The aim of the presented study was to compare the diversity and composition of fungal communities associated with the roots and the rhizosphere soil of P. abies and Larix sp. in mid-age and mature managed forest stands in Lithuania. We also aimed to assess the presence of fungi–host-specific associations, i.e., whether Larix sp. stands could provide habitats for soil fungi currently associated with P. abies. The study sites were 10 Larix sp. and 10 P. abies forest stands in Lithuania. For the study, 100 root samples and 10 organic and 10 mineral soil samples were collected in P. abies stands as well as the same number in Larix sp. stands, and DNA was isolated, amplified using ITS2 rDNA as a marker and subjected to high-throughput sequencing. The results showed that the Shannon diversity index of fungal communities was similar between the two tree species when compared either between root (H = 4.26 P. abies and H = 3.82 Larix sp.), organic soil (H = 5.12 P. abies and H = 5.13 Larix sp.) or mineral soil (H = 4.71 P. abies and H = 4.29 Larix sp.) samples. Multivariate analysis showed that the fungal community composition in the organic and mineral soil samples of both P. abies and Larix sp. were similar, and thus, overlapping. The analysis also showed that the distribution of fungal species was denser in the roots and organic soil but more scattered in mineral soil. Simple Summary: The nun moth (Lymantria monacha L.), a defoliator of conifers and broad-leaved trees, is expanding its range, and outbreaks are increasingly occurring in the forests of central and eastern Europe. The only way to control and eradicate mass outbreaks of the pest is aerial spraying with a biological insecticide, Foray 76B. However, knowledge of variations in non-target insect assemblages following a treatment is limited. The present study aimed to determine the effect of the treatment on the diversity of non-target insects in Pinus sylvestris stands in three regions of Lithuania in the year following a nun moth outbreak. The treatment was found to influence the diversity of psyllids, ants, and beetles. Treated pine forests near the Baltic Sea exhibited lower insect species richness and a decreased relative abundance of beetles on the forest floor. The spraying influenced a reduction in the relative abundance of Carabus arcensis in the forests in the southern part of the country. The treatment also influenced the movement of ants from the tree canopy to the forest floor at all studied locations. Outbreaks of Lymantria monacha are of great concern, as their occurrence is predicted to become more intense and frequent due to a warming climate. A frequent treatment to control mass outbreaks of the pest is with the bioinsecticide Foray 76B. However, knowledge of how this treatment affects non-target insect species is limited. We surveyed the assemblages of non-target epigeal and arboreal insects in Pinus sylvestris forests in the year following bioinsecticide application. A collection of insects using sweep nets and pitfall traps was carried out in L. monacha-infested pine stands, (i) treated with Foray 76B and (ii) untreated, in three regions of Lithuania from May to October 2021. The results revealed that, in Neringa forests, species richness of the epigeal insects was lower in treated than in untreated sampling plots, with 36 and 41 different insect species, respectively. The relative abundance of epigeal Coleoptera in treated plots was 3.6%, while in untreated it was 53.2%. There was a significant decrease in the relative abundance of Carabus arcencis in Kapčiamiestis (by 7.4%) and Marcinkonys (by 16.7%). Simple Summary: Here we investigated fungal communities associated with three species of siricid wood wasps occurring in Lithuania. Siricid wood wasps are pests of weakened forest trees or develop on the stem of dead trees, which may also vector tree-pathogenic fungi. The results demonstrated that, generally, species–specific fungal communities are associated with each species of wood wasps, and that the economically important tree pathogen Amylostereum chailletii was among the most common fungal taxon associated. The study, therefore, provided new insights into the ecology and symbiotic associations of these wood wasps. We investigated the diversity and occurrence of wood wasps in Lithuania and determined communities of associated fungi. Trapping of wood wasps resulted in three different species, including Sirex juvencus, Urocerus gigas, and Tremex fuscicornis. Fungal culturing from adult females of T. fuscicornis mainly resulted in fungi from the genera Penicillium and Trichoderma. High-throughput sequencing of ITS2 rDNA resulted in 59,797 high-quality fungal sequences, representing 127 fungal OTUs. There were 93 fungal OTUs detected in U. gigas, 66 in S. juvencus, and 10 in T. fuscicornis. The most common fungi were Fusarium sporotrichioides (63.1% of all fungal sequences), Amylostereum chailletii (14.9%), Penicillium crustosum (7.8%), Microascus sp. 2261_4 (5.0%), and Pithoascus ater (2.1%). Among these, only A. chailletii was found in all three insect species with the highest relative abundance in U. gigas (15.2%), followed by S. juvencus (7.7%), and the lowest in T. fuscicornis (0.3%) (p < 0.0003). Correspondence analysis of fungal communities showed a distant placement of different species of wood wasps, indicating that fungal communities in each of these were largely different. We assessed the diversity and composition of fungal communities in different functional tissues and the rhizosphere soil of Pinus sylvestris and Picea abies stands along the latitudinal gradient of these tree species distributions in Europe to model possible changes in fungal communities imposed by climate change. For each tree species, living needles, shoots, roots, and the rhizosphere soil were sampled and subjected to high-throughput sequencing. Results showed that the latitude and the host tree species had a limited effect on the diversity and composition of fungal communities, which were largely explained by the environmental variables of each site and the substrate they colonize. The mean annual temperature and mean annual precipitation had a strong effect on root fungal communities, isothermality on needle fungal communities, mean temperature of the warmest quarter and precipitation of the driest month on shoot fungal communities, and precipitation seasonality on soil fungal communities. Fungal communities of both tree species are predicted to shift to habitats with a lower annual temperature amplitude and with increasing precipitation during the driest month, but the suitability of these habitats as compared to the present conditions is predicted to decrease in the future. The aim of the present work was to study seasonal and site-specific patterns of airborne fungal diversity, focusing on plant pathogens. The sampling of fungal spores was carried out for twelve months, i.e., between September 2017 and August 2018, using passive spore traps that were placed at three different sites in western (Lenkimai), central (Dubrava), and eastern (Labanoras) Lithuania. Samples were collected every 7–10 days, resulting in 146 samples altogether. Following DNA isolation, samples were individually amplified using ITS2 rRNA as a marker and subjected to high-throughput sequencing. Clustering and taxonomic classification of 283,006 high-quality reads showed the presence of 805 non-singleton fungal taxa. The detected fungi were 53.4% Ascomycota, 46.5% Basidiomycota, and 0.1% Mucoromycota. The most common fungal taxon at Labanoras and Lenkimai was Hannaella coprosmae (23.2% and 24.3% of all high-quality fungal sequences, respectively), while at Dubrava it was Cladosporium macrocarpum (16.0%). In different sites, plant pathogenic fungi constituted between 1.6% and 14.6% of all fungal taxa and among these the most common were Protomyces inouyei (4.6%) and Sydowia polyspora (1.9%). The results demonstrated that the diversity of airborne fungi was mainly determined by the surrounding vegetation and climatic factors, while the occurrence of pathogenic fungi was affected by the availability of their hosts. The aim of this study was to investigate fungal communities associated with leaves and roots of healthy-looking and declining U. glabra trees. The study was expected to demonstrate whether and how the diversity and composition of fungal communities change in these functional tissues following the infection by Dutch elm disease-causing fungi. The study sites included six U. glabra sites in Lithuania, where leaves and roots were sampled. DNA was isolated from individual samples, amplified using ITS2 rRNA as a marker, and subjected to high-throughput sequencing. The sequence analysis showed the presence of 32,699 high-quality reads, which following clustering, were found to represent 520 non-singleton fungal taxa. In leaves, the fungal species richness was significantly higher in healthy-looking trees than in diseased ones (p < 0.05). In roots, a similar comparison showed that the difference was insignificant (p > 0.05). The most common fungi in all samples of roots were Trichocladium griseum (32.9%), Penicillium restrictum (21.2%), and Unidentified sp. 5238_7 (12.6%). The most common fungi in all samples of leaves were Trichomerium sp. 5238_8 (12.30%), Aureobasidium pullulans (12.03%), Cladosporium sp. 5238_5 (11.73%), and Vishniacozyma carnescens (9.86%). The results showed that the detected richness of fungal taxa was higher in samples collected from healthy-looking trees than from diseased ones, thereby highlighting the negative impact of the Dutch elm disease on the overall fungal diversity. This study aimed to assess the changes in chemical properties and fungal communities in the upper mineral soil layer in managed Scots pine (Pinus sylvestris L.) ecosystems. Study sites were located in the three largest P. sylvestris massifs in Lithuania, and six sampling sites, representing different development stages of the P. sylvestris forest ecosystem, were selected in each of them: mature P. sylvestris forest stands; clear-cuts of former P. sylvestris mature stand; and the P. sylvestris plantations aging from the 1st to 4th year. High-throughput sequencing was performed to evaluate the soil fungus community at clear-cuts, early-stage reforested sites, and mature forests in Lithuania. This study has shown that, among other chemical soil parameters, the mean concentrations of mineral nitrogen (N), total phosphorus (P), and P2O5 were slightly higher in the clear-cut sites, and significantly higher in the 1st year plantations compared to the mature forests. The quality filtering after PacBio sequencing showed the presence of 60,898 high-quality fungal sequences, and 1143 fungal operational taxonomic units (OTUs). The most abundant fungal OTU in our study was Archaeorhizomyces sp. 5425_1. In total, 70 mycorrhizal fungal OTUs were found in the soil samples at the studied sites. The most abundant ectomycorrhizal fungus identified was Amanita fulva (Schaeff.) Fr. The highest amount of ectomycorrhizal fungal OTUs was found in the clear-cut sites and in the mature forests. The concentrations of mineral N and P2O5 in the upper mineral soil layer did not significantly affect fungal OTUs diversity. Conversely, a relatively strong correlation was obtained between the number of mycorrhizal fungal OTUs and the concentrations of total N and soil organic carbon (SOC), as well as between the numbers of saprotrophic fungal OTUs and the concentration of magnesium ions (Mg2+). The plant- and soil-associated microbial communities are critical to plant health and their resilience to stressors, such as drought, pathogens, and pest outbreaks. A better understanding of the structure of microbial communities and how they are affected by different environmental factors is needed to predict and manage ecosystem responses to climate change. In this study, we carried out a country-wide analysis of fungal communities associated with Pinus sylvestris growing under different environmental conditions. Needle, shoot, root, mineral, and organic soil samples were collected at 30 sites. By interconnecting the high-throughput sequencing data, environmental variables, and soil chemical properties, we were able to identify key factors that drive the diversity and composition of fungal communities associated with P. sylvestris. The fungal species richness and community composition were also found to be highly dependent on the site and the substrate they colonize. The results demonstrated that different functional tissues and the rhizosphere soil of P. sylvestris are associated with diverse fungal communities, which are driven by a combination of climatic (temperature and precipitation) and edaphic factors (soil pH), and stand characteristics. Fifty-nine fungal taxa, isolated from re-emerging Fraxinus excelsior sites in Lithuania, were in vitro tested against three strains of Hymenoscyphus fraxineus on agar media to establish their biocontrol properties. All tested fungi were isolated from leaves and shoots of relatively healthy Fraxinus excelsior trees (<30% defoliation), which were affected by ash dieback but their phytosanitary condition has not worsened during the last decade. The inhibition of H. fraxineus growth by tested fungal taxa ranged between 16–87%. Occasionally isolated fungal taxa such as Neonectria coccinea, Nothophorma quercina, and Phaeosphaeria caricis were among the most effective fungi inhibiting the growth of H. fraxineus cultures. Among the more commonly isolated fungal taxa, Cladosporium sp., Fusarium sp., Malassezia sp., and Aureobasidium pullulans showed a strong growth inhibition of H. fraxineus. [ABSTRACT FROM AUTHOR]

In Lithuania, the dieback of European ash (Fraxinus excelsior L.), caused by alien ascomycete Hymenoscyphus fraxineus, started in the mid-1990s, resulting in a large-scale decline of F. excelsior and its dominated forest habitats. Nevertheless, the recent inventories show the presence of several hundred hectares of naturally regenerated F. excelsior stands. We used seven naturally regenerated sites and three planted progeny trials of F. excelsior to collect leaves, shoots, roots, and the surrounding soil to study ash-associated fungal communities based on high-throughput sequencing. Results showed that fungal communities associated with F. excelsior in re-emerging stands in post-dieback areas were composed of 1487 fungal taxa. Among these, 60.5% were Ascomycota, 37.5%—Basidiomycota, 1.7%—Zygomycota, and 0.2% were Chytridiomycota. Revealed mycobiota was largely composed of endophytic fungal communities as these were dominated by Cladosporium sp., Fraxinicola fraxini (syn. Venturia fraxini) and Vishniacozyma foliicola. Identified mycobiota also included a range of ash-specific fungal taxa. Hymenoscyphus fraxineus occurred in all stands but was not frequent. Cladosporium sp. showed strongest negative correlation with the presence of H. fraxineus. This ascomycete, given its dominance in leaves, shoots and in the organic soil layer, might be the limiting factor for the infection rate or spread of H. fraxineus. Although fungal communities in asymptomatic and symptomatic samples of F. excelsior differed significantly from each other, the majority of the most frequently found fungal taxa were not host-specific, suggesting that these were negligibly affected by ash dieback. Investigated stands in natural F. excelsior habitats exhibited larger diversity of fungal taxa (especially ash-specific), than progeny trials planted on former grasslands, indicating the importance of natural habitats in F. excelsior restoration programs. Recent observations suggest that climate change affects the growth conditions and range of tree species distribution in Europe. This may also have a major effect on communities of different organisms associated with these tree species. We aimed to determine whether Larix sp. could provide suitable habitats to insects and lichens associated with P. abies to conserve their biodiversity under climate change. The study sites were 10 Larix sp. and 10 P. abies forest stands in Lithuania. Both living and dead trees were included. Sticky traps, bark sheets, and exit hole methods were used for the assessment of insects. Independent plots on tree stems were established for the assessment of lichens. There were 76 and 67 different insect species on dead and living P. abies, respectively, using sticky traps. Similarly, there were 64 and 68 on dead and living Larix sp., respectively. The overall community of xylophagous insects consisted of nine and eight species, which were detected using the bark sheet and exit hole methods, respectively. The bark area colonized by lichens was 34.3% on dead P. abies and 63.2% on dead Larix sp., and 40.4% on living P. abies and 78.0% on living Larix sp. Taken together, the results demonstrate that native P. abies and introduced Larix sp. support similar diversity of stem-associated insect and lichen species. [ABSTRACT FROM AUTHOR]

Climate change, which leads to higher temperatures, droughts, and storms, is expected to have a strong effect on both health of forest trees and associated biodiversity. The aim of this study was to investigate the diversity and composition of fungal communities associated with the functional tissues and rhizosphere soil of healthy-looking Picea abies to better understand these fungal communities and their potential effect on tree health in the process of climate change. The study sites included 30 P. abies stands, where needles, shoots, roots, and the rhizosphere soil was sampled. DNA was isolated from individual samples, amplified using ITS2 rRNA as a marker and subjected to high-throughput sequencing. The sequence analysis showed the presence of 232,547 high-quality reads, which following clustering were found to represent 2701 non-singleton fungal OTUs. The highest absolute richness of fungal OTUs was in the soil (1895), then in the needles (1049) and shoots (1002), and the lowest was in the roots (641). The overall fungal community was composed of Ascomycota (58.3%), Basidiomycota (37.2%), Zygomycota (2.5%), Chytridiomycota (1.6%), and Glomeromycota (0.4%). The most common fungi based on sequence read abundance were Aspergillus pseudoglaucus (7.9%), Archaeorhizomyces sp. (3.6%), and Rhinocladiella sp. (2.0%). Pathogens were relatively rare, among which the most common were Phacidium lacerum (1.7%), Cyphellophora sessilis (1.4%), and Rhizosphaera kalkhoffii (1.4%). The results showed that the detected diversity of fungal OTUs was generally high, but their relative abundance varied greatly among different study sites, thereby highlighting the complexity of interactions among the host trees, fungi, and local environmental conditions. The horse chestnut leaf-miner (Cameraria ohridella Deschka & Dimić, Lepidoptera, Gracillariidae) is an invasive pest causing extensive damage to leaves of the horse-chestnut (Aesculus hippocastanum L.) in Europe. In Lithuania, C. ohridella invaded in 2002 causing wilting, browning and premature fall of A. hippocastanum leaves. The aim was to get a better understanding of possible linkages between foliar fungal communities and leaf-miner damage in A. hippocastanum. Leaves of A. hippocastanum, differentially damaged by C. ohridella, were collected in 10 sites in Lithuania. The fungal communities were described through DNA isolation and amplification using an ITS rRNA marker and Ion Torrent-sequencing. Clustering of 214,897 high-quality sequences resulted in 1017 non-singleton fungal taxa, among which Aureobasidium pullulans (28.2% of all fungal sequences), Endoconidioma populi (27.7%), Phoma fungicola (11.3%), Cladosporium ramotenellum (7.6%) and Cryptococcus sp. 2185_4 (5.0%) were most common. Correspondence analysis showed that fungal communities from heavily and slightly damaged leaves were largely intermingled, showing that in both types of samples fungal communities were similar. In conclusion, the study demonstrated that the phyllosphere of A. hippocastanum is inhabited by a high diversity of fungal species, the majority of which constitute generalist endophytes, epiphytes and saprotrophic fungi. The aim of this study was to assess the diversity and composition of fungal communities in damaged and undamaged shoots of Norway spruce ( Picea abies) following recent invasion of the spruce bud scale ( Physokermes piceae) in Lithuania. Sampling was done in July 2013 and included 50 random lateral shoots from ten random trees in each of five visually undamaged and five damaged 40-50-year-old pure stands of P. abies. DNA was isolated from 500 individual shoots, subjected to amplification of the internal transcribed spacer of fungal ribosomal DNA (ITS rDNA), barcoded and sequenced. Clustering of 149,426 high-quality sequences resulted in 1193 non-singleton contigs of which 1039 (87.1 %) were fungal. In total, there were 893 fungal taxa in damaged shoots and 608 taxa in undamaged shoots ( p < 0.0001). Furthermore, 431 (41.5 %) fungal taxa were exclusively in damaged shoots, 146 (14.0 %) were exclusively in undamaged shoots, and 462 (44.5 %) were common to both types of samples. Correspondence analysis showed that study sites representing damaged and undamaged shoots were separated from each other, indicating that in these fungal communities, these were largely different and, therefore, heavily affected by P. piceae. In conclusion, the results demonstrated that invasive alien tree pests may have a profound effect on fungal mycobiota associated with the phyllosphere of P. abies, and therefore, in addition to their direct negative effect owing physical damage of the tissue, they may also indirectly determine health, sustainability and, ultimately, distribution of the forest tree species. Simple Summary: Tetropium gabrieli is a longhorn beetle that usually attacks weakened larch trees. During prolonged summer droughts, T. gabrieli can form outbreaks, causing damages to larch stands. Although it is known in several European countries, in Lithuania it was detected for the first time in 2019. The detection of T. gabrieli indicates potential secondary damages to European larch and other larch species growing in the area. Tetropium gabrieli is known to be native to the Alps in Europe where it breeds in European larch (Larix decidua), but it has spread to other areas and was reported in Poland, Sweden, Denmark and Belorussia. Although T. gabrieli is considered an important secondary pest of Larix spp., it can be particularly harmful to trees subjected to abiotic stress. Here we report that in Lithuania, T. gabrieli was for the first time captured in 2019 using sticky traps attached to Larix spp. trees. Two adult beetles were trapped at two different sites in central Lithuania, and this was in the period between 10th of May and 5th of June. The production of tree seedlings in forest nurseries and their use in the replanting of clear-cut forest sites is a common practice in the temperate and boreal forests of Europe. Although conifers dominate on replanted sites, in recent years, deciduous tree species have received more attention due to their often-higher resilience to abiotic and biotic stress factors. The aim of the present study was to assess the belowground fungal communities of bare-root cultivated seedlings of Alnus glutinosa, Betula pendula, Pinus sylvestris, Picea abies and Quercus robur in order to gain a better understanding of the associated fungi and oomycetes, and their potential effects on the seedling performance in forest nurseries and after outplanting. The study sites were at the seven largest bare-root forest nurseries in Lithuania. The sampling included the roots and adjacent soil of 2–3 year old healthy-looking seedlings. Following the isolation of the DNA from the individual root and soil samples, these were amplified using ITS rRNA as a marker, and subjected to high-throughput PacBio sequencing. The results showed the presence of 161,302 high-quality sequences, representing 2003 fungal and oomycete taxa. The most common fungi were Malassezia restricta (6.7% of all of the high-quality sequences), Wilcoxina mikolae (5.0%), Pustularia sp. 3993_4 (4.6%), and Fusarium oxysporum (3.5%). The most common oomycetes were Pythium ultimum var. ultimum (0.6%), Pythium heterothallicum (0.3%), Pythium spiculum (0.3%), and Pythium sylvaticum (0.2%). The coniferous tree species (P. abies and P. sylvestris) generally showed a higher richness of fungal taxa and a rather distinct fungal community composition compared to the deciduous tree species (A. glutinosa, B. pendula, and Q. robur). The results demonstrated that the seedling roots and the rhizosphere soil in forest nurseries support a high richness of fungal taxa. The seedling roots were primarily inhabited by saprotrophic and mycorrhizal fungi, while fungal pathogens and oomycetes were less abundant, showing that the cultivation practices used in forest nurseries secured both the production of high-quality planting stock and disease control. The aim of the present study was to assess fungal and oomycete communities in the irrigation water of forest nurseries, focusing on plant pathogens in the hope of getting a better understanding of potential pathogenic microorganisms and spreading routes in forest nurseries. The study sites were at Anykščiai, Dubrava, Kretinga and Trakai state forest nurseries in Lithuania. For the collection of microbial samples, at each nursery five 100-L water samples were collected from the irrigation ponds and filtered. Following DNA isolation from the irrigation water filtrate samples, these were individually amplified using ITS rDNA as a marker and subjected to PacBio high-throughput sequencing. Clustering in the SCATA pipeline and the taxonomic classification of 24,006 high-quality reads showed the presence of 1286 non-singleton taxa. Among those, 895 were representing fungi and oomycetes. The detected fungi were 57.3% Ascomycota, 38.1% Basidiomycota, 3.1% Chytridiomycota, 0.8% Mucoromycota and 0.7% Oomycota. The most common fungi were Malassezia restricta E. Guého, J. Guillot & Midgley (20.1% of all high-quality fungal sequences), Pezizella discreta (P. Karst.) Dennis (10.8%) and Epicoccum nigrum Link (4.9%). The most common oomycetes were Phytopythium cf. citrinum (B. Paul) Abad, de Cock, Bala, Robideau, Lodhi & Lévesque (0.4%), Phytophthora gallica T. Jung & J. Nechwatal (0.05%) and Peronospora sp. 4248_322 (0.05%). The results demonstrated that the irrigation water used by forest nurseries was inhabited by a species-rich but largely site-specific communities of fungi. Plant pathogens were relatively rare, but, under suitable conditions, these can develop rapidly, spread efficiently through the irrigation system and be a threat to the production of high-quality tree seedlings. The aim was to assess fungal communities associated with living needles and soil of Pinus sylvestris in managed and unmanaged forest stands to get a better understanding of whether and how different intensities of forest management affects fungal diversity and community composition under the north temperate forest zone conditions. The study was carried out in three national parks in Lithuania. Each included five study sites in managed stands and five in unmanaged stands. At each site, three random soil cores and five random last-year needle samples were collected. Following DNA isolation, a DNA fragment of the ITS2 rRNA gene region of each sample was individually amplified and subjected to high-throughput sequencing. Analysis of 195,808 high-quality reads showed the presence of 1909 fungal taxa. Richness and composition of fungal taxa were similar in each substrate (needles and soil) in managed vs. unmanaged sites. The most common fungi in needles were Coleosporium campanulae (12.4% of all fungal sequences), Unidentified sp. 3980_1 (12.4%), Unidentified sp. 3980_4 (4.1%) and Sydowia polyspora (3.1%). In soil: Unidentified sp. 3980_21 (8.6%), Umbelopsis nana (8.2%), Archaeorhizomyces sp. 3980_5 (8.1%) and Penicillium spinulosum (6.3%). The results demonstrated that managed and unmanaged P. sylvestris stands support similar diversity and composition of fungal communities associated with living needles and soil. [ABSTRACT FROM AUTHOR]
