The impact of droughts on vegetation is essentially manifested as the transition of water shortage from the meteorological to ecological stages. Therefore, understanding the mechanism of drought propagation from meteorological to ecological drought is crucial for ecological conservation. This study proposes a method for calculating the probability of meteorological drought to trigger ecological drought at different magnitudes in Northwestern China. In this approach, meteorological and ecological drought events during 1982-2020 are identified using the three-dimensional identification method; the propagated drought events are extracted according to a certain spatio-temporal overlap rule; and propagation probability is calculated by coupling machine learning model and C-vine copula. The results indicate that: (1) 46 drought events are successfully paired by 130 meteorological and 184 ecological drought events during 1982-2020; ecological drought exhibits a longer duration, but smaller affected area and severity than meteorological drought; (2) Quadratic Discriminant Analysis (QDA) classifier performs the best among the 11 commonly used machine learning models which is combined with four-dimensional C-vine copula to construct drought propagation probability model; (3) the hybrid method considers more drought characteristics and more detailed propagation process which addresses the limited applicability of the traditional method to regions with large spatial extent. Agricultural drought is caused by reduced soil moisture and precipitation and affects the growth of crops and vegetation, and in turn agricultural production and food security. For developing measures for drought mitigation, reliable agricultural drought forecasting is essential. In this study, we developed an agricultural drought forecasting model based on canonical vine copulas under three-dimensions (3C-vine model), in which the antecedent meteorological drought and agricultural drought persistence were utilized as predictors. Besides, the meta-Gaussian (MG) model was selected as a reference model to evaluate the forecast skill. The agricultural drought in August of 2018 was selected as a case study, and the spatial patterns of 1-3-month lead forecasts of agricultural drought utilizing the 3C-vine model resembled the corresponding observations, indicating the predictive ability of the model. The performance metrics (NSE, R2, and RMSE) showed that the 3C-vine model outperformed the MG model for August under diverse lead times. Also, the 3C-vine model exhibited excellent forecast skills in capturing the extreme agricultural drought over different selected typical regions. Oxidative stress is implicated in the pathogenesis of diabetic kidney injury. SS-31 is a mitochondria-targeted tetrapeptide that can scavenge reactive oxygen species (ROS). Here, we investigated the effect and molecular mechanism of mitochondria-targeted antioxidant peptide SS-31 on injuries in diabetic kidneys and mouse mesangial cells (MMCs) exposed to high-glucose (HG) ambience. CD-1 mice underwent uninephrectomy and streptozotocin treatment prior to receiving daily intraperitoneal injection of SS-31 for 8 wk. The diabetic mice treated with SS-31 had alleviated proteinuria, urinary 8-hydroxy-2- deoxyguanosine level, glomerular hypertrophy, and accumulation of renal fibronectin and collagen IV. SS-31 attenuated renal cell apoptosis and expression of Bax and reversed the expression of Bcl-2 in diabetic mice kidneys. Furthermore, SS-31 inhibited expression of transforming-growth factor (TGF)-β1, Nox4, and thioredoxin-interacting protein (TXNIP), as well as activation of p38 MAPK and CREB and NADPH oxidase activity in diabetic kidneys. In vitro experiments using MMCs revealed that SS-31 inhibited HG-mediated ROS generation, apoptosis, expression of cleaved caspase-3, Bax/ Bcl-2 ratio, and cytochrome c (cyt c) release from mitochondria. SS-31 normalized mitochondrial potential (▵Ψ m) and ATP alterations, and inhibited the expression of TGF-β1, Nox4, and TXNIP, as well as activation of p38 MAPK and CREB and NADPH oxidase activity in MMCs under HG conditions. SS-31 treatment also could reverse the reduction of thioredoxin (TRX) biologic activity and upregulate expression of thioredoxin 2 (TRX2) in MMCs under HG conditions. In conclusion, this study demonstrates a protective effect of SS-31 against HG-induced renal injury via an antioxidant mechanism in diabetic nephropathy. [ABSTRACT FROM AUTHOR]
