A finite element procedure is developed for the computation of the thermoelastic properties of textile composites with complex and compact two- and three-dimensional woven reinforcement architectures. The purpose of the method is to provide estimates of the properties of the composite with minimum geometrical modeling effort. The software TexGen is used to model simplified representations of complex textiles. This results in severe yarn penetrations, which prevent conventional meshing. A non-conformal meshing strategy is adopted, where the mesh is refined at material interfaces. Penetrations are mitigated by using an original local correction of the material properties of the yarns to account for the true fiber content. The method is compared to more sophisticated textile modeling approaches and successfully assessed towards experimental data selected from the literature. [ABSTRACT FROM AUTHOR]

Stochastic foam models are generated from Voronoi spatial partitioning, using the centers of equi-sized hard spheres in random periodic distributions as seed points. Models with different levels of polydispersity are generated by varying the packing of the spheres. Subsequent relaxation is then performed with the Surface Evolver software which minimizes the surface area for better resemblance with real foam structures. The polydispersity of the Voronoi precursors is conserved when the models are converted into equilibrium models. The relation between the sphere packing fraction and the resulting degree of volumetric polydispersity is examined and the relations between the polydispersity and a number of associated morphology parameters are then investigated for both the Voronoi and the equilibrium models. Comparisons with data from real foams in the literature indicate that the used method is somewhat limited in terms of spread in cell volume but it provides a very controlled way of varying the foam morphology while keeping it periodic and truly stochastic. Stochastic cellular models of rigid foam based on Voronoi spatial partitioning are generated and investigated for potential use in numerical analysis using finite element methods. Such partitions are deterministic once a distribution of cell nuclei has been defined. A drawback is that the models tend to exhibit a significant share of short edges and small faces. Such small geometrical features are not likely to occur in real foams since they are unfavorable from a surface energy point of view and they also generate problems in numerical analysis due to associated meshing challenges. Through minimization of the surface area, using the computer software Surface Evolver, the Voronoi models are brought to better resemblance with ideal dry foam and the occurrence of small geometrical features is strongly reduced. It is generally seen that different seed point distribution algorithms result in different model topologies. The presented methodology is systematic, parameterized and the results are very promising. BACKGROUND: Pancreatic islet transplantation is a promising option for the treatment of diabetic patients; xenotransplantation of porcine islet cells would be a possibility to overcome the shortage of donor organs. Usually the donor pancreas is preserved with University of Wisconsin (UW) solution. A large number of reports have shown that the two-layer method (TLM), which employs oxygenated perfluorochemical and UW solution, is superior to simple cold storage. However, the extensive use of TLM is cost intensive and there is evidence that TLM only oxygenates small parts of the organ preserved. Another possibility to increase the oxygen supply during organ preservation would be the use of hyperbaric oxygenation (HBO) which increases the oxygen tension in fluids. The aim of this study was to evaluate the effect of pre-oxygenation of different preservation solutions on organ quality in terms of high energy phosphate levels as well as the occurrence of apoptosis and the induction of heat shock proteins and nitrosative stress induced cell death in porcine pancreatic tissue. METHODS: Porcine pancreatic tissue was preserved in different preservation solutions with or without pre-oxygenation for 6 hours of cold ischemic time (CIT). Then, tissue specimens were harvested and high energy phosphate levels were determined by HPLC. Moreover, immunohistochemistry was performed in order to detect occurrence of apoptosis, heat shock protein 70 (HSP70) as well as nitrosative stress induced cell death. RESULTS: Organs stored in pre-oxygenated UW solution showed best results in terms of high energy phosphate levels; apoptotic cells per islet as well as HSP70 positivity were significantly less when compared to simple UW storage and all other organ preservation solutions with or without pre-oxygenation. CONCLUSIONS: Pre-oxygenation of UW solution is a simple and promising method to improve islet cell quality after cold organ storage. Endotoxemia leads to the induction of inducible nitric oxide synthase (NOS-2) and increased expression of numerous inflammatory mediators contributing to endotoxin-induced acute lung injury. We tested the hypothesis that supplementation of nitric oxide (NO) by the novel NO donor S-nitroso human serum albumin (S-NO-HSA) given after lipopolysaccharide (LPS) challenge may reduce NOS-2 expression, lung inflammation and acute lung injury. Rats were divided into four groups: sham-operated (no treatment), LPS, LPS+HSA (human serum albumin), and LPS+S-NO-HSA. LPS was administered intravenously (20 mg kg−1) resulting in acute lung injury and a high mortality rate within 6 h (>90%). LPS-induced lung injury was characterized by an increased lung edema (lung wet/dry weight ratio), pulmonary neutrophil infiltration (myeloperoxidase activity, MPO) as well as a robust inflammatory response [increased expression of intercellular adhesion molecule-1 (ICAM-1), NOS-2, and cyclooxygenase-2 (COX-2)]. Infusion of S-NO-HSA or HSA was started 2 h after LPS and continued for 4 h (total dose of 72 mg kg−1) at a rate of 300 μg kg−1 min−1. S-NO-HSA but not HSA prolonged survival of endotoxemic rats, reduced the hypotensive response to LPS, minimized LPS-induced lung edema and injury, normalized MPO activity as well as diminished lung expression of pro-inflammatory molecules such as ICAM-1, NOS-2, and COX-2. Continuous supplementation of NO by S-NO-HSA after LPS challenge prevents induction of NOS-2, provides significant protection of endotoxin-induced acute lung injury, and prevents early mortality in endotoxic shock in rats. Neutrophil dysfunction in alcoholic hepatitis is associated with endotoxemia and an increased incidence of infection, but the mechanism is unclear. We aimed to investigate the role of Toll-like-receptors (TLR)2, 4, and 9 in mediating neutrophil dysfunction in alcoholic hepatitis. Neutrophils from healthy volunteers were incubated with alcoholic hepatitis patients' plasma (n = 12) with and without TLR2, 4, or 9 antagonists and with and without human albumin. TLR2, 4, and 9 expression, neutrophil oxidative burst, phagocytosis, and CXCR1+2 expression were measured by FACS analysis. Patients' plasma increased oxidative burst, decreased CXCR1+2 expression, and decreased phagocytosis of normal neutrophils in association with increased expression of TLR2, 4, and 9 and depletion of ATP. Inhibition of TLR2, 4, and 9 prevented the increase in oxidative burst and the decrease in CXCR1 and CXCR2 expression but did not prevent phagocytic dysfunction. Incubation with albumin completely prevented the patient plasma induced neutrophil dysfunction. Increased expression of TLR2, 4, and 9 is associated with neutrophil dysfunction, endotoxemia, arid energy depletion. TLR2, 4, and 9 inhibition does not improve phagocytosis, indicating that TLR overexpression may be the result and not the cause of neutrophil activation. The vascular endothelium of the coronary arteries has been identified as the important organ that locally regulates coronary perfusion and cardiac function by paracrine secretion of nitric oxide (NO) and vasoactive peptides. NO is constitutively produced in endothelial cells by endothelial nitric oxide synthase (eNOS). NO derived from this enzyme exerts important biological functions including vasodilatation, scavenging of superoxide and inhibition of platelet aggregation. Routine cardiac surgery or cardiologic interventions lead to a serious temporary or persistent disturbance in NO homeostasis. The clinical consequences are “endothelial dysfunction”, leading to “myocardial dysfunction”: no- or low-reflow phenomenon and temporary reduction of myocardial pump function. Uncoupling of eNOS (one electron transfer to molecular oxygen, the second substrate of eNOS) during ischemia-reperfusion due to diminished availability of L-arginine and/or tetrahydrobiopterin is even discussed as one major source of superoxide formation. Therefore maintenance of normal NO homeostasis seems to be an important factor protecting from ischemia/reperfusion (I/R) injury. Both, the clinical situations of cardioplegic arrest as well as hypothermic cardioplegic storage are followed by reperfusion. However, the presently used cardioplegic solutions to arrest and/or store the heart, thereby reducing myocardial oxygen consumption and metabolism, are designed to preserve myocytes mainly and not endothelial cells. Curved sandwich beams subjected to opening bending moment are studied. Face-core debonds of varying size are introduced at the compressively loaded face sheet and the structural integrity is investigated. Analytical and finite element models are compared in order to identify the governing failure modes of the beams. A simple expression is presented as a tool for getting a quick estimate of the severity of an interface crack in a curved sandwich beam. An investigation of the response of sandwich structures subjected to impact velocities of virtually 0 m/s and approximately 1000 m/s is conducted. The higher velocity exceeds both the longitudinal and the transverse wave propagation velocities of the core material in the sandwich panels. The objective is to investigate the possibility to simulate the damage from ballistic impact of sandwich panels through quasi-static experiments. Panels are impacted using a 40 mm Bofors AA gun and, using a similar projectile, other panels are indented quasi-statically. Energy absorption is measured in both test series. After impact, the panels are tested in in-plane compression together with one undamaged panel for reference. Residual strength of impacted panels is analyzed by finite element analysis. It is shown that the damage from high-velocity impact is limited, and it is possible to regain most of the undamaged strength by repair. [ABSTRACT FROM AUTHOR]

Although often not considered, the heart is one of the targets of multiple organ failure in sepsis and septic shock, with myocardial depression being a prominent component of this 'acute septic cardiomyopathy'. Hypotheses concerning the etiology of this depression are increasingly elucidated on a cellular level, including dysfunction of the β-adrenoceptor/G protein/adenylate cyclase system, calcium channel blockade by cardiodepressant factor, contractile impairment by activated leucocytes, as well as inhibition of protein synthesis by Pseudomonas exotoxin A. In the search for 'mechanisms of myocardial depression in sepsis', isolated cardiomyocytes may play a role as research tools with respect to: a) discrimination between direct and indirect cardiodepressant effects; b) identifying not only the acute, but also chronic toxin- and mediator-induced cardiodepression; c) clarification of the mechanism of action of cardiodepressant bacterial toxins and sepsis mediators; d) establishment of in vitro models of leucocyte-mediated cardiodepressann in sepsis. A qualitative analysis is presented of failure, perpendicular to the grain, in laminated timber reinforced with a glass fibre composite. The study is focused on beams with holes of different shape. The stress by corners, infinitesimal cracks and finite cracks are investigated. An initial crack model is suggested that brings about some of the phenomena observed in earlier performed experiments. A crack appears to propagate in the wood but is retarded in the reinforced beams. Eventually, the composite will fracture and failure of the beam follows. Finite element computations suggest that the reinforcement decreases the stress intensity at cracks in the wood and acts as a crack stopper. The reinforcing effect increases with the crack length. An experimental investigation of glass fibre reinforcement of glued laminated timber beams is presented. A polyester resin is used both as matrix and adhesive between the reinforcement and the wood. The main part of the work considers beams with large holes tested in three point bending. Circular and rectangular holes, centred at quarter length of the beams make the strength of wood perpendicular to the grain become critical. Great improvements of strength are obtained with the glass fibres. A comparison between various kinds and combinations of glass fibre reinforcement is made. Further, the reinforcement applied as repair of earlier cracked beams is investigated with positive results. One series of beams without holes is reinforced and tested in four-point bending.
