, the in vivo functions of miR-379 remain unclear. We created miR-379 knockout (KO) mice using CRISPR-Cas9 nickase and dual guide RNA technique and characterized their phenotype in diabetes. We screened for miR-379 targets in renal mesangial cells from WT vs. miR-379KO mice using AGO2-immunopreciptation and CLASH (cross-linking, ligation, sequencing hybrids) and identified the redox protein thioredoxin and mitochondrial fission-1 protein. miR-379KO mice were protected from features of DKD as well as body weight loss associated with mitochondrial dysfunction, ER- and oxidative stress. These results reveal a role for miR-379 in DKD and metabolic processes via reducing adaptive mitophagy. Strategies targeting miR-379 could offer therapeutic options for DKD.
