Endothelial dysfunction develops gradually with age, and is the foundation of many age-related diseases in the elderly. The purpose of this study was to investigate the role of the cyclic GMP-AMP synthase (cGAS)-stimulator of interferon genes (STING) pathway in aging-related endothelial dysfunction. Endothelial functional parameters and biochemical indices of vascular function were examined in 2-, 6-, 12- and 24-monthold mice. Then, 6-month-old mice were administered RU.521, a specific inhibitor of cGAS, for 6 months, and endothelial functional parameters and biochemical indices of vascular function were re-examined. An in vitro model of cell senescence was established by treating human aortic endothelial cells (HAECs) with D-Galactose (D-GAL). Using inhibitors or siRNA interference, cGAS and STING were suppressed or silenced in senescent HAECs, and changes in the expression of eNOS, the senescence markers, p53, p21 and p16, components of the cGAS-STING pathway and Senescence-Associated β-galactosidase (SA-β-gal) staining were examined. Finally, cGAS, STING and p-IRF3 levels were measured in aorta tissue sections from eight patients. A decline in endothelial function, up-regulation of p53, p21 and p16 expression, and activation of the cGAS-STING pathway were observed in aging mice. Inhibition of cGAS was found to improve endothelial function and reverse the increased expression of aging markers. Our in vitro data demonstrated that D-GAL induced a decrease in eNOS expression and cell senescence, which could be partly reversed by cGAS inhibitor, STING inhibitor, siRNA-cGAS and siRNA-STING treatment. Higher expression levels of cGAS, STING and p-IRF3 were observed in aged human aortic intima tissue compared to young aortic intima tissue. Our study demonstrated that activation of the cGAS-STING pathway played a vital role in aging-related endothelial dysfunction. Blood-brain barrier (BBB) breakdown and inflammation occurring at the BBB have a key, mainly a deleterious role in the pathophysiology of ischemic stroke. Neddylation is a ubiquitylation-like pathway that is critical in various cellular functions by conjugating neuronal precursor cell-expressed developmentally down-regulated protein 8 (NEDD8) to target proteins. However, the roles of neddylation pathway in ischemic stroke remain elusive. Here, we report that NEDD8 conjugation increased during acute phase after ischemic stroke and was present in intravascular and intraparenchymal neutrophils. Inhibition of neddylation by MLN4924, also known as pevonedistat, inactivated cullin-RING E3 ligase (CRL), and reduced brain infarction and improved functional outcomes. MLN4924 treatment induced the accumulation of the CRL substrate neurofibromatosis 1 (NF1). By using virus-mediated NF1 silencing, we show that NF1 knockdown abolished MLN4924-dependent inhibition of neutrophil trafficking. These effects were mediated through activation of endothelial P-selectin and intercellular adhesion molecule-1 (ICAM-1), and blocking antibodies against P-selectin or anti-ICAM-1 antibodies reversed NF1 silencing-induced increase in neutrophil infiltration in MLN4924-treated mice. Furthermore, we found that NF1 silencing blocked MLN4924-afforded BBB protection and neuroprotection through activation of protein kinase C 6 (PKC6), myristoylated alanine-rich C-kinase substrate (MARCKS), and myosin light chain (MLC) in cerebral microvessels after ischemic stroke, and treatment of mice with the PKC6 inhibitor rottlerin reduced this increased BBB permeability. Our study demonstrated that increased neddylation promoted neutrophil trafficking and thus exacerbated injury of the BBB and stroke outcomes. Nine herbivore-induced plant volatiles (HIPVs) and one methyl jasmonate were field-tested for their attractiveness to the main predators in cotton fields of North China in 2 yr. The main predators including ladybird beetles (Propylaea japonica (Thunberg), Harmonia axyridis (Pallas)), green lacewings (Chrysoplera sinica (Tjeder), Chrysopa spp.), predatory bugs (Geocoris pallidipennis (Costa), Orius spp., Nabis spp.) and spiders (Misumenops tricuspidatus (Fabricius), Erigonidium graminicolum (Sundevall)) were investigated. Two-way ANOVA indicated that the volatile compound, year, and the volatile compound x year interaction affected the behavioral responses of predators. It was found that indole significantly attracted the ladybird beetle P. japonica, H. axyridis. Linalool could attract P. japonica. Green lacewing C. sinica was significantly attracted by α-pinene and β-pinene, whereas indole significantly attracted Chrysopa spp. Methyl jasmonate and α-pinene showed significant attraction to small-flower bug Orius spp. In addition, the attraction of α-humulene to C. sinica, attractiveness of β-pinene to Orius spp. and Chrysopa spp., were observed only in one of the two years. However, the big-eyed bug G. pallidipennis, damsel bug Nabis spp., spiders M. tricuspiata and E. graminicolum did not respond to any of the tested HIPVs. Microplitis mediator Haliday (Hymenoptera: Braconidae) is an important larval endoparasitoid of various lepidopteran pests, including Helicoverpa armigera (Hübner). In China, H. annigera is a key pest of cotton and is currently the focus of several biological control efforts that use M. mediator as principal natural enemy of this pest. To improve the success of biological control efforts, behavioral studies are needed that shed light on the interaction between M. mediator and H. armigera. In this study, we determined M. mediator response to volatile compounds from undamaged, mechanically injured, or H. armigera--damaged plants and identified attractive volatiles. In Y-tube olfactometer assays, we found that mechanically damaged plants and/or plants treated with H. arinigera oral secretions did not attract wasps. However, volatiles from H. armigeradamaged plants elicited a strong attraction of both M. mediator sexes. Headspace extracts from H. armigera-damaged cotton were analyzed by coupled gas chromatographyelectroantennographic detection (GC-EAD), and a total of seven different compounds were found to elicit electroantennogram (EAG) responses, including an unknown compound. Six different EAD-active volatiles were identified from caterpillar-damaged cotton plants, of which 3, 7-dimethyl-1, 3, 6-octatriene and (Z)-3-hexenyl acetate were the principal compounds. Olfactometer assays indicated that individual synthetic compounds of 3, 7-dimethyl-1, 3, 6-octatriene, (Z) -3-hexenyl acetate, and nonanal were attractive to M. mediator. Field cage studies showed that parasitism of H. armigera larvae by M. mediator was higher on cotton plants to which 3,7-dimethyl-1,3, 6-octatriene was applied. Our results show that the combination of terpenoids and green leaf volatiles may not only facilitate host, mate, or food location but may also increase H. armigera parasitism by M. mediator. Seven synthetic herbivore-induced plant volatiles (HIPVs) and a mixture of nonanal + (Z)-3-hexen-1-ol were field tested for their ability to attract beneficial insects in an open cotton field. Eleven species of the main natural enemies of insect pests in cotton fields were studied. Significantly greater numbers of the ladybird beetle Coccinella septemptunctata were trapped on (Z)-3-hexenyl acetate-baited cards than on others that were HIPV baited or the control cards. Erigonidium graminicolum was attracted to traps baited with nonanal, (Z)-3-hexenyl acetate, and methyl salicylate (MeSA). The predatory bug Deraeocoris punctulatus was only attracted to traps baited with octanal, The predatory bug Onus similis responded to traps baited with 3,7-dimethyl,1,3,6-octatriene, nonanal, (Z)-3-hexenyl acetate, nonanal + (Z)-3-hexen-1-ol, and MeSA. Dimethyl octatriene, nonanal + (Z)-3-hexen-1-ol, and octanal significantly attracted the syrphid fly Paragus quadrifasciatus. The ladybird beetle Propylaea japonica, the green lacewing Chrysopa sinica, the bigeyed bug Geocoris pallidipennis, the syrphid fly Epistrophe balteata, and the parasitic wasp Campoletis chlorideae did not respond to any of the HIPVs tested.
