This paper discusses co-housing and its role as sustainable architecture to support various aspects of the life of communities and the environment. This research is important because the potential of co-housing settlements to improve the social and mental well-being of their residents has not been widely discussed. In addition, housing that uses sustainable technology tends to be very expensive, which, in the end, makes it more difficult for cultural communities to access settlements. For this reason, this study aims to demonstrate how the co-housing concept can address the issues mentioned earlier. To achieve this, a qualitative approach is employed, with the Miss Tjitjih (MT) community selected as the case study. The MT community was chosen because it has been living in co-housing concept housing for more than four decades. Open-ended questions were given to reveal opinions from the participant's side. The answers from the participants are then analyzed with the help of Nvivo. The results of the analysis are then grouped to obtain a code that explains the role of co-housing for this community. The resulting diagram reveals the role of co-housing as a settlement model that can answer various problems of cultural communities in the city. [ABSTRACT FROM AUTHOR]

The government of the Education Office of North Maluku Province is trying to overcome the problem of student distribution for schools in the city of Ternate, especially SMA/SMK/equivalent which aims to make all schools quality and there are no more superior schools. However, new problems arise related to the zoning system itself, sometimes some parents of prospective students and prospective students themselves do not know the schools in their domicile zone. The problem formulation is how to create and compile a Geographic Information System Mapping SMA and SMK in Ternate City. Designing a geographic information system that can provide information about web-based SMA and SMK Mapping in Ternate City and become a reference for the community in choosing their or their children's schools according to the zoning system. For testing the system is in accordance with the stages tested where for testing the system is used black box testing, namely testing to see the suitability of data accessed. The high school and vocational recession geographic information system was built using the PHP programming language, Mysql as a database, while for making locations of high school and vocational high school vulnerable places using google maps, maps API. All high school and vocational tracking information system uploads use black boxes, run as expected on the system upload carried out. The Information and Communication Technology Unit (ICTU) is a technical implementation unit in the field of information and communication technology development and management. Currently, Khairun University's ICTU is undertaking a software development project, one of which involves creating an application called "Permata". In the process of developing this application, one of the common challenges faced is estimating the project costs for software development, as the developers do not yet have a standard method to determine the costs of software development. The objective of this research is to produce a Use Case Point application for estimating the costs of software development at ICTU, Khairun University. The Use Case Point (UCP) method is derived from the Function Point Analysis (FPA) method and aims to provide a simple estimation method oriented toward software project objects. The software objects involved in this method are Actor Types (UAW), Use Case Weight (UUCW), Technical System Development Complexity (TF), and Development Environment Complexity (EF). The estimated time value for developing the Permata application is 1,389.844855 man-hours, equivalent to approximately 1.904 months. The estimated cost for the Permata application is Rp129,099,127. In the pursuit of technologically advanced governance, the Ministry ABC has integrated technology into daily operations, particularly through web applications like an employee administration information system. However, challenges persist in the implementation of online administration, especially in terms of technology adoption among the older generation. This study employs the Technology Acceptance Model (TAM) to investigate the usage of the administrative information system, focusing on System Quality (SQ), Perceived Ease of Use (EU), Perceived Usefulness (PU), and Behavioral Intention (BI). Surveying 147 respondents via questionnaires, the findings reveal that system quality positively influences perceived ease of use and perceived usefulness, while unexpectedly establishing a negative relationship between system quality, perceived ease of use, perceived usefulness, and behavioral intention. These insights hold significance for both theory and practice, reaffirming the crucial roles of perceived ease of use and perceived usefulness in shaping behavioral intentions and augmenting the TAM model. Background: There are thousands of species of known medicinal plants in the world. Ruta angustifolia L. has been widely used in traditional medication for jaundice and liver disease. Previous studies have shown that R. angustifolia leaves can inhibit the hepatitis C virus in Huhit culture cells, reduce the value of NS3 protein, and possess a synergistic effect in combination with antiviral drugs. Therefore, this plant is potential to be developed as a drug candidate. Characteristics of plants including microscopic, physicochemical properties, and phytochemical profiles are necessary information to ensure the quality of raw material in drug development. Objective: This study was carried out to examine the microscopic and physicochemical including the standardized parameter of R. angustifolia leaves to fulfil the quality raw materials as traditional medicine. Methods: Simplicia of R. angustifolia leaves obtained from Jombang, East Java, were observed under a microscope and determined its physicochemical properties referred to the Materia Medica Indonesia V. The TLC and HPLC profiles of extract were determined as well. Results: Microscopic analysis were conducted by transfection sections and the presence of epidermis cells, palisade, mesophyll with stomata, and Ca-oxalate crystal were found. The standard parameter obtained value of loss of drying, extractive value in water and ethanol, and ash value. The TLC and HPLC profiles of leaves extract demonstrated to contain with flavonoid, terpenoids, and alkaloids. Conclusion: Ruta angustifolia obtained from Jombang, east Java, has a specific character in microscopic analysis. Background: Hepatitis c virus infection is a global health problem which chronically infected 71 million people in the world. This infection has a risk of becoming liver cirrhosis and hepatocellular carcinoma. Since the current HCV therapy has been developed by direct-acting antivirals (DAA), however, most patients get limited access due to the high cost. Therefore, further development anti-HCV agent still greatly needed. Ruta angustifolia is a natural resource which was reported to possess anti-HCV activity. Ribavirin is an antiviral agent used to treat several virus infections, either DNA or RNA. Ribavirin was known to inhibit HCV infection by regulated immune system in host cells and interfering the replication of HCV by inhibit HCV RdRp. Objective: The current study evaluated the combination treatment of R. angustifolia extracts and ribavirin by in vitro culture cells of Huh 7it. Method: The study was conducted under an invitro cell culture of Huh 7it and infected with JFH1a. Result: The result demonstrated an enhancement effect of the extract by increasing the anti-HCV activity 3.5-fold higher compared to ribavirin alone. The 50% inhibitory concentration of ribavirin by single treatment was 10.43 ± 0.18 µg/mL, while in combination with Ruta angustifolia extract was 2.80 ± 0.03 µg/mL. Further analysis of the combination by CompuSyn software mediated a synergistic effect among the combination with a combination index value of 0.691. Conclusion: These results suggested that combination of Ribavirin and Ruta angustifolia should be considered in developing anti-hepatitis C virus agents. [ABSTRACT FROM AUTHOR]

Background: Medicinal plants are known to perform many pharmacological actions due to their chemical metabolites, which include antiviral effects. Previously, the extract of Ruta angustifolia was shown to have potential anti-hepatitis C virus (HCV) activity without any cytotoxicity, with a 50% inhibitory concentration of 3.0 μg/mL and a 50% cytotoxicity concentration of >100 μg/mL. Furthermore, the combination of medicinal plants and current anti-HCV agents, such as a direct-acting antiviral agent, was shown to potentiate their overall effectiveness. In the course of this study, the ethanolic extract of R. angustifolia was evaluated for its anti-HCV effects; specifically, the mechanism of action on HCV NS3 and NS5A protease was investigated. Methods: Analysis of the use of this extract in conjunction with current NS3 inhibitor drugs, simeprevir (SMV) and telaprevir (TVR), was performed. Anti-HCV activity was performed by in vitro culture of hepatocyte cells. The cells were infected and treated with various concentrations of the sample. HCV inhibition was calculated and CompuSyn software analysis was used to determine the synergistic effect of the combination. Results: Results demonstrated that R. angustifolia extract inhibited the post-entry step and decreased the protein levels of HCV NS3 and NS5A. The combination of extract and SMV and TVR mediated a synergistic effect. Conclusions: These findings suggest that combining R. angustifolia extract with current anti-HCV drugs should be considered when developing alternative and complementary anti-HCV medicines. "Megacity Skeleton" is a housing prototype project aimed at improving the urban living environment. The project site is located at Kampong Cikini, a high‐density informal settlement situated in the center of Jakarta Megacity. Although this kind of area is always at risk of slum clearance owing to its poor hygiene and environmental conditions, it has fostered attractive lifestyles with a low global environmental load and rich co‐help systems among the intimate community over time. Our challenge is to propose a new approach to sustain urban regeneration through a series of micro architectural interventions respecting the existing urban structure and social capital. The proposed architecture "Megacity Skeleton" is clearly constituted by the skeleton and the skin. How do we reveal hidden structures within a complicated web of social interactions? How can we explain the complex nexus of relationships in a public space? In this paper, we offer a different perspective on urban structures by using a multi-layer perspective to understand the structure of space that emerges from the complex uses of spaces. We particularly benefit from the concept of nesting as the theoretical lens in our research. We use a popular urban leisure space in Jakarta as a case study: The Hotel Indonesia (HI) Roundabout during Car Free Day Sunday events. We reveal that within this particular event, there are nested smaller events. The nesting structure also demonstrates how events and activities overlap within a single setting. Background: Hepatitis C virus (HCV) is a major cause of liver disease and a potential cause of substantial morbidity and mortality worldwide. The overall prevalence of HCV infection is 2%, representing 120 million people worldwide. Current standard treatment using pegylated interferon and ribavirin is effective in only 50% of the patients infected with HCV genotype 1, and is associated with significant side effects. Therefore, it is still of importance to develop new drugs for treatment of HCV. Antiviral substances obtained from natural products, including medicinal plants, are potentially good targets to study. In this study, we evaluated Indonesian medicinal plants for their anti-HCV activities. Methods: Ethanol extracts of 21 samples derived from 17 species of medicinal plants explored in the East Java region were tested. Anti-HCV activities were determined by a cell culture method using Huh7.5 cells and HCV strains of 9 different genotypes (1a to 7a, 1b and 2b). Results: Four of the 21 samples tested showed antiviral activities against HCV: Toona sureni leaves (TSL) with 50% inhibitory concentrations (IC50) of 13.9 and 2.0 μg/ml against the HCV J6/JFH1-P47 and -P1 strains, respectively, Melicope latifolia leaves (MLL) with IC50 of 3.5 and 2.1 μg/ml, respectively, Melanolepis multiglandulosa stem (MMS) with IC50 of 17.1 and 6.2 μg/ml, respectively, and Ficus fistulosa leaves (FFL) with IC50 of 15.0 and 5.7 μg/ml, respectively. Time-of-addition experiments revealed that TSL and MLL inhibited both at the entry and post-entry steps while MMS and FFL principally at the entry step. TSL and MLL inhibited all of 11 HCV strains of all the genotypes tested to the same extent. On the other hand, FFL showed significantly weaker inhibitory activities against the HCV genotype 1a strain, and MMS against the HCV strains of genotypes 2b and 7a to a lesser extent, compared to the other HCV genotypes. Conclusions: Ethanol extracts of TSL, MLL, MMS and FFL showed antiviral activities against all the HCV genotypes tested with the exception that some genotype(s) showed significant resistance to FFL and to MMS to a lesser extent.
