A compact low-energy and high-intensity x-ray source for radiation biology applications is presented. A laser-induced plasma moves inside a 30 kV diode and produces a beam of 1014 electrons at the anode location. An aluminum foil converts a part of the energy of these electrons into x-ray photons, which are characterized using filtered imaging plates. The dose that would be deposited by these x-ray photons in C. elegans larvae is calculated from Geant4 simulations. It can be set to a value ranging between 10 μGy and 10 mGy per laser shot by simply changing the aluminum foil thickness and the diode voltage. Therefore, this versatile and compact x-ray source opens a new path to explore the radiation effects induced by dose rates varying over several orders of magnitude. [ABSTRACT FROM AUTHOR]

Engineering artificial extracellular matrices, based on the biomimicry of the spatial distribution of proteins and growth factors within their native microenvironment, is of great importance for understanding mechanisms of bone tissue regeneration. Herein, photolithography is used to decorate glass surfaces with subcellular patterns of RGD and BMP-2 ligands; two mimetic peptides recognized to be involved in stem cells osteogenesis. The biological relevance of well-defined RGD and BMP-2 patterned surfaces is evaluated by investigating the differentiation of human mesenchymal stem cells (hMSCs) into osteoblasts, in the absence of induction media. The extent of hMSCs differentiation is revealed to be dependent on both the pattern shape and the ligand type. Indeed, the spatial patterning of BMP-2, but not RGD peptide, significantly enhances the extent of hMSCs differentiation, suggesting that geometric cues guide stem cells specification into specialized cells in a ligand type dependent manner. Such cell culture models provide an interesting tool to investigate how stem cells perceive and respond to their microenvironment and may contribute to the development of next-generation biomaterials capable of producing clinically relevant volume of bone tissue. Techniques that can probe the thermal properties of cells are used in many applications ranging from cryogenic preservation to hyperthermia therapy, and provide powerful tools to investigate diseased conditions. The structural complexity of cells, however, requires innovative modalities operating at a subcell scale. We developed a label-free, non-ionizing technique based on a thermoelastic lens. With this device, we captured images of single cells with a ~2 μm resolution based on thermal properties as the contrast mechanism. To investigate the thermorheological behaviour of cells, we present simultaneous acoustic imaging using an inverted opto-acoustic microscope. Acoustic impedances extracted from the acoustic images support the effusivity obtained from the thermal images. This technique should provide diagnostic tools at the single cell scale.
