In section Leiopetali, the diploid chromosome numbers were reported from only six of total 25 taxa. There is no karyological data relating to other 19 taxa. In this study, it is intended to determine the diploid chromosome numbers of taxa of section Leiopetali. The diploid number of 13 species were detected, four of which were recorded for the first time and nine numbers compatible with the previous reports. Twelve species were diploid with 2n = 2x =30. D. leptopetalus was diploid and polyploid, which reveals only one polyploidy level of tetraploidy (2n = 4x = 60). Polyploidy might have played a role in the karyotype evolution of the genus. One of the most important reasons for this situation is that although polyploidy is observed in the genus Dianthus, the dysploidy mechanism that causes a change in the basic chromosome number has not been found until now. The basic number is x = 15 according to the all chromosomal reports. In conclusion, this study reports new data into the karyological characteristics of section Leiopetali (genus Dianthus) that can be useful for interpreting or understanding relationships among sections. The chromosome number and karyotype analysis of Paroncyhia amani var. amani and P. amani var. minutiflora are described for the first time. The diploid chromosome numbers and karyotype formulae are 2n = 4x = 36 = 32m + 4sm in taxa. The karyotypes are symmetrical type including metacentric and submetacentric chromosomes. The small chromosomes varied from 0.96 μm (var. amani) to 2.74 μm (var. minutiflora). P. amani var. amani and P. amani var. minutiflora are very little different varieties morphologically. There are some similarities between varieties in the karyological data. (i) equal number of chromosomes, (ii) same karyotype formula, (iii) very close karyotype asymmetry values, (iv) polyploidy by ploidy level of 4x. These seem to support morphological closeness. In this study, three hybrids of Origanum (Lamiaceae) were examined in detail using morphological, karyological, palynological and molecular approaches. One of the three hybrids, O. × aytacii, is here described for the first time, and is compared with its putative parents, O. sipyleum and O. vulgare subsp. hirtum. In addition to morphological, karyological and palynological features, we also examined nuclear ribosomal DNA (ITS) sequences of O. × intermedium (O. onites × O. sipyleum), O. × intercedens (O. onites × O. vulgare subsp. hirtum) and their purported parental species that share the same area with the new hybrid. Pollen grains were hexacolpate and isopolar. The range of Polar/Equatorial ratio (P/E) of pollen for investigated taxa varied from 0.74 to 0.99. All investigated taxa are diploid with a chromosome number as 2n = 30. According to molecular results, O. × aytacii was the most polymorphic taxon. In addition, DNA sequences showed that O. × intercedens was more similar to O. vulgare subsp. hirtum than to O. onites, and O. × intermedium was more similar to O. onites than to O. sipyleum. Furthermore, O. onites and O. vulgare subsp. hirtum have many different ITS haplotypes, probably resulting from backcrossing with different individuals. Origanum L. is one of the high-value medicinal and aromatic plants generally used for various purposes in the field of food and health. This study evaluated the effect of aqueous extract, hydrosol and essential oil forms of seven Origanum taxa including three endemic species in fresh-cut mushroom samples in order to prevent polyphenol oxidase (PPO) activity. Food samples were treated with these natural preservatives (aqueous extract, hydrosol, and essantial oil) and stored at 4°C temperature for a maximum of 5 days. It was found that the extract form of Origanum vulgare L. subsp. hirtum (Link) Ietsw. provided maximum PPO inhibition with 64.50% of reduction on 3rd day. Similarly, 3-fold reduction in hydrosol application was observed with Origanum bilgeri P. H. Davis which is the endemic plant. Chromosomal data are valuable and very useful for revealing evolution and speciation processes. Due to its wide distribution throughout the world, morphological differences, and chromosomal alterations, Erodium L'Hér. is an important genus for investigating the relationship between chromosomal alterations and karyotype evolution. In the present study, the chromosome records of 15 taxa are provided; three are reported here for the first time (E. birandianum, E. gaillardotii, and E. hendrikii), seven present new chromosome numbers, and five are in agreement with previous reports. Karyotype evolution is summarized in the light of this data, and four different genomes are presented in the genus. Millions of years ago the ancestral karyotype was x = 9 in Asia (Genome I). Then, karyotypes x = 8 (Genome II) and x = 10 (Genome III) were shaped through dysploidy in Anatolia and Asia. They were distributed in the Mediterranean Basin through the Anatolian land bridge and in North and South America via the Bering land bridge and the North Atlantic land bridge. Chromosomal data can provide very valuable information about karyotypic phylogeny and speciation. This is the first study on karyotype phylogeny and polyploidy variations of the genus Paronychia. In this context, the results are these: (1) in 14 taxa, the first report on chromosomes numbers; (2) in 2 taxa, equal chromosome numbers as in the previous report; (3) in all taxa, the first report of detailed karyotype analyses; (4) karyotype asymmetry data and generally symmetrical karyotypes; (5) karyotypic variations by mechanisms of dysploidy and polyploidy; and (6) phylogenetic relationships in Paronychia. The data indicate that Anatolia is an important area for the distribution of Paronychia. In light of all data, karyotype evolution is briefly summarized. The ancestral karyotype was x = 9 (millions of years ago). The karyotypes (x = 8 and x = 7) were then shaped by dysploidy. The rate of polyploidization then significantly increased in the genus. However, data should be supported by molecular analysis. In addition, the chromosome numbers of 8 species of Turkish Paronychia is still unknown. Chromosomal data and karyological relationships provide valuable information about karyotype evolution and speciation. For the genus Bunium, the chromosomal data are limited. In the present study, the chromosomal data of 10 taxa are provided, 6 of which are given for the first time, 2 present new chromosome numbers, and 2 agree with previous reports. Four different chromosome numbers (2n=18, 20, 22 and 40) were detected, and 2n=40 is a new number in the genus Bunium. B. brachyactis is the first polyploid species of the genus with a ploidy level of 4x. The most asymmetric karyotypes are those of B. pinnatifolium and B. sayae. Regarding karyological relationships, B. pinnatifolium forms a monophyletic group by quite different karyological features such as large chromosomes, more submedian chromosomes and the most asymmetric karyotypes. In addition, the other 5 taxa form a strong monophyletic group. B. verruculosum and B. ferulaceum are cytotaxonomically very close species, as are B. sayae and B. elegans var. elegans. The chromosome numbers of 2 Turkish species, B. nudum and B. sivasicum, remain unknown. The presented results provide important contributions to the cytotaxonomy of Bunium. [ABSTRACT FROM AUTHOR]

Based on combined results of different approaches we describe two new natural hybrids, Origanum × bilgilii Dirmenci, T. Yazıcı & Arabacı and O. × dumanii Dirmenci, Arabacı & T. Yazıcı from southern Turkey (Antalya). They have a common parent, Origanum saccatum. Morphology and pollen micromorphology of the collected plants were studied, and a comparison of one nuclear (nrITS) and one chloroplast (rpl32) DNA marker was performed. The ploidy level of the two hybrids was established by chromosome counts. Both hybrids have some morphological characters in the leaves, bracts, calyces and corollas that are intermediate between those of their parents. Surface sculpturing and pollen grain size and shape of the hybrids vary and differ to a certain degree from those of the parental species. Heterozygous loci were detected in nrITS sequences of both O. × bilgilii and O. × dumanii, while rpl32 sequences were uninformative. Stachys kurdica, well adapted to live in sloping limestone crevices and steep screes, is distributed across particular routes in Southeast Turkey and North Iraq and also extends to Iran. This species belongs to the section Fragilicaulis and contains the following infraspecific taxa; var. kurdica and var. brevidens. These taxa share partly the same habitats and are distinguished by the calyx characters. Micromorphological studies of trichomes on the epidermal surface and nutlet surface by Tabletop Scanning Electron Microscopy and Light Microscopy have revealed non-glandular and glandular trichomes on the taxa. The nutlets were coloured from light to dark-brown, usually obovate, ventral sides flat, rooflike with rib, apex rounded, faintly granulate, wrinkly, glabrous and reticulate. Karyotypes of the taxa have been introduced for the first time to the scientific community and were obtained by the Image Analysis System. The karyotype analysis showed the taxa as diploid, with chromosome numbers of 2n = 34. In addition to these morphological characters, chromosomal and micromorphological differences further serve to distinguish the taxa. Macro- and micromorphological features of fruit and seeds belonging to 12 taxa genera of Matthiola W.T.Aiton were studied via stereomicroscope and scanning electron microscope. In macromorphological studies, the following variables were investigated: the shape, size, and color of fruit and seeds; the length of fruiting pedicel; the structure of the median vein; stigma (horns); and trichome properties and density in fruit. In micromorphological studies, the presence or absence of seed wings, seed coat pattern in disc and wing, and epidermal cell shape in disc and wing were determined. Origanum ayliniae Dirmenci & Yazıcı, which differs significantly from O. dictamnus L., is described for the first time in this paper. In addition, a new hybrid, O. × adae Dirmenci & Yazıcı, between O. ayliniae and O. sipyleum L., is described and investigated in detail. The general morphology, pollen structure, chromosome features, and nuclear (ITS) and chloroplast (rpl32) genomes of collected specimens were studied. O. × adae was determined to have some intermediate properties morphologically and palynologically that place it between the parents. Pollen size and shape were important features in the diagnosis of the hybrid and its parents in this study. The somatic chromosome number of O. ayliniae, O. × adae, and O. sipyleum was counted as 2n = 30. According to the results of these different approaches, O. ayliniae differs significantly from O. dictamnus, which has a close relationship with O. ayliniae, and O. × adae has a hybrid nature. These results support previous reports for the speciation of Origanum members via hybridization. This study aimed to present a new species (O. ayliniae), a new hybrid of O. × adae, and their differences. In conclusion, O. ayliniae, O. × adae, and O. sipyleum provide significant morphological, micromorphological, palynological, cytological, and molecular information about homoploid hybridization. In this study, the morphology, anatomy, pollen morphology, leaf and seed micromorphology and karyology of the Turkish endangered endemic Physoptychis haussknechtii were investigated in detail for the first time. The species was studied in terms of detailed morphology. Basal leaves are narrowly spatulate and cauline leaves are linear to oblanceolate; both leaves are covered with flattened stalked five-armed trichomes. Raceme length was found to be up to 6 cm and 9 cm in the flowering and fruiting stages, respectively. Sepals are linear-oblong, rarely elliptic with obtuse to subacute apex. Petals are spatulate with emarginate apex. Some morphological characteristics such as dimensions of cauline leaves and ovary and length of pistil were measured for the first time. In an anatomical study, root, stem, basal leaf and cauline leaf were examined. The anatomical results showed that the plants have secondary growth in roots, primary growth in stems, amphistomatic and equifacial basal and cauline leaves with anisocytic stomata. Pollen grains are radial symmetrical, isopolar and tricolpate. The ratio of polar axis to equatorial diameter is 1.16, and pollen shape is subprolate. Exine sculpturing is reticulate under light microscope and scanning electron microscope. Seeds are obovatesuborbicular, compressed and pale brown. The chromosome number of P. haussknechtii was defined to be 2n=16. The karyotype formula of P. haussknechtii consists of five metacentric chromosome pairs and three submetacentric chromosome pairs. Bellevalia vuralii B.Şahin & Aslan is described as a new species of Bellevalia from southeast Turkey. As well as its morphological characteristics and classification with regards to sections, its description, ecology, and relationship with related species are presented and discussed. Matthiola W.T.Aiton is a taxonomically complex genus in which there are many problems, mostly with Matthiola longipetala and M. odoratissima. Matthiola species native to Turkey were collected from various locations in Anatolia, and their DNA was isolated. Revision studies performed on the basis of molecular data obtained from studies conducted in recent years have made the phylogenetic relationships and systematic positions of the taxa more apparent and reliable. Consequently, the remaining taxonomic problems among the species have been resolved through the use of DNA-based molecular analysis methods, which, unlike phenotype studies, are not affected by environmental factors. The inter-simple sequence repeat (ISSR) fingerprinting method was used in the study because its properties were considered to be more reliable and consistent than those of the randomly amplified polymorphic DNA method. DNA fragments were amplified through the use of ISSR primers. The phylogenetic relationships among the taxa were represented on a dendrogram constructed through means of NTSYSpc 2.02 software. The infrageneric and intergeneric phylogenetic relationships between Matthiola and other related genera were also characterized. It was determined that the taxa Matthiola odoratissima and M. ovatifolia are separate but closely related. Moreover, it was observed that the Matthiola longipetala complex forms a separate group within the genera. Clearly, the genera Matthiola, Sterigmostemum, Strigosella, Malcolmia, and Chorispora are phylogenetically differentiated on the dendrogram. [ABSTRACT FROM AUTHOR]

In the present study, chromosome morphology of 32 Scorzonera L. taxa, including 19 species endemic to Turkey, was analyzed. The plant materials were collected from different natural populations of Turkey between 2005 and 2011. The classification of chromosomes, the length of the long and short arm, haploid chromosome length, arm ratio, and relative chromosomal length were measured by software image analyses. The basic chromosome numbers were determined as x = 6 and x = 7. Two types of ploidy levels were observed as diploidy and tetraploidy. Karyotype asymmetry indices, TF%, As K%, Syi, Rec, A, A1, and A2 were also calculated. Scorzonera ahmet-duranii S.Makbul & Coskuncelebi, S. laciniata subsp. calcitrapifolia (Vahl) Marie, and Scorzonera suberosa C.Koch subsp. cariensis (Boiss.) Chamberlain had the most symmetrical karyotypes for the subgenera Scorzonera L., Podospermum (L.) DC., and Pseudopodospermum (Lipsch. & Krasch.) Lipsch., respectively. [ABSTRACT FROM AUTHOR]

In this study, the somatic chromosome numbers and karyotype analyses were determined for nine taxa that belong to three different genera of Senecioneae tribe. The studied taxa are; Senecio tauricolus V.A.Matthews, Senecio racemosus (M.Bieb.) DC., Senecio paludosus L., Senecio trapezuntinus Boiss., Senecio inops Boiss. & Balansa subsp. inops, Turanecio pandurifolius (K.Koch) Hamzaoğlu, Turanecio hypochionaeus (Boiss.) Hamzaoğlu, Turanecio eriospermus (DC.) Hamzaoğlu, Tephroseris integrifolia (L.) Holub. subsp. aurantiaca (Hoppe ex Willd.) B.Nord. S. tauricolus, S. trapezuntinus, S. inops subsp. inops and T. hypochionaeus taxa are endemic to Turkey. The chromosomal variability in the tribe Senecioneae has observed the existence of four chromosome numbers: 2 n = 38, 2 n = 40, 2 n = 46 and 2 n = 80. The chromosome number 2 n = 38 is a new record for Turanecio species. All taxa existed karyotypes measured by a set of median to submedian chromosomes and a smaller number of subterminal chromosome pairs. Also their karyotype asymmetry indices (TF %, As K %, Syi, Rec, A, A and A) were calculated. The ideograms and detailed chromosome morphology measurements of the species were performed by the use of image analysis system (Bs200Pro). This is the first account of somatic chromosomes and karyotypes for the studied taxa except for S. paludosus. Centaurea amaena Boiss. & Balansa included in the Phalolepis section of the Compositae (Asteraceae) family is an endemic species that is under critical risk of extinction. The morphological, anatomical, palynological, karyological and autecological features have been studied in this study. In accordance with the new observations, additional information was added to the previous description of the species, and the description was edited. Moreover, threat category of C. amaena has been updated according to International Union for Conservation of Nature criterions as CR B2ab(i,iii) (previously EN). Pollen grains are symmetric and isopolar, tricolporate. The somatic chromosome number is determined as 2 n = 18 in C. amaena. The karyotype formula of this species consists of three median chromosome pairs and six submedian chromosome pairs. In the present study, ISSR markers were employed to determine the phylogenetic relationships among the taxa of Johrenia. The genera Angelica and Xanthogalum were selected as outgroups. Unweighted pair group method with arithmetic mean (UPGMA) and Principal Coordinate Analyses were conducted to view the molecular relationships. Johrenia alpina, J. depauperatum and J. aurea are transferred to the genus Dichoropetalum. This study examined chromosome numbers and karyotypes of four taxa, Hesperis novakii Dvořák, H. bottae Fourn., H. balansae Fourn. and H. syriaca (DC.) Dvořák, from naturally growing Diaplictos (Dvořák) Dvořák section, which is represented by only these four taxa in Turkey, in the genus Hesperis. H. novakii and H. syriaca have 2n=14 chromosomes, H. bottae and H. balansae have 2n=12 chromosomes in somatic cells. Total chromosome lengths are 5.37 μm and 2.17 μm in H. novakii, 5.38 μm and 2.70 μm in H. bottae, 9.59 μm and 4.28 μm in H. balansae, 4.88 μm and 2.32 μm in H. syriaca (no. 4969), 7.89 μm and 3.44 μm in H. syriaca (no. 4971). The karyotypes are as follows: H. novakii consists of 3m+1sm+1st+2T, H. bottae consists of 4m+2st, H. balansae consists of 4m+2sm, H. syriaca (no. 4969) consists of 3m+1sm+1st+2T and H. syriaca (no. 4971) consists of 3m+1sm+1st+2T chromosome pairs.
