To properly repair and maintain implants, which are bone tissue implants that replace natural tooth roots, it is crucial to accurately identify their brand and specification. Deep learning has demonstrated outstanding capabilities in analysis, such as image identification and classification, by learning the inherent rules and degrees of representation of data models. The purpose of this study is to evaluate deep learning algorithms and their supporting application software for their ability to recognize and categorize three dimensional (3D) Cone Beam Computed Tomography (CBCT) images of dental implants. By using CBCT technology, the 3D imaging data of 27 implants of various sizes and brands were obtained. Following manual processing, the data were transformed into a data set that had 13,500 two-dimensional data. Nine deep learning algorithms including GoogleNet, InceptionResNetV2, InceptionV3, ResNet50, ResNet50V2, ResNet101, ResNet101V2, ResNet152 and ResNet152V2 were used to perform the data. Accuracy rates, confusion matrix, ROC curve, AUC, number of model parameters and training times were used to assess the efficacy of these algorithms. These 9 deep learning algorithms achieved training accuracy rates of 100%, 99.3%, 89.3%, 99.2%, 99.1%, 99.5%, 99.4%, 99.5%, 98.9%, test accuracy rates of 98.3%, 97.5%, 94.8%, 85.4%, 92.5%, 80.7%, 93.6%, 93.2%, 99.3%, area under the curve (AUC) values of 1.00, 1.00, 1.00, 1.00, 1.00, 1.00, 1.00, 1.00, 1.00. When used to identify implants, all nine algorithms perform satisfactorily, with ResNet152V2 achieving the highest test accuracy, classification accuracy, confusion matrix area under the curve, and receiver operating characteristic curve area under the curve area. The results showed that the ResNet152V2 has the best classification effect on identifying implants. The artificial intelligence identification system and application software based on this algorithm can efficiently and accurately identify the brands and specifications of 27 classified implants through processed 3D CBCT images in vitro, with high stability and low recognition cost. The shuttle effect and slow REDOX kinetics of lithium polysulfides (LiPSs) lead to low sulfur utilization rate, short cycle life, poor rate performance, which hinder the application of Li–S batteries. Herein, the Ni-NiS/NCF heterojunction composite was prepared with multistage pore structure and a large specific surface area, which can effectively capture LiPSs, provide more active sites for catalyzing LiPSs. Moreover, due to the heterojunction structure of Ni-NiS, in which NiS can effectively capture and catalyze lithium polysulfide, and Ni can effectively accelerate the diffusion and charge transfer of lithium ions, the Ni-NiS/NCF heterojunction composite establishes a high ion and electron conduction network, so as to achieve efficient mass and charge transfer capacity. The mutual coordination of uniformly distributed Ni-NiS heterojunctions inhibits the shuttle effect of LiPSs. When the sulfur load is 1.8 mg/cm2, the initial capacity of the cell with Ni-NiS/NCF-coated separator at 1 C is 1109.6 mAh/g, and the final discharge capacity is maintained at 618.0 mAh/g after 300 cycles. Ischemic stroke activates toll-like receptor 4 (TLR4) signaling, resulting in proinflammatory polarization of microglia and secondary neuronal damage. Herein, we report a novel lipid-nanoparticle (LNP)-mediated knockdown of TLR4 in microglia and amelioration of neuroinflammation in a mouse model of transient middle cerebral artery occlusion (tMCAO). siRNA against TLR4 (siTLR4) complexed to the novel LNP (siTLR4/DoGo310), which was based on a dioleoyl-conjugated short peptidomimetic (denote DoGo310), was readily internalized by the oxygen–glucose-deprived (OGD) mouse primary microglia, knocked-down TLR4, and polarized the cell to the anti-inflammatory phenotype in vitro. Systemic administration of siTLR4/DoGo310 LNPs in the tMCAO mice model resulted in the accumulation of siRNA mainly in the Iba1 positive cells in the peri-infarct. Analysis of the peri-infarct brain tissue revealed that a single injection of siTLR4/DoGo310 LNPs led to significant knockdown of TLR4 gene expression, reversing the pattern of cytokines expression, and improving the neurological functions in tMCAO model mice. Cationic polysaccharides have shown excellent ability of nucleic acids delivery. However, cationic curdlan derivatives with high degree of amination cause damage to the cell membrane and induce considerable cytotoxicity, limiting their in vivo application. Herein, we synthesized PEGylated 6-amino-6-deoxy-curdlan derivatives containing cleavable disulfide bonds. The resulting polymers (denote 6AC-2S PEGx) not only showed high affinity to siRNA but also exhibited significantly decreased cytotoxicity and hemolysis effect, while showing remarkable in vitro transfection efficiency. In vivo study demonstrated that 6AC-2S PEG40, which had a lower LD50 value than that of 6AC-100, did not cause liver damage, as the i.v. injection of 6AC-2S PEG40 to mouse did not increase serum level of ALT/AST. Furthermore, tissue distribution results showed that 6AC-2S PEG40 successfully delivered siRNA to liver, lung and spleen. The Mesozoic to Cenozoic mountain uplift, exhumation, and deformation of the SW Tianshan Mountains (Kyrgyzstan and Northwest China) offer an important window to understand the intra-continental rejuvenation mechanism of the Central Asian Orogenic Belt (CAOB), as response to the far-field effects of the India-Asia collision. This article presents new observation and data for the planation surface and sedimentation and deformation features of the regional intermountain basins to rebuild the orogenic history in Mesozoic to Cenozoic. Three planation surfaces were recognized by field observation, showing that the mountain may have experienced lengthy erosion since the end Cretaceous, and a continuous planation surface may have formed at the Eocene to Oligocene. The filling sequences and deformation character revealed that the orogenic disintegrate and intermountain basin formation likely began in the end of Oligocene. Subsequently, the uniform planation surface in Western Tianshan may have begun to disintegrate, leading to the basin-and-range landform formation. Folds and nappes in the Cenozoic basins, large-scale thrusting of Palaeozoic rocks over Cenozoic sediments at the basin margin associated with the rapid mountain uplift may have occurred at the end of Early Pleistocene, suggesting a tectonic inversion. The Mesozoic-Cenozoic Tianshan uplift and deformation were likely induced by the collision/accretion along the southern margin of Eurasia. Both the northward propagation of the Parmir syntaxis to the SW Tianshan and the oblique dextral faulting of the Talas-Fergana fault have likely played an important role on the formation and deformation of the Cenozoic basins in the SW Tianshan. The Atebayue Sb deposit is hosted in the Silurian clastics in the South Tianshan Orogen in Kyrgyzstan. The Sb ores appear as veins/veinlets and disseminations, with stibnite being the main ore mineral. Gangue minerals comprise quartz, calcite, and clay minerals. The quartz at Atebayue only contains aqueous fluid inclusions with low homogenization temperature (215–336 °C) and salinity (3.4–6.9 wt.% NaCl equiv.), supporting an epizonogenic hydrothermal origin. The minimum trapping pressures estimated from the NaCl─H2O inclusions are 9–14 MPa, suggesting that the Sb mineralization mainly occurred at a depth of 0.9–1.4 km. Sulphur isotopes (δ34S = −0.4 to 6.2‰) suggest that the host rocks within the Silurian system to be a significant source of ore metals. The ores contain average 206Pb/204Pb, 207Pb/204Pb, and 208Pb/204Pb values of 18.112, 15.547, and 38.064, respectively, and 2‐stage model ages of 337–381 Ma, indicating the ores were likely sourced from the Paleozoic strata. Integrating the data obtained from the studies including ore geology, fluid inclusion, and S─Pb isotope geochemistry, we conclude that that the Atebayue Sb deposit is best classified as epizonogenic type formed by the Tarim–Kazakhstan continent–continent collision. Sulfated glucopyranans having long alkyl groups were prepared by the ring-opening copolymerization of benzylated 1,6-anhydroglucopyranose with 3-O-octadecyl 1,6-anhydro-β-d-glucopyranose monomers, and subsequent deprotection and sulfation. Water-soluble sulfated glucopyranans with 2.8 and 4.7 mol% of 3-O-octadecyl group and lower molecular weights of M-n = 2.5 × 103–5.1 × 103 have potent anti-HIV activity at 0.05–1.25 μg/mL, even though sulfated polysaccharides with molecular weights below M-n = 6 × 103 had low anti-HIV activity. The interaction with poly-l-lysine as a model compound of proteins was analyzed by SPR, DSL, and zeta potential, indicating that the sulfated 3-O-octadecyl glucopyranans had high association and low dissociation rate constants, and the particle size increased after addition of poly-l-lysine. The anti-HIV activity was induced by electrostatic interaction between sulfate groups and amino groups of poly-l-lysine and by the synergistic effect of the hydrophobic long alkyl chain and hydrophilic sulfated group. To develop a biomaterial with an influenza virus-adsorptive functionality, an alkyl curdlan sulfate was prepared by ionic interaction between a positively charged didodecyldimethyl ammonium bromide and a negatively charged sulfate group of curdlan sulfate, which has potent anti-HIV activity, and then it was coated on a membrane filter with a 1-μm pore size by hydrophobic interaction with the long alkyl groups in the curdlan sulfate. The alkyl curdlan sulfate with the degree of alkylation (DOA) of 0.03 (one didodecyldimethyl group/12 sugar residues with 36 hydroxyl or sulfate groups) showed potent anti-HIV activity in a 50% effective concentration (EC50) as low as 0.87 μg/mL (standard curdlan sulfate EC50 = 0.3 μg/mL), and the activity decreased with increasing DOA. A DOA higher than 0.1 (one didodecyldimethyl group/three sugar residues with nine hydroxyl or sulfate groups) gave no anti-HIV activity. Although both curdlan sulfate and alkyl curdlan sulfates did not inhibited infection of Madin-Darby canine kidney cells by influenza viruses, the alkyl curdlan sulfate-coated membrane filter was found to have a specific adsorptive functionality for influenza A virus in vitro. When 1.6 mg of the alkyl curdlan sulfate with the DOA of 0.03 was coated on a membrane filter (φ13 mm; pore size, 1 μm), three stacked alkyl curdlan sulfate-coated membrane filters dramatically decreased hemagglutination to 1/4-1/32. However, the membrane filter did not effectively remove on influenza B viruses, and thus a membrane filter without alkyl curdlan sulfate was not effective against influenza viruses. These results can therefore be presumed to demonstrate that the alkyl curdlan sulfate-coated membrane filter removed influenza A viruses by adsorption between the negatively charged sulfate groups and the positively charged envelope protein of the virus. © 2011 Wiley Periodicals, Inc. Cutaneous melanoma is the most aggressive skin cancer with notorious drug resistance. Inhibition of immune checkpoint molecules is one of the most promising approaches for cancer therapy. Herein, we show that RNAi mediated silencing of STAT3 expression in the tumor tissue robustly inhibit tumor growth in B16F10 mouse model of melanoma. We designed a peptidomimetic-based lipid nanoparticles (LNPs) for the delivery of siRNA in mouse model of melanoma. When systemically administered, the novel formulation (denote DoCh) preferentially delivered siRNA to the tumor tissue. Remarkably, sequential intravenous injections of siRNA against STAT3 induced profound silencing of STAT3 expression in tumor tissue, which resulted in significant downregulation of PD-L1, leading to significant inhibition of tumor growth through inhibition of tumor immune checkpoint. Moreover, DoCh-mediated siRNA delivery did not show noticeable damage to the major organs. Collectively, our data demonstrated that DoCh LNP is a promising tumor-targeted siRNA delivery system. Lipid nanoparticles (LNP) are the most potent carriers for the delivery of nucleic acid-based therapeutics. The first FDA approved a short interfering RNA (siRNA) drug that uses a cationic LNP system for the delivery of siRNA against human transthyretin (hTTR). However, preparation of such LNP involves tedious multi-step synthesis with relatively low yields. In the present study, we synthesized cationic peptidomimetic functionalized cholesterol (denote Chorn) in straightforward chemical approaches with high yield. When formulated with helper lipids, Chorn LNPs complexed with siRNA to form nanoparticles with an average diameter of 150 nm to 200 nm. Chorn LNP mediated transfection of a green fluorescence protein (GFP) expressing plasmid resulted in 60% GFP positive cells. Moreover, Chorn LNP delivered siRNA against polo-like kinase 1 (Plk1), a disease related gene in cancer cells and efficiently suppressed the expression of the gene, resulting in significant morphological changes in the cell nuclei. Sugars containing cationic polymers are potential carriers for in vitro and in vivo nucleic acid delivery. Monosaccharides such as glucose and galactose have been chemically conjugated to various materials of synergistic poly-lysine dendrimer systems for efficient and biocompatible delivery of short interfering RNA (siRNA). The synergistic dendrimers, which contain lipid conjugated glucose terminalized lysine dendrimers, have significantly lower adverse impact on cells while maintaining efficient cellular entry. Moreover, the synergistic dendrimers complexed to siRNA induced RNA interference (RNAi) in the cells and profoundly knocked down green fluorescence protein (GFP) as well as the endogenously expressing disease related gene Plk1.
