The aim of this paper is to study and establish precise asymptotics for complete integral convergence in the law of the logarithm under the sub-linear expectation space. The methods and tools in this paper are different from those used to study precise asymptotics theorems in probability space. We extend precise asymptotics for complete integral convergence from the classical probability space to sub-linear expectation space. It is essential to explore the relationship between drugs and transporters in the process of drug development. Strong background signals in nonhuman MDCK or LLC-PK1 cells and overlapping interference of inhibitors or RNAi in human Caco-2 cells mean that an ideal alternative could be to knock out specific transporter genes in Caco-2 cells. However, the application of gene knockout (KO) to Caco-2 cells is challenging because it is still inefficient to obtain rapidly growing Caco-2 subclones with double-allele KO through long-term monoclonal cultivation. Herein, CRISPR/Cas9, a low cost but more efficient and precise gene editing technology, was utilized to singly or doubly knockout the P-gp, BCRP, and MRP2 genes in Caco-2 cells. By combining this with single cell expansion, rapidly growing transporter-deficient subclones were successfully screened and established. Bidirectional transport assays with probe substrates and three protease inhibitors indicated that more reliable and detailed data could be drawn easily with these KO Caco-2 models. The six robust KO Caco-2 subclones could contribute to efficient in vitro drug transport research. [ABSTRACT FROM AUTHOR]

Enterprise BIM aims to optimise business management and asset management throughout the lifecycle of buildings and infrastructures. This follow-up study of the Enterprise BIM concept analyses the potential for international standardisation, mainly focusing on the contribution of the ISO 19650 standard series to further development. The study explores how key ISO 19650 concepts related to different types of information requirements, levels of information need, and common data environments can help strengthen Enterprise BIM. As such, the paper presents an in-depth analysis of how the ISO 19650 series and several adjacent standards can provide a further means of facilitating information management within the Enterprise BIM concept. The study uses a customised Zachman framework by adding the necessary perspectives from ISO 19650 and Enterprise BIM. The paper adds increased knowledge related to understanding the standard series parts 1, 2 and 3 and showing how they can be implemented in an enterprise structure. Based on the ISO 19650 framework, the study emphasises the importance of linking the information requirements to the various information needs of multiple end-users. At the same time, the paper provides new knowledge by showing that the information flow in the operational phase is far more nuanced than in the delivery phase, suggesting that information management must be considered in a completely different light from what previous research shows. 8. Size-controllable carbon spheres doped Ni (II) for enhancing the catalytic oxidation of methanol.

Ni(II)/CSs were prepared using a simple two-step hydrothermal method. The morphology and composition of the catalysts were studied with scanning electron microscope, transmission electron microscope, and X-ray diffraction. Fourier transform infrared spectroscopy and X-ray photoelectron spectroscopy showed that the surface of the prepared carbon spheres was rich in hydroxyl groups, which was beneficial to remove CO intermediates, and therefore, improving the catalytic efficiency and the antipoisoning ability of the catalysts. The results of cyclic voltammetry and chronoamperometry showed that the electrocatalytic activity and stability of Ni(II)/CSs were higher than that of unloaded NiAc under alkaline environment. When the nickel content was 5 wt.%, the peak oxidation current density of methanol on Ni(II)/CSs electrocatalyst reached the maximum of 34.54 mA/cm2, which was about 1.8 times that of unloaded NiAc. These results indicate that Ni(II)/CSs has potential applications in the electrocatalytic oxidation of methanol. [ABSTRACT FROM AUTHOR]

It is well known that both the mutation and integration of the Hepatitis B virus (HBV) are of great significance in liver cancer, however, the relationship between mutation and integration is still unclear. In the present study, sequencing data from 426 previously published samples were analyzed and 5374 specific HBV mutations in cancer tissues were discovered. By comparing integrated samples and non-integrated samples, we found that the integrated samples had higher sample single nucleotide variants (SNVs) positive rates and SNV numbers, as well as higher sample frequency of SNV in the X region of the HBV genome. Samples with HBV integration in the telomerase reverse transcriptase (TERT) region showed higher SNV positive rates and numbers than samples without integration. Moreover, the SNVs (209 [T>G] and 531 [T>C; T>G]) were seen with higher frequency in samples with integration in the TERT region. Our study showed that the occurrence of viral integration events is closely related to the occurrence of SNV, and SNV in the X region should be more directly associated with viral integration. The present study provides an initial exploration of the relationship between HBV mutation and integration to help improve our understanding of the relationship between viral integration and mutation. 10. Comprehensive analysis of long non-coding RNAs and mRNAs in skeletal muscle of diabetic Goto-Kakizaki rats during the early stage of type 2 diabetes.

Skeletal muscle long non-coding RNAs (lncRNAs) were reported to be involved in the development of type 2 diabetes (T2D). However, little is known about the mechanism of skeletal muscle lncRNAs on hyperglycemia of diabetic Goto-Kakizaki (GK) rats at the age of 3 and 4 weeks. To elucidate this, we used RNA-sequencing to profile the skeletal muscle transcriptomes including lncRNAs and mRNAs, in diabetic GK and control Wistar rats at the age of 3 and 4 weeks. In total, there were 438 differentially expressed mRNAs (DEGs) and 401 differentially expressed lncRNAs (DELs) in skeletal muscle of 3-week-old GK rats compared with age-matched Wistar rats, and 1000 DEGs and 726 DELs between GK rats and Wistar rats at 4 weeks of age. The protein-protein interaction analysis of overlapping DEGs between 3 and 4 weeks, the correlation analysis of DELs and DEGs, as well as the prediction of target DEGs of DELs showed that these DEGs (Pdk4, Stc2, Il15, Fbxw7 and Ucp3) might play key roles in hyperglycemia, glucose intolerance, and increased fatty acid oxidation. Considering the corresponding co-expressed DELs with high correlation coefficients or targeted DELs of these DEGs, our study indicated that these dysregulated lncRNA-mRNA pairs (NONRATG017315.2-Pdk4, NONRATG003318.2-Stc2, NONRATG011882.2-Il15, NONRATG013497.2-Fbxw7, MSTRG.1662-Ucp3) might be related to above biological processes in GK rats at the age of 3 and 4 weeks. Our study could provide more comprehensive knowledge of mRNAs and lncRNAs in skeletal muscle of GK rats at 3 and 4 weeks of age. 11. Transcriptomic responses of hypothalamus to acute exercise in type 2 diabetic Goto-Kakizaki rats.

The hypothalamus has an integral role in energy homeostasis regulation, and its dys-functions lead to the development of type 2 diabetes (T2D). Physical activity positively affects the prevention and treatment of T2D. However, there is not much information on the adaptive mechanisms of the hypothalamus. In this study, RNA sequencing was used to determine how acute exercise affects hypothalamic transcriptome from both type 2 diabetic Goto-Kakizaki (GK) and control Wistar rats with or without a single session of running (15 m/min for 60 min). Through pairwise comparisons, we identified 957 differentially expressed genes (DEGs), of which 726, 197, and 98 genes were found between GK and Wistar, exercised GK and GK, and exercised Wistar and Wistar, respectively. The results of Gene Ontology and Kyoto Encyclopedia of Genes and Genomes pathway enrichment revealed that lipid metabolism-related terms and pathways were enriched in GK and exercised GK rats, and nervous system related terms and pathways were enriched in exercised GK and Wistar rats. Furthermore, 45 DEGs were associated with T2D and related phenotypes according to the annotations in the Rat Genome Database. Among these 45 DEGs, several genes (Plin2, Cd36, Lpl, Wfs1, Cck) related to lipid metabolism or the nervous system are associated with the exercise-induced benefits in the hypothalamus of GK rats. 12. Improved Defined Approaches for Predicting Skin Sensitization Hazard and Potency in Humans.

Since the EU banned animal testing for cosmetic products and ingredients in 2013, many defined approaches (DA) for skin sensitization assessment have been developed. Machine learning models were shown to be effective in DAs, but the predictivity might be affected by data imbalance (i.e., more sensitizers than non-sensitizers) and limited information in the databases. To improve the predictivity of DAs, we attempted to apply data-rebalancing ensemble learning (bagging with support vector machine (SVM)) and a novel and comprehensive Cosmetics Europe database. For predicting human hazard and three-class potency, 12 models were built for each using a training set of 96 substances and a test set of 32 substances from the database. The model that predicted hazard with the highest accuracy (90.63% for the test set and 88.54% for the training set, named hazard-DA) used SVM-bagging with combinations of all variables (V6), while the model that predicted potency with the highest accuracy (68.75% for the test set and 82.29% for the training set, named potency-DA) used SVM alone. Both DAs showed better performance than LLNA and other machine learning-based DAs, and the potency-DA provided more in-depth assessment. These findings indicate that SVM-bagging-based DAs provide enhanced predictivity for hazard assessment by further data rebalancing. Meanwhile, the effect of imbalanced data might be offset by more detailed categorization of sensitizers for potency assessment, thus SVM-based DA without bagging could provide sufficient predictivity. Objective(s): microRNA-29 (miR-29) family miRNAs have been mentioned as tumor suppressive genes in several human cancers. The purpose of this study was to investigate the function of miR-29a in nasopharyngeal carcinoma (NPC) cells. Materials and Methods: Human NPC cell line 5-8F was transfected with mimic, inhibitor or scrambled controls specific for miR-29a. Subsequently, cell viability, migration, apoptosis and expression changes of VEGF were assessed by trypan blue staining, MTT assay, transwell assay, flow cytometry, Western blot and RT-qPCR. TargetScan online database was used to predict the targets of miR-29a, and luciferase reporter assay was carried out for testing the targeting relationship between VEGF and miR-29a. Western blot analysis was performed to determine the expression changes of core proteins in PI3K/AKT and JAK/STAT pathways. Results: Overexpression of miR-29a suppressed 5-8F cells viability and relative migration, but increased apoptotic cell rate. Consistently, Bcl-2 was downregulated, Bax was upregulated, and caspase-3 and -9 were cleaved by miR-29a overexpression. VEGF was a target gene of miR-29a. Besides, VEGF silence exerted similar effects like miR-29a, as the viability and migration were repressed and apoptosis was induced. Finally, we found that PI3K/AKT and JAK/STAT pathways were deactivated by miR-29a or VEGF silence. Conclusion: These findings highlighted the tumor suppressive effects of miR-29a on NPC cells, as its overexpression inhibited 5-8F cells viability, migration, and induced apoptosis. miR-29a exerted tumor suppressive functions might be via targeting VEGF and deactivating PI3K/AKT and JAK/STAT pathways. Compared with cylindrical hohlraums, the octahedral spherical hohlraums have natural superiority in maintaining high radiation symmetry during the whole capsule implosion process in indirect drive inertial confinement fusion. However, the narrow space between laser beams and the hohlraum wall may disturb laser propagation inside the spherical hohlraum. In this work, the laser propagation inside the spherical hohlraum and cylindrical hohlraum is investigated experimentally by measuring laser spot movement at the SGIII-prototype laser facility. The experimental results show that the laser propagations inside the spherical hohlraum and the cylindrical hohlraum are totally different from each other due to different hohlraum structures. For the spherical hohlraum, although the laser energy is mainly deposited in the initial position of the laser spot during the whole laser pulse, some laser energies are absorbed by the ablated plasmas from the hohlraum wall. Because the laser beam is refracted by the thin plasmas near the laser entrance hole (LEH) region, the laser spot in the spherical hohlraum moves toward the opposite LEH. In contrast, the laser spot in the cylindrical hohlraum moves toward the LEH along the laser path due to the plasma expansion. When the laser is to be turned off, the accumulated plasmas near the LEH region in the cylindrical hohlraum absorb a majority of laser energy and hinder the laser arriving at the appointed position on the hohlraum wall. Porcine pluripotent cells with the capacity to generate germ line chimeras have not been developed yet. The transcription factor Oct-4 is an important marker of undifferentiating status and a central regulator of pluripotency in cells. Establishment of an Oct-4 promoter-based reporter system, such as that used in mice, will be a useful tool for monitoring the differentiating statuses of porcine cells both in vivo and in vitro. In the present study, we constructed a vector, pOGN2, in which enhanced green fluorescent protein ( EGFP) was driven by the porcine Oct-4 promoter. In pigs containing this vector, EGFP was expected to be specifically expressed in pluripotent cells. We delivered the vectors into porcine fetal fibroblasts (PEFs) using liposomes. After transfected PEFs were selected with G418, we established eight cell lines containing the pOGN2 vector. When transgenic cells were used as donor nuclei to make somatic cell nuclear transfer (SCNT) embryos, SCNT embryos derived from four transgenic cell lines expressed green fluorescence. When PEFs with pOGN2 vectors were infected with retroviral vectors encoding the four transcription factors ( Oct-4, Sox2, Klf4, and c-Myc), EGFP-expressing iPS cell colonies were observed at day 20. This work lays a foundation that can be used to generate a pig strain with an Oct4-EGFP reporter system, which would be greatly helpful in studying the differentiating and reprogramming mechanisms of pig embryos. Air-stable n-type field effect transistors were fabricated with an axially oxygen substituted metal phthalocyanine, tin (IV) phthalocyanine oxide (SnOPc), as active layers. The SnOPc thin films showed highly crystallinity on modified dielectric layer, and the electron field-effect mobility reached 0.44 cm
2 V-1 s-1 . After storage in air for 32 days, the mobility and on/off ratio did not obviously change. The above results also indicated that it is an effective approach of seeking n-type semiconductor by incorporating the appropriate metal connected with electron-withdrawing group into π-π conjugated system.
