Telescopic wing requires changing the wingspan under aerodynamic load. Structural deformation caused by forces and moments will result in increased driving forces or even stagnation. Deformation and distributed contact force of linear motion pair of the load-bearing mechanism which considers the geometric nonlinearity of guide rail is necessary to predict the driving force, the mechanical efficiency, or the stiffness matching in the optimal design process. Aiming at the cantilever load-bearing mechanism, the structural model was established in terms of the flexibility of the components, contact stiffness and other parameters on the friction of moving pairs are studied based on the modified chained-beam constraint model (CBCM). The model prediction accuracy is verified through simulation and experiment. The result shows that the friction force is related to the stiffness of guide rail, slider connector, ball bearing contact as well as the geometric parameters. Background: With the changes in lifestyle and diet structure, the incidence of obesity has increased year by year, and obesity is one of the inducements of many chronic metabolic diseases. Epigallocatechin gallate (EGCG), which is the most abundant component of tea polyphenols, has been used for many years to improve obesity and its complications. Though it has been reported that EGCG can improve obesity through many molecular mechanisms, EGCG may have many mechanisms yet to be explored. In this study, we explored other possible mechanisms through molecular docking and in vitro experiments. Methods: AutoDock Vina was selected for conducting the molecular docking analysis to elucidate the interaction between EGCG and Notch1, while molecular dynamics simulations were employed to validate this interaction. Then, the new regulation mechanism of EGCG on obesity was verified with in vitro experiments, including a Western blot experiment, immunofluorescence experiment, oil red O staining, and other experiments in 3T3-L1 adipocytes. Results: The molecular docking results showed that EGCG could bind to Notch1 protein through hydrogen bonding. In vitro cell experiments demonstrated that EGCG can significantly reduce the sizes of lipid droplets of 3T3-L1 adipocytes and promote UCP-1 expression by inhibiting the expression of Notch1 in 3T3-L1 adipocytes, thus promoting mitochondrial biogenesis. Density is an important parameter for both geological research and geophysical exploration. However, for model-driven seismic inversion methods, high-fidelity density inversion is challenging due to seismic wave traveltime insensitivity to density and crosstalk that density has with velocity. To circumvent the challenge of density inversion, some inversion methods treat density as a constant value or derive density from velocity through empirical equation. On the other hand, deep learning approaches are completely driven by data and have strong target-oriented characteristics, offering a new way to solve multiparameter coupling problems. Nevertheless, the accuracy of the inversion results of data-driven algorithms is directly related to the amount and diversity of the training data, and thus, they lack the universality of model-driven algorithms. To achieve accurate density inversion, we propose a simultaneous inversion algorithm for velocity and density that combines the advantages of data- and model- driven approaches: A neural network model (U-T), combining the U-net and Transformer architectures, is proposed to construct non-linear mappings between seismic data as inputs and the velocity and density as predictions. Next, the model-driven inversion algorithm uses the U-T prediction as the initial model to obtain the final accurate solution. In the model-driven module, envelope-based sparse constrained deconvolution is used to obtain full-band seismic data, while a variable dominant frequency full waveform inversion algorithm is used to perform multiscale inversion, ultimately leading to accurate inversion results of velocity and density. Chloride corrosion is a key factor affecting the life of marine concrete, and surface chloride concentration is the main parameter for analyzing its durability. In this paper, we first introduce six erosion mechanism models for surface chloride ion concentration, reveal the convection effect in the diffusion behavior of chloride ions, and then introduce the corrosion mechanisms that occur in different marine exposure environments. On this basis, the analysis is carried out using empirical formulations and machine learning methods, which provides a clearer understanding of the research characteristics and differences between empirical formulas and emerging machine learning techniques. This paper summarizes the time-varying model and multifactor coupling model on the basis of empirical analysis. It is found that the exponential function and the reciprocal function are more consistent with the distribution law of chloride ion concentration, the multifactor model containing the time-varying law is the most effective, and the Chen model is the most reliable. Machine learning, as an emerging method, has been widely used in concrete durability research. It can make up for the shortcomings of the empirical formula method and solve the multifactor coupling problem of surface chloride ion concentration with strong prediction ability. In addition, the difficulty of data acquisition is also a major problem that restricts the development of machine learning and incorporating concrete maintenance conditions into machine learning is a future development direction. Through this study, researchers can systematically understand the characteristics and differences of different research methods and their respective models and choose appropriate techniques to explore the durability of concrete structures. A friction stir vibration processing (FSVP) with different vibration frequencies (20 Hz, 40 Hz, and 80 Hz) was employed for the fabrication of AZ70/ZrO2–CeO2 surface composite with the aim of improving mechanical and wear performance. Thus, the influence of vibration frequency as a vital parameter on the microstructure and reinforcement distribution, shear punch strength (SPT), and wear behavior of the composites was investigated. The results indicated that the material flow, agglomeration of particles, and dislocation density were influenced by the increasing vibration frequency that can be contributed to the successive mechanical stirring-assisted plastic deformation/strain and further grain refinement. It was also demonstrated that when vibration frequency was increased, the shear punch strength (by about 34%) and wear resistance (by about 44%) of the developed composites improved. Congenital infection caused by vertical transmission of microsporidia N. bombycis can result in severe economic losses in the silkworm-rearing industry. Whole-transcriptome analyses have revealed non-coding RNAs and their regulatory networks in N. bombycis infected embryos and larvae. However, transcriptomic changes in the microsporidia proliferation and host responses in congenitally infected embryos and larvae remains unclear. Here, we simultaneously compared the transcriptomes of N. bombycis and its host B. mori embryos of 5-day and larvae of 1-, 5- and 10-day during congenital infection. For the transcriptome of N. bombycis, a comparison of parasite expression patterns between congenital-infected embryos and larva showed most genes related to parasite central carbon metabolism were down-regulated in larvae during infection, whereas the majority of genes involved in parasite proliferation and growth were up-regulated. Interestingly, a large number of distinct or shared differentially expressed genes (DEGs) were revealed by the Venn diagram and heat map, many of them were connected to infection related factors such as Ricin B lectin, spore wall protein, polar tube protein, and polysaccharide deacetylase. For the transcriptome of B. mori infected with N. bombycis, beyond numerous DEGs related to DNA replication and repair, mRNA surveillance pathway, RNA transport, protein biosynthesis, and proteolysis, with the progression of infection, a large number of DEGs related to immune and infection pathways, including phagocytosis, apoptosis, TNF, Toll-like receptor, NF-kappa B, Fc epsilon RI, and some diseases, were successively identified. In contrast, most genes associated with the insulin signaling pathway, 2-oxacarboxylic acid metabolism, amino acid biosynthesis, and lipid metabolisms were up-regulated in larvae compared to those in embryos. Furthermore, dozens of distinct and three shared DEGs that were involved in the epigenetic regulations, such as polycomb, histone-lysine-specific demethylases, and histone-lysine-N-methyltransferases, were identified via the Venn diagram and heat maps. Notably, many DEGs of host and parasite associated with lipid-related metabolisms were verified by RT-qPCR. Taken together, simultaneous transcriptomic analyses of both host and parasite genes lead to a better understanding of changes in the microsporidia proliferation and host responses in embryos and larvae in N. bombycis congenital infection. The present study was conducted to develop a green process that provides access to the development of Schiff base derivatives of chitosan with the heterocyclic moiety as a novel class of anti-gastric cancer agent. In the present study, we have synthesized these derivatives by reacting various pyrazoles with chitosan using CAN in PEG400. The compounds were synthesized in 20 min in excellent yield by using CAN at 5% in PEG400 at 80°C in the shortest reaction time of 20 min. The PEG400 could be efficiently recycled for the three consecutive runs. The developed compounds were tested for EGFR-TK inhibition using a Kinase-Glo Plus luminescence kinase assay kit where they exhibited significant activity revealing compound 2d as the most potent analog, while other compounds showed mild to moderate inhibitory activity. MTT assay was conducted to determine the effect of the three most potent EGFR inhibitors (2b, 2c, and 2d) on the proliferation of gastric cancer cells (SGC-7901). The results showed compound 2d as the most potent anticancer agent against SGC7901 cells. The effect of compound 2d was also quantified on the apoptosis and cell phase of SGC7901 cells using flow cytometry assay at various concentrations ranging from 0, 10, 20, and 30 µM. Results suggest that compound 2d showed significant inhibition of SGC-7901 by inducing apoptosis and arresting G0/G1 cell phase. The western blot analysis also revealed that compound 2d significantly inhibited the overexpression of EGFR in SGC-7901 cells. The development of cost‐effective and stable electrocatalysts that can replace the prevailing Pt‐based and Ir‐based catalysts for water splitting remains a formidable challenge. The electrocatalytic performance of a catalyst depends on its chemical composition and environment during catalysis. Herein, a Ru–Mo composite is used as an example to highlight the significance of tailoring the chemical environments of the active‐sites. This customization enhances the cathodic and anodic reactions involved in water splitting, ultimately leading to an improved full reaction efficiency. Specifically, the chemically reduced state of Ru–Mo demonstrates promising hydrogen evolution activity and exhibits low overpotentials of 48 and 34 mV at the current density of 10 mA cm−2 in acidic and alkaline electrolytes, respectively. The chemically oxidized state of Ru–Mo, derived from the same precursor, demonstrates proficient oxygen evolution activity and exhibits low overpotentials of 260 and 270 mV at 10 mA cm−2 in acidic and alkaline electrolytes, respectively. Additionally, in both cases, the Ru–Mo composites exhibit significantly improved stability under typical water‐splitting conditions. Despite the close chemical compositions of these two catalysts, they show poor performance in the counter electrode reactions, demonstrating the importance of establishing a suitable chemical environment for efficient electrocatalysis. Aiming at the test requirements of specified path coverage of fully automatic operation (FAO) system test, this paper proposes an algorithm for automatic generation of test cases for FAO based on the modeling theory of timed automata to meet the specified path coverage and edge coverage. Firstly, the modeling theory of timed automata is studied, and the FAO timed automata model is established. Then, a modeling method to add marker variables to the d automata model is proposed to characterize the FAO test requirements of specified path coverage. Thirdly, combined with the test case generation mechanism of Yggdrasil, a test case generation method of FAO is proposed, which satisfies both the specified path coverage and edge coverage criteria of FAO. Finally, taking the mode conversion function of FAO as an example, the timed automata model is established and test cases are generated. The results show that the test cases cover 100 % of the test requirements specified by the tester and all the edges of the timed automata model, which can meet the test requirements of the specified path coverage of FAO. Microsporidia are a group of obligate intracellular parasites that infect almost all animals, causing serious human diseases and major economic losses to the farming industry. Nosema bombycis is a typical microsporidium that infects multiple lepidopteran insects via fecal-oral and transovarial transmission (TOT); however, the underlying TOT processes and mechanisms remain unknown. Here, we characterized the TOT process and identified key factors enabling N. bombycis to invade the ovariole and oocyte of silkworm Bombyx mori. We found that the parasites commenced with TOT at the early pupal stage when ovarioles penetrated the ovary wall and were exposed to the hemolymph. Subsequently, the parasites in hemolymph and hemolymph cells firstly infiltrated the ovariole sheath, from where they invaded the oocyte via two routes: (I) infecting follicular cells, thereby penetrating oocytes after proliferation, and (II) infecting nurse cells, thus entering oocytes following replication. In follicle and nurse cells, the parasites restructured and built large vacuoles to deliver themselves into the oocyte. In the whole process, the parasites were coated with B. mori vitellogenin (BmVg) on their surfaces. To investigate the BmVg effects on TOT, we suppressed its expression and found a dramatic decrease of pathogen load in both ovarioles and eggs, suggesting that BmVg plays a crucial role in the TOT. Thereby, we identified the BmVg domains and parasite spore wall proteins (SWPs) mediating the interaction, and demonstrated that the von Willebrand domain (VWD) interacted with SWP12, SWP26 and SWP30, and the unknown function domain (DUF1943) bound with the SWP30. When disrupting these interactions, we found significant reductions of the pathogen load in both ovarioles and eggs, suggesting that the interplays between BmVg and SWPs were vital for the TOT. In conclusion, our study has elucidated key aspects about the microsporidian TOT and revealed the key factors for understanding the molecular mechanisms underlying this transmission. Author summary: In addition to horizontal transmission, certain species of microsporidia possess the capability of vertically transmitting to host offspring by invading parental oocytes, a process known as transovarial transmission (TOT). However, the pathways and mechanisms through which these parasites penetrate the host barrier system and enter oocytes remain unclear. This study elucidates a comprehensive process in which the microsporidian Nosema bombycis infects various components of the silkworm Bombyx mori ovariole sheath, including follicular cells (FCs) and nurse cells (NCs), ultimately reaching their destination within the oocytes. Within FCs and NCs, N. bombycis undergoes restructuring and forms large vacuoles for delivering themselves into the oocyte. Furthermore, we have identified B. mori vitellogenin (BmVg) and specific spore wall proteins (SWP12, SWP26, SWP30) from N. bombycis as key factors involved in TOT. Additionally, our findings showed that specific domains within BmVg (VWD and DUF1943) are essential for binding with these spore wall proteins. Simple Summary: Lepidopteran insects are serious agricultural and forest pests that cause enormous global economic losses. The development of gene editing methods will significantly improve our ability to study gene function and greatly facilitate the control of lepidopteran pests. The application of gene editing technology in the model organism of lepidopteran insects, Bombyx mori, is mainly achieved by the microinjection of early embryos, but this approach is challenging in other lepidopteran insects. In this study, Cas9-mediated gene editing was established using the Receptor-Mediated Ovary Transduction of Cargo (ReMOT) control technique by injection into B. mori female pupae. We identified a B. mori oocytes-targeting peptide ligand (BmOTP) that mediated the transduction of Cas9 ribonucleoprotein into the oocytes, resulting in heritable gene editing of the offspring. Because the BmOTP ligand was highly conserved among lepidopteran, our results will significantly facilitate genetic manipulation of other lepidopteran insects, advancing pest control or economic insect breeding. Lepidoptera is one of the most speciose insect orders, causing enormous damage to agricultural and forest crops. Although genome editing has been achieved in a few Lepidoptera for insect controls, most techniques are still limited. Here, by injecting female pupae of the Lepidoptera model species, Bombyx mori, gene editing was established using the Receptor-Mediated Ovary Transduction of Cargo (ReMOT) control technique. We identified a B. mori oocytes-targeting peptide ligand (BmOTP, a 29 aa of vitellogenin N-terminal of silkworms) with a highly conserved sequence in lepidopteran insects that could efficiently deliver mCherry into oocytes. When BmOTP was fused to CRISPR-associated protein 9 (Cas9) and the BmOTP-Cas9 ribonucleoprotein complex was injected into female pupae, heritable editing of the offspring was achieved in the silkworms. Compared with embryo microinjection, individual injection is more convenient and eliminates the challenge of injecting extremely small embryos. In this study, a composite modifier for wood impregnation is prepared, which is functional and environmentally friendly. The surface of silica sol was modified by using KH-560. The modified silica sol, melamine, and glyoxal were used as raw materials. The silica sol/melamine glyoxal resin (from now on referred to as Silica sol/MG) composite modifier was prepared based on in-situ polymerization. The physicochemical properties (viscosity, solid content and etc.) of the composite modifier were evaluated. The structural and thermal properties were characterized and analyzed by FTIR spectroscopy, particle size distribution, TG and DSC. The results showed that the viscosity and solid content of the composite modifier decreased with the increase in the mass of the silica sol. The FTIR spectroscopy showed peaks at 473 cm−1 and 1101 cm−1, which were assigned to bending and stretching vibrations of the Si-O-Si bond, respectively, indicating that the modified silica sol was successfully introduced into the MG resin. When the modified silica sol mass fraction was 30%-40%, the particle size distribution of the composite modifier was relatively uniform. TG analysis found that the thermal stability of the composite modifier was significantly improved compared with the unmodified resin. Electroreduction of oxygen driven by renewable electricity holds significant promise for the sustainable production of value‐added hydrogen peroxide (H2O2). While water is a desirable source of protons and electrons for this reaction, its low gas solubility often limits the transportation of gas molecules and consequently leads to large concentration overpotential, resulting in unsatisfactory energy efficiencies. Herein, a facile and effective strategy to promote the 2e− oxygen reduction reaction (ORR) through microenvironment modulation is presented. In this work, it is specifically aimed to facilitate oxygen transportation at the reaction interface, particularly at high rates. To achieve this, hydrophobic polytetrafluoroethylene (PTFE) particles are introduced into a catalyst layer containing amino‐group‐functionalized carbon nanotube (CNT‐NH2) as the ORR catalyst for H2O2 production. As a result, the PTFE‐modified CNT‐NH2‐based gas‐diffusion electrode (GDE) substantially improves ORR activity. At 100 mA cm−2, the PTFE‐modified CNT‐NH2 achieves a high cathodic energy efficiency of 92%, 1.5 times higher than the pristine CNT‐NH2‐based GDE (63%). Detailed kinetic analysis reveals that this enhanced ORR performance is indeed due to the enhanced oxygen transportation induced by the persistent hydrophobic microenvironment created by the PTFE‐modified catalyst layer, reducing the concentration overpotential during ORR. In order to overcome the defects of fast-growing poplar wood, such as low strength and poor toughness, this paper introduces a method of modifying poplar wood via impregnation with silica sol/melamine–glyoxal (silica sol/MG) resin and explores its effects on the physical, mechanical, and thermal properties of poplar wood. It was found via scanning electron microscopy that the composite modifier covered and filled the cell lumen, cell interstitial space, and cell wall pores of poplar wood. Further, infrared spectroscopy and X-ray photoelectron spectroscopy analyses confirmed that chemical cross-linking occurred between the silica sol/MG resin composite modifier and the internal groups of poplar wood and that the Si-O-Si flexible long chains introduced in the composite modifier formed a cross-linking network with poplar wood such as Si-O-Si and Si-O-C, which led to the improvement of the physical and mechanical properties and the enhancement of the thermal stability of poplar wood. Phenolic resin-modified materials partially reduce the toughness of the wood. In this study, organic–inorganic composite modifiers were used to modify the wood. Silica sol/phenolic resin was prepared through in-situ polymerization, and poplar wood was modified using a vacuum pressure impregnation process, enhancing its toughness. Orthogonal experiments were conducted, and the impact toughness of the modified poplar wood was used as the evaluation index. Through orthogonal experiments, using the impact toughness of modified poplar as the evaluation indicator, it was found that when the average particle size of the silica sol is 8–15 nm, the pressure is 1.2 MPa, and the pressurization time is 3 h, the impregnation-modified poplar's impact toughness reaches its optimum, improving by 84.1% and 135.4% compared to the raw material and phenolic resin impregnated wood, respectively. The Fourier Transform Infrared Spectroscopy (FT-IR) results indicated that the characteristic absorption peak of Si-O-Si appears in the poplar wood after impregnation, confirming the formation of new silicon-oxygen (Si-O) chemical bonds. X-ray Photoelectron Spectroscopy (XPS) analysis revealed that a chemical reaction occurs between the impregnation liquid and the wood, generating Si-O-C. Subsequently, through Dynamic Mechanical Analysis (DMA) and Thermogravimetric (TGA) analysis, it was understood that this chemical reaction significantly enhances the thermal stability and toughness of the impregnated material, making it superior to the original poplar material. The TGA further unveiled that, compared to untreated poplar, the thermal stability of the impregnated material has been notably improved. The present study investigates the influence of micron‐sized diatomite dosage on the physical, mechanical, rheological, and creep properties of poplar wood/polypropylene (PW/PP) composites to enhance their toughness and practicality. The diatomite‐PW/PP composites were prepared using the melt extrusion method, replacing different proportions of PW with diatomite, and subsequently evaluated for their properties. The results indicate that adding diatomite enhances the density while reducing the brightness and water absorption of the composites. Scanning electron microscopy, analysis reveals a uniform distribution of diatomite in the range of 5–10 wt%, which effectively serves as a filler and reinforcing agent, consequently improving the toughness of the composite system. Specifically, the impact strength, tensile strength, and elongation at break are enhanced by 25.9%, 9.5%, and 35.7%, respectively. Alkaline rock-type rare-earth (RE) ores have significant utilisation value. However, the exploitation of such resources faces great challenges owing to the complex mineral and element assemblages. Composite collectors exhibit excellent performances, which may provide solutions to the flotation problem of alkaline rock-type RE ores. Therefore, 16 collectors typically used in RE ores flotation were selected. Flotation tests were performed to identify collectors with high selectivity and collection ability for RE minerals, then nine composite collectors were prepared by combining the satisfactory collectors. The flotation performances of single and composite collectors for RE minerals were examined, and the composite collector FA301 with different carbon chain lengths was identified as the best one. When FA301 was applied in optimal conditions of slurry temperature, grinding size, collector and inhibitor dosage, RE concentrate yield of 6.29%, REO grade of 32.013%, and recovery of 59.02% were achieved. According to the results of the zeta potential, FTIR, and XPS test, the functional groups (dominated by carboxyl groups) in FA301 chemically adsorbed onto the main active sites (La, Ce, Y, etc.) on the surface of RE minerals. Background: The interaction networks between coding and non-coding RNAs (ncRNAs) including long non-coding RNA (lncRNA), covalently closed circular RNA (circRNA) and miRNA are significant to elucidate molecular processes of biological activities and interactions between host and pathogen. Congenital infection caused by vertical transmission of microsporidia N. bombycis can result in severe economic losses in the silkworm-feeding industry. However, little is known about ncRNAs that take place in the microsporidia congenital infection. Here we conducted whole-transcriptome RNA-Seq analyses to identify ncRNAs and regulatory networks for both N. bombycis and host including silkworm embryos and larvae during the microsporidia congenital infection. Results: A total of 4,171 mRNAs, 403 lncRNA, 62 circRNAs, and 284 miRNAs encoded by N. bombycis were identified, among which some differentially expressed genes formed cross-talk and are involved in N. bombycis proliferation and infection. For instance, a lncRNA/circRNA competing endogenous RNA (ceRNA) network including 18 lncRNAs, one circRNA, and 20 miRNAs was constructed to describe 14 key parasites genes regulation, such as polar tube protein 3 (PTP3), ricin-B-lectin, spore wall protein 4 (SWP4), and heat shock protein 90 (HSP90). Regarding host silkworm upon N. bombycis congenital infection, a total of 14,889 mRNAs, 3,038 lncRNAs, 19,039 circRNAs, and 3,413 miRNAs were predicted based on silkworm genome with many differentially expressed coding and non-coding genes during distinct developmental stages. Different species of RNAs form interacting network to modulate silkworm biological processes, such as growth, metamorphosis and immune responses. Melanin is a complex natural pigment that is widely present in fungi. The mushroom Ophiocordyceps sinensis has a variety of pharmacological effects. The active substances of O. sinensis have been extensively studied, but few studies have focused on the O. sinensis melanin. In this study, the production of melanin was increased by adding light or oxidative stress, namely, reactive oxygen species (ROS) or reactive nitrogen species (RNS), during liquid fermentation. Subsequently, the structure of the purified melanin was characterized using elemental analysis, ultraviolet-visible absorption spectrum, Fourier transform infrared (FTIR), electron paramagnetic resonance (EPR), and pyrolysis gas chromatography and mass spectrometry (Py-GCMS). Studies have shown that O. sinensis melanin is composed of C (50.59), H (6.18), O (33.90), N (8.19), and S (1.20), with maximum absorbance at 237 nm and typical melanin structures such as benzene, indole, and pyrrole. Additionally, the various biological activities of O. sinensis melanin have been discovered; it can chelate heavy metals and shows a strong ultraviolet-blocking ability. Moreover, O. sinensis melanin can reduce the levels of intracellular reactive oxygen species and counteract the oxidative damage of H2O2 to cells. Hole is the most common symmetrical geometric structure of mechanical parts, and aperture and hole center distance are important measured dimensions in machining. However, existing visual measurement methods often require high equipment installation accuracy and low measurement accuracy. To solve the problem of projection deformation of the measured hole: Firstly, a local coordinate system is created, and the coordinate plane of the coordinate is parallel to the end face of the measured hole. Secondly, a cone is established, the end face of the measured hole is alike to the bottom face of the cone, and the optical center of the camera is the vertex. The space equation of the cone is represented by the coordinates of the center point of the hole. Finally, a cutting plane is established near the imaging plane of the camera, and the intersection line equation between the plane and the cone is obtained, and the diameter of the measured hole and the coordinates of the center of the circle are obtained through the parameters of the intersection line equation. In order to solve the problem of errors in edge points of the hole, geometric constraints based on the center of the circle are used to improve measurement accuracy. Line-structured light calibration is an important step in the process of the visual measurement of structured light, and in the calibration process, the light plane equation is fitted by the camera coordinates of the data points on the light plane. There is error in solving the spatial coordinates of the data points, and the existing methods generally cannot effectively find the deviation points in the fitted data points, which leads to deviation when solving the parameters of the light plane equation. To correct the error caused by the deviation point in the calibration, this paper established a calibration method based on geometric constraints. In the method, the first constraint was that all fit points involved in the fitting had a certain consistency, and a part of the error points could be filtered by the distance from the data point to the light plane. In order to further improve the calibration accuracy, the laser was moved on a linear guide, the optimized objective function was established by the second geometric constraint, and the constraint was the relationship that the light plane at each position was parallel. In the experiment, a high-precision gauge block was used as the measured object, and the measurement results could be obtained. In order to study the influence of vinyl acetate–ethylene copolymerization emulsions on the bonding performance of fiber and mortar, mortar materials with different polymer contents were prepared. The optimal mix ratio of the matrix was obtained using a pull-out test with a 0° inclination angle. On this basis, polypropylene fibers and alkali-resistant glass fibers were set at different burial depths (6 mm, 12 mm, and 18 mm) and different burial angles (0°, 30°, 45°, and 60°). The load–displacement curves of two types of fibers pulled out from the polymer mortar were obtained. The test results show that polymer contents of 3% and 5% increase the peak pull-out loads of glass fibers and polypropylene fibers by 16.28% and 30.72% and 7.41% and 27.11%, respectively. When the polymer content is 7%, the peak pull-out load decreases by 1.31% and 24.26%, especially for polypropylene fiber, which significantly weakens the bonding performance between the matrix and the fiber. The pull-out load of glass fibers and polypropylene fibers increases with the increase in the buried depth, and both show tensile failure at 18 mm. The molecular mechanism of Chinese cordyceps formation has received a substantial amount of attention because of its usage as traditional Chinese medicine. The formation process of Chinese cordyceps includes two parts: asexual proliferation (Ophiocordyceps sinensis proliferates in the hemolymph of Thitarodes armoricanus larvae) and sexual development (formation and development of fruiting bodies). Therefore, validation of reference genes under different development stages and experimental conditions is crucial for RT-qPCR analysis. However, there is no report on stable reference genes at the development stage of O. sinensis fruiting body. In this study, 10 candidate reference genes, Actin, Cox5, Tef1, Ubi, 18s, Gpd, Rpb1, Try, Tub1 and Tub2, were selected and calculated their expression stability using four methods: geNorm, NormFinder, BestKeeper, and Comparative △Ct. After comprehensive analysis of the results of these four methods with RefFinder, we determined that the most stable reference genes during asexual reproduction of O. sinensis were Tef1 and Tub1, while the most stable reference genes during fruiting body development were Tyr and Cox5, and the most stable reference genes under light-induced conditions were Tyr and Tef1. Our study provides a guidance for reference genes selections at different proliferation processes with light stress of O. sinensis, and represents a foundation for studying the molecular mechanism of Chinese cordyceps formation. Mid‐ocean ridge basalts (MORBs) can record heterogeneity of the upper mantle. Early crystallized minerals and their melt inclusions commonly record greater isotopic variability than whole‐rocks and are powerful tools to investigate the mantle source. In this study, we develop a new in situ Pb analytical protocol for Pb‐poor materials (sub‐ppm level) and apply it to the plagioclase‐hosted melt‐inclusions from IODP Expedition 349 Site U1433, South China Sea (SCS). Together with element mapping, in situ major and trace element analyzes and in situ Sr isotopes of plagioclase phenocrysts, we show that unlike the low and uniform whole‐rock 87Sr/86Sr ratios, the plagioclase phenocrysts record extreme Sr isotopic heterogeneity. Strontium isotopic heterogeneity is observed between crystals even in a single thin section. Based on the high an contents of plagioclase phenocrysts and high Mg# values of their melt inclusions, we propose they crystallized early in the magma chamber and were most likely formed in different batches of mantle‐derived melts. The plagioclase‐hosted melt inclusions show highly variable Pb isotopes, exhibiting a typical linear variation in 207Pb/204Pb versus 206Pb/204Pb. Such a mixing trend indicates an end‐member with highly unradiogenic Pb isotopes. The LREE enrichment and negative correlations between La/Sm and Pb isotopes of the melt inclusions, indicate that ancient cratonic lower continent crustal materials, with low time‐integrated U/Pb ratios, are likely present in the sub‐ridge mantle of the southwest Sub‐basin, SCS. Cordyceps farinosa, an entomopathogenic fungus, infects and leads to high mortality of Thitarodes armoricanus larvae, which die soon after the infection of C. farinose, usually before the colonization of Ophiocordyceps sinensis owing to competitive inhibition and fruiting body formation. Therefore, monitoring C. farinosa in the O. sinensis cultivation environment is critical for minimizing the C. farinosa infection-induced losses. In this study, we initially designed a PCR primer pair (Tar-1F/Tar-1R) through open reading frame prediction and homology comparison of the C. farinosa genome sequence. This primer pair can detect both C. farinosa and Samsoniella hepiali. To further distinguish, primers (ITS5-172/ITS4-95) were then designed to selectively amplify the large ribosomal subunit sequences in the C. farinosa genome. All these primers were applied in combination for detection of C. farinosa in soil samples. The sensitivity reached a detection limit of 1 × 106 spores/g soil. In addition, these primers can detect the presence of C. farinosa in dead T. armoricanus larval samples. The search for hidden minerals with a large, buried depth depends on the guidance of geological and metallogenic theory, but it is difficult to master the geological characteristics of deep deposits. In this paper, taking a survey area as an example, the stimulated polarization geophysical method is used to obtain the information of deep minerals indirectly. Through the analysis and study of IP (induced polarization) anomaly and its characteristics in a tin polymetallic mining area, we delineate the IP anomaly distribution zone of metal ore, identify the distribution of polarized abnormal bodies by IP sounding, and determine the distribution of metallogenic fracture zones in the working area. Through the least square inversion of resistivity and polarizability, the spatial correspondence between the abnormal area of resistivity and polarizability and the location of ore bodies are obtained. Microsporidia are ubiquitous in the environment, infecting almost all invertebrates, vertebrates, and some protists. The microsporidian Nosema bombycis causes silkworms pébrine disease and leads to huge economic losses. Parasite secreted proteins play vital roles in pathogen–host interactions. Serine protease inhibitors (serpins), belonging to the largest and most broadly distributed protease inhibitor superfamily, are also found in Microsporidia. In this study, we characterized 19 serpins (NbSPNs) in N. bombycis; eight of them were predicted with signal peptides. All NbSPN proteins contain a typical conserved serpin (PF00079) domain. The comparative genomic analysis revealed that microsporidia serpins were only found in the genus Nosema. In addition to N. bombycis, a total of 34 serpins were identified in another six species of Nosema including N. antheraeae (11), N. granulosis (8), Nosema sp. YNPr (3), Nosema sp. PM-1 (3), N. apis (4), and N. ceranae (5). Serpin gene duplications in tandem obviously occurred in Nosema antheranae. Notably, the NbSPNs were phylogenetically clustered with serpins from the Chordopoxvirinae, the subfamily of Poxvirus. All 19 NbSPN transcripts were detected in the infected midgut and fat body, while 19 NbSPN genes except for NbSPN12 were found in the transcriptome of the infected silkworm embryonic cell line BmE-SWU1. Nosema bombycis Naegeli (Dissociodihaplophasida: Nosematidae), an obligate intracellular parasite of the silkworm Bombyx mori, causes a devastating disease called pébrine. Every year pébrine will cause huge losses to the sericulture industry worldwide. Until now, there are no effective methods to inhibit the N. bombycis infection in silkworms. In this study, we first applied both the novel protein degradationTrim-Away technology and NSlmb (F-box domain-containing in the N-terminal part of supernumerary limbs from Drosophila melanogaster) to lepidopteran Sf9-III cells to check for specific degradation of a target protein in combination with a single-chain Fv fragment (scFv). Our results showed that the Trim-Away and NSlmb systems are both amenable to Sf9-III cells.We then created transgenic cell lines that overexpressed the protein degradation system and N. bombycis chimeric scFv targeting spore wall protein NbSWP12 and evaluated the effects of the insect transgenic cell lines on the proliferation of N. bombycis. Both methods could be applied to cell lines and bothTrim-Away and NSlmb ubiquitin degradation systems effectively inhibited the proliferation of N. bombycis. Further, either of these degradation systems could be applied to individual silkworms through a transgenic platform, which would yield individual silkworms with high resistance to N. bombycis, thus greatly speeding up the process of acquiring resistant strains. Fe-based single-atom catalysts (SACs) have been recognized as attractive candidates for the oxygen reduction reaction (ORR). However, several limitations such as unsatisfactory intrinsic activity and poor stability hinder their practical applications, motivating further optimization of Fe-SACs which requires precise control of their composition and coordination environment. Herein, we establish a facile solvothermal method to incorporate Cu into Fe-SACs to form heterometallic SACs. We observe that the adjacent Cu atoms can regulate the coordination environment of FeN4 sites and significantly enhance the ORR activity. As a result, the heterometallic SACs show superior ORR catalytic performance compared to the mono-Fe-SAC counterpart (an enhancement factor of ∼5 times), delivering a half-wave potential (E1/2) of 0.889 V vs. RHE and a high kinetic current density (jk) of 4.77 mA cm−2 at 0.9 V vs. RHE, and also outperform the state-of-the-art Pt/C catalyst under identical testing conditions. Sorption-based atmospheric water harvesting has the potential to realize water production anytime, anywhere, but reaching a hundred-gram high water yield in semi-arid climates is still challenging, although state-of-the-art sorbents have been used. Here, we report a portable and modularized water harvester with scalable, low-cost, and lightweight LiCl-based hygroscopic composite (Li-SHC) sorbents. Li-SHC achieves water uptake capacity of 1.18, 1.79, and 2.93 g g−1 at 15%, 30%, and 60% RH, respectively. Importantly, considering the large mismatch between water capture and release rates, a rationally designed batch processing mode is proposed to pursue maximum water yield in a single diurnal cycle. Together with the advanced thermal design, the water harvester shows an exceptional water yield of 311.69 g day−1 and 1.09 g gsorbent−1 day−1 in the semi-arid climate with the extremely low RH of ~15%, demonstrating the adaptability and possibility of achieving large-scale and reliable water production in real scenarios. Background: Recognition of viral invasion by innate antiviral immune system triggers activation of the type I interferon (IFN-I) and proinflammatory signaling pathways. Subsequently, IFN-I induction regulates expression of a group of genes known as IFN-I-stimulated genes (ISGs) to block viral infection. The tripartite motif containing 22 (TRIM22) is an ISG with strong antiviral functions. Results: Here we have shown that the TRIM22 has been strongly upregulated both transcriptionally and translationally upon Zika virus (ZIKV) infection. ZIKV infection is associated with a wide range of clinical manifestations in human from mild to severe symptoms including abnormal fetal brain development. We found that the antiviral function of TRIM22 plays a crucial role in counterattacking ZIKV infection. Overexpression of TRIM22 protein inhibited ZIKV growth whereas deletion of TRIM22 in host cells increased ZIKV infectivity. Mechanistically, TRIM22, as a functional E3 ubiquitin ligase, promoted the ubiquitination and degradation of ZIKV nonstructural protein 1 (NS1) and nonstructural protein 3 (NS3). Further studies showed that the SPRY domain and Ring domain of TRIM22 played important roles in protein interaction and degradation, respectively. In addition, we found that TRIM22 also inhibited other flaviviruses infection including dengue virus (DENV) and yellow fever virus (YFV). Nosema bombycis is a destructive and specific intracellular parasite of silkworm, which is extremely harmful to the silkworm industry. N. bombycis is considered as a quarantine pathogen of sericulture because of its long incubation period and horizontal and vertical transmission. Herein, two single-chain antibodies targeting N. bombycis hexokinase (NbHK) were cloned and expressed in fusion with the N-terminal of Slmb (a Drosophila melanogaster FBP), which contains the F-box domain. Western blotting demonstrated that Sf9-III cells expressed NSlmb–scFv-7A and NSlmb–scFv-6H, which recognized native NbHK. Subsequently, the NbHK was degraded by host ubiquitination system. When challenged with N. bombycis, the transfected Sf9-III cells exhibited better resistance relative to the controls, demonstrating that NbHK is a prospective target for parasite controls and this approach represents a potential solution for constructing N. bombycis-resistant Bombyx mori. Due to the high specific capacity and energy density, lithium-sulfur batteries have been recognized as the most promising energy storage devices for electric vehicles. However, they are also faced with some problems, such as poor electronic conductivity and severe polysulfide migration. In this work, hierarchical porous carbons (HPCs) are prepared and developed as sulfur host materials in the lithium-sulfur batteries. Due to the hierarchical porous structure, the element sulfur can be uniformly dispersed into the HPCs. Besides, the unique structure also can promote the electron transport and inhibit the polysulfide migration. Based on these advantages, the as-prepared HCPs/S cathode exhibits superior electrochemical performance, including high capacity, excellent rate capability, and stable long-term cycling performance. The greenhouse effect and plastic pollution caused by the accumulation of plastics have led to a global concern for environmental protection, as well as the development and application of biodegradable materials. Polypropylene carbonate (PPC) is a biodegradable polymer with the function of "carbon sequestration", which has the potential to mitigate the greenhouse effect and the plastic crisis. It has the advantages of good ductility, oxygen barrier and biocompatibility. However, the mechanical and thermal properties of PPC are poor, especially the low thermal degradation temperature, which limits its industrial use. In order to overcome this problem, PPC can be modified using environmentally friendly materials, which can also reduce the cost of PPC-based products to a certain extent and enhance their competitiveness in terms of improving their mechanical and thermal properties. In this paper, we present different perspectives on the synthesis, properties, degradation, modification and post-modification applications of PPC. The modification part mainly introduces the influence of inorganic materials, natural polymer materials and degradable polymers on the performance of PPC. Continuous outbreaks of viral diseases in humans facilitates a need for the rapid development of viral test kits and vaccines. These require expression systems to produce a pure and high yield of target viral proteins. We utilized a baculovirus–silkworm expression system to produce the receptor binding domain (RBD) of the SARS-CoV-2 spike protein. First, we had to develop a strategy for constructing a recombinant baculovirus for RBD expression. For this, the coding region of the Bombyx mori cypovirus (BmCPV) polyhedron was assembled with the Bombyx mori nuclear polyhedrosis virus (BmNPV) promoter. We demonstrated that the recombinant baculovirus has the ability to form polyhedrons within host silkworm cells. In addition, the encapsulated BVs are able to infect silkworms by ingestion and induce foreign protein expression. In this way, we utilized this novel system to obtain a high yield of the target foreign protein, the RBD of the SARS-CoV-2 S protein. However, the viral infection rate of our recombinant BV needs to be improved. The baculovirus vector expression system is a well-established tool for foreign protein production and gene delivery. In this study, we constructed a recombinant baculovirus vector system. The UAS promotor region and Bombyx mori nucleopolyhedrovirus (BmNPV) polyhedrin coding region were ligated into a pFastBac Dual vector to obtain a BmBac-UPS recombinant bacmid. The recombinant bacmid BmBac-Gal4 was generated by the same strategy which has a Gal4 coding region controlled by the IE2 promoter. BmBac-UPS and BmBac-IGal4 were co-infected into silkworm BmN cells to confirm the ability of the UAS/Gal4 system to form polyhedrons in B. mori cells. Rational design of host materials for element sulfur is critical in the lithium-sulfur batteries to stabilize the polysulfide, which could migrate from the cathode to the anode, leading to the so-called shuttle effect. Herein, we develop a method to prepare the suitable sulfur host materials for the lithium-sulfur batteries. The manganese dioxide nanospheres are successfully prepared and designed as sulfur host in the lithium-sulfur batteries. The as-prepared manganese dioxide/S composites could be formed flexible and self-supported electrode without the current collector. Background: Nosema bombycis is a unicellular eukaryotic pathogen of the silkworm, Bombyx mori, and is an economic and occupational hazard in the silkworm industry. Because of its long incubation period and horizontal and vertical transmission, it is subject to quarantine measures in sericulture production. The microsporidian life-cycle includes a dormant extracellular phase and intracellular proliferation phase, with the proliferation period being the most active period. Methods: In order to find suitable target for the selective breeding of N. bombycis-resistant silkworm strains, we screen highly expressed membrane proteins from the transcriptome data of N. bombycis. The subcellular localization of the candidate protein was verified by Indirect immunofluorescence analysis (IFA) and immunoelectron microscopy (IEM), and its role in N. bombycis proliferation was verified by RNAi. Results: The N. bombycis protein (NBO_76g0014) was identified as a transmembrane protein and named NbTMP1. It is homologous with hypothetical proteins NGRA_1734 from Nosema granulosis. NbTMP1 has a transmembrane region of 23 amino acids at the N-terminus. Indirect immunofluorescence analysis (IFA) results suggest that NbTMP1 is secreted on the plasma membrane as the spores develop. Western blot and qRT-PCR analysis showed that NbTMP1 was expressed in all developmental stages of N. bombycis in infected cells and in the silkworm midgut. Downregulation of NbTMP1 expression resulted in significant inhibition of N. bombycis proliferation. Conclusions: We confirmed that NbTMP1 is a membrane protein of N. bombycis. Reduction of the transcription level of NbTMP1 significantly inhibited N. bombycis proliferation, and this protein may be a target for the selective breeding of N. bombycis-resistant silkworm strains. The rechargeable lithium-sulfur battery is an attractive energy storage device due to the high specific capacity and high energy density. The wide application of lithium-sulfur batteries, however, is hindered owing to the rapid capacity fading caused by the polysulfide dissolution and migration between the cathode and anode side. Herein, we demonstrate a new strategy to address the severe problem of polysulfide migration during the electrochemical cycles by designing titanium carbide nanosheets/S hybrid composites. In this unique hybrid structure, the titanium carbide nanosheets could provide fast lithium-ion and electron paths and keep structural stability during the cycles. Moreover, the space of the titanium carbide nanosheets acts as binding sites for trapping polysulfide. As a result, the hybrid titanium carbide nanosheets/S composites exhibit high performance with specific capacity, rate capability, and cycling stability. [ABSTRACT FROM AUTHOR]

Trough profile is an important structural parameter of the spiral concentrator. However, limited attention has been focused on the influence of trough profile on coal separation. In the present work, 21 sets of experiments were carried out to investigate the impact of operating parameters and trough profile on the separation performance. Results show that particles tend to transport outward with the increase of the flow rate and the solid concentration. The cut specific gravity of separation (SG50) increases with the flow rate and the solid concentration. The SG50 of the spiral concentrator with the synthetic curve is much lower than that of spiral concentrator with the ellipse or cubic parabola. Light particles prefer to be placed in the outer place of the spiral concentrator with the trough profile of ellipse. Heavy particles tend to report to the inner region of the spiral concentrator with the trough profile of cubic parabola. The spiral concentrator, with the trough profile of cubic parabola in inner place and the ellipse in outer place, is more desirable to separate the coal slime used in this paper. The emergency use authorization of two mRNA vaccines in less than a year from the emergence of SARS-CoV-2 represents a landmark in vaccinology1,2. Yet, how mRNA vaccines stimulate the immune system to elicit protective immune responses is unknown. Here we used a systems vaccinology approach to comprehensively profile the innate and adaptive immune responses of 56 healthy volunteers who were vaccinated with the Pfizer–BioNTech mRNA vaccine (BNT162b2). Vaccination resulted in the robust production of neutralizing antibodies against the wild-type SARS-CoV-2 (derived from 2019-nCOV/USA_WA1/2020) and, to a lesser extent, the B.1.351 strain, as well as significant increases in antigen-specific polyfunctional CD4 and CD8 T cells after the second dose. Booster vaccination stimulated a notably enhanced innate immune response as compared to primary vaccination, evidenced by (1) a greater frequency of CD14+CD16+ inflammatory monocytes; (2) a higher concentration of plasma IFNγ; and (3) a transcriptional signature of innate antiviral immunity. Consistent with these observations, our single-cell transcriptomics analysis demonstrated an approximately 100-fold increase in the frequency of a myeloid cell cluster enriched in interferon-response transcription factors and reduced in AP-1 transcription factors, after secondary immunization. Finally, we identified distinct innate pathways associated with CD8 T cell and neutralizing antibody responses, and show that a monocyte-related signature correlates with the neutralizing antibody response against the B.1.351 variant. Collectively, these data provide insights into the immune responses induced by mRNA vaccination and demonstrate its capacity to prime the innate immune system to mount a more potent response after booster immunization.Profiling the immune responses of 56 volunteers vaccinated with BNT162b2 reveals how this mRNA vaccine primes the innate immune system to mount a potent response to SARS-CoV-2 after booster immunization. Background: Nosema bombycis is a unicellular eukaryotic pathogen of the silkworm, Bombyx mori, and is an economic and occupational hazard in the silkworm industry. Because of its long incubation period and horizontal and vertical transmission, it is subject to quarantine measures in sericulture production. The microsporidian life-cycle includes a dormant extracellular phase and intracellular proliferation phase, with the proliferation period being the most active period. Background: Reducing peritoneal recurrence after radical surgery is an important choice to improve the prognosis of patients with advanced gastric cancer. Intraoperative intraperitoneal chemotherapy has the potential to be a promising treatment strategy. In the present study, we conducted a multi-center, randomized, controlled clinical study to evaluate the efficacy and safety of intraoperative intraperitoneal chemotherapy using sustained-release fluorouracil implants plus radical gastrectomy and adjuvant chemotherapy for cTNM stage III gastric cancer. Methods: The patients were randomized into intraperitoneal chemotherapy group (sustained-release fluorouracil implants administration after standard D2 radical gastrectomy, and followed by XELOX adjuvant chemotherapy) and control group (standard D2 radical gastrectomy, and followed by XELOX adjuvant chemotherapy). A total of 122 patients from three centers were enrolled from September 2015 to February 2017. Results: One hundred and two eligible patients completed the treatment course. The median follow-up time was 41.7 months (36.1–52.9 months). The 3-year progression-free survival rate and overall survival of patients in the intraperitoneal chemotherapy group were 43.9% and 49.1%, respectively, which were significantly better than those of the control group, 31.0% and 38.4%. In the intraperitoneal chemotherapy group, the number of cases with peritoneal recurrence was significantly less than that of the control group, 9 cases (17.3%) vs. 19 cases (44.2%). There were neither significant differences between the groups in the incidence of hematogenous metastasis, lymph node metastasis, nor local metastasis. Conclusion: For cTNM stage III gastric cancer, intraoperative sustained-release fluorouracil implants after radical resection combined with postoperative adjuvant chemotherapy, could significantly reduce the risk of peritoneal recurrence and prolong PFS.
